Envisaging antiferromagnetic spintronics pivots on two key criteria of high transition temperature and tuning of underlying magnetic order using straightforward application of magnetic field or electric current. Here, we show that NiSi metal can provide suitable new platform in this quest. First, our study unveils high temperature antiferromagnetism in single crystal NiSi with $T_{N} \geq 700$ K. Antiferromagnetic order in NiSi is accompanied by the non-centrosymmetric magnetic character with small ferromagnetic component in a-c plane. Second, we find that NiSi manifests distinct magnetic and electronic hysteresis responses to field applications due to the disparity in two moment directions. While magnetic hysteresis is characterized by one-step switching between ferromagnetic states of uncompensated moment, electronic behavior is ascribed to metamagnetic switching phenomena between non-collinear spin configurations. Ferroptosis is mediated by lipid peroxidation of phospholipids containing polyunsaturated fatty acyl moieties. Glutathione, the key cellular antioxidant capable of inhibiting lipid peroxidation via the activity of the enzyme glutathione peroxidase 4 (GPX-4), is generated directly from the sulfur-containing amino acid cysteine, and indirectly from methionine via the transsulfuration pathway. Herein we show that cysteine and methionine deprivation (CMD) can synergize with the GPX4 inhibitor RSL3 to increase ferroptotic cell death and lipid peroxidation in both murine and human glioma cell lines and in ex vivo organotypic slice cultures. We also show that a cysteine-depleted, methionine-restricted diet can improve therapeutic response to RSL3 and prolong survival in a syngeneic orthotopic murine glioma model. Finally, this CMD diet leads to profound in vivo metabolomic, proteomic and lipidomic alterations, highlighting the potential for improving the efficacy of ferroptotic therapies in glioma treatment with a non-invasive dietary modification.

SUMMARYGlioma cells hijack developmental transcriptional programs to control cell state. During neural development, lineage trajectories rely on specialized metabolic pathways. However, the link between tumor cell state and metabolic programs is poorly understood in glioma. Here we uncover a glioma cell state-specific metabolic liability that can be leveraged therapeutically. To model cell state diversity, we generated genetically engineered murine gliomas, induced by deletion of p53 alone (p53) or with constitutively active Notch signaling (N1IC), a pathway critical in controlling cellular fate. N1IC tumors harbored quiescent astrocyte-like transformed cell states while p53 tumors were predominantly comprised of proliferating progenitor-like cell states. N1IC cells exhibit distinct metabolic alterations, with mitochondrial uncoupling and increased ROS production rendering them more sensitive to inhibition of the lipid hydroperoxidase GPX4 and induction of ferroptosis. Importantly, treating patient-derived organotypic slices with a GPX4 inhibitor induced selective depletion of quiescent astrocyte-like glioma cell populations with similar metabolic profiles.

We observe linear and nonlinear light localization at the edges and in the corners of truncated moir\'e lattices created by the superposition of periodic mutually-twisted at Pythagorean angles square sublattices. Experimentally exciting corner linear modes in the fs-laser written moir\'e lattices we find drastic differences in their localization properties in comparison with the bulk excitations. We also address the impact of nonlinearity on the corner and bulk modes and experimentally observe the crossover from linear quasi-localized states to the surface solitons emerging at the higher input powers. The burden of coronavirus disease 2019 (COVID-19) in children represents a fraction of cases worldwide, yet a subset of those infected are at risk for severe disease. We measured plasma severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) RNA in a cohort of 103 children hospitalized with COVID-19 with diverse clinical manifestations. SARS-CoV-2 RNAemia was detected in 27 (26%) of these children, lasted for a median of 6 (interquartile range, 2–9) days, and was associated with higher rates of oxygen administration, admission to the intensive care unit, and longer hospitalization.

There is a continuous demand for high-power, high-efficiency, and high-density converters in various existing and emerging applications such as large motor drives, electric vehicles, more electric aircraft, and utility applications. Compared with conventional silicon (Si) insulated gate bipolar transistors (IGBTs), silicon carbide (SiC) MOSFETs can have lower switching loss and higher frequency operation capabilities, which enables higher efficiency and higher power density at the converter level. Meanwhile, to achieve a high power rating such as 500 kW, how to extend the current capabilities of existing SiC power modules, the choice of parallel schemes, the selection of passive components, cooling and mechanical structure design pose clear challenges in maximizing the system power density and efficiency. This article presents a comprehensive design of the world-first air-cooled 500 kW SiC dc/ac three-phase converter, which achieves a record-high power density of 1.246 MW/m3 (1.246 kW/L) and an efficiency over 98.5%. The converter has been tested in the experiment to the full power rating, which provides benchmark results for high power SiC converters and their applications.

Dyslipidemia is an important risk factor of atherosclerotic cardiovascular disease (ASCVD). Statins delay the occurrence and development of ASCVD, and reduce the risk of cardiovascular events and death. Due to safety concerns, there exist insufficient use of lipid-lowering agents and a high withdrawal rate of the agents in the elderly. Chemokine C‐C motif ligand 7 (CCL7), a member of CC chemokine subfamily, plays pivotal roles in numerous inflammatory diseases. Hyper‐activation of inflammation is an important characteristic of abdominal aortic aneurysm (AAA). Therefore, in the present study, we aimed to determine the effect of CCL7 on AAA formation. CCL7 abundance in aortic tissue and macrophage infiltration were both increased in angiotensin II (Ang II)‐induced AAA mice. Ex vivo, CCL7 promoted macrophage polarization towards M1 phenotype. This effect was reversed by the blockage of CCR1, a receptor of CCL7. CCL7 up‐regulated JAK2/STAT1 protein level in macrophage, and CCL7‐induced M1 activation was suppressed by JAK2/STAT1 pathway inhibition. To verify the effect of CCL7 on AAA in vivo, either CCL7‐neutralizing antibody (CCL7‐nAb) or vehicles were intraperitoneally injected 24 hours prior to Ang II infusion and subsequently every three days for 4 weeks. CCL7‐nAb administration significantly attenuated Ang II‐induced luminal and external dilation as well as pathological remodelling. Immunostaining showed that CCL7‐nAb administration significantly decreased aneurysmal macrophage infiltration. In conclusion, CCL7 contributed to Ang II‐induced AAA by promoting M1 phenotype of macrophage through CCR1/JAK2/STAT1 signalling pathway.

CePtSi2 was reported to exhibit an antiferromagnetic order below T*=1.8 K at ambient pressure, a valence state change at ~1.2 GPa, and superconductivity in the range between 1.4 and 2.1 GPa with the maximum transition temperature of 0.14 K [T. Nakano et al., Phys. Rev. B 79, 172507 (2009)]. We have performed polycrystalline and single crystal neutron diffraction experiments to determine the magnetic structure under ambient and high pressures. We found that incommensurate magnetic peaks with the magnetic propagation vector of (0.32, 0, 0.11) at ambient pressure below T_{SDW}~1.25 K. Those magnetic peaks which originate from a spin-density-wave order with the easy axis along the c axis and an averaged ordered moment of 0.45(5) mu_B, suggesting that there may be an intermediate phase between T* and T_{SDW}. Applying pressures, the magnetic propagation vector shows no change and the magnetic order disappears around 1.0 GPa, which is much lower than the critical pressure for the superconducting phase. The results suggest that other than magnetic fluctuations may play a primary role in the superconducting pairing mechanism., 6 pages, 6 figures

The full-waveform data is the core data of GaoFen-7 (GF-7) satellite laser altimeter, and evaluation of waveform data quality is an important step and premise for satellite laser altimetry and quality control. In this paper, the full waveform data quality assessment and analysis of GF-7 laser altimeter is implemented during the period of on-orbit experiment, and the real waveform data of many orbits is used to quantitatively describe the characteristic parameters of the transmitted waveform and the signal-to-noise ratio (SNR), and the result of two beam lasers is compared. The conclusion is validated that the GF-7 laser altimeter can obtain effective waveform data and the echo waveform availability of the experimental data is approximate 72.59%, moreover, the quality of beam 1 is slightly better than that of the beam 2. The laser temperature is an important indication of the quality of transmitted waveform according to the SNR changing. The good SNR value of the waveform and small footprint size will be helpful for the terrain information extraction and analysis, although the repetition frequency is low.

In recent years, audio positioning technology has been more favoured because it is not only highly accurate but also universally applicable to the mass users. In line with this trend, a chip dedicated to indoor positioning, the Kepler A100, has been released lately, which ranges by estimating the arrival time of the audio chirp. As with radio ranging techniques, sound-based ranging methods is subject to significant observation errors due to Non-Line-Of-Sight (NLOS) or multi-path effects, while Pedestrian Dead Reckoning (PDR) based on inertial data have the disadvantage of error accumulation. Both make localization challenging. In this paper, the ranging performance of the Kepler A100 chip is evaluated and experiments are conducted in two typical indoor scenarios to verify the stability and sub-metre accuracy of the ranging results. Meanwhile, this paper implements an indoor positioning system combining audio chirp/Micro-Electro-Mechanical Systems (MEMS)/floor map based on particle filtering algorithm. A new mapping constraint method is applied in the system, which can effectively constrain the particles in a reasonable space. Experiments are conducted in two complex office environments and the results show that the error of the proposed algorithm is about 0.65m in 95% of cases, a reduction of more than 40% compared to only-PDR or only-audio. The effectiveness and stability of the method is demonstrated.

Luteoloside (Lute), a bioactive natural ingredient, widely exists in nature and possesses hepatoprotective and hepatocyte proliferation-promoting properties. This study aimed to investigate whether Lute could counteract non-alcoholic fatty liver disease (NAFLD)-caused hepatocyte damage via its stimulation of hepatocyte regeneration efficacy and to explore the involved mechanism. LO2 cells and primary hepatocytes were used to examine the hepatocyte proliferation effects of Lute under physiological conditions and in the palmitic acid (PA)- induced in vitro model of NAFLD. STAT3 and cell cycle-related proteins (cyclin D1, c-myc and p21) were evaluated by Western blot. Under physiological conditions, LO2 cells and primary hepatocytes treated with various concentration of Lute for 12 and 24 h showed increased hepatocyte proliferation, especially with 20 μM treatment for 24 h. More notably, under the model conditions, co-incubation with 20 μM of Lute also markedly reversed PA-induced inhibition of cell proliferation and viability in primary hepatocytes. Mechanistically, Lute could activate STAT3 and subsequently increase cyclin D1 and cmyc expression, which positively regulates cell cycle progression, and decrease expression of p21, an inhibitor of cell cycle progression. Furthermore, Luteinduced hepatocyte proliferation-promoting efficacy was abolished by STAT3 inhibitor stattic. Collectively, Lute can alleviate PA-induced hepatocyte damage via activating STAT3-mediated hepatocyte regeneration.

White matter injury is the most common form of brain injury in premature infants, which is highly associated with poor brain development. White matter segmentation in brain MR images is very important for early detection of brain injury in premature infants. In this paper, a deep learning based approach is proposed to segment the white matter region of premature infants. We retrospectively analyzed 63 preterm infants with gestational age < 37 weeks who underwent MRI examination. Firstly, the U-Net deep learning neural network model is used to automatically segment white matter, U-Net++ and ResU-Net are used as controlled experimental groups, and the Dice coefficient and Iou coefficient are used as evaluation indicators for white matter segmentation. In the above experiments, the U-Net network has the best experimental results. Segmenting white matter on the training set with a Dice coefficient of 0.90 and an Iou of 0.82, respectively. The test set was used to validate the performance of the proposed model, with a Dice coefficient of 0.83 and an Iou of 0.71, respectively. Secondly, we performed image binarization step on the segmented white matter, and used Gaussian filtering, flood filling and connected component analysis to process the automatically segmented white matter region, and finally obtain a white matter region with clear boundaries.

Large time-stepping is important for efficient long-time simulations of deterministic and stochastic Hamiltonian dynamical systems. Conventional structure-preserving integrators, while being successful for generic systems, have limited tolerance to time step size due to stability and accuracy constraints. We propose to use data to innovate classical integrators so that they can be adaptive to large time-stepping and are tailored to each specific system. In particular, we introduce NySALT, Nystr\"{o}m-type inference-based schemes adaptive to large time-stepping. The NySALT has optimal parameters for each time step learnt from data by minimizing the one-step prediction error. Thus, it is tailored for each time step size and the specific system to achieve optimal performance and tolerate large time-stepping in an adaptive fashion. We prove and numerically verify the convergence of the estimators as data size increases. Furthermore, analysis and numerical tests on the deterministic and stochastic Fermi-Pasta-Ulam (FPU) models show that NySALT enlarges the maximal admissible step size of linear stability, and quadruples the time step size of the St\"{o}rmer--Verlet and the BAOAB when maintaining similar levels of accuracy., Comment: 26 pages, 7 figures

There is a compelling need for new therapeutic strategies for glioblastoma multiforme (GBM). Preclinical target and therapeutic discovery for GBMs is primarily conducted using cell lines grown in serum-containing media, such as U-87 MG, which do not reflect the gene expression profiles of tumors found in GBM patients. To address this lack of representative models, we sought to develop a panel of patient-derived GBM models and characterize their genomic features, using RNA sequencing (RNA-seq) and growth characteristics, both when grown as neurospheres in culture, and grown orthotopically as xenografts in mice. When we compared these with commonly used GBM cell lines in the Cancer Cell Line Encyclopedia (CCLE), we found these patient-derived models to have greater diversity in gene expression and to better correspond to GBMs directly sequenced from patient tumor samples. We also evaluated the potential of these models for targeted therapy, by using the genomic characterization to identify small molecules that inhibit the growth of distinct subsets of GBMs, paving the way for precision medicines for GBM.

Cardiac ankyrin repeat protein (CARP) is a cardiac-specific stress-response protein which exerts diverse effects to modulate cardiac remodeling in response to pathological stimuli. We examined the role of CARP in postnatal cardiac development and function under basal conditions in mice. Transgenic mice that selectively overexpressed CARP in heart (CARP Tg) exhibited dilated cardiac chambers, impaired heart function, and cardiac fibrosis as assessed by echocardiography and histological staining. Furthermore, the mice had a shorter lifespan and reduced survival rate in response to ischemic acute myocardial infarction. Immunofluorescence demonstrated the overexpressed CARP protein was predominantly accumulated in the nuclei of cardiomyocytes. Microarray analysis revealed that the nuclear localization of CARP was associated with the suppression of calcium-handling proteins. In vitro experiments revealed that CARP overexpression resulted in decreased cell contraction and calcium transient. In post-mortem cardiac specimens from patients with dilated cardiomyopathy and end-stage heart failure, CARP was significantly increased. Taken together, our data identified CARP as a crucial contributor in dilated cardiomyopathy and heart failure which was associated with its regulation of calcium-handling proteins.

Efficient simulation of SDEs is essential in many applications, particularly for ergodic systems that demand efficient simulation of both short-time dynamics and large-time statistics. However, locally Lipschitz SDEs often require special treatments such as implicit schemes with small time-steps to accurately simulate the ergodic measure. We introduce a framework to construct inference-based schemes adaptive to large time-steps (ISALT) from data, achieving a reduction in time by several orders of magnitudes. The key is the statistical learning of an approximation to the infinite-dimensional discrete-time flow map. We explore the use of numerical schemes (such as the Euler-Maruyama, a hybrid RK4, and an implicit scheme) to derive informed basis functions, leading to a parameter inference problem. We introduce a scalable algorithm to estimate the parameters by least squares, and we prove the convergence of the estimators as data size increases. We test the ISALT on three non-globally Lipschitz SDEs: the 1D double-well potential, a 2D multi-scale gradient system, and the 3D stochastic Lorenz equation with degenerate noise. Numerical results show that ISALT can tolerate time-step magnitudes larger than plain numerical schemes. In order to study the effect of the layup structure on the static strength and low-velocity impact strength of carbon fiber/epoxy composite (CFRP) laminates, theoretical simulation analysis under different laying angles have been carried out. In this study, Finite Element Analysis (FEA) models for different CFRP laminate specimens are created using ANSYS Workbench by changing the relative volume fraction of 0°, 45° and 90° plies in each specimen and their relative location. The FEA results revealed that the increase of relative volume of 90° ply will improve the impact the impact resistance performance, while the increase of relative volume of 45° ply will take the opposite effect. Moreover, when the relative volume fraction of 0°, 45° and 90° plies are the same, the strength performance of the laminate cannot be improved by changing the thickness of the outermost layer. The study illustrated the significant effects of different stacking sequences and laying angles on the tensile and flexural failure mechanisms in composite laminates, leading to some suggestions to improve the design of composite laminates.

Cellular responses to environmental changes are encoded in the complex temporal patterns of signaling proteins. However, quantifying the accumulation of information over time to direct cellular decision-making remains an unsolved challenge. This is, in part, due to the combinatorial explosion of possible configurations that need to be evaluated for information in time-course measurements. Here, we develop a quantitative framework, based on inferred trajectory probabilities, to calculate the mutual information encoded in signaling dynamics while accounting for cell-cell variability. We use it to understand NFκB transcriptional dynamics in response to different immune threats, and reveal that some threats are distinguished faster than others. Our analyses also suggest specific temporal phases during which information distinguishing threats becomes available to immune response genes; one specific phase could be mapped to the functionality of the IκBα negative feedback circuit. The framework is generally applicable to single-cell time series measurements, and enables understanding how temporal regulatory codes transmit information over time., Understanding how cells discriminate between stimuli is an ongoing challenge. Here, the authors propose a mathematical framework for inferring the mutual information encoded in temporal signaling dynamics and use it to study how information is transmitted over time in response to different stimuli in NFκB, MAPK and p53 signaling pathways.

Because of the limited treatment options available, oral lopinavir/ritonavir (LPR) was used for treating coronavirus disease (COVID-19) in pediatric patients. This study aimed to assess the efficacy and safety of LPR in COVID-19 pediatric patients with mild symptoms.This retrospective multicenter analysis included hospitalized children with mild COVID-19 who received LPR at one of 13 hospitals in China from January 1, 2020, to June 1, 2020. Patients treated with LPR were matched with patients not treated with LPR (1:4) according to age, sex, and length of symptom onset and hospitalization. Descriptive statistics and non-parametric tests were applied to compare differences between groups. Kaplan-Meier probability curves and Cox regression models were used to analyze nasal swab turning negative time (recovery time) and hospital discharge days.In total, 23 patients treated with LPR were matched with 92 untreated controls. The median age of patients was 6 years, and 56.52% of them were male. All patients were discharged from the hospital after being cured. The treatment group had a longer nasal swab turning negative time (hazard ratio [HR] 5.33; 95% CI: 1.94-14.67; p = 0.001) than the control group. LPR treatment was also associated with a longer hospitalization time (HR 2.01; 95% CI: 1.24-3.29; p = 0.005). After adjusting for the influence of LPR treatment, adverse drug reaction events were associated with a longer nasopharyngeal swab negative time (HR 4.67; 95% CI 1.35-16.11; p = 0.015).For children with mild COVID-19, LPR is inferior to conventional treatment in reducing virus shedding time and hospitalization duration and is associated with increased adverse reactions.
