Liraglutide is a new generation lipopeptide drug used for the treatment of type II diabetes. In this work, we describe new approaches for its preparation fully by chemical methods. The key step of these strategies is the synthesis in solution of the Lys/γ-Glu building block, Fmoc-Lys-(Pal-γ-Glu-OtBu)-OH, in which Lys and Glu residues are linked through their side chains and γ-Glu is N(α) -palmitoylated. This dipeptide derivative is then inserted into the peptide sequence on solid phase. As liraglutide is obtained with great purity and high yield, our approach can be particularly attractive for an industrial production. We also report here the results of a circular dichroism conformational analysis in a membrane mimetic environment that offers new insights into the mechanism of action of liraglutide. Total syntheses and complete characterizations of singly substituted PheCN -based analogs of alamethicin AlaP, which is active on model and natural membranes, and the TM peptide, which inserts in a transmembrane orientation in lipid bilayers, are reported. The syntheses of the AlaP analogs were performed in solution, while those of TM and its analogs were carried out by solid phase. Using the cyanophenyl fluorescence and infrared (IR) absorption probe, an in-depth investigation of the self-association, membrane-interacting, permeabilizing, and orientation properties of these peptides were conducted. The aromatic residue incorporated induces only a negligible modification to the properties of the parent peptides. The PheCN IR absorption band was located between 2228 and 2230cm(-1) for all peptides, irrespective of the position of labeling. By contrast, as the width of this band varied significantly with the depth of probe insertion in the bilayer, it could represent a good marker of the PheCN position in phospholipid membranes. 2015 Wiley Periodicals, Inc. Short proline-rich peptides interact with SH3 domains, exhibiting little or no secondary structure before their binding to the cognate proteintargets. Under these conditions the binding process of a proline-rich peptide with the SH3 domain shows unfavorable binding entropy, likely resulting from a loss of rotational freedom on the formation of the PPII helix. With the aim of stabilizing the PPII helix conformation in SH3 binding motifs, in the previous years, we replaced the proline residues of the HPK1 proline-rich decapeptide, PPPLPPKPKF (P2), either with 4-R- (Fp) or with 4-S- (fp) fl uoro-L-proline at different i, i+3 positions. The interactions of the fluoroproline-peptides with the SH3 domain of cortactin protein were analyzed quantitatively by Non-Immobilized Ligand Interactions Assay by Circular Dichroism (NILIA-CD), whereas CD thermal transitions were measured to correlate their propensity to adopt PPII helix with their affi nity for SH3. Results show that although the introduction of the Fp residue stabilizes the PPII helix conformation of peptides in a position-dependent manner, the induction of a stable peptide conformation does not increase the ligand affinity towards the SH3 domain of cortactin. To explore the effect of electron-withdrawing substituent in the Pro residue, the Fp residues of P2 were replaced by the natural amino acid 4-R-hydroxy-proline (Hyp). Unexpectedly, NILIACD and CD thermal transitions results showed that the Hyp-containing peptides exhibit a stable conformation in aqueous buffer and Kd values lower than the corresponding Fp peptide-analogues. In particular, Hyp3 peptide containing Hyp residues at i, i+3 and i+6 positions adopts the greater percentage of PPII helix conformation over the entire studied temperature range and shows a Kd value comparable to that of the parent P2 peptide.

Peptaibols are peculiar peptides produced by fungi as weapons against other microorganisms. Previous studies showed that peptaibols are promising peptide-based drugs because they act against cell membranes rather than a specific target, thus lowering the possibility of the onset of multi-drug resistance, and they possess non-coded α-amino acid residues that confer proteolytic resistance. Trichogin GA IV (TG) is a short peptaibol displaying antimicrobial and cytotoxic activity. In the present work, we studied thirteen TG analogues, adopting a multidisciplinary approach. We showed that the cytotoxicity is tuneable by single amino-acids substitutions. Many analogues maintain the same level of non-selective cytotoxicity of TG and three analogues are completely non-toxic. Two promising lead compounds, characterized by the introduction of a positively charged unnatural amino-acid in the hydrophobic face of the helix, selectively kill T67 cancer cells without affecting healthy cells. To explain the determinants of the cytotoxicity, we investigated the structural parameters of the peptides, their cell-binding properties, cell localization, and dynamics in the membrane, as well as the cell membrane composition. We show that, while cytotoxicity is governed by the fine balance between the amphipathicity and hydrophobicity, the selectivity depends also on the expression of negatively charged phospholipids on the cell surface.
