Vulvovaginal candidiasis is a common mucosal infection affecting a large proportion of women with some of them affected by recurrent often intractable forms of the disease. Thus, there is an increasing interest in understanding the pathogenesis of this disease. The aim of our work was to characterize, in animal models of vaginal candidiasis, the components of the host-fungus interaction at the mucosal level. The evidence of an immune response in the vaginal compartment was very encouraging to identify the proper targets for new strategies for vaccination or immunotherapy of vaginal candidiasis. Aspartyl-proteinase (Sap2), which is an important immunodominant antigens and virulence factors of C.albicans acting in mucosal infections, was assembled with virosomes and a vaccine PEV7 was obtained. The results obtained in the mouse model and in the clinical trial conducted by Pevion on women have evidenced that the vaccine PEV7, intravaginally administered, has an encouraging therapeutic potential for the treatment of recurrent vulvovaginal candidiasis. This opens the way to a modality for anti-Candida protection at mucosal level. © The Author(s) 2017. © 2017 IOP Publishing Ltd and Sissa Medialab. We present the temperature and polarization angular power spectra measured by the Atacama Cosmology Telescope Polarimeter (ACTPol). We analyze night-time data collected during 2013-14 using two detector arrays at 149 GHz, from 548 deg2of sky on the celestial equator. We use these spectra, and the spectra measured with the MBAC camera on ACT from 2008-10, in combination with planck and wmap data to estimate cosmological parameters from the temperature, polarization, and temperature-polarization cross-correlations. We find the new ACTPol data to be consistent with the ΛCDM model. The ACTPol temperature-polarization cross-spectrum now provides stronger constraints on multiple parameters than the ACTPol temperature spectrum, including the baryon density, the acoustic peak angular scale, and the derived Hubble constant. The new ACTPol data provide information on damping tail parameters. The joint uncertainty on the number of neutrino species and the primordial helium fraction is reduced by 20% when adding ACTPol to Planck temperature data alone.

A large sub-mm survey with Herschel will enable many exciting science opportunities, especially in an era of wide-field optical and radio surveys and high resolution cosmic microwave background experiments. The Herschel-SPIRE Legacy Survey (HSLS), will lead to imaging data over 4000 sq. degrees at 250, 350, and 500 micron. Major Goals of HSLS are: (a) produce a catalog of 2.5 to 3 million galaxies down to 26, 27 and 33 mJy (50% completeness; 5 sigma confusion noise) at 250, 350 and 500 micron, respectively, in the southern hemisphere (3000 sq. degrees) and in an equatorial strip (1000 sq. degrees), areas which have extensive multi-wavelength coverage and are easily accessible from ALMA. Two thirds of the of the sources are expected to be at z > 1, one third at z > 2 and about a 1000 at z > 5. (b) Remove point source confusion in secondary anisotropy studies with Planck and ground-based CMB data. (c) Find at least 1200 strongly lensed bright sub-mm sources leading to a 2% test of general relativity. (d) Identify 200 proto-cluster regions at z of 2 and perform an unbiased study of the environmental dependence of star formation. (e) Perform an unbiased survey for star formation and dust at high Galactic latitude and make a census of debris disks and dust around AGB stars and white dwarfs.

The expression of the Candida albicans complement-binding C3d protein (MP60) was investigated both in vitro and in vivo by immunogold labelling and electron microscopy. In vivo expression was determined in a rat vaginitis model. Reactivity of in vitro-grown cells to an anti-MP60 rabbit serum was associated with both cytoplasmic and cell wall sites. Immunostaining in the cell wall of both yeast and hyphae was most concentrated in the inner, electron-lucid layer. Immunogold stained preparations of C. albicans from vaginal smears of infected animals also showed intense localization of the MP60 in the inner cell wall, plasma membrane. However, immunogold label was also intense at the cell surface in these samples, mostly in the area of close adherence with the keratinocytes of the vaginal epithelia. These observations indicate that MP60 is expressed both in vitro and in vivo, but to a different degree in the different cell wall layers.

We present a method for selecting z > 4 dusty, star-forming galaxies (DSFGs) using Herschel/Spectral and Photometric Imaging Receiver 250/350/500 μm flux densities to search for red sources. We apply this method to 21 deg2 of data from the HerMES survey to produce a catalog of 38 high-z candidates. Follow-up of the first five of these sources confirms that this method is efficient at selecting high-z DSFGs, with 4/5 at z = 4.3-6.3 (and the remaining source at z = 3.4), and that they are some of the most luminous dusty sources known. Comparison with previous DSFG samples, mostly selected at longer wavelengths (e.g., 850 μm) and in single-band surveys, shows that our method is much more efficient at selecting high-z DSFGs, in the sense that a much larger fraction are at z > 3. Correcting for the selection completeness and purity, we find that the number of bright (S 500 μm ≥ 30 mJy), red Herschel sources is 3.3 ± 0.8 deg-2. This is much higher than the number predicted by current models, suggesting that the DSFG population extends to higher redshifts than previously believed. If the shape of the luminosity function for high-z DSFGs is similar to that at z ∼ 2, rest-frame UV based studies may be missing a significant component of the star formation density at z = 4-6, even after correction for extinction. © 2014. Detection and ultrastructural localization of aspartyl proteinase (Sap) in Candida albicans experimentally infecting rat vagina were studied. Two Sap-positive (Sap+) and one Sap-negative (Sap-) strains of the fungus, endowed with high and low experimental vaginopathic potential, respectively, were used. Both Sap+ strains produced consistent Sap levels in the rat vagina, while the Sap- strain did not produce any measurable Sap. Electron microscopy of thin sections of chemically-fixed vaginal scrapings showed clear evidence of hyphae of proteolitic strains of C. albicans invading the keratinized epithelial cell layer of the vagina. The fungal cells exhibited a pronounced fibrillar layer on the cell wall with a marked intermixing of fungal and vaginal materials especially pronunced at the hyphal tip. Post-embedding immunogold techniques with the use of anti-Sap polyclonal and the specifically generated monoclonal antibody GF1 showed that Sap was essentially localized in the cell wall of C. albicans early during infection, in a cytological pattern mirroring Sap localization in C. albicans cells grown in Sap-inductive media in vitro. In summary, the data offer a new biochemical and ultrastructural evidence that Sap is actively secreted during experimental rat vaginitis by C. albicans. Cell wall localization of Sap is probably inherent to this active secretion process.

We present Submillimeter Array (SMA) high-spatial resolution imaging and Gemini-South and Multiple Mirror Telescope optical spectroscopy of strong lens candidates discovered in the two widest extragalactic surveys conducted by the Herschel Space Observatory: the Herschel-Astrophysical Terahertz Large Area Survey (H-ATLAS) and the Herschel Multi-tiered Extragalactic Survey (HerMES). From a sample of 30 Herschel sources with S 500 > 100 mJy, 21 are strongly lensed (i.e., multiply imaged), 4 are moderately lensed (i.e., singly imaged), and the remainder require additional data to determine their lensing status. We apply a visibility-plane lens modeling technique to the SMA data to recover information about the masses of the lenses as well as the intrinsic (i.e., unlensed) sizes (r half) and far-infrared luminosities (L FIR) of the lensed submillimeter galaxies (SMGs). The sample of lenses comprises primarily isolated massive galaxies, but includes some groups and clusters as well. Several of the lenses are located at z lens > 0.7, a redshift regime that is inaccessible to lens searches based on Sloan Digital Sky Survey spectroscopy. The lensed SMGs are amplified by factors that are significantly below statistical model predictions given the 500 μm flux densities of our sample. We speculate that this may reflect a deficiency in our understanding of the intrinsic sizes and luminosities of the brightest SMGs. The lensed SMGs span nearly one decade in L FIR (median L FIR = 7.9 × 10 12 L ) and two decades in FIR luminosity surface density (median ΣFIR = 6.0 × 1011 L kpc-2). The strong lenses in this sample and others identified via (sub-)mm surveys will provide a wealth of information regarding the astrophysics of galaxy formation and evolution over a wide range in redshift. © 2013. Modern cosmology has sharpened questions posed for millennia about the origin of our cosmic habitat. The age-old questions have been transformed into two pressing issues primed for attack in the coming decade: How did the Universe begin? and What physical laws govern the Universe at the highest energies? The clearest window onto these questions is the pattern of polarization in the Cosmic Microwave Background (CMB), which is uniquely sensitive to primordial gravity waves. A detection of the special pattern produced by gravity waves would be not only an unprecedented discovery, but also a direct probe of physics at the earliest observable instants of our Universe. We use galaxy positions and the continuity equation to obtain a reconstruction of the line-of-sight velocity field. We stack the microwave temperature at the location of each halo, weighted by the corresponding reconstructed velocity. We vary the size of the aperture photometry filter used, thus probing the free electron profile of these halos from within the virial radius out to three virial radii, on the scales relevant for investigating the missing baryons problem. The resulting best fit kSZ model is preferred over the no-kSZ hypothesis at 3.3 and 2.9σ for two independent velocity reconstruction methods, using 25,537 galaxies over 660 square degrees. The data suggest that the baryon profile is shallower than the dark matter in the inner regions of the halos probed here, potentially due to energy injection from active galactic nucleus or supernovae. Thus, by constraining the gas profile on a wide range of scales, this technique will be useful for understanding the role of feedback in galaxy groups and clusters. The effect of foregrounds that are uncorrelated with the galaxy velocities is expected to be well below our signal, and residual thermal Sunyaev-Zel'dovich contamination is controlled by masking the most massive clusters. Finally, we discuss the systematics involved in converting our measurement of the kSZ amplitude into the mean free electron fraction of the halos in our sample.

Advanced ACTPol is a polarization-sensitive upgrade for the 6 m aperture Atacama Cosmology Telescope (ACT), adding new frequencies and increasing sensitivity over the previous ACTPol receiver. In 2016, Advanced ACTPol will begin to map approximately half the sky in five frequency bands (28-230 GHz). Its maps of primary and secondary cosmic microwave background (CMB) anisotropies -- imaged in intensity and polarization at few arcminute-scale resolution -- will enable precision cosmological constraints and also a wide array of cross-correlation science that probes the expansion history of the universe and the growth of structure via gravitational collapse. To accomplish these scientific goals, the Advanced ACTPol receiver will be a significant upgrade to the ACTPol receiver, including four new multichroic arrays of cryogenic, feedhorn-coupled AlMn transition edge sensor (TES) polarimeters (fabricated on 150 mm diameter wafers); a system of continuously rotating meta-material silicon half-wave plates; and a new multiplexing readout architecture which uses superconducting quantum interference devices (SQUIDs) and time division to achieve a 64-row multiplexing factor. In recent years there have been significant improvements in the sensitivity and the angular resolution of the instruments dedicated to the observation of the Cosmic Microwave Background (CMB). ACTPol is the first polarization receiver for the Atacama Cosmology Telescope (ACT) and is observing the CMB sky with arcmin resolution over about 2000 sq. deg. Its upgrade, Advanced ACTPol (AdvACT), will observe the CMB in five frequency bands and over a larger area of the sky. We describe the optimization and implementation of the ACTPol and AdvACT surveys. The selection of the observed fields is driven mainly by the science goals, that is, small angular scale CMB measurements, B-mode measurements and cross-correlation studies. For the ACTPol survey we have observed patches of the southern galactic sky with low galactic foreground emissions which were also chosen to maximize the overlap with several galaxy surveys to allow unique cross-correlation studies. A wider field in the northern galactic cap ensured significant additional overlap with the BOSS spectroscopic survey. The exact shapes and footprints of the fields were optimized to achieve uniform coverage and to obtain cross-linked maps by observing the fields with different scan directions. We have maximized the efficiency of the survey by implementing a close to 24 hour observing strategy, switching between daytime and nighttime observing plans and minimizing the telescope idle time. We describe the challenges represented by the survey optimization for the significantly wider area observed by AdvACT, which will observe roughly half of the low-foreground sky. Two recombinant strains of Streptococcus gordonii, secreting or displaying a microbicidal single-chain antibody (H6), and stably colonizing rat vagina, were used to treat an experimental vaginitis caused by Candida albicans. A post-challenge intravaginal delivery of the H6-secreting strain was as efficacious as fluconazole in rapidly abating the fungal burden. Three weeks after challenge, 75% and 37.5% of the rats treated with the H6-secreting or displaying bacteria, respectively, were cured of the infection, which persisted in 100% of the animals treated with a S. gordonii strain expressing an irrelevant single-chain antibody. Thus, a human commensal bacterium can be suitably engineered to locally release a therapeutic antibody fragment.

A prospective, multicenter, open study of fluconazole prophylaxis was performed in AIDS patients to evaluate the efficacy and toxicity of the drug in preventing relapses of esophageal candidiasis. To this aim, 99 AIDS patients who presented a first episode of clinically and microbiologically confirmed esophageal candidiasis were enrolled in eleven clinical centers scattered throughout the Italian territory. After resolution of this initial esophagitis, all subjects were given fluconazole, 100 mg/die, and followed up for a 6 month period. Only 7 out of the 99 patients enrolled had a relapse of Candida esophagitis, during a mean follow-up period of 138.5 days. All relapsing patients had CD4+ cell number < 100/microliters at baseline. Mild side effects were reported in only eight patients. However, 14 of the 27 subjects from whom serial serum samples were available became (12) or remained (2) antigenemic during fluconazole prophylaxis, independently from relapse, suggesting the persistence of tissue-invasive, proliferating Candida cells. Overall, the data of this study suggest a beneficial effect of prophylactic maintenance therapy with fluconazole against Candida esophagitis, particularly in the population with > 100 CD4+/microliters. However, the data on Candida antigenemia in these patients invite the consideration of a relative inefficiency of the drug to eradicate the microrganism from the esophageal tissue

Background: The IgA1 protease of Streptococcus pneumoniae is a proteolytic enzyme that specifically cleaves the hinge regions of human IgA1, which dominates most mucosal surfaces and is the major IgA isotype in serum. This protease is expressed in all of the known pneumococcal strains and plays a major role in pathogen's resistance to the host immune response. The present work was focused at identifying the immunodominant regions of pneumococcal IgA1 protease recognized by the human antibody response. Results: An antigenic sequence corresponding to amino acids 420–457 (epiA) of the iga gene product was identified by screening a pneumococcal phage display library with patients' sera. The epiA peptide is conserved in all pneumococci and in two out of three S. mitis strains, while it is not present in other oral streptococci so far sequenced. This epitope was specifically recognized by antibodies present in sera from 90% of healthy adults, thus representing an important target of the humoral response to S. pneumoniae and S. mitis infection. Moreover, sera from 68% of children less than 4 years old reacted with the epiA peptide, indicating that the human immune response against streptococcal antigens occurs during childhood. Conclusion: The broad and specific recognition of the epiA polypeptide by human sera demonstrate that the pneumococcal IgA1 protease contains an immunodominant B-cell epitope. The use of phage display libraries to identify microbe or disease-specific antigens recognized by human sera is a valuable approach to epitope discovery.
