The aim of the paper is to describe autocompact objects in Ab5-categories, i.e. objects in cocomplete abelian categories with exactness preserving filtered colimits of exact sequences, whose covariant Hom-functor commutes with copowers of the object itself. A characterization of non-autocompact object is given, a general criterion of autocompactness of an object via the structure of its endomorphism ring is presented and a criterion of autocompactness of products is proven.

The weather conditions formed in troposphere causes the greatest signal attenuation in satellite communication systems especially at frequencies above 10 GHz. This paper describes possible signal attenuations on the satellite-earth path due to rain and clouds. It was measured whether it is advantageous to use the Ku band for data transmission over other bands. The measurement was carried out in the Czech Republic using a beacon signal from Eutelsat 12W satellite at 12.5 GHz. Clouds and the rainfall rate at the measured location were obtained from the CHMI portal. The measurements show that the clouds cause negligible attenuation. Significant attenuation was caused by rainfall. The measured values of slant path rain show a significant decrease in signal strength even in light rain. The measured cumulative rainfall rate was found to be close to the ITU-R model. The measurements show that the Ku band is advantageous for data transmission in rain poorer regions including Central Europe.

The study of molecular genetic variability in genetic resources of farm animals is necessary for optimal conduct of their breeding and for understanding of relations between molecular markers and traits. The determination of variability in the CRC and the MYF4 genes and the evaluation of associations between these genetic markers and the total number born, the number born alive and the number weaned of piglets, weaning weight of piglets and farrowing interval were carried out in population of 102 Přeštice Black-Pied sows. The frequency of the allele CRCn (0.083) was found out to be markedly lower than the frequency of the allele CRCN and there was observed no sow of homozygous recessive genotype in the investigated sample population. In MYF4 locus, the higher frequency of the MYFB allele was detected (0.711). A significant influence of the CRC gene polymorphism on number of piglets alive born in the 1st and the 1st–6th litters was discovered. In the total number of born piglets the significant disparity were ascertained between the MYF4A/MYF4A and MYF4A/MYF4B, MYF4A/MYF4A and MYF4B/MYF4B sows in the 2nd–6th litters and the 1st–6th litters. The obtained data are exploitable in genetic structure mapping, management of breeding and improvement of this breed and comparison with other pig genetic resources.

The social and economic changes that began over 25 years ago in post-communist Eastern Europe and the countries of the former Soviet Union also affected the forestry sector. Forested areas were privatised in many countries, and timber harvesting, also in state-owned forests, was contracted out to private sector logging companies. An analysis was conducted of the following countries: Belarus, Bulgaria, Czech Republic, Estonia, Latvia, Lithuania, Poland, Romania, Slovakia, and Ukraine. The basic parameters of forestry, like the characteristics of forest resources, the volume of harvested timber and logging processes used, were given for each country. Special attention was paid to the methods of timber harvesting. The main findings of the study are that various methods are used in Eastern Europe depending on site and forest conditions. In some countries, especially the wealthier ones, a dynamic increase in the cut-to-length method is observed, with the use of harvesters and forwarders.

© 2016 by The American Society for Biochemistry and Molecular Biology, Inc. Kunitz-type (KT) protease inhibitors are low molecular weight proteins classically defined as serine protease inhibitors. We identified a novel secreted KT inhibitor associated with the gut and parenchymal tissues of the infective juvenile stage of Fasciola hepatica, a helminth parasite of medical and veterinary importance. Unexpectedly, recombinant KT inhibitor (rFhKT1) exhibited no inhibitory activity toward serine proteases but was a potent inhibitor of the major secreted cathepsin L cysteine proteases of F. hepatica, FhCL1 and FhCL2, and of human cathepsins L and K (Ki ô 0.4-27 nM). FhKT1 prevented the autocatalytic activation of FhCL1 and FhCL2 and formed stable complexes with the mature enzymes. Pulldown experiments from adult parasite culture medium showed that rFhKT1 interacts specifically with native secreted FhCL1, FhCL2, and FhCL5. Substitution of the unusual P1 Leu15 within the exposed reactive loop of FhKT1 for the more commonly found Arg (FhKT1Leu15 / Arg15 ) had modest adverse effects on the cysteine protease inhibition but conferred potent activity against the serine protease trypsin (Ki ô 1.5 nM). Computational docking and sequence analysis provided hypotheses for the exclusive binding of FhKT1 to cysteine proteases, the importance of the Leu15 in anchoring the inhibitor into the S2 active site pocket, and the inhibitor's selectivity toward FhCL1, FhCL2, and human cathepsins L and K. FhKT1 represents a novel evolutionary adaptation of KT protease inhibitors by F. hepatica, with its prime purpose likely in the regulation of the major parasite-secreted proteases and/or cathepsin L-like proteases of its host.

Certainty, fairness and control over contracts: the DCFR and the draft Civil Code.

The focus of this study was to analyze the content, distribution, and comparative genome relationships of 996 chromosome bin-mapped expressed sequence tags (ESTs) accounting for 2266 restriction fragments (loci) on the homoeologous group 3 chromosomes of hexaploid wheat (Triticum aestivum L.). Of these loci, 634, 884, and 748 were mapped on chromosomes 3A, 3B, and 3D, respectively. The individual chromosome bin maps revealed bins with a high density of mapped ESTs in the distal region and bins of low density in the proximal region of the chromosome arms, with the exception of 3DS and 3DL. These distributions were more localized on the higher-resolution group 3 consensus map with intermediate regions of high-mapped-EST density on both chromosome arms. Gene ontology (GO) classification of mapped ESTs was not significantly different for homoeologous group 3 chromosomes compared to the other groups. A combined analysis of the individual bin maps using 537 of the mapped ESTs revealed rearrangements between the group 3 chromosomes. Approximately 232 (44%) of the consensus mapped ESTs matched sequences on rice chromosome 1 and revealed large- and small-scale differences in gene order. Of the group 3 mapped EST unigenes ∼21 and 32% matched the Arabidopsis coding regions and proteins, respectively, but no chromosome-level gene order conservation was detected.

A total of 1918 loci, detected by the hybridization of 938 expressed sequence tag unigenes (ESTs) from 26 Triticeae cDNA libraries, were mapped to wheat (Triticum aestivum L.) homoeologous group 4 chromosomes using a set of deletion, ditelosomic, and nulli-tetrasomic lines. The 1918 EST loci were not distributed uniformly among the three group 4 chromosomes; 41, 28, and 31% mapped to chromosomes 4A, 4B, and 4D, respectively. This pattern is in contrast to the cumulative results of EST mapping in all homoeologous groups, as reported elsewhere, that found the highest proportion of loci mapped to the B genome. Sixty-five percent of these 1918 loci mapped to the long arms of homoeologous group 4 chromosomes, while 35% mapped to the short arms. The distal regions of chromosome arms showed higher numbers of loci than the proximal regions, with the exception of 4DL. This study confirmed the complex structure of chromosome 4A that contains two reciprocal translocations and two inversions, previously identified. An additional inversion in the centromeric region of 4A was revealed. A consensus map for homoeologous group 4 was developed from 119 ESTs unique to group 4. Forty-nine percent of these ESTs were found to be homoologous to sequences on rice chromosome 3, 12% had matches with sequences on other rice chromosomes, and 39% had no matches with rice sequences at all. Limited homology (only 26 of the 119 consensus ESTs) was found between wheat ESTs on homoeologous group 4 and the Arabidopsis genome. Forty-two percent of the homoeologous group 4 ESTs could be classified into functional categories on the basis of blastX searches against all protein databases.

A novel and quick high-performance liquid chromatography (HPLC) method with UV spectrophotometric detection was developed and validated for the determination of five compounds in topical gel. The described method is suitable for simultaneous determination of active component ketoprofen, two preservatives methylparaben and propylparaben and two degradation products of ketoprofen - 3-acetylbenzophenone and 2-(3-carboxyphenyl) propionic acid - in a topical cream after long-term stability tests using ethylparaben as an internal standard. The chromatographic separation was performed on a 5 μm Supelco Discovery C18 column (125 mm × 4 mm i.d., Sigma-Aldrich); the optimal mobile phase for separation of ketoprofen, methylparaben, propylparaben, degradation products 3-acetylbenzophenone and 2-(3-carboxyphenyl) propionic acid and ethylparaben as internal standard consists of a mixture of acetonitril, water and phosphate buffer pH 3.5 (40:58:2, v/v/v). At a flow rate of 1.0 ml min -1 and detection at 233 nm, the total time of analysis was less than 10 min. The method was applied for routine analysis (batch analysis and stability tests) of these compounds in topical pharmaceutical product. © 2004 Elsevier B.V. All rights reserved.

Targeted homoeologous recombination mediated by the absence of the Phi locus is currently the most efficient technique by which foreign genes can be introgressed into polyploid wheat species. Because intra-arm homoeologous double cross-overs are rare, introgressed foreign genes are usually on terminal foreign chromosome segments. Since the minimum length of such a segment is determined by the position of a gene in the chromosome, large chromosome segments with undesirable genetic effects are often introgressed. Introgression of foreign genes on short interstitial segments based on two cycles of homoeologous recombination is described here. The utility of the technique is demonstrated by the introgression of the Kna1 locus, which controls K+/Na+selectivity in T. aestivum L., on short interstitial segments of chromosome 4D into chromosome 4B of Triticum turgidum L. The level of recombination in a homoeologous segment is not significantly affected by a juxtaposed proximal homologous segment in the absence of the Ph1 locus.

In most allopolyploid plants, only homogenetic chromosome pairing occurs in meiosis, as a result of the recognition of genome differentiation by the genetic system regulating meiotic chromosome pairing. The nature of differentiation between chromosomes of closely related genomes is examined here by investigating recombination between wheat chromosome 1A and the closely related homoeologous chromosome 1Am of Triticum monococcum. The recognition of the differentiation between these chromosomes by the Ph1 locus, which prevents heterogenetic chromosome pairing in wheat, is also investigated. Chromosomes 1A and 1Am are shown to be colinear, and it is concluded that they are differentiated 'substructurally.' This substructural differentiation is argued to be recognized by the Ph1 locus. In the absence of Ph1, the distribution and frequencies of crossing over between the 1A and 1Am homoeologues were similar to the distribution and frequencies of crossing over between 1A homologues. The cytogenetic and evolutionary significance of these findings is discussed.

A genetic map of diploid wheat, Triticum monococcum L., involving 335 markers, including RFLP DNA markers, isozymes, seed storage proteins, rRNA, and morphological loci, is reported. T. monococcum and barley linkage groups are remarkably conserved. They differ by a reciprocal translocation involving the long arms of chromosomes 4 and 5, and paracentric inversions in the long arm of chromosomes 1 and 4; the latter is in a segment of chromosome arm 4L translocated to 5L in T. monococcum. The order of the markers in the inverted segments in the T. monococcum genome is the same as in the B and D genomes of T. aestivum L. The T. monococcum map differs from the barley maps in the distribution of recombination within chromosomes. The major 5S rRNA loci were mapped on the short arms of T. monococcum chromosomes 1 and 5 and the long arms of barley chromosomes 2 and 3. Since these chromosome arms are colinear, the major 5S rRNA loci must be subjected to positional changes in the evolving Triticeae genome that do not perturb chromosome colinearity. The positional changes of the major 5S rRNA loci in Triticeae genomes are analogous to those of the 18S-5.8S-26S rRNA loci.

The Common European Research Information Format (CERIF) is an established standard for Current Research Information Systems (CRISs) facing the increasing need for information sharing and exchange. euroCRIS released the first official CERIF XML exchange format in 2007; it followed the structure of the relational data model. Based on experience with the format and consulting with the CRIS community on newer interoperation and exchange concepts, the authors proposed an update to the CERIF XML exchange format. This updated CERIF XML aimed at compactness with expression and backwards compatibility. With this article, we provide insight into the motivation for change, present the updated format, and finally outline possible next steps.

The harvester technology is increasingly used in Central and Eastern Europe, yet is still costly and thus has be used efficiently. This study focused on the analysis of the development of operational costs of two mid-performance harvesters (JD770D and JD1070D) over a ten- year period, from 2006 to 2017. The costs were aggregated into four categories: depreciation, services, material, and personnel costs) and were analy- sed annually. The Harvesters were used in thinning under different conditions, which has to be considered when interpreting our results. The mean stem size processed by the JD770D was 0.11 m3 under bark (u.b.) and it harvested 16, 793 m3 u.b. per year of timber operating for 3, 166 machine hours (MH) per year. The mean costs per m3 u.b. of produced timber ranged from 6.6 to 10.2 €/m3 u.b. The JD1070D was harvesting trees with a mean stem size of 0.29 m3 u.b. and it harvested 26, 837 m3 u.b. per year on average, with an operational time of 3, 067 MH. The mean costs per m3 u.b. of produced timber ranged from 5.4 to 6.9 €/m3 u.b. Personnel costs were important in driving costs and their share on the total operational costs was 40 % for JD770D and 31 % for JD1070D. Material costs were also comparably high with 23 and 28 % of the total ope- rational costs of JD770D and JD1070D, respectively. Depreciation accounted for 18 % and 23 % for the two studied harvesters. The temporal utilization of the harvesters ranged from 63 to 96 %, with higher values for the JD770D. With increasing number of operators, the utilization rate decreased. Given the importance of personnel costs in the studied harvester further analyses should focus on optimizing this decisive driver of costs in mid-performance harvesters in Central Europe
