Background: Peritoneal loose body (PLB) which is also called "peritoneal mouse" is a rare disease, with only a few cases reported. PLB is mostly found due to unplanned examination, surgery, or autopsy. With the increase in abdominal imaging examinations, it was gradually recognized by us. The diagnosis of PLB has three major characteristics: free movement, no blood supply, and pathological exclusion of other tumors. Case Presentation: We reported a 66-year-old male patient with an asymptomatic freely moving peritoneal loose body. The size of the lesion was about 4*4cm. The imaging data were complete including Ultrasound, Magnetic resonance (MR), Computed tomography (CT) plain scan and enhancement. Interestingly, changes in the location of the lesion were found on follow-up CT. The lesion showed concentric circles changes on CT without obvious enhancement. MR also appeared as a multi-layer signal of concentric circles. In addition, the imaging manifestations were compared with histological pathology in this article to explain imaging characteristics. Conclusion: PLB is an uncommon disease. Through the complete imaging examination of this case, we can further understand the mobility of the lesion by changing the position. The CT and MR have characteristic concentric circle changes without obvious enhancement. The diagnosis could be confirmed before surgery, which prompted the clinical early diagnosis of the disease, and an appropriate choice between surgery and conservative treatment.

BACKGROUND: Although the internal fixation technique for scoliosis is effective, there is a great risk of nail placement in actual operation. OBJECTIVE: To compare the effects of three different nail placement strategies on LenkeC patients with scoliosis under cyclic loading. METHODS: Firstly, the finite element model was established by using CT scanning images and X-ray images of patients with LenkeC scoliosis. Secondly, the validity of the model was verified. Finally, the harmonic response analysis and transient dynamic analysis were carried out. Results: The results showed that the maximum amplitude of each vertebral body appeared in the Z direction at the third natural frequency and the amplitudes of each corresponding vertebral body were very close under three kinds of nail placement. Under different nail placement methods, the uneven distribution of nails would make the model produce obvious stress concentration, but the principal stress of vertebral body and nail rod was far lower than its own yield strength. This showed that under the axial cyclic load, the vertebral bodies of scoliosis tended to have larger impact in Z direction. From the point of view of dynamics, it was feasible to reduce a certain number of nails in operation. CONCLUSION: This paper revealed that it was feasible to reduce a certain number of nails during surgery.

The current mainstay therapeutic strategy for advanced prostate cancer is to suppress androgen receptor (AR) signaling. However, castration-resistant prostate cancer (CRPC) invariably arises with restored AR signaling activity. To date, the AR ligand-binding domain (LBD) is the only targeted region for all clinically available AR signaling antagonists, such as enzalutamide (ENZ). Major resistance mechanisms have been uncovered to sustain the AR signaling in CRPC despite these treatments, including AR amplification, AR LBD mutants, and the emergence of AR splice variants (AR-Vs) such as AR-V7. AR-V7 is a constitutively active truncated form of AR that lacks the LBD; thus, it can not be inhibited by AR LBD-targeting drugs. Therefore, an approach to inhibit AR through the regions outside of LBD is urgently needed. In this study, we have discovered a novel small molecule SC428, which directly binds to the AR N-terminal domain (NTD) and exhibits pan-AR inhibitory effect. SC428 potently decreased the transactivation of AR-V7, ARv567es, as well as full-length AR (AR-FL) and its LBD mutants. SC428 substantially suppressed androgen-stimulated AR-FL nuclear translocation, chromatin binding, and AR-regulated gene transcription. Moreover, SC428 also significantly attenuated AR-V7–mediated AR signaling that does not rely on androgen, hampered AR-V7 nuclear localization, and disrupted AR-V7 homodimerization. SC428 inhibited in vitro proliferation and in vivo tumor growth of cells that expressed a high level of AR-V7 and were unresponsive to ENZ treatment. Together, these results indicated the potential therapeutic benefits of AR-NTD targeting for overcoming drug resistance in CRPC.

Streptococcus mutans, found in the human oral cavity, is a significant contributor to the pathogenesis of dental caries. This bacterium expresses three genetically distinct types of glucosyltransferases named GtfB (GTF-I), GtfC (GTF-SI) and GtfD (GTF-S) that play critical roles in the development of dental plaque. The catalytic domains of GtfB, GtfC and GtfD contain conserved active-site residues for the overall enzymatic activity that relate to hydrolytic glycosidic cleavage of sucrose to glucose and fructose, release of fructose and generation of a glycosyl-enzyme intermediate in the reducing end. In a subsequent transglycosylation step, the glucosyl moiety is transferred to the nonreducing end of an acceptor to form a growing glucan polymer chain made up of glucose molecules. It has been proposed that both sucrose breakdown and glucan synthesis occur in the same active site of the catalytic domain, although the active site does not appear to be large enough to accommodate both functions. These three enzymes belong to glycoside hydrolase family 70 (GH70), which shows homology to glycoside hydrolase family 13 (GH13). GtfC synthesizes both soluble and insoluble glucans (α-1,3 and α-1,6 glycosidic linkages), while GtfB and GtfD synthesize only insoluble or soluble glucans, respectively. Here, crystal structures of the catalytic domains of GtfB and GtfD are reported. These structures are compared with previously determined structures of the catalytic domain of GtfC. With this work, apo structures and inhibitor-complex structures with acarbose are now available for the catalytic domains of GtfC and GtfB. The structure of GtfC with maltose allows further identification and comparison of active-site residues. A model of sucrose binding to GtfB is also included. The new structure of the catalytic domain of GtfD affords a structural comparison of the three S. mutans glycosyltransferases. Unfortunately, the catalytic domain of GtfD is not complete since crystallization resulted in the structure of a truncated protein lacking approximately 200 N-terminal residues of domain IV.

Since Wuhan became a comprehensive pilot zone for cross-border e-commerce, the city has seen a significant rise in import and export trade by virtue of its superior geographical location and continued ability to sustain talent. However, with the continuous development of the digital economy, related positions have tended to become more complex and refined, and cross-border e-commerce practitioners need to continuously improve their comprehensive ability and professionalism. Although there are many universities in Wuhan, which can deliver a large number of cross-border e-commerce and related professional graduates every year, a large amount of data shows that there is a large discrepancy between the supply of talents and the demand of enterprises. This study focuses on investigating and analysing the skills of employees in cross-border e-commerce enterprises in Wuhan, through conducting surveys on supply chain management positions, trade service positions, sales positions, operation positions and design positions, to understand and analyse the skills mastery of employees working in cross-border enterprises in Wuhan, and to summarise the situation, so as to provide advice on the curriculum setting and talent training programmes of cross-border e-commerce majors in Wuhan and universities in other regions. The survey will provide advice on the adjustment of the curriculum and talent training programmes for cross-border e-commerce majors in Wuhan and other regional universities, so as to promote the better and sustainable development of cross-border e-commerce in China.

Abstract: Large volume variation during charge/discharge of silicon (Si) nanostructures applied as the anode electrodes for high energy lithium-ion batteries (LIBs) has been considered the most critical problem, inhibiting their commercial applications. Searching for alternative high performance anodes for LIBs has been emphasized. Silicon carbide (SiC) nanomaterials, a wide bandgap semiconductor with excellent mechanical properties, have been investigated as anode electrode materials even as active materials, protective layers, or inactive buffer stuff. In this mini review, we briefly summarize the synthesis of SiC nanostructures, the application of SiC/C anode materials, and SiC/Si composite anodes in LIBs.

To evaluate whether same-day discharge increased the incidence of 30-day readmission (30dR) after conventional transarterial chemoembolization (TACE) for hepatocellular carcinoma (HCC) at a single institution.In this retrospective study, 253 patients with HCC underwent 521 transarterial chemoembolization procedures between 2013 and 2020. TACE was performed with 50-mg doxorubicin/10-mg mitomycin C/5-10-mL ethiodized oil/particles. Patients not requiring intravenous pain medications were discharged after a 3-hour observation, and 30dR was tracked. The primary objective was to determine the incidence of 30dR in same-day discharge patients versus patients admitted for observation using the chi-square test. Secondary objectives assessed factors associated with overnight admission and factors predictive of 30dR using generalized estimated equation calculations and logistic regression.In the cohort, 24 readmissions occurred within 30 days (4.6%). Same-day discharge was completed after 331 TACE procedures with sixteen 30dRs (4.8%). Patients admitted overnight were readmitted 8 times after 190 TACE procedures (4.2%). This difference was not statistically significant (P = .4). Factors predicting overnight admission included female sex (58/190 [30.5%] vs 58/331 [17.5%], P.001) and tumor size of ≥3.8 cm (104/190 [55%] vs 85/190 [45%]). Factors predicting 30dR included female sex (10/116 [8.6%] vs 14/405 [0.2%]) and younger age (median [interquartile range], 63 years [55-65 years] vs 65 years [59-71 years]). At regression, factors predictive of 30dR were Child-Pugh Class B/C (odds ratio [OR], 2.1; P = .04) and female sex (OR, 2.9; P = .004).Same-day discharge after conventional TACE is a safe and effective strategy with 30dR rate of5%, similar to overnight observation.

Lung ultrasound scores (LUSs) have been demonstrated to accurately predict moderate-to-severe bronchopulmonary dysplasia (msBPD). This study attempted to explore the additional value of LUSs for predicting msBPD compared to clinical multivariate models in different gestational age (GA) groups. The study prospectively recruited preterm infants with GA P Conclusion: The contribution of LUSs to predict msBPD in infants with different GAs remains controversial and requires further investigation. What is Known:• Lung ultrasound scores (LUSs) have been demonstrated to accurately predict moderate-to-severe bronchopulmonary dysplasia in infants with gestational age (GA)＜32 weeks. What is New:• The LUSs evolution differed between extremely preterm infants born before 28 weeks and preterm infants born at 28–32 weeks of gestation.• LUSs provided similar moderate predictive performance as GA-adjusted LUS and clinical multivariate models in infants born after 28 weeks, while LUSs seem to be less helpful in infants born before 28 weeks.

Angiogenesis is vital for tumor growth and metastasis. Emerging evidence suggests that metabolic reprogramming in endothelial cells (EC) may affect angiogenesis. Here, we showed that multiple regulators in the fructose metabolism pathway, especially fructose transporter SLC2A5 and fructose-metabolizing enzyme ketohexokinase (KHK), were upregulated in tumor endothelial cells from hepatocellular carcinoma (HCC). In mouse models with hepatoma xenografts or with Myc/sgp53-induced liver cancer, dietary fructose enhanced tumor angiogenesis, tumor growth, and metastasis, which could be attenuated by treatment with an inhibitor of SLC2A5. Furthermore, vessel growth was substantially increased in fructose-containing Matrigel compared with PBS-Matrigel. Inhibiting fructose metabolism in EC cells in vivo using EC-targeted nanoparticles loaded with siRNA against KHK significantly abolished fructose-induced tumor angiogenesis. Fructose treatment promoted the proliferation, migration, and tube formation of ECs and stimulated mitochondrial respiration and ATP production. Elevated fructose metabolism activated AMPK to fuel mitochondrial respiration, resulting in enhanced EC migration. Fructose metabolism was increased under hypoxic conditions as a result of HIF1α-mediated upregulation of multiple genes in the fructose metabolism pathway. These findings highlight the significance of fructose metabolism in ECs for promoting tumor angiogenesis. Restricting fructose intake or targeting fructose metabolism is a potential strategy to reduce angiogenesis and suppress tumor growth. Significance: Fructose metabolism in endothelial cells fuels mitochondrial respiration to stimulate tumor angiogenesis, revealing fructose metabolism as a therapeutic target and fructose restriction as a dietary intervention for treating cancer.

Circular RNAs are closed endogenous RNAs that are involved in the progression of diverse tumors. Even with the most advanced combined treatments, patients with glioblastoma multiforme have a median survival time of15 months. This study aimed to investigate the roles of circular PLOD2 (circPLOD2) in glioma tumorigenesis and tumor development and to clarify its tumor-promoting effects by bioinformatics analysis and molecular experiments.To determine the characteristics of circPLOD2 expression, quantitative real-time polymerase chain reaction was conducted. Stable knockdown of circPLOD2 was implemented for functional assays. Cell Counting Kit-8 and colony formation assays were used to measure cell proliferation. Transwell assays and tube formation assays were used to evaluate cell invasion and angiogenesis abilities, respectively. An intracranial xenograft model was established to determine the function of circPLOD2 in vivo. Further biochemical and Western blot analyses were conducted to evaluate proteins associated with circPLOD2.circPLOD2 was upregulated in glioma tissues and cells. High expression of circPLOD2 was significantly associated with tumor size, World Health Organization grade, and molecular characteristics of glioma. circPLOD2 deregulation affected glioblastoma multiforme cell proliferation, invasion, and angiogenesis. Knockdown of circPLOD2 inhibited tumorigenesis in vivo. Further biochemical analysis showed that circPLOD2 was involved in oncogenic pathways and correlated with the expression of proteins related to proliferation, invasion, and angiogenesis.Our data indicate that circPLOD2 promotes glioma tumorigenesis and tumor development in vitro and in vivo and that suppressing circPLOD2 could be a novel therapeutic strategy for glioma.

Hard-to-dissolve polymers provide next-generation alternatives for high-performance filter materials owing to their intrinsically high chemical stability, superior mechanical performance, and excellent high-temperature resistance. However, the mass production of hard-to-dissolve nanofibers still remains a critical challenge. A simple, scalable, and low-cost ionic solution blow-spinning method has herein been provided for the large-scale preparation of hard-to-dissolve Nomex polymeric nanofibers with an average diameter of nearly 100 nm. After rapidly dissolving Nomex microfibers in the lithium chloride/dimethylacetamide (LiCl/DMAc) solution system, the conductive solution can be stably and conductivity-independently processed into nanofibers. The method optimizes electrospinning and avoids spinnability degradation and potential safety hazards caused by high electrical conductivity. Owing to nanofibrous structure and high dipole moment, Nomex nanofibrous filters show a stable high filtration efficiency of 99.92% for PMHard-to-dissolve nanofibers provide alternatives for high-efficiency and low-resistant air filtration but are limited by the universality and economics of fabrication methods. Background: Accumulating evidence has demonstrated the immunomodulatory effects of tumor necrosis factor-related apoptosis-inducing ligand (TRAIL) in rheumatoid arthritis and the tumor microenvironment, besides its known capacity of specifically inducing the apoptosis of cancer cells. Mice are common available animal models for studying the roles of TRAIL. However, mice express only a single TRAIL receptor (mTRAILR) with an intracellular death domain, in contrast to the two TRAIL receptors (TRAILR1 and TRAILR2) in humans. Moreover, human TRAIL binds weakly to mTRAILR, whereas mouse TRAIL has high affinity for the human TRAIL-Rs. Therefore, we considered that murine TRAIL would be more suitable than human TRAIL for exploring the immunoregulatory effect of TRAIL in immunocompetent mice or when using mouse cells as the target. To our knowledge, the detailed method for production of recombinant murine TRAIL has not been reported. Objective: In this study, we aimed to design and express two soluble forms of murine TRAIL and verify the properties of the protein. Methods: Recombinant murine TRAILs were expressed in Escherichia coli BL21 (DE3, and Ni-chelating affinity chromatography was used for protein purification. SDS-PAGE, GDS-PAGE and HPLC were applied to analyze the protein structure. The cytotoxicity of our purified murine TRAILs was evaluated in the TRAIL-sensitive human breast cancer ZR-75-30 cells and murine breast cancer 4T1 cells. Finally, validation of the tumor-killing ability of the murine protein in vivo. Results: Two soluble forms of murine TRAILs (mT_N99 and mT_N188) were purified and demonstrated with high purity and trimeric structure. In addition, Zn2+ supplement was essential to produce soluble murine TRAILs in E.coli BL21 (DE3). The two purified soluble mTRAILs showed similar cytotoxicity to cancer cells, moreover, mT_N99 also showed a good anti-tumor effect in vivo and is more suitable for the treatment of murine tumor models. Conclusion: A production approach for recombinant murine TRAIL was determined, which covered the design of shortened forms, expression, purification and characterization.

According to the alarming statistical analysis of global cancer, there are over 19 million new diagnoses and more than 10 million deaths each year. One such cancer is the oral squamous cell carcinoma (OSCC), which requires new therapeutic strategies. Ficus septica extract has been used in traditional medicine to treat infectious diseases. In this study, we examined the anti-proliferative effects of an extract of F. septica bark (FSB) in OSCC cells. Our results showed that FSB caused a concentration-dependent reduction in the viability of SCC2095 OSCC cells, as determined by 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide assays, and was less sensitive to fibroblasts. In addition, FSB induced apoptosis by activating caspases, accompanied by the modulation of Akt/mTOR/NF-κB and mitogen-activated protein kinase signaling. Moreover, FSB increased reactive oxygen species generation in a concentration-dependent manner in SCC2095 cells. Furthermore, FSB inhibited cell migration and modulated the levels of the cell adhesion molecules including E-cadherin, N-cadherin, and Snail in SCC2095 cells. Pinoresinol, a lignan isolated from FSB, showed antitumor effects in SCC2095 cells, implying that this compound might play an important role in FSB-induced OSCC cell death. Taken together, FSB is a potential anti-tumor agent against OSCC cells.

This study aimed to evaluate the effects of curcumin by co-administration of arsenic trioxide (AsUsing Pubchem database, Traditional Chinese Medicine Information Database (TCMID) database, and Swiss target prediction database to predict compound-related targets, AML-associated targets were determined using GeneCards and Online Mendelian Inheritance in Man (OMIM) databases. We identify overlapping common targets by comparing Compounds-related and AML-associated targets and using these targets to perform GO and KEGG functional enrichment analyses. Subsequently, these targets were input into the STRING database, and we used Cytoscape to construct protein-protein interaction (PPI) network. Finally, we used KG1-a cells and the AML mouse model to measure the anti-leukemia effects of curcumin and AsCompounds and targets screening hinted that 85 intersection targets were predicted in the curcumin treatment of AML, 75 targets in the AsOur results suggested that curcumin and As
