3. TGF-β regulates invasive behavior of human pancreatic cancer cells by controlling Smad expression

Abstract

Introduction: To investigate the role of Smads in tumor cell activation, we examined changes in Smad expression as well as changes in proliferative and invasive behaviors in transforming growth factor-β (TGF-β) – stimulated pancreatic cancer cells. Material and methods: Expression of TGF-β receptor type I (TβR-I) and type II (TβR-II) was determined using RT-PCR and Western blot analysis in the human pancreatic cancer cell lines BxPC-3, Capan-2, and PANC-1. TGF-β-mediated changes in Smad mRNA expression were examined using quantitative real-time RT-PCR. Proliferation of pancreatic cancer cells was monitored using an MTT assay and cell counting. Invasive behavior was examined using a Matrigel double-chamber assay. Results: TβR-I and TβR-II were expressed in all three cell lines studied here at the mRNA and protein level. Smad2/3 mRNA expression was decreased after TGF-β stimulation in all three cell lines, while Smad4 mRNA expression remained unchanged. Smad6/7 mRNA expression was also attenuated in all three cell lines. TGF-β enhanced the invasive capacity of all three cell lines, but had no effect on the proliferative behavior. Anti-TβR-II antibody inhibited this TGF-β-enhanced invasive potential in pancreatic cancer cells. Conclusions: The Smad pathway, particularly down-regulation of Smad2/3 and Smad6/7, may be responsible for TGF-β-induced invasion of human pancreatic cancer cells.

Abstract

The Theta+ was searched for via the K+p-->pi+X reaction using the 1.2 GeV/c K+ beam at the K6 beam line of the KEK-PS 12 GeV Proton Synchrotron. In the missing mass spectrum of the K+p-->pi+X reaction, no clear peak structure was observed. Therefore a 90% C.L. upper limit of 3.5 µb/sr was derived for the differential cross section averaged over 2° to 22° in the laboratory frame of the K+p-->pi+Theta+ reaction. This upper limit is much smaller than the theoretical calculation for the t-channel process where a K0* is exchanged. From the present result, either the t-channel process is excluded or the coupling constant of gK*NTheta is quite small.

Abstract

Abstract

PHASE; A hyperon–nucleon scattering experiment, KEK-PS E289, was carried out at the K2 beam line in KEK. We adopted a scintillating fiber active target as a hydrogen target for hyperon production and subsequent hyperon–nucleon scattering. We collected more data than in a previous experiment, KEK-PS E251. In this paper, the differential cross sections for Σ+p elastic scattering in the incident momentum range from 350 to 750 MeV/c are presented and compared with theoretical predictions from the Nijmegen group (Nijmegen Soft Core 97) and the Kyoto–Niigata group (RGM-FSS and fss2).

Abstract

Invariant mass spectra of e+e- pairs have been measured in 12 GeV p+A reactions to detect possible in-medium modification of vector mesons. Copper and carbon targets are used to study the nuclear-size dependence of e+e- invariant mass distributions. A significant excess on the low-mass side of the phi meson peak is observed in the low beta gamma (=beta/sqrt(1-beta^2)) region of phi mesons (beta gamma < 1.25) with copper targets. However, in the high beta gamma region (beta gamma > 1.25), spectral shapes of phi mesons are well described by the Breit-Wigner shape when experimental effects are considered. Thus, in addition to our earlier publication on rho / omega modification, this study has experimentally verified vector meson mass modification at normal nuclear density., 4 pages, 4 figures

Abstract

We have recently demonstrated that nuclear factor kappaB (NF-kappaB) mediates the tumour necrosis factor alpha (TNF-alpha)-dependent expression of the gene encoding interleukin 6 (IL-6) in rat thyroid FRTL-5 cells cultured in the presence of thyrotropin (TSH). In the present study we investigated how TSH is involved in the activation of NF-kappaB by TNF-alpha in the cells. Electrophoretic mobility-shift assay revealed that, in the absence of TSH, TNF-alpha activated a single protein-DNA complex containing the p50 subunit but not other NF-kappaB subunits such as p65. In contrast, two distinct protein-DNA complexes were activated in the presence of TSH: the faster-migrating complex contained only p50 subunit; the slower-migrating complex consisted of p65-p50 heterodimer. This TSH effect was mimicked by forskolin and thyroid-stimulating antibodies obtained from patients with Graves's disease, suggesting that an increase in intracellular cAMP is responsible for the induction of different NF-kappaBs by TNF-alpha. A transient transfection study with a luciferase reporter gene driven by multimerized NF-kappaB sites demonstrated that TNF-alpha increased the luciferase activities only in the presence of TSH, and that this increase was inhibited by the co-transfection of mutant p65, which prevented the function of wild-type p65 in a dominant-negative manner. Accordingly, TNF-alpha activated the expression of the IL-6 gene in the presence of TSH but not in its absence. Although the expression of the p105 gene, another known target for NF-kappaB, was increased by TNF-alpha in the absence of TSH, the presence of TSH further increased the mRNA level. Taken together, these observations indicate that the presence of TSH is crucial for the NF-kappaB-mediated actions of TNF-alpha on thyroid follicular cells.

Abstract

We report on the present progress in development of the dark matter axion search experiment with Rydberg-atom-cavity detectors in Kyoto, CARRACK I and CARRACK II. The axion search has been performed with CARRACK I in the 8 % mass range around $ 10 \mu {\rm eV} $, and CARRACK II is now ready for the search in the wide range $ 2 \mu {\rm eV} - 50 \mu {\rm eV} $. We have also developed quantum theoretical calculations on the axion-photon-atom system in the resonant cavity in order to estimate precisely the detection sensitivity for the axion signal. Abstract

A coupled microwave-cavity system of cylindrical TM$_{010}$ single-mode has been developed to search for dark matter axions around 10 $\mu {\rm eV}$(2.4 GHz) with the Rydberg-atom cavity detector at 10 mK range temperature. One component of the coupled cavity (conversion cavity) made of oxygen-free high-conductivity copper is used to convert an axion into a single photon with the Primakoff process in the strong magnetic field, while the other component (detection cavity) made of Nb is utilized to detect the converted photons with Rydberg atoms passed through it without magnetic field. Top of the detection cavity is attached to the bottom flange of the mixing chamber of a dilution refrigerator, thus the whole cavity is cooled down to 10 mK range to reduce the background thermal blackbody-photons in the cavity. The cavity resonant frequency is tunable over $\sim$ 15% by moving dielectric rods inserted independently into each part of the cavities along the cylindrical axis. In order to reduce the heat load from the higher temperature side to the most cooled dilution refrigerator part, the tuning rod at the conversion cavity is especially driven via the Kevlar strings with a stepping motor outside the cryostat at room temperature. The lowest temperature achieved up to now is 12 $\sim$ 15 mK for the long period operation and the loaded Q value at low temperature is 3.5 $\sim$ 4.5 $\times$ $10^4$ for the whole range of frequency tuning. Abstract

Coherent time evolution of highly excited Rydberg states in Rb (98 < n < 150) under pulsed electric field in high slew-rate regime was investigated with the field ionization detection. The electric field necessary to ionize the Rydberg states was found to take discrete values successively depending on the slew rate. Specifically the slew-rate dependence of the ionization field varies with the quantum defect value of the states, i.e. with the energy position of the states relative to the adjacent manifold. This discrete transitional behavior of the ionization field observed for the first time is considered to be a manifestation of the strong coherence effect in the time evolution of the Rydberg states in pulsed electric field and opens a new effective way to stringently select a low-l state from the nearby states by field ionization., 4 pages, 4 figures

Abstract

The considerable polarization of hyperons produced at high x(F) has been known for a long time and has been interpreted with various theoretical models in terms of the constituents' spin. Recently, the analyzing power in inclusive Lambda(0) hyperon production has also been measured using the 200 GeV/c Fermilab polarized proton beam. The covered kinematic range is 0.2 less than or equal to x(F) less than or equal to 1.0 and 0.1 less than or equal to p(T) less than or equal to 1.5 GeV/c. The data indicate a negative asymmetry at large x(F) and moderate p(T). These results can further test the current ideas on the underlying mechanisms for hyperon polarization.

Abstract

Stopped-Ξ-12C reactions have been investigated with respect to H-dibaryon and hypernucleus formation with a scintillating fiber detector. Out of 5.5×103 tagged Ξ- particles via the (K-,K+) reaction we obtained 72 stopping Ξ- events with 6.7+1.1-1.4 background events after imposing kinematic constraints. Hypernuclear production has been studied by observing free Λ decays in the stopped-Ξ- reactions. From the observed Λ momentum spectrum, we have found the hypernuclear production rates to be 18±13, 65±6, and 17±12 % for double/twin-A hypernucleus, single-Λ hypernucleus, and no hypernucleus cases, respectively. With the same sample, we set upper limits on R (stopped-Ξ-12C→HX /stopped-Ξ-12C→ all) for H decaying into Λn, Σ-p, and Λpπ-, for H masses just below ΛΛ mass threshold and for H lifetime range 10-11≤τH≤10-8 s. The upper limits on R are ∼0.1-0.2, assuming all of the H decay into each decay mode. Our results give a tight constraint on the existence of the bound H, in comparison with the theoretical estimates of Aerts and Dover, albeit with different target nuclei.
