Preterm premature rupture of membranes (PPROM) remote from term is an important obstetric cause of maternal and fetal adverse outcomes. The aim of our study is to examine the efficacy of ampicillin and Lactobacillus casei rhamnosus treatment in cases of PPROM remote from term.The study was carried out by examining the results of cases who were given Ampicillin and Lactobacillus casei rhamnosus treatment. The patients were divided into two groups. Group 1 who didn't develop clinical chorioamnionitis and Group 2 who developed clinical chorioamnionitis. Obstetric characteristics, neonatal outcomes, adverse events were recorded.A total of 46 pregnant women, 40 in Group 1 and six in Group 2, were included in the study. The frequency of clinical chorioamnionitis developing during the treatment was found to be 13.0%. Mean gestational age at diagnosis was 28.43 ± 2.38 and 28.17 ± 1.33 for Groups 1 and Group 2, respectively. Mean gestational age at the time of delivery was 32.38 ± 2.07 31.33 ± 1.63 for Group 1 and Group 2, respectively. The mean latency period for Group 1 and Group 2 was 27.45 ± 1.71 days, 23.66 ± 4.53, respectively. Sepsis developed in six newborns (15%) in Group 1, while it developed in three newborns (50%) in Group 2. While 90% of the babies in Group 1 were discharged from the hospital, this rate was 66.7% in Group 2.Ampicillin + Lactobacillus casei rhamnosus is an effective treatment method in PPROM cases and positively affects perinatal outcomes.

3. Evaluation of the relationship of intuitive eating behaviour between body mass index and waist circumference in university students

Objective: This study was conducted to examine the relationship between intuitive eating behaviour, body mass index (BMI), and waist-hip circumference among university students. Methods: This study was conducted on 146 students studying at the Department of Nutrition and Dietetics of a foundation university on the European side of Istanbul in the 2020-2021 academic year. Socio-demographic characteristics, anthropometric measurements, and intuitive eating habits of the students were taken through an online questionnaire. Intuitive Eating Scale-2 (IES-2) was used to determine students’ intuitive eating behaviour. Results: 146 students with a mean age of 20.64±1.69 participated in the study. 62.3% of the students were female and 37.7% were male. Intuitive eating scale total score mean ± standard deviation values were found to be 3.40±0.55. BMI mean ± standard deviation values were found to be 21.89 ± 3.94 kg / m2 while waist circumference mean ± standard deviation values were 69.78±9.32 cm. eating due to physical rather than emotional causes sub-dimension has a positive and significant correlation with the eating sub-dimension related to hunger and fullness signals, body-food choice congruity, and intuitive eating scale-total score. There is a significant and positive relationship between the eating sub-dimension related to hunger and fullness signals and the body-food choice congruity sub-dimension and the total score of the intuitive eating scale. A significant and positive correlation was found between the body-food choice congruity sub-dimension and the total score of the intuitive eating scale. Comparing the intuitive eating scale total scores and sub-dimensions according to BMI groups, there is a significant difference in the mean score of the body-food choice congruity sub-dimension according to the BMI classification. Conclusion: As intuitive eating increases in university students, the body mass index decreases.

Disrupting bacterial quorum sensing (QS) signaling is a promising strategy to combat pathogenic biofilms without the development of antibiotic resistance. Here, we report that food-associated bacteria can interfere with the biofilm formation of a Gram-negative pathogenic bacterium by targeting its AHL (acyl-homoserine lactone) QS system. This was demonstrated by screening metabolic end-products of different lactobacilli and propionibacteria using Gram-negative and biofilm-forming Chromobacterium violaceum as the QS reporter and our anti-QS microscale screening platform with necessary modifications. The method was optimized in terms of the inoculation technique and the concentrations of D-glucose and L-tryptophan, two key factors controlling the synthesis of violacein, a purple pigment indicating the activation of the QS system in C. violaceum. These improvements resulted in ca. 16-times higher violacein yields and enabled revealing anti-QS effects of Lactobacillus acidophilus, Lentilactobacillus kefiri, Lacticaseibacillus rhamnosus and Propionibacterium freudenreichii, including new cheese-associated strains. Our findings also suggest that acetate and propionate excreted by these species are the main factors that interrupt the QS-mediated signaling and subsequent biofilm growth without affecting the cell viability of the C. violaceum reporter. Thus, the present study reports a revised anti-QS screening method to accurately define new bacteria with an ability to combat pathogens in a safe and sustainable way. publishedVersion

6. Neonatal Isoflurane Exposure in Rats Impairs Short-Term Memory, Cell Viability, and Glutamate Uptake in Slices of the Frontal Cerebral Cortex, But Not the Hippocampus, in Adulthood

Neonatal exposure to general anesthetics has been associated with neurotoxicity and morphologic changes in the developing brain. Isoflurane is a volatile anesthetic widely used in pediatric patients to induce general anesthesia, analgesia, and perioperative sedation. In the present study, we investigated the effects of a single neonatal isoflurane (3% in oxygen, 2 h) exposure in rats at postnatal day (PND) 7, in short-term (24 h - PND8) and long-term (adulthood) protocols. In PND8, ex vivo analysis of hippocampal and frontal cortex slices evaluated cell viability and susceptibility to in vitro glutamate challenge. In adult rats, behavioral parameters related to anxiety-like behavior, short-term memory, and locomotor activity (PND60-62) and ex vivo analysis of cell viability, membrane permeability, glutamate uptake, and susceptibility to in vitro glutamate challenge in hippocampal and cortical slices from PND65. A single isoflurane (3%, 2 h) exposure at PND7 did not acutely alter cell viability in cortical and hippocampal slices of infant rats (PND8) per se and did not alter slice susceptibility to in vitro glutamate challenge. In rat's adulthood, behavioral analysis revealed that the neonatal isoflurane exposure did not alter anxiety-like behavior and locomotor activity (open field and rotarod tests). However, isoflurane exposure impaired short-term memory evaluated in the novel object recognition task. Ex vivo analysis of brain slices showed isoflurane neonatal exposure selectively decreased cell viability and glutamate uptake in cortical slices, but it did not alter hippocampal slice viability or glutamate uptake (PND65). Isoflurane exposure did not alter in vitro glutamate-induced neurotoxicity to slices, and isoflurane exposure caused no significant long-term damage to cell membranes in hippocampal or cortical slices. These findings indicate that a single neonatal isoflurane exposure did not promote acute damage; however, it reduced cortical, but not hippocampal, slice viability and glutamate uptake in the adulthood. Additionally, behavioral analysis showed neonatal isoflurane exposure induces short-term recognition memory impairment, consolidating that neonatal exposure to volatile anesthetics may lead to behavioral impairment in the adulthood, although it may damage brain regions differentially.

The thesis aims to bridge a gap in the current re-evaluation of women’s contribution to the literature of Renaissance Italy, by focussing on the reception of Ariosto’s epic poem Orlando Furioso - published in its ‘final’ version in 1532 - by three female poets of the sixteenth century: Isabella di Morra (1516/18-45/46), Laura Bacio Terracina (c.1519-77) and Moderata Fonte (1555-92). The thesis examines how the ‘Ariostism’ of all three women writers challenges the model of Petrarch as benchmark for female literary production during the Cinquecento. Chapter One outlines some of the gender-related issues raised in the Furioso, with which the three female poets may subsequently be said to engage. The chapter looks at both narratorial statements and constructions of masculinity and femininity within the text, revealing Ariosto’s complex relationship with the question of gender. Chapter Two considers the Ariostan echoes present within Isabella di Morra’s Rime, first published in full in an anthology of 1559. Her small canzoniere - not typically Petrarchan in its themes and tone - is re-considered in light of the Furioso, especially with regard to the topos of the ‘abandoned woman’ and the twin themes of harsh landscape and malevolent Fortune which are central to her poetics. Chapter Three discusses Laura Bacio Terracina’s Discorso sopra tutti i primi canti d’Orlando Furioso (1549), a moralizing commentary in ottava rima which constitutes one of the most direct responses to Ariosto’s poem. Terracina’s bold appropriation of the model of the Furioso incorporates aims of both a poetic and an ideological nature, since her engagement also revolves around moral issues that are mostly gendered in their emphasis. Chapter Four considers Moderata Fonte’s Tredici canti del Floridoro (1581), the first ‘original’ romance epic to be written and published by a woman. The Floridoro contains strong thematic and structural parallels with the Furioso; yet the chapter also highlights the ways in which Fonte diverges from the Ariostan precedent re-writing the epic genre in feminine - and ‘feminist’ - key. The thesis concludes that the poetic model of the Furioso would play an increasingly important role in the development of the female literary canon as the century progressed.

Aim Previous studies reported conflicting evidence on the effects of obesity on outcomes after gastrointestinal surgery. The aims of this study were to explore the relationship of obesity with major postoperative complications in an international cohort and to present a meta-analysis of all available prospective data.Methods This prospective, multicentre study included adults undergoing both elective and emergency gastrointestinal resection, reversal of stoma or formation of stoma. The primary end-point was 30-day major complications (Clavien-Dindo Grades III-V). A systematic search was undertaken for studies assessing the relationship between obesity and major complications after gastrointestinal surgery. Individual patient meta-analysis was used to analyse pooled results.Results This study included 2519 patients across 127 centres, of whom 560 (22.2%) were obese. Unadjusted major complication rates were lower in obese vs normal weight patients (13.0% vs 16.2%, respectively), but this did not reach statistical significance (P = 0.863) on multivariate analysis for patients having surgery for either malignant or benign conditions. Individual patient meta-analysis demonstrated that obese patients undergoing surgery for malignancy were at increased risk of major complications (OR 2.10, 95% CI 1.49-2.96, P Conclusions In our international data, obesity was not found to be associated with major complications following gastrointestinal surgery. Meta-analysis of available prospective data made a novel finding of obesity being associated with different outcomes depending on whether patients were undergoing surgery for benign or malignant disease.

Since the start of the COVID-19 pandemic in March 2020, the Inter-American Commission of Women (CIM/OAS) and the European Union (EU), through the Gender Equality area of the EUROsociAL+ program, have articulated a process of work to strengthen the public policies of the States on the rights and economic autonomy of women. In this route, of incidence to position new gender pacts, we started from the analysis of the legal and normative framework on the economic rights of women, with the purpose of, on the one hand, to formulate and support the implementation of practical recommendations and, on the other hand, to identify standards for the creation of normative and legal tools that support the full exercise of the economic rights of the women and their empowerment and autonomy, with power to transform the lives of women in the Region. As a result of this dynamic, the Inter-American Model Law on Care arises, which includes key elements of international instruments and is based on existing standards in terms of rights, non-discrimination and equality between men and women. Its purpose is to recognize, redistribute, regulate, promote and generate new forms of attention to unpaid care and domestic work, as well as to make visible and recognize the historical contribution of women in this matter. Through this Model Law, we offer a concrete tool to the States to strengthen economic autonomy and enhance the contribution and human talent of women in the productive world and towards sustainable economic growth. This work is framed within the mandates of the Assembly of Delegates of the CIM, which has declared the importance of making visible the economic and social value of unpaid work in the home, particularly care work, and of promoting that laws and public policies incorporate the social co-responsibility of care to create the necessary conditions so that women can participate and remain in the labor market under equal conditions and fully enjoy their rights.

Assim, para que seja possível cumprir com tal objetivo, a primeira etapa consiste em caracterizar o gênero fantástico a partir do que Roas (2014) e Bozzetto (2001) qualificam como um problema de linguagem enquanto fundamento do fantástico contemporâneo, assim como seu diálogo constante com a irrepresentabilidade da alteridade. A definição do (neo)fantástico feita por Alazraki também foi cara ao trabalho. Após uma primeira etapa de alicerce conceitual, a pesquisa se debruça diretamente sobre a obra de Silvina Ocampo, explorando o contexto em que tornou-se escritora e sua escrita propriamente dita. Assim, a leitura feita dos contos de A Fúria considera as intersecções com a perspectiva transgressora do (neo)fantástico, especialmente em seu trato da linguagem, no qual a exageração e o absurdo estão a serviço de uma experiência do insólito. A tomada de conceituações psicanalíticas, como é o caso do unheimlich freudiano, é a base para uma inerdisciplinaridade que não extingue as questões de análise literária, mas pelo contrário, propõe novos olhares e possibilidades de singificação. Com a leitura crítica da bibliografia acerca do trabalho de Silvina Ocampo, somada às considerações teóricas a partir de propostas conceituais psicanalíticas e de teoria literária, fez-se possível pensar o texto ocampiano a partir de temas fundamentais, como o feminino, a religiosidade, os atravessamentos sociais e de classe, traduzido nas relações afetivas com objetos inventariados. Para os fins pretendidos, a base bibliográfica da pesquisa é composta majoritariamente por textos de estudiosos de Silvina Ocampo, como Sylvia Molloy e Mariana Enriquez; textos de teóricos da psicanálise, como Freud, Lacan e Maria Rita Kehl; e teóricos do fantástico, como David Roas, Roger Bozzetto e Jaime Alazraki. Apesar da grande quantidade de textos teóricos, a coletânea A Fúria e outros contos é, do começo ao fim, o principal objeto e razão de ser desta dissertação. Abstract: This work proposes a reading of five short stories by Silvina Ocampo in her collection of texts A Fúria e outros contos, her first work translated and published in Brazil, having as main goal to think and critically analyze the (neo)fantastic strategy in the author’s writing. Thus, in order to achieve this goal, the first step consists in characterizing the fantastic genre from what Roas (2014) and Bozzetto (2001) qualify as a language problem as the foundation of the contemporary fantastic, as well as its constant dialogue with the unrepresentability of otherness. Alazraki's definition of the (neo)fantastic was also significant to this work. After a first stage of conceptual foundation, the research focuses directly on the work of Silvina Ocampo, exploring the context in which she became a writer and her writing itself. The reading of the short stories extracted from of A Fúria considers the intersections with the transgressive perspective of the (neo)fantastic, especially in its treatment of language, in which exaggeration and absurdity are at the service of an experience of the unusual. The use of psychoanalytic concepts, as is the case of the Freudian unheimlich, is the basis for an interdisciplinary approach that does not extinguish questions of literary analysis, but on the contrary, proposes new perspectives and possibilities of meaning. With the critical reading of the bibliography about Silvina Ocampo's work, added to theoretical considerations from psychoanalytic conceptual ideas and literary theory, it was possible to think about the Ocampian text considering fundamental themes, such as the feminine, religiosity, social and class issues, translated into the meaningful relationships with maintained with inventoried objects. For the intended purposes, the bibliographic base of the research is mainly composed of texts by Silvina Ocampo scholars, such as Sylvia Molloy and Mariana Enriquez; texts by psychoanalysis theorists, such as Freud, Lacan and Maria Rita Kehl; and fantastic literature theorists such as David Roas, Roger Bozzetto and Jaime Alazraki. Despite the large amount of theoretical texts, the collection A Fúria e outros contos is, from beginning to end, the main object and reason for being of this dissertation. Resumen: El presente trabajo es una propuesta de lectura de cinco cuentos de Silvina Ocampo de su colección de textos La furia y otros cuentos, su primera obra traducida y publicada en Brasil, teniendo como principal objetivo pensar y analizar críticamente la estrategia (neo)fantástica en escrito de la autora. Así, para lograr este objetivo, el primer paso consistió en caracterizar el género fantástico a partir de lo que Roas (2014) y Bozzetto (2001) califican como un problema de lenguaje como fundamento de lo fantástico contemporáneo, así como su diálogo constante con la irrepresentabilidad de la alteridad. La definición de Alazraki de lo (neo)fantástico también fue fundamental para el trabajo. Después de una primera etapa de fundamentación conceptual, la investigación se centró directamente en la obra de Silvina Ocampo, explorando el contexto en el que se convirtió en escritora y su escritura como tal. Así, la lectura realizada de los cuentos de La furia consideró las intersecciones con la perspectiva transgresora del (neo)fantástico, especialmente en su tratamiento del lenguaje, en el que la exageración y el absurdo están al servicio de una experiencia de lo insólito. La tomada de conceptualizaciones psicoanalíticas, como el unheimlich freudiano, son la base para un abordaje interdisciplinario que no extingue cuestiones de análisis literario, sino por el contrario, propone nuevas perspectivas y posibilidades de sentido. Con la lectura crítica de la bibliografía sobre la obra de Silvina Ocampo, sumada a consideraciones teóricas provenientes de las propuestas conceptuales psicoanalíticas y de la teoría literaria, fue posible pensar el texto ocampeano a partir de temas fundamentales, como lo femenino, la religiosidad, los cruces sociales y de clase, traducidos en relaciones afectivas con objetos inventariados. Para los fines previstos, la base bibliográfica de la investigación está compuesta principalmente por textos de intelectuales que estudian a Silvina Ocampo, como Sylvia Molloy y Mariana Enríquez; textos de teóricos del psicoanálisis, como Freud, Lacan y Maria Rita Kehl; y teóricos del fantástico como David Roas, Roger Bozzetto y Jaime Alazraki. A pesar de la gran cantidad de textos teóricos, la colección La furia y otros cuentos es, del principio al fin, el objeto principal y la razón de ser de esta disertación.

Denna studie undersöker om ett chock-moment i ett så kallat serious game kan användas som medel för att påverka spelare positivt och få dem att reflektera över sina konsumtionsvanor och miljöpåverkan i verkliga livet. Vad som framgår är att målgrupp, spels design samt hur chocken presenteras är oerhört viktigt för att få önskvärda resultat. Denna studie gör ett försök att fylla i luckor som upptäckts under förundersökningen och testar om chock går att framkalla via ett brädspel på ett etiskt och betydelsefullt sätt. Detta leder till frågeformuleringen: Hur väl fungerar chock som en motivator till beteendeförändring efter ett serious game? Hur uppfattar spelare chocken i ett miljöspel? Det finns övrigt digitalt material (t.ex. film-, bild- eller ljudfiler) eller modeller/artefakter tillhörande examensarbetet som ska skickas till arkivet.

La Ley Modelo Interamericana de Cuidados (LMIC) nace de la competencia de la Organización de los Estados Americanos (OEA) de avanzar hacia el fortalecimiento de los marcos jurídicos y normativos de sus Estados Miembros, desarrollando instrumentos que sirvan para este fin. La Ley Modelo Interamericana de Cuidados (LMIC) representa una herramienta de alcance regional que busca establecer las bases de un nuevo pacto en la organización social de los cuidados que resignifique su rol en la sociedad, permita hacer frente a su crisis actual e impulse el salto evolutivo hacia sociedades que cuidan. Ello permite crear y mantener comunidades de vida libres, justas, sostenibles y prósperas, que respeten y protejan los derechos humanos de mujeres y hombres. La Guía de Implementación que acompaña la LMIC recupera los más altos estándares en esta materia, así como doctrinas y prácticas de la región, haciendo un recorrido de las normas y de las experiencias de los países que ya vienen avanzando acciones en el ámbito de los cuidados. La Unión Europea, a través de su programa EUROsociAL+, contribuyó también con la Guía de Implementación, ilustrando los avances y las experiencias de países europeos en el tema de los cuidados. Las experiencias europeas nos permiten afianzar la noción de tender puentes para consolidar pactos sociales que promuevan el empoderamiento de las mujeres. Desde el inicio de la pandemia del COVID-19 en marzo de 2020, la Comisión Interamericana de Mujeres (CIM/OEA) y la Unión Europea (UE), a través del área de Igualdad de Género del programa EUROsociAL+, hemos articulado un proceso de trabajo para fortalecer las políticas públicas de los Estados sobre los derechos y autonomía económica de las mujeres. En esta ruta, de incidencia para posicionar nuevos pactos de género, se partió del análisis del marco legal y normativo sobre los derechos económicos de las mujeres, con el propósito de, por un lado, formular y apoyar la implementación de recomendaciones prácticas y, por otro lado, identificar estándares para la creación de herramientas normativas y jurídicas que apoyen el pleno ejercicio de los derechos económicos de las mujeres y su empoderamiento y autonomía, con poder para transformar la vida de las mujeres de la Región. Producto de esta dinámica surge la Ley Modelo Interamericana de Cuidados recoge elementos clave de instrumentos internacionales y se basa en estándares existentes en materia de derechos, no discriminación e igualdad entre hombres y mujeres. Tiene como objeto reconocer, redistribuir, regular, promocionar y generar nuevas formas de atención del trabajo de cuidados y doméstico no remunerado, así como visibilizar y reconocer la contribución histórica de las mujeres en esta materia. A través de esta Ley Modelo, ofrecemos una herramienta concreta a los Estados para fortalecer la autonomía económica y potenciar el aporte y el talento humano de las mujeres en el mundo productivo y hacia el crecimiento económico sostenible. Este trabajo se enmarca dentro de los mandatos de la Asamblea de Delegadas de la CIM, quien ha declarado la importancia de visibilizar el valor económico y social del trabajo no remunerado en el hogar, en particular el trabajo de cuidado, y de promover que las leyes y políticas públicas incorporen la corresponsabilidad social del cuidado para crear las condiciones necesarias para que las mujeres puedan participar y permanecer en el mercado laboral en igualdad de condiciones y disfrutar plenamente de sus derechos. Denna studie syftar till att undersöka skildringen av hur maktdiskursen i dokumentären "Pornfluencers" gestaltas även hur makten tar sig form utifrån dokumentären. Dokumentären "Pornfluencers" på SVT som publicerades den 15 juni 2021 och är publicerad av Erik Galli och Lisa Östberg. Studien genomförs med hjälp av en foucauldiansk diskursanalytisk metod, materialet analyseras med hjälp a Simone de Beauvoirs begrepp transcendens, immanens samt ond tro, även Rosalind Gills teorier om hur postfeminism ger sig uttryck i media. Materialet diskuteras också utifrån tidigare forskning av Nicholas de Villiers samt kommer dokumentären ha analyserats med hjälp av Lars Gustav Andersson och Erik Hedling (1999) Filmanalys. Med dessa metoder kommer jag fram till att makten som framkommer via dokumentären har en central roll och ger sig uttryck i två olika maktformer. De maktformer som dokumentären påvisar är en maktposition hos kreatörerna som använder sig av onlineplattformen Onlyfans samt en ekonomiskmaktposition.

This master thesis has two main objectives, the first is to develop a list of impressionistic key features and the second is to apply those key features when writing idiomatic music for the guitar. The composers of the impressionist era tried to capture the sense of an emotion through the musical language, something this master thesis investigates further. During the process of gathering information about impressionistic key features, I found important building blocks that made me aware of the impressionistic musical language. The four methods I used were arranging and analyzing sheet music, interviews with composers, literature studies and video interpretation. When I found enough material, I started to write music using the impressionistic key features. It resulted in three impressionistic sounding pieces. The pieces are presented with sheet music and an audio recording. As a reader and musician, one can gain knowledge about the impressionistic sound, how to write idiomatic music for the guitar and methods how one can intertwine these two subjects. The discussion part of the thesis addresses problems I struggled with during the process and how I could move away from these difficulties using composing methods and utilizing the impressionistic key features in a creative manor.

The Inter-American Model Law on Care (LMIC) represents a tool of regional scope that seeks to establish the bases of a new pact in the social organization of care that resignifies its role in society, allows it to face its current crisis and promote the leap evolutionary towards caring societies. The Implementation Guide that accompanies the LMIC recovers the highest standards in this matter, as well as doctrines and practices of the region, making a tour of the standards and experiences of countries that are already advancing actions in the field of care. The European Union, through its EUROsociAL+ program, also contributed with the Implementation Guide, illustrating the progress and experiences of European countries on the subject of care. European experiences allow us to strengthen the notion of building bridges to consolidate social pacts that promote the empowerment of women. The purpose of this study is to analyze the RRF antenna tower under wind load action, the wind load is viewed from an angle of 0( to the towing cable to the largest angle on the two types of antenna towers, triangle and Square. Both types will produce different wind loads so that we can find out which load is the biggest from a combination of wind directions. The research method is by empirical analysis, calculating the wind load alone while other loads are not counted. Due to the wind load that works horizontally and will be provided by the tower that stands vertically, the tower requires several retaining cables in a diagonal position, the force that propagates on the diagonal cable will cause a force in the vertical direction or lift, so the lift must be placed using anchors that are embedded in the soil, the large load from the soil that will hold the anchor is determined based on the value of the analysis, the criteria are too light so the anchor will be lifted and friendly, it will cause waste.

Glial cells are involved in multiple cerebral functions that profoundly influence brain tissue viability during ischemia, and astrocytes are the main source of extracellular purines as adenosine and guanosine. The endogenous guanine-based nucleoside guanosine is a neuromodulator implicated in important processes in the brain, such as modulation of glutamatergic transmission and protection against oxidative and inflammatory damage. We evaluated if the neuroprotective effect of guanosine is also observed in cultured cortical astrocytes subjected to oxygen/glucose deprivation (OGD) and reoxygenation. We also assessed the involvement of A1 and A2A adenosine receptors and phosphatidylinositol-3 kinase (PI3K), MAPK, and protein kinase C (PKC) signaling pathways on the guanosine effects. OGD/reoxygenation decreased cell viability and glutamate uptake and increased reactive oxygen species (ROS) production in cultured astrocytes. Guanosine treatment prevented these OGD-induced damaging effects. Dipropyl-cyclopentyl-xanthine (an adenosine A1 receptor antagonist) and 4-[2-[[6-amino-9-(N-ethyl-β-D-ribofuranuronamidosyl)-9H-purin-2-yl]amino]ethyl] benzenepropanoic acid hydrochloride (an adenosine A2A receptor agonist) abolished guanosine-induced protective effects on ROS production, glutamate uptake, and cell viability. The PI3K pathway inhibitor 2-morpholin-4-yl-8-phenylchromen-4-one, the extracellular-signal regulated kinase kinase (MEK) inhibitor 2'-amino-3'-methoxyflavone, or the PKC inhibitor chelerythrine abolished the guanosine effect of preventing OGD-induced cells viability reduction. PI3K inhibition partially prevented the guanosine effect of reducing ROS production, whereas MEK and PKC inhibitions prevented the guanosine effect of restoring glutamate uptake. The total immunocontent of the main astrocytic glutamate transporter glutamate transporter-1 (GLT-1) was not altered by OGD and guanosine. However, MEK and PKC inhibitions also abolished the guanosine effect of increasing cell-surface expression of GLT-1 in astrocytes subjected to OGD. Then, guanosine prevents oxidative damage and stimulates astrocytic glutamate uptake during ischemic events via adenosine A1 and A2A receptors and modulation of survival signaling pathways, contributing to microenvironment homeostasis that culminates in neuroprotection.

The guanine-based purines (GBPs) have essential extracellular functions such as modulation of glutamatergic transmission and trophic effects on neurons and astrocytes. We previously showed that GBPs, such as guanosine-5'-monophosphate (GMP) or guanosine (GUO), promote the reorganization of extracellular matrix proteins in astrocytes, and increase the number of neurons in a neuron-astrocyte co-culture protocol. To delineate the molecular basis underlying these effects, we isolated cerebellar neurons in culture and treated them with a conditioned medium derived from astrocytes previously exposed to GUO or GMP (GBPs-ACM) or, directly, with GUO or GMP. Agreeing with the previous studies, there was an increase in the number of β-tubulin III-positive neurons in both conditions, compared with controls. Interestingly, the increase in the number of neurons in the neuronal cultures treated directly with GUO or GMP was more prominent, suggesting a direct interaction of GBPs on cerebellar neurons. To investigate this issue, we assessed the role of adenosine and glutamate receptors and related intracellular signaling pathways after GUO or GMP treatment. We found an involvement of A2A adenosine receptors, ionotropic glutamate N-methyl-D-aspartate (NMDA), and non-NMDA receptors in the increased number of cerebellar neurons. The signaling pathways extracellular-regulated kinase (ERK), calcium-calmodulin-dependent kinase-II (CaMKII), protein kinase C (PKC), phosphatidilinositol-3'-kinase (PI3-K), and protein kinase A (PKA) are also potentially involved with GMP and GUO effect. Such results suggest that GMP and GUO, and molecules released in GBPs-ACM promote the survival or maturation of primary cerebellar neurons or both via interaction with adenosine and glutamate receptors.

Os meningiomas correspondem aos tumores cerebrais primários benignos mais comuns, representando de 13% a 26 % dos tumoresintracranianos. Apresentam maior incidência no sexo feminino, sendo a exposição a radiação ionizante o principal fator de risco.São divididos em 3 grupos pela OMS, sendo o grau I o mais comum e menos agressivo. Possuem crescimento lento e são muitas vezessilenciosos, podendo cursar com alterações psiquiátricas quando localizados na fossa anterior com efeito compressivo sobre os lobosfrontais, porém esta manifestação é rara. Apresentamos o caso de uma paciente de 68 anos que apresentava sintomas psiquiátricosdesde 2012 e, após 6 anos, evoluiu com deterioração neurológica súbita, tendo o diagnóstico tardio de meningioma frontal gigante.Destacamos a importância da pesquisa de causa orgânica para manifestação psiquiátrica em determinado grupo de pessoas, nointuito de diagnóstico e tratamento precoce, melhorando sobrevida e qualidade de vida dos pacientes.

To evaluate the effect of cognitive and sociodemographic characteristics of healthcare and non-healthcare workers on their traumatic responses to the COVID-19 pandemic.Data were collected using an online survey between August-September 2020. The survey included the following scales: Beck Anxiety Inventory (BAI), Anxiety Sensitivity Index (ASI), and Impact of Event Scale-Revised (IES-R). Traumatic responses were categorized into three types: avoidance (IES-R_A), intrusion (IES-R_I), and hyperarousal (IES-R_H).The study included a total of 672 participants, comprised of 399 (59.4%) men, and 273 (40.6%) women with a mean age of 39.25 ± 933 years. The results indicated that women had higher IES-R_I (Female gender and a positive history of psychiatric diseases were found to lead to an increase in the frequency of all three traumatic responses to COVID-19. Contrary to expectation, being a healthcare worker was not found as a factor facilitating trauma response formation in our study.Évaluer l’effet des caractéristiques cognitives et sociodémographiques des travailleurs de la santé et des travailleurs non médicaux sur leurs réponses traumatiques à la pandémie de COVID-19.Les données ont été recueillies à l’aide d’une enquête en ligne entre août et septembre 2020. L’enquête comprenait les échelles suivantes : Inventaire d’anxiété de Beck (BAI), Indice de sensibilité à l’anxiété (ASI) et Échelle d’impact de l’événement - révisée (IES-R). Les réponses traumatiques ont été classées en trois types : évitement (IES-R_A), intrusion (IES-R_I) et hyperexcitation (IES-R_H).L’étude a inclus un total de 672 participants, composés de 399 (59,4 %) hommes et 273 (40,6 %) femmes avec un âge moyen de 39,25 ± 933 ans. Les résultats ont indiqué que les femmes avaient des scores IES-R_I (On a constaté une augmentation de la fréquence de trois réponses traumatiques à la COVID-19 chez les femmes et celles ayant des antécédents positifs de maladies psychiatriques. Contrairement aux attentes, le fait d’être un travailleur de la santé ne s’est pas avéré être un facteur facilitant la formation de réponses traumatiques dans notre étude.

Prostate cancer (PCa) is a common type of cancer in western countries and prominent cause of mortality in men. The aim of the study was to analyze circulating miRNAs as biomarkers in the sera of healthy individuals and prostate cancer cases without biopsy.Twenty prostate cases, age (mean and range) 61,4±12.1 (45-73), and twenty healthy men, age 59,3±11.2 (44-70) were included to the study. The mean and range of prostate spesific antigen (PSA) in cancer cases and healthy individuals were 6.79±2.84 ng/ml (2.25-14.7) and 3.8±2.2 ng/ml (1.3-7.8) respectively.Seven miRNAs including two internal controls (Let7c, miR125b, miR141, miR145, miR 155, miR181 ve miR192) were evaluated in two groups. The level of miR141 was significantly lower in PCa cases than healthy individuals (p=0,004), and miR155 was significantly higher (p=0,005) in PCa cases. Both miRNAs were explored sensitive and spesific in the ROC analysis. Tumor mass were found to be associated with the level of miR-125b and miR-145. Conclusion; validation studies are required in wider patient groups in the subject of tumor effect and miRNA biomarkers in prostate cancer.

To evaluate the maternal serum ischemia-modified albumin (IMA) concentration as an oxidative stress biomarker in pregnancies complicated by preterm pre-labor rupture of membranes (PPROM) without maternal clinical infection and compare these results with healthy pregnancies.The present cohort study included 40 pregnancies complicated by PPROM and 49 similar gestational age healthy pregnancies in the third trimester of gestation. Maternal venous blood specimens were obtained at the day of first diagnosis. Maternal serum IMA level was assayed with an Albumin Cobalt Binding test. The subjects were followed up until delivery and perinatal outcomes were recorded.The maternal serum IMA concentrations were significantly higher in the study group (0.56 ± 0.05 absorbance units) as compared to controls (0.54 ± 0.03 absorbance units) (p = 0.020). The maternal serum IMA concentrations were not significantly correlated with the initial maternal white blood cell count (r: 0.118, p = 0.269) and C-reactive protein levels (r: 0.066, p = 0.541). The maternal serum IMA concentrations were negatively correlated with gestational age at delivery (r: -0.248, p = 0.019), birthweight (r: -0.247, p = 0.020) and Apgar scores (r: -0.200, p = 0.049; r: -0.245, p = 0.020). The threshold value of maternal serum IMA concentration above 0.55 absorbance units indicated the pregnancy complicated by PPROM by 57.5% sensitivity and 57.1% specificity (Area under curve 0.613, confidence interval 0.50-0.73).The current study supported for the first time that there is an association between increased maternal serum IMA levels and the development of PPROM in the third trimester of gestation without maternal clinical infection. Elevated maternal serum IMA levels may alert the obstetrician about poor ongoing perinatal outcomes in the early phase of PPROM before increased maternal C-reactive protein and white blood cell count.

The main objective of this study was to analyze the productivity and efficiency of concrete casting using a mini crane with a capacity of 200 kg as designed by the researcher, compared to using the manual method. The method used in this study is the assembly of mini cranes with a capacity of 200 kg, then field or work tests are carried out and accompanied by observations and subsequent data analysis. The results obtained in this study are that the engine from motorcycle waste can lift loads up to 100 kg even though its ability can lift 200 kg, working for 10 hours only consumes 3 liters of gasoline. The results of the analysis are that using a mini-crane is more profitable compared to the manual method. The authenticity of this research is that mini-cranes are designed and made by researchers with style and models adapted to the conditions of small towns and inland areas in Indonesia, made based on Appropriate Technology not using imported materials and prioritizing scrap metal waste which is the authenticity of this research , thus our research has the principle of producing innovations and works that are useful for human life.

Background and Objective: Cisplatin is an antineoplastic, platinum derivative used in the treatment of various cancers. Mucositis is a notable side effect of cisplatin treatment. Rutin (vitamin P1) is a drug with antioxidant, anticancer, antidiabetic and antimicrobial properties. The aim of this study was to biochemically, histopathologically and immunohistochemically investigate the effect of rutin on cisplatin-induced mucositis of the small intestine (jejunum) in rats. Materials and Methods: Twenty four rats were divided into four groups with six animals in each group: healthy group (HG), cisplatin-only group (CCG), 50 mg kg–1 of rutin plus cisplatin group (RG-50) and 100 mg kg–1 of rutin plus cisplatin group (RG-100). Rutin or distilled water were administered via an oral gavage. One hour after the administration of rutin or distilled water, the CCG, RG-50 and RG-100 were intraperitoneally injected with 5 mg kg–1 of cisplatin once every 2 for 8 days. At the end of this period, all the animals were sacrificed using a high-dose anesthetic and their small intestines (jejunum) were removed for biochemical and histopathological procedures. Differences between the groups were analyzed using a one-way analysis of variance, followed by Dunnett’s multiple comparisons test. Results: The levels of oxidants increased in all the cisplatin-treated groups, whereas those of antioxidants decreased. Rutin administered at a dose of 100 but not 50 mg kg–1 reduced oxidant levels and increased antioxidant levels to those of healthy tissues. Histopathologically, tissue damage was observed in jejunal tissue of the rats administered only cisplatin, whereas treatment with 100 mg kg–1 of rutin prevented cisplatin-induced histopathological damage. In the group administered 50 mg kg–1 of rutin, jejunal tissue showed a near-normal appearance, except for mildly dilated congested blood vessels. As shown immunohistochemically, rutin prevented cisplatin-induced jejunal damage more effectively when administered at a dose of 100 mg kg–1 than 50 mg kg–1. Conclusion: Rutin may be useful in the prevention of cisplatin-induced jejunal mucositis.

The neonatal exposure to general anesthetics has been associated with neuronal apoptosis and dendritic spines morphologic changes in the developing brain. Ketamine, a noncompetitive N-methyl-D-aspartate (NMDA) receptor antagonist, is widely used in pediatric patients to induce general anesthesia, analgesia, and perioperative sedation. In the present study, we investigated short- and long-term effects of a single ketamine (20 mg/kg, s.c.) neonatal exposure at postnatal day 7 in rats on the hippocampal and frontal cortical cellular viability. Additionally, putative neurochemical alterations and neurobehavioral impairments were evaluated in the adulthood. Ketamine neonatal administration selectively decreased cellular viability in the hippocampus, but not in the frontal cortex, 24 h after the treatment. Interestingly, a single ketamine neonatal exposure prevented the vulnerability to glutamate-induced neurotoxicity in the frontal cortex of adult rats. No short- or long-term damage to cellular membranes, as an indicative of cell death, was observed in hippocampal or cortical slices. However, ketamine induced a long-term increase in hippocampal glutamate uptake. Regarding behavioral analysis, neonatal ketamine exposure did not alter locomotor activity and anxiety-related parameters evaluated in the open-field test. However, ketamine administration disrupted the hippocampal-dependent object recognition ability of adult rats, while improved the motor coordination addressed on the rotarod. These findings indicate that a single neonatal ketamine exposure induces a short-term reduction in the hippocampal, but not in cortical, cellular viability, and long-term alterations in hippocampal glutamate transport, improvement on motor performance, and short-term recognition memory impairment.

The purpose of this study was to identify possible delays between the suspected diagnosis, the definitive diagnosis and the beginning of treatment in patients with cervical cancer treated by the Unified Health System in a reference center for cancer treatment in southern Brazil. This is a retrospective descriptive study of a series of 222 cervical cancer cases referred for treatment between August 2003 and July 2013. Data were collected from medical records, including sociodemographic, clinical and time intervals between suspicion, diagnosis and the beginning of treatment. Among the 95 patients who started the clinical picture with an abnormal Pap smear, only 27.4% performed a biopsy within 60 days of the examination. Only 19.3% of the 57 patients who started the clinical symptoms of the disease performed the biopsy within 60 days after the onset of symptoms. From the total number of patients, only 37.8% started treatment within 60 days after the biopsy result, and 15.3% took more than 180 days to start treatment. It was concluded that there was a delay between the suspected diagnosis and the biopsy, as well as the beginning of the treatment. Strategies to reduce the time for diagnosis and treatment of the disease should be prioritized.

Amaç: Bu çalışmada, Obsesif Kompülsif Bozukluğu (OKB) olan bireylerde örtük bellek performansı ile bilişsel süreçlerin ilişkisinin değerlendirilmesi amaçlandı.Materyal ve Metot: Bu çalışma; Yüzüncü Yıl Üniversitesi Tıp Fakültesi Hastanesi Psikiyatri Polikliniğine başvuran; 30 Obsesif Kompülsif Bozukluk (OKB) tanılı hasta ve rastgele seçilen 40 kişilik sağlıklı kontrol grubunda örtük bellek performansı ile bilişsel süreçler değerlendirildi. Her iki grup için; örtük bellek performansını değerlendiren Kelime Kökü Tamamlama Testi (KKTT) ve Sosyo-demografik bilgi formu uygulandı. Hasta grubuna ise bu teste ilave olarak, Yale– Brown Obsesyon Kompulsiyon Ölçeği (Y-BOCS), Boyutsal Yale- Brown Obsesyon Kompulsiyon Ölçeği (DY-BOCS), Obsesyonel İnanışlar Ölçeği (OİÖ), Düşünce Eylem Kaynaşması Ölçeği (TAF) ve Üst Biliş Ölçeği (ÜBÖ) uygulandı.Bulgular: Yapılan değerlendirme neticesinde; Y-BOCS ve bilişsel parametrelerin (TAF-OİÖ-ÜBÖ) puanlarındaki artışın KKTT performansını ters yönde etkilediği saptandı. Ortalama KKTT skoru hasta grubunda 7,63±3,05, kontrol grubunda 6,30±2,09 idi (p=0,034). Örtük bellek performansını en fazla etkileyen bilişsel parametre ise TAF parametresi idi.Sonuç: OKB’da, hastalığın şiddeti arttıkça örtük bellek performansı olumsuz etkilediği saptandı.

35. Evaluation of an Extended Stroke Rehabilitation Service (EXTRAS)

- Abstract
Background and Purpose— There is limited evidence to guide rehabilitation to meet the longer term needs of stroke survivors. The clinical effectiveness and cost-effectiveness of an extended stroke rehabilitation service (EXTRAS) provided following early supported discharge were determined. Methods— EXTRAS was a pragmatic parallel-group observer-blind randomized controlled trial involving 19 UK centers. Patients with stroke were individually randomized to receive EXTRAS or usual care at discharge from early supported discharge. Five EXTRAS reviews were provided by an early supported discharge team member between one and 18 months, usually by telephone. Reviews consisted of a semi-structured interview assessing progress, rehabilitation needs, and service provision, with goal setting and action planning. The primary outcome was performance in extended activities of daily living (Nottingham EADL Scale) at 24 months post-randomization. The Nottingham EADL Scale is scored 0 to 66, with higher scores indicating better performance in these activities. Cost-effectiveness was estimated using resource utilization costs and Quality Adjusted Life Years. Analyses were intention to treat. Results— Between January 9, 2013 and October 26, 2015, 573 participants were randomized (EXTRAS, n=285; usual care, n=288). Mean 24 month Nottingham EADL Scale scores were EXTRAS (n=219) 40.0 (SD 18.1) and usual care (n=231) 37.2 (SD 18.5) giving an adjusted mean difference of 1.8 (95% CI, –0.7 to 4.2). 1155/1338 (86%) of expected EXTRAS reviews were undertaken. Over 24 months, the mean cost of resource utilization was lower in the intervention group: –£311 (–$450 [95% CI, −£3292 to £2787; −$4764 to $4033]). EXTRAS provided more Quality Adjusted Life Years (0.07 [95% CI, 0.01 to 0.12]). At current conventional thresholds of willingness to pay (£20 000 [$28 940] per Quality Adjusted Life Years), there was a 90% chance that EXTRAS could be considered cost-effective. Conclusions— EXTRAS did not significantly improve stroke survivors’ performance in extended activities of daily living. However, given the impact on costs and Quality Adjusted Life Years, EXTRAS may be an affordable addition to improve stroke care. Clinical Trial Registration— URL: www.isrctn.com. Folic acid (folate) is a vitamin of the B-complex group crucial for neurological function. Considering that excitotoxicity and cell death induced by glutamate are involved in many disorders, the potential protective effect of folic acid on glutamate-induced cell damage in rat hippocampal slices and the possible intracellular signaling pathway involved in such effect were investigated. The treatment of hippocampal slices with folic acid (100 μM) significantly abrogated glutamate (1 mM)-induced reduction of cell viability measured by MTT reduction assay and inhibited glutamate-induced D-[3H]-aspartate release. To investigate the putative intracellular signaling pathways implicated in the protective effect of folic acid, we used a PI3K inhibitor, LY294002, which abolished the protective effects of folic acid against glutamate-induced cell damage and D-[3H] aspartate release. Moreover, hippocampal slices incubated with folic acid alone for 30 min presented increased phosphorylation of GSK-3β at Ser9, indicating an inhibition of the activity of this enzyme. Furthermore, folic acid in the presence of glutamate insult in hippocampal slices maintained for an additional period of 6 h in fresh culture medium without glutamate and/or folic acid induced phosphorylation of GSK-3β and β-catenin expression. In addition, glutamate-treated hippocampal slices showed increased iNOS expression that was reversed by folic acid. In conclusion, the results of this study show that the protective effect of folic acid against glutamate-induced excitotoxicity may involve the modulation of PI3K/GSK-3β/β-catenin pathway and iNOS inhibition.

Atorvastatin is a cholesterol-lowering statin that has been shown to exert several pleiotropic effects in the nervous system as a neuroprotective and antidepressant-like agent. Antidepressant-like effect of atorvastatin in mice is mediated by glutamatergic and serotoninergic receptors, although the precise intracellular signaling pathways involved are unknown. PI3K/Akt/GSK-3β/mTOR signaling pathway has been associated to neurobiology of depression and seems to be modulated by some pharmacological antidepressant strategies. The present study investigated the participation of the PI3K/Akt/GSK-3β/mTOR signaling pathway in the antidepressant-like effect of an acute atorvastatin treatment in mice. Atorvastatin sub-effective (0.01 mg/kg) or effective (0.1 mg/kg) doses in the tail suspension test (TST) was administered orally alone or in combination with PI3K, GSK-3β or mTOR inhibitors. The administration of PI3K inhibitor, LY294002 (10 nmol/site, i.c.v) completely prevented the antidepressant-like effect of atorvastatin (0.1 mg/kg, p.o.). The participation of GSK-3β in the antidepressant-like effect of atorvastatin was demonstrated by co-administration of a sub-effective dose of atorvastatin (0.01 mg/kg, p.o.) with AR-A014418 (0.01 μg/site, i.c.v., a selective GSK-3β inhibitor) or with lithium chloride (10 mg/kg, p.o., a non-selective GSK-3β inhibitor). The mTOR inhibitor, rapamycin (0.2 nmol/site, i.c.v.) was also able to prevent atorvastatin (0.1 mg/kg, p.o.) antidepressant-like effect. These behavioral findings were supported by neurochemical observations, as atorvastatin treatment increased the immunocontent of the phosphorylated isoforms of Akt, GSK-3β and mTOR in the hippocampus of mice. Taken together, our results suggest an involvement of the PI3K/Akt/GSK-3β/mTOR signaling pathway in the antidepressant-like effect of atorvastatin in mice.

Intracerebroventricular (icv) amyloid-beta (Aβ)1–40 infusion to mice has been demonstrated to cause neurotoxicty and depressive-like behavior and it can be used to evaluate antidepressant and neuroprotective effect of drugs. Atorvastatin is a widely used statin that has demonstrated antidepressant-like effect in predictable animal behavioral models and neuroprotective effect against Aβ1–40 infusion. The purpose of this study was to determine the effect of in vivo atorvastatin treatment against Aβ1–40-induced changes in mood-related behaviors and biochemical parameters in ex vivo hippocampal slices from mice. Atorvastatin treatment (10 mg/kg, p.o., once a day for seven consecutive days) abolished depressive-like and anhedonic-like behaviors induced by Aβ1–40 (400 pmol/site, icv) infusion. Aβ1–40-induced hippocampal cell damage was reversed by atorvastatin treatment. Aβ1–40 infusion decreased glutamate uptake in hippocampal slices, and atorvastatin did not altered it. Glutamine synthetase activity was not altered by any treatment. Atorvastatin also increased hippocampal mature brain-derived neurotrophic factor (mBDNF)/precursor BDNF (proBDNF) ratio, suggesting an increase of proBDNF to mBDNF cleavage. Accordingly, increased tissue-type plasminogen activator (tPA) and p11 genic expression were observed in hippocampus of atorvastatin-treated mice. Atorvastatin displays antidepressant-like and neuroprotective effects against Aβ1–40-induced toxicity, and these effects may involve tPA- and p11-mediated cleavage of proBDNF to mBDNF.

Guanosine, the endogenous guanine nucleoside, prevents cellular death induced by ischemic events and is a promising neuroprotective agent. During an ischemic event, nitric oxide has been reported to either cause or prevent cell death. Our aim was to evaluate the neuroprotective effects of guanosine against oxidative damage in hippocampal slices subjected to an in vitro ischemia model, the oxygen/glucose deprivation (OGD) protocol. We also assessed the participation of nitric oxide synthase (NOS) enzymes activity on the neuroprotection promoted by guanosine. Here, we showed that guanosine prevented the increase in ROS, nitric oxide, and peroxynitrite production induced by OGD. Moreover, guanosine prevented the loss of mitochondrial membrane potential in hippocampal slices subjected to OGD. Guanosine did not present an antioxidant effect per se. The protective effects of guanosine were mimicked by inhibition of neuronal NOS, but not of inducible NOS. The neuroprotective effect of guanosine may involve activation of cellular mechanisms that prevent the increase in nitric oxide production, possibly via neuronal NOS.

Amyloid-beta (Aβ) peptides are the major neuropathological hallmarks related with Alzheimer's disease (AD). Aβ peptides trigger several biochemical mechanisms of neurotoxicity, including neuroinflammation and glutamatergic neurotransmission impairment. Guanosine is the endogenous guanine-derived nucleoside that modulates the glutamatergic system and the cellular redox status, thus acting as a neuroprotective agent. Here, we investigated the putative neuroprotective effect of guanosine in an AD-like mouse model. Adult mice received a single intracerebroventricular injection of Aβ1-40 (400 pmol/site) or vehicle and then were treated immediately, 3 h later, and once a day during the subsequent 14 days with guanosine (8 mg/kg, intraperitoneally). Aβ1-40 or guanosine did not alter mouse locomotor activity and anxiety-related behaviors. Aβ1-40-treated mice displayed short-term memory deficit in the object location task that was prevented by guanosine. Guanosine prevented the Aβ1-40-induced increase in latency to grooming in the splash test, an indicative of anhedonia. Aβ1-40 increased Na+-independent glutamate uptake in ex vivo hippocampal slices, and guanosine reversed it to control levels. The repeated administration of guanosine increased hippocampal GDP levels, which was not observed in the group treated with Aβ plus guanosine. Aβ1-40 induced an increase in hippocampal ADP levels. Aβ1-40 decreased GFAP expression in the hippocampal CA1 region, an effect not modified by guanosine. No differences were observed concerning synaptophysin and NeuN immunolabeling. Together, these results show that guanosine prevents memory deficit and anhedonic-like behavior induced by Aβ1-40 that seem to be linked to glutamate transport unbalance and alterations on purine and metabolite levels in mouse hippocampus.

Glial cells are involved in multiple cerebral functions that profoundly influence brain tissue viability during ischemia, and astrocytes are the main source of extracellular purines as adenosine and guanosine. The endogenous guanine-based nucleoside guanosine is a neuromodulator implicated in important processes in the brain, such as modulation of glutamatergic transmission and protection against oxidative and inflammatory damage. We evaluated if the neuroprotective effect of guanosine is also observed in cultured cortical astrocytes subjected to oxygen/glucose deprivation (OGD) and reoxygenation. We also assessed the involvement of A(1) and A(2A) adenosine receptors and phosphatidylinositol-3 kinase (PI3K), MAPK, and protein kinase C (PKC) signaling pathways on the guanosine effects. OGD/reoxygenation decreased cell viability and glutamate uptake and increased reactive oxygen species (ROS) production in cultured astrocytes. Guanosine treatment prevented these OGD-induced damaging effects. Dipropyl-cyclopentyl-xanthine (an adenosine A(1) receptor antagonist) and 4-[2-[[6-amino-9-(N-ethyl-β-d-ribofuranuronamidosyl)-9H-purin-2-yl]amino]ethyl] benzenepropanoic acid hydrochloride (an adenosine A(2A) receptor agonist) abolished guanosine-induced protective effects on ROS production, glutamate uptake, and cell viability. The PI3K pathway inhibitor 2-morpholin-4-yl-8-phenylchromen-4-one, the extracellular-signal regulated kinase kinase (MEK) inhibitor 2′-amino-3′-methoxyflavone, or the PKC inhibitor chelerythrine abolished the guanosine effect of preventing OGD-induced cells viability reduction. PI3K inhibition partially prevented the guanosine effect of reducing ROS production, whereas MEK and PKC inhibitions prevented the guanosine effect of restoring glutamate uptake. The total immunocontent of the main astrocytic glutamate transporter glutamate transporter-1 (GLT-1) was not altered by OGD and guanosine. However, MEK and PKC inhibitions also abolished the guanosine effect of increasing cell-surface expression of GLT-1 in astrocytes subjected to OGD. Then, guanosine prevents oxidative damage and stimulates astrocytic glutamate uptake during ischemic events via adenosine A(1) and A(2A) receptors and modulation of survival signaling pathways, contributing to microenvironment homeostasis that culminates in neuroprotection.

Syftet med studien var att synliggöra hur små barn interagerar med ett mikroskop-ägg i utforskandet av naturvetenskapliga fenomen. I förskolans uppdrag ingår att arbeta med natur-vetenskap och teknik i barnens vardag. Förskollärarens kompetens och deras förhållningssätt gentemot barnen är betydelsefull för att möjliggöra för små barns digitala utforskande. Tidigare forskning visar att utforskande handlar om att få pröva, undersöka och på ett lekfullt sätt ta reda på mer om något. Det teoretiska ramverk som legat till grund för studien är social construction of technology, SCOT, vilket gett möjlighet att beskriva hur mening skapas i barns interaktion med olika tekniska artefakter. Studien genomfördes på en medelstor förskola i Luleå kommun med tre grupper barn i två-års ålder. Data samlades in med hjälp av filminspelning och ljudupptagning vid planerade aktiviteter. Bearbetningen har sketti en abduktiv analysprocess där en växelverkan mellan att härleda materialet till teoretiska utgångspunkter och en öppenhet för nya förklaringar och upp-täckter har varit vägledande. I resultatet framträder olika interaktionsramar som visar på vilket sätt barnen interagerar med den digitala teknologin utifrån tidigare erfarenheter och upplevelser. Resultatet visar även hur mikroskop-ägget på olika sätt kan möjliggöra för små barns naturvetenskapliga utforskande.

The Planetary Radio Interferometry and Doppler Experiment (PRIDE) is a technique that can enhance the sciencereturn of planetary missions. By shadow tracking the spacecraft signal using radio telescopes from VLBI networks,the PRIDE technique provides precise open-loop Doppler and near-field VLBI observables (Duev et al. 2012,Bocanegra-Bahamon et al. 2018a) to find the radial velocity of the spacecraft and its position in the plane of thesky. This information is not only important for navigation, but it can also be used for many science applications.One such case is the study of planetary atmospheres by means of radio occultation experiments.

Sömnbrist är ett ämne som givits allt mer uppmärksamhet i dagens samhälle. Sömnen har betydelse för människan i den bemärkelse att kroppen får återhämtning och hjärnan kan lagra och bearbeta minnen. Sjuksköterskor har i dagens samhälle en stor roll i det patientnära arbetet. Men vad händer med en sjuksköterska som har sömnbrist och hur påverkar detta hens arbete och hälsa? Syftet med detta arbete är att beskriva hur sjuksköterskor erfar sömnbrist, trötthet och fatigue samt hur det påverkar sjuksköterskans arbete och hälsa. En litteraturöversikt användes till arbetet för att få en överblick om det nuvarande kunskapsläget. Tio artiklar inkluderades i arbetet och analyserades. Studier visar på att sömnbrist, trötthet och fatigue är vanligt förekommande hos sjuksköterskor och studierna visade även på konsekvenser som uppstått till följd av sjuksköterskors trötthet. Dessa konsekvenser kan innebära att sjuksköterskor begår misstag i det patientnära arbetet, men tröttheten kan också gå ut över det privata livet. Detta innebär att sjuksköterskan kan utsätta sig för onödiga risker utanför arbetsplatsen i form av att köra bil medan hen är trött. Resultaten visar att det behövs mer forskning kring vilka orsaker som gör att sjuksköterskan lider av sömnbrist och därav upplever trötthet. Likaså studier som visar hur sömnbrist påverkar den vårdande vården vore värdefullt. En slutsats är att man bör lyfta frågan om sömnbrist och trötthet hos personalen för att på så sätt kunna se om rätt förutsättningar finns för en rimlig återhämtning.

47. Guanosine and GMP increase the number of granular cerebellar neurons in culture: dependence on adenosine A

The guanine-based purines (GBPs) have essential extracellular functions such as modulation of glutamatergic transmission and trophic effects on neurons and astrocytes. We previously showed that GBPs, such as guanosine-5′-monophosphate (GMP) or guanosine (GUO), promote the reorganization of extracellular matrix proteins in astrocytes, and increase the number of neurons in a neuron-astrocyte co-culture protocol. To delineate the molecular basis underlying these effects, we isolated cerebellar neurons in culture and treated them with a conditioned medium derived from astrocytes previously exposed to GUO or GMP (GBPs-ACM) or, directly, with GUO or GMP. Agreeing with the previous studies, there was an increase in the number of β-tubulin III-positive neurons in both conditions, compared with controls. Interestingly, the increase in the number of neurons in the neuronal cultures treated directly with GUO or GMP was more prominent, suggesting a direct interaction of GBPs on cerebellar neurons. To investigate this issue, we assessed the role of adenosine and glutamate receptors and related intracellular signaling pathways after GUO or GMP treatment. We found an involvement of A(2A) adenosine receptors, ionotropic glutamate N-methyl-D-aspartate (NMDA), and non-NMDA receptors in the increased number of cerebellar neurons. The signaling pathways extracellular-regulated kinase (ERK), calcium-calmodulin-dependent kinase-II (CaMKII), protein kinase C (PKC), phosphatidilinositol-3′-kinase (PI3-K), and protein kinase A (PKA) are also potentially involved with GMP and GUO effect. Such results suggest that GMP and GUO, and molecules released in GBPs-ACM promote the survival or maturation of primary cerebellar neurons or both via interaction with adenosine and glutamate receptors.

Guanosine (GUO) has been shown to act as a neuroprotective agent against glutamatergic excitotoxicity by increasing glutamate uptake and decreasing its release. In this study, a putative effect of GUO action on glutamate transporters activity modulation was assessed in hippocampal slices subjected to oxygen and glucose deprivation (OGD), an in vitro model of brain ischemia. Slices subjected to OGD showed increased excitatory amino acids release (measured by D-[(3)H]aspartate release) that was prevented in the presence of GUO (100 µM). The glutamate transporter blockers, DL-TBOA (10 µM), DHK (100 µM, selective inhibitor of GLT-1), and sulfasalazine (SAS, 250 µM, Xc(-) system inhibitor) decreased OGD-induced D-aspartate release. Interestingly, DHK or DL-TBOA blocked the decrease in glutamate release induced by GUO, whereas SAS did not modify the GUO effect. GUO protected hippocampal slices from cellular damage by modulation of glutamate transporters, however selective blockade of GLT-1 or Xc- system only did not affect this protective action of GUO. OGD decreased hippocampal glutamine synthetase (GS) activity and GUO recovered GS activity to control levels without altering the kinetic parameters of GS activity, thus suggesting GUO does not directly interact with GS. Additionally, the pharmacological inhibition of GS activity with methionine sulfoximine abolished the effect of GUO in reducing D-aspartate release and cellular damage evoked by OGD. Altogether, results in hippocampal slices subjected to OGD show that GUO counteracts the release of excitatory amino acids, stimulates the activity of GS, and decreases the cellular damage by modulation of glutamate transporters activity.

Stress and excessive glutamatergic neurotransmission have been implicated in the pathophysiology of depression. Therefore, this study was aimed at investigating the influence of zinc on depressive-like behavior induced by chronic unpredictable stress (CUS), on alterations in glutamate-induced toxicity and immunocontent of proteins involved in the control of glutamatergic neurotransmission in the hippocampus of mice. Mice were subjected to CUS procedure for 14 days. From the 8th to the 14th day, mice received zinc chloride (ZnCl2) (10 mg/kg) or fluoxetine (10 mg/kg, positive control) once a day by oral route. CUS caused a depressive-like behavior evidenced by the increased immobility time in the tail suspension test (TST), which was prevented by treatment with ZnCl2 or fluoxetine. Ex vivo exposure of hippocampal slices to glutamate (10 mM) resulted in a significant decrease on cell viability; however, neither CUS procedure nor drug treatments altered this reduction. No alterations in the immunocontents of GLT-1 and GFAP or p-Akt were observed in any experimental group. The ratio of p-Akt/AKT was also not altered in any group. However, Akt immunocontent was increased in stressed mice and in animals treated with ZnCl2 (stressed or non-stressed mice) and EAAC1 immunocontent was increased in stressed mice treated with ZnCl2, fluoxetine or vehicle and in non-stressed mice treated with ZnCl2 and fluoxetine. These findings indicate a robust effect of zinc in reversing behavioral alteration induced by CUS in mice, through a possible modulation of the glutamatergic neurotransmission, extending literature data regarding the mechanisms underlying its antidepressant-like action.
