A simple direct method for the synthesis of lumazine-6,7-diamines (9, 25-27) from uracil-5,6-diamines (10-13) and methyl cyanoformimidate (24) was found. Various nucleophilic displacement reactions were performed with 7-chloro-1,3-dimethyllumazine-6-amine (17) leading to new 6,7-disubstituted 1,3- dimethyllumazine derivatives (18-23).

The pathophysiology of Ventricular Septal Defect (VSD) is determined by the size of the defect and the state of the pulmonary vascular resistance. We assessed the morphology of VSD using 3D transthoracic echocardiography (3D-TTE) and the ability to estimate the pathophysiology.MethodsWe prospectively enrolled 48 children with isolated unique muscular (77%) and membranous VSD (23%). Severity of the VSD was rated according to their pathophysiology. (Type 1: minor left-to-right shunt; 2a: significant left-to-right shunt (left ventricular end diastolic diameter (LVEDD) Zscore >2); 2b: VSD associated with pulmonary hypertension). VSD measurements were obtained after a multi-planar reconstruction of a TTE 3D full volume (X5-1 or X7-2 matrix probes, ie33, Philips). Diagnosis properties of the ratio of 3D-VSD area to aortic annulus area (3DA/AAA), 3D-VSD area to body surface area (3DA/BSA) and 2Dmax diameter to BSA (2DD/BSA) were compared.ResultsMedian age was 8.5 month-old (min 1 max 123). LVEDD Zscore was >2 in 19 children (40%), of whom 12 (25%) had pulmonary hypertension. 3D-VSD systolic area, 3D and 2D maximal diameters were correlated with LVEDD. (r=0.71, r=0.52, r=0.55, p0.39 has a sensitivity of 92% and a specificity of 97% to diagnose a 2b-VSD.Conclusion3D-TTE allows a morphological and a quantitative assessment of muscular and membranous VSD. Dimethyl heptalene-4,5-dicarboxylates† undergo preferentially a Michael addition reaction at C(3) with α-lithiated alkyl phenyl sulfones at temperatures below −50°, leading to corresponding cis-configured 3,4-dihydroheptalene-4,5-dicarboxylates (cf. Table 1, Schemes 3 and 4). The corresponding heptalenofuran-1-one-type pseudoesters of dimethyl heptalene-4,5-dicarboxylates (Scheme 5) react with [(phenylsulfonyl)methyl]lithium almost exclusively at C(1) of the furanone group (Scheme 6). In contrast to this expected behavior, the uptake of 1-[phenylsulfonyl)ethyl]lithium occurs at C(5) of the heptalenofuran-1-ones as long as they carry a Me group at C(11) (Schemes 6 and 7). The 1,4- as well as the 1,6-addition products eliminate, on treatment with MeONa/MeOH in THF, benzenesulfinate, thus leading to 3- and 4-alkylated dimethyl heptalene-4,5-dicarboxylates, respectively (Schemes 8–13). The configuration of the addition reaction of the nucleophiles to the inherently chiral heptalenes is discussed in detail (cf. Schemes 14–19) on the basis of a number of X-ray crystal-structure determinations as well as by studies of the temperature-dependence of the 1H-NMR spectra of the addition products.
