The study was aimed to evaluate the performance of a newly developed spectroscopy-based non-invasive and noncontact device (SAMIRA) for the simultaneous measurement of hemoglobin, bilirubin and oxygen saturation as an alternative to the invasive biochemical method of blood sampling. The accuracy of the device was assessed in 4318 neonates having incidences of either anemia, jaundice, or hypoxia. Transcutaneous bilirubin, hemoglobin and blood saturation values were obtained by the newly developed instrument which was corroborated with the biochemical blood tests by expert clinicians. The instrument is trained using Artificial Neural Network Analysis to increase the acceptability of the data. The artificial intelligence incorporated within the instrument determines the disease condition of the neonate. The Pearson’s correlation coefficient, r was found to be 0.987 for hemoglobin estimation and 0.988 for bilirubin and blood gas saturation respectively. The bias and the limits of agreement for the measurement of all the three parameters were within the clinically acceptance limit.

The diverse excitabilities of cells often produce various spiking-bursting oscillations that are found in the neural system. We establish the ability of a fractional-order excitable neuron model with Caputo’s fractional derivative to analyze the effects of its dynamics on the spike train features observed in our results. The significance of this generalization relies on a theoretical framework of the model in which memory and hereditary properties are considered. Employing the fractional exponent, we first provide information about the variations in electrical activities. We deal with the 2D class I and class II excitable Morris-Lecar (M-L) neuron models that show the alternation of spiking and bursting features including MMOs & MMBOs of an uncoupled fractional-order neuron. We then extend the study with the 3D slow-fast M-L model in the fractional domain. The considered approach establishes a way to describe various characteristics similarities between fractional-order and classical integer-order dynamics. Using the stability and bifurcation analysis, we discuss different parameter spaces where the quiescent state emerges in uncoupled neurons. We show the characteristics consistent with the analytical results. Next, the Erdös-Rényi network of desynchronized mixed neurons (oscillatory and excitable) is constructed that is coupled through membrane voltage. It can generate complex firing activities where quiescent neurons start to fire. Furthermore, we have shown that increasing coupling can create cluster synchronization, and eventually it can enable the network to fire in unison. Based on cluster synchronization, we develop a reduced-order model which can capture the activities of the entire network. Our results reveal that the effect of fractional-order depends on the synaptic connectivity and the memory trace of the system. Additionally, the dynamics captures spike frequency adaptation and spike latency that occur over multiple timescales as the effects of fractional derivative, which has been observed in neural computation.

The isometry in crisp graph theory is a well-known fact. But, isometry under a fuzzy environment was developed recently and studied many facts. In a m-polar fuzzy graph, we have to think m components for each node and edge. Since, in our consideration, we consider m components for each nodes as well as edges, therefore we can not handle this type of situation using fuzzy model as their is a single components for this concept. Again, we can not apply bipolar or intuitionistic fuzzy graph model as each edges or nodes have just two components. Thus, these mPFG models give more efficient fuzziness results than other fuzzy model. Also, it is very interesting to develop and analyze such types of mPFGs with examples and related theorems. Considering all those things together, we have presented isometry under a m-polar fuzzy environment. In this paper, we have discussed the isometric m-polar fuzzy graph along with many exciting facts about it. Metric space properties have also been implemented on m-polar fuzzy isometric graph. We also have initiated a generalized fuzzy graph, namely antipodal m-polar fuzzy graphs, along with several issues. The degree of it is also presented along with edge regularity properties. We also give a relation between m-polar fuzzy antipodal graphs and their underlying crisp graphs. Its properties have also been discussed on m-polar fuzzy odd as well as even cycles, complete graphs, etc. Finally, a real-life application on a road network system in a m-polar fuzzy environment using the $$\mu$$ μ -distance concept is also presented.

Ectopic fat deposition is more strongly associated with obesity-related health problems including type 2 diabetes mellitus (T2DM), cardiovascular diseases (CVD), hypothyroidism, arthritis, etc. Our study aimed at identifying the cumulative role of several risk factors in developing obesity and the role of ectopic fat (visceral fat) in predicting cardiovascular disease risk in varied age groups among urban adult academic professionals in West Bengal. 650 adults (Male = 456; Female = 194) associated with the academic job (age 20–65 years) in urban West Bengal were randomly selected for anthropometric, blood biochemical, and questionnaire-based analyses. Optoelectric biosensors measure the conformational changes of biomolecules and their molecular interactions, allowing researchers to use them in different biomedical diagnostics and analysis activities. Among different biosensors, surface plasmon resonance (SPR)-based biosensors utilize label-free and gold-based plasmonic principles with high precision and accuracy, allowing these gold-based biosensors as one of the preferred methods. The dataset generated from these biosensors are being used in different machine learning (ML) models for disease diagnosis and prognosis, but there is a scarcity of models to develop or assess the accuracy of SPR-based biosensors and ensure a reliable dataset for downstream model development. Current study proposed innovative ML-based DNA detection and classification models from the reflective light angles on different gold surfaces of biosensors and associated properties. We have conducted several statistical analyses and different visualization techniques to evaluate the SPR-based dataset and applied t-SNE feature extraction and min-max normalization to differentiate classifiers of low-variances. We experimented with several ML classifiers, namely support vector machine (SVM), decision tree (DT), multi-layer perceptron (MLP), k-nearest neighbors (KNN), logistic regression (LR) and random forest (RF) and evaluated our findings in terms of different evaluation metrics. Our analysis showed the best accuracy of 0.94 by RF, DT and KNN for DNA classification and 0.96 by RF and KNN for DNA detection tasks. Considering area under the receiver operating characteristic curve (AUC) (0.97), precision (0.96) and F1-score (0.97), we found RF performed best for both tasks. Our research shows the potentiality of ML models in the field of biosensor development, which can be expanded to develop novel disease diagnosis and prognosis tools in the future.

This paper presents a novel neuro-adaptive augmented distributed nonlinear dynamic inversion (N-DNDI) controller for consensus of nonlinear multi-agent systems in the presence of unknown external disturbance. N-DNDI is a blending of neural network and distributed nonlinear dynamic inversion (DNDI), a new consensus control technique that inherits the features of Nonlinear Dynamic Inversion (NDI) and is capable of handling the unknown external disturbance. The implementation of NDI based consensus control along with neural networks is unique in the context of multi-agent consensus. The mathematical details provided in this paper show the solid theoretical base, and simulation results prove the effectiveness of the proposed scheme. Background: Purulia is a malaria-prone district in West Bengal, India, with approximately half of the jurisdictional blocks defined as malaria endemic. Methods: We examined the malaria incidence in each block of the district of Purulia for the period between January 1, 2016 and December 31, 2020. Per the Annual Parasite Incidence (API), 20 blocks of Purulia district were assigned to four different categories (0-3) and mapped using ArcGIS software version 10.8. An exponential decay model was fitted to forecast the trend of malaria cases for each block of the district for the 2021-2025 timeline. Findings: There was a sharp decrease in total malaria cases and API from 2016 to 2020. Plasmodium vivax (Pv) cases were decreased by 96.43%, Plasmodium falciparum (Pf) by 90.34%, and mixed infection cases by 92.78%. The majority of cases (72.63%) were found in the 15 year and older age group. Males were more prone to malaria (60.09%) than females. Malaria was highly prevalent among Scheduled Tribes (ST) (48.44%). Six blocks were reported in Category 3 (high risk) and none in Category 0 (no risk) in 2016 while no blocks were determined to be in Category 3 and three blocks were in Category 0 in 2020. The exponential decay model prediction is oriented towards gaining malaria-free status in thirteen blocks of the Purulia district by 2025. Interpretation: By efficiently diagnosing malaria, implementing a GIS system to track malaria incidences, fair usage of Long-lasting insecticidal nets (LLINs), and conducting effective awareness campaigns, malaria control could be possible. In this article, we demonstrate the solution methodology of start-up electrokinetic flow of non-Newtonian fluids in a microfluidic channel having square cross-section using Spreadsheet analysis tool. In order to incorporate the rheology of the non-Newtonian fluids, we take into consideration the Ostwald-de Waele power law model. By making a comprehensive discussion on the implementation details of the discretized form of the transport equations in Spreadsheet analysis tool, and establishing the analytical solution for a special case of the start-up flow, we compare the results both during initial transience as well as in case of steady-state scenario. Also, to substantiate the efficacy of the proposed spreadsheet analysis in addressing the detailed flow physics of rheological fluids, we verify the results for several cases with the corresponding numerical results. It is found that the solution obtained from the Spreadsheet analysis is in good agreement with the numerical results—a finding supporting spreadsheet analysis's suitability for capturing the fine details of microscale flows. We strongly believe that our analysis study will open up a new research scope in simulating microscale transport process of non-Newtonian fluids in the framework of cost-effective and non-time consuming manner.

Globally 3 billion people are consuming water with moderately high concentrations of fluoride and arsenic. The development of a simple point of care (PoC) device or home device for the detection of fluoride/arsenic ensures safety before consuming water. Till date, lateral flow assay (LFA) based PoC devices can detect nucleic acids, viruses and diseases. An aluminium complex of rhodamine B functionalized oxacalix[4]arene (L) was designed to execute the LFA-based PoC device. Initially, Al3+ and Fe3+ ions were involved in complexation with the rhodamine B functionalized oxacalix[4]arene (L), resulting C1 (L-Al3+) and C2 (L-Fe3+) complexes respectively. The receptor L, as well as the probes (C1, C2), were characterized thoroughly using mass spectroscopy, FTIR, NMR, and EA. C1 and C2 were further utilized as recyclable probes for the detection of aqueous fluoride (21 ppb) and arsenate (1.92 ppb) respectively. The computational calculation indicates that upon complexation, the spirolactam ring opening at the rhodamine B site leads to optoelectronic changes. The consistency of LFA-based portable sensing device has been tested with water samples, synthetic fluoride standards and dental care products like toothpaste and mouthwash with concentrations ≥ 3 ppm. Moreover, fixed cell imaging experiments were performed to ascertain the in-vitro sensing phenomena.

Marker-assisted breeding and tagging of important quantitative trait loci for beneficial traits are two important strategies for the genetic improvement of plants. However, the scarcity of diverse and informative genetic markers covering the entire tea genome limits our ability to achieve such goals. In the present study, we used a comparative genomic approach to mine the tea genomes of Camellia sinensis var. assamica (CSA) and C. sinensis var. sinensis (CSS) to identify the markers to differentiate tea genotypes. In our study, 43 and 60 Camellia sinensis miniature inverted-repeat transposable element (CsMITE) families were identified in these two sequenced tea genomes, with 23,170 and 37,958 putative CsMITE sequences, respectively. In addition, we identified 4912 non-redundant, Camellia sinensis intron length polymorphic (CsILP) markers, 85.8% of which were shared by both the CSS and CSA genomes. To validate, a subset of randomly chosen 10 CsMITE markers and 15 CsILP markers were tested and found to be polymorphic among the 36 highly diverse tea genotypes. These genome-wide markers, which were identified for the first time in tea plants, will be a valuable resource for genetic diversity analysis as well as marker-assisted breeding of tea genotypes for quality improvement.

Wnt1 is the first mammalian Wnt gene, which is discovered as proto-oncogene and in human the gene is located on the chromosome 12q13. Mutations in Wnt1 are reported to be associated with various cancers and other human diseases. The structural and functional consequences of most of the non-synonymous SNPs (nsSNPs), present in the human Wnt1 gene, are not known. In the present work, extensive bioinformatics analyses are used to screen 292 nsSNPs of Wnt1 for predicting pathogenic and harmless polymorphisms. We have identified 10 highly deleterious nsSNPs among which 7 are located within the highly conserved areas. These 10 nsSNPs are also predicted to affect the post-translational modifications of Wnt1. Further, structure based stability analyses of these 10 highly deleterious nsSNPs revealed 8 variants as highly destabilizing. These 8 highly destabilizing variants were shown to have high BC score and high RMSIP score from normal mode analyses. Based on the deformation energies, obtained from the normal mode analyses, variants like G169A, G169S, G331R and G331S were found to be unstable. Molecular Dynamics (MD) simulations revealed structural stability and fluctuation of WT Wnt1 and its prioritized variants. RMSD remained fluctuating mostly between 4 and 5 Å and occasionally between 3.5 and 5.5 Å ranges. RMSF in the CTD region (residues 330–360) of the binding pocket were lower compared to that of WT. Studying the impacts of nsSNPs on the binding interface of Wnt1 and seven Frizzled receptors have predicted substitutions which can stabilize or destabilize the binding interface. We have found that Wnt1 and FZD8-CRD is the best docked complex in our study. MD simulation based analyses of wild type Wnt1-FZD8-CRD complex and the 8 prioritized variants revealed that RMSF was higher in the unstructured regions and RMSD remained fluctuating in the region of 5 Å ± 1 Å. We have also observed differential Wnt1 gene expression pattern in normal, tumor and metastatic conditions across different tissues. Wnt1 gene expression was significantly higher in metastatic tissues of lungs, colon and skin; and was significantly lower in metastatic tissues of breast, esophagus and kidney. We have also found that Wnt1 deregulation is associated with survival outcome in patients with gastric and breast cancer. Furthermore, these computationally screened highly deleterious nsSNPs of Wnt1 can be analyzed in population based genetic studies and may help understand the Wnt1 associated diseases.

Anti-microbial resistant infection is predicted to be alarming in upcoming years. In the present study, we proposed co-localization of two model drugs viz., rifampicin and benzothiazole used in anti-tuberculosis and anti-fungal agents respectively in a nanoscopic cationic micelle (cetyl triethyl ammonium bromide) with hydrodynamic diameter of 2.69 nm. Sterilization effect of the co-localized micellar formulation against a model multi-drug resistant bacterial strain viz., Methicillin resistant Staphylococcus aureus was also investigated. 99.88% decrease of bacterial growth in terms of colony forming unit was observed using the developed formulation. While Dynamic Light Scattering and Forsters Resonance Energy Transfer between benzothiazole and rifampicin show co-localization of the drugs in the nanoscopic micellar environment, analysis of time-resolved fluorescence decays by Infelta-Tachiya model and the probability distribution of the donor–acceptor distance fluctuations for 5 μM,10 μM and 15 μM acceptor concentrations confirm efficacy of the co-localization. Energy transfer efficiency and the donor acceptor distance are found to be 46% and 20.9 Å respectively. We have also used a detailed computational biology framework to rationalize the sterilization effect of our indigenous formulation. It has to be noted that the drugs used in our studies are not being used for their conventional indication. Rather the co-localization of the drugs in the micellar environment shows a completely different indication of their use in the remediation of multi-drug resistant bacteria revealing the re-purposing of the drugs for potential use in hospital-born multi-drug resistant bacterial infection.

The imidazolium compound Ym155 was first reported to be a survivin inhibitor. Ym155 potently induces cell death of many types of cancer cells in preclinical studies. However, in phase II clinical trials Ym155 failed to demonstrate a significant benefit. Studies have suggested that the cytotoxic effects of Ym155 in cancer cells are not mediated by the inhibition of survivin. Understanding the mechanism by which Ym155 induces cell death would provide important insight how to improve its efficacy as a cancer therapeutic. We demonstrate a novel mechanism by which Ym155 induces cell death by localizing to the mitochondria causing mitochondrial dysfunction. Our studies suggest that Ym155 binds mitochondrial DNA leading to a decrease in oxidative phosphorylation, decrease in TCA cycle intermediates, and an increase in mitochondrial permeability. Furthermore, we show that mitochondrial stress induced by Ym155 and other mitochondrial inhibitors activates AMP-activated kinase leading to the downregulation to bone morphogenetic protein (BMP) signaling. We provide first evidence that Ym155 initiates cell death by disrupting mitochondrial function.

Phosphorus (P) is a crucial structural component of living systems and central to modern bioenergetics. P cycles through terrestrial geochemical reservoirs via complex physical and chemical processes. Terrestrial life has altered these fluxes between reservoirs as it evolved, which is why it is of interest to explore planetary P flux evolution in the absence of biology. This is especially true, since environmental P availability affects life’s ability to alter other geochemical cycles, which could then be an example of niche construction. Understanding how P reservoir transport affects environmental P availability helps parameterize how the evolution of P reservoirs influenced the emergence of life on Earth, and potentially other planetary bodies. Geochemical P fluxes likely change as planets evolve, and element cycling models that take those changes into account can provide insights on how P fluxes evolve abiotically. There is considerable uncertainty in many aspects of modern and historical global P cycling, including Earth’s initial P endowment and distribution after core formation and how terrestrial P interactions between reservoirs and fluxes and their rates have evolved over time. We present here a dynamical box model for Earth’s abiological P reservoir and flux evolution. This model suggests that in the absence of biology, long term planetary geochemical cycling on planets similar to Earth with respect to geodynamism tends to bring P to surface reservoirs, and biology, including human civilization, tends to move P to subductable marine reservoirs.

An increased nitrate (NO3−) concentration in groundwater has been a rising issue on a global scale in recent years. Different consumption mechanisms clearly illustrate the adverse effects on human health. The goal of this present study is to assess the natural and anthropogenic NO3− concentrations in groundwater in a semi arid area of Rajasthan and its related risks to human health in the different groups of ages such as children, males, and females. We have found that most of the samples (n = 90) were influenced by anthropogenic activities. The background level of NO3− had been estimated as 7.2 mg/L using a probabilistic approach. About 93% of nitrate samples exceeded the background limit, while 28% of the samples were beyond the permissible limit of 45 mg/L as per the BIS limits. The results show that the oral exposure of nitrate was very high as compare to dermal contact. With regard to the non-carcinogenic health risk, the total Hazard Index (HITotal) values of groundwater nitrate were an average of 0.895 for males, 1.058 for females, and 1.214 for children. The nitrate health risk assessment shows that about 38%, 46%, and 49% of the samples constitute the non-carcinogenic health risk to males, females, and children, respectively. Children were found to be more prone to health risks due to the potential exposure to groundwater nitrate.

Complex interactions in tumor microenvironment between ECM (extra-cellular matrix) and cancer cell plays a central role in the generation of tumor supportive microenvironment. In this study, the expression of ECM-related genes was explored for prognostic and immunological implication in clear cell renal clear cell carcinoma (ccRCC). Out of 964 ECM genes, higher expression (z-score > 2) of 35 genes showed significant association with overall survival (OS), progression-free survival (PFS) and disease-specific survival (DSS). On comparison to normal tissue, 12 genes (NUDT1, SIGLEC1, LRP1, LOXL2, SERPINE1, PLOD3, ZP3, RARRES2, TGM2, COL3A1, ANXA4, and POSTN) showed elevated expression in kidney tumor (n = 523) compared to normal (n = 100). Three gene families in plants viz. Argonaute (AGOs), Dicer-like (DCLs) and RNA dependent RNA polymerase (RDRs) constitute the core components of small RNA mediated gene silencing machinery. The present study endeavours to identify members of these gene families in tea and to investigate their expression patterns in different tissues and various stress regimes. Using genome-wide analysis, we have identified 18 AGOs, 5 DCLs and 9 RDRs in tea, and analyzed their phylogenetic relationship with orthologs of Arabidopsis thaliana. Gene expression analysis revealed constitutive expression of CsAGO1 in all the studied tissues and stress conditions, whereas CsAGO10c showed most variable expression among all the genes. CsAGO10c gene was found to be upregulated in tissues undergoing high meristematic activity such as buds and roots, as well as in Exobasidium vexans infected samples. CsRDR2 and two paralogs of CsAGO4, which are known to participate in biogenesis of hc-siRNAs, showed similarities in their expression levels in most of the tea plant tissues. This report provides first ever insight into the important gene families involved in biogenesis of small RNAs in tea. The comprehensive knowledge of these small RNA biogenesis purveyors can be utilized for tea crop improvement aimed at stress tolerance and quality enhancement.

Indigenous folk rice cultivars often possess remarkable but unrevealed potential in terms of nutritional attributes and biotic stress tolerance. The unique cooking qualities and blissful aroma of many of these landraces make it an attractive low-cost alternative to high priced Basmati rice. Sub-Himalayan Terai region is bestowed with great agrobiodiversity in traditional heirloom rice cultivars. In the present study, ninety-nine folk rice cultivars from these regions were collected, purified and characterized for morphological and yield traits. Based on traditional importance and presence of aroma, thirty-five genotypes were selected and analyzed for genetic diversity using micro-satellite marker system. The genotypes were found to be genetically distinct and of high nutritive value. The resistant starch content, amylose content, glycemic index and antioxidant potential of these genotypes represented wide variability and ‘Kataribhog’, ‘Sadanunia’, ‘Chakhao’ etc. were identified as promising genotypes in terms of different nutritional attributes. These cultivars were screened further for resistance against blast disease in field trials and cultivars like ‘Sadanunia’, ‘T4M-3-5’, ‘Chakhao Sampark’ were found to be highly resistant to the blast disease whereas ‘Kalonunia’, ‘Gobindabhog’, ‘Konkanijoha’ were found to be highly susceptible. Principal Component analysis divided the genotypes in distinct groups for nutritional potential and blast tolerance. The resistant and susceptible genotypes were screened for the presence of the blast resistant pi genes and association analysis was performed with disease tolerance. Finally, a logistic model based on phenotypic traits for prediction of the blast susceptibility of the genotypes is proposed with more than 80% accuracy.

Ultrafast plasma dynamics play a pivotal role in the relativistic high harmonic generation, a phenomenon that can give rise to intense light fields of attosecond duration. Controlling such plasma dynamics holds key to optimize the relevant sub-cycle processes in the high-intensity regime. Here, we demonstrate that the optimal coherent combination of two intense ultrashort pulses centered at two-colors (fundamental frequency, $$\omega$$ ω and second harmonic, $$2\omega$$ 2 ω ) can lead to an optimal shape in relativistic intensity driver field that yields such an extraordinarily sensitive control. Conducting a series of two-dimensional (2D) relativistic particle-in-cell (PIC) simulations carried out for currently achievable laser parameters and realistic experimental conditions, we demonstrate that an appropriate combination of $$\omega -2\omega$$ ω - 2 ω along with a precise delay control can lead to more than three times enhancement in the resulting high harmonic flux. Finally, the two-color multi-cycle field synthesized with appropriate delay and polarization can all-optically suppress several attosecond bursts while favourably allowing one burst to occur, leading to the generation of intense isolated attosecond pulses without the need of any sophisticated gating techniques.

In this study, the influence of a pulsed fiber laser of 250 W power with a spot size of 40 µm was successfully analyzed during scabbling of six types of cement mortar and three types of ultra-high-performance concrete (UHPC). Confocal microscopy on the surface of the scabbled samples elucidated the formation of three distinct zones: glassy layer (GL), partially melted zone (PMZ), and heat-affected zone (HAZ) with unique morphological appearances. The glassy layer exhibited bubble formation, whereas cracks were spotted alongside the scabbled area. The difference in scabbling depth between the beginning and end of the process was revealed by using 3D topography images. Moreover, the development of pores and the changes in the microstructure of each zone were observed by using scanning electron microscopy (SEM). Further energy dispersive X-ray (EDX) analysis also revealed significant changes in the percentage of silicon and calcium inside the glassy layer and non-processed zone (NPZ).

Assessment of morphodynamic groundwater reserves is important for the sustainable management of water resources. It is a truth that groundwater resource evaluation is anxious with the ambiguity of its several factors and employing methods. Thus, an information-based model has been hypothesized to assess natural groundwater reserves in a morphodynamic system in a part of the Ganga basin of Northern India, where the spatial variability in natural groundwater reserve exists. Marginal information of rainfall data, and transinformation among the rainfall, and monthly depth to groundwater level measurement at 50 wells in a dense monitoring network were used for evaluating natural groundwater reserve. The results indicate that an average recharge rate is about 246 mm/monsoon and or 32.65% of the seasonal rainfall, and its values are well-correlated with the soil infiltration rate. It has been found that the estimated recharge rates are about 54.08, 45.85, 33.77, 32.48, and 32.14% of the seasonal rainfall in an active flood plain, back swamp, natural levees, flood plain, and palaeochannel, respectively. The calculated annual rainfall input to groundwater reserve is found about 127.98 MCM/monsoon rainfall, which could be employed for sustainable management of groundwater resources in the morphodynamic system of the Ganga river basin.

Dense data acquisition for 3-D high-resolution aquifer mapping through heliborne transient electromagnetic (HTEM) survey is continually not possible due to various technical and administrative constraints. Consequently, we apply ground geophysical surveys at possibly closer spacing to collect the sub-surface information in the no-fly area, which provides only a regional aquifer picture. In the area near Patna of Northern India, an extent of 18% was covered under the HTEM survey, and the rest was surveyed by ground geophysical methods. Both data are integrated using the theory of regionalized variables. The parameters of multi-aquifers i.e., top of the first aquifer, top of the separating clay layer, top and the bottom of second aquifer, are estimated together with their respective resistivities. The estimations are made at an interval of 250 m, practically an appropriate distance at which dense data generation was carried out using the HTEM survey. The integrated approach generates the data in the no-fly area with the same spatial density as the flown area. With this, we achieved the goal of completing the 3-D aquifer mapping of the entire area with dense data at high spatial resolution. This is a unique finding to manage the handicapped situation in this HTEM surveys, and an aide to overcome such constraints with cost-effectiveness.

Conservation agriculture (CA), which encompasses minimum soil disturbance, residue retention either through crop residue, or cover crops and crop diversification-based crop management practices can modify the status of pest dynamics and activities under the changing climatic scenarios. CA has been advocated extensively to optimize the use of available resources, maintain the environmental quality, enhance crop productivity, and reduce the climate change impacts. Information related to the impacts of long-term CA-production systems under rice-based cropping systems on pest status is lacking, particularly in middle Indo-Gangetic Plains (MIGP). Under CA, puddling is completely avoided, and rice is directly sown or transplanted to maintain better soil health. Different sets of experimentations including farmers practice, partial CA and full CA (CA) as treatments in rice-based cropping systems, were established from 2009, 2015 and 2016 to understand the long-term impacts of CA on pest dynamics. In this study, direct and indirect effects of tillage (zero, reduced and conventional tillage), residue retention and cropping sequences on abundance and damage by pests were investigated. After 4–5 years of experimentation, populations of oriental armyworm [Mythinma (Leucania) (Pseudaletia) separata (Wlk.)] in wheat, mealybug [Brevennia rehi (Lindinger)] and bandicoot rat [Bandicota bengalensis (Gray)] in rice were found to increase abnormally in CA-based production systems. Conventionally tilled plots had a significant negative effect while residue load in zero-tilled plots had a significant positive effect on larval population build-up of M. separata. Zero tillage had a higher infestation of mealybug (52–91% infested hills) that used grassy weeds (Echinochloa colona, Echinochloa crusgalli, Cynodon dactylon, Leptochloa chinensis and Panicum repense) as alternate hosts. Cropping sequences and no disturbance of soil and grassy weeds had higher live burrow counts (4.2 and 13.7 burrows as compared to 1.47 and 7.53 burrows per 62.5 m2 during 2019–2020 and 2020–2021, respectively) and damaged tillers (3.4%) in CA-based practices. Based on the present study, pest management strategies in CA need to be revisited with respect to tillage, residue retention on soil surface, grassy weeds in field and cropping sequences to deliver the full benefits of CA in MIGP to achieve the sustainable development goals under the climate change scenarios.

The COVID-19 pandemic has resulted in significant diversion of human and material resources to COVID-19 diagnostics, to the extent that influenza viruses and co-infection in COVID-19 patients remains undocumented and pose serious public-health consequences. We optimized and validated a highly sensitive RT-PCR based multiplex-assay for the detection of SARS-CoV-2, influenza A and B viruses in a single-test. This study evaluated clinical specimens (n = 1411), 1019 saliva and 392 nasopharyngeal swab (NPS), tested using two-assays: FDA-EUA approved SARS-CoV-2 assay that targets N and ORF1ab gene, and the PKamp-RT-PCR based assay that targets SARS-CoV-2, influenza viruses A and B. Of the 1019 saliva samples, 17.0% (174/1019) tested positive for SARS-CoV-2 using either assay. The detection rate for SARS-CoV-2 was higher with the multiplex assay compared to SARS-specific assay [91.9% (160/174) vs. 87.9% (153/174)], respectively. Of the 392 NPS samples, 10.4% (41/392) tested positive for SARS-CoV-2 using either assay. The detection rate for SARS-CoV-2 was higher with the multiplex assay compared to SARS-specific assay [97.5% (40/41) vs. 92.1% (39/41)], respectively. This study presents clinical validation of a multiplex-PCR assay for testing SARS-CoV-2, influenza A and B viruses, using NPS and saliva samples, and demonstrates the feasibility of implementing the assay without disrupting the existing laboratory workflow.

Fractional-order dynamics of excitable systems can be physically described as a memory dependent phenomenon. It can produce diverse and fascinating oscillatory patterns for certain types of neuron models. To address these characteristics, we consider a nonlinear fast-slow FitzHugh-Rinzel (FH-R) model that exhibits elliptic bursting at a fixed set of parameters with a constant input current. The generalization of this classical order model provides a wide range of neuronal responses (regular spiking, fast-spiking, bursting, mixed-mode oscillations, etc.) in understanding the single neuron dynamics. So far, it is not completely understood to what extent the fractional-order dynamics may redesign the firing properties of excitable systems. We investigate how the classical order system changes its complex dynamics and how the bursting changes to different oscillations with stability and bifurcation analysis depending on the fractional exponent (0 α ≤ 1). This occurs due to the memory trace of the fractional-order dynamics. The firing frequency of the fractional-order FH-R model is less than the classical order model, although the first spike latency exists there. Further, we investigate the responses of coupled FH-R neurons with small coupling strengths that synchronize at specific fractional-orders. The interesting dynamical characteristics suggest various neurocomputational features that can be induced in this fractional-order system which enriches the functional neuronal mechanisms.

In this article, we highlight the alterations in the photoinduced electron transfer (ET) and hydrogen atom transfer (HAT) pathways between an anti-tumor drug vitamin-K3 (MQ) and a nucleobase adenine (ADN) in the presence of gold (Au) and iron (Fe) nanoparticles (NPs). Inside the confined micellar media, with laser flash photolysis corroborated with an external magnetic field (MF), we have detected the transient geminate radicals of MQ and ADN, photo-generated through ET and HAT. We observe that the presence of AuNP on the MQ-ADN complex (AuMQ-ADN) assists HAT by limiting the ET channel, on the other hand, FeNP on the MQ-ADN complex (FeMQ-ADN) mostly favors a facile PET. We hypothesize that through selective interactions of the ADN molecules with AuNP and MQ molecules with FeNP, a preferential HAT and PET process is eased. The enhanced HAT and PET have been confirmed by the escape yields of radical intermediates by time-resolved transient absorption spectroscopy in the presence of MF.

Presently, rice-fallows are targeted for cropping intensification in South Asia. Rice-fallows a rainfed mono-cropping system remain fallow after rice due to lack of irrigation facilities and poor socio-economic condition of the farmers. Nevertheless, there is the scope of including ecologically adaptable winter crops in water-limited rice-fallow conditions with effective moisture conservation practices. The study aimed to identify the winter-crops that are adaptable and productive in rice-fallow conditions and to evaluate the different tillage-based crop establishment practices for soil moisture conservation, grain yield, economics, and sustainability parameters. Six different crop establishment and residue management (CERM) practices viz., zero-tillage direct seeded rice (ZTDSR), zero-tillage transplanted rice (ZTTPR), puddled transplanted rice (PTR), ZTDSR with rice residue retention (ZTDSRR+), ZTTPR with rice residue retention (ZTTPRR+), PTR with rice residue retention (PTRR+) as main-plot treatment and five winter crops (chickpea, lentil, safflower, linseed, and mustard) as sub-plot treatment were evaluated in a split-plot design. The productivity of grain legumes (chickpea and lentil) was higher over oilseed crops in rice-fallow conditions with an order of chickpea > lentil > safflower > mustard > linseed. Among the CERM practices, ZTDSRR+ and ZTDSR treatments increased the grain yield of all the winter crops over PTR treatment, which was primarily attributed to higher soil moisture retention for an extended period. Grain yield increment with conservation tillage practices was highly prominent in safflower (190%) followed by lentil (93%) and chickpea (70%). Rice grain yield was higher (7–35%) under PTR treatment followed by ZTDSR treatment. Conservation tillage practices (ZTDSR, ZTTPR) reduced energy use (11–20%) and increased the energy ratio over conventional tillage practice (PTR), higher in rice-safflower, rice-lentil and rice-chickpea rotations. Higher net return was attained in rice-safflower and rice-chickpea rotations with ZTDSRR+ treatment. Predicted emission of greenhouse gases was markedly reduced in ZTDSR treatment (30%) compared to ZTTPR and PTR treatments. Hence, the study suggests that cropping intensification of rice-fallows with the inclusion of winter crops like chickpea, lentil, and safflower following conservation tillage practices (ZTDSRR+ in particular) could be the strategic options for achieving the higher system productivity, economic returns, and energy use efficiency with the reduced emission of greenhouse gases.

This study analyzes and forecasts the long-term Spatio-temporal changes in rainfall using the data from 1901 to 2015 across India at meteorological divisional level. The Pettitt test was employed to detect the abrupt change point in time frame, while the Mann-Kendall (MK) test and Sen’s Innovative trend analysis were performed to analyze the rainfall trend. The Artificial Neural Network-Multilayer Perceptron (ANN-MLP) was employed to forecast the upcoming 15 years rainfall across India. We mapped the rainfall trend pattern for whole country by using the geo-statistical technique like Kriging in ArcGIS environment. Results show that the most of the meteorological divisions exhibited significant negative trend of rainfall in annual and seasonal scales, except seven divisions during. Out of 17 divisions, 11 divisions recorded noteworthy rainfall declining trend for the monsoon season at 0.05% significance level, while the insignificant negative trend of rainfall was detected for the winter and pre-monsoon seasons. Furthermore, the significant negative trend (−8.5) was recorded for overall annual rainfall. Based on the findings of change detection, the most probable year of change detection was occurred primarily after 1960 for most of the meteorological stations. The increasing rainfall trend had observed during the period 1901–1950, while a significant decline rainfall was detected after 1951. The rainfall forecast for upcoming 15 years for all the meteorological divisions’ also exhibit a significant decline in the rainfall. The results derived from ECMWF ERA5 reanalysis data exhibit that increasing/decreasing precipitation convective rate, elevated low cloud cover and inadequate vertically integrated moisture divergence might have influenced on change of rainfall in India. Findings of the study have some implications in water resources management considering the limited availability of water resources and increase in the future water demand.

Patients with Parkinson’s disease and focal dystonia have difficulty in generating and preventing movement. Reaction time (RT) and stop signal reaction time (SSRT) measure the speed to initiate and stop a movement respectively. We developed a portable device to assess RT and SSRT. This incorporated a novel analysis to measure SSRT more efficiently (optimal combination SSRT, ocSSRT). After validation ocSSRT was measured in Parkinson’s disease patients without dyskinesia (PD), cervical dystonia (CD) and writer’s cramp. We also assessed how ocSSRT responded to L-dopa in PD patients and botulinum toxin injections in CD patients. Participants were instructed to release a button following a green LED flash on the device. We examined the association between socioeconomic status (SES) and continuum of care (CoC) completion rate in maternal, neonatal, and child health among mothers in Bangladesh. We used data from the Bangladesh Demographic Health Survey (BDHS) from 2017 to 2018. Our findings were based on the responses of 1527 married women who had at least one child aged 12 to 23 months at the time of the survey. As a measure of SES, we focused on the standard of living (hereinafter referred to as wealth). The CoC for maternal and child health (MNCH) services is the study's outcome variable. The CoC was calculated using seven MNCH interventions: four or more antenatal care (ANC) visits with a skilled practitioner, delivery by a skilled birth attendant, post-natal care for mothers (PNCM) within two days of giving birth, post-natal care for newborns (PNCM) within two days of birth, immunization, age-appropriate breastfeeding, and maternal current use of modern family planning (FP) methods. Only 18.1% of Bangladeshi women completed all seven MNCH care interventions during the reproductive life span. Participants in the high SES group were 2.30 times (95% confidence interval [CI] 1.61–3.28) more likely than those in the low SES group to have higher composite care index (CCI) scores. Women with secondary or higher secondary education, as well as women who were exposed to mass media at least once a week, women who lived in an urban setting, women who had an intended pregnancy, and women with one parity, are associated with high CCI scores when other sociodemographic variables are considered. The complete CoC for MNCH reveals an extremely low completion rate, which may suggest that Bangladeshi mothers, newborns, and children are not receiving the most out of their present health care. Participants in the high SES group displayed higher CCI values than those in the low SES group, indicating that SES is one of the primary drivers of completion of CoC for MNCH services.

Evolutionary trends may underlie some aspects of the risk for common, non-communicable disorders, including psychiatric disease. We analyzed whole exome sequencing data from 80 unique individuals from India coming from families with two or more individuals with severe mental illness. We used Population Branch Statistics (PBS) to identify variants and genes under positive selection and identified 75 genes as candidates for positive selection. Of these, 20 were previously associated with Schizophrenia, Alzheimer’s disease and cognitive abilities in genome wide association studies. We then checked whether any of these 75 genes were involved in common biological pathways or related to specific cellular or molecular functions. We found that immune related pathways and functions related to innate immunity such as antigen binding were over-represented. We also evaluated for the presence of Neanderthal introgressed segments in these genes and found Neanderthal introgression in a single gene out of the 75 candidate genes. However, the introgression pattern indicates the region is unlikely to be the source for selection. Our findings hint at how selection pressures in individuals from families with a history of severe mental illness may diverge from the general population. Further, it also provides insights into the genetic architecture of severe mental illness, such as schizophrenia and its link to immune factors.

Imaging modalities combined with a multimodal nanocomposite contrast agent hold great potential for significant contributions in the biomedical field. Among modern imaging techniques, photoacoustic (PA) and fluorescence (FL) imaging gained much attention due to their non-invasive feature and the mutually supportive characteristic in terms of spatial resolution, penetration depth, imaging sensitivity, and speed. In this present study, we synthesized IR783 conjugated chitosan-polypyrrole nanocomposites (IR-CS-PPy NCs) as a theragnostic agent used for FL/PA dual-modal imaging. A customized FL and photoacoustic imaging system was constructed to perform required imaging experiments and create high-contrast images. The proposed nanocomposites were confirmed to have great biosafety, essentially a near-infrared (NIR) absorbance property with enhanced photostability. The in vitro photothermal results indicate the high-efficiency MDA-MB-231 breast cancer cell ablation ability of IR-CS-PPy NCs under 808 nm NIR laser irradiation. The in vivo PTT study revealed the complete destruction of the tumor tissues with IR-CS-PPy NCs without further recurrence. The in vitro and in vivo results suggest that the demonstrated nanocomposites, together with the proposed imaging systems could be an effective theragnostic agent for imaging-guided cancer treatment.

Marine mammal strandings provide vital information on species’ life histories, population health and status of marine ecosystems. Opportunistic reporting of strandings also serve as a powerful low-cost tool for monitoring these elusive mammals. We collated data over ~ 270 years available through various open access databases, reports and publications. Annual strandings along the Indian coast (mean = 11.25 ± SE 9.1) increased in the last two years of the study (2015–2017, mean = 27.66 ± SE 8.5 strandings /year). We found that stranding events spike during June—September along the west coast and during December–January along the east coast. We identified several sections of the coastline, such as Mumbai (0.38 strandings/km), Kozhikode (0.28 strandings/km), Tuticorin (0.4 strandings/km), Rameswaram (1.82 strandings/km), Chennai (0.32 strandings/km) and Bhubaneshwar (0.26 strandings/km) with a higher number of stranded animals reported. Emerging Hotspot Analysis located new and consecutive hotspots along the north-west coast, and sporadic hotspots along the south-east coast. We recommend establishing regional stranding response centres at the identified hotspots coordinated by a National Stranding Centre with adequately trained personnel and central funding support. Regular stranding response training programs for field veterinarians, and frontline personnel of State Forest Departments near stranding hotspots would provide an improved understanding of marine mammal health and threats in Indian waters. Further, the suggested National Stranding Centre needs to maintain a ‘National Stranding Database’ for long-term marine mammal conservation planning in India.

A microscale ecological study using the closure approach to understand the impact of productivity controlled by geographical and seasonal variations on the intermittency of phytoplankton is done in this paper. Using this approach for a nutrient–phytoplankton model with Holling type III functional response, it has been shown how the dynamics of the system can be affected by the environmental fluctuations triggered by the impact of light, temperature, and salinity, which fluctuate with regional and seasonal variations. Reynold’s averaging method in space, which results in expressing the original components in terms of its mean (average value) and perturbation (fluctuation) has been used to determine the impact of growth fluctuation in phytoplankton distribution and in the intermittency of phytoplankton spreading (variance). Parameters are estimated from the nature of productivity and spread of phytoplankton density during field observation done at four different locations of Tokyo Bay. The model validation shows that our results are in good agreement with the field observation and succeeded in explaining the intermittent phytoplankton distribution at different locations of Tokyo Bay, Japan, and its neighboring coastal regions.

An essential attribute of many fractal structures is self-similarity. A Sierpinski gasket (SPG) triangle is a promising example of a fractal lattice that exhibits localized energy eigenstates. In the present work, for the first time we establish that a mixture of both extended and localized energy eigenstates can be generated yeilding mobility edges at multiple energies in presence of a time-periodic driving field. We obtain several compelling features by studying the transmission and energy eigenvalue spectra. As a possible application of our new findings, different thermoelectric properties are discussed, such as electrical conductance, thermopower, thermal conductance due to electrons and phonons. We show that our proposed method indeed exhibits highly favorable thermoelectric performance. The time-periodic driving field is assumed through an arbitrarily polarized light, and its effect is incorporated via Floquet-Bloch ansatz. All transport phenomena are worked out using Green’s function formalism following the Landauer–Büttiker prescription.

Shigellosis, caused by Shigella bacterial spp., is one of the leading causes of diarrheal morbidity and mortality. An increasing prevalence of multidrug-resistant Shigella species has revived the importance of bacteriophages as an alternative therapy to antibiotics. In this study, a novel bacteriophage, Sfk20, has been isolated from water bodies of a diarrheal outbreak area in Kolkata (India) with lytic activity against many Shigella spp. Phage Sfk20 showed a latent period of 20 min and a large burst size of 123 pfu per infected cell in a one-step growth analysis. Phage-host interaction and lytic activity confirmed by phage attachment, intracellular phage development, and bacterial cell burst using ultrathin sectioning and TEM analysis. The genomic analysis revealed that the double-stranded DNA genome of Sfk20 contains 164,878 bp with 35.62% G + C content and 241 ORFs. Results suggested phage Sfk20 to include as a member of the T4 myoviridae bacteriophage group. Phage Sfk20 has shown anti-biofilm potential against Shigella species. The results of this study imply that Sfk20 has good possibilities to be used as a biocontrol agent.

Adjuvant trastuzumab has been associated with superior survival in women with ≥ T1c or node-positive HER2-positive early-stage breast cancer; however, there is a lack of phase III trials in women with T1a/bN0 disease. Our study aimed to assess the outcomes of women with HER2-positive T1a/bN0 breast cancer who received adjuvant trastuzumab in Saskatchewan, Canada. We evaluated all women diagnosed with HER2-positive T1a/bN0 breast cancer in Saskatchewan between 2008 and 2017. We performed Cox proportional multivariable analysis to determine factors correlated with survival. In addition, inverse probability treatment weighting (IPTW) using propensity score was performed to assess benefit of adjuvant trastuzumab. Ninety-one eligible women with a median age of 61 years (range 30–89) were identified. Thirty-nine (43%) women received adjuvant trastuzumab. Women who received trastuzumab were younger and had a higher rate of T1b disease. Overall, 3% of women who received trastuzumab compared to 12% of women who did not receive trastuzumab developed breast cancer recurrence (p = 0.23). Five-year disease-free survival (DFS) of women who received adjuvant trastuzumab was 94.8% compared to 82.7% of women who did not receive trastuzumab (p = 0.22). Five-year overall survival was 100% of women who received trastuzumab compared to 90.4% of women who did not receive adjuvant trastuzumab (p = 0.038). In the multivariable analysis, grade III tumors were correlated with inferior DFS (hazard ratio [HR] 5.5, 95% CI [1.7–17.7]). The propensity score using the inverse probability of treatment weighting showed that lack of adjuvant trastuzumab was correlated inferior DFS, with an HR of 4 (95% CI 1.05–15.5). Women with HER2-positive T1a/bN0 breast cancer had overall low recurrence of breast cancer. However, the results of this exploratory analysis indicate that women who received adjuvant trastuzumab had better survival.

We report spin-wave excitations in annular antidot lattice fabricated from 15 nm-thin Ni80Fe20 flm. The nanodots of 170 nm diameters are embedded in the 350 nm (diameter) antidot lattice to form the annular antidot lattice, which is arranged in a square lattice with edge-to-edge separation of 120 nm. A strong anisotropy in the spin-wave modes are observed with the change in orientation angle ({\phi}) of the in-plane bias magnetic field by using Time-resolved Magneto-optic Kerr microscope. A fattened fourfold rotational symmetry, mode hopping and mode conversion leading to mode quenching for three prominent spin-wave modes are observed in this lattice with the variation of the bias field orientation. Micromagnetic simulations enable us to successfully reproduce the measured evolution of frequencies with the orientation of bias magnetic field, as well as to identify the spatial profles of the modes. The magnetostatic field analysis, suggest the existence of magnetostatic coupling between the dot and antidot in annular antidot sample. Further local excitations of some selective spin-wave modes using numerical simulations showed the anisotropic spin-wave propagation through the lattice., Comment: 9 pages, 7 figures, 8 pages of supplementary material

Betulinic acid, a plant secondary metabolite, has gained significant attention due to its antiproliferative activity over a range of cancer cells. A promising betulinic acid analogue (2c) with better therapeutic efficacy than parent molecule to colon carcinoma cells has been reported. Despite impressive biological applications, low aqueous solubility and bioavailability create difficulties for its therapeutic applications. To overcome these lacunae and make it as a promising drug candidate we have encapsulated the lead betulinic acid derivative (2c) in a polymeric nanocarrier system (2c-NP) and evaluated its in vitro and in vivo therapeutic efficacy. Apoptosis that induces in vitro antiproliferative activity was significantly increased by 2c-NP compared to free-drug (2c), as assured by MTT assay, Annexin V positivity, JC1 analysis and cell cycle study. The therapeutic potential measured in vitro and in vivo reflects ability of 2c-NP as an effective therapeutic agent for treatment of colon carcinoma and future translation to clinical trials.

In this article, we predict the emergence of non-trivial band topology in the family of XX′Bi compounds having $$P\overline{6}2m$$ P 6 ¯ 2 m (# 189) space group. Using first principles calculations within hybrid functional framework, we demonstrate that NaSrBi and NaCaBi are strong topological insulator under controlled band engineering. Here, we propose three different ways to engineer the band topology to get a non-trivial order: (i) hydrostatic pressure, (ii) biaxial strain (due to epitaxial mismatch), and (iii) doping. Non-triviality is confirmed by investigating bulk band inversion, topological Z2 invariant, surface dispersion and spin texture. Interestingly, some of these compounds also show a three dimensional topological nodal line semi-metal (NLS) state in the absence of spin orbit coupling (SOC). In these NLS phases, the closed loop of band degeneracy in the Brillouin zone lie close to the Fermi level. Moreover, a drumhead like flat surface state is observed on projecting the bulk state on the [001] surface. The inclusion of SOC opens up a small band gap making them behave like a topological insulator.

Assessment of morphodynamic groundwater reserves is important for the sustainable management of water resources. It is a truth that groundwater resource evaluation is anxious with the ambiguity of its several factors and employing methods. Thus, an information-based model has been hypothesized to assess natural groundwater reserves in a morphodynamic system in a part of the Ganga basin of Northern India, where the spatial variability in natural groundwater reserve exists. Marginal information of rainfall data, and transinformation among the rainfall, and monthly depth to groundwater level measurement at 50 wells in a dense monitoring network were used for evaluating natural groundwater reserve. The results indicate that an average recharge rate is about 246 mm/monsoon and or 32.65% of the seasonal rainfall, and its values are well-correlated with the soil infiltration rate. It has been found that the estimated recharge rates are about 54.08, 45.85, 33.77, 32.48, and 32.14% of the seasonal rainfall in an active flood plain, back swamp, natural levees, flood plain, and palaeochannel, respectively. The calculated annual rainfall input to groundwater reserve is found about 127.98 MCM/monsoon rainfall, which could be employed for sustainable management of groundwater resources in the morphodynamic system of the Ganga river basin.

Studies related to loading ability and delivery of clinically used first-line anti-tuberculosis drugs (ATDs) such as isoniazid, rifampicin, pyrazinamide and streptomycin on the surface of starch-derived bulk and nanopolyurethanes (SBPUs and SNPUs) as drug delivery systems (DDS) have been focused to minimise or remove the drug-associated adverse effects. The efficiencies of nanopolyurethanes obtained from the differently substituted cyclic aliphatic and aromatic isocyanates have been studied for drug loading and release purposes. Different advanced instrumental techniques analysed the structural and morphological properties, thermal stability and crystallinity of the starch nanopolyurethans. Average particle sizes ranging from 27.35–42.38 nm to 126.89–218.60 nm for starch nanopolyurethans, SNPU3i and SNPU4i, respectively, were determined by high-resolution transmission electron microscopy. Similarly, the loading efficiency of ATDs to the surfaces of SNPUs and SBPUs was observed in the range of 60–97% while ATDs-loaded SNPUs showed a sustainable release profile for all ATDs except for streptomycin. However, most SBPUs provided burst-release for all the above-mentioned ATDs in pH-dependent studies. The anti-tuberculosis assay against the Mycobacterium tuberculosis H37Rv strain revealed that streptomycin-loaded SNPU4i and isoniazid-loaded SNPU7i are approximately 42 and 7 times more active than the native streptomycin and isoniazid, respectively.

Wheat grain yield (GY) improvement using genomic tools is important for achieving yield breakthroughs. To dissect the genetic architecture of wheat GY potential and stress-resilience, we have designed this large-scale genome-wide association study using 100 datasets, comprising 105,000 GY observations from 55,568 wheat lines evaluated between 2003 and 2019 by the International Maize and Wheat Improvement Center and national partners. We report 801 GY-associated genotyping-by-sequencing markers significant in more than one dataset and the highest number of them were on chromosomes 2A, 6B, 6A, 5B, 1B and 7B. We then used the linkage disequilibrium (LD) between the consistently significant markers to designate 214 GY-associated LD-blocks and observed that 84.5% of the 58 GY-associated LD-blocks in severe-drought, 100% of the 48 GY-associated LD-blocks in early-heat and 85.9% of the 71 GY-associated LD-blocks in late-heat, overlapped with the GY-associated LD-blocks in the irrigated-bed planting environment, substantiating that simultaneous improvement for GY potential and stress-resilience is feasible. Furthermore, we generated the GY-associated marker profiles and analyzed the GY favorable allele frequencies for a large panel of 73,142 wheat lines, resulting in 44.5 million datapoints. Overall, the extensive resources presented in this study provide great opportunities to accelerate breeding for high-yielding and stress-resilient wheat varieties.

Accurate prognostic biomarkers in early-stage melanoma are urgently needed to stratify patients for clinical trials of adjuvant therapy. We applied a previously developed open source deep learning algorithm to detect tumor-infiltrating lymphocytes (TILs) in hematoxylin and eosin (H&E) images of early-stage melanomas. We tested whether automated digital (TIL) analysis (ADTA) improved accuracy of prediction of disease specific survival (DSS) based on current pathology standards. ADTA was applied to a training cohort (n = 80) and a cutoff value was defined based on a Receiver Operating Curve. ADTA was then applied to a validation cohort (n = 145) and the previously determined cutoff value was used to stratify high and low risk patients, as demonstrated by Kaplan–Meier analysis (p ≤ 0.001). Multivariable Cox proportional hazards analysis was performed using ADTA, depth, and ulceration as co-variables and showed that ADTA contributed to DSS prediction (HR: 4.18, CI 1.51–11.58, p = 0.006). ADTA provides an effective and attainable assessment of TILs and should be further evaluated in larger studies for inclusion in staging algorithms.

A series of stable and ready-to-use bioinks have been developed based on the xeno-free and tunable hydrogel (VitroGel) system. Cell laden scaffold fabrication with optimized polysaccharide-based inks demonstrated that Ink H4 and RGD modified Ink H4-RGD had excellent rheological properties. Both bioinks were printable with 25–40 kPa extrusion pressure, showed 90% cell viability, shear-thinning and rapid shear recovery properties making them feasible for extrusion bioprinting without UV curing or temperature adjustment. Ink H4-RGD showed printability between 20 and 37 °C and the scaffolds remained stable for 15 days at temperature of 37 °C. 3D printed non-small-cell lung cancer (NSCLC) patient derived xenograft cells (PDCs) showed rapid spheroid growth of size around 500 µm in diameter and tumor microenvironment formation within 7 days. IC50 values demonstrated higher resistance of 3D spheroids to docetaxel (DTX), doxorubicin (DOX) and erlotinib compared to 2D monolayers of NSCLC-PDX, wild type triple negative breast cancer (MDA-MB-231 WT) and lung adenocarcinoma (HCC-827) cells. Results of flow property, shape fidelity, scaffold stability and biocompatibility of H4-RGD suggest that this hydrogel could be considered for 3D cell bioprinting and also for in-vitro tumor microenvironment development for high throughput screening of various anti-cancer drugs.

Schistosoma spindale and Schistosoma indicum are ruminant-infecting trematodes of the Schistosoma indicum group that are widespread across Southeast Asia. Though neglected, these parasites can cause major pathology and mortality to livestock leading to significant welfare and socio-economic issues, predominantly amongst poor subsistence farmers and their families. Here we used mitogenomic analysis to determine the relationships between these two sympatric species of schistosome and to characterise S. spindale diversity in order to identify possible cryptic speciation. The mitochondrial genomes of S. spindale and S. indicum were assembled and genetic analyses revealed high levels of diversity within the S. indicum group. Evidence of functional changes in mitochondrial genes indicated adaptation to environmental change associated with speciation events in S. spindale around 2.5 million years ago. We discuss our results in terms of their theoretical and applied implications.

Based upon the morphological characteristics, tea is classified botanically into 2 main types i.e. Assam and China, which are morphologically very distinct. Further, they are so easily pollinated among themselves, that a third category, Cambod type is also described. Although the general consensus of origin of tea is India, Burma and China adjoining area, yet specific origin of China and Assam type tea are not yet clear. Thus, we made an attempt to understand the origin of Indian tea through the comparative analysis of different chloroplast (cp) genomes under the Camellia genus by performing evolutionary study and comparing simple sequence repeats (SSRs) and codon usage distribution patterns among them. The Cp genome based phylogenetic analysis indicated that Indian Tea, TV1 formed a different group from that of China tea, indicating that TV1 might have undergone different domestications and hence owe different origins. The simple sequence repeats (SSRs) analysis and codon usage distribution patterns also supported the clustering order in the cp genome based phylogenetic tree.
