Core-fucosylation is an essential biological modification by which a fucose is transferred from GDP-β-L-fucose to the innermost N-acetylglucosamine residue of N-linked glycans. A single human enzyme α1,6-fucosyltransferase (FUT8) is the only enzyme responsible for this modification via the addition of an α-1,6-linked fucose to N-glycans. To date, the details of substrate recognition and catalysis by FUT8 remain unknown. Here, we report the crystal structure of FUT8 complexed with GDP and a biantennary complex N-glycan (G0), which provides insight into both substrate recognition and catalysis. FUT8 follows an SN2 mechanism and deploys a series of loops and an α-helix which all contribute in forming the binding site. An exosite, formed by one of these loops and an SH3 domain, is responsible for the recognition of branched sugars, making contacts specifically to the α1,3 arm GlcNAc, a feature required for catalysis. La propuesta de innovación educativa Flipped laboratorio clínico y biomédico aplica la metodología de aula invertida a las prácticas de laboratorio. Su aplicación ha supuesto un cambio en la manera de enseñar del docente, obteniendo mejoras en el aprendizaje de los alumnos, permitiendo un mayor aprovechamiento del tiempo en las sesiones experimentales y aumentando el control del grupo. En el desarrollo de la propuesta se describe el uso de videos explicativos de corta duración y la introducción de preguntas en los mismos para el trabajo más profundo de los contenidos teóricos relacionados con la práctica de laboratorio que se va a realizar. De este modo, se ha invertido la forma tradicional de entender la clase, transfiriendo los conceptos ligados a la explicación de contenidos al entorno fuera del aula. El trabajo previo realizado por parte de los alumnos les confiere capacidad de reflexión y favorece el aprendizaje autónomo del alumno. Finalmente, su puesta en práctica ha servido para afrontar nuevos retos de la profesión docente y necesidades de la sociedad actual derivadas de los protocolos COVID. Ha supuesto un análisis del contexto que rodea y condiciona el desempeño docente, así como una contribución en las actividades desarrollas en el centro, a través de la introducción de las TAC en la preparación previa de las prácticas de laboratorio y de la adaptación de la docencia a la situación de semipresencialidad.

Se pretende dibujar, con un enfoque proteómico, un mapa general de las alteraciones metabólicas producidas por ambos estreses nutricionales en las plantas. El estudio de estos procesos permitirá incrementar el conocimiento existente sobre la homeostasis de estos metales y así asentar las bases para mejorar la calidad y productividad de los cultivos, elaborar posibles estrategias de fitoremediación y desarrollar fertilizantes eficientes y respetuosos con el medio ambiente., Se plantean los siguientes objetivos específicos: 1. Describir la categorización funcional de los perfiles proteicos de los tres fluidos principales en plantas, fluido apoplástico y las savias de xilema y floema, considerando los estudios proteómicos recientemente publicados. 2. Caracterizar, usando técnicas de 2-DE, los perfiles proteicos del fluido apoplástico de remolacha (Beta vulgaris), tanto en plantas control como afectadas por deficiencia de Fe y, en las mismas plantas, tras el suministro de Fe a las raíces. 3. Caracterizar, mediante técnicas de “shotgun proteomics”, los cambios inducidos por las deficiencias de Fe y Mn en el perfil proteico de la savia de xilema de plantas de tomate (Solanum lycopersicum). 4. The phloem sap, xylem sap and apoplastic fluid play key roles in long and short distance transport of signals and nutrients, and act as a barrier against local and systemic pathogen infection. Among other components, these plant fluids contain proteins which are likely to be important players in their functionalities. However, detailed information about their proteomes is only starting to arise due to the difficulties inherent to the collection methods. This review compiles the proteomic information available to date in these three plant fluids, and compares the proteomes obtained in different plant species in order to shed light into conserved functions in each plant fluid. Inter-species comparisons indicate that all these fluids contain the protein machinery for self-maintenance and defense, including proteins related to cell wall metabolism, pathogen defense, proteolysis, and redox response. These analyses also revealed that proteins may play more relevant roles in signaling in the phloem sap and apoplastic fluid than in the xylem sap. A comparison of the proteomes of the three fluids indicates that although functional categories are somewhat similar, proteins involved are likely to be fluid-specific, except for a small group of proteins present in the three fluids, which may have a universal role, especially in cell wall maintenance and defense. 12 Pags.- 2 Tabls.- 5 Figs.- Supp. Data., Iron deficiency is a yield-limiting factor with major implications for crop production, especially in soils with high CaCO3. Because stems are essential for the delivery of nutrients to the shoots, the aim of this work was to study the effects of Fe deficiency on the stem proteome of Medicago truncatula. Two-dimensional electrophoresis separation of stem protein extracts resolved 276 consistent spots in the whole experiment. Iron deficiency in absence or presence of CaCO3 caused significant changes in relative abundance in 10 and 31 spots, respectively, and 80% of them were identified by mass spectrometry. Overall results indicate that Fe deficiency by itself has a mild effect on the stem proteome, whereas Fe deficiency in the presence of CaCO3 has a stronger impact and causes changes in a larger number of proteins, including increases in stress and protein metabolism related proteins not observed in the absence of CaCO3. Both treatments resulted in increases in cell wall related proteins, which were more intense in the presence of CaCO3. The increases induced by Fe-deficiency in the lignin per protein ratio and changes in the lignin monomer composition, assessed by pyrolysis-gas chromatography–mass spectrometry and microscopy, respectively, further support the existence of cell wall alterations. Biological significance: In spite of being essential for the delivery of nutrients to the shoots, our knowledge of stem responses to nutrient deficiencies is very limited. The present work applies 2-DE techniques to unravel the response of this understudied tissue to Fe deficiency. Proteomics data, complemented with mineral, lignin and microscopy analyses, indicate that stems respond to Fe deficiency by increasing stress and defense related proteins, probably in response of mineral and osmotic unbalances, and eliciting significant changes in cell wall composition. El Fe y el Mn están clasificados como micronutrientes que participan en funciones esenciales para el desarrollo y crecimiento de las plantas. El organismo no puede crecer sin un suministro adecuado de estos metales, pero por otro lado, si los niveles de exposición son demasiado elevados pueden resultar potencialmente tóxicos. El rango de concentraciones considerado fisiológico para los metales esenciales es muy estrecho y varía en función de la especie, el tejido y las condiciones de crecimiento. El conjunto de mecanismos implicados en el mantenimiento de los niveles adecuados de estos nutrientes es lo que se conoce como homeostasis. Desde un punto de vista fisiológico, la homeostasis de metales requiere la coordinación a nivel de toda la planta de los mecanismos de adquisición en la raíz, translocación a la parte aérea, almacenamiento y su posterior removilización a órganos sumidero. En la regulación de la homeostasis el sistema vascular juega un papel fundamental puesto que participa en la traslocación via xilema, en la removilización via floema y en la distribución y almacenamiento via apoplasto. Cuando este equilibrio se pierde, se producen pérdidas en la productividad causadas por disminuciones tanto en rendimiento como en la calidad de los cultivos que conllevan un alto impacto económico al generar productos con menor valor comercial y nutricional e incrementa los costes en el manejo del cultivo. La disponibilidad de ambos metales en la corteza terrestre depende del pH y las características redox del suelo, de forma que cuando el pH del suelo es elevado ambos metales se encuentran en formas con niveles de solubilidad mínimos que resultan difícilmente asimilables por las plantas. El 30% de los suelos cultivables presentan un pH elevado, indicando que la deficiencia de de estos metales resulta uno de los factores limitantes en el área mediterránea, en cultivos en el centro de Asia, el medio oeste de estados unidos o el sur de Australia. Por otro lado, en el caso del Mn, la cantidad del metal asimilable por las plantas aumenta considerablemente conforme el pH disminuye, de forma que en suelos con pH ácido puede llegar a resultar tóxico. Las actividades antropogénicas, como las actividades mineras y los residuos de incineración o de procesos industriales, han provocado el enriquecimiento en metales del medio ambiente, originándose una acumulación de 100 a 1.000 veces más alta en relación con su proporción natural en la corteza terrestre. La toxicidad de Mn supone un problema agronómico cada vez más común y es prevalentes en zonas del norte de Europa y Asia, el norte de EEUU y Canadá, gran parte de sudamérica o en algunas zonas de Australia. Aunque la información de la homeostasis en metales ha aumentado en los últimos años, todavía hay muchos interrogantes al respecto. Por ello, la proteómica puede resultar una herramienta muy útil a la hora de identificar y caracterizar nuevas proteínas que participan en la homeostasis de metales. El estrés por Fe ha sido más estudiado desde un punto de vista proteómico que el estrés por Mn. Los primeros estudios sobre Fe fueron en tejidos vegetales completos como la raíz y las hojas, y aún hoy en día los trabajos en estos tejidos son mayoritarios. En el caso del Mn, los trabajos publicados hasta el momento a nivel de proteoma se han centrado en plantas con características hiperacumuladoras o en estudios en hoja. Sin embargo, el análisis de subproteomas, entre los que se incluyen los fluidos de la planta, que resultan cruciales para el entendimiento de la homeostasis de estos metales, resultan todavía limitados. Con estos antecedentes el objetivo general de esta Tesis Doctoral ha sido incrementar el conocimiento existente acerca de los efectos que causan las deficiencias de Fe y Mn y la toxicidad por Mn en los proteomas vegetales implicados en la absorción (raíz) y el transporte (savias de xilema y floema y fluido apoplástico) de estos nutrientes mediante la aplicación de aproximaciones proteómicas. El uso de la proteómica diferencial en estos estreses y subproteomas puede proporcionar información sobre los procesos de adaptación de las plantas, así como apuntar hacia posibles estrategias para combatirlos. Además, el uso de técnicas de proteómica clásicas (2-DE) y avanzadas (“shotgun proteomics”) permite llevar a cabo la comparación entre técnicas y establecer las ventajas y limitaciones de ambas en el análisis de estos proteomas.

El incremento del impacto humano sobre las aguas de nuestro planeta, está modificando fuertemente la disponibilidad de agua de calidad para amplios grupos de la población mundial. Las cianobacterias del fitoplancton juegan un papel muy relevante en la preservación de la calidad de las aguas (degradación de compuestos, reciclado de nutrientes…) pero además, algunos grupos de estos organismos pueden producir toxinas que provocan serios problemas se salud pública y ambiental. Las numerosas actividades humanas desarrolladas en las cuencas fluviales desde la Revolución Industrial, además de reducir la cantidad del caudal circulante de nuestros ríos, provocan la llegada de numerosos contaminantes que provocan severas alteraciones de la ecología de los ríos. Estos cambios afectan especialmente al fitoplancton, biofilms y bacterias. El objetivo de este trabajo ha sido estudiar el posible impacto de un pesticida, el lindano, sobre la producción de una de las cianotoxinas más ubicuas y abundantes, la microcistina. Para ello, se estudiaron los cambios inducidos por el pesticida en los niveles de RNA mensajero de genes responsables de la síntesis de la microcistina, operon mcy, genes reguladores de este operon y niveles de toxina producidos. El método utilizado para llevar a cabo este estudio fue la RT-PCR semicuantitativa por la que se cuantificó la expresión de los transcritos de los genes en diferentes condiciones, comparándolo con una muestra control. Como gen de referencia se utilizó el gen rrs. La cantidad de microcistina producida se determinó por el ensayo de inhibición de la proteín fosfatasa 2A. Con este estudio se pretende aportar datos que ayuden a poner en claro los efectos ocasionados por los lixiviados de lindano (que se producen periódicamente en los vertederos en que se depositaron los residuos de fabricación de este pesticida) en la población de cianobacterias y en la toxicidad de los blooms.

Inhibiting the main protease 3CLpro is the most common strategy in the search for antiviral drugs to fight the infection from SARS-CoV-2. We report that the natural compound eugenol is able to hamper in vitro the enzymatic activity of 3CLpro, the SARS-CoV-2 main protease, with an inhibition constant in the sub-micromolar range (Ki = 0.81 μM). Two phenylpropene analogs were also tested: the same effect was observed for estragole with a lower potency (Ki = 4.1 μM), whereas anethole was less active. The binding efficiency index of these compounds is remarkably favorable due also to their small molecular mass (MW < 165 Da). The development of new antiviral drugs against SARS-CoV-2 is a valuable long-term strategy to protect the global population from the COVID-19 pandemic complementary to the vaccination. Considering this, the viral main protease (Mpro) is among the most promising molecular targets in light of its importance during the viral replication cycle. The natural flavonoid quercetin 1 has been recently reported to be a potent Mpro inhibitor in vitro, and we explored the effect produced by the introduction of organoselenium functionalities in this scaffold. In particular, we report here a new synthetic method to prepare previously inaccessible C-8 seleno-quercetin derivatives. By screening a small library of flavonols and flavone derivatives, we observed that some compounds inhibit the protease activity in vitro. For the first time, we demonstrate that quercetin (1) and 8-(p-tolylselenyl)quercetin (2d) block SARS-CoV-2 replication in infected cells at non-toxic concentrations, with an IC50 of 192 μM and 8 μM, respectively. The pandemic, due to severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2), has stimulated the search for antivirals to tackle COVID-19 infection. Molecules with known pharmacokinetics and already approved for human use have been demonstrated or predicted to be suitable to be used either directly or as a base for a scaffold-based drug design. Among these substances, quercetin is known to be a potent in vitro inhibitor of 3CLpro, the SARS-CoV-2 main protease. However, its low in vivo bioavailability calls for modifications to its molecular structure. In this work, this issue is addressed by using rutin, a natural flavonoid that is the most common glycosylated conjugate of quercetin, as a model. Combining experimental (spectroscopy and calorimetry) and simulation techniques (docking and molecular dynamics simulations), we demonstrate that the sugar adduct does not hamper rutin binding to 3CLpro, and the conjugated compound preserves a high potency (inhibition constant in the low micromolar range, Ki = 11 μM). Although showing a disruption of the pseudo-symmetry in the chemical structure, a larger steric volume and molecular weight, and a higher solubility compared to quercetin, rutin is able to associate in the active site of 3CLpro, interacting with the catalytic dyad (His41/Cys145). The COVID-19 pandemic outbreak prompts an urgent need for efficient therapeutics, and repurposing of known drugs has been extensively used in an attempt to get to anti-SARS-CoV-2 agents in the shortest possible time. The glycoside rutin shows manifold pharmacological activities and, despite its use being limited by its poor solubility in water, it is the active principle of many pharmaceutical preparations. We herein report our in silico and experimental investigations of rutin as a SARS-CoV-2 Mpro inhibitor and of its water solubility improvement obtained by mixing it with l-arginine. Tests of the rutin/l-arginine mixture in a cellular model of SARS-CoV-2 infection highlighted that the mixture still suffers from unfavorable pharmacokinetic properties, but nonetheless, the results of this study suggest that rutin might be a good starting point for hit optimization., This research was in part founded with POR FESR 2014/2020, and thus, Regione Umbria is gratefully acknowledged (S.M.). 13 Pags.- 3 Tabls.- 4 Figs., The aim of this work was to assess the effects of manganese (Mn) toxicity on the proteome of tomato roots using two proteomic approaches, shotgun and two-dimensional electrophoresis. The shotgun approach yielded 367 reliable proteins, whereas the 2-DE approach detected 340 consistent spots. The 2-DE method found 54 proteins changing in relative abundance in the excess Mn treatment, whereas the shotgun detected changes in 118 proteins. Only 7% of the differential proteins were found by both methods, illustrating their complementary nature. Metabolic pathways most affected were protein metabolism, oxido-reductases and signaling. Results support that Mn toxicity alters the protein turnover and impairs energy production in roots, leading to changes in glycolysis, pyruvate metabolism, TCA and oxidative phosphorylation. Excess Mn also induced changes in peroxidases and hydrolases participating in cell wall lignification and suberization and activated plant defense mechanisms, with changes occurring via pathogenesis-related proteins as well as peroxidases. Finally, Mn toxicity elicited regulatory mechanisms and affected the abundance of root nutrient reservoir proteins. The overall analysis of the differential root proteome upon Mn toxicity suggests a general slowdown of metabolic activities, especially energy production, cell wall integrity and protein turnover, which occurs in parallel with increases in stress related proteins., Supported by the Spanish Ministry of Economy and Competitiveness (MINECO; project AGL2016-75226-R, co-financed with FEDER).
