Background: Alterations of sexual functions are frequent in major depressive disorder (MDD), however there are little data on sexual functions in patients with treatment-resistant depression (TRD).Aim: To assess sexual desire impairment in patients with TRD.Methods: Baseline data were analyzed from an ongoing cohort study from an outpatient clinic specialized in TRD treatment in Brazil. The cohort comprised consecutive patients with the diagnosis of TRD that sought treatment in this center.Main outcome measures: The Hamilton Depression Rating Scale (HAMD) Genital Symptoms Item (item 14) was used as a proxy to assess sexual desire impairmentResults: Sixty-five participants with TRD were included. There was sexual desire impairment in 67.7% of patients. Men (87.5%) were more affected than women (61.2%) and depression severity was associated with greater complaints of this aspect of sexual function.Clinical Implications: Our findings suggest that health care professionals should systematically assess sexual desire in patients with TRD in daily clinical practice.Strengths & Limitations: To our knowledge, this is the first study to assess impairment in sexual desire in TRD. A major limitation that need to be addressed in future studies is the reliance on item 14 of the HAMD, which was not designed specifically for assessing sexual dysfunction.Conclusion: Participants with TRD had a high prevalence of sexual desire impairment, this being associated with greater depressive symptoms severity and male gender. Further longitudinal studies are needed, in larger samples, using specific instruments for assessing sexual dysfunction comparing TRD and non-TRD populations.

Sexual health is relevant throughout a person’s life; however, studying human sexuality is complex and requires particular care when working with individuals from different cultural and social backgrounds. Much of the research addressing this subject has been conducted in Western countries, and that in non-Western countries is generally based on small sample sizes. The biopsychosocial nature of sexuality and its dysfunctions should be taken into consideration given that it is indispensable when conducting and assessing sexual studies in different countries and cultures. Therefore, culturally sensitive studies that consider cultural contexts and determinants as well as social markers are needed. The topic of sexuality in Arab culture is still enigmatic. This enigma has impacted the advancement of sexual science and limited researchers, health care practitioners, and patients. Thus, the aim of this systematic literature review was to find and assemble all scales and questionnaires regarding human sexual health that have been translated into Arabic and validated in order to promote a critical analysis of the methods used in each instrument and to inform readers and researchers of the limits and potential of each scale. Electronic databases were systematically searched, and eight instruments were selected for inclusion: the Arabic Index of Premature Ejaculation (AIPE), the Sexual Health Inventory for Men (SHIM), the Arabic Female Sexual Function Index (ArFSFI), the Female Genital Self-Image Scale (AVFGSIS), the Arabic Arizona Sexual Experiences Scale (ASEX), the Egyptian Pelvic Organ Prolapse/Incontinence Sexual Questionnaire (PISQ-IR), the Saudi Arabian Pelvic Organ Prolapse/Urinary Incontinence Sexual Questionnaire in Arabic (PSIQ-IR), and the Arabic Female Sexual Distress Scale (FSDS). All included instruments showed good validity and reliability for the target population. Future studies are needed to develop culturally sensitive instruments

The physical barrier of the intestine and associated mucosal immunity maintains a delicate homeostatic balance between the host and the external environment by regulating immune responses to commensals, as well as functioning as the first line of defense against pathogenic microorganisms. Understanding the orchestration and characteristics of the intestinal mucosal immune response during commensal or pathological conditions may provide novel insights into the mechanisms underlying microbe-induced immunological tolerance, protection, and/or pathogenesis. Over the last decade, our knowledge about the interface between the host intestinal mucosa and the gut microbiome has been dominated by studies focused on bacterial communities, helminth parasites, and intestinal viruses. In contrast, specifically how commensal and pathogenic protozoa regulate intestinal immunity is less well studied. In this review, we provide an overview of mucosal immune responses induced by intestinal protozoa, with a major focus on the role of different cell types and immune mediators triggered by commensal (Blastocystisspp. andTritrichomonasspp.) and pathogenic (Toxoplasma gondii,Giardia intestinalis,Cryptosporidium parvum) protozoa. We will discuss how these various protozoa modulate innate and adaptive immune responses induced in experimental models of infection that benefit or harm the host.

Mycobacterium tuberculosis (Mtb) infection induces pulmonary expression of the heme-degrading enzyme heme oxygenase-1 (HO-1). We have previously shown that pharmacological inhibition of HO-1 activity in experimental tuberculosis results in decreased bacterial loads and unexpectedly that this outcome depends on the presence of T lymphocytes. Here, we extend these findings by demonstrating that IFNγ production by T lymphocytes and NOS2 expression underlie this T-cell requirement and that HO-1 inhibition potentiates IFNγ-induced NOS2-dependent control of Mtb by macrophages in vitro. Among the products of heme degradation by HO-1 (biliverdin, carbon monoxide, and iron), only iron supplementation reverted the HO-1 inhibition-induced enhancement of bacterial control and this reversal was associated with decreased NOS2 expression and NO production. In addition, we found that HO-1 inhibition results in decreased labile iron levels in Mtb-infected macrophages in vitro and diminished iron accumulation in Mtb-infected lungs in vivo. Together these results suggest that the T-lymphocyte dependence of the therapeutic outcome of HO-1 inhibition on Mtb infection reflects the role of the enzyme in generating iron that suppresses T-cell-mediated IFNγ/NOS2-dependent bacterial control. In broader terms, our findings highlight the importance of the crosstalk between iron metabolism and adaptive immunity in determining the outcome of infection.

This paper provides an integrative review of psychometric instruments targeting sexual function and clinical measures of sexuality available to the Brazilian population. Our aim was to summarize the existing measures, indicating current development needs related to the quality of psychometric evidence, research design and untapped theoretical ground that could be relevant for clinical practice and research in sexuality. Electronic databases were searched. Additional records were found through a manual search. The general search terms and operators were (scale OR inventory OR questionnaire) AND (validation) AND (sexual beliefs OR sexuality OR sexual behavior OR sexual function OR sexual satisfaction) AND (Brazil or Brazilian). The results indicate a total of 18 developed or transculturally adapted and/or validated scales and questionnaires available in Brazilian Portuguese, covering sexual dysfunctions and clinical measures. Despite development, adaptation and validation efforts, there is a critical need for better psychometric and sampling strategies as well as norms for the clinical interpretation of test results. Ecological validity is also a challenge. At present, sexual assessments in Brazil offer some useful information, but reaching the applied clinical field remains a challenge.Supplemental data for this article is available online at https://doi.org/10.1080/0092623X.2022.2035869 .

The apicomplexan intracellular parasiteToxoplasma gondiiis a major food borne pathogen with significant impact in children and during pregnancy. The majority of theT. gondiiproteome remains uncharacterized and the organization of proteins into complexes is unclear. To overcome this knowledge gap, we utilize a biochemical fractionation strategy coupled with mass spectrometry to predict interactions by correlation profiling. Key to this approach is the integration of additional datasets based on gene co-expression as well as phylogenetic profiles that eliminate poorly supported interactions and reduce the number of false positive interactions. In addition to a supervised machine learning strategy, we employed an unsupervised approach in data integration, based on similarity network fusion, to overcome the deficit of high-quality training data in non-model organisms. The resulting high confidence network, we term ToxoNet, comprises 2,063 interactions connecting 652 proteins. Clustering of this network identifies 93 protein complexes, predicting both novel complexes as well as new components for previously known complexes. In particular, we identified clusters enriched in mitochondrial machinery that include previously uncharacterized proteins that likely represent novel adaptations to oxidative phosphorylation. Furthermore, complexes enriched in proteins localized to secretory organelles and the inner membrane complex, predict additional novel components representing novel targets for detailed functional characterization. We present ToxoNet as a publicly available resource with the expectation that it will help drive future hypotheses within the research community.

The objective of this project was to evaluate the supply of narasin on the performance of ewes and their lambs in the lactation phase (Exp. I) and feedlot lambs (Exp. II). Experiment I: Thirty-two ewes with ten days of lactation were used, together with their lambs (s). The experimental design was randomized complete blocks. In this complex, carbohydrate recognition domains of MIC1 and MIC4 are exposed and interact with terminal sialic acid and galactose residues, respectively, of host cell glycans. Recently, we demonstrated that MIC1 and MIC4 binding to the N-glycans of Toll-like receptor (TLR) 2 and TLR4 on phagocytes triggers cell activation and pro-inflammatory cytokine production. Herein, we investigated the requirement for TLR2 heterodimerization and co-receptors in MIC-induced responses, as well as the signaling molecules involved. We used MICs to stimulate macrophages and HEK293T cells transfected with TLR2 and TLR1 or TLR6, both with or without the co-receptors CD14 and CD36. Then, the cell responses were analyzed, including nuclear factor-kappa B (NF-&kappa, B) activation and cytokine production, which showed that (1) only TLR2, among the studied factors, is crucial for MIC-induced cell activation, (2) TLR2 heterodimerization augments, but is not critical for, activation, (3) CD14 and CD36 enhance the response to MIC stimulus, and (4) MICs activate cells through a transforming growth factor beta-activated kinase 1 (TAK1)-, mammalian p38 mitogen-activated protein kinase (p38)-, and NF-&kappa, B-dependent pathway. Remarkably, among the studied factors, the interaction of MIC1 and MIC4 with TLR2 N-glycans is sufficient to induce cell activation, which promotes host protection against T. gondii infection.

Despite developments in panic disorder (PD) research, a significant percentage of patients do not benefit from conventional treatments. Interpersonal factors have been identified as potential predictors of treatment failures. We aimed to evaluate assertiveness in a sample of patients with PD and its implications for treatment. Forty-six symptomatic patients with PD and 46 college students responded to assessment scales regarding assertiveness and clinical data. Seventy-five percent of the patients had a secondary diagnosis of agoraphobia. We found that the PD group was characterized as nonassertive and slightly less assertive than control subjects. Furthermore, the deficit in the level of assertiveness correlated with the severity of the PD. The diagnosis of agoraphobia was correlated with unassertiveness (p < 0.05). Agoraphobia predisposes individuals to dependency and insecurity about their ability to overcome anxiogenic situations. These data demonstrate the importance of managing assertiveness in patients with PD accompanied by agoraphobia.

A geração fotovoltaica (FV) tem crescido cada vez mais em uma escala mundial e tudo indica que continuará crescendo com o tempo. Este trabalho tem como objetivo analisar as políticas de inovação em relação aos modelos adotados a fim de disseminar a energia FV nos cinco maiores países produtores desta energia e no Brasil, visando possibilidades e perspectivas para o mercado brasileiro. Esta pesquisa foi realizada na base Scopus a partir do portal capes com intuito de reunir trabalhos referentes aos sistemas FV, políticas inovadoras e incentivos utilizados para que esta tecnologia fosse difundida nos países escolhidos, sendo possível analisar o cenário brasileiro neste contexto. Nos países onde esta tecnologia foi disseminada com sucesso foi percebido um grande apoio de políticas e incentivos governamentais. O Brasil, embora possua recentes incentivos fiscais, foi identificado que para o crescimento desta ciência, carece de maiores investimentos públicos.

Infection of host cells by Toxoplasma gondii is an active process, which is regulated by secretion of microneme (MICs) and rhoptry proteins (ROPs and RONs) from specialized organelles in the apical pole of the parasite. MIC1, MIC4 and MIC6 assemble into an adhesin complex secreted on the parasite surface that functions to promote infection competency. MIC1 and MIC4 are known to bind terminal sialic acid residues and galactose residues, respectively and to induce IL-12 production from splenocytes. Here we show that rMIC1- and rMIC4-stimulated dendritic cells and macrophages produce proinflammatory cytokines, and they do so by engaging TLR2 and TLR4. This process depends on sugar recognition, since point mutations in the carbohydrate-recognition domains (CRD) of rMIC1 and rMIC4 inhibit innate immune cells activation. HEK cells transfected with TLR2 glycomutants were selectively unresponsive to MICs. Following in vitro infection, parasites lacking MIC1 or MIC4, as well as expressing MIC proteins with point mutations in their CRD, failed to induce wild-type (WT) levels of IL-12 secretion by innate immune cells. However, only MIC1 was shown to impact systemic levels of IL-12 and IFN-γ in vivo. Together, our data show that MIC1 and MIC4 interact physically with TLR2 and TLR4 N-glycans to trigger IL-12 responses, and MIC1 is playing a significant role in vivo by altering T. gondii infection competency and murine pathogenesis., Author summary Toxoplasmosis is caused by the protozoan Toxoplasma gondii, belonging to the Apicomplexa phylum. This phylum comprises important parasites able to infect a broad diversity of animals, including humans. A particularity of T. gondii is its ability to invade virtually any nucleated cell of all warm-blooded animals through an active process, which depends on the secretion of adhesin proteins. These proteins are discharged by specialized organelles localized in the parasite apical region, and termed micronemes and rhoptries. We show in this study that two microneme proteins from T. gondii utilize their adhesion activity to stimulate innate immunity. These microneme proteins, denoted MIC1 and MIC4, recognize specific sugars on receptors expressed on the surface of mammalian immune cells. This binding activates these innate immune cells to secrete cytokines, which promotes efficient host defense mechanisms against the parasite and regulate their pathogenesis. This activity promotes a chronic infection by controlling parasite replication during acute infection.

Objetivo: O estudo com terapia cognitivo-comportamental (TCC) procurou demonstrar a preparaçãodo paciente com transtorno do pânico para exposições agorafóbicas. Foram usadas as técnicas dasexposições interoceptivas, às próprias sensações corporais, e in vivo, a locais ou situações temidas.Metodologia: 50 indivíduos, divididos em dois grupos de 25 participantes cada um. O grupo 1 realizou 10 sessões de TCC semanais e individuais com uma hora de duração e fez uso de medicamentos e o grupo 2, controle, usou apenas medicação sem TCC. A medicação prescrita consistiu em antidepressivos tricíclicos e inibidores seletivos de recaptação da serotonina. Foram aplicados instrumentos de avaliação no início e ao fim das intervenções médica e psicológica. Resultados: Observamos uma diferença significativa entre a avaliação inicial e final no grupo 1, como: redução nos ataques de pânico, ansiedade antecipatória, esquiva agorafobia e medo das sensações corporais. Na escala de avaliação global do funcionamento, um aumento do bem-estar global de 60.8% a 72.5% entre pacientes do grupo 1 com terapia, diferentemente do grupo 2 sem terapia. Conclusões: As técnicas de TCC para as exposições foram consideradas essenciais na preparação dos pacientes com transtorno de pânico, para enfrentarem os ataques de pânico e as situações agorafóbicas subsequentes.

Metabolic syndrome (MS) seems to be associated with both depression and increased cardiovascular risk. This article emphasizes the most relevant findings on the link between MS and psychological cardiovascular risk factors, focusing on the impact of anxiety. There is evidence for an association between psychological disorders and the development of MS, which could eventually increase cardiovascular risk. However, the coexistence of anxiety in MS patients seems to be a byproduct of anxiety-depression comorbidity, stress and negative health behaviors. Endocannabinoid receptors, and hypothalamic-pituitary-adrenal axis dysregulation and sympathetic hyperactivation are the most commonly mentioned plausible underlying pathways.

INTRODUÇÃO: Há um crescente interesse e necessidade em avaliar a qualidade da produção científica dos pesquisadores. Para tal, as métricas mais importantes são: número de citações, número médio de citações por artigo indexado e o índice-H. Contudo, essas métricas apresentam limitações na avaliação do potencial de impacto das publicações de um pesquisador, especialmente dentre aqueles mais produtivos. OBJETIVO: Propor e demonstrar a utilização de uma nova métrica científica - índice-H dos artigos citantes, que permite refinar a discriminação do impacto das publicações de pesquisadores experientes sobre o conhecimento existente em sua área. MÉTODOS: Foram analisados dados da Web of Science de 13 dentre os pesquisadores doutores brasileiros mais produtivos na área de exercício físico e esporte, incluindo: número de artigos publicados, número de citações, número médio de citações por artigos, índice-H e o índice-H dos artigos citantes, ou seja, o índice-H obtido a partir da ordenação decrescente dos artigos que citaram artigos publicados pelo pesquisador. Dados de quatro outros pesquisadores - brasileiros e estrangeiros - foram usados como marcos de referência para comparações. RESULTADOS: Os índice-H do pesquisador e o dos artigos citantes são associados (r = 0,92; p > 0,01), porém, quando são analisados os seis pesquisadores mais produtivos, com índices-H acima de 7, a associação entre os dois índices-H desaparece (r = 0,35; p = 0,49). CONCLUSÃO: O índice-H dos artigos citantes pode contribuir para diferenciar a produção científica de pesquisadores com um grande número de artigos publicados. Sugere-se a adoção de sua mensuração pelas agências brasileiras e estrangeiras de fomento e de avaliação da produção científica. BACKGROUND: there has been an increasing interest and need in evaluating the quality of scientific papers written by researchers. Therefore, the most important measurements used are the number of citations, the average number of citations per article published in indexed journals and the H-index. Those bibliometric indicators present, however, limitations when assessing the potential impact of the publications of a given researcher, particularly among the most productive ones. OBJECTIVE: To propose and to demonstrate the use of a new scienciometric strategy - the H-index of the citing articles-, that allow to better discriminate the impact of a given researcher to the body of knowledge in the respective research area. METHODS: Research data - Web of Science - from 13 of the most productive Brazilian PhD researchers in the field of exercise and sport sciences were analyzed, including: number of publications, number of citations, average number of citations per article, H-index and the proposed H-index of citing articles (the H-index extracted from the ranking of articles that cited the papers published by a given researcher). Data from four other researchers - Brazilian and foreign - were used as reference for comparisons. RESULTS: The researcher's H-index and the H-index of the citing articles were associated (r=.92; p>.01). However, when the six most productive authors were analyzed with H-index above 7, the association between the two indexes disappears (r=.35; p=.49). CONCLUSIONS: The H-index of the citing articles can be useful to discriminate the scientific production of authors with a high number of published papers. It is suggested that this algorithm should be adopted by Brazilian and foreign financing and scientific production evaluation agencies.

Psychological factors such as stress and depression have already been established as primary and secondary cardiovascular risk factors. More recently, the role of anxiety in increasing cardiac risk has also been studied. The underlying mechanisms of increased cardiac risk in panic disorder patients seem to reflect the direct and indirect effects of autonomic dysfunction, as well as behavioral risk factors associated with an unhealthy lifestyle. Implications of the comorbidity between panic and cardiovascular disease include higher morbidity, functional deficits, increased cardiovascular risk, and poor adherence to cardiac rehabilitation or exercise programs. This article probes the most recent evidence on the association between coronary artery disease, anxiety and panic disorder, and discusses the potential role of incorporating regular physical exercise and cognitive behavioral therapy in the treatment of this condition.

INTRODUÇÃO: O medo de dirigir pode restringir a vida do indivíduo e gerar sérios problemas pessoais, sociais e ocupacionais. Usar um instrumento adequado para a avaliação de cognições do paciente é importante, já que contribui para uma formulação dos problemas apresentados, podendo guiar um plano de tratamento coerente. O objetivo deste estudo foi apresentar as etapas de tradução e adaptação para a língua portuguesa do instrumento Driving Cognitions Questionnaire, que mede as cognições que podem estar presentes no medo de dirigir MÉTODOS: Três traduções e três retrotraduções foram realizadas por seis avaliadores independentes. Realizou-se a equivalência semântica e avaliação das versões, elaborando-se uma versão síntese. Comentários dos 10 participantes sobre a versão preliminar do questionário foram examinados. Foi construída a versão brasileira preliminar do questionário. RESULTADOS: A maioria dos participantes entendeu as descrições das cognições expostas no questionário em português. CONCLUSÃO: A utilização de três versões de tradução e retrotradução, a discussão sobre a versão síntese preliminar e a interlocução com a população-alvo proporcionaram viabilidade ao processo de equivalência semântica da versão final brasileira.INTRODUCTION: The fear of driving may restrict the life of patients and cause serious personal, social, and occupational problems. It is important to use a suitable tool while assessing patients’ cognitions, once these findings may help formulate the problems observed and may guide the selection of a consistent treatment plan. The objective of this study was to describe the stages of translation and adaptation into Brazilian Portuguese of the Driving Cognitions Questionnaire, designed to measure cognitions that may be present in the fear of driving. METHODS: Three translations and three back translations were conducted by six independent evaluators. Semantic equivalence assessment and evaluation of the different versions were conducted and resulted in a synthesized version of the instrument. Comments made by 10 participants on the preliminary version of the questionnaire were examined. The preliminary Brazilian version of the questionnaire was developed. RESULTS: Most participants understood the cognitions as described in the Brazilian Portuguese version of the questionnaire. CONCLUSION: The use of three different translations and back translations, discussion of a preliminary synthetic version and interaction with the target population contributed to the viability of the process aimed to assess the semantic equivalence of the Brazilian final version.

Este trabalho consiste no relato da utilização de um programa de exercícios físicos aeró-bicos intervalados no contexto da terapia cognitivo-comportamental em um caso de transtorno de pânico e agorafobia. São descritos os procedimentos utilizados, bem como os resultados positivos obtidos na redução da ansiedade cardíaca e na ansiedade cotidiana em relação a situações que exigem esforço físico. The present article reports the use of an interval aerobic exercise program as an adjunct intervention in the context of cognitive behavioural therapy in a patient with panic disorder and agoraphobia. We describe the procedures used in the exercise program and also the positive results achieved regarding cardiac anxiety reduction and everyday anxiety reduction concerning activities that requested physical effort.

CONTEXTO: Aspectos psicológicos como estresse e depressão já são reconhecidos como fatores de risco cardiovascular. Mais recentemente, o impacto da ansiedade passou a ser objeto de estudo. OBJETIVO: Identificar a prevalência de transtornos psiquiátricos e a presença de ansiedade relacionada à saúde e de ansiedade cardíaca em coronariopatas participantes de um programa de exercício supervisionado. MÉTODOS: Quarenta e dois homens coronariopatas foram entrevistados com o Mini International Neuropsychiatric Interview (MINI), versão 5.0, e solicitados a preencher a Escala de Sensibilidade à Ansiedade, o Questionário de Cognições Agorafóbicas, a Escala de Sensações Corporais e o Questionário de Ansiedade Cardíaca. RESULTADOS: Dentre os participantes, 38% apresentaram um ou mais diagnósticos psiquiátricos pelo MINI, mas apenas 19% apresentaram múltiplas comorbidades. A presença de transtornos psiquiátricos encontra-se associada a escores mais altos de ansiedade relacionada à saúde e à ansiedade cardíaca (p < 0,05). Não foi encontrada relação entre os escores de ansiedade e a gravidade da doença cardiovascular (p > 0,05). CONCLUSÕES: Ansiedade relacionada à saúde parece estar mais fortemente associada à presença de comorbidades psiquiátricas do que à gravidade do quadro cardiovascular. Recomenda-se a triagem regular de pacientes coronariopatas para transtornos psiquiátricos. Estratégias terapêuticas complementares como exercício físico e psicoterapia podem ser alternativas terapêuticas complementares.

OBJETIVO: A traducao, a afericao da equivalencia semântica e uma aplicacao piloto (n = 30), sem pretensao psicometrica, do Questionario de Atividade Fisica Habitual, visando sua utilizacao na populacao brasileira de diferentes niveis de escolaridade. METODOS: O processo envolveu duas traducoes e retrotraducoes realizadas por avaliadores independentes, avaliacao das versoes seguida da elaboracao de uma versao sintese e pre-teste comentado. RESULTADOS: A maioria dos participantes (91%) nao apresentou dificuldades de compreensao com o questionario. Para cada item do instrumento, apresentam-se os resultados das quatro etapas. Mais estudos sao necessarios para determinar a adequacao para populacoes de baixa escolaridade. Os autores recomendam que sujeitos menos instruidos sejam supervisionados ao preencher o questionario. CONCLUSOES: A utilizacao de duas versoes de traducao e retrotraducao, a discussao sobre a versao sintese e a interlocucao com a populacao-alvo proporcionam maior seguranca ao processo de equivalencia semântica.

However, there is a lack of self-report instruments to assess levels of physical activity adapted to Brazilian Portuguese. OBJECTIVE: To translate, assess the semantic equivalence of the Habitual Physical Activity Questionnaire and perform a non-psychometric pre-test with subjects (n = 30) from the Brazilian population, with different educational backgrounds. METHODS: The cross-cultural adaptation process consisted of two translations and back translations performed by two independent evaluators; an evaluation of the versions and the development of a synthetic version; and a commented pretest of the questionnaire. RESULTS: For each item of the instrument, the results of the four stages are reported. Most of the participants (91%) did not present any difficulties comprehending the items of the instrument. Further studies should be addressed to determine the adequacy of using this instrument in the less-educated population. We recommend that less instructed subjects be supervised while responding the questionnaire. DISCUSSION: The use of two translations versions, their critical appraisal and the assessment of the target population conceives more safety to the process of semantic equivalence.

Individuals with PD experience unexpected panic attacks, characterized by anxiety and fear, resulting in a number of autonomic and respiratory symptoms. Respiratory stimulation is a common event during panic attacks. The respiratory abnormality most often reported in PD patients is increased CO2 sensitivity, which has given rise to the hypothesis of fundamental abnormalities in the physiological mechanisms that control breathing in PD. There is evidence that PD patients with dominant respiratory symptoms are more sensitive to respiratory tests than are those who do not manifest such symptoms, and that the former group constitutes a distinct subtype. Patients with PD tend to hyperventilate and to panic in response to respiratory stimulants such as CO2, triggering the activation of a hypersensitive fear network. Although respiratory physiology seems to remain normal in these subjects, recent evidence supports the idea that they present subclinical abnormalities in respiration and in other functions related to body homeostasis. The fear network, composed of the hippocampus, the medial prefrontal cortex, the amygdala and its brain stem projections, might be oversensitive in PD patients. This theory might explain why medication and cognitive-behavioral therapy are both clearly effective. Our aim was to review the relationship between respiration and PD, addressing the respiratory subtype of PD and the hyperventilation syndrome, with a focus on respiratory challenge tests, as well as on the current mechanistic concepts and the pharmacological implications of this relationship.

Indivíduos com TP experimentam ataques de pânico inesperados, caracterizados por ansiedade, medo e diversos sintomas autonômicos e respiratórios. A estimulação respiratória é um fenômeno comum durante os ataques de pânico. A anormalidade respiratória mais citada em pacientes com TP é a sensibilidade aumentada para o CO2, que originou a hipótese de uma disfunção fundamental nos mecanismos fisiológicos de controle da respiração no TP. Há evidências de que pacientes com TP com sintomas respiratórios predominantes são mais sensíveis a testes respiratórios do que aqueles sem a manifestação de tais sintomas, representando um subtipo distinto. Pacientes com TP tendem a hiperventilar e a reagir com pânico como resposta a estimulantes respiratórios como o CO2, gerando uma ativação de um circuito de medo hipersensível. Apesar de a fisiologia respiratória desses pacientes permanecer normal, algumas evidências recentes apontam a presença de disfunções subclínicas na respiração e em outras funções relacionadas à homeostase corporal. O circuito do medo, composto pelo hipocampo, córtex pré-frontal medial, amígdala e projeções do tronco cerebral, pode estar hipersensível em pacientes com TP. Essa teoria pode explicar porque os medicamentos e a terapia cognitivocomportamental são claramente eficazes. Nosso objetivo foi revisar a relação entre respiração e TP, especialmente o subtipo respiratório de TP e a síndrome da hiperventilação, focalizando os testes respiratórios, bem como as hipóteses mecanísticas e as implicações farmacológicas dessa relação.

OBJETIVO: Estresse e depressão já são considerados fatores de risco para o desenvolvimento e o agravamento de doenças cardiovasculares. Os transtornos de ansiedade têm sido fortemente associados às cardiopatias nos últimos anos. O transtorno de pânico em cardiopatas representa um desafio em termos de diagnóstico e tratamento. Atualizar o leitor quanto ao status da associação entre transtornos de ansiedade, especialmente transtorno de pânico, e cardiopatias. MÉTODO: Foi realizada uma busca nas bases de dados ISI e Medline, com as palavras-chave: "heart disease", "coronary disease", "anxiety", "panic disorder" e "autonomic function". Foram selecionados os artigos publicados a partir de 1998. DISCUSSÃO: O padrão autonômico encontrado em pacientes com transtorno de pânico, em particular a redução da variabilidade cardíaca, é apontado como o provável fator mediador do impacto cardiovascular do transtorno de pânico. CONCLUSÕES: Apesar de a associação entre transtornos de ansiedade e doenças cardiovasculares estar atualmente bastante estabelecida, existem ainda diversas lacunas no estado atual do conhecimento. São recomendadas a terapia cognitivo-comportamental e a prática de exercícios físicos supervisionados como potenciais coadjuvantes na intervenção terapêutica.OBJECTIVE: Psychosocial stress and depression have already been established as risk factors for developing and worsening cardiovascular diseases. Anxiety disorders are been strongly associated to cardiac problems nowadays. Panic disorder in cardiac patients represents a challenge for diagnose and treatment. Update the reader on the status of the association between anxiety disorders, particularly panic disorder, in cardiac patients. METHOD: Were retrieved papers published at ISI and Medline databases since 1998. Key-words used were: "heart disease", "coronary disease", "anxiety", "panic disorder" and "autonomic function". DISCUSSION: The characteristic autonomic pattern found in patients with panic disorder, particularly the reduction in heart rate variability, are discussed as potential mediators of the cardiovascular impact of panic disorder. CONCLUSIONS: Despite de established association between panic disorder and cardiac diseases, current literatures data points to a knowledge gap on treating and diagnosing the comorbidity between panic disorder and cardiovascular diseases. Cognitive-behavioral therapy and regular supervised exercise are recommended as potential adjunct therapeutic interventions.

O estudo desenvolveu e analisou as características psicométricas de um Inventário de Empatia (IE). A partir da identificação de 16 situações de interação social, fornecidas pela literatura, foi construída uma escala inicial com 74 itens, baseados nos componentes cognitivos, afetivos e comportamentais da empatia, com afirmativas do tipo Likert, e aplicada em 715 universitários de ambos os sexos, das áreas: humana, biomédica, ciências sociais e exatas. As análises fatoriais dos componentes principais, com rotação oblíqua, identificaram quatro fatores representativos da estrutura interna do instrumento: Tomada de Perspectiva, Flexibilidade Interpessoal, Altruísmo e Sensibilidade Afetiva. Tais fatores apresentaram índices de consistência interna que variaram de 0,72 a 0,85, calculados através do coeficiente Alfa de Cronbach. Com base nas características psicométricas do IE, recomenda-se o seu uso tanto em pesquisa quanto em clínica.

Algumas alterações semânticas foram realizadas com o intuito de facilitar a compreensão do instrumento. We also examined comments provided by participants on the preliminary version of the questionnaire and used them for the development of the final version. Participants with tertiary education had no difficulties comprehending the translated items of the questionnaire, only pointing item 5 as ambiguous. Participants from the lower educational level reported comprehension problems regarding items 2, 4, 6, 7, 10, 11 and 14. Some small changes were made in our first version to enhance comprehensibility. CONCLUSION: The use of two versions of translations, a critical examination of the two versions, and suggestions made by participants resulted in a final Brazilian version with a satisfactory degree of semantic accuracy and semantic equivalence with the original version.

Os transtornos psiquiátricos relacionados à distorção da auto-imagem corporal são marcados por elevado grau de sofrimento físico e psíquico, associado a perdas sociais e ocupacionais significativas, além de produzirem comportamentos danosos para a saúde física do paciente. A dismorfia muscular (DISMUS) é um subtipo do transtorno dismórfico corporal, que se caracteriza por uma preocupação específica com o tamanho do corpo e com o desenvolvimento dos músculos. É um transtorno presente quase exclusivamente em homens que, apesar da visível hipertrofia, procuram ativamente aumentar sua massa muscular, através de exercícios excessivos, do uso de substâncias ergogênicas e de dietas hiperprotéicas. O presente estudo teve como objetivo avaliar a validade de constructo de um critério antropométrico (Índice B/P - relação entre a perimetria do braço contraído e da perna), anteriormente proposto, para suspeita diagnóstica de DISMUS. Uma amostra (n = 100) de homens, praticantes de treinamento de força, com idade entre 18 e 35 anos, teve suas medidas antropométricas comparadas os os escores obtidos em uma escala usada para o diagnóstico da DISMUS (MASS - Muscle Appearance Satisfaction Scale) e também com as medidas de um grupo de indívíduos de características semelhantes, avaliados com o objetivo de obter orientação para a prática de exercícios físicos (n = 313). A análise dos resultados indica que um indivíduo com índice B/P > 1 possuem alta probabilidade de apresentar um escore indicativo de DISMUS no instrumento MASS e sugere ser coerente a aplicação do índice B/P como um indicador antropométrico válido para o diagnóstico de DISMUS. O critério B/P pode ser um potente instrumento usado por profissionais da área de exercício para auxiliar no diagnóstico e tratamento precoce desses indivíduos.Psychiatric disorders related to body image distortions are highly associated with physical and psychological distress and significant social and ocupacional disfunction. Also, they are caracterized for inducing behaviors that can be harmful for the patient's health. Muscle Dysmorfia (MD) is a subtype of Body Dysmorfic Disorder, marked by specific and recurrent worring about muscle appearance and hipertrophy. This disorder is significantly more prevalent in men and, despite the actual level of muscle hipertrophy, the subjects seek increasing their body mass by excessive weight lifting, engaging in hiperproteic diets and using anabolic substances. The present study aimed to evaluate the construct validity of an antropometric criteria (The B/P index - the ratio between the perimeter of the contracted arm and the perimeter of the leg), previously proposed, as an antropometric indicator to suspect of MD in weight lifters. A sample (n = 100) of male body builders, between 18 and 35 years old, were measured and responded the Muscle Appearance Satisfaction Scale (MASS). Their cineantropometric measures were also compared to the ones from another sample (n = 313), with similar demografic caracteristics, evaluated to start an exercise program. Data analysis sugest that a subjetc with B/P > 1 have a higher probability of presenting a score that would indicate MD in the MASS. This results indicate that the B/P index can be used as a valid antropometric criteria to help exercise professionals suspect of MD in individuals with B/P > 1 and reffer them to especialized professionals for diagnose and treatment.

According to cognitive behavioral models (CBT), panic attacks (PA) arise from distorted and catastrophic interpretations of bodily symptoms. As exercising involves exposure to physiological stimuli similar to those experienced during PAs, patients often experience anxiety and avoid exercising. Exercise avoidance and low levels of everyday physical activity turn out to promote a sedentary lifestyle as an indirect effect of Panic Disorder (PD), with deleterious health impacts. Interoceptive exposure techniques – the voluntary exposure of the patient to autonomic manifestations – have a therapeutic effect in the treatment of panic by promoting habituation to physiological cues contributing to break the hypervigilance-anxiety-panic-avoidance cycle. Preliminary results indicating a potential positive role of aerobic exercise interventions designed to promote interoceptive habituation in the treatment of PD have been reported. The inclusion of an exercise protocol in the context of CBT interventions, may enhance motivation to participate and endure the discomfort and anxiety provoked by exercise using exposure therapy rationale. This chapter reviews and discusses the main results available in the literature of including exercise in the treatment of PD, as well as provides an unique cognitive behavioral perspective to the understanding of the relationship between panic disorder and exercise. We also provide specific recommendations for exercise testing, adherence promotion and particular issues that need to be addresses to successfully include PD patients in exercise protocols, based on authors experience.

Toxoplasma gondii is an obligate intracellular protozoan parasite found worldwide that is able to chronically infect almost all vertebrate species, especially birds and mammalians. Chitinases are essential to various biological processes, and some pathogens rely on chitinases for successful parasitization. Here, we purified and characterized a chitinase from T. gondii. The enzyme, provisionally named Tg_chitinase, has a molecular mass of 13.7 kDa and exhibits a Km of 0.34 mM and a Vmax of 2.64. The optimal environmental conditions for enzymatic function were at pH 4.0 and 50 °C. Tg_chitinase was immunolocalized in the cytoplasm of highly virulent T. gondii RH strain tachyzoites, mainly at the apical extremity. Tg_chitinase induced macrophage activation as manifested by the production of high levels of pro-inflammatory cytokines, a pathogenic hallmark of T. gondii infection. In conclusion, to our knowledge, we describe for the first time a chitinase of T. gondii tachyzoites and provide evidence that this enzyme might influence the pathogenesis of T. gondii infection.

Sexual activity (SA) encompasses several behaviors such as kissing (Ki), touching (T), oral (O) stimulation, masturbation (M), and vaginal/anal intercourse (I). The acronym KiTOMI is proposed here to represent these behaviors. SA, particularly coitus, is a major aspect of health-related quality of life and is often considered the most pleasant and rewarding exercise performed during an entire lifetime. Although several studies have been conducted on sexuality, relatively limited information is available regarding SA in patients with heart disease. Moreover, the level of evidence of this limited information is nearly always B or C. This article provides a comprehensive and updated review of the relevant literature and offers evidence and expert-based practical messages regarding SA in patients with heart disease. Considering the rationale for exercise prescription, SA is typically well tolerated by most clinically stable patients with heart disease. Even in more debilitated and sicker individuals, KiT activities would most likely be feasible and desirable. The absolute risk of major adverse cardiovascular events during SA is typically very low. Even lower death rates have been reported for specific groups, such as women in general, aerobically fit men, and asymptomatic young adults with congenital heart disease. Finally, we emphasize the relevance of sexual counselling for patients and their partners, including the proper use of medications to treat erectile dysfunction. Counselled patients will be reassured and adequately informed regarding how to gradually resume habitual SA after a major cardiac event or procedure, starting with KiT and progressively advancing to KiTOM until all KiTOMI activities are allowed.

Toxoplasmosis, a zoonotic disease caused by Toxoplasma gondii, is an important public health problem and veterinary concern. Although there is no vaccine for human toxoplasmosis, many attempts have been made to develop one. Promising vaccine candidates utilize proteins, or their genes, from microneme organelle of T. gondii that are involved in the initial stages of host cell invasion by the parasite. In the present study, we used different recombinant microneme proteins (TgMIC1, TgMIC4, or TgMIC6) or combinations of these proteins (TgMIC1-4 and TgMIC1-4-6) to evaluate the immune response and protection against experimental toxoplasmosis in C57BL/6 mice. Vaccination with recombinant TgMIC1, TgMIC4, or TgMIC6 alone conferred partial protection, as demonstrated by reduced brain cyst burden and mortality rates after challenge. Immunization with TgMIC1-4 or TgMIC1-4-6 vaccines provided the most effective protection, since 70% and 80% of mice, respectively, survived to the acute phase of infection. In addition, these vaccinated mice, in comparison to non-vaccinated ones, showed reduced parasite burden by 59% and 68%, respectively. The protective effect was related to the cellular and humoral immune responses induced by vaccination and included the release of Th1 cytokines IFN-γ and IL-12, antigen-stimulated spleen cell proliferation, and production of antigen-specific serum antibodies. Our results demonstrate that microneme proteins are potential vaccines against T. gondii, since their inoculation prevents or decreases the deleterious effects of the infection.

Panic disorder (PD) patients often report respiratory symptoms and tend to perform poorly during maximal cardiopulmonary exercise testing (CPX), at least partially, due to phobic anxiety. Thus, we hypothesized that a submaximal exercise variable, minimum VE/VO2 - hereafter named cardiorespiratory optimal point (COP) -, may be useful in their clinical assessment. Data from 2,338 subjects were retrospectively analyzed and 52 (2.2%) patients diagnosed with PD (PDG) (70% women; aged 48±13 years). PD patients were compared with a healthy control group (CG) precisely matched to number of cases, age and gender profiles. PDG was further divided into two subgroups, based on having achieved a maximal or a submaximal CPX (unwilling to continue until exhaustion). We compared COP, VO2 max, maximum heart rate (HR max) between PDG and CG, and also COP between maximal and submaximal PD subgroups. COP was similar between PDG and CG (21.9±0.5 vs. 23.4±0.6; p = 0.07), as well as, for PD subgroups of maximal and submaximal CPX (22.0±0.5 vs. 21.6±1.3; p = 0.746). Additionally, PD patients completing a maximal CPX obtained VO2 max (mL x kg-1 x min-1) (32.9±1.57 vs 29.6±1.48; p = 0.145) and HR max (bpm) similar to controls (173±2.0 vs 168±2.7; p = 0.178). No adverse complications occurred during CPX. Although clinically safe, it is sometimes difficult to obtain a true maximal CPX in PD patients. Normalcy of cardiorespiratory interaction at submaximal effort as assessed by COP may contribute to reassure both patients and physicians that there is no physiological substrate for exercise-related respiratory symptoms often reported by PD patients.

OBJECTIVES: The anxiolytic effect of regular aerobic exercise in panic disorder patients is well known. However, a protocol for aerobic exercise intervention as an adjunct non-pharmacologic treatment for panic disorder is still lacking. Our aim was to propose and present a pilot study about an aerobic training protocol that could be replicable, safe and viable for other clinical trials with panic disorder patients. METHODS: A total of 24 exercise sessions (twice/week) of treadmill walking at controlled intensity (75% VO2max) were completed by four panic disorder patients. RESULTS: No major complications were observed. The benefits of the aerobic training intervention were reflected in favorable changes in the Panic and Agoraphobia Scale, the Cardiac Anxiety Questionnaire, the Anxiety Sensitivity Index and in the Body Sensations Questionnaire scores, in spite of no significant differences in physiological variables. CONCLUSIONS: The proposed protocol of aerobic training intervention was shown to be a safe and potentially useful tool as adjunct non-pharmacologic treatment of panic disorder. Further studies are needed in order to determine whether higher intensities and/or longer exercise interventions would induce physiological benefits while still being feasible and safe. RESUMO OBJETIVO: O efeito ansiolítico do exercício aeróbio regular em pacientes com Transtorno de Pânico é bem conhecido. No entanto, permanece a lacuna de um protocolo de intervenção com exercícios aeróbios como um tratamento adjunto não farmacológico para o transtorno de pânico. Nosso objetivo foi apresentar estudo piloto de um protocolo de treinamento aeróbio que pudesse ser replicável, seguro e viável para outras triagens clínicas com pacientes com transtorno de pânico. MÉTODO: Pacientes com transtorno de pânico completaram 24 sessões (duas vezes por semana) de caminhada na esteira, com intensidade controlada (75% VO2max). RESULTADOS: Não foram observadas complicações importantes. Os benefícios da intervenção com treinamento aeróbio refletiram-se favoravelmente através de mudanças nos escores da Escala de Pânico e Agorafobia, do Questionário de Ansiedade Cardíaca, do Índice de Sensibilidade à Ansiedade e do Questionário de Sensações Corporais, apesar de nenhuma diferença ter se mostrado significativa nas variáveis fisiológicas. CONCLUSÕES: O protocolo de treinamento aeróbio proposto mostrou ser seguro e uma ferramenta potencialmente útil para o tratamento adjunto não farmacológico do transtorno de pânico. Mais estudos são necessários a fim de avaliar se intensidades mais elevadas e/ou intervenções mais longas com exercícios poderiam induzir benefícios fisiológicos adicionais, ao mesmo tempo permanecendo viáveis e seguros.

BACKGROUND: Cardiac Anxiety (CA) is the fear of cardiac sensations, characterized by recurrent anxiety symptoms, in patients with or without cardiovascular disease. The Cardiac Anxiety Questionnaire (CAQ) is a tool to assess CA, already adapted but not validated to Portuguese. OBJECTIVE: This paper presents the three phases of the validation studies of the Brazilian CAQ. METHODS: To extract the factor structure and assess the reliability of the CAQ (phase 1), 98 patients with coronary artery disease were recruited. The aim of phase 2 was to explore the convergent and divergent validity. Fifty-six patients completed the CAQ, along with the Body Sensations Questionnaire (BSQ) and the Social Phobia Inventory (SPIN). To determine the discriminative validity (phase 3), we compared the CAQ scores of two subgroups formed with patients from phase 1 (n = 98), according to the diagnoses of panic disorder and agoraphobia, obtained with the MINI - Mini International Neuropsychiatric Interview. RESULTS: A 2-factor solution was the most interpretable (46.4% of the variance). Subscales were named "Fear and Hypervigilance" (n=9; alpha = 0.88), and "Avoidance", (n = 5; alpha = 0.82). ArtinM is a D-mannose-binding lectin extracted from the seeds of Artocarpus heterophyllus that interacts with TLR2 N-glycans and activates antigen-presenting cells (APCs), as manifested by IL-12 production. In vivo ArtinM administration induces Th1 immunity and confers protection against infection with several intracellular pathogens. In the murine model of Candida albicans infection, it was verified that, in addition to Th1, ArtinM induces Th17 immunity manifested by high IL-17 levels in the treated animals. Herein, we investigated the mechanisms accounting for the ArtinM-induced IL-17 production. We found that ArtinM stimulates the IL-17 production by spleen cells in BALB/c or C57BL/6 mice, a response that was significantly reduced in the absence of IL-23, MyD88, or IL-1R. Furthermore, we showed that ArtinM directly induced the IL-23 mRNA expression and the IL-1 production by macrophages. Consistently, in cell suspensions depleted of macrophages, the IL-17 production stimulated by ArtinM was reduced by 53% and the exogenous IL-23 acted synergistically with ArtinM in promoting IL-17 production by spleen cell suspensions. We verified that the absence of IL-23, IL-1R, or MyD88 inhibited, but did not block, the IL-17 production by ArtinM-stimulated spleen cells. Therefore, we investigated whether ArtinM exerts a direct effect on CD4+ T cells in promoting IL-17 production. Indeed, spleen cell suspensions depleted of CD4+ T cells responded to ArtinM with very low levels of IL-17 release. Likewise, isolated CD4+ T cells under ArtinM stimulus augmented the expression of TGF-β mRNA and released high levels of IL-17. Considering the observed synergism between IL-23 and ArtinM, we used cells from IL-23 KO mice to assess the direct effect of lectin on CD4+ T cells. We verified that ArtinM increased the IL-17 production significantly, a response that was inhibited when the CD4+ T cells were pre-incubated with anti-CD3 antibody. In conclusion, ArtinM stimulates the production of IL-17 by CD4+ T cells in two major ways: (I) through the induction of IL-23 and IL-1 by APCs and (II) through the direct interaction with CD3 on the CD4+ T cells. This study contributes to elucidation of mechanisms accounting for the property of ArtinM in inducing Th17 immunity and opens new perspectives in designing strategies for modulating immunity by using carbohydrate recognition agents.

Regular physical exercise has been shown to favorably influence mood and anxiety; however, there are few studies regarding psychiatric aspects of physically active patients with coronary artery disease (CAD). The objective of the present study was to compare the prevalence of psychiatric disorders and cardiac anxiety in sedentary and exercising CAD patients. A total sample of 119 CAD patients (74 men) were enrolled in a case-control study. The subjects were interviewed to identify psychiatric disorders and responded to the Cardiac Anxiety Questionnaire. In the exercise group (N = 60), there was a lower prevalence (45 vs 81%; P < 0.001) of at least one psychiatric diagnosis, as well as multiple comorbidities, when compared to the sedentary group (N = 59). Considering the Cardiac Anxiety Questionnaire, sedentary patients presented higher scores compared to exercisers (mean ± SEM = 55.8 ± 1.9 vs 37.3 ± 1.6; P < 0.001). In a regression model, to be attending a medically supervised exercise program presented a relevant potential for a 35% reduction in cardiac anxiety. CAD patients regularly attending an exercise program presented less current psychiatric diagnoses and multiple mental-related comorbidities and lower scores of cardiac anxiety. These salutary mental effects add to the already known health benefits of exercise for CAD patients.

Toxoplasma gondii é um protozoário coccídio intracelular obrigatório conhecido por sua habilidade em parasitar uma ampla gama de espécies hospedeiras. A região apical do parasito é rica em organelas que, em função dos produtos liberados, estão envolvidas no processo de adesão e invasão da célula hospedeira. As proteínas liberadas por micronemas (MICs), solúveis e transmembrana, possuem domínios adesivos essenciais para a virulência do parasita. Algumas dessas proteínas são encontradas associadas entre si na superfície do taquizoíta, formando complexos como TgMIC1/MIC4/MIC6 e TgMIC3/MIC8. Em estudos anteriores demonstramos a que o subcomplexo TgMIC1/MIC4 (Fração LAC+) liga-se à lactose e estimula macrófagos a secretar IL-12. Verificamos, utilizando células HEK293 transfectadas, que um dos principais mecanismos responsáveis pela produção de IL-12 decorria do reconhecimento de N-glicanos do ectodomínio de TLR2 pelo domínio de reconhecimento de carboidrato de TgMIC1. O objetivo do presente estudo foi o de investigar a capacidade de TgMIC1 e TgMIC4 de ativar macrófagos murinos e qual o papel desempenhado por TLR2 e/ou TLR4 no desencadeamento dessa ativação. Mostramos que macrófagos derivados de medula óssea de camundongos C57Bl/6, estimulados com TgMIC1 e TgMIC4, utilizadas isoladamente ou em combinação, secretam altos níveis de citocinas pró-inflamatórias, como TNF-?, IL-6, IL-12 e IL-1?, produzem altos níveis de óxido nítrico, e têm aumentadas suas capacidades migratória e fagocítica. Os ensaios que utilizaram macrófagos de camundongos C57Bl/6 nocauteados revelaram que a ausência de expressão de TLR2 ou de TLR4 prejudicou os efeitos ativadores exercidos por TgMIC1 e TgMIC4. Macrófagos TLR2-/- tiveram as manifestações de ativação celular significantemente reduzidas em relação aos macrófagos selvagens. Por outro lado, esses efeitos foram mais afetados pela ausência de TLR4, uma vez que as respostas obtidas frente ao estímulo com TgMIC1 ou TgMIC4 eram similares às verificadas em células não estimuladas (controle negativo). Concluímos que TgMIC1 e TgMIC4 interagem com os receptores do tipo toll 2 e 4 expressos por macrófagos, levando à ativação celular manifesta por alta produção de citocinas e outros mediadores inflamatórios, e aumento das capacidades migratória e fagocítica. A interação com TLR4 é preponderante em relação à estabelecida com TLR2 no desencadeamento de ativação celular Toxoplasma gondii is an obligate intracellular coccidian protozoan known for its ability to parasitize a wide range of host species. The parasite\'s apical region is rich in organelles that, because of the products it releases, are involved in the processes of adhesion and invasion of the host cell. The soluble and transmembrane proteins released by the micronemes (MICs), have adhesive domains that are essential for the parasite virulence. Some of these proteins can be found associated with each other on the taquizoite surface, as it is the case of TgMIC1/MIC4/MIC6 or TgMIC3/TgMIC8 complexes. Our group has demonstrated in previous studies that the TgMIC1/TgMIC4 subcomplex (LAC+ fraction) binds to lactose and stimulates macrophages to release IL-12. Using HEK293 transfected cells, we showed that one of the main mechanisms leading to IL-12 release by macrophages, was the recognition of N-glycans of the TLR2 ectodomain by the carbohydrate recognition domain of TgMIC1. Thus, the objective of this study was to address whether TgMIC1 and TgMIC4 activate murine macrophages and what is the role of TLR2 and TLR4 in macrophage activation. Our results show that the stimulation of C57BL/6 mice bone marrow derived macrophages with TgMIC1 and TgMIC4, alone or in combination, induce the release of high levels of proinflammatory cytokines such as TNF-?, IL-6, IL-12 and IL-1?, and also nitric oxide; and increases phagocytic activity and cell migration activity. The assays using knockout C57Bl/6 macrophages showed that the absence of TLR2 or TLR4 expression impaired the activating effects of TgMIC1 e TgMIC4. TLR2-/- macrophages presented significantly reduced cell activation manifestations compared to WT macrophages. On the other hand, these effects were more affected in the absence of TLR4, once the responses obtained with TgMIC1 or TgMIC4 stimulation were similar to those observed in non stimulated cells (negative control). We conclude that TgMIC1 and TgMIC4 interact with the receptors Toll like 2 and 4 expressed in macrophages, leading to cell activation, resulting in high cytokine and inflammatory mediators production, and increased migratory and phagocytic capacity.
