Public decisions are typically related to large investments leaving long legacies. We should therefore strive for wide societal agreement regarding such decisions, which meet the diversity of preferences between stakeholders and over time. But if, how and why do stakeholder preferences change over time? In decision analysis, these questions received little attention. We explored them using three real-world public decision processes, based on Multi-Criteria Decision Analysis (MCDA). We used repeatedly elicited ranking of objectives over time. These were obtained during three to five moderated workshops we organised several months apart (total N = 200 questionnaires, and 100 stakeholders). We analysed individual and aggregated (group) preferences, their changes and potential drivers including demographic and experience variables. We also analysed the effect of preference evolution on the performance of decision-alternatives with MCDA over time. We found that stakeholder preferences often changed over time, both on an individual and group level. These changes did not systematically diminish over time, but some convergence of preferences was observed for stakeholders who repeatedly participated in workshops. High-ranking objectives were relatively stable and similar between stakeholders. While preference changes could not be explained by demographics and personal experiences, repeated interaction with the decision problem might play a role. Neither the observed disagreement between stakeholders, nor the preference changes over time affected the best and worst performing alternatives in our decision problems. Thus, despite changing stakeholder preferences over time, public decision-makers can contrive robust solutions to complex public decision problems in the present.

BACKGROUND: At present, more than 8000 sesquiterpene lactones have been isolated and described from natural sources, a significant part of which has cytotoxicity and antitumor activity. One of the practically available sesquiterpene lactones is arglabin, which, as a renewable material, is used for the synthesis of new compounds. The article presents data on the study of cytotoxicity and antitumor activity of the arglabin and its derivatives using molecular modeling methods and, in the experiment in vitro and in vivo. AIM: The aim of this work is to study the cytotoxicity and antitumor activity of new compounds based on the sesquiterpene lactone arglabin using molecular modeling and experimental pharmacology. METHODS: ChemDraw programs and a set of AutoDock programs were used for computer simulation. Molecular docking was carried out using the Maestro graphical interface of the Schrödinger Suite software package (Schrödinger, LLC, New York, NY, 2017). Docking modes standard precision and XP (extra precision) were used. In in vitro experiments, the antitumor activity of compound samples was studied in models of 60 human tumor cell lines, and clonogenic C6 rat glioma cells. The antitumor activity of the samples was studied in experiments in vivo on white outbred rats with transplanted tumors and was evaluated by the inhibition of tumor growth and the magnitude of the increase in average life expectancy. CONCLUSION: When studying the antitumor activity on 60 cell lines of tumor cells (NCI60), clonogenic cells of C6 rat glioma, a high antitumor activity of some arglabin derivatives was established. The connection between the structure of arglabin derivatives and their inhibitory effect on farnesyl protein transferase, topoisomerases -I and -II was studied.

In this paper we present an extension to the $\texttt{matryoshka}$ suite of neural-network-based emulators. The new editions have been developed to accelerate EFTofLSS analyses of galaxy power spectrum multipoles in redshift space. They are collectively referred to as the $\texttt{EFTEMU}$. We test the $\texttt{EFTEMU}$ at the power spectrum level and achieve a prediction accuracy of better than 1\% with BOSS-like bias parameters and counterterms on scales $0.001\ h\ \mathrm{Mpc}^{-1} \leq k \leq 0.19\ h\ \mathrm{Mpc}^{-1}$. We also run a series of mock full shape analyses to test the performance of the $\texttt{EFTEMU}$ when carrying out parameter inference. Through these mock analyses we verify that the $\texttt{EFTEMU}$ recovers the true cosmology within $1\sigma$ at several redshifts ($z=[0.38,0.51,0.61]$), and with several noise levels (the most stringent of which is Gaussian covariance associated with a volume of $5000^3 \ \mathrm{Mpc}^3 \ h^{-3}$). We compare the mock inference results from the $\texttt{EFTEMU}$ to those obtained with a fully analytic EFTofLSS model and again find no significant bias, whilst speeding up the inference by three orders of magnitude. In the strive for the climate-neutral and ultra-low emission vehicle powertrains of the future, synthetic fuels produced from renewable sources will play a major role. Polyoxymethylene dimethyl ethers (POMDME or “OME”) produced from renewable hydrogen are a very promising candidate for zero-impact emissions in future CI engines. To optimize the utilisation of these fuels in terms of efficiency, performance and emissions, it is not only necessary to adapt the combustion parameters, but especially to optimize the injection and mixture formation process. In the present work, the spray break-up behavior and mixture formation of OME fuel is investigated numerically in 3D CFD and validated against experimental data from optical measurements in a high pressure/high temperature chamber using Schlieren and Mie scattering. For comparison, the same operating points using conventional diesel fuel were measured in the optical chamber, and the CFD modeling was optimized based on these data. To model the spray-breakup phenomena reliably, the primary break-up model according to Fischer is used, taking into account the nozzle internal flow in a detailed calculation of the disperse droplet phase. As OME has not yet been investigated very intensively with respect to its chemico-physical properties, chemical analyses of the substance properties were carried out to capture the most important parameters correctly in the simulation. With this approach, the results of the optical spray measurement could be reproduced well by the numerical model for the cases studied here, laying the basis for further numerical studies of OME sprays, including real engine operation.

The development of highly active anti-retroviral therapy (HAART) has markedly prolonged the life expectancy of individuals with human immunodeficiency virus (HIV). The prevalence of age-related cardiovascular disease (CVD) and arrhythmias is therefore expected to increase among the HIV-positive population.We aimed to assess the trends in prevalence, and inpatient outcomes among patients with HIV and atrial fibrillation (AF).Using ICD-9-CM coding, we identified 38,252,858 HIV-negative and 31,224 HIV-positive encounters with AF from the National Inpatient Sample (NIS) database from January 2005 to September 2015. Trends in prevalence of HIV in AF patients, length and cost of hospital stay, and inpatient mortality, were determined.Among the 31,224 HIV-positive encounters, 78.6% were males. The median age was 56 years for HIV-positive patients and 78 years for HIV-negative patients. Black patients were markedly overrepresented among HIV-positive as compared to HIV-negative hospitalisations (48.6The prevalence of HIV among patients with AF increased between 2005 and 2015. As compared to HIV-negative individuals with AF, a diagnosis of HIV was associated with increased inpatient mortality, length of stay, and cost of care. Future research on the underlying mechanisms of these findings is warranted to inform the treatment of AF in patients with HIV.

The diprovocims, a new class of toll-like receptor (TLR) agonists, bear no similarity to prior TLR agonists, act through a well-defined mechanism (TLR1/TLR2 agonist), exhibit exquisite structure-activity relationships, and display in vivo adjuvant activity. They possess potent and efficacious agonist activity toward human TLR1/TLR2 but modest agonism toward the murine receptor. A manner by which diprovocims can be functionalized without impacting hTLR1/TLR2 activity is detailed, permitting future linkage to antigenic, targeting, or delivery moieties. Improvements in both potency and its low efficacy in the murine system were also achieved, permitting more effective use in animal models while maintaining the hTLR1/TLR2 activity. The prototypical member diprovocim-X exhibits the excellent potency/efficacy of diprovocim-1 in human cells, displays substantially improved potency/efficacy in mouse macrophages, and serves as an adjuvant in mice when coadministered with a nonimmunogenic antigen, indicating stimulation of the adaptive as well as innate immune response.

We use angular clustering of luminous red galaxies from the Dark Energy Spectroscopic Instrument (DESI) imaging surveys to constrain the local primordial non-Gaussianity parameter fNL. Our sample comprises over 12 million targets, covering 14,000 square degrees of the sky, with redshifts in the range 0.2< z < 1.35. We identify Galactic extinction, survey depth, and astronomical seeing as the primary sources of systematic error, and employ linear regression and artificial neural networks to alleviate non-cosmological excess clustering on large scales. Our methods are tested against log-normal simulations with and without fNL and systematics, showing superior performance of the neural network treatment in reducing remaining systematics. Assuming the universality relation, we find fNL $= 47^{+14(+29)}_{-11(-22)}$ at 68\%(95\%) confidence. With a more aggressive treatment, including regression against the full set of imaging maps, our maximum likelihood value shifts slightly to fNL$ \sim 50$ and the uncertainty on fNL increases due to the removal of large-scale clustering information. We apply a series of robustness tests (e.g., cuts on imaging, declination, or scales used) that show consistency in the obtained constraints. Despite extensive efforts to mitigate systematics, our measurements indicate fNL > 0 with a 99.9 percent confidence level. This outcome raises concerns as it could be attributed to unforeseen systematics, including calibration errors or uncertainties associated with low-\ell systematics in the extinction template. Alternatively, it could suggest a scale-dependent fNL model--causing significant non-Gaussianity around large-scale structure while leaving cosmic microwave background scales unaffected. Our results encourage further studies of fNL with DESI spectroscopic samples, where the inclusion of 3D clustering modes should help separate imaging systematics., 19 pages, 15 figures, 6 tables (Appendix excluded). Successfully replacing meat with plant-based options will require not only replicating the texture of muscular fibres, but also imitating the taste, aroma, and juiciness of meat as closely and realistically as possible. This study examines the impact of rapeseed oil on the textural properties of meat analogues. Pea protein and soy protein are chosen as model systems to assess the effect of rapeseed oil. Optical, mechanical, and rheological characterisation tests are conducted to investigate the oil droplet distribution, the gel strength, Young’s modulus, and the length of the LVE region. The hypothesis is that oil droplets will act as active fillers in the protein matrix, and thus, diminish the strength of the protein gel network. The results of this study show that rapeseed oil droplets act as inactive fillers, as they are not bound to both examined protein matrices. Soy protein extrudates display minimal changes, while pea protein extrudates are significantly affected by the addition of oil. For example, oil addition decreased the G′ in the LVE region of pea protein meat analogues by 50%, while soy protein samples showed no significant changes. Despite the similar interfacial activities of the investigated proteins, the decreased encapsulation efficiency of pea protein was confirmed by the higher amount of extracted oil from the respective extrudates.

We present results from a set of high-fidelity simulated lightcones for the DESI One-Percent Survey, created from the Uchuu simulation. This 8 (Gpc/h)^3 N-body simulation comprises 2.1 trillion particles and provides high-resolution dark matter (sub)haloes in the framework of the Planck base-LCDM cosmology. Employing the subhalo abundance matching (SHAM) technique, we populate the Uchuu (sub)haloes with all four DESI tracers (BGS, LRG, ELG and QSO) to z = 2.1. Our method accounts for redshift evolution as well as the clustering dependence on luminosity and stellar mass. The two-point clustering statistics of the DESI One-Percent Survey align reasonably well with our predictions from Uchuu across scales ranging from 0.1 Mpc/h to 100 Mpc/h. Some discrepancies arise due to cosmic variance, incompleteness in the massive end of the stellar mass function, and a simplified galaxy-halo connection model. We find that the Uchuu BGS and LRG samples are adequately described using the standard 5-parameter halo occupation distribution model, while the ELGs and QSOs show agreement with an adopted Gaussian distribution for central halos with a power law for satellites. We observe a fair agreement in the large-scale bias measurements between data and mock samples, although the data exhibits smaller bias values, likely due to cosmic variance. The bias dependence on absolute magnitude, stellar mass and redshift aligns with that of previous surveys. These results improve simulated lightcone construction from cosmological models and enhance our understanding of the galaxy-halo connection, with pivotal insights from the first DESI data for the success of the final survey., 23 pages, 15 figures, 5 tables, submitted to MNRAS. The Dark Energy Spectroscopic Instrument (DESI) completed its five-month Survey Validation in May 2021. Spectra of stellar and extragalactic targets from Survey Validation constitute the first major data sample from the DESI survey. This paper describes the public release of those spectra, the catalogs of derived properties, and the intermediate data products. In total, the public release includes good-quality spectral information from 466,447 objects targeted as part of the Milky Way Survey, 428,758 as part of the Bright Galaxy Survey, 227,318 as part of the Luminous Red Galaxy sample, 437,664 as part of the Emission Line Galaxy sample, and 76,079 as part of the Quasar sample. In addition, the release includes spectral information from 137,148 objects that expand the scope beyond the primary samples as part of a series of secondary programs. We report a new test of modified gravity theories using the large-scale structure of the Universe. This paper is the first attempt to (1) apply a joint analysis of the anisotropic components of galaxy two- and three-point correlation functions (2 and 3PCFs) to actual galaxy data and (2) constrain the nonlinear effects of degenerate higher-order scalar-tensor (DHOST) theories on cosmological scales. Applying this analysis to the Baryon Oscillation Spectroscopic Survey (BOSS) data release 12, we obtain the lower bounds of $-1.655 < \xi_{\rm t}$ and $-0.504 < \xi_{\rm s}$ at the $95\%$ confidence level on the parameters characterising the time evolution of the tidal and shift terms of the second-order velocity field. These constraints are consistent with GR predictions of $\xi_{\rm t}=15/1144$ and $\xi_{\rm s}=0$. Moreover, they represent a $35$-fold and $20$-fold improvement, respectively, over the joint analysis with only the isotropic 3PCF. We ensure the validity of our results by investigating various quantities, including theoretical models of the 3PCF, window function corrections, cumulative ${\rm S/N}$, Fisher matrices, and statistical scattering effects of mock simulation data. We also find statistically significant discrepancies between the BOSS data and the Patchy mocks for the 3PCF measurement. Microglia play a role in the pathogenesis of many retinal diseases. Fundus spots in mice often correlate with the accumulation of activated subretinal microglia. Here we use a semiquantitative fundus spot scoring scale in combination with an unbiased, state-of-the-science forward genetics pipeline to identify causative associations between chemically induced mutations and fundus spot phenotypes. Among several associations, we focus on a missense mutation in Lipe linked to an increase in yellow fundus spots in C57BL/6J mice. Lipe−/− mice generated using CRISPR-Cas9 technology are found to develop accumulation of subretinal microglia, a retinal degeneration with decreased visual function, and an abnormal retinal lipid profile. We establish an indispensable role of Lipe in retinal/RPE lipid homeostasis and retinal health. Further studies using this new model will be aimed at determining how lipid dysregulation results in the activation of subretinal microglia and whether these microglia also play a role in the subsequent retinal degeneration.

In the Cerrado, the palm swamps ( veredas ) are characterized by being humid and stable environments that lead to the formation of Histosols ( Organossolos ). and soils with surface horizons of organic constitution, which are fragile and sensitive to anthropic action. This study aimed to evaluate the impact of anthropization (recurrent forest fires and livestock farming) on the chemical, physical and morphological properties of soils in two palm swamps in the Environmental Preservation Area (EPA) of Pandeiros River, Minas Gerais, namely: Água Doce, in preserved condition, and Taboa, in anthropized condition. Four soil profiles were morphologically described, two profiles in each palm swamp, with subsequent chemical and physical analyses, calculations of organic carbon stock and identification of the origin of organic matter. The results were analyzed using descriptive statistics and Pearson’s correlation coefficient. Soil morphological properties were influenced by vegetation cover, drainage and anthropization conditions. As for the physical and chemical properties, adequate values were observed in the preserved palm swamp, including lower bulk density values and higher cation contents. Anthropic actions in the anthropized palm swamp caused degradation of soils, revealed by subsidence, reduction in organic carbon content, increase in bulk density and decrease in fertility. Changes promoted in the soils of the palm swamps compromise ecosystem services, indicating that actions at either local or governmental level should be stimulated for the preservation and conservation of these environments.

Mountainous watersheds are characterized by variability in functional traits, including vegetation, topography, geology, and geomorphology, which together determine nitrogen (N) retention, and release. Coal Creek and East River are two contrasting catchments within the Upper Colorado River Basin that differ markedly in total nitrate (NO3-) export. The East River has a diverse vegetation cover, sinuous floodplains, and is underlain by N-rich marine shale, resulting in a three to twelve times greater total NO3- export relative to the conifer-dominated Coal Creek. While this can partly be explained by the larger size of the East River, the distinct watershed traits of these two catchments imply different mechanisms controlling the aggregate N-export signal. A causality analysis shows biogenic and geogenic processes were critical in determining NO3- export from the East River catchment. Stable isotope ratios of NO3- (δ15NNO3 and δ18ONO3) show the East River catchment is a strong hotspot for biogeochemical processing of NO3- at the soil-saprolite interface and within the floodplain prior to export. By contrast, the conifer-dominated Coal Creek retained nearly all (~97 %) atmospherically-deposited NO3-, and its export was controlled by catchment hydrological traits (i.e., snowmelt periods and water table depth). The conservative N-cycle within Coal Creek is likely due to the abundance of conifer trees, and a smaller riparian region, retaining more NO3- overall and reduced processing prior to export. This study highlights the value of integrating isotope systematics to link watershed functional traits to mechanisms of watershed element retention and release.

Tailgut cysts (TGC) are rare lesions occurring in the retrorectal space, a potential space anterior to the sacrum and coccyx and posterior to the rectum. They are developmental lesions that have potential for malignant transformation into neuroendocrine tumors, carcinomas, or adenocarcinomas. However, they present with non-specific complaints and are often challenging to diagnose and can lead to misdiagnosis and delay of treatment. We present a case of a a 27-year-old female patient with history of a myelomeningocele who was found to have a left sided pre-sacral cystic mass. Histologic examination of the lesion following surgical removal were consistent with TGC. The presence of a TGC in a patient with myelomeningocele is a rare occurrence. There is some evidence that disturbances in the Sonic Hedge Hog (SHH) signaling pathway can lead to abnormal branching and overgrowth of the notochord contributing to abnormal separation from the hindgut endoderm. Clinicians should have a heightened clinical suspicion for a TGC in patients with known neural tube defects who present with a perirectal mass lesion.

DESI will determine precise redshifts and employ the baryon acoustic oscillation method to measure distances from the nearby universe to beyond redshift z > 3.5, and employ redshift space distortions to measure the growth of structure and probe potential modifications to general relativity. We describe the significant instrumentation we developed to conduct the DESI survey. This includes: a wide-field, 3fdg2 diameter prime-focus corrector; a focal plane system with 5020 fiber positioners on the 0.812 m diameter, aspheric focal surface; 10 continuous, high-efficiency fiber cable bundles that connect the focal plane to the spectrographs; and 10 identical spectrographs. Each spectrograph employs a pair of dichroics to split the light into three channels that together record the light from 360–980 nm with a spectral resolution that ranges from 2000–5000. We describe the science requirements, their connection to the technical requirements, the management of the project, and interfaces between subsystems. DESI was installed at the 4 m Mayall Telescope at Kitt Peak National Observatory and has achieved all of its performance goals. Some performance highlights include an rms positioner accuracy of better than 0farcs1 and a median signal-to-noise ratio of 7 of the [O ii] doublet at 8 × 10−17 erg s−1 cm−2 in 1000 s for galaxies at z = 1.4–1.6. We conclude with additional highlights from the on-sky validation and commissioning, key successes, and lessons learned. © 2022. The Author(s). Early-onset pancreatic cancer (EOPC) - defined as pancreatic cancer diagnosed before the age of 50 years - is associated with a poor prognosis as compared to later-onset pancreatic cancer (LOPC). Emerging evidence suggests that EOPC may exhibit a genetic signature and tumor biology that is distinct from that of LOPC. We review genetic mutations that are more prevalent in EOPC relative to LOPC and discuss the potential impact of these mutations on treatment and survival.Using PubMed and Medline, the following terms were searched and relevant citations assessed: "early onset pancreatic cancer," "late onset pancreatic cancer," "pancreatic cancer," "pancreatic cancer genes," and "pancreatic cancer targeted therapy."Mutations inGenetic mutations associated with EOPC are distinct from those of LOPC. The preponderance of the evidence suggest that poor outcomes in EOPC are related both to advanced stage of presentation and unique tumor biology. The molecular and genetic features of EOPC warrant further investigation in order to optimize management.

O objetivo da pesquisa foi avaliar a influência do protetor, tratamento químico com fungicida e inseticida e períodos de armazenamento de sementes das cultivares de arroz irrigado IRGA 424 RI e BR IRGA 409, na germinação e emergência. Para cada cultivar, o delineamento experimental foi inteiramente casualizado, em esquema fatorial 3 x 5, com quatro repetições. Os três tratamentos foram: 1- fungicida + inseticida + protetor de sementes; 2- fungicida + inseticida; 3- sem tratamento de sementes. Os cinco períodos de armazenamento foram: 0, 5, 15, 30 e 60 dias, antes de realizar o teste de germinação das sementes em laboratório e emergência no campo. Em laboratório, avaliou-se também o número de plantas normais, anormais, comprimento de raiz e parte aérea para a cultivar IRGA 424 RI. O tempo de armazenamento das sementes influenciou a germinação de sementes em laboratório, porém, a porcentagem de germinação se manteve acima de 82,5%. Os tratamentos de sementes promovem maior emergência de plântulas e o tempo de armazenamento de até 60 dias não alterou a emergência no campo, nas duas cultivares. A utilização do protetor de sementes não alterou o estabelecimento inicial de plântulas no campo, nas duas cultivares.

The aim of this study was to assess the diagnostic yield and complication rate of 18 F-FDG PET/CT ultrasound (US) fusion for percutaneous biopsy of FDG-avid lesions among patients with known or suspected malignancy.We describe the clinical, imaging, and histopathologic features of 36 patients who underwent percutaneous biopsy using real-time PET/CT US fusion. In addition, we review the literature on PET/CT US fusion. Using Medline, the following MeSH terms were searched and relevant citations assessed: "fusion imaging," "PET/CT fusion," "PET/CT-guided biopsy," "PET/US fusion," "ultrasound fusion," and "ultrasound fusion-guided biopsy."A total of 36 patients (15 men, 21 women) with known or suspected malignancy and prior PET/CT imaging underwent percutaneous biopsy of FDG-avid lesions using PET/CT US fusion between October 2014 and July 2020. Coregistration was achieved using General Electric LOGIQ E9 software. Adequate tissue for analysis was obtained in all 36 patients. Histologic evaluation revealed malignancy in 14 patients (38.9%) and nonneoplastic tissue in 22 patients (61.1%). No intraprocedural or postprocedural complications were recorded.Fusion of PET/CT and US for percutaneous biopsy of FDG-avid lesions can be used to achieve excellent diagnostic yield with a low risk of complications.

Neurofibromatosis type 1 (NF1) is the most common genetic disease affecting the nervous system. Patients typically develop many tumors over their lifetime, leading to increased morbidity and mortality. The NF1 gene, mutated in NF1, is also commonly mutated in sporadic glioblastoma multiforme (GBM). Because both NF1 and GBM are currently incurable, new therapeutic approaches are clearly needed. Natural products represent an opportunity to develop new therapies, as they have been evolutionarily selected to play targeted roles in organisms. Schweinfurthin A is a prenylated stilbene natural product that has previously shown specific inhibitory activity against brain and hematopoietic tumor lines. We show that patient-derived GBM and NF1 malignant peripheral nerve sheath tumor (MPNST) lines, as well as tumor lines derived from the Nf1−/+;Trp53−/+ (NPcis) mouse model of astrocytoma and MPNST are highly sensitive to inhibition by schweinfurthin A and its synthetic analogs. In contrast, primary mouse astrocytes are resistant to the growth inhibitory effects of schweinfurthin A, suggesting that schweinfurthin A may act specifically on tumor cells. Stable transfection of the GTPase-activating protein related domain of Nf1 into Nf1−/−;Trp53−/− astrocytoma cells confers resistance to schweinfurthin A. In addition, the profound effect of schweinfurthin A on dynamic reorganization of the actin cytoskeleton led us to discover that schweinfurthin A inhibits growth factor–stimulated Rho signaling. In summary, we have identified a class of small molecules that specifically inhibit growth of cells from both central and peripheral nervous system tumors and seem to act on NF1-deficient cells through cytoskeletal reorganization correlating to changes in Rho signaling. Supplementary Data from Schweinfurthin A Selectively Inhibits Proliferation and Rho Signaling in Glioma and Neurofibromatosis Type 1 Tumor Cells in a NF1-GRD–Dependent Manner

The conjugation of proteins with ubiquitin plays numerous regulatory roles through both proteasomal-dependent and nonproteasomal-dependent functions. Alterations in ubiquitylation are observed in a wide range of pathologic conditions, including numerous malignancies. For this reason, there is great interest in targeting the ubiquitin-proteasome system in cancer. Several classes of proteasome inhibitors, which block degradation of ubiquitylated proteins, are widely used in research, and one, Bortezomib, is now in clinical use. Despite the well-defined and central role of the ubiquitin-activating enzyme (E1), no cell permeable inhibitors of E1 have been identified. Such inhibitors should, in principle, block all functions of ubiquitylation. We now report 4[4-(5-nitro-furan-2-ylmethylene)-3,5-dioxo-pyrazolidin-1-yl]-benzoic acid ethyl ester (PYR-41) as the first such inhibitor. Unexpectedly, in addition to blocking ubiquitylation, PYR-41 increased total sumoylation in cells. The molecular basis for this is unknown; however, increased sumoylation was also observed in cells harboring temperature-sensitive E1. Functionally, PYR-41 attenuates cytokine-mediated nuclear factor-κB activation. This correlates with inhibition of nonproteasomal (Lys-63) ubiquitylation of TRAF6, which is essential to IκB kinase activation. PYR-41 also prevents the downstream ubiquitylation and proteasomal degradation of IκBα. Furthermore, PYR-41 inhibits degradation of p53 and activates the transcriptional activity of this tumor suppressor. Consistent with this, it differentially kills transformed p53-expressing cells. Thus, PYR-41 and related pyrazones provide proof of principle for the capacity to differentially kill transformed cells, suggesting the potential for E1 inhibitors as therapeutics in cancer. SummaryDevelopment of vaccines and therapeutics that are broadly effective against known and emergent coronaviruses is an urgent priority. Current strategies for developing pan-coronavirus countermeasures have largely focused on the receptor binding domain (RBD) and S2 regions of the coronavirus Spike protein; it has been unclear whether the N-terminal domain (NTD) is a viable target for universal vaccines and broadly neutralizing antibodies (Abs). Additionally, many RBD-targeting Abs have proven susceptible to viral escape. We screened the circulating B cell repertoires of COVID-19 survivors and vaccinees using multiplexed panels of uniquely barcoded antigens in a high-throughput single cell workflow to isolate over 9,000 SARS-CoV-2-specific monoclonal Abs (mAbs), providing an expansive view of the SARS-CoV-2-specific Ab repertoire. We observed many instances of clonal coalescence between individuals, suggesting that Ab responses frequently converge independently on similar genetic solutions. Among the recovered antibodies was TXG-0078, a public neutralizing mAb that binds the NTD supersite region of the coronavirus Spike protein and recognizes a diverse collection of alpha- and beta-coronaviruses. TXG-0078 achieves its exceptional binding breadth while utilizing the same VH1-24 variable gene signature and heavy chain-dominant binding pattern seen in other NTD supersite-specific neutralizing Abs with much narrower specificity. We also report the discovery of CC24.2, a pan-sarbecovirus neutralizing mAb that targets a novel RBD epitope and shows similar neutralization potency against all tested SARS-CoV-2 variants, including BQ.1.1 and XBB.1.5. A cocktail of TXG-0078 and CC24.2 provides protection againstin vivochallenge with SARS-CoV-2, suggesting potential future use in variant-resistant therapeutic Ab cocktails and as templates for pan-coronavirus vaccine design.

Understanding the universe in its pristine epoch is crucial in order to obtain a concise comprehension of the late-time universe. Although current data in cosmology are compatible with Gaussian primordial perturbations whose power spectrum follows a nearly scale-invariant power law, this need not be the case when a fundamental theoretical construction is assumed. These extended models lead to sharp features in the primordial power spectrum, breaking its scale invariance. In this work, we obtain combined constraints on four primordial feature models by using the final data release of the BOSS galaxies and eBOSS quasars. By pushing towards the fundamental mode of these surveys and using the larger eBOSS volume, we were able to extend the feature parameter space (i.e. the feature frequency $\omega$) by a factor of four compared to previous analyses using BOSS. While we did not detect any significant features, we show that next-generation galaxy surveys such as DESI will improve the sensitivity to features by a factor of 7, and will also extend the parameter space by a factor of 2.5., Comment: 28 + 4 pages, 16 figures

In this case study, we describe a 25-year-old male who was admitted due to a severe traumatic brain injury, requiring invasive intracranial pressure monitoring. At 48 hours posttrauma, he developed intracranial hypertension refractory to medical treatment without tomographic changes in the brain. Subsequently, intra-abdominal hypertension and tomographic signs of abdominal surgical pathology were observed. An exploratory laparotomy was performed with an intraoperative diagnosis of acute mesenteric ischemia. After surgical intervention for the abdominal pathology, intracranial pressure was restored to physiological values with a favorable recovery of the patient. In this report, the relationship between intracranial pressure and intra-abdominal pressure is discussed, highlighting the delicate association between the brain, abdomen, and thorax. Measures should be taken to avoid increases in intra-abdominal pressure in neurocritical patients. When treating intracranial hypertension refractory to conventional measures, abdominal causes and multiple compartment syndrome must be considered. The cranial compartment has physiological interdependence with other body compartments, where one can be modified by variations from another, giving rise to the concept of multiple compartment syndrome. Understanding this relationship is fundamental for a comprehensive approach of the neurocritical patient. To the best of our knowledge, this is the first report of a comatose patient post-traumatic brain injury, who developed medically unresponsive intracranial hypertension secondary to acute mesenteric ischemia, in which surgical resolution of intra-abdominal pathology resulted in intracranial pressure normalization and restitutio ad integrum of neurological status.

Questionnaire on Treatment Satisfaction in Inpatient Child and Adolescent Psychiatry (FBZ-KJP) - Results of a Swiss Pilot Study Abstract: Objectives: Patient satisfaction is an established indicator for medical interventions. Existing questionnaires for the assessment of patient satisfaction in child and adolescent psychiatry are too global to target quality improvement in child and adolescent psychiatric hospitals. The assessment of patient satisfaction in child and adolescent psychiatry is very challenging because specific demands (development status, role of parents in treatment) have been taken into account. Therefore, an expert team leaded by the Swiss Society of Child and Adolescent Psychiatry developed a targeted instrument to assess patient satisfaction in both language regions (i. e., German and French). Methods: The article reviews the development of a new child and adolescent psychiatric questionnaire for the assessment of patient satisfaction as well as the findings of a survey conducted in a representative sample of 174 patients and 145 parents in six hospitals. Results: The internal consistency (Cronbach's α = .93) is excellent. The questionnaire has high levels of both convergence and face validity, and the correlation with the Client Satisfaction Questionnaire (CSQ-8) is ρ = .80 (patient-report) and .83 (parent-report). Furthermore, this questionnaire reveals the relative strengths and weaknesses of individual hospitals. The correlation between patient and parent assessment is, as expected, moderate (ρ = .29, for the total score and ρ = .39 for the CSQ-8). Conclusions: The Patient Satisfaction Questionnaire can be recommended to professionals as a standard for collecting data on client satisfaction within child and adolescent psychiatry.

Zusammenfassung: Fragestellung: Die Patientenzufriedenheit ist ein etablierter Qualitätsindikator für medizinische Interventionen, welchen die Kostenträger im Gesundheitssystem zunehmend als Indikator heranziehen, um Unterschiede zwischen Kliniken sichtbar zu machen. Die Erfassung der Patientenzufriedenheit stellt die Kinder- und Jugendpsychiatrie/-psychotherapie (KJPP) vor besondere Herausforderungen (Entwicklungsstand, Rolle der Eltern). Die bestehenden Patientenzufriedenheitsinstrumente erwiesen sich als zu unspezifisch, um die verschiedenen Aspekte einer stationären KJPP-Behandlung abbilden zu können. Deshalb setzte sich eine Arbeitsgruppe im Auftrag der Schweizerischen Gesellschaft für Kinder und Jugendpsychiatrie (SGKJPP) zum Ziel, ein psychometrisches Instrument für die Patienten- und Elternzufriedenheit zu entwickeln, welches in den deutsch- und französischsprachigen Sprachregionen eingesetzt werden kann. Methodik: Die erste Anwendung und Überprüfung dieses Fragebogens in einer multizentrischen Pilotstudie mit einer Stichprobe von 174 Patientinnen und Patienten und 145 Eltern aus sechs Kliniken wird in diesem Artikel vorgestellt. Ergebnisse: Die Ergebnisse zeigen hervorragende Testgütekriterien des Instrumentariums (Cronbachs α = .93 bzw. α = .97 (Eltern-/Patientenversion). Die Fragebogenvarianten besitzen eine hohe konvergente Validität; die Korrelation zum Client Satisfaction Questionnaire (CSQ-8) beträgt p = .80 bzw. .83 (Patienten-, Elternurteil). Zudem differenzieren sie zwischen den einzelnen Kliniken. Die Korrelationen zwischen Patienten- und Elternurteil liegen bei moderaten p = .29 für den Gesamttest-Score und p = .39 für den CSQ-8. Schlussfolgerungen: Der Fragebogen eignet sich für die Beschreibung der Qualitätsentwicklung in der KJPP und kann als Standardverfahren für die Erfassung der Patientenzufriedenheit empfohlen werden.

Background Sepsis is marked by elevated histamine, which is a vasodilator that increases vascular permeability. Although human studies are lacking, murine models of sepsis have indicated potential protective effects of histamine 2 receptor antagonist administration (H2RAs). Objective To assess any association between H2RA use in sepsis-3 patients admitted to the ICU and mortality, mechanical ventilation, length of stay, and markers of renal, liver, and lung dysfunction. Design Retrospective cohort study. Setting Intensive care units of the Beth Israel Deaconess Medical Center (BIDMC) accessed via the MIMIC-IV database spanning an 11-year period from 2008 to 2019. Patients (or participants) A total of 30,591 patients met the inclusion criteria for sepsis-3 on admission (mean age 66.49, standard deviation 15.92). Main measures We collected patient age, gender, ethnicity, comorbidities (contained within the Charlson comorbidity index), SOFA score, OASIS score, APS III score, SAPS II score, H2RA use, creatinine, BUN, ALT, AST, and P/F ratios. Primary outcomes were mortality, mechanical ventilation, and ICU length of stay. Key results A total of 30,591 patients met inclusion criteria over the 11-year sample period. The 28-day in hospital mortality rate was significantly lower among patients who received an H2RA (12.6% vs 15.1%, p < 0.001) as compared to those who did not receive an H2RA. Patients receiving an H2RA had significantly lower adjusted odds of mortality (0.802, 95% CI 0.741–0.869, p < 0.001), but significantly higher adjusted odds of invasive mechanical ventilation (4.426, 95% CI 4.132–4.741, p < 0.001) and significantly higher ICU LOS (3.2 days vs. 2.4 days, p < 0.001) as compared to the non-H2RA group. H2RA use was also associated with decreased severity of acute respiratory distress syndrome (ARDS) and lower serum creatinine. Conclusion Among patients hospitalized in the ICU for sepsis, the use of an H2RA was associated with significantly lower odds of mortality, decreased severity of ARDS, and a lower incidence of renal insufficiency.

The association between knee injury and knee osteoarthritis (OA) is understudied relative to its importance, particularly in younger populations. This study examined the association of knee injury with radiographic features of knee OA in military officers, who have a physically-demanding profession and high rates of knee injury.Participants were recruited in 2015-2017 from an existing program that enrolled 6,452 military officers during 2004-2009. Officers with a history of knee ligament or meniscal injuries (n=117 via medical record review) were compared to officers with no history of knee injury (n=143). Bilateral posteroanterior knee radiographs were obtained using a standardized fixed-flexion positioning frame. All images were read for Kellgren-Lawrence grade (KLG), osteophyte (OST), and joint space narrowing (JSN) scores. Data were analyzed using linear-risk regression models with generalized estimating equations.Injured and non-injured participants were similar (mean age=28 years, mean body mass index=25 kg/mAt the midpoint of a projected 20-year military career, officers with a history of traumatic knee injury have a markedly increased prevalence of knee rOA compared to officers without injury. Bacterial leaf streak (BLS) of barley is caused by the Gram-negative bacterial pathogen Xanthomonas translucens (Sapkota et al. 2020). In 2021, we observed multiple hill plots with BLS symptomatic plants in a barley stripe rust nursery in Vancouver, BC, Canada. We collected 29 leaf samples showing typical BLS symptoms (e.g. necrotic lesions; Fig. S1) and stored at 4 oC until bacterial isolation. Samples were surface-sterilized in 10% NaOCl for 20 sec and rinsed twice. About 1 cm2 of leaf tissue containing BLS characteristic lesions was macerated in 200 μL sterile H2O on a petri dish, incubated for 15 min, and 10 μl of the homogenates was streaked onto Wilbrink’s - Boric Acid - Cephalexin (WBC) agar medium. Plates were incubated at 28-30 oC for 48 hrs. Four single colonies were obtained: BC10-1-2a (USask BC10-2a), BC10-1-2b (USask BC10-2b), UBC026 and UBC028. Colonies were grown in WBC broth and gDNA was extracted using E.Z.N.A. Bacterial DNA Kit (Omega Bio-Tek) or DNeasy Plant Pro Kit® (Qiagen) following manufacturer protocols. Genus-level identification was achieved using 16S rRNA sequencing with 27F/1492R primers (Lane 1991) of UBC026 (1,399 bp; NCBI # OP327375) and UBC028 (1,415 bp; NCBI #OP327376). Complete 16S rRNA sequences (1,533bp) of BC10-2a and BC10-2b (1,533 bp) were extracted from the draft whole-genome sequences (WGS) generated in this study. The 16S rRNA sequence homology values of 99.0-100% were recorded between the 4 strains. BLAST analyses of the 16S rRNA sequences to GenBank entries exhibited 99.5-100% similarity values (100% coverage) with the pathotype strains of Xtt DSM 18974T (LT604072) and X. translucens pv. undulosa (Xtu) CFBP 2055 (CP074361). Whole genomes of BC10-2a (JANUQY01) and BC10-2b (JANUQZ01) were sequenced (150-bp; reads 33.1 million; mean coverage 2125x) using NovaSeq Illumina, assembled (Unicycler v0.4.8; Wick et al. 2017) and analyzed to identify the strains to the species-level (Tambong et al. 2021). WGS of strains USask BC10-2a and USask BC10-2b exhibited genome-based DNA-DNA hybridization (dDDH; Meier-Kolthoff et al. 2013) and BLAST-based average nucleotide identity (ANIb; Richter et al. 2015) of 100%. The two strains also showed dDDH and ANIb of 90.4% (species-leel cut-off of 70%) and 98.780% and 98.80% (cut-off of 96%), respectively, with Xtt DSM 18974T (LT604072). In contrast, the WGS of BC10-2a and BC10-2b exhibited only 78.2% dDDH homology values with Xtu CFBP 2055T, suggesting that the strains are genetically more similar to Xtt. The assignment of these strains to Xtt is corroborated by phylogenomic analysis (Fig. S2; Meier-Kolthoff and Göker 2019) that showed the two strains clustering together (100% bootstrap) with the type strain DSM 18974T. These data suggest that these strains are taxonomically members of Xtt. Identification was also confirmed to the genus-level by LAMP assay using published X. translucens primers (Langlois et al. 2017). Pathovar-level identification was confirmed using a cbsA and S8.pep multiplex PCR diagnostic assay (Roman-Reyna et al. 2022). Koch’s postulates were verified by greenhouse inoculation via leaf infiltration of UBC026 and UBC028 on 21-day old barley plants (line HB522) using an inoculum of 108 CFU ml-1 followed by re-isolation of the bacteria on WBC. The inoculated plants showed typical BLS symptoms similar to those observed in the field (Fig. S1). Water-inoculated plants had no symptoms. To our knowledge, this is the first published report of BLS of barley in British Columbia.
