Abstract

Systemic sclerosis (SSc) is an autoimmune disease that shows one of the highest mortality rates among rheumatic diseases. We perform a large genome-wide association study (GWAS), and meta-analysis with previous GWASs, in 26,679 individuals and identify 27 independent genome-wide associated signals, including 13 new risk loci. The novel associations nearly double the number of genome-wide hits reported for SSc thus far. We define 95% credible sets of less than 5 likely causal variants in 12 loci. Additionally, we identify specific SSc subtype-associated signals. Functional analysis of high-priority variants shows the potential function of SSc signals, with the identification of 43 robust target genes through HiChIP. Our results point towards molecular pathways potentially involved in vasculopathy and fibrosis, two main hallmarks in SSc, and highlight the spectrum of critical cell types for the disease. Abstract

Introduction: RECIPE study was designed to assess the impact of dietary supplementation with fermented milk enriched with plant sterols (FMPS) on the nutritional behavior, lifestyle and the joint therapeutic responsibility of hypercholesterolemic patients. Methods: Observational, prospective, multicenter, international study, was conducted to assess dietary habits and other lifestyle components in hypercholesterolemic patients in whom the general practitioner indicated FMPS supplementation. Nutritional, lifestyle and clinical data were collected at the initial visit and after a 4 months follow-up. The biochemical and anthropometric data were retrieved from medical records obtained at baseline and 4 months. Results: We report the results of Spain. Two hundred and one physicians who provided valid data of 1044 patients participated. The addition of FMPS was associated with improved overall nutritional index, being adequate initially in 28.2% of patients, and increasing up to 75.2% at the end of the study (P< 001). This nutritional change was associated with an improvement in the lipid profile and anthropometric data. Patients more adherent to therapy generally achieved a better result in all parameters compared to non-compliant ones. Conclusions: The addition of FMPS to a diet designed to reduce the LDL enhances the patient's attitude regarding changes in lifestyle, leading to better overall control of dyslipidemia and anthropometric improvement. © 2012 Elsevier España, S.L. and SEA. more...

Abstract

In the last years, significant research in the context of bond of near-surface mounted (NSM) fibre reinforced polymer (FRP) systems in concrete has been conducted at the Department of Civil Engineering of the University of Minho. This paper presents a brief summary of the major results obtained in that research, namely in terms of durability, quality control and design topics. Accelerated ageing tests on NSM FRP bond specimens were conducted to simulate different environmental conditions. A new method was developed and applied to investigate the evolution of the adhesive stiffness and the bond behaviour of NSM systems for different curing conditions used for quality control of FRP installations. Abstract

The constant changes in the world market demand flexible and fluid organizational structures, such as rotating and engaging human effort, to provide high performance. Therefore, organizations make use of distributed multicultural teams, meetings, and online lead projects. However, these social categorization processes can become a disadvantage if trigger potential conflicts during task performance. Distributed teams can also be difficult to manage, and their members can face extra adversities in communication. Architecture, Engineering and Construction Industry (AEC), is a context where the use of distributed teams is growing significantly, particularly through the enabling features of Building Information Modelling (BIM) methodologies. This paper was aimed at the diagnosis of communication behaviour in distributed teams in the context of a PBL methodology that requested students to work in distributed teams on two distinct locations. For that, the authors managed a workshop on Lean Project Management and Collaborative Tools in the European Master in Building Information Modelling (BIM A+) using a Lego for Scrum activity, adapted to a team of students distributed in Portugal and Slovenia. After that, nine distributed teams of students had to design exposition pavilions in BIM platform and using collaborative tools. Abstract

Early age cracking of structures often leads to aesthetic problems and service life reduction. Among the parameters involved in the stress build-up that causes this cracking process, the stiffness evolution is of major importance for models and numerical computations. This paper reports the use of six different techniques aimed for stiffness evolution assessment, applied on the same concrete mix, in a round robin experimental test within three laboratories. The observations are compared after having expressed the results at the same maturity. Some of the reported techniques provide original means for Young's modulus monitoring of concrete at early age both for industrial and research applications. Two sets of results emerge. Ultrasonic measurements provide values of Young's modulus much higher than the values provided by the static or quasi static tests at the time of the concrete setting. Abstract

Useful to define the stiffness of geomaterials at very small strains, the assessment of seismic wave velocities in the laboratory is currently a worldwide common practice. The combined use of piezoelectric transducers such as bender elements and accelerometers, already proved to be a simple but powerful laboratory tool for seismic wave measurements, helping decrease some subjectivity inherent to BE when used individually. Additionally, the simultaneous use of time and frequency domain signal analysis techniques is considered a useful alternative to reduce error in seismic wave measurements. Abstract

A normalização é um dos elementos mais importantes para a implementação dos processos BIM. De facto, trata-se de fator primordial para colaboração eficaz entre os intervenientes da indústria AECO. Um dos importantes vértices a ter em conta na normalização diz respeito à definição da informação alfanumérica relevante, bem como a sua respetiva nomenclatura. Este tipo de definições corresponde a estruturas normalizadas de dados para produtos de construção, designados na língua inglesa por: 'Product Data Template' (PDT). Apesar de já existirem algumas iniciativas a nível internacional para definição de PDTs, trata-se de um processo ainda em fase de maturação a nível mundial, mas que reconhecidamente carece de definições específicas ao contexto de cada país (conforme determinado na ISO 23386:2020, por exemplo). No contexto da CT197 (comissão de normalização nacional BIM, sob alçada do Instituto Português da Qualidade) e de projetos de investigação em curso (p.ex. SECClasS, REV@Construction, e GrowingCircle), a equipa do presente artigo está a encetar esforços comuns rumo ao estabelecimento de PDT’s a nível nacional. O presente artigo descreve esses esforços, com enfoque particular na perspetiva de mobilização da indústria. 10. Desafios BIM na implementação do plano geral de drenagem de Lisboa

Abstract

Com a finalidade de mitigar as consequências provocadas pelas graves inundações que têm fustigado Lisboa devido a eventos pluviais extremos, a Câmara Municipal de Lisboa promoveu a sua maior obra dos últimos 100 anos, a empreitada de execução dos túneis de drenagem da cidade de Lisboa e intervenções associadas. Sendo uma obra geotécnica complexa e audaciosa, projetada para um período de vida útil mínimo de 100 anos, não poderia deixar de acompanhar as melhores práticas internacionais e, como tal, está a ser desenvolvida com recurso à metodologia BIM. O concurso público foi preparado tendo em conta que o projeto de execução seria desenvolvido pelo adjudicatário, no regime usualmente conhecido de conceção‑construção, e deste modo o projeto está a ser realizado com o apoio de uma equipa experiente em obras geotécnicas e sistemas de informação, constituída pelo empreiteiro (Mota‑Engil e Spie Batignolles) e o projetista (LCW, AQUALOGUS, Spie Batignolles e SUEZ), contando com o apoio da fiscalização (TPF – CONSULTORES DE ENGENHARIA E ARQUITETURA, S.A.) e do Dono de Obra (EPPGDL/CML) e seus consultores (Iscte – Instituto Universitário de Lisboa). Para retirar a máxima potencialidade desta metodologia, o projeto e os modelos têm evoluído significativamente, facto que só é possível devido à enorme colaboração entre todos os intervenientes, coadunando‑se assim com a metodologia adotada. Com uma flexibilidade e evolução próprias de uma metodologia que, em Portugal, ainda se encontra numa fase de implementação e expansão, regista‑se já um avanço significativo em relação às práticas usuais, que culminará no objetivo final de apoiar a manutenção do sistema de drenagem em fase de operação. Durante o processo de preparação e construção esta metodologia permitirá definir e gerir os fluxos de comunicação e informação, assegurar a qualidade, apoiar o planeamento da obra e assegurar que a informação relevante é armazenada no cadastro tridimensional da obra. Abstract

O projeto SECClasS – Sustainability Enhanced Construction Classification System – financiado pelo EEA Grants – pretende facilitar a Economia Circular na Construção introduzindo um Sistema de Classificação de Informações sobre Construção otimizado para a Sustentabilidade. Este sistema será orientado para a metodologia BIM e servirá não só a componente de sustentabilidade, mas também os restantes usos BIM, como a gestão do processo BIM, extração de quantidades, compatibilização de especialidades ou planeamento de obra, e todas as fases do ciclo de vida. É objetivo do projeto que o sistema de classificação seja usado pelos profissionais do projeto, construção e responsáveis pela gestão e manutenção de edifícios e outras infraestruturas. Um sistema de classificação permite unificar a terminologia a todas as escalas, facilitando a comunicação, seleção de materiais e componentes, bem como uma avaliação precisa dos impactos dos edifícios ao longo do seu ciclo de vida, com o objetivo de melhorar o desempenho dos edifícios e redução dos resíduos através de ferramentas digitais de gestão e seleção informada dos elementos construtivos. Abstract

Platinum-based drugs are popular in clinics as chemotherapeutic agents to treat solid tumors. However, severe side effects such as nephro- and neurotoxicity impose strict dosage limitations that can lead to the development of drug resistance and tumor relapse. To overcome these issues Pt(IV) prodrugs and platinum delivery systems might represent the next generation of platinum-based drugs. In this study four novel Pt(II) complexes (namely, PEG-Glu-Pt-EDA, PEG-Glu-Pt-DACH, PEG-Mal-Pt-EDA and PEG-Mal-Pt-DACH) were synthesized and a general strategy to covalently bind them to iron oxide nanoparticles was developed. The intracellular uptake and cell distribution studies of Pt-tethered magnetic nanoparticles on breast and ovarian cancer cell line models indicate that binding of the Pt complexes to the nanoparticles facilitates, for all the complexes, cellular internalization. Moreover, the magnetic nanoparticles (MNPs), as shown in a magnetofection experiment, enhance the uptake of MNP-Pt conjugates if a magnet is placed beneath the culture dish of tumor cells. As shown by a Pt release experiment, intranuclear platinum quantification and TEM analysis on cell sections, the presence of a pH-sensitive dicarboxylic group coordinating the Pt complex, triggers platinum dissociation from the NP surface. In addition, the triazole moiety facilitates endosomal swelling and the leakage of platinum from the endosomes with intranuclear localization of platinum release by the NPs. Abstract

A cyclic hexapeptide with three pyridyl moieties connected to its backbone forms a hydrogen-bonded dimer, which tightly encapsulates a single xenon atom, like a pearl in its shell. The dimer imprints its shape and symmetry to the captured xenon atom, as demonstrated by 129 Xe NMR spectroscopy, single-crystal X-ray diffraction, and computational studies. Abstract

As instalações das instituições de Ensino Superior, que no caso do ISCTE-IUL estão concentradas num campus no centro de Lisboa, são usadas diariamente por vários milhares de utentes, acolhem inúmeros eventos e o exercício próprio da instituição e requerem investimentos constantes de manutenção, renovação e adaptação. Ao contrário de outros ativos estratégicos a sua gestão é frequentemente suportada por instrumentos informais: folhas de cálculo, documentação em papel e registos pessoais, tornando-se difícil controlar e planear operações e custos globais de manutenção por equipamento ou sistema. Por outro lado, os desenhos técnicos das instalações estão dispersos e não são fiéis à realidade construída, o que também dificulta as operações de manutenção, modernização e estudo de cenários de melhoria. Perante este cenário, o ISCTE-IUL montou uma equipa multidisciplinar, tendo-se definido uma metodologia para o desenvolvimento de uma solução híbrida de gestão de edifícios e apoio ao utilizador. A primeira etapa consiste na uniformização e verificação no terreno dos desenhos dos projetos incluindo equipamentos fixos e móveis e redes, numa Base Digital de Desenhos em 2D CAD. A segunda etapa consiste na migração desta informação para um modelo BIM, mais completo e fácil de atualizar, principalmente no que diz respeito à extração de métricas.
