Die gegenständliche Diplomarbeit setzt sich mit der sozialen Betreuung von Strafgefangenen, Untersuchungshäftlingen sowie jugendlichen Strafgefangenen und deren Kontaktmöglichkeiten mit der Außenwelt auseinander. Darüber hinaus werden die Rechte und Pflichten der in Haft befindlichen Personen einer genaueren Betrachtung unterzogen. Näher beleuchtet wird sodann die Wechselwirkung zwischen dem Außenkontakt der in Haft befindlichen Personen und dem damit verbundenen Resozialisierungserfolg. Im Zuge der Covid-19-Pandemie kam es zu massiven Einschränkungen der Kontaktmöglichkeiten mit der Außenwelt. Diese werden zum einen anhand der gesetzlichen Vorgaben dargestellt. Zum anderen wird mittels diesbezüglicher Erfahrungen der Justizanstalt Garsten ein Einblick gewährt, welche Optionen eine in Haft befindliche Person im Verlauf der Pandemie zur Verfügung hatte, den Kontakt mit der Außenwelt fortbestehen zu lassen und wie dies in der Praxis umgesetzt wurde. This diploma thesis deals with the social care of prisoners, pre-trial detainees and juvenile prisoners and their possibilities of contact with the outside world. In addition, the rights and duties of persons in custody are examined in detail. The interaction between the contact of persons in custody with the outside world and the associated resocialisation success is then analysed in more detail. In the course of the Covid-19 pandemic, there were massive restrictions on the possibilities of contact with the outside world. On the one hand, these are described on the basis of the legal regulations. On the other hand, the experiences of the prison in Garsten in this regard will provide an insight into the options available to a person in custody during the pandemic to maintain contact with the outside world and how this was implemented in practice. The compartmentalization of metabolic and regulatory pathways is a common pattern of living organisms. Eukaryotic cells are subdivided into several organelles enclosed by lipid membranes. Organelle proteomes define their functions. Yeasts, as simple eukaryotic single cell organisms, are valuable models for higher eukaryotes and frequently used for biotechnological applications. While the subcellular distribution of proteins is well studied in Saccharomyces cerevisiae, this is not the case for other yeasts like Komagataella phaffii (syn. Pichia pastoris). Different to most well-studied yeasts, K. phaffii can grow on methanol, which provides specific features for production of heterologous proteins and as a model for peroxisome biology. We isolated microsomes, very early Golgi, early Golgi, plasma membrane, vacuole, cytosol, peroxisomes and mitochondria of K. phaffii from glucose- and methanol-grown cultures, quantified their proteomes by liquid chromatography-electrospray ionization-mass spectrometry of either unlabeled or tandem mass tag-labeled samples. Classification of the proteins by their relative enrichment, allowed the separation of enriched proteins from potential contaminants in all cellular compartments except the peroxisomes. We discuss differences to S. cerevisiae, outline organelle specific findings and the major metabolic pathways and provide an interactive map of the subcellular localization of proteins in K. phaffii., About 2000 intracellular proteins of the methylotrophic yeast Komagataella phaffii were quantified and for most of them the organelle localization was attributed.

Peer-to-peer accommodation networks have been described as disruptive innovations, as revolutions in tourist accommodation, where demand is driven by new factors such as living like a local, authenticity and meeting new people. If indeed reasons for trading on these networks are so fundamentally different, prices should reflect that. This chapter investigates what drives the price of Airbnb listings in Vienna, and asks whether these price drivers are indeed new, or whether they reflect those in established commercial accommodation.

Einleitung: Das Nichteinhalten des verordneten Behandlungsplans führt bei rheumatoider Arthritis zu einer negativen Auswirkung auf die Funktion des Bewegungsapparats und den radiologischen Status. Um effektiv gegen Nonadhärenz vorgehen zu können sind Assessments notwendig die dieses Verhalten messen. In dieser Studie werden aktuelle Assessments zur Adhärenzmessung recherchiert und mit der ICF verlinkt. Methodik: Um dieses Ziel zu erreichen wurde in den Datenbanken Cinahl, Pubmed und Cochrane im Zeitraum von 12/2016 bis 02/2017 eine Recherche mit den Mesh-Terms arthritis, rheumatoid und patient compliance durchgeführt. Insgesamt konnten 17 Assessments identifiziert werden. Davon konnten neun mit den Regeln von Cieza et al. (2016) mit der ICF gelinkt werden, um im Anschluss mit Daten aus Interviews von PatientInnen mit RA verglichen zu werden. Resultate: Die Aufteilung und die Anzahl der Assessments konnte aufgrund der aktuell gewonnenen Ergebnisse zur Vergleichsstudie um neun Messmethoden erweitert werden. Der durchgeführte Linkingprozess der neun linkbaren Assessments ergab 110 Items die zu 32 verschiedenen ICF Kategorien verbunden werden konnten. Der Vergleich mit den Linkingdaten, die von Ritschl et al. (2017) aus den Interviews mit RA PatientInnen gewonnen Items, lässt lediglich bei den zwei Assessments CQ und MMAS-4 eine annähernde Übereinstimmungen mit den Perspektiven, Werten und Bedürfnissen der PatientInnen erkennen. Conclusio: Die Fragebögen CQ und MMAS-4, die annähernd ähnliche Linkingergebnisse mit den gelinkten Interviewdaten abbilden, sind kurz, prägnant und einfach in der Handhabung und können dadurch sehr gut in den klinischen Alltag impliziert werden. Ein wichtiger Forschungsansatz wäre, anhand der Linkingergebnisse der Interviewdaten, einen kompatiblen Fragebogen zu erstellen und die derzeit nicht im Detail übereinstimmenden Assessments abzulösen. Introduction: Non-compliance of prescribed treatment plan in case of rheumatoid arthritis leads to negative impacts on the functionality of the musculoskeletal system and as well radiological status. To work in an effective way against this non-adherence it is essential to implement assessments which provide measurements of this behavior. Within this study current assessments concerning measurement of adherence should be researeched and linked with ICF. Methods: To finally reach that goal researches with Mesh-Terms arthritis, rheumatoid and patient compliance were executed from 12/2016 until 02/2017 in following databases Cinahl, Pubmed and Cochrane. Within that research in total 17 assessments could have been identified. Form these assessments nine could have been linked with the rules from Cieza et al. (2016), which were afterwards compared with data gathered from interviews with RA patients. Results: Because of obtained findings within the current research it was possible to expand distribution and amount of assessments compared to the comparative study by nine measurement methods. The result of the executed linking process of these nine linkable assessments were 110 items which could have been linked to 32 different ICF-categories. A comparison with linking data from Ritschl et al. (2017) out of interviews with RA-patients only provides an approximate match among the assessments CQ and MMAS-4 with perspectives, values and needs of the patients. Conclusion: CQ and MMAS-4 questionnaires, which show an approximate match with the linked data from the interviews are short, precise and simple to handle. Because of these characteristics they can be easily implemented into the clinical daily routine. An important research approach would be to create a compatible questionnaire which is based on linking results coming from interview data. In der Biologie, Linguistik, Psychologie und Soziologie wird versucht, (menschliches) Interaktionsverhalten zu verstehen und zu beschreiben. In der Robotik ist ein Schwerpunkt, dieses (menschliche) Interaktionsverhalten zu modellieren, damit eine natürliche Interaktion mit Robotern möglich ist. Ein Bestandteil der natürlichen Interaktion ist unter anderem, zu erkennen, wann ein Interaktionspartner die Sprecherrolle übernehmen darf, ohne unfreundlich zu wirken und den anderen Interaktionspartner zu unterbrechen. Ein weiterer Schwerpunkt ist die Analyse, wie verschiedene Menschen beim Sprechen gestikulieren, um z. B. gleiche Sachinhalte mittels Sprache und sprachbezogener Gesten zu beschreiben. Sind aus solchen Analysen Verhaltensmuster erkannt worden und wurden diese Interaktionsverhaltensweisen implementiert, muss verifiziert werden, ob Menschen das z. B. von einem Roboter oder sozialen Agenten ausgeführte Verhalten als natürlich empfinden. Eine gängige Methode, ein solches Verhalten zu analysieren, ist die Aufzeichnung in verschiedenen multimedialen Daten wie Audio und Video, sodass diese anschließend im Detail analysiert werden können. Leider ist dieser Videoanalyseprozess sehr zeitintensiv, da er manuell durch Menschen durchgeführt werden muss. Um eine Bewegung in einem Video analysieren zu können, muss diese erst aus dem Video extrahiert werden, wobei dieses nicht immer genau durchgeführt werden kann. Dieses kann der Fall sein, wenn Gelenk- und andere Körperteilepositionen nicht genau bekannt sind, da diese Körperteile verdeckt sein können. Da diese Analyse ein zeitintensiver Prozess ist, der durch viele Arbeitsstunden teuer wird, gibt es Bemühungen, möglichst Mechanismen zu finden, durch die diese Arbeiten automatisch durchgeführt werden können. Als erstes Problem muss bei einer Analyse von Videodaten ermittelt werden, was Personen sind und in welcher Körperhaltung sie sich befinden. Allgemein funktioniert dieses, ist allerdings fehleranfällig. Um genauere Daten der Interaktionen zu erhalten und um auch automatische Analysen durchführen zu können, geht ein Trend dazu über, weitere modale Daten wie Motion-Capture-Daten zusätzlich aufzuzeichnen. Dadurch kann die Bewegung der interagierenden Personen viel genauer in räumlicher Relation zueinander analysiert werden. Um dieses durchführen zu können, stellen sich die Fragen, "wie die Motion-Capture-Daten sinnvoll mit angemessenem Arbeitsaufwand für die Untersuchungen genutzt werden können" und "wie die Interaktionen mehrerer Personen über eine längere Zeitspanne robust aufgezeichnet werden können". Beim Motion-Capturing ist eine lange Aufnahme mit einem Vielfachen dieser Zeit als Nachbearbeitungsphase verbunden. In dieser Nachbearbeitungsphase werden die Daten aufgearbeitet, damit einzelne Marker immer den zugehörigen Körperteilen zugeordnet werden können. Um einen deutlichen Nutzen aus dem Motion-Capturing ziehen zu können, darf die Zeit, die für das zusätzliche Motion-Capturing aufgewendet wird, nicht höher sein als die Zeit, die für das Annotieren der Video-Analyse aufgewendet würde. In dieser Arbeit wird gezeigt, wie das Motion-Capturing mit einem angemessenen Zeiteinsatz verwendet werden kann, um automatische Analysemöglichkeiten nutzbringend durchführen zu können. Dabei wird auf die Fragestellung eingegangen, "was die Motion-Capture-Daten für Möglichkeiten bei der Verhaltensforschung bei Interaktionen bieten". Dazu wird gezeigt, dass diese neuen Möglichkeiten in einer automatischen detaillierten Analyse liegen, die eine standardisierte Basis für Analysen mit einer immer gleichbleibend guten Qualität liefern. Um die Nützlichkeit der Motion-Capture-Daten hervorzuheben, wird gezeigt, wie diese im Forschungsalltag eingesetzt werden können. Die hierbei gesammelten Erfahrungen sind in die Entwicklung eines Annotationstools "PAMOCAT" eingegangen, bei dem verschiedene elementare Verhaltensbestandteile als abstrakte Kategorien (wie z.B. Bewegung in elementaren Gelenken, etwas angucken, Handbewegungen oder Posen) automatisch annotiert werden können. Dabei haben sich verschiedene elementare Kategorien herauskristallisiert, die ein breites Spektrum von möglichen Einsatzbereichen in der Verhaltensforschung bieten. Dazu wird eine Basis von elementaren Interaktionsphänomenen bereitgestellt, die durch Kombinationen mit anderen Interaktionsphänomenen als Suche nach Zeitpunkten, bei denen diese zusammen auftreten, angesetzt werden kann. Dadurch ist eine detailliertere Analyse komplexen Verhaltens einfacher und schneller möglich, als es zuvor möglich war. Um diese Analysefunktionalität einem möglichst großen Anwenderkreis bereitzustellen, ist ein Graphical User Interface - GUI entwickelt worden, welches in Zusammenarbeit mit Endnutzern optimiert wurde. Damit ergeben sich neue Möglichkeiten bei der Analyse großer Korpora und es kann viel Zeit eingespart werden, sodass die Aufmerksamkeit auf eine detaillierte Analyse fokussiert werden kann.

Hintergrund: Herpes Zoster (HZ) ist ein schmerzhafter Hautausschlag verursacht durch Reaktivierung des Varizella-Zoster-Virus. Die häufigste Komplikation ist die postherpetische Neuralgie (PHN), ein lang anhaltender Schmerz in der betroffenen Hautregion. In Europa erhielt 2006 ein Impfstoff zur Prävention von HZ und PHN bei Personen ≥50 Jahren die Zulassung, der aber erst Ende 2013 in Deutschland verfügbar wurde. Ziel dieser Dissertation war es, neben der Erhebung der HZ- und PHN-assoziierten epidemiologischen und ökonomischen Krankheitslast eine gesundheitsökonomische Evaluation der neuen Impfung im Kontext des gesetzlichen Krankenversicherungssystems (GKV) in Deutschland durchzuführen. Methodik: Die Dissertation wurde in 3 aufeinander aufbauenden Publikationen bearbeitet. 1) Zur Erhebung der alters-spezifischen HZ Inzidenz und der HZ bedingten Sterblichkeit wurde eine Analyse von Abrechnungsdaten für die Jahre 2006-08 der kassenärztlichen Vereinigungen (KV) und der Gesundheitsberichterstattung des Bundes durchgeführt. HZ Patienten ≥50 Jahre wurden über die ICD-10 Diagnosecodes identifiziert. 2) Behandlungskosten des HZ einschließ-lich der PHN sowie der Anteil an PHN Fällen an HZ Patienten wurde für die Jahre 2004-09 mit-tels einer Stichprobe von 240.000 Personen aus Krankenkassenabrechnungsdaten erhoben. Alle relevanten Kostenfaktoren wurden berücksichtigt und getrennt nach der Perspektive der GKV und der gesellschaftlichen Perspektive ausgewertet. 3) Ein mathematisches Modell wurde entwickelt, mit dem die gesundheitsökonomischen Effekte der Impfung vorausberechnet und Impfstrategien miteinander verglichen werden konnten. Die Ergebnisse der 1. und 2. Publikation fungierten hierbei als Inputdaten. Ergebnisse: Bei ≥50-Jährigen wurde eine HZ Inzidenz von 9,6/1.000 Personen und Jahre (PJ) erhoben. Etwa 5% der HZ Fälle entwickeln eine PHN. Für die etwa 400.000 HZ- und 20.000 PHN-Fälle p.a. entstehen aus GKV- (gesellschaftlicher) Perspektive Kosten in Höhe von ca. 105 (180) Millionen Euro. Wenn 20% einer hypothetischen Kohorte von 1 Million 50-Jähriger im Alter von 60 Jahren geimpft würden, könnten in dieser Kohorte ca. 20.000 HZ-Fälle verhindert werden. Um ein qualitäts-adjustiertes Lebensjahr (QALY) zu gewinnen müssen 28.146 Euro (gesellschaftliche Perspektive) aufgewendet werden. Neben dem Impfstoff-Preis ist auch die Dauer des Impfschutzes ein kritischer Faktor. Verschiedene Sub-Analysen zeigten, dass das Impfen im Alter von 60 Jahren am kosten-effektivsten wäre. Schlussfolgerung: Die medizinische und auch ökonomische Krankheitslast aufgrund von HZ und PHN ist in Deutschland hoch. Die HZ Impfung kann eine substantielle Zahl von Fällen verhindern, jedoch zu höheren Kosten im Vergleich zum Nicht-Impfen., Background: Herpes zoster (HZ) is a painful skin-rash caused by a reactivation of the varicella-zoster-virus. The main complication of HZ is postherpetic neuralgia (PHN), a long-term persisting pain in the affected skin area. In 2006, a vaccine for the prevention of HZ and PHN got licensed in Europe for individuals aged ≥50 years but was not available on the German market before the end of 2013. Objective of this dissertation was the assessment of the HZ- and PHN-related epidemiological and economic burden in Germany and the health economic evaluation of the new vaccine in the context of the German statutory health insurance (SHI) system. Methods: The dissertation consisted of three consecutive publications. 1) For the estima-tion of age-specific HZ incidence and HZ related mortality data from the Association of Statutory Health Insurance Physicians (ASHIP) and the Federal Health Monitoring System (FHM) for the years 2006-08 were analyzed. HZ cases were identified via ICD-10 codes. 2) Treatment costs for HZ and PHN cases were analyzed for the years 2004-09 from a study sample (240,000 individuals) of one SHI fund. All treatment sectors and patient co-payments were considered and evaluated separately according to the SHI- and the societal perspective (SP). 3): A mathematical model was developed to predict the health economic effects of the vaccination and to identify most critical and uncertain input factors. Results from 1st and 2nd publications served as input data for the mathematical model. Results: HZ incidence in ≥50 year old individuals is 9.6/1,000 persons per year (PY). About 5% of HZ cases develop a PHN. Costs for the estimated total 400,000 HZ and 20,000 PHN cases p.a. range between 105 and 180 million Euros from SHI and societal perspective. If 20% of a cohort of 1 million individuals aged 50 years gets vaccinated at age 60, about 20,000 HZ cases can be prevented. To gain one quality-adjusted life year (QALY) 28,146 Euros have to be invested (SP). Besides vaccine price, the duration of vaccine induced immunity is a critical factor. Several analyses indicated 60 to be the most cost-effective age at vaccination. Conclusion: The medical and economic burden caused by HZ/PHN in Germany is high. The vaccination can avoid a substantial number of cases, however, at higher costs compared to no vaccination.

Therefore it is fruitful to combine qualitative (Conversation Analysis) and quantitative analytical (Motion Capturing) methods. Tools for data visualization and annotation are important since they constitute a central interface between different research approaches and methodologies. With this aim we have developed the pre-annotation tool "PAMOCAT - Pre Annotation Motion Capture Analysis Tool" [2]. This has a sophisticated user interface that easily allows the annotating person to find correlations between different joints and/or phenomena, to analyze the corresponding 3D pose in a reconstructed virtual environment, and to export combined qualitative and quantitative annotations to standard annotation tools. The tool has the functionality to easily search for distinct constellations of different phenomena and display frames in which these could be found in an annotation overview.

A sophisticated user interface easily allows the annotating person to find correlations between different joints, analyze the corresponding 3D pose in a reconstructed virtual environment, and to export combined qualitative and quantitative annotations to standard annotation tools. Using this technique we are able to examine complex setups with three persons in tight conversion or largely unconstrained engagement situations of humans and robots.

Motion Capturing (MoCap) ermöglicht realistische Bewegungen virtueller Agenten. Hier wird ein System für die optische Bewegungserfassung und GPU-beschleunigte Bewegungssynthese vorgestellt. Das Verfahren berücksichtigt Clusterumgebungen heutiger VR Systeme. Mittels eines optischen Trackingsystems werden die Bewegungen eines Benutzers in einer immersiven virtuellen Umgebung auf Basis der Positions- und Ausrichtungsdaten einiger Marker an signifikanten Körperpunkten erfasst. Die Übertragung auf einen virtuellen Agenten erfolgt interaktiv und unterstützt die Anpassung unterschiedlicher Skelettproportionen. Signifikante Teile der benötigten kinematischen Berechnungen auf Basis einer Denavit- Hartenberg-Repräsentation sowie die für die Animation des Agenten notwendigen Meshdeformationen werden dabei in Echtzeit über eine GPU-basierte (GPU-Graphical Processing Unit) Implementierung der Algorithmen realisiert.

If we want to model the dynamic and contingent nature of human social interaction (e.g. for the design of human robot interaction), analysis and description of natural interaction is required that combines different methodologies and research tools (qualitative/quantitative; manual/automated). In this paper, we pinpoint the requirements and technical challenges for constituting and managing multimodal corpora that arise when linking Conversation Analysis with novel 3D motion capture technologies: i.e. to robustly track multiple participants over an extended period of time. We present and evaluate a solution to by-pass the limits of the current standard Vicon system (using rigid bodies) and ways of mapping the obtained coordinates to a human skeleton model (inverse kinematics) and to export the data into a format that is supported by standard annotation tools (such as ANVIL).

With this aim we have developed the pre-annotation tool "PAMOCAT - Pre Annotation Motion Capture Analysis Tool" that detects different phenomena. These phenomena are in the category single person and person overlapping phenomena. Included are functions for the analysis of head focused objects, hand activities, single DOF- degree of freedom activity, posture detection and intrusions into the co-participant's space. These detected phenomena related to the frames will be displayed in an overview. The phenomena can be chosen to search for a specific constellation between these different phenomena. A sophisticated user interface easily allows the annotating person to find correlations between different joints and phenomena, to analyze the corresponding 3D pose in a reconstructed virtual environment, and to export combined qualitative and quantitative annotations to standard annotation tools. Using this technique we are able to examine complex setups with three participants engaged in conversation. In this paper we propose how PAMOCAT can be integrated in the research cycle by showing a concrete PAMOCAT-based micro-analysis of a multimodal phenomenon, which deals with kinetic procedures to claim the floor.

In mobile SCR applications, the most widespread reducing agent is ammonia. However, due to its toxicity it is not stored directly as pressurized or liquefied gas. Instead, an aqueous solution of 32.5 wt % urea is commonly used as ammonia precursor. The urea solution can be dosed into the main exhaust pipe, where it decomposes in the hot exhaust gas and on the SCR catalyst to yield ammonia and carbon dioxide. Urea as ammonia precursor and its decomposition will be discussed in the first part of this chapter, with a focus on catalytic decomposition and byproduct formation. In the second part, alternative ammonia precursor compounds, including solid ammonia precursors will be presented. These exhibit a higher ammonia storage density, a lower melting point and/or a higher stability when stored at elevated temperatures compared to urea solution.

This thesis is separated in three main parts including a general introduction, three chapters based on research projects and the last chapters with the experimental procedures and analytical data. The focus of this work is based on the isolation of natural products and their application in chemistry and biology. Chapter 1: The first chapter consists in a general introduction about natural products, where their importance in drug discovery and in the interaction between living organisms is highlighted. Several examples of natural products from different origins are described. Chapter 2: The study of a system living in obligatory symbiosis is addressed in this chapter. Psychotria kirkii is a plant having the particularity to be in symbiosis with a bacterial strain named Candidatus Burkholderia kirkii and the presence of this microorganism was found to be essential for the plant’s survival. Interestingly, the bacterial genomic sequencing and analysis revealed the presence of a putative C7N aminocyclitol. This project led to the isolation, total synthesis and biological evaluation of kirkamide, a new C7N aminocyclitol from Candidatus Burkholderia kirkii. In the course of the chemical composition analysis of Psychotria kirkii, another new natural product (streptol glucoside) was isolated and its structure was elucidated. The last part of this chapter focuses on detection and quantification methods for the analysis of these natural products. Chapter 3: After completing a genome-driven bioassay-guided fractionation, a natural product possessing an unusual N-nitrosohydroxylamine functional group was isolated. The formation of the N-N bond in biological system has yet to be elucidated. Chapter 3 addresses the question regarding the biosynthetic analysis of the N-nitrosohydroxylamine compound. Furthermore, the mechanism of the N-N bond formation was investigated in detail using model compounds. Chapter 4: The potential of secondary metabolites has been recognized for the discovery of biological active compounds. The strength of natural products resides in the diversity and complexity of their chemical structures. In this chapter the isolation, structure elucidation and biological evaluation of new natural products from cyanobacteria are described. Three new cyanobactins were isolated from the cyanobacteria Microcystis aeruginosa EAWAG 251. The stereoassignement of these natural products, named balgacyclamides A–C, is presented with their biological activity investigations against the parasite Plasmodium falciparum. The last part of the chapter describes a bioassay-guided fractionation leading to the discovery of two new glycolipopeptides from cyanobacteria. The efforts achieved towards the structure elucidation using HRMS tandem mass spectrometry, NMR and amino acid analysis are presented.
