The aim of the present study was to optimize the preparation of an immobilized acetylcholinesterase (AChE)-based micro-immobilized enzyme reactor (IMER) for inhibition studies. For this purpose two polymeric monolithic disks (CIM, 3 mm x 12 mm i.d.) with different reactive groups (epoxy and ethylendiamino) and a packed silica column (3 mm x 5 mm i.d.; Glutaraldehyde-P, 40 microm) were selected as solid chromatographic supports. All these reactors were characterized in terms of rate of immobilization, stability, conditioning time for HPLC analyses, optimum mobile phase and peak shape, aspecific interactions and costs. Advantages and disadvantages were defined for each system. Immobilization through Schiff base linkage gave more stable reactors without any significant change in the enzyme behaviour; monolithic matrices showed very short conditioning time and fast recovery of the enzymatic activity that could represent very important features in high throughput analysis and satisfactory reproducibility of immobilization yield. Unpacked silica material allowed off-line low costs studies for the optimization of the immobilization step.

Amphiphilic copolymers have been the object of growing scientific interest due to their ability to form polymeric micelles in aqueous environments entrapping lipophilic drugs in their inner core. In this study, polyvinylalcohol substituted with oleic acid was employed as an amphiphilic micellar carrier for folic acid (FA), a model drug similar for its chemical-physical characteristics to methotrexate. In order to investigate the stability of the polymeric micelles, the drug incorporation and the kinetic aspects of drug release from these systems, selective analytical methods are required. The development of three analytical methods suitable for selectively identifying and reliably determining FA contained in the micelles and in the delivery systems is reported. UV derivative (first and second order) spectrophotometry was first applied to the aqueous solution of the FA containing micelles obtained at pH 9.0 and provided a characteristic spectral profiling with sharp peaks, related to the analyte, whose amplitude was used for quantitative application. A second approach involved a solid phase extraction (strong anion exchanger), which provided an effective clean up of the FA micelles solution, allowing accurate analysis to be performed also by a conventional spectrophotometric method. A RP-HPLC method, selectively supplying the FA separation from the micelles' components, was then used as a reference method to determine the accuracy of the spectrophotometric methods. L'analisi volumetrica o volumetria comprende vari metodi di analisi quantitativa, tutti caratterizzati dall'applicazione di un procedimento pratico definito con il termine "titolazione". Si tratta di un procedimento che permette di determinare la quantità di un analita in una soluzione (campione o titolato), basandosi sulla misura del volume di un reagente a concentrazione nota (titolante) che viene aggiunto al campione fino a che la sua reazione con l'analita arriva a completamento.

Il capitolo tratta i principi generali e le applicazioni in campo farmaceutico della tecnica separativa elettroforesi capillare. In particolare, sono sviluppati gli aspetti teorici e quelli strumentali assieme alle diverse modalità in cui tale tecnica può essere realizzata.

La maggior parte delle analisi viene effettuata prelevando una parte o porzione del materiale/prodotto da esaminare (formulazione farmaceutica, estratto da pianta medicinale, prodotto cosmetico, etc.) per sottoporlo ad un procedimento di analisi di laboratorio o talvolta anche sul posto. Questa porzione viene detta campione ed è costituita dagli analiti (principi attivi), di cui si vuole verificare la presenz e7o il contenuto e dalla matrice (insieme dei componenti inattivi) che racchiude in forma dispersa gli analiti. L'intero processo di analisi prevede quindi varie fasi, ognuna delle quali richiede la dovuta attenzione. La prima fase è il campionamento cioè il prelievo del campione dal materiale in esame. Di solitp c'è un certo ritardo tra il momento della raccolta del campione e la sua analisi; pertanto la coservazione del campione può costituire un punto critico del processo di lavoro analitico. Infine, la terza importante fase è la preparazione del campione. Questa fase che precede la misura analitica è di estrema importanza, infatti, il campione non puà essere introdotto direttamente tal quale nello strumneto di analisi ma deve essere reso "presentabile" allo strumento analitico.In altre parole preparare il campione significa sottoporre il campione ad un tratatmento appropriato per portatre gli analiti nelle condizioni adatte di solvente, pH, concentrazione, forma molecolare rilevabile ed assenza di interferenze, assicurando nel contempo la loro stabilità ed il loro recupero quantitativo e/o riproducibile dalla matrice in cui si trovano dispersi. La preparzione del campione quindi prevede solitamente un processo di estrazione degli analiti dalla loro matrice mediante l'impiego di gas inerti, solventi, fluidi supercritici o adsorbenti solidi. Le principali tecniche impiegate per estrarre un soluto (principio attivo) dalla matrice di un campione di interesse farmaceutico possono essere riassunte nel modo seguente: (i) estrazioni liquido-solido in cui si cerca di rimuovere il soluto da una matrice solida mediante un solvente o un fluido supercritico, (ii) estrazione liquido-liquido che consente di trasferire in modo selettivo un soluto da una fase liquida ad un'altra fase liquida immiscibile con la prima, (iii) estrazione in fase solida che consente di estrarre un soluto da un campione liquido o gassoso mediante un materiale solido adsorbente sul quale il soluto viene selettivamente concentrato e poi rimosso con quantità minime di solvente o per desorbimento termico.

The amyloid beta (Abeta) peptide is responsible for toxic amyloid plaque formation and is central to the aetiology of Alzheimer's disease (AD). It is generated by proteolytic processing of the amyloid precursor protein (APP) by beta-secretase (BACE-1) and gamma-secretase. Consequently, inhibition of BACE-1, a rate-limiting enzyme in the production of Abeta, is an attractive therapeutic approach to the treatment of Alzheimer's disease. This paper reports on improved microtiter plate-based fluorescence and colorimetric assays for the high-throughput screening (HTS) of BACE-1 inhibitors achieved by employing, for the first time, casein fluorescein isothiocyanate (casein-FITC) and N-alpha-benzoyl-D,L-arginine p-nitroanilide (BAPNA) as substrates, since they are known to be readily available and convenient substrates for proteases. The methods are based on the fluorescence enhancement following casein-FITC proteolysis and the visible absorbance of the p-nitroaniline (pNA) produced by BAPNA hydrolysis, with both reactions catalysed by BACE-1. Casein-FITC is a high-affinity substrate (K (m) = 110 nM) for BACE-1, more so than the Swedish (SW) type peptide (a peptide containing the Swedish mutant of APP, a familiar mutation that enhances Abeta production). BACE-1 catalysis of casein-FITC proteolysis exhibited Michaelis-Menten kinetic. Therefore, it was found that BACE-1 was saturable with casein-FITC that was processed in a time- and pH-dependent manner with greater catalytic efficiency than observed for the SW peptide. The enantioselective hydrolysis of L-BAPNA by BACE-1 was observed. L-BAPNA was hydrolysed ten times more efficiently by BACE-1 than the WT (wild-type peptide). The novel methods were validated using a FRET assay as an independent reference method. Therefore, in order to select new leads endowed with multifunctional activities, drugs for Alzheimer's disease (AD) - potent acetylcholinesterase (AChE) inhibitors - were tested for BACE-1 inhibition using the proposed validated assays. Among these, donepezil, besides being an acetylcholinesterase inhibitor, was also found to be a BACE-1 inhibitor that displayed submicromolar potency (170 nM).

Il presente testo, rinnovato ed ampliato rispetto alla precedente edizione, viene proposto come guida all'analisi farmaceutica, in quanto, da un lato riunisce indispensabili nozioni di base di chimica analitica, chimica organica e chimica farmaceuticae, dall'altro, introduce lo studente alla conoscenza di avanzate ed aggiornate tecniche analitiche mirate ad affrontare le specifiche problematiche dell'analisi di farmaci di origine sintetica o naturale.

A GC/FID/MS method was developed for the identification and quantification of d-allethrin (DA) and its major impurities in commercial samples. Optimisation of the experimental conditions was carried out considering such important requirements as resolution, reproducibility, detection limits of 0.1% (m/m) for the impurities, and short analysis time. Under the optimised final conditions the method was validated for specificity, precision (CV% = 0.133 at 2.10 mg/mL and CV% = 0.035 at 3.00 mg/mL), linearity (0-3.00 microg injected), limits of detection (0.09 ng injected) and quantitation (0.28 ng injected), and robustness. The DA related impurities were identified by using a GC/MS method with ion trap mass detection and also by comparison with synthesised standards. The most abundant impurities were crysolactone, allethrolone, chrysanthemic acid, and chloro-derivatives of DA.
