The purpose of this study was to establish the correlates of sports confidence among high school athletes. The study examined Vealey’s (1986) nine sources of sports confidence in relation to contextual factors of gender, age, playing experience and type of sport. A descriptive survey design was used, and data were collected using Vealey’s Sources of sports confidence questionnaire (SSCQ) from high school athletes (n=175). Results showed that coaches’ leadership was the highest source of sports confidence, followed by mastery and physical and mental preparation, while situational favourableness appeared to be the weakest source of confidence amongst the athletes. There were significant differences in physical self-presentation (t=-2.14, df = 173, p =0.034) in favour of male athletes. Athletes participating in individual sports had superior scores in mastery, while coaches’ leadership was the most important source of sports confidence for athletes in team sports. There were significant differences in coaches’ leadership (t=-3.118, df = 175, p = 0.002) and social support (t= -2.236, df = 175, p = 0.027) in favour of athletes in a team sport. Athletes who did not engage in their current sport while in a primary school rated coaches’ leadership highly, while mastery of the skill was scored highly by athletes who did participate in the sport while in primary school. It is concluded that coaches’ leadership skills, mastery of skill execution and physical or mental preparedness were the major sources of sports confidence. High schools should utilize coaches who can be emulated by the athletes.

Active people in sports or regular exercises make up a significant portion of individuals using nutritional supplements (NS). Therefore the purpose of this study was to investigate the prevalence of NS (types, sources of information), motives for going to the gym, reasons for consumption of nutritional supplements and attitudes towards nutritional supplements among gymnasium users in Eldoret Town, Kenya. Data was collected using a questionnaire from 210 participants who were sampled through systematic convenience sampling from 11 gyms in Eldoret Town, Kenya. The study found that majority (55.3%) of the respondents used nutritional supplements and most of them used the supplements believing that they enhance their performance at the gym and therefore perceived usage of supplements as acceptable. The major reasons for going to gym were to enhance health, body building and to stay fit. Most (91%) of the gym users got the information about NS from the internet and the most consumed NS were protein supplements (72%), followed by sport drinks (69%) and fish oils (60%). It is concluded that gym goers use nutritional supplements and have positive attitudes towards their use. Therefore, this study recommends that users should be sensitized on the value, precautionary measures and side effects of using nutritional supplements.

Inadequate access to public bariatric surgical services has favoured the growth of bariatric tourism. This study analysed data extracted from bariatric surgical centres that care for patients travelling from abroad. The research highlights apparent deficits in accreditation, communication, perioperative care, and travel health advice. An international registry of accredited bariatric tourism providers and patient education may be indicated.

Nationally-representative surveys provide an opportunity to assess trends in recent HIV infection based on assays for recent HIV infection.We assessed HIV incidence in Kenya in 2018 and trends in recent HIV infection among adolescents and adults in Kenya using nationally representative household surveys conducted in 2007, 2012 and 2018. To assess trends, we defined a recent HIV infection testing algorithm (RITA) that classified as recently infected (12 months) those HIV-positive participants that were recent on the HIV-1 limiting antigen (LAg)-avidity assay without evidence of antiretroviral use. We assessed factors associated with recent and long-term (≥12 months) HIV infection versus no infection using a multinomial logit model while accounting for complex survey design.Of 1,523 HIV-positive participants in 2018, 11 were classified as recent. Annual HIV incidence was 0.14% in 2018 (95% confidence interval [CI] 0.057-0.23), representing 35,900 (95% CI 16,300-55,600) new infections per year in Kenya among persons aged 15-64 years. The percentage of HIV infections that were determined to be recent was similar in 2007 and 2012 but fell significantly from 2012 to 2018 (adjusted odds ratio [aOR]=0.31, p0.001). Compared to no HIV infection, being aged 25-34 versus 35-64 years (aOR=4.2, 95% CI 1.4-13), having more lifetime sex partners (aOR=5.2, 95% CI 1.6-17 for 2-3 partners and aOR=8.6, 95% CI 2.8-26 for ≥4 partners versus 0-1 partners), and never having tested for HIV (aOR=4.1, 95% CI 1.5-11) were independently associated with recent HIV infection.Though HIV remains a public health priority in Kenya, HIV incidence estimates and trends in recent HIV infection support a significant decrease in new HIV infections from 2012 to 2018, a period of rapid expansion in HIV diagnosis, prevention, and treatment.

Human skin disease, the most infectious dermatological ailment globally, is initially diagnosed by sight. Some clinical screening and dermoscopic analysis of skin biopsies and scrapings for accurate classification are medically compulsory. Classification of skin diseases using medical images is more challenging because of the complex formation and variant colors of the disease and data security concerns. Both the Convolution Neural Network (CNN) for classification and a federated learning approach for data privacy preservation show staggering performance in the realm of medical imaging fields. In this paper, a custom image dataset was prepared with four classes of skin disease, a CNN model was suggested and compared with several benchmark CNN algorithms, and an experiment was carried out to ensure data privacy using a federated learning approach. An image augmentation strategy was followed to enlarge the dataset and make the model more general. The proposed model achieved a precision of 86\%, 43\%, and 60\%, and a recall of 67\%, 60\%, and 60\% for acne, eczema, and psoriasis. In the federated learning approach, after distributing the dataset among 1000, 1500, 2000, and 2500 clients, the model showed an average accuracy of 81.21\%, 86.57\%, 91.15\%, and 94.15\%. The CNN-based skin disease classification merged with the federated learning approach is a breathtaking concept to classify human skin diseases while ensuring data security.

PURPOSE Patients with progressive or recurrent meningiomas have limited systemic therapy options. Focal adhesion kinase (FAK) inhibition has a synthetic lethal relationship with NF2 loss. Given the predominance of NF2 mutations in meningiomas, we evaluated the efficacy of GSK2256098, a FAK inhibitor, as part of the first genomically driven phase II study in recurrent or progressive grade 1-3 meningiomas. PATIENTS AND METHODS Eligible patients whose tumors screened positively for NF2 mutations were treated with GSK2256098, 750 mg orally twice daily, until progressive disease. Efficacy was evaluated using two coprimary end points: progression-free survival at 6 months (PFS6) and response rate by Macdonald criteria, where PFS6 was evaluated separately within grade-based subgroups: grade 1 versus 2/3 meningiomas. Per study design, the FAK inhibitor would be considered promising in this patient population if either end point met the corresponding decision criteria for efficacy. RESULTS Of 322 patients screened for all mutation cohorts of the study, 36 eligible and evaluable patients with NF2 mutations were enrolled and treated: 12 grade 1 and 24 grade 2/3 patients. Across all grades, one patient had a partial response and 24 had stable disease as their best response to treatment. In grade 1 patients, the observed PFS6 rate was 83% (10/12 patients; 95% CI, 52 to 98). In grade 2/3 patients, the observed PFS6 rate was 33% (8/24 patients; 95% CI, 16 to 55). The study met the PFS6 efficacy end point both for the grade 1 and the grade 2/3 cohorts. Treatment was well tolerated; seven patients had a maximum grade 3 adverse event that was at least possibly related to treatment with no grade 4 or 5 events. CONCLUSION GSK2256098 was well tolerated and resulted in an improved PFS6 rate in patients with recurrent or progressive NF2-mutated meningiomas, compared with historical controls. The criteria for promising activity were met, and FAK inhibition warrants further evaluation for this patient population.

The Perseverance rover landed in Jezero crater, Mars, in February 2021. We used the Scanning Habitable Environments with Raman and Luminescence for Organics and Chemicals (SHERLOC) instrument to perform deep-ultraviolet Raman and fluorescence spectroscopy of three rocks within the crater. We identify evidence for two distinct ancient aqueous environments at different times. Reactions with liquid water formed carbonates in an olivine-rich igneous rock. A sulfate-perchlorate mixture is present in the rocks, which probably formed by later modifications of the rocks by brine. Fluorescence signatures consistent with aromatic organic compounds occur throughout these rocks and are preserved in minerals related to both aqueous environments.

Approximately half of the patients with diabetes develop diabetic bladder dysfunction (DBD). The initiation and progression of DBD is largely attributed to inflammation due to dysregulated glucose and the production of toxic metabolites that activate the NOD-, LRR-, and pyrin domain-containing protein 3 (NLRP3) inflammasome. NLRP3 activation leads to the production and release of proinflammatory cytokines and causes urothelial pyroptosis, a form of programmed cell necrosis, which we hypothesize compromises urothelial barrier integrity. Here, we investigated how NLRP3-dependent inflammation impacts barrier function during the progression of diabetes using a type 1 diabetic female Akita mouse model that progresses from an early overactive to a late underactive detrusor phenotype at 15 and 30 wk, respectively. Non-finite verbs in Turkic are typically categorised as participles, converbs, and sometimes infinitives, with multiple uses of a form within one category considered to constitute multiple functions. This multifunctionality approach predicts that all non-finite verb forms within each of the categories should have the same range of syntactic functions. We show that this is not the case. Based on analysis of a representative set of Turkic languages (Kazakh, Kyrgyz, Sakha, Tatar, Turkish, and Tuvan), we propose a categorisation based on morphological and syntactic properties of non-finite verbs, resulting in four categories: verbal nouns, verbal adjectives, verbal adverbs, and infinitives. Under this approach, forms that are typically labelled as participles end up categorised as verbal nouns, verbal adjectives, or both, and forms that are typically labelled as converbs end up categorised as verbal adverbs, infinitives, or both. Some forms even span these two divisions. When a non-finite verb form appears to exist in multiple categories, we consider this to be a case of syncretism; this is, there is a member of one category that has the same form as a member of another category. We propose historical trajectories that may have led to the types of situations that are attested, examine the limitations of this approach, and discuss its wider implications.

17 Pág., Ectomycorrhizas are an intrinsic component of tree nutrition and responses to environmental variations. How epigenetic mechanisms might regulate these mutualistic interactions is unknown. By manipulating the level of expression of the chromatin remodeler DECREASE IN DNA METHYLATION 1 (DDM1) and two demethylases DEMETER-LIKE (DML) in Populus tremula × Populus alba lines, we examined how host DNA methylation modulates multiple parameters of the responses to root colonization with the mutualistic fungus Laccaria bicolor. We compared the ectomycorrhizas formed between transgenic and wild-type (WT) trees and analyzed their methylomes and transcriptomes. The poplar lines displaying lower mycorrhiza formation rate corresponded to hypomethylated overexpressing DML or RNAi-ddm1 lines. We found 86 genes and 288 transposable elements (TEs) differentially methylated between WT and hypomethylated lines (common to both OX-dml and RNAi-ddm1) and 120 genes/1441 TEs in the fungal genome suggesting a host-induced remodeling of the fungal methylome. Hypomethylated poplar lines displayed 205 differentially expressed genes (cis and trans effects) in common with 17 being differentially methylated (cis). Our findings suggest a central role of host and fungal DNA methylation in the ability to form ectomycorrhizas including not only poplar genes involved in root initiation, ethylene and jasmonate-mediated pathways, and immune response but also terpenoid metabolism., MDS and JV received PhD grants from the MRES and Conseil Régional Région Centre Val de Loire, respectively. Doping is a worldwide problem that harms athletes' health and undermines the spirit of sport. Studies have shown that male athletes are more prone to doping than female athletes. Athletes with mastery climate have been associated with anti-doping attitudes, while those with performance climate have pro-doping attitudes. However, it is unclear whether motivational climate is equally important to attitude towards doping for males and females. Data were collected from 323 runners in Elgeyo-Marakwet County, Kenya, using cross-sectional survey design. Runners self-reported their motivational climate using Perceived Motivational Climate in Sport Questionnaire and attitudes towards doping using Performance Enhancement Attitude Scale. Correlational analysis indicated significant inverse relationship between mastery climate and doping attitude (rho = -.242; p < .001) and significant positive correlation between performance climate and doping attitude, (rho = .362; p < .001). Hierarchical regression analysis revealed performance and mastery climate were significant predictors of attitudes towards doping (F (3, 319) = 28.24, p = .001), and gender did not moderate the relations between motivational climate and doping attitudes (β = -.028, p = .621). MANOVA results showed male athletes were significantly lower in performance climate scores (p = .045) and non-significantly low in mastery climate scores (p =.075) and doping attitude scores (p = .595) than females. In conclusion, performance climate was associated with doping attitudes in females- but not in males. Therefore, policy frameworks that buttresses the aspects of mastery climate as opposed to performance climate in females is likely to promote anti-doping attitudes.

To evaluate bacterial contamination of conjunctiva and aqueous humor in dogs undergoing phacoemulsification following asepsis with 0.5% povidone iodine and determine the influence of intravenous antibiotics on outcome of contamination.Client-owned dogs were prospectively enrolled and randomly assigned to a control group, receiving 22 mg/kg intravenous cefazolin at induction prior to sampling, or experimental group receiving no antibiotic prior to sampling, masked to the surgeon. Dogs receiving antimicrobials in the pre-operative period were excluded. Asepsis was performed on all operated eyes using 0.5% iodine with minimum 3 min contact time at induction of anesthesia and repeated before surgery. A conjunctival swab and aqueous humor sample were collected prior to incision and following incision closure, respectively. Samples were submitted for aerobic and anaerobic bacterial culture and susceptibility.Seventy-one eyes of 42 dogs were included. Median age was 9 years. Thirty-nine and 32/71 eyes received intravenous cefazolin and no antibiotic, respectively. Median procedure time was 40 min per eye. Conjunctival cultures were positive in 6 eyes (8.5%): Serratia marcescens (5 eyes) and Cutibacterium acnes (1 eye). Aqueous humor cultures were positive in 5 eyes (7.0%): S. marcescens (2 eyes), Pseudomonas aeruginosa (2 eyes), Staphylococcus pseudointermedius (1 eye). Prevalence of positive culture did not differ between groups (p = .74), order of eyes for bilateral procedures (p = .74) and diabetic status (p = 1).Bacterial contamination of the conjunctiva and aqueous humor was present in 8.5% and 7.0% of dogs undergoing phacoemulsification after asepsis. Lack of IV cefazolin was not significantly associated with positive culture.

Immunotherapy is used to treat almost all patients with advanced non-small cell lung cancer (NSCLC); however, identifying robust predictive biomarkers remains challenging. Here we show the predictive capacity of integrating medical imaging, histopathologic and genomic features to predict immunotherapy response using a cohort of 247 patients with advanced NSCLC with multimodal baseline data obtained during diagnostic clinical workup, including computed tomography scan images, digitized programmed death ligand-1 immunohistochemistry slides and known outcomes to immunotherapy. Using domain expert annotations, we developed a computational workflow to extract patient-level features and used a machine-learning approach to integrate multimodal features into a risk prediction model. Our multimodal model (area under the curve (AUC) = 0.80, 95% confidence interval (CI) 0.74–0.86) outperformed unimodal measures, including tumor mutational burden (AUC = 0.61, 95% CI 0.52–0.70) and programmed death ligand-1 immunohistochemistry score (AUC = 0.73, 95% CI 0.65–0.81). Our study therefore provides a quantitative rationale for using multimodal features to improve prediction of immunotherapy response in patients with NSCLC using expert-guided machine learning.

To describe the effects of gabapentin on ocular and behavioral parameters following oral administration in healthy cats.Masked, placebo-controlled, randomized crossover-design study. Ten young, healthy cats were scheduled for two veterinary visits 7 days apart and randomly assigned to receive a compounded capsule containing 100 mg of gabapentin or placebo (100 mg lactose powder) at the first visit and the opposite treatment at the second visit. Respiratory rate, heart rate, stress score, sedation score, compliance score, horizontal pupil diameter, intraocular pressure, and Schirmer tear test-1 were measured prior to and 1.5, 3, and 6 h following capsule administration. Stress score, sedation score, and compliance score were assigned based on established behavioral scales. Results of the two treatments were statistically compared with a p-value0.05 considered significant.Respiratory rate was significantly reduced at 1.5 (p = 0.049) and 3 (p = 0.03) hours following gabapentin administration. Stress score was significantly reduced at 1.5 (p = 0.01) hours following gabapentin administration. Sedation score was significantly increased at 1.5 (p = 0.015) and 3 (p = 0.03) hours following gabapentin administration. Gabapentin had no significant effect on heart rate, compliance score, or ocular values measured in this study.Gabapentin reduces stress and increases sedation at 1.5 h after treatment, with no significant effect on horizontal pupil diameter, intraocular pressure or Schirmer tear test-1 results.

Based on the cold spray technique, the solvent-free and solid-state deposition of glassy polymers is envisioned. Adiabatic inelastic deformation mechanisms in the cold spray technique are studied through high-velocity collisions (1000 m/s) of polystyrene microparticles against stationary target substrates of polystyrene and silicon. During extreme collisions, a brittle-to-ductile transition occurs, leading to either fracture- or shear-dominant inelastic deformation of the colliding microparticles. Due to the nonlinear interplay between the adiabatic shearing and the thermal softening of polystyrene, the plastic shear flow becomes the dominant deformation channel over brittle fragmentation when increasing the rigidity of the target substrate. High molecular weights (20 kDa) are essential to hinder the evolution of brittle fracture and promote shear-induced heating beyond the glass transition temperature of polystyrene. However, an excessively high molecular weight (∼100 kDa) reduces the adhesion of the microparticles to the substrate due to insufficient wetting of the softened polystyrene. Due to the two competing viscoelastic effects, proper selection of molecular weight becomes critical for the cold spray technique of glassy polymers.

Key messageWe investigate the genetic basis of panicle architecture in switchgrass in two mapping populations across a latitudinal gradient, and find many stable, repeatable genetic effects and limited genetic interactions with the environment.AbstractGrass species exhibit large diversity in panicle architecture influenced by genes, the environment, and their interaction. The genetic study of panicle architecture in perennial grasses is limited. In this study, we evaluate the genetic basis of panicle architecture including panicle length, primary branching number, and secondary branching number in an outcrossed switchgrass QTL population grown across ten field sites in the central USA through multi-environment mixed QTL analysis. We also evaluate genetic effects in a diversity panel of switchgrass grown at three of the ten field sites using genome-wide association (GWAS) and multivariate adaptive shrinkage. Furthermore, we search for candidate genes underlying panicle traits in both of these independent mapping populations. Overall, 18 QTL were detected in the QTL mapping population for the three panicle traits, and 146 unlinked genomic regions in the diversity panel affected one or more panicle trait. Twelve of the QTL exhibited consistent effects (i.e., no QTL by environment interactions or no QTL × E), and most (four of six) of the effects with QTL × E exhibited site-specific effects. Most (59.3%) significant partially linked diversity panel SNPs had significant effects in all panicle traits and all field sites and showed pervasive pleiotropy and limited environment interactions. Panicle QTL co-localized with significant SNPs found using GWAS, providing additional power to distinguish between true and false associations in the diversity panel.

Complications from placental malaria cause poor pregnancy outcomes, including low birthweight, preterm delivery, and stillbirths. Many of these complications are driven by maternal innate proinflammatory responses to the sequestration of Plasmodium falciparum in the placenta. However, recent studies show that, in reaction to maternal innate immune responses that are detrimental to the fetus, the fetus mounts innate immune counter-responses that ameliorate pregnancy outcomes. Such fetal-maternal conflict in placental malaria has potential for pharmacologic modulation for better pregnancy outcomes. Here, we discuss placental malaria pathogenesis, its complications, and the role of innate immunity and fetal-maternal innate immune conflict in placental malaria. Finally, we discuss pharmacologic immunomodulatory strategies and agents with the potential to improve placental malaria outcomes.

Polymorphisms in immunity genes can have large effects on susceptibility to infection. To understand the origins of this variation, we have investigated the genetic basis of resistance to the parasitoid wasp Leptopilina boulardi in Drosophila melanogaster. We discovered that increased expression of the gene Lectin-24A after infection by parasitic wasps was associated with a faster cellular immune response and greatly increased rates of killing the parasite. Lectin-24A encodes a protein that is strongly upregulated in the fat body after infection and localizes to the surface of the parasite egg. In certain susceptible lines a deletion upstream of the Lectin-24A has largely abolished expression. Other mutations predicted to abolish the function of this gene have arisen recurrently in this gene, with multiple loss-of-expression alleles and premature stop codons segregating in natural populations. The frequency of these alleles varies greatly geographically, and in some southern African populations natural selection has driven them near to fixation. We conclude that natural selection has favored the repeated loss of an important component of the immune system, suggesting that in some populations a pleiotropic cost to Lectin-24A expression outweighs the benefits of resistance.

Osteosarcoma is the most common type of bone cancer that tends to occur in teenagers and young adults. Due to crowded context, inter-class similarity, inter-class variation, and noise in H&E-stained (hematoxylin and eosin stain) histology tissue, pathologists frequently face difficulty in osteosarcoma tumor classification. In this paper, we introduced a hybrid framework for improving the efficiency of three types of osteosarcoma tumor (nontumor, necrosis, and viable tumor) classification by merging different types of CNN-based architectures with a multilayer perceptron (MLP) algorithm on the WSI (whole slide images) dataset. We performed various kinds of preprocessing on the WSI images. Then, five pre-trained CNN models were trained with multiple parameter settings to extract insightful features via transfer learning, where convolution combined with pooling was utilized as a feature extractor. For feature selection, a decision tree-based RFE was designed to recursively eliminate less significant features to improve the model generalization performance for accurate prediction. Here, a decision tree was used as an estimator to select the different features. Finally, a modified MLP classifier was employed to classify binary and multiclass types of osteosarcoma under the five-fold CV to assess the robustness of our proposed hybrid model. Moreover, the feature selection criteria were analyzed to select the optimal one based on their execution time and accuracy. The proposed model achieved an accuracy of 95.2% for multiclass classification and 99.4% for binary classification. Experimental findings indicate that our proposed model significantly outperforms existing methods; therefore, this model could be applicable to support doctors in osteosarcoma diagnosis in clinics. In addition, our proposed model is integrated into a web application using the FastAPI web framework to provide a real-time prediction.

Meteoritical Bulletin 111 contains the 3094 meteorites approved by the Nomenclature Committee of the Meteoritical Society in 2022. It includes 11 falls (Antonin, Botohilitano, Cranfield, Golden, Great Salt Lake, Longde, Msied, Ponggo, Qiquanhu, Tiglit, Traspena), with 2533 ordinary chondrites, 165 HED, 123 carbonaceous chondrites (including 4 ungrouped), 82 lunar meteorites, 28 Rumuruti chondrites, 27 iron meteorites, 23 ureilites, 22 mesosiderites, 22 Martian meteorites, 21 primitive achondrites (one ungrouped), 17 ungrouped achondrites, 13 pallasites, 7 enstatite achondrites, 6 enstatite chondrites, and 5 angrites. Of the meteorites classified in 2022, 1787 were from Antarctica, 1078 from Africa, 180 from South America, 34 from Asia, 6 from North America, 4 from Europe, and 1 from Oceania.

Human tooth functionality is the most important for the human body to become fit and healthy. Due to the disease attacks in human teeth, parts may lead to different fatal diseases. A spectroscopy-based photonic crystal fiber (PCF) sensor was simulated and numerically analyzed for the detection of dental disorders in the human body. In this sensor structure, SF11 is used as the base material, gold (Au) is used as the plasmonic material, and TiO2 is used within the gold and sensing analyte layer, and the sensing medium for the analysis of the teeth parts is the aqueous solution. The maximum optical parameter values for the human tooth parts enamel, dentine, and cementum in terms of wavelength sensitivity and confinement loss were obtained as 28,948.69 nm/RIU and 0.00015 dB/m for enamel, 33,684.99 nm/RIU and 0.00028 dB/m, and 38,396.56 nm/RIU and 0.00087 dB/m, respectively. The sensor is more precisely defined by these high responses. The PCF-based sensor for tooth disorder detection is a relatively recent development. Due to its design flexibility, robustness, and wide bandwidth, its application area has been spreading out. The offered sensor can be used in the biological sensing area to identify problems with human teeth.

To develop standard optical biosensors, the simulation procedure takes a lot of time. For reducing that enormous amount of time and effort, machine learning might be a better solution. Effective indices, core power, total power, and effective area are the most crucial parameters for evaluating optical sensors. In this study, several machine learning (ML) approaches have been applied to predict those parameters while considering the core radius, cladding radius, pitch, analyte, and wavelength as the input vectors. We have utilized least squares (LS), LASSO, Elastic-Net (ENet), and Bayesian ridge regression (BRR) to make a comparative discussion using a balanced dataset obtained with the COMSOL Multiphysics simulation tool. Furthermore, a more extensive analysis of sensitivity, power fraction, and confinement loss is also demonstrated using the predicted and simulated data. The suggested models were also examined in terms of R2-score, mean average error (MAE), and mean squared error (MSE), with all of the models having an R2-score of more than 0.99, and it was also shown that optical biosensors had a design error rate of less than 3%. This research might pave the way for machine learning-based optimization approaches to be used to improve optical biosensors.

Wolbachia is a common endosymbiont that can protect insects against viral pathogens. However, whether the antiviral effects of Wolbachia have a significant effect on fitness remains unclear. We have investigated the interaction between Drosophila melanogaster, Wolbachia and two viruses that we recently isolated from wild flies, La Jolla virus (LJV; Iflaviridae) and Newfield virus (NFV; Permutotetraviridae). Flies infected with these viruses have increased mortality rates, and NFV partially sterilizes females. These effects on fitness were reduced in Wolbachia-infected flies, and this was associated with reduced viral titres. However, Wolbachia alone also reduces survival, and under our experimental conditions these costs of the symbiont can outweigh the benefits of antiviral protection. In contrast, protection against the sterilizing effect of NFV leads to a net benefit of Wolbachia infection after exposure to the virus. These results support the hypothesis that Wolbachia is an important defense against the natural pathogens of D. melanogaster. Furthermore, by reducing the cost of Wolbachia infection, the antiviral effects of Wolbachia may aid its invasion into populations and help explain why it is so common in nature.

Background: In vitro, the molecular docking method has been suggested for estimating the biological affinity of the pharmacophores with physiologically active compounds. It is the latter stage in molecular docking, and the docking scores are examined using the AutoDock 4.2 tool program. The chosen compounds can be evaluated for in vitro activity based on the binding scores, and the IC50 values can be computed. Objective: The purpose of this work was to create methyl isatin compounds as potential antidepressants, compute physicochemical characteristics, and carry out docking analysis. Methods: The protein data bank of the RCSB (Research Collaboratory for Structural Bioinformatics) was used to download the PDB structures of monoamine oxidase (PDB ID: 2BXR) and indoleamine 2,3-dioxygenase (PDB ID: 6E35). Based on the literature, methyl isatin derivatives were chosen as the lead chemicals. By determining their IC50 values, the chosen compounds were tested for in vitro anti-depressant activity. Results: The binding scores for the interactions of SDI 1 and SD 2 with indoleamine 2,3 dioxygenase were found to be -10.55 kcal/mol and -11.08 kcal/mol, respectively, while the scores for their interactions with monoamine oxidase were found to be -8.76 kcal/mol and -9.28 kcal/mol, respectively, using AutoDock 4.2. The relationship between biological affinity and pharmacophore electrical structure was examined using the docking technique. The chosen compounds were tested for their ability to inhibit MAO, and the IC50 values for each were found to be 51.20 and 56, respectively. Conclusion: This investigation has identified many novel and effective MAO-A inhibitors from the family of chemicals known as methyl isatin derivatives. Lead optimization was applied to the SDI 1 and SDI 2 derivatives. The superior bioactivity, pharmacokinetic profile, BBB penetration, pre-ADMET profiles, such as HIA (human intestinal absorption) and MDCK (Madin-Darby canine kidney), plasma protein binding, toxicity assessment, and docking outcomes, have been obtained. According to the study, synthesised isatin 1 and SDI 2 derivatives exhibited a stronger MAO inhibitory activity and effective binding energy, which may help prevent stress-induced depression and other neurodegenerative disorders caused by a monoamine imbalance.

Metallization of polymers and fiber-reinforced polymer composites is gaining attention due to the widespread application of these components in various industries, such as wind energy, aerospace, and automotive industries. Cold spray is a promising new technique to achieve the metallization of polymer and fiber-reinforced polymer composites. This work investigates the deposition mechanisms of polymer-coated metallic particles on polymer-based substrates by finite element analyses. Impact mechanics of PEEK-coated nickel particles impacting PEEK and carbon fiber-reinforced PEEK substrates are modeled. Results show the prominence of mechanical interlocking of metallic particles in the substrate, which occurs due to their entrapment inside the substrate, caused by the high energy impact-induced welding of scraped PEEK coating. The PEEK coating acts as a cushioning component, effectively mitigating the impact energy of the metallic component. The scraped PEEK coating also accumulates on the upper half of the particle, forming a cap welded to the substrate and sealing the metallic particle inside. It is observed that the depth of the carbon fiber mat in the substrate affects the mechanism and the success of deposition.

High-performance polymers such as poly(ether ether ketone) (PEEK) are appealing for a wide variety of industrial and medical applications due to their excellent mechanical properties. However, these applications are often limited by relatively low thermal stability and conductivity compared to metals. Many methods developed to metallize polymers, including vapor deposition and thermal spray processes, can lead to poor quality control, low deposition rate, and high cost. Thus, cold spray is a promising potential alternative to rapidly and inexpensively produce polymer-metal composites. In this study, we investigated the deposition characteristics of metalpolymer composite feedstock, composed of PEEK powder with varying volume fractions of copper (Cu) flake added, onto a PEEK substrate. We prepared the Cu-PEEK composite powder in varying compositions by two methods: hand-mixing the powders and cryogenically milling the powders. Scanning electron microscopy (SEM) of the feed mixtures shows that cryogenically milling the polymer and metal powders together created uniformly distributed micron-scale domains of Cu on PEEK particle surfaces, and vice versa, as well as consolidating much of the porous Cu flake. In lowpressure cold spray, the relatively large volume fractions of PEEK in the composite mixtures allowed for lower operating temperatures than those commonly used in PEEK metallization (300-500 °C). While the deposition efficiencies of each mixture were relatively similar in single-layer experiments, deposits formed after multiple passes showed significant changes in deposition efficiency and composition in PEEK-rich feedstock mixtures. SEM of deposit surfaces and cross-sections revealed multiple co-dominant mechanisms of deposition, which affect both the porosity and final composition of the deposit. Though present in all samples analyzed, the effects of cryogenic milling were more prevalent at lower Cu concentrations.

Polymers have proven to be challenging to cold spray, particularly with high efficiency and quality when using inexpensive nitrogen (N2) and air propellants. Helium (He) when used as a process propellant can improve spray deposit properties but is often undesirable due to its limited availability and high cost. In this study, additives of multiple particle sizes and materials were mixed with polymer powder in an effort to improve the performance of polymer sprays using mainly N2 as a process propellant. The effects of additives on deposit microstructure were investigated by precise ion-beam polishing of deposit cross sections and subsequent electron microscope imaging. Additional metrics including the density and post - spray composition of deposits were investigated to quantify the peening effect and the amount of embedded additive. Additives, regardless of size, were observed to embed in the spray deposits. Additionally, hard-phase additives demonstrated nozzle-cleaning properties that continually remove polymer fouling on the nozzle walls. Inversely, sprays with polymer powder and no additives tended to clog the nozzle throat and diverging section as a result of continual fouling.

Diabetic Bladder Dysfunction (DBD) is a prevalent diabetic complication that is recalcitrant to glucose control. Using the Akita mouse model (type 1) bred to be NLRP3+/+ or NLRP3-/-, we previously found that females (mild hyperglycemia) progress from an overactive to an underactive bladder phenotype and this progression was dependent on NLRP3-induced inflammation. Here we examined DBD in the male Akita (severe hyperglycemia) and found by urodynamics only a compensated underactive-like phenotype (increased void volume and decreased frequency but unchanged efficiency). Surprisingly, this phenotype was still present in the NLRP3-/- strain and so was not dependent on NLRP3 inflammasome-induced inflammation. Examining the cause of the compensated underactive-like phenotype, we assessed overall nerve density and afferent nerves (Aδ-fibers). Both were decreased in density during diabetes but denervation was absent in the diabetic NLRP3-/- strain so it was deemed unlikely to cause the underactive-like symptoms. Changes in bladder smooth muscle (BSM) contractility to cell depolarization and receptor activation were also not responsible as KCl (depolarizing agent), carbachol (muscarinic agonist) and α, β-methylene-ATP (purinergic agonist) elicited equivalent contractions in denuded bladder strips in all groups. However, electrical field stimulation revealed a diabetes-induced decrease in contractility that was not blocked in the NLRP3-/- strains suggesting that bladder compensated underactive-like phenotype in the male Akita is likely through a decrease in efferent neurotransmitter release.

This paper proposes a method to determine harmonic pollution of consumers connected to single-phase low voltage distribution system. Today, the power quality of the system is greatly affected due to the penetration of non-linear loads. As per the IEEE standard 519-2014, both utility and consumers are responsible for reducing the harmonic pollution. This necessitates the quantification of harmonic contribution of individual consumers. The proposed method defines harmonic distortion indices for current and voltage separating consumer harmonic contribution at the point of common coupling (PCC). These indices are used to determine non-fundamental apparent power associated with each consumer connected to the PCC. Simulation results point to the effectiveness of this method in separating the harmonic contribution of consumer and utility. Experimental investigations are also carried out with several linear and non-linear loads comparing the performance of the method with the power definitions mentioned in IEEE standard 1459-2010.

The vacuolar H+-ATPase is an enzymatic complex that functions in an ATP-dependent manner to pump protons across membranes and acidify organelles, thereby creating the proton/pH gradient required for membrane trafficking by several different types of transporters. We describe heterozygous point variants in ATP6V0C, encoding the c-subunit in the membrane bound integral domain of the vacuolar H+-ATPase, in 27 patients with neurodevelopmental abnormalities with or without epilepsy. Corpus callosum hypoplasia and cardiac abnormalities were also present in some patients. In silico modelling suggested that the patient variants interfere with the interactions between the ATP6V0C and ATP6V0A subunits during ATP hydrolysis. Consistent with decreased vacuolar H+-ATPase activity, functional analyses conducted in Saccharomyces cerevisiae revealed reduced LysoSensor fluorescence and reduced growth in media containing varying concentrations of CaCl2. Knockdown of ATP6V0C in Drosophila resulted in increased duration of seizure-like behaviour, and the expression of selected patient variants in Caenorhabditis elegans led to reduced growth, motor dysfunction and reduced lifespan. In summary, this study establishes ATP6V0C as an important disease gene, describes the clinical features of the associated neurodevelopmental disorder and provides insight into disease mechanisms. ispartof: BRAIN vol:146 issue:4 ispartof: location:England status: Published online
