Oculocutaneous albinism (OCA) could be either non-syndromic or syndromic. There are significant challenges in clinically recognizing and differentiating Hermansky-Pudlak syndrome (HPS) from non-syndromic OCA.In a prospective consecutive case series, 63 patients (less than 18 years old) with a molecular genetic diagnosis of albinism (exceptA total of 63 patients were evaluated. Forty-five patients had non-syndromic OCA (11 OCA1B, 24 OCA2, 9 OCA4, and 1 OCA6), 5 patients had OA and 13 patients had HPS. All 3 BLOC-related HPS categories were seen (1 with BLOC1, 7 with BLOC-2 and 5 with BLOC-3 related HPS). All patients with OA were hyperopic, had darker fundus pigmentation, but had poor foveal development. All HPS patients had lighter fundus pigmentation. The degree of fundus pigmentation correlated positively with the iris pigmentation and also with the foveal development only in OCA2.Careful observation of the phenotype by comparison of the skin, hair, iris colour, with the degree of fundus pigmentation and foveal development may help clinically differentiate HPS from OCA patients of Chinese ethnicity even in the absence of any bleeding tendency.

4. Effect of Oxygen and Water on the Stability of Imipenem and Cilastatin Sodium for Injection

The study aimed to investigate the factors affecting the stability of imipenem and cilastatin sodium for injection (IMI/CIL) to improve the quality and stability in IMI/CIL preparation. In this study, the effects of headspace oxygen (HO), water content, particle shape, and particle size on the stability of IMI/CIL were investigated. IMI/CIL was purged with air, premixed oxygen/nitrogen gas (5%/95%), or high-purity nitrogen (99.999%) at 20, 5, or 2% oxygen levels to prepare IMI/CIL with different HO levels. IMI/CIL was stored at 30, 45, and 75% relative humidity for 30 days to prepare IMI/CIL with different water contents. High-performance liquid chromatography method was used for analysis. The results showed that oxygen, water, particle shape, and particle size had significant effects on the stability of IMI/CIL, and free water content is a better predictor of the safety and stability of imipenem and cilastatin sodium than the total water content. The optimization scheme of the above parameters is proposed, which significantly improves the stability of IMI/CIL. This study led to a better understanding of the degradation mechanism of imipenem and cilastatin sodium, and could provide a reference for the selection and control of IMI/CIL process conditions. This study would contribute to the development of IMI/CIL with improved stability.

Elagolix sodium is a gonadotropin-releasing hormone (GnRH) receptor antagonist that inhibits endogenous GnRH signaling by competitively binding to GnRH receptors in the pituitary gland to treat moderate to severe pain associated with endometriosis. To keep the safety and quality of the drug, a fast quantitative method by reversed-phase ultrahigh-performance liquid chromatography coupled with tandem mass spectrometry has been developed and validated for the identification, assay and estimation of potential genotoxic impurities trimethyl phosphate and triisopropyl phosphate in commercial batches of this active pharmaceutical ingredient in accordance with International Conference on Harmonization guidelines Q2 and M7. The method was validated by assessing specificity, sensitivity, linearity, the limit of quantification and detection, accuracy, precision and robustness for above analytes at a very low concentration, whose quantification and detection limits reached to 24 and 4.8 pg/ml, respectively, and the total run time for a single injection was 6 min.

7. Characterization of host and escherichia coli strains causing recurrent urinary tract infections based on molecular typing

Background Escherichia coli is the leading pathogen responsible for urinary tract infection (UTI) and recurrent UTI (RUTI). Few studies have dealt with the characterization of host and bacteria in RUTI caused by E. coli with genetically identical or different strains. This study aimed to investigate the host and bacterial characteristics of E. coli RUTI based on molecular typing. Results Patients aged 20 years or above who presented with symptoms of UTI in emergency department or outpatient clinics between August 2009 and December 2010 were enrolled. RUTI was defined as patients had 2 or more infections in 6 months or 3 or more in 12 months during the study period. Host factors (including age, gender, anatomical/functional defect, and immune dysfunction) and bacterial factors (including phylogenicity, virulence genes, and antimicrobial resistance) were included for analysis. There were 41 patients (41%) with 91 episodes of E. coli RUTI with highly related PFGE (HRPFGE) pattern (pattern similarity > 85%) and 58 (59%) patients with 137 episodes of E. coli RUTI with different molecular typing (DMT) pattern, respectively. There was a higher prevalence of phylogenetic group B2 and neuA and usp genes in HRPFGE group if the first episode of RUTI caused by HRPFGE E. coli strains and all episodes of RUTI caused by DMT E. coli strains were included for comparison. The uropathogenic E. coli (UPEC) strains in RUTI were more virulent in female gender, age E. coli RUTI. The use of fluoroquinolones was more likely associated with subsequent antimicrobial resistance in most types of antibiotics. Conclusions This study demonstrated that the uropathogens in RUTI were more virulent in genetically highly-related E. coli strains. Higher bacterial virulence in young age group (E. coli RUTI.

The high harmonic generation (HHG) by few-cycle laser pulses is essential for research in strong-field solid-state physics. Through comparison of high harmonic spectra of solids generated by laser pulses with varying durations, we discovered that lasers with good dispersion compensation are capable of producing a broad spectrum of high harmonics. As the pulse duration is further compressed, several interference peaks appear in the broad spectrum. Moreover, we conducted simulations using the semiconductor Bloch equation, considering the effect of Berry curvature, to better understand this process. Our work provides a valuable approach for studying HHG by few-cycle laser pulses in solid materials, expanding the application of HHG in attosecond physics.

Background Urinary tract infections (UTIs) caused by uropathogenic Escherichia coli (UPEC) are one of the most common outpatient bacterial infections. Although bacterial and host factors are reported to be associated with UTI pathogenesis, little is known about the host age-related differences in bacterial virulence factors and antimicrobial susceptibility. Methods PCRs were carried out to detect K1 capsule antigen, 15 virulence factors, and phylogenetic groups in E. coli isolates. Antimicrobial susceptibility of selected agents was determined by the disk diffusion method. Isolates were divided into 6 groups based on their host age. Results The results showed that virulence factors PapGII, PapGIII, Cnf1, Aer, Usp, Iha, OmpT, HlyA, and Sat, had highest frequencies in the host age group 0–3. Phylogenetic group B2 dominated in our isolates (59.6%) followed by group D (20.7%). In addition, 77.4% of strains isolated from 0 to 3 age group belonged to phylogenetic group B2. Antimicrobial susceptibility tests showed that E. coli strains isolated were significantly more resistant to antimicrobial agents as host age increased. Phylogenetic group B2 isolates were more susceptible to antimicrobial agents, compared to A, B1, and D isolates. Conclusion We found E. coli isolated from elders were more resistant to antimicrobial agents and had less virulence factors.

10. Implementation of a Four-Class Motor Imagery Brain Computer Interface Using Long Short-Term Memory Neural Network

We have developed multi-channel dry-electrode electroencephalography (EEG) system to implement a brain computer interface (BCI) and discriminate four-class motor imagery signals. The EEG channels recorded from Fz, F3, F4, C3, Cz, C4, P3, and P4 positions, according to the international 10‒20 EEG system, were acquired and wirelessly transmitted to a personal computer. Five subjects were recruited to perform right hand, left hand, right foot, and left foot imagery movements with 60 trials in each movement type. EEG data were segmented into epochs from 0 to 8 s and anchored to time points of imagery movement cues. The segmented EEG epochs were pre-processed using the Morlet wavelet. A long short-term memory (LSTM) neural network with 64 LSTM cells was constructed to discriminate EEG signals recorded from different imagery movements. Among total EEG epochs, 80% were randomly chosen as training data and the rest 20% were used as validation data. The detection accuracy reached 89%.

Background: FimH adhesin is proposed to enhance Escherichia coli kidney infection by acting with PapGII adhesin, but genetic epidemiology study and animal study have not been widely conducted to confirm this hypothesis. Methods: We compared the prevalence of adhesin gene and their coexistent pattern between upper and lower urinary tract infection (UTI) strains. fimH mutant (EC114FM), papGII mutant (EC114PM) and fimH/papGII double mutant (EC114DM) were constructed from a pylonephritogenic strain (EC114). We compared among these strains for the infection ability in bladders and kidneys of female BALB/c mice challenged transurethrally with these bacteria and assessed 1, 3, and 7 days after inoculation. 14. Accurate and Efficient SOH Estimation for Retired Batteries

There will be an increasing number of retired batteries in the foreseeable future. Retired batteries can reduce pollution and be used to construct a battery cycle ecosystem. To use retired batteries more efficiently, it is critical to be able to determine their State of Health (SOH) precisely and speedily. SOH can be estimated accurately through a comprehensive and inefficient charge-and-discharge procedure. However, the comprehensive charge and discharge is a time-consuming process and will make the SOH assessment for many retired batteries unrealistic. This paper proposes an accurate and efficient SOH Estimation (SOH-E) method using the actual data of retired batteries. A battery data acquisition system is designed to acquire retired batteries’ comprehensive discharge and charge data. The acquired discharge data are separated into various time interval-segregated sub-data. Then, the specially designed features for SOH-E are extracted from the sub-data. Neural Networks (NNs) are trained using these sub-data. The retired batteries’ SOH levels are then estimated after the NNs’ training. The experiments described herein use retired lead–acid batteries. The batteries’ rated voltage and capacity are 12 V and 90 Ah, respectively. Different feature value extractions and time intervals that might affect the SOH-E accuracy and are tested. The Backpropagation NN (BPNN) and Long-Short-Term-Memory NN (LSTMNN) are designed to estimate SOH in this paper. The experimental results indicate that SOH can be calculated in 30 min. The Root-Mean-Square Errors (RMSEs) are less than 3%. The proposed SOH-E can help decrease pollution, extend the life cycle of a retired battery, and establish a battery cycle ecosystem.

Silk fibroin has been widely used in biological fields due to its biocompatibility, mechanical properties, biodegradability, and safety. Recently, silk fibroin as a drug carrier was developed rapidly and achieved remarkable progress in cancer treatment. The silk fibroin-based delivery system could effectively kill tumor cells without significant side effects and drug resistance. However, few studies have been reported on silk fibroin delivery systems for antitumor therapy. The advancement of silk fibroin-based drug delivery systems research and its applications in cancer therapy are highlighted in this study. The properties, applications, private opinions, and future prospects of silk fibroin carriers are discussed to understand better the development of anti-cancer drug delivery systems, which may also contribute to advancing silk fibroin innovation.

19. Real-Time Cognitive Load Measurement in Classroom Environment using a Dry-Electrode EEG system

Cognitive load (CL) theory suggests that instructional materials need to be designed for reducing unnecessary CL and has been regarded as one of the most influential theories in science education. How to measure individual CL is still under investigation. In this study, we developed an eight-channel dry-electrode electroencephalogram (EEG) system and proposed an algorithm to real-time measure the depth of working memory of the N-back task in a classroom environment. The ocular artifact was removed by using the recursive least-square (RLS) method. Time-frequency analysis was applied to extract event-related theta- band activities in the artifact-suppressed EEG signals. Eight participants had the active duration for theta-band activities as 1.44±0.36 mv, 1.70±0.22 mv, and 1.97±0.04 mv for 0-back, 1-back, and 2-back tasks, respectively. In contrast to the previous research that has used spectral power of particular frequency bands as signal features, we found the detection of active duration provides better discrimination power in classifying different CL levels, compared to that of the classification using features of spectral power. The result in this study demonstrates the feasibility of theta-band EEG as an indicator to measure students’ cognitive load in a classroom environment.

The mechanism of annealing-induced amorphization of metallic glass is investigated in this study via molecular dynamics simulation. Spherical nucleuses of Cu–Ni–Al alloy with a face-centered cubic structure are embedded to simulate nanograins in Cu–Ni–Al amorphous alloy; subsequently, the material is annealed at different temperatures. The results show that the critical radius for nucleation at temperatures above the glass transition temperature (Tg) affected the behavior, grain growth, and annihilation of nanograins in the Cu–Ni–Al amorphous alloy during annealing. When the temperature increased, the critical radius for nucleation increased as well. This causes the small nanograins to annihilate quickly and the large nanograins to develop rapidly. When the annealing temperature is higher than Tg, part of the crystal nuclei, which is smaller than the critical radius, can be eliminated. The crystallinity of the metallic glass decreased, and the minimum crystallinity is attained after a period of annealing simulation. Subsequently, as the residual effective nanograins began developing, the crystallinity of the amorphous metal increased again. Therefore, the annealing duration time is critical to the crystallinity of the amorphous alloy after annealing.

Hybrid structures from metal and fiber-reinforced polymer (FRP) hybrid laminates offer an attractive solution to enhance strength and structural stiffness as well as to achieve lightweight effect. The behavior of steel-FRP hybrid laminates subject to axial compression is not well understood due to lack of experimental and analytical investigations. The axial compression performance of steel-FRP hybrid laminates was studied by using axial compression test in this study. Hybrid laminates from DP980-CFRP (carbon fiber-reinforced polymer) and DP980-AFRP (aramid fiber-reinforced polymer) were fabricated and studied. An analytical model was deduced to analyze the maximum axial compressive load of steel-FRP hybrid laminates. The results demonstrate that steel-FRP hybrid laminates exhibit significantly higher maximum compressive loads and initial stiffnesses in axial compression in comparison with the single steel sheet.

Diagnosing aging for preventative intervention generally relies on the tracking of aging biomarkers in the resting state. However, the static marker levels are insufficient to fully evaluate aging, particularly given that the stress response capacity (SRC) decay is currently viewed as a critical feature of aging. Therefore, we have developed a dual-channel fluorescent probe ROKS capable of the logic-based visualization of thiophenol (stressor) and HOCl (thiophenol-activated stress response product) in vivo, which provides a new strategy from the time dimension to precisely assess the SRC of individuals under stress using the dual-channel fluorescence ratio. Using ROKS we observed that the SRC of live cells decayed with senescence, and that a higher SRC was found for young vs. aged Caenorhabditis elegans. As such, our study offers a promising strategy for the fluorescence-guided diagnosis of aging and paves the way for accurate evaluation of the efficacy of anti-aging drugs., Rather than tracking aging using the resting state, ROKS, an optical probe, was developed for evaluating the degree of aging dynamically by precisely monitoring the stress response of individuals under stress.

A high-energy narrow-linewidth Ti:sapphire laser with widely tunable wavelength was investigated. The Littman cavity was seeded by an extended prism cavity, and they were coupled together by sharing a partial reflection mirror. The widely wavelength tunability of the prism cavity and the linewidth compression of Littman cavity were incorporated together, which resulted in a significantly increased tunable wavelength range from 720 nm to 884 nm with linewidth of less than 100 MHz. The coupling effect and the synchronization between the two cavities in temporal and spectral domain were discussed. The narrow-linewidth laser centered at 786 nm was further amplified to 36 mJ and frequency-doubled to 393 nm with pulse energy of 18.8 mJ while maintaining the narrow linewidth at a repetition rate of 10 Hz. This widely tunable narrow-linewidth laser is a promising light source for high-resolution fluorescence spectroscopy.

Due to the limited service life of new energy vehicle power batteries, a large number of waste power batteries are facing “retirement”, so it will soon be important to effectively improve the recycling and reprocessing of waste power batteries. Consumer environmental protection responsibility awareness affects the recycling of waste power batteries directly. Therefore, under the two recycling modes of new energy vehicle manufacturers and third-party recycling enterprises, this study analyzes the impact of consumer environmental protection responsibility awareness on the recycling price of waste power batteries and profit in the supply chain. The influence of factors such as recycling income, recycling input cost, and black-market recycling prices on consumer awareness of responsibility is also analyzed. Through theoretical research, it was found that: Under the model that third-party recycling enterprises are responsible for recycling, it can obtain better overall supply chain benefits; consumer environmental protection responsibility awareness and recycling benefits are positively correlated with supply chain benefits overall; and recycling benefits have a certain role in promoting consumer awareness of responsibility, while the increase in informal recycling prices inhibits consumer awareness of responsibility.

Auditory steady-state response (ASSR) is a translational biomarker for several neurological and psychiatric disorders, such as hearing loss, schizophrenia, bipolar disorder, autism, etc. The ASSR is sinusoidal electroencephalography (EEG)/magnetoencephalography (MEG) responses induced by periodically presented auditory stimuli. Traditional frequency analysis assumes ASSR is a stationary response, which can be analyzed using linear analysis approaches, such as Fourier analysis or Wavelet. However, recent studies have reported that the human steady-state responses are dynamic and can be modulated by the subject’s attention, wakefulness state, mental load, and mental fatigue. The amplitude modulations on the measured oscillatory responses can result in the spectral broadening or frequency splitting on the Fourier spectrum, owing to the trigonometric product-to-sum formula. Accordingly, in this study, we analyzed the human ASSR by the combination of canonical correlation analysis (CCA) and Holo-Hilbert spectral analysis (HHSA). The CCA was used to extract ASSR-related signal features, and the HHSA was used to decompose the extracted ASSR responses into amplitude modulation (AM) components and frequency modulation (FM) components, in which the FM frequency represents the fast-changing intra-mode frequency and the AM frequency represents the slow-changing inter-mode frequency. In this paper, we aimed to study the AM and FM spectra of ASSR responses in a 37 Hz steady-state auditory stimulation. Twenty-five healthy subjects were recruited for this study, and each subject was requested to participate in two auditory stimulation sessions, including one right-ear and one left-ear monaural steady-state auditory stimulation. With the HHSA, both the 37 Hz (fundamental frequency) and the 74 Hz (first harmonic frequency) auditory responses were successfully extracted. Examining the AM spectra, the 37 Hz and the 74 Hz auditory responses were modulated by distinct AM spectra, each with at least three composite frequencies. In contrast to the results of traditional Fourier spectra, frequency splitting was seen at 37 Hz, and a spectral peak was obscured at 74 Hz in Fourier spectra. The proposed method effectively corrects the frequency splitting problem resulting from time-varying amplitude changes. Our results have validated the HHSA as a useful tool for steady-state response (SSR) studies so that the misleading or wrong interpretation caused by amplitude modulation in the traditional Fourier spectrum can be avoided.

Understanding the regime shifts of Social–Ecological Systems (SES) and their local effects and driving factors over a long period of time is significant for future sustainability. We provide a perspective on the processes unfolding over time in order to identify the regime shifts of an SES based on changes in the relationships between the SES components. In addition, we investigate their driving factors and local effects. The applicability of this approach is demonstrated by analyzing the evolution of the SES in Guizhou Province, China, over the past 600 years. Six evolutionary phases are identified: the slow expansion of cultivation, the rapid expansion of cultivation, the continuous expansion of cultivation, the slower expansion of cultivation, the transformation of ecological protection driven by returning farmland to forest, and green development driven by urbanization. Our study establishes the empirical relationship between the state (phase) of the SES and its driving factors and effects. This study on the evolution, driving factors, and effects of the SES in Guizhou Province, China, provides an important reference for long-term regional planning and policy making.

This study addresses the challenges associated with incomplete or missing information in obstacle detection methods that employ a single sensor. Additionally, it tackles the issue of motion distortion in LiDAR point cloud data during synchronization and mapping in complex environments. The research introduces two significant contributions. Firstly, a novel obstacle detection method, named the point-map fusion (PMF) algorithm, was proposed. This method integrates point cloud data from the LiDAR, camera, and odometer, along with local grid maps. The PMF algorithm consists of two components: the point-fusion (PF) algorithm, which combines LiDAR point cloud data and camera laser-like point cloud data through a point cloud library (PCL) format conversion and concatenation, and selects the most proximate point cloud to the quadruped robot dog as the valid data; and the map-fusion (MF) algorithm, which incorporates local grid maps acquired using the Gmapping and OctoMap algorithms, leveraging Bayesian estimation theory. The local grid maps obtained by the Gmapping and OctoMap algorithms are denoted as map A and map B, respectively. This sophisticated methodology enables seamless map fusion, which significantly enhances the precision and reliability of the approach. Secondly, a motion distortion removal (MDR) method for LiDAR point cloud data based on odometer readings was proposed. The MDR method utilizes legged odometer data for linear data interpolation of the original distorted LiDAR point cloud data, facilitating the determination of the corresponding pose of the quadruped robot dog. Subsequently, the LiDAR point cloud data are then transformed to the quadruped robot dog coordinate system, efficiently mitigating motion distortion. Experimental results demonstrated that the proposed PMF algorithm achieved a 50% improvement in success rate compared to using only LiDAR or the PF algorithm in isolation, while the MDR algorithm enhanced mapping accuracy by 45.9% when motion distortion was taken into account. The effectiveness of the proposed methods was confirmed through rigorous experimentation.

34. Multi-Scenario Land Use Change Simulation and Spatial Response of Ecosystem Service Value in Black Soil Region of Northeast China

Simulating the spatial response of ecosystem service value (ESV) caused by land use change in Heilongjiang Province under different scenarios in the future is of great significance for ensuring ecological security and sustainable development in the region. Heilongjiang Province, an important grain-producing region in China, is taken as the research area in this study. Five land use maps (2000, 2005, 2010, 2015, and 2020) were used to evaluate the change of ecosystem service value in Heilongjiang Province in the past 20 years. In addition, the area of each land use type under different future scenarios was predicted by Markov model and MOP model, the future land use pattern was simulated based on PLUS model, the ESV under different scenarios was calculated, and the spatial distribution and the degree of ESV clustering were further explored. The results show that: (1) During 2000–2020, the built-up land in Heilongjiang Province continued to grow, with a total increase of 5076 km2 during the 20-year period, the area of water continued to grow, the area of unused land gradually decreased, and the area of farmland and forest land changed less. (2) During the study period, the ESV in 2000, 2005, 2010, 2015, and 2020 were 1320.8 billion yuan, 1334.5 billion yuan, 1342.1 billion yuan, 1327.6 billion yuan, and 1328.4 billion yuan, respectively. Generally, it shows a fluctuating trend. (3) The ESV of natural development scenario (NDS), economic priority scenario (ERS) and ecological protection scenario (EPS) are 1334.3 billion yuan, 1254.8 billion yuan and 1455.6 billion yuan, respectively. The ESV of different scenarios is quite different. (4) The spatial distribution of ESV was higher in the northwest, central, and southeast, and lower in the east and west. The hot and cold spots of ESV are widely distributed and the degree of polymerization is high. The methods and conclusions of this study can provide scientific reference for the optimization of national spatial pattern and the formulation of sustainable development policy.

35. Three-Stage Dead-Time Adjustment Scheme for Conversion Efficiency Enhancement of Phase-Shift Full-Bridge Converters at Light Loads

This article proposes a three-stage dead-time adjustment scheme for the leading-leg and lagging-leg switches of phase-shift full-bridge converter (PSFBC) according to the load conditions to enhance the conversion efficiencies at light loads. This article also investigates an iterative rigorous loss calculation for light loads under dead-time adjustment. The proposed iterative calculation can be used to modify the duty ratio of the transition region and estimate the operating currents more accurately; therefore, the losses of a PSFBC at light loads and under the dead-time adjustments can be effectively estimated. A PSFBC prototype with an input voltage of 380 V, an output voltage of 48 V, and a rated output power of 480 W is designed and implemented in this article. Experimental results show that comparing with the PSFBC without the dead-time adjustment and with the conventional dead-time adjustment, the conversion efficiency enhancements of 12.13% and 5.97% can be achieved by the proposed three-stage dead-time adjustment scheme at about 5% load condition, respectively. For a small-scale data center with 100 designed PSFBCs, simulated results indicate that about 3.21 MWh of electricity and 1.03 tons of carbon dioxide emission can be saved and reduced per year, respectively, by the proposed dead-time adjustment scheme.

Recurrent urinary tract infection (RUTI) can damage renal function and has impact on healthcare costs and patients’ quality of life. There were 2 stages for development of prediction models for RUTI. The first stage was a scenario in the clinical visit. The second stage was a scenario after hospitalization for urinary tract infection caused by Escherichia coli. Three machine learning models, logistic regression (LR), decision tree (DT), and random forest (RF) were built for the RUTI prediction. The RF model had higher prediction accuracy than LR and DT (0.700, 0.604, and 0.654 in stage 1, respectively; 0.709, 0.604, and 0.635 in stage 2, respectively). The decision rules constructed by the DT model could provide high classification accuracy (up to 0.92 in stage 1 and 0.94 in stage 2) in certain subgroup patients in different scenarios. In conclusion, this study provided validated machine learning models and RF could provide a better accuracy in predicting the development of single uropathogen (E. coli) RUTI. Both host and bacterial characteristics made important contribution to the development of RUTI in the prediction models in the 2 clinical scenarios, respectively. Based on the results, physicians could take action to prevent the development of RUTI.

Rationale and ObjectiveGut microbiota have been targeted by alternative therapies for non-communicable diseases. We examined the gut microbiota of a healthy Taiwanese population, identified various bacterial drivers in different demographics, and compared them with dialysis patients to associate kidney disease progression with changes in gut microbiota.Study DesignThis was a cross-sectional cohort study.Settings and ParticipantsFecal samples were obtained from 119 healthy Taiwanese volunteers, and 16S rRNA sequencing was done on the V3-V4 regions to identify the bacterial enterotypes. Twenty-six samples from the above cohort were compared with fecal samples from 22 peritoneal dialysis and 16 hemodialysis patients to identify species-level bacterial biomarkers in the dysbiotic gut of chronic kidney disease (CKD) patients.ResultsSpecific bacterial species were identified pertaining to different demographics such as gender, age, BMI, physical activity, and sleeping habits. Dialysis patients had a significant difference in gut microbiome composition compared to healthy controls. The most abundant genus identified in CKD patients was Bacteroides, and at the species level hemodialysis patients showed significant abundance in B. ovatus, B. caccae, B. uniformis, and peritoneal dialysis patients showed higher abundance in Blautia producta (p ≤ 0.05) than the control group. Pathways pertaining to the production of uremic toxins were enriched in CKD patients. The abundance of the bacterial species depended on the type of dialysis treatment.ConclusionThis study characterizes the healthy gut microbiome of a Taiwanese population in terms of various demographics. In a case-control examination, the results showed the alteration in gut microbiota in CKD patients corresponding to different dialysis treatments. Also, this study identified the bacterial species abundant in CKD patients and their possible role in complicating the patients’ condition.

Single domain spectral/pseudospectral integration preconditioning matrices have been shown to be effective operators for solving differential equations. In this study we extend the integration precondition methodology to a multidomain computational framework, and construct global inverse matrices for the advection operator discretized by multidomain Gauss-Lobatto-Legendre pseudospectral methods. These inverse operators can be used as solution operators for solving first order boundary value problems even with piecewise continuous variable coefficients. Moreover, they are effective integration preconditioning matrices for the second order diffusion operator, in the sense that a model diffusion problem can be solved by an iterative method with the number of iteration steps being proportional to the number of subdomains but independent of the degree of the solution polynomial. Numerical experiments were conducted and we observed the performance of the inverse operators as expected.

Resilience, which has rapidly become an area of interest in multiple disciplines, is regarded as being key in disaster mitigation and adaptation. The objective-indicator framework is a common way to evaluate resilience, but limited attention has been paid to measuring the risk perceptions and adaptation behaviors of individuals. In addition, due to limitations related to predicting potential earthquake events, past studies have placed more emphasis on predisaster discussions. Fortunately, this paper explores the changes in risk perceptions and adaptation behaviors in different socioeconomic groups through a comparative analysis between pre- and postearthquake disasters and through one-way analysis of variance (ANOVA) with a post hoc test applied to examine the changes in risk perceptions and adaptation behaviors. The results show that people tend to have greater risk perceptions of future earthquakes but are less willing to retrofit their houses after a serious disaster. Females show greater fear and worry accompanied by a higher willingness to retrofit their houses compared to males. In addition, people with a higher education level and a better occupation might be more willing than others to adopt adaptation behaviors. The results can serve as a reference to provide risk communication, risk education, and diverse disaster adaptation options. Although limitations exist, the results of comparative analysis between the predisaster and postdisaster conditions could serve as a reference for adequate strategies and government decisions on the prioritization of risk management policies.

The design of hybrid structure offers an attractive solution to enhance strength and structural stiffness as well as to achieve lightweight effect and cost reduction. The applications of steel–FRP (fiber-reinforced polymer) composites in transportation and civil engineering have been comprehensively reviewed. In order to apply hybrid structures to car body parts such as B-pillar, flexural performance of steel–FRP composites is investigated by means of three-point bending test in this study. An analytical model is deduced to calculate the initial stiffness, the bending load and the energy absorption of steel–FRP composites. Steel–CFRP (carbon fiber-reinforced polymer) and steel–AFRP (aramid fiber-reinforced polymer) composites are experimentally studied and discussed. The results demonstrate that the steel–FRP composites exhibit significantly higher load-carrying capabilities and initial stiffnesses along with larger energy absorptions in the bending process compared to the single steel sheet.

All patients underwent D2 lymphadenectomy in the hospital. A prognostic nomogram based on the integration of traditional prognostic factors and NLR (neutrophil-to-lymphocyte ratio) and PLR (platelet-to-lymphocyte ratio) was established and compared with the nomogram based on the traditional prognostic factors alone. ROC curves were further applied to verify the predictive accuracy of the established model.Results: Both NLR (P=0.00) and PLR (P=0.01) predicted the 5-year OS. In multivariate analysis, age, T3 category, T4 category, N2 category, N3 category, Pgp (P-glycoprotein), NLR and PLR are proven to be independent (all P≤ 0.05). The established nomogram showed better predictive power than that of traditional profile (c-index: 0.66 versus 0.63) in both training and validation cohorts. External assessment by ROC curve analysis demonstrated that the established model had a good prediction accuracy of 5-year OS in stage III CRC patients, with area under curve values of 0.657 and 0.629 in training and validating sets, respectively.Conclusion: A nomogram based on the integration of traditional prognostic factors and inflammatory markers (NLR and PLR) could provide more precise long-term prognosis information for lymph node-metastatic CRC patients than the model based on traditional profile alone. Spectral/pseudospectral integration preconditioning matrices are useful tools for solving differential equations involving pure differential operators d m / d x m . In this study we construct well-conditioned inverse pseudospectral matrices for the basic differential operator and the mixed differential operator d d r a ( r ) d d r on Gauss-Radau-Legendre points based on stepwise integrations and low-rank updates. The inverse matrices can be used either as a solution operator or an effective preconditioner for variable coefficient differential equations of first and second order in polar coordinates. Numerical experiments were conducted and we observed the performance of the inverse operator as expected.

Electric load analysis is an important task in early new stage for design of shipboard power systems (SPSs). The installation generation capacity onboard depends on the accurate result of the analysis, which significantly affects the system economic and secure operations. In the traditional electrical load analysis, the demand factor is often obtained from engineering experiences which may lead to inaccurate analysis results. In order to obtain better results, a methodology for electrical load analysis in SPSs is proposed in this article. The load analysis spreadsheet is adopted to find the power consumption of key electric loads in SPSs. The actual power consumptions of selected loads are collected by intelligent meters. The statistical analysis is then performed to find the critical parameter of each selected load in the load analysis under different operating conditions based on the power measurement of field test and load analysis is performed for estimating system power demand accordingly. The large practical maritime vessel power systems are selected for numerical analysis to ensure the performance of the proposed method. The proposed method has been adopted by local ship builders in Taiwan to better understand the load characteristics in shipboard power systems to support various functions of system planning and operation in a more effective manner.

The porous and biomimetic cobalt silicate@diatomite (Co2SiO4@diatomite) was successfully synthesized by a two-step method, including the hydrothermal method and calcination to improve the electromagnetic wave absorption property. Different hydrothermal times were well-tuned for Co2SiO4@diatomite composites with different loadings of Co2SiO4. Interestingly, the Co2SiO4@diatomite composites (6 h, 25 wt%) had a smaller minimum reflection loss. Moreover, the minimum reflection loss (RLmin) could reach −12.03 dB at 16.64 GHz and the matched absorber thickness was 10 mm, while the effective absorption bandwidth (EAB, RL ≤ −10 dB) could be 1.92 GHz. In principle, such findings indicate that Co2SiO4@diatomite nanocomposites could be a promising candidate for high-efficiency microwave absorption capability.

50. Niclosamide Revitalizes Sorafenib through Insulin-like Growth Factor 1 Receptor (IGF-1R)/Stemness and Metabolic Changes in Hepatocellular Carcinoma

Sorafenib is the first approved systemic targeting agent for advanced HCC; however, when used alone, drug resistance can result in considerably reduced efficacy. Here, we demonstrate that niclosamide, an antihelminthic agent approved by the US Food and Drug Administration, can be repurposed to increase sorafenib sensitivity in sorafenib-resistant HCC cells. We generated sorafenib-resistant HCC cell lines (HepG2215_R and Hep3B_R) with elevated IGF-1R levels and strong properties in terms of stemness and epithelial–mesenchymal transition. Niclosamide was found to increase sorafenib sensitivity effectively in both cell lines and their organoids. The underlying mechanism involves the modulation of cancer stemness, IGF-1R/p-IGF1R/OCT4, and metabolic changes. The combination of sorafenib and niclosamide, but not linsitinib, effectively suppressed the IGF-1R/OCT4 expressions, yielded a synergistic combination index (CI), and attenuated stemness-related properties such as secondary tumor sphere formation and cell migration in sorafenib-resistant HCC cells. Notably, niclosamide significantly suppressed the sorafenib-induced IGF-1R phosphorylation prompted by IGF-1 treatment. Niclosamide effectively downregulated the sorafenib-induced gene expression associated with glycolysis (GLUT1, HK2, LDHA, and PEPCK), stemness (OCT4), and drug resistance (ABCG2) and enhanced the ability of sorafenib to reduce the mitochondrial membrane potential in vitro. The synergistic effect of a combination of niclosamide and sorafenib in vivo was further demonstrated by the decreased tumor size and tumor volume resulting from apoptosis regulation. Our results suggest that niclosamide can enhance sorafenib sensitivity in sorafenib-resistant HCC cells through IGF-1R/stemness regulation and metabolic changes. Our findings highlight a practical clinical strategy for enhancing sorafenib sensitivity in HCC.
