1. Triazole pesticides exposure impaired steroidogenesis associated to an increase in AHR and CAR expression in testis and altered sperm parameters in chicken

Abstract: Since several decades, we observe the decline of various bird populations that could be partly linked to the agricultural intensification and the use of large amount of pesticides. Even if triazoles compounds are the most widely used fungicides, their effects on the reproductive parameters in birds are not clearly known. In the present study, we investigated the in vitro effects of 8 triazoles compounds alone (propiconazole (PP, from 0 to 10 µM), prothioconazole (PT), epoxiconazole (Epox), tetraconazole (TT), tebuconazole (TB), difenoconazole (Dif), cyproconazole (Cypro), metconazole (MC) (from 0 to 1 mM)) on the male chicken reproductive functions by using testis explants, primary Sertoli cells and sperm samples. In testis, all triazoles at the higher concentrations for 48 h inhibited lactate and testosterone secretion mostly in association with reduced expression of HSD3B and/or STAR mRNA levels. These data were also associated with increased expression of the nuclear receptors Aryl Hydrocarbon Receptor (AHR) and Constitutive Androstane Receptor (CAR) mRNA levels in testis and for all triazoles except for PP a reduction in Sertoli cell viability. When focusing on the sperm parameters, we demonstrated that most of the triazoles (MC, Epox, Dif, TB, TT and Cypro) at 0.1 or 1 mM for either 2, 12 or 24 min of exposure decreased sperm motility and velocity and increased the percentage of spermatozoa abnormal morphology. At the opposite, PP increased sperm motility in a dose dependent manner after 2 min of exposure whereas no significant effect was observed in response to PT whatever the dose and the time of exposure. Moreover, these effects were associated with an increase in the production of reactive oxygen species in spermatozoa. Taken together, most of the triazoles compounds impair testis steroidogenesis and semen parameters potentially through an increase in AHR and CAR expression and in oxidative stress, respectively. Abstract: Glyphosate-based herbicides (GBHs) are massively used in agriculture. However, few studies have investigated the effects of glyphosate-based herbicides on avian species although they are largely exposed via their food. Here, we investigated the potential reversibility of the effects of chronic dietary exposure to glyphosate-based herbicides in broiler hens. For 42 days, we exposed 32-week-old hens to glyphosate-based herbicides via their food (47 mg/kg/day glyphosate equivalent, glyphosate-based herbicides, n = 75) corresponding to half glyphosate’s no-observed-adverse-effect-level in birds. We compared their performance to that of 75 control animals (CT). Both groups (glyphosate-based herbicides and control animals) were then fed for 28 additional days without glyphosate-based herbicides exposure (Ex-glyphosate-based herbicides and Ex-control animals). Glyphosate-based herbicides temporarily increased the plasma glyphosate and AMPA (aminomethylphosphonic acid) concentrations. Glyphosate and aminomethylphosphonic acid mostly accumulated in the liver and to a lesser extent in the leg muscle and abdominal adipose tissue. Glyphosate-based herbicides also temporarily increased the gizzard weight and plasma oxidative stress monitored by TBARS (thiobarbituric acid reactive substances). Glyphosate-based herbicides temporarily decreased the cecal concentrations of propionate, isobutyrate and propionate but acetate and valerate were durably reduced. The cecal microbiome was also durably affected since glyphosate-based herbicides inhibited Barnesiella and favored Alloprevotella. Body weight, fattening, food intake and feeding behavior as well as plasma lipid and uric acid were unaffected by glyphosate-based herbicides. Taken together, our results show possible disturbances of the cecal microbiota associated with plasma oxidative stress and accumulation of glyphosate in metabolic tissues in response to dietary glyphosate-based herbicides exposure in broiler hens. Luckily, glyphosate-based herbicides at this concentration does not hamper growth and most of the effects on the phenotypes are reversible.

Abstract: International audience; Metformin is a drug used for the treatment of type 2 diabetes and disorders associated with insulin resistance. Metformin is also used in the treatment of pregnancy disorders such as gestational diabetes. However, the consequences of foetal exposure to metformin on the fertility of exposed offspring remain poorly documented. In this study, we investigated the effect of in utero metformin exposure on the fertility of female and male offspring. We observed that metformin is detectable in the blood of the mother and in amniotic fluid and blood of the umbilical cord. Metformin was not measurable in any tissues of the embryo, including the gonads. The effect of metformin exposure on offspring was sex specific. The adult females that had been exposed to metformin in utero presented no clear reduction in fertility. However, the adult males that had been exposed to metformin during foetal life exhibited a 30% reduction in litter size compared with controls. The lower fertility was not due to a change in sperm production or the motility of sperm. Rather, the phenotype was due to lower sperm head quality – significantly increased spermatozoa head abnormality with greater DNA damage – and hypermethylation of the genomic DNA in the spermatozoa associated with lower expression of the ten-eleven translocation methylcytosine dioxygenase 1 (TET1) protein. In conclusion, while foetal metformin exposure did not dramatically alter gonad development, these results suggest that metabolic modification by metformin during the foetal period could change the expression of epigenetic regulators such as Tet1 and perturb the genomic DNA in germ cells, changes that might contribute to a reduced fertility.

Abstract: International audience; Genetic selection in parental broiler breeders has increased their susceptibility to metabolic disorders and reproductive dysfunction. We have recently shown that maternal dietary grape seed extract (GSE) supplementation in hens improves fertility parameters, egg quality, oxidative stress in different tissues and the quality of F1 chicks. Here, we analysed the growth and fertility (both female and male) of the F1 generation animals and the quality of their offspring (F2 generation). Eggs issued from hens supplemented with GSE presented lower ROS production than control hens, suggesting a change in the embryonic environment. However, this did not affect the growth nor the body composition of male and female F1s from hatching to adulthood (37 weeks of age). At 37 weeks of age, the biochemistry analysis of the GSE-F1 muscle has revealed an increase in sensitivity to oxidative stress and a slight change in lipid composition. Both male and female F1-GSE groups presented a delay in puberty with a lower testis volume at 30 weeks of age and lower ovary development at 26 weeks of age. Adult GSE-F1 males did not present histological alterations of seminiferous tubules or semen production, but the semen quality was degraded due to higher oxidative stress and DNA-damaged spermatozoa compared with control F1 animals. In adult GSE-F1 females, despite the delay in puberty, the females laid more eggs of better quality (fewer broken eggs and a higher hatching rate). At hatching, the weight of the chicks from GSE-F1 females was reduced, and this effect was stronger in F2 male chicks (F2) compared with F2 control chicks (F2), because of the lower muscle volume. In conclusion, we can raise the hypothesis that maternal dietary GSE supplementation produces eggs with change in embryonic metabolism, which may affect in adulthood the fertility. The data obtained from the F1-GSE group pointed to a sex-specific modification with higher egg quality in females but semen sensitive to stress in males. Finally, male F2 chicks were leaner than control chicks. Thus, maternal dietary grape seed extract (GSE) supplementation in hens may impact on the fertility of the offspring in a sex-specific manner in subsequent generations.

Abstract: International audience; It is well known that adipokines are endocrine factors that are mainly secreted by white adipose tissue. Their central role in energy metabolism is currently accepted. More recently, their involvement in fertility regulation and the development of some reproductive disorders has been suggested. Data concerning the role of leptin and adiponectin, the two most studied adipokines, in the control of the reproductive axis are consistent. In recent years, interest has grown about some novel adipokines, chemerin, visfatin, resistin and apelin, which have been found to be strongly associated with obesity and insulin-resistance. Here, we will review their expression and role in male and female reproduction in humans and animal models. According to accumulating evidence, they could regulate the secretion of GnRH (Gonadotropin-Releasing Hormone), gonadotropins and steroids. Furthermore, their expression and that of their receptors (if known), has been demonstrated in the human and animal hypothalamo-pituitary-gonadal axis. Like leptin and adiponectin, these novel adipokines could thus represent metabolic sensors that are able to regulate reproductive functions according to energy balance changes. Therefore, after investigating their role in normal fertility, we will also discuss their possible involvement in some reproductive troubles known to be associated with features of metabolic syndrome, such as polycystic ovary syndrome, gestational diabetes mellitus, preeclampsia and intra-uterine growth retardation in women, and sperm abnormalities and testicular pathologies in men.

Abstract: International audience; Adipokines are secreted by adipose tissue and could be the link between obesity and infertility. Different studies investigated the involvement of adipokines in reproductive functions but only a few have looked into the male part. This review assesses adipokine functions on male reproductive parameters. Adiponectin seems to have a positive effect on sperm parameters, whereas other adipokines such as resistin or chemerin would have a rather deleterious effect on spermatogenesis. Semen parameters seem to be impacted when resistin and chemerin are increased: indeed, there is a decrease of sperm motility. Sperm morphology is improved when adiponectin is increased. The most studied adipokine, leptin, has a dual effect with a positive effect on sperm at physiological levels and a negative one for high seminal concentrations. Many semen parameters and fertility itself are disturbed according to semen adipokine levels, even if it is not the only interfering element. Taken together, adipokines are found in human and animal semen and most of them or their receptors are expressed in male genital tract. Although the pathophysiological role of adipokines in semen is not clearly elucidated, the adipokines could influence sperm functionality and could be potential biomarkers of male fertility.

Metabolic homeostasis influences the reproductive functions, and its alteration leads to disturbances in the reproductive functions of humans as well as animals. For a long time, adipose tissue has been recognised as an endocrine organ but its ability to secrete and release hormones called adipokines is now emerging. Adipokines have been found to play a major role in the regulation of metabolic and reproductive processes at both central and peripheral levels. Leptin was initially the first adipokine that has been described to be the most involved in the metabolism/reproduction interrelation in mammals. In avian species, the role of leptin is still under debate. Recently, three novel adipokines have been discovered: adiponectin (ADIPOQ, ACRP30), visfatin (NAMPT, PBEF), and chemerin (RARRES2, TIG2). However, their mode of action between mammalian and nonmammalian species is different due to the different reproductive and metabolic systems. Herein, we will provide an overview of the structure and function related to metabolic and reproductive mechanisms of the latter three adipokines with emphasis on avian species.

Abstract: To survive in temperate latitudes, species rely on the photoperiod to synchronize their physiological functions, including reproduction, with the predictable changes in the environment. In sheep, exposure to decreasing day length reactivates the hypothalamo-pituitary-gonadal axis, while during increasing day length, animals enter a period of sexual rest. Neural stem cells have been detected in the sheep hypothalamus and hypothalamic neurogenesis was found to respond to the photoperiod. However, the physiological relevance of this seasonal adult neurogenesis is still unexplored. This longitudinal study, therefore aimed to thoroughly characterize photoperiod-stimulated neurogenesis and to investigate whether the hypothalamic adult born-cells were involved in the seasonal timing of reproduction. Results showed that time course of cell proliferation reached a peak in the middle of the period of sexual activity, corresponding to decreasing day length period. This enhancement was suppressed when animals were deprived of seasonal time cues by pinealectomy, suggesting a role of melatonin in the seasonal regulation of cell proliferation. Furthermore, when the mitotic blocker cytosine-b-D-arabinofuranoside was administered centrally, the timing of seasonal reproduction was affected. Overall, our findings link the cyclic increase in hypothalamic neurogenesis to seasonal reproduction and suggest that photoperiod-regulated hypothalamic neurogenesis plays a substantial role in seasonal reproductive physiology.

Abstract: International audience; Filamentous fungi asymptomatically colonize the inner tissues of macroalgae, yet their ecological roles remain largely underexplored. Here, we tested if metabolites produced by fungal endophytes might protect their host against a phylogenetically broad spectrum of protistan pathogens. Accordingly, the cultivable fungal endophytes of four brown algal species were isolated and identified based on LSU and SSU sequencing. The fungal metabolomes were tested for their ability to reduce the infection by protistan pathogens in the algal model Ectocarpus siliculosus. The most active metabolomes effective against the oomycetes Eurychasma dicksonii and Anisolpidium ectocarpii, and the phytomixid Maullinia ectocarpii were further characterized chemically. Several pyrenocines isolated from Phaeosphaeria sp. AN596H efficiently inhibited the infection by all abovementioned pathogens. Strikingly, these compounds also inhibited the infection of nori (Pyropia yezoensis) against its two most devastating oomycete pathogens, Olpidiopsis pyropiae, and Pythium porphyrae. We thus demonstrate that fungal endophytes associated with brown algae produce bioactive metabolites which might confer protection against pathogen infection. These results highlight the potential of metabolites to finely-tune the outcome of molecular interactions between algae, their endophytes, and protistan pathogens. This also provide proof-of-concept toward the applicability of such metabolites in marine aquaculture to control otherwise untreatable diseases.

Abstract: Adipokines are secreted by adipose tissue and could be the link between obesity and infertility. Different studies investigated the involvement of adipokines in reproductive functions but only a few have looked into the male part. This review assesses adipokine functions on male reproductive parameters. Adiponectin seems to have a positive effect on sperm parameters, whereas other adipokines such as resistin or chemerin would have a rather deleterious effect on spermatogenesis. Semen parameters seem to be impacted when resistin and chemerin are increased: indeed, there is a decrease of sperm motility. Sperm morphology is improved when adiponectin is increased. The most studied adipokine, leptin, has a dual effect with a positive effect on sperm at physiological levels and a negative one for high seminal concentrations. Many semen parameters and fertility itself are disturbed according to semen adipokine levels, even if it is not the only interfering element. Taken together, adipokines are found in human and animal semen and most of them or their receptors are expressed in male genital tract. Although the pathophysiological role of adipokines in semen is not clearly elucidated, the adipokines could influence sperm functionality and could be potential biomarkers of male fertility.

Abstract: International audience; In order to follow the composition of parental broilers (Cobb500) during growth from hatching to adulthood (32 weeks of age), we evaluated kinetics of fattening, growth rate, reproduction parameters and body composition of the animals by using non-invasive tools such as medical imaging and blood sample analysing. The use of CT scanner allowed us to monitor the development of body composition on the same animals who are sleeping (fatness, bone, muscle, ovary and testis growth). These analyses were associated with biochemical blood analysis. Parental broilers fed the recommended diet by breeders. The analyses were performed with a CT scanner of bio-medical grade (Siemens As Definition ; 100K Volta and 120 mA / s). Five hundred images were performed every 0.6 mm, 0.45 Pitch and reconstruction filter Safire I26 was used to characterize fat tissues, skeletal and muscle. Image processing by thresholding was used to quantify and monitor the tissues from 4 weeks to 32 weeks of age. The acquired data were used to compare males and females. A significant difference in volume of adipose tissue was observed between male and female. From 20 weeks of age, shortly before the onset of lay, the females had 1.6 fold more adipose tissue than males (P < 0,001). In the female group, the increase in adipose tissue is associated to elevated plasma triglycerides levels (average 8 fold more in comparison to male at the beginning of the laying period. In addition, females, from 16 weeks of age, present a slow bone development in comparison to male (P < 0,001). The ratio bone/bodyweight is 30% lower in female compared to male chicken at 32 weeks of age. A section of the pectoral muscle volume was also analysed but no difference has been noted between both sexes. The correlation between the number of yellow follicles of hens and volume of ovarian adipose tissue determined in vivo was 0.80. In conclusion, the use of CT scanner and ultrasound system has permitted to follow the body composition of an actual parental breed without dissection

Abstract: Energy metabolism (in connection with growth, adipose tissue, etc.) is an important modulator of fertility. In order to improve performance in fertility, we have evaluated the feasibility to analyze the adipose tissue composition in 3 poultry species: chicken, turkey and guinea fowl by using a non-invasive method such as medical imaging (computed tomography, CTScan). In these 3 species, the adipose tissue composition varies between 10 and 15% depending the age. In particular, we carried out kinetics of fattening in the hen to associate it with the feeding, growth, and egg quality. In the experiment, meat-type chickens (Cobbgenetics) were bred with 2 diets conditions (ad libitum and restricted diet) from 3 weeks to 35 weeks of age. During this period, analyzes were performed with a biomedical scanner (Siemens, 100K Volta and 120mA / s). 500 images were taken every 0.6 mm. Image processing by thresholding was used to quantify adipose tissue. We calculated a correlation r² of 0.94 between the weight of the abdominal adipose tissue measured and the weight of the adipose tissue determined throughout the organism in vivo by computed tomography. The correlation between the chicken body weight and the volume of adipose tissue determined in vivo is r²= 0.82. The location and development of adipose tissue revealed significant differences as early as 6 weeks between ad libitum and restricted groups. In the ad libitum group, the increase in volume of adipose tissue is also associated with a high plasma triglyceride concentration and early onset of the laying period. Preliminary measurements of muscle, bone and liver volume have been measured, and allow opening new perspectives on the acquisition of new data that could be extrapolated with the feed and fertility parameters. In conclusion, this work has shown the importance of the CTscan to follow the evolution of the body composition in a non-invasive method, and to associate it with the growth parameters. Similar experiments are underway in turkey and guinea fowl., Le métabolisme énergétique (en lien avec la croissance, la masse du tissu adipeux,...) est un modulateur important de la fertilité. Afin d'améliorer les performances de fertilité, nous avons évalué la faisabilité d’analyser la composition en tissu adipeux de 3 espèces avicoles d’importance autour de la période de stimulation lumineuse : la poule, la dinde et la pintade en utilisant des outils non invasifs tels que l'imagerie médicale (tomodensitométrie, CTScan) qui permettent de localiser le tissu. Chez ces 3 espèces, lacomposition en tissu adipeux varie entre 10 et 15 % en fonction de l’âge. Plus particulièrement, nous avons réalisé une cinétique d'engraissement chez la poule afin de la mettre en lien avec l’alimentation, la croissance, et la qualité des oeufs. Pour cela, des poules de type chair (génétique Cobb) ont été nourries selon 2 régimes (ad libitum; régime restreint) de 3 semaines à 35 semaines d'âge. Sur cette période, des analyses ont été effectuées avec un scanner biomédical (Siemens Comme Définition; 100K Volta et 120 mA / s). 500 images ont été prises tous les 0,6 mm. Un traitement de l'image par seuillage a été utilisé pour quantifier le tissuadipeux. Nous avons calculé une corrélation r² de 0,94 entre le poids du tissu adipeux abdominal mesuré et le poids du tissu adipeux déterminé dans tout l’organisme in vivo par tomodensitométrie. La corrélation entre le poids corporel des poules et le volume de tissu adipeux déterminé in vivo est de r²=0,82. La localisation et le développement du tissu adipeux a révélé des différences significatives dès l’âge de 6 semaines entre les lots ad libitum et restreint. Dans le groupe ad libitum, l'augmentation de volume du tissu adipeux est également associée à une concentration élevée en triglycérides plasmatiques et un début de la période de ponte plus précoce. Des mesures préliminaires de masse musculaire, de volume osseux et du foie ont été réalisées, etpermettent d’ouvrir sur l’acquisition de nouvelles données qui pourraient être extrapolées avec les paramètres de fertilité ou de l’alimentation utilisée. En conclusion, ce travail montre l’importance du CTscan pour suivre l'évolution de la composition corporelle de façon non invasive, et de l’associer aux paramètres de croissance. Des expériences similaires sont en cours chez la dinde et la pintade.

Abstract: Session : Santé animale; The biological activities of water-soluble sulfated polysaccharides of green algae (Ulvans) are explored with the aim of their being used as bioactive molecules for human and animal health. A purified fraction of ulvans was prepared from the green algae Ulva armoricana harvested in the Brittany region (France) and tested for its capacity to stimulate the immune response of the gut using an in vitro system of porcine intestinal epithelial cells (IPEC-1). RT-qPCR and ELISA analysis showed a significant increase in mRNA and protein expression of cytokines such as CCL20, IL8, and TNFalpha. By using human embryonic kidney HEK293 reporter cell lines for pattern recognition receptors, ulvan was found to stimulate mostly TLR4. We also examined this fraction’s effect on different signaling pathways involved in activating cytokine gene expression. Western blot analysis of ulvan-treated HEK293-TLR4 cells showed an increase in Akt and the p65 subunit of the nuclear factor-κB (NFκB) phosphorylation. Inhibition of Akt phosphorylation with specific inhibitor abrogated ulvan enhancement of IL-8 secretion. The overall results showed that ulvan is an immunostimulator compound by itself, and furthermore, it could be used to encapsulate and deliver TLR ligands to relevant immune cells to strongly enhance adjuvanticity and immunity in vaccination strategies.

Abstract: In broiler hens, rapid growth is often associated with a disruption of reproduction. One of the methods used byfarmers to limit the decline of fertility is feed restriction. The aim of this study is to assess the consequences ofthis type of feed intake management and polyunsaturated fatty acid supplementation (FAS) known to increaseadipokines (hormones produced by the adipose tissue) production, on fertility, steroidogenesis and eggs quantity and quality. Hence, we followed 320 hens divided into 4 equal groups fed with a restricted (Rt) or ad libitum (Ad, receiving an amount of feed to reach a 1.7 times heavier weight than the animals of the restricted diet), supplemented or not with FAS (1%). Abstract: Since ancient times, humans have used fungi as food sources ( 1 , 2 ). The edible sexual structures of basidiomycetes and ascomycetes (e.g., truffles), the so-called mushrooms, are produced mostly in wood because many fungi are tree symbionts or decayers of tree tissues. These fruiting bodies represent a rich source of proteins, with low fat content and otherwise nutritionally quite poor. In some soils, they accumulate pollution (heavy metals and radioactivity) and should only be eaten in moderate quantities. Some mushroom species are considered delicacies (e.g., truffles, boletus, morels), but cultivation attempts have been unsuccessful, with a few exceptions (e.g., Morchella rufobrunnea). Only a few saprobic species can be industrially produced, such as Agaricus bisporus ( 3 ), Lentinus edodes (shiitake), and Pleurotus ostreatus, with production having mainly taken place in Asia for thousands of years ( 4 ). Some other fungi, while not really cultured, are inoculated on trees grown in appropriate natural habitats to increase the production of fruiting bodies, such as for shiitake and oyster mushrooms, with, however, sometimes unpredictable success, as is the case for truffles ( 5 ).

Abstract: VISFATIN is a novel adipokine, also known as a Nicotinamide phosphorybosyltransferase (NAMPT) that is able to modulate different processes including lipid and glucose metabolism, oxidative stress, inflammation and insulin resistance. Recent data suggest that it also plays a role in reproductive function in rats, humans and chickens Here we identified VISFATIN in the bovine ovary and investigated the in vitro effects of this hormone on granulosa cell steroidogenesis and proliferation and oocyte maturation. By RT-PCR, immunoblotting and immunohistochemistry, we found VISFATIN in various ovarian cells, including granulosa and theca cells, corpus luteum, and oocytes. In cultured bovine granulosa cells, we showed that IGF1 (10-8M) and VISFATIN (10 and 100 ng/ml) but not FSH (10-8M) increased mRNA expression levels of NAMPT after 48h of stimulation. Moreover, we observed that human recombinant VISFATIN (hVisf, 10 ng/ml, 48h) increased the release of progesterone and estradiol secretion and this was associated with an increase in the protein level of STAR, the HSD3B activity and the phosphorylation levels of IGF1R and MAPK ERK1/2 in the presence or absence of IGF1 (10-8 M). All these effects were abolished when NAMPT was knocked down and when the sirtuin pharmacological inhibitors, CHIC-35 (60 nM) and EX-527 (0.5 M) were pre-incubated in bovine granulosa cells. Thus, in cultured bovine granulosa cells, VISFATIN improves basal and IGF1-induced steroidogenesis and IGF1 receptor signaling through SIRT1.
