Abstract

Senescent cells secrete several molecules, collectively named senescence-associated secretory phenotype (SASP). In the SASP of cells that became senescent following several in vitro chemical and physical stress, we identified the IGFBP-4 protein that can be considered a general stress mediator. This factor appeared to play a key role in senescence-paracrine signaling. We provided evidences showing that genotoxic injury, such as low dose irradiation, may promote an IGFBP-4 release in bloodstream both in mice irradiated with 100 mGy X-ray and in human subjects that received Computer Tomography. Increased level of circulating IGFBP-4 may be responsible of pro-aging effect. We found a significant increase of senescent cells in the lungs, heart, and kidneys of mice that were intraperitoneally injected with IGFBP-4 twice a week for two months. We then analyzed how genotoxic stressors may promote the release of IGFBP-4 and the molecular pathways associated with the induction of senescence by this protein.

5. Hybrid complexes of high and low molecular weight hyaluronan delay in vitro replicative senescence of mesenchymal stromal cells: a pilot study for future therapeutic application

Abstract

Mesenchymal stem cells, a subpopulation of mesenchymal stromal cells (MSCs), are present in the stroma of several tissues. MSC in vitro cultivation for clinical treatments may greatly affect MSC properties. A primary handicap is replicative senescence that impairs MSC functions. Hyaluronan (HA) is present in the extracellular matrix that composes the stem cell niche environment and is under investigation as a key factor for in vitro stem cell growth. We evaluated the effect on MSC cultivation of HA hybrid cooperative complexes (HCC) that are obtained from high (H) and low (L) weight molecules (NAHYCO(TM)). We compared this HCC with H-HA and L-HA. We investigated the effects of these HAs on proliferation, cell cycle, apoptosis, senescence, and differentiation following the addition of the polymer solutions in the culture media at concentrations that did not drastically modify the medium viscosity. Interestingly, 0,16% HCC significantly delayed the senescence compared with the controls. This occurred without alteration of the cell cycle, cytotoxicity, or apoptosis. HCCs also promoted adipogenic and chondrogenic differentiation. Our finding could suggest a potential functional role of HCC above the updated scientific reports of its effects and pave the way to optimization of MSC cultivation for therapeutic application.

La promozione e la valorizzazione della Dieta Mediterranea come strumento di prevenzione primaria delle principali malattie cardiovascolari, metaboliche, oncologiche e degenerative è l’obiettivo di Idimed e questo opuscolo è uno degli strumenti della sua strategia di divulgazione. Idimed, fedele alla sua impostazione sistemica, ha sempre parlato di alimentazione a 360°, integrando i diversi aspetti del fenomeno: culturali, salutistici, agronomici, botanici, gastronomici, psicologici, relazionali e della comunicazione. Far conoscere l’alimento, descrivere la realizzazione della ricetta, fornire indicazioni su quantità, uso dei condimenti, specificità della tradizione e raccomandazioni salutistiche costituisce il fondamento di questo progetto editoriale che integra cultura, gusto e salute. Una pubblicazione per tutti: per le famiglie, per i ristoratori, per gli appassionati, per i visitatori/viaggiatori/turisti. Per sperimentare un viaggio nel gusto e nella tradizione siciliana e ritrovare o riscoprire antichi sapori dimenticati. Abstract

Les procédés rencontrés dans la valorisation des résidus et des huiles lourdes font appel à des équipements dont le dimensionnement optimal exige la connaissance précise des propriétés rhéologiques de la phase hydrocarbonée impliquée et ce, dans un large domaine de température et de pression. En conséquence, le Laboratoire de Thermodynamique du Centre Réacteurs et Processus de l'École Nationale Supérieure des Mines (ENSM) de Paris s'est attaché au développement d'un viscosimètre approprié à ces mesures. Il est basé sur la détermination du couple résistant provoqué par le fluide à étudier, cisaillé entre deux cylindres coaxiaux en rotation relative. Le domaine de viscosité balayé s'étend de 10 à 40 000 cP, les température et pression maximales d'utilisation étant respectivement de 500°C et 30 MPa. La qualité des résultats fournis par cet appareillage a été testée, à des températures et dilution variables, sur quelques coupes lourdes issues des bruts Safaniya et Boscan après désasphaltage ou hydroviscoréduction. On obtient ainsi des valeurs fiables, reproductibles et précises de la viscosité des coupes lourdes hydrocarbonées sur un large domaine de température. Au-delà de 430-440°C toutefois, la pression opératoire de la cellule, fixée par la quantité du gaz admis, augmente brusquement, révélant un craquage thermique important des échantillons et offrant ainsi un moyen d'étude et de simulation des procédés industriels de viscoréduction. Processes used for the upgrading of residues and heavy oils makes use of equipment for which the optimal sizing requires the exact understanding of the rheological properties of the hydrocarbon phase involved over a wide temperature and pressure range. Therefore, the thermodynamics laboratory of the ENSM Center for Reactors and Processes has carried out the development of a viscosimeter suited for such measurements. It is based on determining the resistant torque caused by the fluid to be analyzed, sheared between twocoaxial cylinders with relative rotation. The viscosity range covered goes from 10 to 40,000 cP, with maximum temperatures and pressures of use being respectively 500°C and 30 MPa. The quality of the results obtained with this device was tested at varying temperatures and dilutions with several heavy cuts coming from Safaniya and Boscan crudes after deasphalting or hydrovisbreaking. Reliable, reproducible and accurate values were thus obtained for the viscosity of heavy hydrocarbon cuts over a wide temperature range. Above 430-440°C, however, the operating pressure of the cell, determined by the amount of gas admitted, increases abruptly, revealing extensive thermal cracking of the samples and thus providing a way of studying and simulating industrial visbreaking processes.

Abstract

We report on the first Fermi Large Area Telescope (LAT) measurements of the so-called "extra-galactic" diffuse gamma-ray emission (EGB). This component of the diffuse gamma-ray emission is generally considered to have an isotropic or nearly isotropic distribution on the sky with diverse contributions discussed in the literature. The derivation of the EGB is based on detailed modelling of the bright foreground diffuse Galactic gamma-ray emission (DGE), the detected LAT sources and the solar gamma-ray emission. We find the spectrum of the EGB is consistent with a power law with differential spectral index g = 2.41+/-0.05 and intensity, I(> 100 MeV) = (1.03+/-0.17) 10^-5 cm^-2 s^-1 sr^-1, where the error is systematics dominated. Abstract

We perform a consistent modeling of cosmic ray electrons, positrons and of the radio emission of the Galaxy. For the time we reproduce all relevant data sets between 1 GeV and 1 TeV including the recent AMS-02 positron fraction results. We show that below few GeV cosmic ray and radio data require that electron primary spectrum to be drastically suppressed and the propagated spectrum be dominated by secondary particles. Above 10 GeV an electron + positron extra-component with a hard spectrum is required. The positron spectrum measured below few GeV is consistently reproduced only within low reacceleration models. We also constrain the scale-height of the cosmic-ray distribution showing that a thin halo ($z_t \lsim 2 \kpc$) is excluded., 4 pages, 6 figs. Abstract

We present the results of our analysis of cosmic-ray electrons using about 8 million electron candidates detected in the first 12 months on-orbit by the Fermi Large Area Telescope. This work extends our previously-published cosmic-ray electron spectrum down to 7 GeV, giving a spectral range of approximately 2.5 decades up to 1 TeV. We describe in detail the analysis and its validation using beam-test and on-orbit data. In addition, we describe the spectrum measured via a subset of events selected for the best energy resolution as a cross-check on the measurement using the full event sample. Our electron spectrum can be described with a power law $\propto {\rm E}^{-3.08 \pm 0.05}$ with no prominent spectral features within systematic uncertainties. Within the limits of our uncertainties, we can accommodate a slight spectral hardening at around 100 GeV and a slight softening above 500 GeV., 20 pages, 23 figures, 2 tables, published in Physical Review D 82, 092004 (2010) - contact authors: C. Abstract

Designed as a high-sensitivity gamma-ray observatory, the Fermi Large Area Telescope is also an electron detector with a large acceptance exceeding 2 m2 sr at 300 GeV. Building on the gamma-ray analysis, we have developed an efficient electron detection strategy which provides sufficient background rejection for measurement of the steeply falling electron spectrum up to 1 TeV. Our high precision data show that the electron spectrum falls with energy as E-3.0 and does not exhibit prominent spectral features. Interpretations in terms of a conventional diffusive model as well as a potential local extra component are briefly discussed.

Abstract

We describe histochemical techniques for detecting DNA within the osteocytic lacunae of ancient bones. The bones examined were fragments of femurs from two human individuals found in the Pompeian C. I. Polybius house and fragments of metacarpals from two horses (Equus sp.) found in the Pompeian "Casti Amanti" house. Both buildings were buried by the 79 A. D. Vesuvius eruption. Fragments of femurs from a modern horse, a modern swine and a modern amphibian also were studied as controls. Some bone sections were stained with two different DNA-specific fluorochromes, 4'-'6-diamidino-2-phenylindole (DAPI) and chromomycin A3 (CMA), while others were stained by the Feulgen reaction. All of the techniques gave a positive reaction within the osteocytic lacunae. Histological analysis of the undecalcified, ground and unstained sections agreed well with results of bone sections stained with either the fluorochromes or the Feulgen reaction. Bones showing good histology also were positive by our DNA-specific stain. Histochemical and histological analyses correlated well with the success of DNA extraction and amplification. Using conventional DNA-specific histochemical techniques in conjunction with histological analysis can be useful in the study of DNA extracted from ancient bone remains while reducing both the amount of time and cost.

Abstract

To investigate whether unusual allele segregation might explain the dominant negative effect of the expanded allele for myotonic dystrophy on myotonin protein kinase mRNA metabolism, which is suggested to cause the disease, we determined the number of CTG repeats at the DM locus in the nonaffected alleles of 64 DM (dystrophia myotonia) patients. The relative distribution was then compared with the distributions obtained from alleles of the normal parents and normal siblings of DM patients. Comparison was also made with the allele distribution of normal subjects from the same geographic area. It appears that the CTG repeat number of the nonaffected allele in DM patients is not critical for the expression of the disease.

Abstract

Men’s partner-sexual objectification has been linked to increased self-objectification and diminished well-being in women. Some recent findings have also demonstrated that men’s partner-sexual objectification is related to increased violence in the relationship. However, mechanisms driving this association remain unexplored. In the present research, we collected data on women and men involved in heterosexual romantic relationships and investigated the associations between men’s partner-sexual objectification, women’s self-objectification, and both partners’ attitudes toward dating violence. Study 1 ( N = 171 heterosexual couples) provided first evidence for the link between men’s partner-sexual objectification and their attitudes toward dating violence. Furthermore, men’s attitudes toward dating violence mediated the relationship between sexual objectification of their partners and women’s attitudes toward dating violence. These results were replicated in Study 2 ( N = 235 heterosexual couples). Findings of this study also revealed that, along with men’s attitudes toward dating violence, women’s self-objectification acted as a mediating mechanism linking experiences of being sexually objectified by the romantic partner and attitudes toward dating violence in women. Implications of our findings for the issue of dating violence are discussed.

Abstract

Senescent cells secrete inflammatory cytokines, proteases, and other factors, which are indicated as senescence-associated secretory phenotype (SASP). There are contrasting studies on the role of the SASP in cancer. Studies suggested that cancer cells may misuse the senescent secretome for their growth. Other investigations evidenced that the SASP may induce cancer growth arrest, senescence, or apoptosis. These conflicting data can be reconciled considering that cancer cells can coax senescent cells to secrete factors for their survival, thus abrogating the SASP’s anti-cancer effect. Cancer stage may also have an impact on the capacity of the SASP to block tumor proliferation and promote senescence. Indeed, senescence is associated with a permanent cell cycle arrest, which needs functional cell cycle checkpoints. We evaluated the SASP effect on the in vitro biological properties of PNT2 and PC3 cells, which are immortalized prostate cells and metastatic prostatic cancer cells, respectively. We evidenced that SASPs, coming either from mesenchymal stromal cells treated with H202 or with low X-ray doses, induced senescence of immortalized cells but not of cancer cells. Hence, the SASP released by acute senescent cells should be considered as an effective weapon against pre-tumorigenesis events rather than an anti-cancer mechanism acting on malignant cells.

Abstract

The cells present in the stromal compartment of many tissues are a heterogeneous population containing stem cells, progenitor cells, fibroblasts, and other stromal cells. A SSEA3(+) cell subpopulation isolated from human stromal compartments showed stem cell properties. These cells, known as multilineage-differentiating stress-enduring (MUSE) cells, are capable of resisting stress and possess an excellent ability to repair DNA damage. We isolated MUSE cells from different mouse stromal compartments, such as those present in bone marrow, subcutaneous white adipose tissue, and ear connective tissue. These cells showed overlapping in vitro biological properties. The mouse MUSE cells were positive for stemness markers such as SOX2, OCT3/4, and NANOG. They also expressed TERT, the catalytic telomerase subunit. The mouse MUSE cells showed spontaneous commitment to differentiation in meso/ecto/endodermal derivatives. The demonstration that multilineage stem cells can be isolated from an animal model, such as the mouse, could offer a valid alternative to the use of other stem cells for disease studies and envisage of cellular therapies.

Abstract

During their life span, cells have two possible states: a non-cycling, quiescent state (G0) and a cycling, activated state. Cells may enter a reversible G0 state of quiescence or, alternatively, they may undergo an irreversible G0 state. The latter may be a physiological differentiation or, following a stress event, a senescent status. Discrimination among the several G0 states represents a significant investigation, since quiescence, differentiation, and senescence are progressive phenomena with intermediate transitional stages. We used the expression of Ki67, RPS6, and beta-galactosidase to identify healthy cells that progressively enter and leave quiescence through G0-entry, G0 and G0-alert states. We then evaluated how cells may enter senescence following a genotoxic stressful event. We identified an initial stress stage with the expression of beta-galactosidase and Ki67 proliferation marker. Cells may recover from stress events or become senescent passing through early and late senescence states. Discrimination between quiescence and senescence was based on the expression of RPS6, a marker of active protein synthesis that is present in senescent cells but absent in quiescent cells. Even taking into account that fixed G0 states do not exist, our molecular algorithm may represent a method for identifying turning points of G0 transitional states that continuously change.

Muse cells are adult stem cells that are present in the stroma of several organs and possess an enduring capacity to cope with endogenous and exogenous genotoxic stress. In cell therapy, the peculiar biological properties of Muse cells render them a possible natural alternative to mesenchymal stromal cells (MSCs) or to in vitro-generated pluripotent stem cells (iPSCs). Indeed, some studies have proved that Muse cells can survive in adverse microenvironments, such as those present in damaged/injured tissues. We performed an evaluation of Muse cells' proteome under basic conditions and followed oxidative stress treatment in order to identify ontologies, pathways, and networks that can be related to their enduring stress capacity. We executed the same analysis on iPSCs and MSCs, as a comparison. The Muse cells are enriched in several ontologies and pathways, such as endosomal vacuolar trafficking related to stress response, ubiquitin and proteasome degradation, and reactive oxygen scavenging. In Muse cells, the protein-protein interacting network has two key nodes with a high connectivity degree and betweenness: NFKB and CRKL. The protein NFKB is an almost-ubiquitous transcription factor related to many biological processes and can also have a role in protecting cells from apoptosis during exposure to a variety of stressors. CRKL is an adaptor protein and constitutes an integral part of the stress-activated protein kinase (SAPK) pathway. The identified pathways and networks are all involved in the quality control of cell components and may explain the stress resistance of Muse cells. Abstract

This work was supported by The Scientific and Technological Research Council of Turkey (TUBITAK) (Project Number: 117S216) to S.O. Senotherapeutics are new drugs that can modulate senescence phenomena within tissues and reduce the onset of age-related pathologies. Senotherapeutics are divided into senolytics and senomorphics. The senolytics selectively kill senescent cells, while the senomorphics delay or block the onset of senescence. Metformin has been used to treat diabetes for several decades. Recently, it has been proposed that metformin may have anti-aging properties as it prevents DNA damage and inflammation. We evaluated the senomorphic effect of 6 weeks of therapeutic metformin treatment on the biology of human adipose mesenchymal stromal cells (MSCs). The study was combined with a proteome analysis of changes occurring in MSCs' intracellular and secretome protein composition in order to identify molecular pathways associated with the observed biological phenomena. The metformin reduced the replicative senescence and cell death phenomena associated with prolonged in vitro cultivation. The continuous metformin supplementation delayed and/or reduced the impairment of MSC functions as evidenced by the presence of three specific pathways in metformin-treated samples: 1) the alpha-adrenergic signaling, which contributes to regulation of MSCs physiological secretory activity, 2) the signaling pathway associated with MSCs detoxification activity, and 3) the aspartate degradation pathway for optimal energy production. The senomorphic function of metformin seemed related to its reactive oxygen species (ROS) scavenging activity. In metformin-treated samples, the CEBPA, TP53 and USF1 transcription factors appeared to be involved in the regulation of several factors (SOD1, SOD2, CAT, GLRX, GSTP1) blocking ROS.
