Origanum minutiflorum, an endemic species in Turkiye, is used for both food and medicinal purposes. 70% ethanol extract of aerial parts of O. minutiflorum was subjected to liquid-liquid extraction using; n-hexane, dichloromethane, ethyl acetate and n-butanol. The inhibitory activity of these fractions on lipoxygenase (LOX), cyclooxygenase (COX) and matrix metalloproteinase-9 (MMP-9) was investigated at 100 µg/mL concentration. The LOX enzyme was most effectively inhibited by the dichloromethane fraction (82.33%), whereas the COX-1, COX-2 and MMP-9 enzymes were most effectively inhibited by the ethyl acetate fraction (78.50%, 83.96% and 45.11%, respectively). The ethyl acetate fraction was selected for activity-guided fractionation based on the COX enzymes’ inhibitory activities by column chromatography. Only the ethyl acetate sub-fraction-1 showed inhibition on the COX-1 with 65.78%. The COX-2 inhibitory activities of the sub-fractions ranged from 59.96% to 97.62%. The main components were determined by HPLC-MS/MS as jaceidine in the dichloromethane fraction and rosmarinic acid in the ethyl acetate fraction. As far as we know, this study is the first to correlate the anti-inflammatory activity of O. minutiflorum extracts with LOX, COX and MMP-9 enzyme pathways and their secondary metabolites. These data showed that O. minutiflorum may contribute to other studies on natural origin anti-inflammatory agents.

We describe the earliest measurements of the DT fusion cross section commissioned by the Manhattan Project, first at Purdue University in 1943 and then at Los Alamos 1945-6 and later, in 1951-2. The Los Alamos measurements led to the realization that a 3/2$^+$ resonance in the DT system enhances the fusion cross section by a factor of one hundred at energies relevant to applications. This was a transformational discovery, making the quest for terrestrial fusion energy possible. The earliest measurements were reasonably accurate given the technology of the time and the scarcity of tritium, and were quickly improved to provide cross section data accurate to just a few percent. We provide a previously-unappreciated insight: that DT fusion was first reported in Ruhlig's 1938 University of Michigan experiment and likely influenced Konopinski in 1942 to suggest its usefulness for thermonuclear technologies. We report on preliminary work to repeat the 1938 measurement, and our simulations of that experiment. We also present some work by Fermi, from his 1945 Los Alamos lectures, showing that he used the S-factor concept about a decade before it was introduced by nuclear astrophysicists., 10 pages, 8 figures

Why was recovery from the euro area crisis delayed for a decade? The explanation lies in the absence of credible and timely policies to backstop financial intermediaries and sovereign debt markets. In this paper we add light and color to this analysis, contrasting recent experience with the 1992-3 crisis in the European Monetary System, when national central banks and treasuries more successfully provided this backstop. In the more recent episode, the incomplete development of the euro area constrained the ability of the ECB and other European institutions to do likewise.

Significant progress has been made to experimentally determine a complete set of the parity-violating (PV) weak-interaction amplitudes between nucleons. In this paper we describe the design, construction and operation of the n$^3$He experiment that was used to measure the PV asymmetry $A_{\mathrm{PV}}$ in the direction of proton emission in the reaction $\vec{\mathrm{n}} + {^3}\mathrm{He} \rightarrow {^3}\mathrm{H} + \mathrm{p}$, using the capture of polarized cold neutrons in an unpolarized gaseous $^3\mathrm{He}$ target. This asymmetry has was recently calculated \cite{Viviani,Viviani2}, both in the traditional style meson exchange picture, and in effective field theory (EFT), including two-pion exchange. The high precision result (published separately) obtained with the experiment described herein forms an important benchmark for hadronic PV (HPV) theory in few-body systems, where precise calculations are possible. To this day, HPV is still one of the most poorly understood aspects of the electro-weak theory. The calculations estimate the size of the asymmetry to be in the range of $(-9.4 \rightarrow 3.5)\times 10^{-8}$, depending on the framework or model. The small size of the asymmetry and the small overall goal uncertainty of the experiment of $\delta A_{\mathrm{PV}} \simeq 1\times10^{-8}$ places strict requirements on the experiment, especially on the design of the target-detector chamber. In this paper we describe the experimental setup and the measurement methodology as well as the detailed design of the chamber, including results of Garfield++ and Geant4 simulations that form the basis of the chamber design and analysis. We also show data from commissioning and production and define the systematic errors that the chamber contributes to the measured $A_{\mathrm{PV}}$. We give the final uncertainty on the measurement.

We report the first precision measurement of the parity-violating asymmetry in the direction of proton emission with respect to the neutron spin, in the reaction $^{3}\mathrm{He}(\mathrm{n},\mathrm{p})^{3}\mathrm{H}$, using the capture of polarized cold neutrons in an unpolarized active $^3\rm{He}$ target. The asymmetry is a result of the weak interaction between nucleons, which remains one of the most poorly understood aspects of electro-weak theory. The measurement provides an important benchmark for modern effective field theory (EFT) calculations. Measurements like this are necessary to determine the spin-isospin structure of the hadronic weak interaction. Our asymmetry result is $A_{PV} = \left( 1.58 \pm 0.97 ~\mathrm{(stat)} \pm 0.24~\mathrm{(sys)}\right)\times10^{-8}$, which has the smallest uncertainty of any parity-violating asymmetry measurement so far.

Background In collaboration with state, tribal, local, and territorial health departments, CDC established the U.S. Zika Pregnancy Registry (USZPR) in early 2016 to monitor pregnant women with laboratory evidence of possible recent Zika virus infection and their infants. Methods This report includes an analysis of completed pregnancies (which include live births and pregnancy losses, regardless of gestational age) in the 50 U.S. states and the District of Columbia (DC) with laboratory evidence of possible recent Zika virus infection reported to the USZPR from January 15 to December 27, 2016. Birth defects potentially associated with Zika virus infection during pregnancy include brain abnormalities and/or microcephaly, eye abnormalities, other consequences of central nervous system dysfunction, and neural tube defects and other early brain malformations. Results During the analysis period, 1,297 pregnant women in 44 states were reported to the USZPR. Zika virus-associated birth defects were reported for 51 (5%) of the 972 fetuses/infants from completed pregnancies with laboratory evidence of possible recent Zika virus infection (95% confidence interval [CI] = 4%-7%); the proportion was higher when restricted to pregnancies with laboratory-confirmed Zika virus infection (24/250 completed pregnancies [10%, 95% CI = 7%-14%]). Birth defects were reported in 15% (95% CI = 8%-26%) of fetuses/infants of completed pregnancies with confirmed Zika virus infection in the first trimester. Among 895 liveborn infants from pregnancies with possible recent Zika virus infection, postnatal neuroimaging was reported for 221 (25%), and Zika virus testing of at least one infant specimen was reported for 585 (65%). Conclusions and implications for public health practice These findings highlight why pregnant women should avoid Zika virus exposure. Because the full clinical spectrum of congenital Zika virus infection is not yet known, all infants born to women with laboratory evidence of possible recent Zika virus infection during pregnancy should receive postnatal neuroimaging and Zika virus testing in addition to a comprehensive newborn physical exam and hearing screen. Identification and follow-up care of infants born to women with laboratory evidence of possible recent Zika virus infection during pregnancy and infants with possible congenital Zika virus infection can ensure that appropriate clinical services are available.

Objectives. The prevalence of atherosclerotic risk factors and disease in Takayasu's arteritis (TAK) has not been well defined. We aimed to assess the frequency of cardiovascular (CV) risk factors and the incidence of CV events (CVEs) in patients with TAK from two ethnically different populations.

The PREDICTS project—Projecting Responses of Ecological Diversity In Changing Terrestrial Systems (www.predicts.org.uk)—has collated from published studies a large, reasonably representative database of comparable samples of biodiversity from multiple sites that differ in the nature or intensity of human impacts relating to land use. We have used this evidence base to develop global and regional statistical models of how local biodiversity responds to these measures. We describe and make freely available this 2016 release of the database, containing more than 3.2 million records sampled at over 26,000 locations and representing over 47,000 species. We outline how the database can help in answering a range of questions in ecology and conservation biology. To our knowledge, this is the largest and most geographically and taxonomically representative database of spatial comparisons of biodiversity that has been collated to date; it will be useful to researchers and international efforts wishing to model and understand the global status of biodiversity.

In a pilot study, seven patients with multiple sclerosis were treated with CAMPATH-IH which targets the CD52 antigen present on lymphocytes and monocytes. There was a substantial reduction in disease activity as measured by gadoliunium-enhancing lesions on MRI. Encouraged by this result a further seven patients have been treated with CAMPATH-IH; four also received anti-CD4 antibody. Lymphopaenia developed rapidly and was sustained for at least one year. In 12 patients, the first infusion of antibody was characterised by significant exacerbation or re-awakening of pre-existing symptoms lasting several hours. These clinical effects of antibody treatment correlated with increased levels of circulating cytokines. Peak levels of tumour necrosis factor alpha (TNF alpha) and interferon gamma (IFN gamma) occurred at 2 h whereas the rise in interleukin-6 (IL-6) was significantly delayed and peaked at 4 h after starting antibody treatment. The neurological symptoms could not be attributed directly to pyrexia and were not provoked (in one patient) by an artificial rise in temperature. In the remaining two patients, a single pre-treatment with intravenous methylprednisolone (500 mg) prevented both the transient increase in neurological symptoms and the cytokine release. Our results suggest that soluble immune mediators contribute to symptom production in multiple sclerosis by directly or indirectly blocking conduction through partially demyelinated pathways.

Between January 1985 and March 1992, 48 patients with chronic phase CML underwent BMT from volunteer unrelated donors (MUD) serologically identical at HLA-A, B and DR loci. 19 patients received donor marrow ex vivo T-cell depleted (EX-TCD) with Campath monoclonal antibodies. 29 patients received unmanipulated donor marrow with CsA/MTX GVHD prophylaxis; 28 received additional intravenous antilymphocyte therapy from day +1 to +5 (IN-TCD). Overall 26 patients survive at median follow up of 362 days; actuarial survival at 3 years is 50%. 3 patients have sustained haematological relapse; actuarial leukaemia-free survival is 38%. There is no difference in overall survival between the EX-TCD and IN-TCD groups, but primary graft failure (n = 4) occurred only in the EX-TCD group, while GVHD (grade II or greater) occurred more frequently in the IN-TCD group (61% vs. 29%, p = 0.084). The optimum method for GVHD prophylaxis in MUD BMT remains uncertain.

Autologous transplantation has an established role in the treatment of lymphoproliferative disorders, but allogeneic transplantation remains controversial. In an attempt to reduce the high procedure-related mortality reported with allografting in lymphoma, we have used BEAM (BCNU, etoposide, cytarabine and melphalan), a standard conditioning regimen for autologous transplantation. As BEAM may be insufficiently immunosuppressive to permit durable engraftment in the allogeneic setting, patients received additional pretransplant immunosuppression with the anti-CD52 antibody CAMPATH-1G from day -5 to day -1. Twelve patients (median age 46 years) underwent allogeneic transplantation for lymphoma (n = 11) or chronic lymphocytic leukaemia (n = 1) from HLA-identical (n = 9) or mismatched (n = 3) sibling donors. Cyclosporin A and methotrexate were used as graft-versus-host disease (GVHD) prophylaxis. One patient died of progressive lymphoma at day +12, the remaining 11 patients engrafted rapidly, with eight demonstrating full donor chimerism. One patient had an episode of rejection and received a further stem cell infusion with sustained recovery. Only one patient developed GVHD (grade I). The low incidence of acute GVHD may be in part related to persisting levels of in vivo CAMPATH-IG at the time of transplantation. Of 11 evaluable patients, nine achieved complete remission (CR), and a further patient achieved CR after donor lymphocyte infusion at 5 months. Our preliminary experience is that this regimen was well tolerated with a low risk of GVHD and appears no more toxic than a BEAM autograft. Further follow-up is required to see whether the low incidence of GVHD impacts upon relapse risk.

Fifty-eight consecutive children with high-risk malignancies were treated with CY, and targeted topotecan followed by autologous hematopoietic cell transplantation (AHCT) in a phase I/II Institutional Review Board-approved study. Twelve participants enrolled in phase I; 5 received dose level 1 of topotecan 3 mg/m(2) per day, with subsequent doses targeted to total systemic exposure of 100±20 ng h/mL and CY 750 mg/m(2) per day. Seven participants received dose level 2. CY dose escalation to 1 g/m(2) per day was considered excessively toxic; one died from irreversible veno-occlusive disease and two experienced reversible hepatotoxicity. These adverse events halted further dose escalation. A total of 46 participants were enrolled in phase II; results are on the 51 participants who received therapy at dose level 1, the maximum tolerated dose. Diagnoses included neuroblastoma (26), sarcoma (9), lymphoma (8), brain tumors (5), Wilms (2) and retinoblastoma (1). Twenty participants (39.3%) were in CR1 at enrollment; median age was 5.1 years. Most common non-hematological grade III-IV toxicity was gastrointestinal (n=37). Neutrophil and platelet engraftment occurred at a median of 15 and 24 days, respectively. Twenty-six (51%) participants remain alive at a median of 6.4 years after AHCT. CY 3.75 g/m(2), and targeted topotecan followed by AHCT are feasible and produce acceptable toxicity in children with high-risk malignancies.Bone Marrow Transplantation advance online publication, 19 March 2012; doi:10.1038/bmt.2012.51.

Aims/Hypothesis Insulin resistance has been proposed as a risk factor for type 1 diabetes. We investigated whether adiponectin, an insulin sensitiser, can serve as an additional predictive marker for type 1 diabetes in first-degree relatives of known patients. Methods Adiponectin was followed in 211 persistently islet antibody-positive (Ab+) first-degree relatives of type 1 diabetic patients and in 211 age- and sex-matched persistently antibody-negative relatives, and correlated with antibody status, random proinsulin:C-peptide ratio and HLA-DQ genotype. During follow-up, 37 Ab+ relatives developed type 1 diabetes. Results In the group of 422 relatives, baseline adiponectin correlated inversely with age and BMI and was lower in male than in female participants, especially after 15 years of age (pConclusions/Interpretation Adiponectin levels do not contribute to the prediction of type 1 diabetes in Ab+ relatives.

Work is reported here on the process and present results of an international evaluation of the neutron cross section standards. The evaluations include the Hn,n, 3Hen,p, 6Lin,t, 10Bn, 10Bn,1, 197Aun,235Un,f, and 238Un, f standard reactions as well as the 238Un, and 239Pun,f reactions. This evaluation was performed to include new experiments on the standards that have been made since the ENDF/B-VI evaluation was completed and to improve the evaluation process. Evaluations have been completed for the 6Lin,t, 197Aun,and 238Un, cross sections. Also below 20 MeV the Hn,n, 235Un,f, 238Un,f and 239Pun,f cross sections are completed. Many of the cross sections being evaluated are used in neutron dosimetry for fluence determination. The general trend observed for the evaluations is an increase in the cross sections for most of the reactions from fractions of a percent to several percent compared with the ENDF/B-VI results., JRC.D.5-Neutron physics
