Next generation telescopes, like Euclid, Rubin/LSST, and Roman, will open new windows on the Universe, allowing us to infer physical properties for tens of millions of galaxies. Machine learning methods are increasingly becoming the most efficient tools to handle this enormous amount of data, because they are often faster and more accurate than traditional methods. We investigate how well redshifts, stellar masses, and star-formation rates (SFR) can be measured with deep learning algorithms for observed galaxies within data mimicking the Euclid and Rubin/LSST surveys. We find that Deep Learning Neural Networks and Convolutional Neutral Networks (CNN), which are dependent on the parameter space of the training sample, perform well in measuring the properties of these galaxies and have a better accuracy than methods based on spectral energy distribution fitting. CNNs allow the processing of multi-band magnitudes together with $H_{\scriptscriptstyle\rm E}$-band images. We find that the estimates of stellar masses improve with the use of an image, but those of redshift and SFR do not. Our best results are deriving i) the redshift within a normalised error of less than 0.15 for 99.9$\%$ of the galaxies with S/N>3 in the $H_{\scriptscriptstyle\rm E}$-band; ii) the stellar mass within a factor of two ($\sim0.3 \rm dex$) for 99.5$\%$ of the considered galaxies; iii) the SFR within a factor of two ($\sim0.3 \rm dex$) for $\sim$70$\%$ of the sample. 4. Possible evidence for a large-scale enhancement in the Lyman-α forest power spectrum at redshiftz≥ 4

Inhomogeneous reionization enhances the 1D Ly α forest power spectrum on large scales at redshifts z ≥ 4. This is due to coherent fluctuations in the ionized hydrogen fraction that arise from large-scale variations in the post-reionization gas temperature, which fade as the gas cools. It is therefore possible to use these relic fluctuations to constrain inhomogeneous reionization with the power spectrum at wavenumbers $\log _{10}(k/{\rm km^{-1}\, s})\lesssim -1.5$. We use the Sherwood-Relics suite of hybrid radiation hydrodynamical simulations to perform a first analysis of new Ly α forest power spectrum measurements at 4.0 ≤ z ≤ 4.6. These data extend to wavenumbers $\log _{10}(k/{\rm km^{-1}\, s})\simeq -3$, with a relative uncertainty of 10–20 per cent in each wavenumber bin. Our analysis returns a 2.7σ preference for an enhancement in the Ly α forest power spectrum at large scales, in excess of that expected for a spatially uniform ultraviolet background. This large-scale enhancement could be a signature of inhomogeneous reionization, although the statistical precision of these data is not yet sufficient for obtaining a robust detection of the relic post-reionization fluctuations. We show that future power spectrum measurements with relative uncertainties of ≲ 2.5 per cent should provide unambiguous evidence for an enhancement in the power spectrum on large scales.

Despite their unprecedented success in relapsed/refractory (R/R) large B-cell lymphoma (LBCL), anti-CD19 CAR T cells are associated with significant toxicity, and more than half of patients relapse. As monocytes emerged as key players in CAR therapy, we sought to evaluate the evolution of HLA-DR expression on monocytes (mHLA-DR) before and after commercial anti-CD19 CAR T-cell infusion in a large cohort (n = 103) of patients with R/R LBCL and its association with adverse events and treatment response. Cy-Flu-based lymphodepletion (LD) upregulated mHLA-DR in 79% of the cases, whereas in 2l% of cases (15 patients), the mHLA-DR level decreased after LD, and this decrease was associated with poorer outcome. The Aria cryogenic distillation plant, located in Sardinia, Italy, is a key component of the DarkSide-20k experimental program for WIMP dark matter searches at the INFN Laboratori Nazionali del Gran Sasso, Italy. Aria is designed to purify the argon, extracted from underground wells in Colorado, USA, and used as the DarkSide-20k target material, to detector-grade quality. In this paper, we report the first measurement of argon isotopic separation by distillation with the 26 m tall Aria prototype. We discuss the measurement of the operating parameters of the column and the observation of the simultaneous separation of the three stable argon isotopes: $${}^{36}\hbox {Ar}$$ 36 Ar , $${}^{38}\textrm{Ar}$$ 38 Ar , and $${}^{40}\textrm{Ar}$$ 40 Ar . We also provide a detailed comparison of the experimental results with commercial process simulation software. This measurement of isotopic separation of argon is a significant achievement for the project, building on the success of the initial demonstration of isotopic separation of nitrogen using the same equipment in 2019.

The Cosmology and Astrophysics with Machine Learning Simulations (CAMELS) project was developed to combine cosmology with astrophysics through thousands of cosmological hydrodynamic simulations and machine learning. CAMELS contains 4233 cosmological simulations, 2049 N-body simulations, and 2184 state-of-the-art hydrodynamic simulations that sample a vast volume in parameter space. In this paper, we present the CAMELS public data release, describing the characteristics of the CAMELS simulations and a variety of data products generated from them, including halo, subhalo, galaxy, and void catalogs, power spectra, bispectra, Lyα spectra, probability distribution functions, halo radial profiles, and X-rays photon lists. We also release over 1000 catalogs that contain billions of galaxies from CAMELS-SAM: a large collection of N-body simulations that have been combined with the Santa Cruz semianalytic model. We release all the data, comprising more than 350 terabytes and containing 143,922 snapshots, millions of halos, galaxies, and summary statistics. We present the Sherwood-Relics simulations, a new suite of large cosmological hydrodynamical simulations aimed at modelling the intergalactic medium (IGM) during and after the cosmic reionization of hydrogen. The suite consists of over 200 simulations that cover a wide range of astrophysical and cosmological parameters. It also includes simulations that use a new lightweight hybrid scheme for treating radiative transfer effects. This scheme follows the spatial variations in the ionizing radiation field, as well as the associated fluctuations in IGM temperature and pressure smoothing. It is computationally much cheaper than full radiation hydrodynamics simulations and circumvents the difficult task of calibrating a galaxy formation model to observational constraints on cosmic reionization. Using this hybrid technique, we study the spatial fluctuations in IGM properties that are seeded by patchy cosmic reionization. We investigate the relevant physical processes and assess their impact on the z > 4 Lyman-alpha forest. Our main findings are: (i) Consistent with previous studies patchy reionization causes large scale temperature fluctuations that persist well after the end of reionization, (ii) these increase the Lyman-alpha forest flux power spectrum on large scales, and (iii) result in a spatially varying pressure smoothing that correlates well with the local reionization redshift. The valorization of poultry byproducts, like feathers (processed to feather meal), in animal feed could contribute to the presence of veterinary drugs, including antibiotics. An animal study was carried out to study the fate of sulfadiazine, trimethoprim, and oxytetracycline in feathers, plasma, and droppings of broiler chickens. Cage and floor housing, different from current farm practices, were studied. Samples were analyzed by liquid chromatography-tandem mass spectrometry (LC-MS/MS). A longer presence of antibiotics was observed in feathers compared to plasma, with sulfadiazine being present the most. Skipping of BRCA2 exon 3 (∆E3) is a naturally occurring splice event, complicating clinical classification of variants that may alter ∆E3 expression. This study used multiple evidence types to assess pathogenicity of 85 variants in/near BRCA2 exon 3. Bioinformatically predicted spliceogenic variants underwent mRNA splicing analysis using minigenes and/or patient samples. ∆E3 was measured using quantitative analysis. A mouse embryonic stem cell (mESC) based assay was used to determine the impact of 18 variants on mRNA splicing and protein function. For each variant, population frequency, bioinformatic predictions, clinical data and existing mRNA splicing and functional results were collated. Variant class was assigned using a gene-specific adaptation of ACMG/AMP guidelines, following a recently proposed points-based system. mRNA and mESC analysis combined identified six variants with transcript and/or functional profiles interpreted as loss of function. Cryptic splice site use for acceptor site variants generated a transcript encoding a shorter protein that retains activity. Overall, 69/85 (81%) variants were classified using the point-based approach. Our analysis shows the value of applying gene-specific ACMG/AMP guidelines using a points-based approach and highlights the consideration of cryptic splice site usage to appropriately assign PVS1 code strength. Indigenous microorganisms are important components of the complex ecosystem of many dairy foods including cheeses, and they are potential contributors to the development of a specific cheese's sensory properties. Among these indigenous microorganisms are the yeasts Cyberlindnera jadinii, Pichia kudriavzevii, and Kazachstania servazzii, which were previously detected using traditional microbiological methods in both raw milk and some artisanal specialty cheeses produced in the province of Québec, Canada. However, their levels across different cheese varieties are unknown. A highly specific and sensitive real-time quantitative PCR assay was developed to quantitate these yeast species in a variety of specialty cheeses (bloomy-rind, washed-rind, and natural-rind cheeses from raw, thermized, and pasteurized milks). The specificity of the quantitative PCR assay was validated, and it showed no cross-amplification with 11 other fungal microorganisms usually found in bloomy-rind and washed-rind cheeses. The Complete Calibration of the Color-Redshift Relation survey (C3R2) is a spectroscopic programme designed to empirically calibrate the galaxy color-redshift relation to the Euclid depth (I_E=24.5), a key ingredient for the success of Stage IV dark energy projects based on weak lensing cosmology. A spectroscopic calibration sample as representative as possible of the galaxies in the Euclid weak lensing sample is being collected, selecting galaxies from a self-organizing map (SOM) representation of the galaxy color space. Here, we present the results of a near-infrared H- and K-bands spectroscopic campaign carried out using the LUCI instruments at the LBT. Euclid's photometric galaxy cluster survey has the potential to be a very competitive cosmological probe. The main cosmological probe with observations of clusters is their number count, within which the halo mass function (HMF) is a key theoretical quantity. We present a new calibration of the analytic HMF, at the level of accuracy and precision required for the uncertainty in this quantity to be subdominant with respect to other sources of uncertainty in recovering cosmological parameters from Euclid cluster counts. Our model is calibrated against a suite of N-body simulations using a Bayesian approach taking into account systematic errors arising from numerical effects in the simulation. First, we test the convergence of HMF predictions from different N-body codes, by using initial conditions generated with different orders of Lagrangian Perturbation theory, and adopting different simulation box sizes and mass resolution. Then, we quantify the effect of using different halo-finder algorithms, and how the resulting differences propagate to the cosmological constraints. In order to trace the violation of universality in the HMF, we also analyse simulations based on initial conditions characterised by scale-free power spectra with different spectral indexes, assuming both Einstein--de Sitter and standard $Λ$CDM expansion histories. Based on these results, we construct a fitting function for the HMF that we demonstrate to be sub-percent accurate in reproducing results from 9 different variants of the $Λ$CDM model including massive neutrinos cosmologies. The calibration systematic uncertainty is largely sub-dominant with respect to the expected precision of future mass-observation relations; with the only notable exception of the effect due to the halo finder, that could lead to biased cosmological inference., 25 pages, 21 figures, 5 tables, 3 appendixes; v2 matches published version

Los objetivos de este trabajo son: (I) establecer valores de retrodispersión para los blancos agua superficial libre de hielo y recubierta por hielo en lagos –con el soporte de datos de campo– utilizando las imágenes TerraSar-X (TSX) y Sentinel 1A - EW (S-1), teniendo como estudio de caso la península Fildes, Antárctica Marítima; (II) evaluar la relación entre los datos de retrodispersión y los datos meteorológicos; (III) evaluar las potencialidades de TSX y de S-1para el mapeo de la fenología de los lagos. El procesamiento de las imágenes SAR fue realizado en el SNAP y los datos meteorológicos fueron obtenidos en la Base Eduardo Frei Montalva. Se obtuvo el umbral de retrodispersión de -32 dB a -21 dB para el agua libre de hielo en los lagos estudiados en la imagen TSX, y para la Sentinel-1 hubo una variación de -18 dB a -15 dB para el agua libre de hielo de los lagos. La imagen TSX es la más adecuada para el monitoreo de la fenología de los lagos por su resolución espacial, mientras la S-1 se limita a los lagos con más de 10.000 m2. Es necesario tener en cuenta las condiciones meteorológicas, especialmente el viento, las precipitaciones de nieve y los cambios de temperatura, que representan alteraciones significativas en los valores de retrodispersión. Se puede utilizar las recomendaciones metodológicas referentes al uso de las imágenes TSX y S-1 para el mapeo de la cobertura de hielo en los lagos y el monitoreo de los eventos de quiebra de hielo y de derretimiento de la superficie de lagos en ambientes glaciales y periglaciales.

19. Forensic applications of micro-computed tomography: a systematic review

Purpose The aim of this systematic review was to provide a comprehensive overview of micro-CT current applications in forensic pathology, anthropology, odontology, and neonatology. Methods A bibliographic research on the electronic databases Pubmed and Scopus was conducted in the time frame 01/01/2001–31/12/2021 without any language restrictions and applying the following free-text search strategy: “(micro-computed tomography OR micro-CT) AND (forensic OR legal)”. The following inclusion criteria were used: (A) English language; (B) Application of micro-CT to biological and/or non-biological materials to address at least one forensic issue (e.g., age estimation, identification of post-mortem interval). The papers selected by three independent investigators have been then classified according to the investigated materials. Results The bibliographic search provided 651 records, duplicates excluded. After screening for title and/or abstracts, according to criteria A and B, 157 full-text papers were evaluated for eligibility. Ninety-three papers, mostly (64) published between 2017 and 2021, were included; considering that two papers investigated several materials, an overall amount of 99 classifiable items was counted when referring to the materials investigated. It emerged that bones and cartilages (54.55%), followed by teeth (13.13%), were the most frequently analyzed materials. Moreover, micro-CT allowed the collection of structural, qualitative and/or quantitative information also for soft tissues, fetuses, insects, and foreign materials. Conclusion Forensic applications of micro-CT progressively increased in the last 5 years with very promising results. According to this evidence, we might expect in the near future a shift of its use from research purposes to clinical forensic cases.

The communication services of future trains require hyperconnectivity between trains and track. Future Railway Mobile Communication System (FRMCS) reserved bands will not be able to cover all digital train requirements including passengers’ services. The 5GMED project adopts the idea where Infrastructure Managers (IMs) will collaborate with Railway Undertaking (RU) to facilitate the connectivity to Mobile Network Operator (MNO) communication services and vice versa (complemented with the use of satellite networks), with an architecture open to build new business models and relationships between stakeholders. This approach encompasses an adaptative Gigabit train-to-track connectivity solution able to fit Digital Train requirements. The 5GMED project will demonstrate these capabilities through four representative services deployed on a TGV of SNCF (French Train Operator) in a challenging scenario: a cross-border section of the Mediterranean corridor between Figueres (Spain) and Perpignan (France).

International audience; Mucosal healing has emerged as a therapeutic goal to achieve lasting clinical remission in ulcerative colitis. Intestinal repair in response to inflammation presumably requires higher energy supplies for the restoration of intestinal barrier and physiological functions. However, epithelial energy metabolism during intestinal mucosal healing has been little studied, whereas inflammation-induced alterations have been reported in the main energy production site, the mitochondria. The aim of the present work was to assess the involvement of mitochondrial activity and the events influencing their function during spontaneous epithelial repair after colitis induction in mouse colonic crypts. The results obtained show adaptations of colonocyte metabolism during colitis to ensure maximal ATP production for supporting energetic demand by both oxidative phosphorylation and glycolysis in a context of decreased mitochondrial biogenesis and through mitochondrial function restoration during colon epithelial repair. In parallel, colitis-induced mitochondrial ROS production in colonic epithelial cells was rapidly associated with transient expression of GSH-related enzymes. Mitochondrial respiration in colonic crypts was markedly increased during both inflammatory and recovery phases despite decreased expression of several mitochondrial respiratory chain complex subunits after colitis induction. Rapid induction of mitochondrial fusion was associated with mitochondrial function restoration. Finally, in contrast with the kinetics expression of genes involved in mitochondrial oxidative metabolism and in glycolysis, the expression of glutaminase was markedly reduced in the colonic crypts both during colitis and repair phases. Overall, our data suggest that the epithelial repair after colitis induction is characterized by a rapid and transient increased capacity for mitochondrial ATP production in a context of apparent restoration of mitochondrial biogenesis and metabolic reorientation of energy production. The potential implication of energy production adaptations within colonic crypts to sustain mucosal healing in a context of altered fuel supply is discussed.

International audience; Background: Gain of function (GOF) variants of JAK1 drive a rare immune dysregulation syndrome associated with atopic dermatitis, allergy and eosinophilia.Objectives: To describe the clinical and immunological characteristics associated with a new GOF variant of JAK1 and report the therapeutic efficacy of JAK inhibition.Methods: We identified a family affected by JAK1 associated autoinflammatory disease and performed clinical assessment and immunological monitoring on 9 patients. JAK1 signalling was studied by flow and mass cytometry in patients' cells at basal state, or after immune stimulation. A molecular disease signature in the blood was studied at the transcriptomic level. Patients were treated with one of two JAK inhibitors; either baricitinib or upadacitinib. Clinical, cellular, and molecular response were evaluated over a 2-year period.Results: Affected individuals displayed a syndromic disease with prominent allergy including atopic dermatitis, ichthyosis, arthralgia, chronic diarrhoea, disseminated calcifying fibrous tumours and elevated whole blood histamine levels. A variant of JAK1 localized in the pseudokinase domain was identified in all 9 affected tested patients. Hyper-phosphorylation of STAT3 was found in 5 out of 6 patients tested. Treatment of patients' cells with baricitinib controlled most of the atypical hyper-phosphorylation of STAT3. Administration of baricitinib to patients led to rapid improvement of the disease in all adults and was associated with reduction of systemic inflammation.Conclusions: Patients with this new JAK1 GOF pathogenic variant displayed very high levels of blood histamine and showed a variable combination of atopy with articular and gastrointestinal manifestations as well as calcifying fibrous tumours. The disease, that appears to be linked to STAT3 hyper-activation, was well controlled under treatment by JAK inhibitors in adult patients.

Résumé Le site de Sion-Avenue de Tourbillon a été découvert en bordure sud du cône alluvial de la Sionne, à la faveur de la construction d’un immeuble d’habitations et de commerces. Les vestiges mis au jour montrent que les lieux ont été fréquentés à plusieurs reprises entre le Néolithique et la période antique. Les restes les plus significatifs sont représentés par cinq sépultures en ciste, que les datations radiocarbone situent dans le dernier tiers du 5ième millénaire avant notre ère. Quatre coffres étaient groupés dans la partie sud-est de l’emprise du chantier. Le cinquième était isolé, à quelques dizaines de mètres à l’ouest du groupe. Cet ensemble présente les caractères spécifiques des sépultures de type Chamblandes et vient s’ajouter à la dizaine d’ensembles funéraires connus à Sion et dans les alentours. Par leurs caractéristiques et leur excellente conservation, les sépultures de Sion-Avenue de Tourbillon contribuent de manière significative à une meilleure compréhension du phénomène Chamblandes en Valais., Summary in German, French, Italian, English

Childhood is composed of biological and psychosocial transitions, which enable the acquisition of motor, cognitive, and affective-social domains of development. With the onset of the covid-19 pandemic in 2020 the WHO recommended that preventive measures be taken, such as closing schools as well as public places, a fact that culminated in distancing from friends and family. This has led to a greater proximity of children to technology, thus increasing their exposure to screens. According to the Brazilian Society of Pediatrics, the use of television, tablets, and computers should be equivalent to the child's age, monitored by those responsible in order to avoid excessive use. In this context, the objective of this project was to analyze the technological addictions of 4th and 5th grade students from Elementary School I in the city of Votuporanga-SP, developed before and during the Pandemic by Covid-19, and its impacts on the children's mental health. The data were collected through a semi-structured questionnaire designed to evaluate the behavior of the children regarding the use of technology and lifestyle habits, in a qualitative and quantitative way. Our results showed that the pandemic period favored a greater access to digital equipment by children, due to their use for school activities. The increased use of cell phones created the possibility of distraction with games, videos, and participation in social networks. However, they emphasized that they prefer games involving physical activity to simply using screens. We also observed that it is of fundamental importance that parents/guardians accompany the children in stimulating family interaction.

International audience; BACKGROUND AND AIMS: The diagnosis of alcoholic steatohepatitis (ASH) is based on liver biopsy, which is costly and invasive with non-negligible morbidity. The aim of this study was to evaluate the accuracy of circulating cytokeratin 18 M65 fragment (K18-M65) alone or in association with other markers for the non-invasive diagnosis of ASH in patients ongoing alcohol withdrawal. METHODS: This study examined the serum level of K18-M65 in a test cohort of 196 patients. All patients underwent liver biopsy, transient elastography (TE) and serum collection. The diagnostic accuracy of K18-M65 alone or combined with clinico-biological data was assessed and the best defined cut-offs were validated in an independent validation cohort of 58 patients. RESULTS: K18-M65 had an area under the curve (AUC) of 0.82 (test cohort) and 0.90 (validation cohort). Using two cut-off decision points, K18-M65 was able to classify 46.9% (test cohort) and 34.5% (validation cohort) of patients with 95% sensitivity or specificity. Combining K18-M65, alpha-2-macroglobulin, TE, body mass index, and age, we created a score allowing accurate diagnosis of ASH with an AUC of 0.93 (test cohort) and 0.94 (validation cohort). This new score was able to rule out or rule in the diagnosis of steatohepatitis for probability ≤0.135 or ≥0.667 respectively in more than two-thirds of patients. CONCLUSIONS: We propose a new validated non-invasive score for the diagnosis of ASH in patients ongoing alcohol withdrawal. This score can help to identify patients that may benefit from potential therapeutics or motivate them to reduce alcohol consumption.

International audience; BACKGROUND AIMS: Hepatocellular carcinoma (HCC), the third cause of cancer-related death, arises in the context of liver fibrosis (LF). Although HCC is generally poorly fibrogenic, some tumors harbor focal intratumor extracellular matrix (ECM) deposits, called fibrous nests. To date, the molecular composition and the clinical relevance of these ECM deposits has not been fully defined. APPROACH RESULTS: We performed quantitative matrisome analysis by tandem mass tags mass spectrometry (TMT-MS) in 20 human HCCs, with high- or low-grade intratumor fibrosis, and matched non-tumor (NT) tissues, as well as in 12 livers from mice treated with vehicle, CCl4 or diethylnitrosamine (DEN). We found 94 ECM proteins differentially abundant between high- and low-grade fibrous nests, including interstitial and basement membrane components, such as several collagens, glycoproteins, proteoglycans, enzymes involved in ECM stabilization and degradation, and growth factors. Pathway analysis revealed a metabolic switch in high-grade fibrosis, with enhanced glycolysis and decreased oxidative phosphorylation. Integrating our quantitative proteomics data with the transcriptomes from HCCs and NT livers (n = 2,285 samples), we identified a subgroup of fibrous nest HCCs, characterized by cancer-specific ECM remodeling, expression of the WNT/TGFB (S1) subclass signature, and poor patient outcome. Fibrous nest HCCs, abundantly expressed an 11 fibrous nest proteins’ signature, associated with poor patient outcome, by multivariate Cox analysis, and validated by multiplex immunohistochemistry. CONCLUSION: Matrisome analysis highlighted cancer-specific ECM deposits, typical of the WNT/TGFB HCC subclass, associated with poor patient outcome. Hence, histological reporting of intratumor fibrosis in HCC is of clinical relevance.

The contribution of germline copy number variants (CNVs) to risk of developing cancer in individuals with pathogenic BRCA1 or BRCA2 variants remains relatively unknown. We conducted the largest genome-wide analysis of CNVs in 15,342 BRCA1 and 10,740 BRCA2 pathogenic variant carriers. We used these results to prioritise a candidate breast cancer risk-modifier gene for laboratory analysis and biological validation. Notably, the HR for deletions in BRCA1 suggested an elevated breast cancer risk estimate (hazard ratio (HR) = 1.21), 95% confidence interval (95% CI = 1.09-1.35) compared with non-CNV pathogenic variants. In contrast, deletions overlapping SULT1A1 suggested a decreased breast cancer risk (HR = 0.73, 95% CI 0.59-0.91) in BRCA1 pathogenic variant carriers. Functional analyses of SULT1A1 showed that reduced mRNA expression in pathogenic BRCA1 variant cells was associated with reduced cellular proliferation and reduced DNA damage after treatment with DNA damaging agents. Therapies for metastatic SDHB-dependent pheochromocytoma and paraganglioma (PPGL) are limited and poorly efficient. New targeted therapies and identification of early non-invasive biomarkers of response are thus urgently needed for these patients. We characterized an in vivo allograft model of spontaneously immortalized murine chromaffin cells (imCC) with inactivation of the Sdhb gene by dynamic contrast-enhanced MRI (DCE-MRI) and 18FDG-PET. We evaluated the response to several therapies: IACS-010759 (mitochondrial respiratory chain complex I inhibitor), sunitinib (tyrosine kinase inhibitor with anti-angiogenic activity), talazoparib (poly ADP ribose polymerase (PARP) inhibitor) combined or not to temozolomide (alkylating agent), pharmacological inhibitors of HIF2a (PT2385 and PT2977 (belzutifan)) and molecular inactivation of HIF2a (imCC Sdhb−/− shHIF2a). Multimodal imaging was performed, including magnetic resonance spectroscopy (1H-MRS) to monitor the level of succinate in vivo. The allografted model of Sdhb−/− imCC reflected SDHB-deficient tumors, with increased angiogenesis and a particular avidity for 18FDG. After 14 days of treatment, IACS-010759, sunitinib and talazoparib at high doses allowed a significant reduction of the tumor volumes. In contrast to the tumor growth inhibition observed in Sdhb−/− shHIF2a imCC tumors, pharmacological inhibitors of HIF2a (PT2385 and belzutifan) showed no antitumor action in this model, alone or in combination with sunitinib. 1H-MRS, but not DCE-MRI, enabled the monitoring response to sunitinib, which was the best treatment in this study, promoting a decrease in succinate levels detected in vivo. This study paves the way for new therapeutic options and reveals a potential new early biomarker of response to treatment in SDHB-dependent PPGL.

Infections complicate 5%-10% of cerebrospinal fluid (CSF) shunts. We aimed to describe the characteristics and contemporary pathogens of shunt infections in children in Canada and the United States.Descriptive case series at tertiary care hospitals in Canada (N = 8) and the United States (N = 3) of children up to 18 years of age with CSF shunt infections from July 1, 2013, through June 30, 2019.There were 154 children (43% female, median age 2.7 years, 50% premature) with ≥1 CSF shunt infections. Median time between shunt placement and infection was 54 days (interquartile range, 24 days-2.3 years). Common pathogens were coagulase-negative staphylococci (N = 42; 28%), methicillin-susceptible Staphylococcus aureus (N = 24; 16%), methicillin-resistant S. aureus (N = 9; 5.9%), Pseudomonas aeruginosa (N = 9; 5.9%) and other Gram-negative bacilli (N = 14; 9.0%). Significant differences between pathogens were observed, including timing of infection (P = 0.023) and CSF leukocyte count (P = 0.0019); however, differences were not sufficient to reliably predict the causative organism based on the timing of infection or discriminate P. aeruginosa from other pathogens based on clinical features. Empiric antibiotic regimens, which included vancomycin (71%), cefotaxime or ceftriaxone (29%) and antipseudomonal beta-lactams (33%), were discordant with the pathogen isolated in five cases. There was variability between sites in the distribution of pathogens and choice of empiric antibiotics. Nine children died; 4 (44%) deaths were attributed to shunt infection.Staphylococci remain the most common cause of CSF shunt infections, although antibiotic resistant Gram-negative bacilli occur and cannot be reliably predicted based on clinical characteristics.

The class II lanthipeptide mersacidin, a ribosomally synthesized and post-translationally modified peptide (RiPP), displays unique intramolecular structures, including a very small lanthionine ring. When applied in the growing field of RiPP engineering, these can add unique features to new-to-nature compounds with novel properties. Recently, a heterologous expression system for mersacidin in Escherichia coli was developed to add its modification enzymes to the RiPP engineering toolbox and further explore mersacidin biosynthesis and leader-processing. The dedicated mersacidin transporter and leader protease MrsT was shown to cleave the leader peptide only partially upon export, transporting GDMEAA-mersacidin out of the cell. The extracellular Bacillus amyloliquefaciens protease AprE was shown to release active mersacidin in a second leader-processing step after transport. The conserved LanT cleavage site in the mersacidin leader is present in many other class II lanthipeptides. In contrast to mersacidin, the leader of these peptides is fully processed in one step. This difference with mersacidin leader-processing raises fundamentally interesting questions about the specifics of mersacidin modification and processing, which is also crucial for its application in RiPP engineering. Here, mutational studies of the mersacidin leader-core interface were performed to answer these questions. Results showed the GDMEAA sequence is crucial for both mersacidin modification and leader processing, revealing a unique leader layout in which a LanM recognition site is positioned downstream of the conserved leader-protease LanT cleavage site. Moreover, by identifying residues and regions that are crucial for mersacidin-type modifications, the wider application of mersacidin modifications in RiPP engineering has been enabled.

En este artículo presentamos los resultados de la fase inicial de una investigación desarrollada por un grupo de estudio e investigación en el ámbito de una Maestría Profesional (MP), basada y enmarcada en la metodología de la Investigación Basada en el Diseño (IBD). Específicamente, en este trabajo, nos enfocamos en los primeros cuatro encuentros dedicados a discusiones teóricas sobre el contenido matemático de la potenciación, con el objetivo de analizar el Conocimiento Especializado del Contenido (SCK) movilizado/refinado por los docentes mientras trabajan colaborativamente en la etapa de preparación de un IBD y, luego, elaborar una conjetura sobre la dimensión del contenido de potenciación para el 6° año de primaria. A partir de los datos producidos en las grabaciones de las reuniones del grupo de estudio y investigación, concluimos que la experiencia brindada por el IBD favoreció la producción de conocimiento para los docentes, ya que lograron movilizar/refinar su SCK respecto a la potenciación cuando se permitieron entender la potenciación como una representación y cuando pudieron ampliar su perspectiva sobre el tema matemático, en la dirección de establecer relaciones entre la aritmética (trabajada en los primeros años) y el álgebra, a través del pensamiento algebraico, proporcionando un entorno favorable para nuevas reflexiones y aprendiendo sobre pensamiento algebraico y la potenciación.

Artículo escrito por un elevado núnmero de autores, sólo se referencian el qque aparece en primer lugar, los autores pertenecientes a la UAM y el nombre del grupo de colaboración, si lo hubiere, We present a new infrared survey covering the three Euclid deep fields and four other Euclid calibration fields using Spitzer Space Telescope's Infrared Array Camera (IRAC). We combined these new observations with all relevant IRAC archival data of these fields in order to produce the deepest possible mosaics of these regions. In total, these observations represent nearly 11 % of the total Spitzer Space Telescope mission time. The resulting mosaics cover a total of approximately 71.5 deg2 in the 3.6 and 4.5 μm bands, and approximately 21.8 deg2 in the 5.8 and 8 μm bands. They reach at least 24 AB magnitude (measured to 5σ, in a 2″.5 aperture) in the 3.6 μm band and up to ∼5 mag deeper in the deepest regions. The astrometry is tied to the Gaia astrometric reference system, and the typical astrometric uncertainty for sources with 16 "< "[3.6]< 19 is ≲ 0″.15. The photometric calibration is in excellent agreement with previous WISE measurements. We extracted source number counts from the 3.6 μm band mosaics, and they are in excellent agreement with previous measurements. Given that the Spitzer Space Telescope has now been decommissioned, these mosaics are likely to be the definitive reduction of these IRAC data. This survey therefore represents an essential first step in assembling multi-wavelength data on the Euclid deep fields, which are set to become some of the premier fields for extragalactic astronomy in the 2020s

The combination and cross-correlation of the upcoming $Euclid$ data with cosmic microwave background (CMB) measurements is a source of great expectation since it will provide the largest lever arm of epochs, ranging from recombination to structure formation across the entire past light cone. In this work, we present forecasts for the joint analysis of $Euclid$ and CMB data on the cosmological parameters of the standard cosmological model and some of its extensions. This work expands and complements the recently published forecasts based on $Euclid$-specific probes, namely galaxy clustering, weak lensing, and their cross-correlation. With some assumptions on the specifications of current and future CMB experiments, the predicted constraints are obtained from both a standard Fisher formalism and a posterior-fitting approach based on actual CMB data. Compared to a $Euclid$-only analysis, the addition of CMB data leads to a substantial impact on constraints for all cosmological parameters of the standard $\Lambda$-cold-dark-matter model, with improvements reaching up to a factor of ten. For the parameters of extended models, which include a redshift-dependent dark energy equation of state, non-zero curvature, and a phenomenological modification of gravity, improvements can be of the order of two to three, reaching higher than ten in some cases. While Euclid is an ESA mission specifically designed to investigate the nature of dark energy and dark matter, the planned unprecedented combination of survey area (∼15 000 deg2), spatial resolution, low sky-background, and depth also make Euclid an excellent space observatory for the study of the low surface brightness Universe. Scientific exploitation of the extended low surface brightness structures requires dedicated calibration procedures that are yet to be tested. Aims. We investigate the capabilities of Euclid to detect extended low surface brightness structure by identifying and quantifying sky-background sources and stray-light contamination. We test the feasibility of generating sky flat-fields to reduce large-scale residual gradients in order to reveal the extended emission of galaxies observed in the Euclid survey. Methods. We simulated a realistic set of Euclid/VIS observations, taking into account both instrumental and astronomical sources of contamination, including cosmic rays, stray-light, zodiacal light, interstellar medium, and the cosmic infrared background, while simulating the effects of background sources in the field of view. Results. We demonstrate that a combination of calibration lamps, sky flats, and self-calibration would enable recovery of emission at a limiting surface brightness magnitude of μlim = 29.5−0.27+0.08 mag arcsec−2 (3σ, 10 × 10 arcsec2) in the Wide Survey, and it would reach regions deeper by 2 mag in the Deep Surveys. Conclusions.Euclid/VIS has the potential to be an excellent low surface brightness observatory. Covering the gap between pixel-to-pixel calibration lamp flats and self-calibration observations for large scales, the application of sky flat-fielding will enhance the sensitivity of the VIS detector at scales larger than 1″, up to the size of the field of view, enabling Euclid to detect extended surface brightness structures below μlim = 31 mag arcsec−2 and beyond. © ESO 2022., A. Dark matter lighter than 10 GeV/c2 encompasses a promising range of candidates. A conceptual design for a new detector, DarkSide-LowMass, is presented, based on the DarkSide-50 detector and progress toward DarkSide-20k, optimized for a low-threshold electron-counting measurement. Sensitivity to light dark matter is explored for various potential energy thresholds and background rates. These studies show that DarkSide-LowMass can achieve sensitivity to light dark matter down to the solar neutrino fog for GeV-scale masses and significant sensitivity down to 10 MeV/c2 considering the Migdal effect or interactions with electrons. Requirements for optimizing the detector’s sensitivity are explored, as are potential sensitivity gains from modeling and mitigating spurious electron backgrounds that may dominate the signal at the lowest energies. We examine the column density distribution function (CDDF) and Doppler parameter distribution from hydrodynamical simulations and Cosmic Origins Spectrograph (COS) observations of the Lyman-alpha forest at redshift $0\leq z\leq 0.2$. Allowing for a factor of two uncertainty in the metagalactic HI photoionisation rate, our hydrodynamical simulations are in good agreement ($1$-$1.5\sigma$) with the shape and amplitude of the observed CDDF at HI column densities $10^{13.3}\rm\,cm^{-2}\leq N_{\rm HI}\leq 10^{14.5}\rm\,cm^{-2}$. However, the Doppler widths of the simulated lines remain too narrow with respect to the COS data. We argue that invoking AGN feedback does not resolve this discrepancy. We also disfavour enhanced photoheating rates as a potential solution, as this requires an unphysically hard UV background spectrum. If instead appealing to a non-canonical source of heating, an additional specific heat injection of $u \lesssim 6.9\rm\,eV\,m_{\rm p}^{-1}$ is required at $z\lesssim 2.5$ for gas that has $N_{\rm HI}\simeq 10^{13.5}\rm\,cm^{-2}$ by $z=0.1$. Alternatively, there may be an unresolved line of sight turbulent velocity component of $v_{\rm turb}\lesssim 8.5\rm\,km\,s^{-1}(N_{\rm HI}/10^{13.5}\rm\,cm^{-2})^{0.21}$ for the coldest gas in the diffuse IGM., Comment: 21 pages, 13 figures, MNRAS accepted following revision. 41. Entrapment within an ottoman storage bed: an unusual accidental asphyxial death

Herein, we present an uncommon forensic case of death by asphyxia. The victim was a woman whose body at death scene investigation (DSI) was discovered beside an ottoman storage bed. According to the rescue team, who had moved the body before our arrival, the body was originally found in the prone position and stuck with the neck, thorax and arms within the bed. Examination of the body showed hypostasis that was mainly distributed to the face and the lower chest while sparing the neck and the upper chest. The face was markedly swollen, and the eyes were congested with blood. Dissection and histology revealed pulmonary oedema and emphysema of both lungs. Integrating circumstantial, radiology and autopsy data, it was established that the victim, while trapped between the mattress and the edge of the ottoman storage bed, died by mechanical asphyxia due to cervical-thoracic compression and postural asphyxia acting simultaneously.

Background Physical activity may increase the risk of cardiotoxicity (myocardial ischemia, major arrhythmias) of 5-Fluorouracil, but this risk has never been investigated for its prodrug capecitabine. Patients and Methods One hundred and ninety-two consecutive patients undergoing capecitabine chemotherapy from December 1, 2010 through July 31, 2016 were prospectively evaluated. The baseline evaluation included electrocardiography (ECG) and echocardiography (2DE); a follow-up evaluation, including ECG and exercise stress testing (2DE in case of ECG abnormalities), was done after ≥10 days of treatment. Cardiotoxicity was suspected from ischemic ECG changes, new kinetic abnormalities at 2DE, Lown classification ≥2 ventricular arrhythmia, symptomatic arrhythmias, or positive stress test, and confirmed by a negative stress test after capecitabine washout. Results Cardiotoxicity was diagnosed in 32 patients (16.7%): six at rest and 26 during exercise. All 32 patients had ECG abnormalities: ST-segment changes (24 patients), negative T-waves (2) and/or arrhythmias: ventricular arrhythmias (14 cases), supraventricular tachycardia (2), complete heart block (1). Eight patients had typical symptoms, 6 had atypical symptoms, 1 had syncope, 17 (53%) were asymptomatic. Cardiotoxicity was more common in patients with atypical symptoms during daily life (OR = 15.7) and in those on a therapeutic schedule of 5 days/week (OR = 9.44). Conclusion Capecitabine cardiotoxicity is frequent, and often elicited by physical effort. Oncologists, cardiologists, and general practitioners should be aware of this risk. Active cardiotoxicity surveillance with ECG (and echocardiogram and/or stress testing in suspected cases) during therapy is recommended. Clinical Trials registration number CRO-2010-17.

OBJETIVO: Demonstrar a mudança no estilo de vida dos brasileiros frente à pandemia da COVID-19, com a finalidade de compreender seus principais impactos na saúde da população. MÉTODOS: Foi realizada uma revisão da literatura com busca no PubMed, Medline e SciELO. Utilizaram-se os descritores “Life Style AND COVID-19”, pesquisados no MeSH. Foram pesquisados artigos que configuraram estudos transversais descritivos que estavam no idioma português e que foram publicados entre 2020 e 2021. RESULTADOS: Saúde mental: Aumento dos sintomas psicopatológicos durante a pandemia, potencializados por sentimentos de ansiedade ocasionados pela mudança na rotina devido às medidas de distanciamento social. Atividade física: Diminuição da prática de atividades físicas e crescente de hábitos sedentários, como tempo em frente a televisores, celulares e monitores, tendo diversos efeitos negativos à saúde. Alimentação: Grande aumento do volume de compras em supermercados de alimentos ultraprocessados e com alta taxa energética, como pipoca, chocolate, congelados, doces, batatas fritas e sorvetes. Consumo de bebidas alcoólicas e tabagismo: Aumento do consumo de bebida alcoólica e na quantidade do consumo diário de cigarro durante esta fase, mediado pelos efeitos estressores da pandemia. CONCLUSÃO: O distanciamento social se constitui como uma medida essencial de proteção à vida, reduzindo a exposição, número de casos e consequentemente a morbimortalidade. Entretanto, essa medida pode resultar em consequências negativas no campo psicossocial e no estilo de vida populacional.

OBJETIVO: As neoplasias esofágicas são um tipo de doença que apresentam considerável morbidade e mortalidade mesmo com o tratamento adequado. Tendo em vista essa perspectiva, esse estudo visa promover uma análise do prognóstico e da abordagem multidisciplinar em relação a essa temática. MÉTODOS: Foi realizada uma revisão da literatura com busca no PubMed e BVSalud. Utilizou-se o descritor “Esophageal Neoplasms”, pesquisados no MeSH. Foram pesquisados artigos originais, trabalhos de conclusão de curso e letras para o editor que estavam nos idiomas inglês e português, os quais foram publicados entre 2006 e 2021. RESULTADOS: O câncer de esôfago possui um prognóstico insatisfatório e deficiente quando detectado tardiamente visto que na maioria dos casos é descoberto em fase de metástase 1. Seu tratamento busca uma interação multidisciplinar e um projeto terapêutico singular visando atender as necessidades do enfermo. Outra finalidade dessa intervenção terapêutica é um aprimoramento na sobrevida do paciente e redução nos níveis e índices de mortalidade e morbidade dos pacientes que apresentam o quadro clínico de neoplasia esofágica. CONCLUSÃO: Observou-se que por ser uma patologia com alta morbimortalidade, o desenvolvimento de uma abordagem multidisciplinar dos pacientes pode promover um prognóstico mental e físico com maior qualidade.

- Abstract
The tumor microenvironment is highly heterogeneous. For gliomas, the tumor-associated inflammatory response is pivotal to support growth and invasion. Factors of glioma growth, inflammation, and invasion, such as the translocator protein (TSPO) and matrix metalloproteinases (MMP), may serve as specific imaging biomarkers of the glioma microenvironment. In this study, noninvasive imaging by PET with [18F]DPA-714 (TSPO) and [18F]BR-351 (MMP) was used for the assessment of localization and quantification of the expression of TSPO and MMP. Imaging was performed in addition to established clinical imaging biomarker of active tumor volume ([18F]FET) in conjunction with MRI. We hypothesized that each imaging biomarker revealed distinct areas of the heterogeneous glioma tissue in a mouse model of human glioma. Tracers were found to be increased 1.4- to 1.7-fold, with [18F]FET showing the biggest volume as depicted by a thresholding-based, volumes of interest analysis. Tumor areas, which could not be detected by a single tracer and/or MRI parameter alone, were measured. Specific compartments of [18F]DPA-714 (14%) and [18F]BR-351 (11%) volumes along the tumor rim could be identified. [18F]DPA-714 (TSPO) and [18F]BR-351 (MMP) matched with histology. Glioma-associated microglia/macrophages (GAM) were identified as TSPO and MMP sources. Multitracer and multimodal molecular imaging approaches may allow us to gain important insights into glioma-associated inflammation (GAM, MMP). Moreover, this noninvasive technique enables characterization of the glioma microenvironment with respect to the disease-driving cellular compartments at the various disease stages.
