1. ATF4 mediates fetal globin upregulation in response to reduced β-globin

Abstract

Fetal development and anemias such as β-hemoglobinopathies trigger rapid production of red blood cells in a process known as stress erythropoiesis. Cellular stress prompts differentiating erythroid precursors to express high levels of fetal γ-globin, which has suggested strategies to treat hemoglobinopathies such as thalassemia and sickle cell disease. However, the mechanisms underlying γ-globin production during cellular stress are still poorly defined. Here we use CRISPR-Cas genome editing and CRISPRi transcriptional repression to model the stress caused by reduced levels of adult β-globin. We find that loss of β-globin is sufficient to induce widespread globin compensation, including robust re-expression of γ-globin. Time-course RNA-seq of differentiating isogenic erythroid precursors identified the ATF4 transcription factor as a causal regulator of this response. ChIP-seq of multiple erythroid precursor genotypes and differentiation states revealed that β-globin knockout leads to reduced engagement of ATF4 targets involved in the unfolded protein response. This ATF4 program indirectly regulates the levels of BCL11A, a key repressor of γ-globin. Identification of ATF4 as a key regulator of globin compensation adds mechanistic insight to the poorly understood phenomenon of stress-induced globin compensation and could be relevant for proposed gene editing strategies to treat hemoglobinopathies.

Abstract

This work uses the algorithmically di erentiated (AD) version of the the open-source Computer Aided Design (CAD) library Open CASCADE Technology (OCCT) to compute geometric derivatives. Here the di erentiated OCCT is applied to two di erent gradient- based shape optimisation test-cases. First, the di erentiated OCCT is utilised to develop a gradient-based re-parametrisation tool that ts a B-spline surface to an optimised mesh of a duct used in an automotive application. This step is performed in CAD-free shape optimisation context in order to nd a parametric model that approximates the optimised mesh. Second, the di erentiated OCCT is coupled with an adjoint CFD solver to run a CAD-based shape optimisation of a compressor stator blade.

Abstract

Numerical optimisation is becoming an essential industrial method in engineering design for shapes immersed in uids. High-delity optimisation requires ne design spaces with many design variables, which can only be tackled with gradient-based optimisation methods. CAD packages that are open-source or commercially available do not provide shape derivatives, but impose to compute them with expensive, inaccurate and nonrobust nite-dierences. The present work is the rst demonstration of obtaining exact shape derivatives with respect to CAD design parametrisation by applying algorithmic dierentiation to a complete CAD system, in this case the Open Cascade Technology (OCCT) CAD-kernel. The extension of OCCT to perform shape optimisation is shown by using parametric models based on explicit parametrisations of the CAD model and on implicit parametrisations based on the BRep (NURBS). In addition, we demonstrate the imposition of geometric constraints for both approaches, a salient part of industrial design, and an intuitive method of storing them in standard CAD format. The proposed method is applied to redesign TU Berlin Stator Testcase.

Abstract

Numerical shape optimisation is becoming an essential tool in aeronautic design. Highfidelity optimisation with many design variables will require gradient-based optimisation, adjoint methods can compute gradients very efficiently. CAD models are used on every stage of engineering product development from design to manufacturing. Ideally, the CAD system is kept inside the optimisation loop to maintain a consistent CAD model during the optimisation, as often practised in gradient-free optimisation. However, typical commercial CAD systems do not offer derivative computation, and standard CAD parametrisations may not define a suitable design space for the optimisation. In this work, we use the automatically differentiated (AD) version of Open Cascade Technology (OCCT) CAD-kernel, which robustly provides accurate derivatives with respect to CAD parameters. The kernel is used to handle implicit CAD parametrisations, e.g. move control points of NURBS surfaces, automatically re-parametrise these surfaces, compute intersections between them and perform corresponding mesh morphing. We apply the differentiated CAD tool and parametrisation to high-fidelity aerodynamic shape optimisation of the NASA CRM wing-body intersection and exercise the CAD method for the pylon-nacelle modification.

Abstract

Altres ajuts: This work was supported by Celyad, SA (Mont-Saint-Guibert, Belgium). Celyad has received research grants from the Walloon Region (Belgium, DG06 funding). Cardiopoietic cells, produced through cardiogenic conditioning of patients' mesenchymal stem cells, have shown preliminary efficacy. The Congestive Heart Failure Cardiopoietic Regenerative Therapy (CHART-1) trial aimed to validate cardiopoiesis-based biotherapy in a larger heart failure cohort. This multinational, randomized, double-blind, sham-controlled study was conducted in 39 hospitals. Patients with symptomatic ischaemic heart failure on guideline-directed therapy (n = 484) were screened; n = 348 underwent bone marrow harvest and mesenchymal stem cell expansion. Those achieving > 24 million mesenchymal stem cells (n = 315) were randomized to cardiopoietic cells delivered endomyocardially with a retention-enhanced catheter (n = 157) or sham procedure (n = 158). Procedures were performed as randomized in 271 patients (n = 120 cardiopoietic cells, n = 151 sham). The primary efficacy endpoint was a Finkelstein–Schoenfeld hierarchical composite (all-cause mortality, worsening heart failure, Minnesota Living with Heart Failure Questionnaire score, 6-min walk distance, left ventricular end-systolic volume, and ejection fraction) at 39 weeks. The primary outcome was neutral (Mann–Whitney estimator 0.54, 95% confidence interval [CI] 0.47–0.61 [value > 0.5 favours cell treatment], P = 0.27). Exploratory analyses suggested a benefit of cell treatment on the primary composite in patients with baseline left ventricular end-diastolic volume 200–370 mL (60% of patients) (Mann–Whitney estimator 0.61, 95% CI 0.52–0.70, P = 0.015). No difference was observed in serious adverse events. One (0.9%) cardiopoietic cell patient and 9 (5.4%) sham patients experienced aborted or sudden cardiac death. The primary endpoint was neutral, with safety demonstrated across the cohort. Further evaluation of cardiopoietic cell therapy in patients with elevated end-diastolic volume is warranted.

Abstract

Malvasia istriana (Vitis vinifera) is a domestic and widespread grape cultivar in the Istrian wine region that gives the characteristic white wine. Istrian region is situated in the northwestern part of Croatia (Adriatic coast). In the ageing process, Malvasia istriana loses its aroma, (freshness and fruitness) and hence it is usually consumed as a young wine. Therefore, blending is used to improve the quality of Malvasia istriana wine, to enrich its aroma and to maintain its varietal recognizability during the consumption period. Malvasia istriana base wine (85%) was used by blending with Chardonnay, Sauvignon blanc, Pinot blanc, Prosecco and Muscat, from grape varieties grown in the Istrian region. The change in volatile compounds of the blends was observed throughout the year by using HS-SPME headspace solid phase microextraction. Based on the analysis of the volatile compounds and sensory evaluation of the wine quality, the best result was obtained by blending Malvasia istriana, Sauvignon blanc and Pinot blanc. Chardonnay, Pinot blanc, Sauvignon blanc and Prosecco proved to be suitable for blending with Malvasia istriana, giving blends of high quality of the desired aroma profile. Muscat wine blended independently with Malvasia istriana or in combination with other wines proved to be unsuitable for blending due to its specific muscat aroma which dominates over the base wine aroma in the blend. Blending Malvasia istriana with other selected wines produced wines of better sensory quality, richest in volatile compounds, which can justify the blending.

Abstract

Malvazija is a domestic and most widespread sort of white grapes in the wine area of Istra. It regularly gives high crops. Malvazija wines are characterized by nice bouquet and distinguished sort character. Instrumental analysis was applied on Malvazija wines produced in 1999 to point out variability of aroma key compounds and to correlate them with sensory judgment. Higher alcohols, esters and carbonyl compounds were detected by gas chromatography / headspace analysis (GC-HSS). Ethanol content, total extract, sugar-free extract, total acids, reducing sugar, total and free SO2 were also determined in all samples. Important differences were found in concentration of aroma compounds in the analyzed samples. Concentration of acetaldehyde, ethyl acetate, and isoamyl alcohol was negatively correlated with sensory judgment. No significant correlation was found between concentration of diacetyl, isoamyl acetate, ethyl propionate, ethyl lactate, ethyl hexanoate, ethyl octanoate and sensory judgment.

13. DIAGNOSTIC VALUE OF HE4, CA125, ROMA INDEX AND ULTRASOUND FINDINGS IN EPITHELIAL OVARIAN CANCER

Abstract

Background and aims: The aim of this study was to investigate the diagnostic values of risk of ovarian malignancy algorithm (ROMA), human epididymis protein 4 (HE4), cancer antigen 125 (CA125), menopausal status and ultrasound (US) findings for distinguishing benign from malignant pelvic mass. Methods: Serum CA 125 and HE4 levels were determined and the ROMA values were calculated in 40 women scheduled for surgery due to adnexal mass and in 30 healthy women, using Elecsys 2010 (Roche). Ultrasound findings were documented by experienced examiners. We used receiver-operator characteristic curve analysis. Sensitivity and specificity were calculated and the area under the curve was also assessed. Correlation between all methods was estimated using Spearman's rank correlation coefficient r. Results: The mean age of women with an adnexal mass was 52±14 years and the mean age of healthy women was 48±15 years. Only ROMA index was significantly higher in group of patients with adnexal mass compared to healthy patients (t-test: 2, 201 ; df=29, P=0, 036). In group of patients with adnexal mass ROMA index and serum HE4 level were significantly higher in postmenopausal patients, in patients with epithelial ovarian cancer (EOC) and in patients with three or more features from US findings (p=0.01-0.001), while serum CA125 level was significantly higher in patients with EOC and in patients with three or more features from US findings (p=0.001). Conclusion: Our study showed that using menopausal status and US findings in combination with ROMA index, HE4 and CA125, improve diagnostic accuracy for EOC.

Abstract

Aroma is a phenomenon that occurrs when food and live organism mutually react during the process of consumption. Many factors influence the making of aroma of wine distillates (e.g. cognac) during the technological process of distillation. It is extremely difficult to bring an objective judgement on the influence of individual factors since aroma is very subjective in its nature. The possibility of objective assessment of the influence of grape varieties on wine distillate was performed in this work using a computer in processing the results of cromatographic analyses of aroma substances. The given results were verified by sensor analyses. The achieved results have shown that mathematical model for calculating aroma chromatogram similarity can be successfully used for objective assessment of the influence of individual factors on aroma of wine distillates and that grape varieties have significant impact on aroma and the quality of wine distillates.

Abstract

Background and aims: The aim of this study was to evaluate the risk of endometrial cancer (EC) in postmenopausal breast cancer patients according to duration of tamoxifen treatment. Methods: The current study utilized tissue samples from 65 postmenopausal tamoxifen-treated breast cancer patients who underwent hysteroscopy with pathohistological evaluation in the period from 2012 to 2014. The Human Ethics Committee of the institution approved this study. The patients were divided, according to duration of tamoxifen treatment, 45 in Group I (patients treated for less than 3 years) and 20 in Group II (patients treated for longer than 3 years) and correlation with endometrial pathology was evaluated. Results: The mean age of patients was 54.5±8.8 years (range, 44 to 87 years). Vaginal bleeding was present in 18 patients (27.3%) and thickened endometrium in 47 patients (72.3%). The mean endometrial thickness was 13.23±4.68 mm (range, 6 to 36 mm). We found benign disease in 62 patients (95.4%). Non-pathologic endometrium was present in 6 patients (9.2%), endometrial polyps in 35 patients (53.8%), endometrial hyperplasia without atypia in 17 patients (26.2%) and with atypia in 4 patients (6.1%). Abstract

Background and purpose: Blood platelets represent an attractive peripheral model in neuropsychopharmacology owing to serotonin transmembrane uptake mechanism - target for different psychoactive drugs, mainly antidepressants. The transport kinetics of serotonin (5-hydroxytryptamine, 5HT) through the plasma membrane is interpreted by the Michaelis-Menten equation and results expressed in terms of Km and Vmax. Published values of kinetic parameters for the 5HT transport into rat platelets vary considerably. The purpose of this paper was to evaluate possible reasons of these varioations, with special emphasis given on methodology of kinetic measurements. Material and methods: Platelet rich plasma was separated from anticoagulated blood samples (1 mL) obtained by jugular venipuncture from ether-anesthetized rats. Kinetic measurements were performed at 37 C by incubating various number of rat platelets with several 14C-5HT concentrations during different time intervals. The uptake was terminated by rapid cooling of incubating mixture and separation of platelets by filtration at different vacuum settings. Platelet rich plasma was stored at room temperature for different time periods prior to kinetic measurements. Serotonin, taken up by platelets, was quantified as beta counts of 14C in liquid scintillator. Calculation of kinetic parameters, Km and Vmax, was performed using Eadie-Hofstee plot. Results: Based on the results of methodological studies, experimental conditions which, by our experience, assure reliable and reproducible kinetic measurements of 5HT tranasport into rat platelets were defined as follows: 1) concentration of platelets in incubating mixture: 25-30×10^6/uL of incubating medium ; b) concentrations of substrate (14C-5HT) in incubating medium: 0.05, 0.07, 0.10, 0.15, 0.25, 0.50 and 1.00 uM ; c) uptake interval: 30 seconds ; d) vacuum setting for filtration: 600 mmHg ; e) recommended storage of platelet suspension prior to kinetic determinations: up to three hours at room temperature. Values of kinetic parameters for serotonin uptake into rat platelets obtained by this methodology were: Vmax = 225 +/- 37 pmol 5HT ×10^8 plt/min and Km = 0.161 +/- 0.023 uM (M +/- SD, N = 27). Conclusion: Based on thorough methodological studies the procedure for reliable measurement of kinetic parameters of serotonin uptake into rat platelets was described with critical review of available literature in the field. Such methodology can serve as a basis for further development of the experimental model for ex vivo monitoring of platelet serotonin uptake in rats.
