Fifty per cent of neurons in the rat enteric nervous system are lost between 12 and 18 months of age in ad libitum (AL) fed rats. Caloric restriction (CR) protects almost entirely against this neuron loss. Using the ROS-sensitive dyes, dihydrorhodamine (DHR) and 2-[6-(4'-hydroxy)phenoxy-3H-xanthen-3-on-9-yl]benzoic acid (HPF) in vitro, we show that the onset of cell death is linked with elevated intraneuronal levels of reactive oxygen species (ROS). Treatment with the neurotrophic factors NT3 and GDNF enhances neuronal antioxidant defence in CR rats at 12-15 months and 24 months but not in adult or aged AL-fed animals. To examine the link between elevated ROS and neuronal cell death, we assessed apoptotic cell death following in vitro treatment with the redox-cycling drug, menadione. Menadione fails to increase apoptosis in 6-month neurons. However, in 12-15mAL fed rats, when age-related cell death begins, menadione induces a 7- to 15-fold increase in the proportion of apoptotic neurons. CR protects age-matched neurons against ROS-induced apoptosis. Treatment with neurotrophic factors, in particular GDNF, rescues neurons from menadione-induced cell death, but only in 12-15mCR animals. We hypothesize that CR enhances antioxidant defence through neurotrophic factor signalling, thereby reducing age-related increases in neuronal ROS levels and in ROS-induced cell death.

As from 1997, the Journal of Anatomy will be published in 8 issues per year instead of the current 6. The purpose is to reduce the time between acceptance and publication. Otherwise there will be no change in editorial policy. The Journal will continue to publish high quality papers on human and comparative anatomy and cell biology, both experimental and descriptive. Articles on experimental pathology are also acceptable if they contribute to the understanding of normal structure and function. Review articles, if topical and authoritative, are welcome. Short reports are published: these should not exceed 5 pages of printed text. Reports on isolated anatomical variations, if of sufficient interest and relevance, are now included in the Correspondence section, to which letters on matters arising out of previously published papers or on questions of general anatomical interest can be submitted.

Intracellular injection of a biotinylated probe in fixed superior cervical ganglia followed by confocal microscopy was used to investigate the effects of age on the dendritic arborisation of sympathetic neurons in rats aged 6 wk (young adult), 7 months (fully grown adult) and 24 months (aged). In accordance with other studies considerable dendritic growth was observed during postnatal development. However, in old age dendritic growth did not continue, and significant atrophy was observed. Quantitation of neuronal morphology showed significant reductions in soma size, total dendritic length, number of branch points and total area of dendritic arborisation in old age. Unexpectedly, significant reductions in the numbers of primary dendrites were observed in maturity and in old age. Concomitant with this atrophy there was an increase in age-related morphological abnormalities. The similarities between the atrophy and dendritic abnormalities shown by our aged neurons and those seen in other studies of young adult sympathetic neurons following axotomy or trophic factor deprivation are discussed.

The present study shows that cerebral blood vessels of the gerbil resemble other species in being richly supplied by NA-, 5-HT- and NPY-containing nerves. By contrast, the peptidergic nerves containing SP, CGRP and VIP are less dense. The results of superior cervical ganglionectomy show that the SCG is the principal source of NA-, NPY- and 5-HT containing cerebrovascular nerve fibres in the gerbil. The substances seem likely to coexist in some perivascular nerve fibres.

By combining molecular modelling and electrochemistry we envision the creation of modified electrodes tailored for a more sensitive and selective detection of a single analyte. In this study we report on a graphite screen printed electrode modified with electropolymerized o-phenylenediamine, selected by rational design, which promotes the detection of nafcillin (NAF), an antibiotic. Parameters such as monomer concentration, pH and number of electropolymerization cycles were optimized to obtain the highest current signal for the target upon amperometric detection. NAF identification was based on the redox process at +1.1 V (vs pseudo Ag), ascribed to the oxidation of the C-7 side chain. With the optimized modification protocol, a two-fold increase in nafcillin signal could be obtained: the calibration plot in 0.1 M Britton-Robinson buffer pH 4 showed a limit of detection of 80 nM with improved sensitivity and reproducibility (RSD

A biomimetic sensor for cefquinome (CFQ) was designed at multi-walled carbon nanotubes modified graphite screen-printed electrodes (MWCNTs-G-SPEs) as a proof-of-concept for the creation of a sensors array for beta-lactam antibiotics detection in milk. The sensitive and selective detection of antibiotic residues in food and environment is a fundamental step in the elaboration of prevention strategies to fight the insurgence of antimicrobial resistance (AMR) as recommended by authorities around the world (EU, WHO, FDA). The detection strategy is based on the characteristic electrochemical fingerprint of the target antibiotic cefquinome. A conducive electropolymerized molecularly imprinted polymer (MIP) coupled with MWCNTs was found to be the optimal electrode modifier, able to provide an increased selectivity and sensitivity for CFQ detection. The design of CFQ-MIP was facilitated by the rational selection of the monomer, 4-aminobenzoic acid (4-ABA). The electropolymerization process of 4-ABA have not been fully elucidated yet; for this reason a thorough study and optimization of electropolymerization conditions was performed to obtain a conducive and stable poly(4-ABA) film. The modified electrodes were characterized by electrochemical impedance spectroscopy (EIS), scanning electron microscopy (SEM) and cyclic voltammetry (CV). CFQ-MIP were synthesized at MWCNT-G-SPEs by electropolyrnerization in pH approximate to 1 (0.1 M sulphuric acid) with a monomer:template ratio of 5:1. Two different analytical protocols were tested (single and double step detection) to minimize unspecific adsorptions and improve the sensitivity.
