PURPOSE Optimized strategies for risk classification are essential to tailor therapy for patients with biologically distinctive disease. Risk classification in pediatric acute myeloid leukemia (pAML) relies on detection of translocations and gene mutations. Long noncoding RNA (lncRNA) transcripts have been shown to associate with and mediate malignant phenotypes in acute myeloid leukemia (AML) but have not been comprehensively evaluated in pAML. METHODS To identify lncRNA transcripts associated with outcomes, we evaluated the annotated lncRNA landscape by transcript sequencing of 1,298 pediatric and 96 adult AML specimens. Upregulated lncRNAs identified in the pAML training set were used to establish a regularized Cox regression model of event-free survival (EFS), yielding a 37 lncRNA signature (lncScore). Discretized lncScores were correlated with initial and postinduction treatment outcomes using Cox proportional hazards models in validation sets. Predictive model performance was compared with standard stratification methods by concordance analysis. RESULTS Training set cases with positive lncScores had 5-year EFS and overall survival rates of 26.7% and 42.7%, respectively, compared with 56.9% and 76.3% with negative lncScores (hazard ratio, 2.48 and 3.16; P < .001). Pediatric validation cohorts and an adult AML group yielded comparable results in magnitude and significance. lncScore remained independently prognostic in multivariable models, including key factors used in preinduction and postinduction risk stratification. Subgroup analysis suggested that lncScores provide additional outcome information in heterogeneous subgroups currently classified as indeterminate risk. Concordance analysis showed that lncScore adds to overall classification accuracy with at least comparable predictive performance to current stratification methods that rely on multiple assays. CONCLUSION Inclusion of the lncScore enhances predictive power of traditional cytogenetic and mutation-defined stratification in pAML with potential, as a single assay, to replace these complex stratification schemes with comparable predictive accuracy.

Massive open online courses (MOOCs) , which offer open access and widespread interactive participation through the internet, are quickly becoming the preferred method for online and remote learning. Several MOOC platforms offer the service of course recommendation to users, to improve the learning experience of users. Despite the usefulness of this service, we consider that recommending courses to users directly may neglect their varying degrees of expertise. To mitigate this gap, we examine an interesting problem of concept recommendation in this paper, which can be viewed as recommending knowledge to users in a fine-grained way. We put forward a novel approach, termed HinCRec-RL, for C oncept Rec ommendation in MOOCs, which is based on H eterogeneous I nformation N etworks and R einforcement L earning . In particular, we propose to shape the problem of concept recommendation within a reinforcement learning framework to characterize the dynamic interaction between users and knowledge concepts in MOOCs. Furthermore, we propose to form the interactions among users, courses, videos, and concepts into a heterogeneous information network (HIN) to learn the semantic user representations better. We then employ an attentional graph neural network to represent the users in the HIN, based on meta-paths. Extensive experiments are conducted on a real-world dataset collected from a Chinese MOOC platform, XuetangX , to validate the efficacy of our proposed HinCRec-RL. Experimental results and analysis demonstrate that our proposed HinCRec-RL performs well when compared with several state-of-the-art models.

This research investigated the impacts of model prediction on the optimization of hybrid energy systems using a system consisting of solar panels, batteries, a proton exchange membrane fuel cell (PEMFC), and a chemical hydrogen generation system. A PEMFC has several advantages, such as low operating temperatures, fast response times, high power density, and environmental friendliness, and it can convert hydrogen into electricity. However, because hydrogen costs are an important consideration, the PEMFC is usually integrated with hybrid energy systems to guarantee system sustainability. Therefore, in this study, a whole-year household load and solar radiation data were applied to optimize the system components and power management, thereby reducing the system cost by 42.43% and improving system sustainability by 7.05%. The system responses showed that some hydrogen consumption might be saved if the solar and load profiles could be foreseen. Two prediction models were developed that could accurately forecast the radiation and load profiles. Next, a second-year dataset was employed to verify the effectiveness of the model prediction. The results showed that the system cost was reduced by 40.20% without model prediction and by 44.06% with model prediction compared to the original system settings. Finally, experiments to illustrate the feasibility of the hybrid energy system were conducted using prediction models. Based on the results, the model prediction was deemed effective in improving the performance of hybrid energy systems.

In order to identify more high order modal from a small amount of vibration response signal sampling, this paper proposes a method for identifying the OMA of sub-sampled vibration response signals based on compression sensing (CS) and fast independent component analysis (FastICA). Firstly, the method subsamples a large number of vibrational signals to retain the original key information of the signal; Secondly, the subsampled signal is restored by using the compressive sampling matching pursuit (CoSaMP) algorithm of the compression-aware reconstruction algorithm. Finally, the source signal is separated by FastICA to obtain the multi-order natural frequencies and modal mode shapes of the structure. According to Shannon’s theorem, if the analog signal is reproduced without distortion, the sampling frequency should be no less than 2 times the highest frequency in the analog signal spectrum. This method can reduce the amount of original signal acquisition and the requirements for the frequency of the sampled signal, and the recovery signal has a high similarity with the original signal and the natural frequency is not changed. It can effectively identify the higher-order modes of the structure on the basis of breaking through the Nyquist sampling frequency. Experimental results of the subsampled vibration response signal on the uniform steel cantilever beam show that the method can reconstruct the original signal from a small number of subsampled vibration response signals, and identify the high-order nature frequency and modal mode shape that break through the Nyquist sampling frequency.

BACKGROUND: Abdominal or back pain is a common symptom in pancreatic diseases. However, the role of pain in pancreatic neuroendocrine neoplasm (PNENs) has not been clarified. OBJECTIVE: In this study, we aimed to show the association between the pain and the grade of PNENs. METHODS: A total of 186 patients with pathologically confirmed PNENs were included in this study. Clinical features and histological or radiological findings (size, location, and vascular invasion and local organs invasion and distal metastasis) were collected. Logistic regression analyses were used to show the association between pain and grade of PNENs. Nomogram was developed based on associated factors to predict the higher grade of PNENs. Receiver operating characteristic (ROC) curve was used to evaluate the diagnostic performance of size and nomogram model. RESULTS: The prevalence of pain in the cohort was 30.6% (n= 57). The vascular invasion and G3 PNENs were more common in the pain group (P= 0.02, P< 0.01). The tumor size was larger and incident of higher grade of PNENs was higher in the pain group than the non-pain group (p< 0.01). Age, pain and size were independent risk factors for G2/G3 or G3 PNENs. The odds ratio was 3.03 (95% CI: 1.67–7.91) and 3.32 (95% CI: 1.42–7.79) for pain, respectively. The nomogram model was developed to predict the G2/G3 or G3 PNENs. The area under the curve (AUC) of the nomogram model was 0.84 (95% CI, 0.77–0.91) in predicting the G2/G3 PNENs, and was 0.84 (95% CI, 0.78–0.91) in predicting the G3 PNENs. CONCLUSION: Abdominal or back pain is associated with the grade of PNENs. The nomograms based on clinical features may be a powerful numerical tool for predicting the grade of PNENs.
