Under natural conditions in Spain, the fruiting bodies of Pleurotus eryngii var. eryngii, a basidomycete of great economic and gastronomic importance, appears at the base of the stem of (mainly) Eryngium campestre, surrounded by the plant’s dried-out basal leaves. Traditionally collected in the wild of Mediterranean areas, this fungus is now cultivated all over the world for its culinary and even medicinal properties. However, controversy exists regarding its lifestyle. Some authors indicate it is a saprotroph, while others suggest it is a weak or even virulent parasite that causes male sterility in E. campestre. The present work aims to clarify whether P. eryngii behaves as a saprotroph, a weak or virulent parasite to E. campestre. The soil beside the roots of naturally growing E. campestre plants was inoculated with a commercial strain of P. eryngii, and the trophic mode of the fungus recorded. Soil and root samples were subjected to molecular analysis to confirm the absence/presence of the fungus. The death of the plants and the molecular analysis showed P. eryngii is a facultative necrotrophic parasite of E. campestre in natural conditions and a virulent parasite in greenhouse.

This experiment focused on whether preconditioning of leaf litter by microfungi that were confined to one leaf (Unit-Restricted) made leaf litter less likely to be colonized and decomposed by basidiomycetes that bind litter into mats (Non-Unit-Restricted) than non-preconditioned litter. Leaves of Manilkara bidentata in litterbags were preconditioned by incubating them for 0, 1, 2 or 3 months in flat litter/seed rain baskets 10 cm above the forest floor to avoid colonization by basidiomycete fungi. Preconditioned and non-preconditioned leaves were transferred to 5 replicate basidiomycete fungal mats of Gymnopus johnstonii for 6 weeks. Both attachment by basidiomycete fungi and percent mass loss after 6 weeks decreased significantly with increasing preconditioning time. In non-preconditioned leaves, gamma irradiation did not affect mass loss or percent white-rot despite having significantly increased numbers of basidiomycete fungal connections as compared to non-irradiated leaves. In non-preconditioned leaves, more basidiomycetes attachmented to non-irradiated than irradiated leaves suggest facilitation by phyllosphere microfungi. While basidiomycete colonization was initially facilitated by phyllosphere fungi, we inferred that degradation of resource quality led to fewer fungal attachments and less mass loss after 1-3 months of preconditioning by microfungi. The date suggest there is a 1-month time window for basidiomycete fungi to incorporate fallen leaves into their litter mats.

Carbon storage and cycling in boreal forests—the largest terrestrial carbon store—is moderated by complex interactions between trees and soil microorganisms. However, existing methods limit our ability to predict how changes in environmental conditions will alter these associations and the essential ecosystem services they provide. To address this, we developed a metatranscriptomic approach to analyze the impact of nutrient enrichment on Norway spruce fine roots and the community structure, function, and tree–microbe coordination of over 350 root-associated fungal species. In response to altered nutrient status, host trees redefined their relationship with the fungal community by reducing sugar efflux carriers and enhancing defense processes. This resulted in a profound restructuring of the fungal community and a collapse in functional coordination between the tree and the dominant Basidiomycete species, and an increase in functional coordination with versatile Ascomycete species. As such, there was a functional shift in community dominance from Basidiomycetes species, with important roles in enzymatically cycling recalcitrant carbon, to Ascomycete species that have melanized cell walls that are highly resistant to degradation. These changes were accompanied by prominent shifts in transcriptional coordination between over 60 predicted fungal effectors, with more than 5,000 Norway spruce transcripts, providing mechanistic insight into the complex molecular dialogue coordinating host trees and their fungal partners. The host–microbe dynamics captured by this study functionally inform how these complex and sensitive biological relationships may mediate the carbon storage potential of boreal soils under changing nutrient conditions.

The present study reports a sudden fungal outbreak that occurred in the corridor near the entrance of the Catacombs of SS. Marcellino and Pietro in Rome (Italy) observed after 1 year of a restoration treatment that interested the walls of the entrance of the Catacombs and some artifacts placed in situ. The colonization was observed on the vault at the entrance and in correspondence with the restored marble pieces displayed on the left side of the corridor. No growth was observed on the right side where similarly treated marble slabs were placed. Samples taken in correspondence with fungal biofilm were analyzed through the combined use of microscopical, cultural, and molecular tools and showed that the vault and the left side of the corridor entrance were colonized by a complex fungal biofilm consisting mainly of Coniophora sp. and other genera, such as Hypomyces, Purpureocillium, Acremonium, Penicillium, and Alternaria, many of which are well known as responsible of biodeterioration of stone surfaces. Regarding the brown-rot basidiomycete Coniophora, it was able to form very large colonies on the substrata with a diameter of up to 57 cm. Although the direct observation under a light microscope evidenced the presence of abundant brown fungal conidia, several attempts to cultivate the microorganism failed, therefore only through DNA sequencing analyses, it was possible to identify and characterize this fungus. There is very little literature on the genus Coniophora which is reported as one of the causes of wet-rot decay of wood in buildings. A connection with calcium-containing materials such as bricks and mortars was demonstrated, but no data were available about the possible role of this species in the biodeterioration of stones. This study features the first finding of a strain related to the basidiomycetous genus of Coniophora in the order Boletales in association with evident phenomena of biodeterioration.

Ganoderma lucidum is a medicinal fungus that has a worldwide demand due to its numerous bioactive components such as polyphenols, triterpenes, and polysaccharides, which have antioxidant potentials. This study assessed the total polyphenol content through the Folin-Ciocalteu method and analyzed structural functional groups using Fourier-transmform infrared spectroscopy (FTIR). Ethanolic and aqueous solutions were extracted from fruiting bodies of G. lucidum growing in lignocellulosic substrates of sawdust and straw obtained from Pandala talaumanariñensis and Avena sativa, respectively. The total polyphenol content was higher in the aqueous solutions (32,44 ± 0,7023 mg GAE/g), which would indicate that other bioactive compounds of G. lucidum can be extracted with hot water, enriching this solution. The IR spectra of G. lucidum showed absorption bands between the 3372,59 to 670,27 cm-1 range, which were similar to IR spectral information of the genus Ganoderma. These results demonstrate an interesting potential of G. lucidum as a source of natural antioxidants that can be used for the food and medicinal industry, given its total polyphenol content., Ganoderma lucidum es un hongo medicinal con gran demanda en el mercado mundial, reconocido por poseer numerosos compuestos bioactivos con potencial antioxidante, principalmente por sus polifenoles, triterpenos y polisacáridos. En el presente estudio se determinó el contenido de polifenoles totales mediante el método de Folin-Ciocalteu y la evaluación de grupos funcionales estructurales mediante espectroscopía infrarroja por transformada de Fourier (FTIR) en extractos etanólicos y acuosos del cuerpo fructífero de G. lucidum obtenidos del cultivo en sustrato lignocelulósico de aserrín de Pandala talaumanariñensis y paja de Avena sativa. El contenido de polifenoles totales fue mayor en los extractos acuosos (32,44 ± 0,7023 mg EAG/g) lo que indicaría que otros compuestos bioactivos de G. lucidum con actividad antioxidante podrían ser arrastrados en el agua caliente, enriqueciendo el extracto. Los espectros IR de G. lucidum presentaron bandas de absorción en el rango de 3372,59 a 670,27 cm-1, las cuales resultaron comparables con información espectral IR de productos comerciales del género Ganoderma. Los resultados encontrados muestran un interesante potencial de G. lucidum como fuente de antioxidantes naturales que pueden usarse en la industria alimentaria y medicinal, dado su contenido de polifenoles totales.

Low-water soluble metal fluorides such as magnesium fluoride (MgF2) and calcium fluoride (CaF2) were evaluated for decay protection of wood. Initially, the biocidal efficacy of nano metal fluorides (NMFs) against wood destroying fungi was assessed with an in-vitro agar test. The results from the test showed that agar medium containing MgF2 and CaF2 was more efficient in preventing fungal decay than stand-alone MgF2 or CaF2. These metal fluorides, in their nanoscopic form synthesized using fluorolytic sol-gel synthesis, were introduced into the sapwood of Scots pine and beech wood and then subjected to accelerated ageing by leaching (EN 84). MAS 19F NMR and X-ray micro CT images showed that metal fluorides were present in treated wood, unleached and leached. Decay resistance of Scots pine and beech wood treated with NMFs was tested against wood destroying fungi Rhodonia placenta and Trametes versicolor in accordance with EN 113. Results revealed that mass losses were reduced to below 3% in wood treated with the combination of MgF2 and CaF2. Wood-decaying fungi tend to have characteristic substrate ranges that partly define their ecological niche. Fomitopsis pinicola is a brown rot species of Polyporales that is reported on 82 species of softwoods and 42 species of hardwoods. We analyzed gene expression levels of F. pinicola from submerged cultures with ground wood powder (sampled at 5 days) or solid wood wafers (sampled at 10 and 30 days), using aspen, pine, and spruce substrates (aspen was used only in submerged cultures). Fomitopsis pinicola expressed similar sets of wood-degrading enzymes typical of brown rot fungi across all culture conditions and time points. Nevertheless, differential gene expression was observed across all pairwise comparisons of substrates and time points. Genes exhibiting differential expression encode diverse enzymes with known or potential function in brown rot decay, including laccase, benzoquinone reductase, aryl alcohol oxidase, cytochrome P450s, and various glycoside hydrolases. Comparing transcriptomes from submerged cultures and wood wafers, we found that culture conditions had a greater impact on global expression profiles than substrate wood species. These findings highlight the need for standardization of culture conditions in studies of gene expression in wood-decaying fungi. IMPORTANCE All species of wood-decaying fungi occur on a characteristic range of substrates (host plants), which may be broad or narrow. Understanding the mechanisms that allow fungi to grow on particular substrates is important for both fungal ecology and applied uses of different feedstocks in industrial processes. We grew the wood-decaying polypore Fomitopsis pinicola on three different wood species—aspen, pine, and spruce—under various culture conditions. We found that F. pinicola is able to modify gene expression (transcription levels) across different substrate species and culture conditions. Traditional smoking generates not only the impact flavor compound 4-vinylguaiacol, but concurrently many unwanted and potent toxic compounds such as polycyclic aromatic hydrocarbons. Enzyme technology provides a solution without any side-product formation. A feruloyl esterase from Rhizoctonia solani (RspCAE) liberated ferulic acid from low-priced sugar beet fiber. Decarboxylation of ferulic acid to 4-vinylguaiacol was achieved by a second enzyme from Schizophyllum commune (ScoFAD). Both enzymes were covalently immobilized on agarose to enable reusability in a fixed-bed approach. The two enzyme cascades showed high conversion rates with yields of 0.8 and 0.95, respectively, and retained activity for nearly 80 h of continuous operation. The overall productivity of the model process with bed volumes of 300 µL and a substrate flow rate of 0.25 mL min−1 was 3.98 mg 4-vinylguaiacol per hour. A cold online solid phase extraction using XAD4 was integrated into the bioprocess and provided high recovery rates during multiple elution steps. Attempting to facilitate the bioprocess, a fused gene coding for the two enzymes and a set of different linker lengths and properties was constructed and introduced into Komagataella phaffii. Longer and rigid linkers resulted in higher activity of the fusion protein with a maximum of 67 U L−1

Invasive mold infections caused by molds other than Aspergillus spp. or Mucorales are emerging. The reported prevalences of infection due to these rare fungal pathogens vary among geographic regions, driven by differences in climatic conditions, susceptible hosts, and diagnostic capabilities. These rare molds-Fusarium, Lomentospora, and Scedosporium species and others-are difficult to detect and often show intrinsic antifungal resistance. Now, international societies of medical mycology and microbiology have joined forces and created the "Global guideline for the diagnosis and management of rare mould infections: an initiative of the European Confederation of Medical Mycology in cooperation with the International Society for Human and Animal Mycology and the American Society for Microbiology" (published in Lancet Infect Dis, https://doi.org/10.1016/S1473-3099(20)30784-2), with the goal of improving the diagnosis, treatment, prevention, and survival of persons with rare mold infections. The guideline provides cutting-edge guidance for the correct utilization and application of established and new diagnostic and therapeutic options.

The ancestor of termites relied on gut symbionts for degradation of plant material, an association that persists in all termite families.1,2 However, the single-lineage Macrotermitinae has additionally acquired a fungal symbiont that complements digestion of food outside the termite gut.3 Phylogenetic analysis has shown that fungi grown by these termites form a clade—the genus Termitomyces—but the events leading toward domestication remain unclear.4 To address this, we reconstructed the lifestyle of the common ancestor of Termitomyces using a combination of ecological data with a phylogenomic analysis of 21 related non-domesticated species and 25 species of Termitomyces. We show that the closely related genera Blastosporella and Arthromyces also contain insect-associated species. Furthermore, the genus Arthromyces produces asexual spores on the mycelium, which may facilitate insect dispersal when growing on aggregated subterranean fecal pellets of a plant-feeding insect. The sister-group relationship between Arthromyces and Termitomyces implies that insect association and asexual sporulation, present in both genera, preceded the domestication of Termitomyces and did not follow domestication as has been proposed previously. Specialization of the common ancestor of these two genera on an insect-fecal substrate is further supported by similar carbohydrate-degrading profiles between Arthromyces and Termitomyces. We describe a set of traits that may have predisposed the ancestor of Termitomyces toward domestication, with each trait found scattered in related taxa outside of the termite-domesticated clade. This pattern indicates that the origin of the termite-fungus symbiosis may not have required large-scale changes of the fungal partner.

Standing dead trees (snags) decompose more slowly than downed dead wood and provide critical habitat for many species. The rate at which snags fall therefore influences forest carbon dynamics and biodiversity. Fall rates correlate strongly with mean annual temperature, presumably because warmer climates facilitate faster wood decomposition and hence degradation of the structural stability of standing wood. These faster decomposition rates coincide with turnover from fungal-dominated wood decomposer communities in cooler forests to codomination by fungi and termites in warmer regions. A key question for projecting forest dynamics is therefore whether temperature effects on wood decomposition arise primarily because warmer conditions facilitate faster decomposer metabolism, or are also influenced indirectly by belowground community turnover (e.g., termites exert additional influence beyond fungal-plus-bacterial mediated decomposition). To test between these possibilities, we simulate standing dead trees with untreated wooden posts and follow them in the field across 5 yr at 12 sites, before measuring buried, soil–air interface and aerial post sections to quantify wood decomposition and organism activities. High termite activities at the warmer sites are associated with rates of postfall that are three times higher than at the cooler sites. Termites primarily consume buried wood, with decomposition rates greatest where termite activities are highest. However, where higher microbial and termite activities co-occur, they appear to compensate for one another first, and then to slow decomposition rates at their highest activities, suggestive of interference competition. If the range of microbial and termite codomination of wood decomposer communities expands under climate warming, our data suggest that expansion will accelerate snag fall with consequent effects on forest carbon cycling and biodiversity in forests previously dominated by microbial decomposers.

The lichenicolous 'heterobasidiomycetes' belong in the Tremellomycetes (Agaricomycotina) and in the Pucciniomycotina. In this paper, we provide an introduction and review of these lichenicolous taxa, focusing on recent studies and novelties of their classification, phylogeny and evolution. Lichen-inhabiting fungi in the Pucciniomycotina are represented by only a small number of species included in the genera Chionosphaera, Cyphobasidium and Lichenozyma. The phylogenetic position of the lichenicolous representatives of Chionosphaera has, however, never been investigated by molecular methods. Phylogenetic analyses using the nuclear SSU, ITS, and LSU ribosomal DNA markers reveal that the lichenicolous members of Chionosphaera form a monophyletic group in the Pucciniomycotina, distinct from Chionosphaera and outside the Chionosphaeraceae. The new genus Crittendenia is described to accommodate these lichen-inhabiting species. Crittendenia is characterized by minute synnemata-like basidiomata, the presence of clamp connections and aseptate tubular basidia from which 4-7 spores discharge passively, often in groups. Crittendenia, Cyphobasidium and Lichenozyma are the only lichenicolous lineages known so far in the Pucciniomycotina, whereas Chionosphaera does not include any lichenicolous taxa.

Fungal laccases are multicopper oxidases with high catalytic versatility and low catalytic requirements (only O2 of the air is required for activation). Their high redox potential, especially of certain basidiomycete laccases, significantly increases their oxidation capacity compared to bacterial laccases. These characteristics together provide these enzymes with great potential for applications as biocatalysts in a range of synthetic or degrading reactions. We review here some of the main properties and biotechnological applications of fungal laccases, underlining their overexpression limitations for industrial scale application. We focus on the use of laccases as biocatalysts for organic synthesis, with special emphasis on polymer synthesis.

The ancestor of termites relied on gut symbionts for degradation of plant material, an association that persists in all termite families.1,2 However, the single-lineage Macrotermitinae has additionally acquired a fungal symbiont that complements digestion of food outside the termite gut.3 Phylogenetic analysis has shown that fungi grown by these termites form a clade—the genus Termitomyces—but the events leading toward domestication remain unclear.4 To address this, we reconstructed the lifestyle of the common ancestor of Termitomyces using a combination of ecological data with a phylogenomic analysis of 21 related non-domesticated species and 25 species of Termitomyces. We show that the closely related genera Blastosporella and Arthromyces also contain insect-associated species. Furthermore, the genus Arthromyces produces asexual spores on the mycelium, which may facilitate insect dispersal when growing on aggregated subterranean fecal pellets of a plant-feeding insect. The sister-group relationship between Arthromyces and Termitomyces implies that insect association and asexual sporulation, present in both genera, preceded the domestication of Termitomyces and did not follow domestication as has been proposed previously. Specialization of the common ancestor of these two genera on an insect-fecal substrate is further supported by similar carbohydrate-degrading profiles between Arthromyces and Termitomyces. We describe a set of traits that may have predisposed the ancestor of Termitomyces toward domestication, with each trait found scattered in related taxa outside of the termite-domesticated clade. This pattern indicates that the origin of the termite-fungus symbiosis may not have required large-scale changes of the fungal partner.

Lichen-inhabiting fungi in the Pucciniomycotina are represented by only a small number of species included in the genera Chionosphaera, Cyphobasidium and Lichenozyma. The phylogenetic position of the lichenicolous representatives of Chionosphaera has, however, never been investigated by molecular methods. Phylogenetic analyses using the nuclear SSU, ITS, and LSU ribosomal DNA markers reveal that the lichenicolous members of Chionosphaera form a monophyletic group in the Pucciniomycotina, distinct from Chionosphaera and outside the Chionosphaeraceae. Invasive mold infections caused by molds other than Aspergillus spp. or Mucorales are emerging. The reported prevalences of infection due to these rare fungal pathogens vary among geographic regions, driven by differences in climatic conditions, susceptible hosts, and diagnostic capabilities. These rare molds-Fusarium, Lomentospora, and Scedosporium species and others-are difficult to detect and often show intrinsic antifungal resistance. Now, international societies of medical mycology and microbiology have joined forces and created the "Global guideline for the diagnosis and management of rare mould infections: an initiative of the European Confederation of Medical Mycology in cooperation with the International Society for Human and Animal Mycology and the American Society for Microbiology" (published in Lancet Infect Dis, https://doi.org/10.1016/S1473-3099(20)30784-2), with the goal of improving the diagnosis, treatment, prevention, and survival of persons with rare mold infections. The guideline provides cutting-edge guidance for the correct utilization and application of established and new diagnostic and therapeutic options.

Standing dead trees (snags) decompose more slowly than downed dead wood and provide critical habitat for many species. The rate at which snags fall therefore influences forest carbon dynamics and biodiversity. Fall rates correlate strongly with mean annual temperature, presumably because warmer climates facilitate faster wood decomposition and hence degradation of the structural stability of standing wood. These faster decomposition rates coincide with turnover from fungal-dominated wood decomposer communities in cooler forests to codomination by fungi and termites in warmer regions. A key question for projecting forest dynamics is therefore whether temperature effects on wood decomposition arise primarily because warmer conditions facilitate faster decomposer metabolism, or are also influenced indirectly by belowground community turnover (e.g., termites exert additional influence beyond fungal-plus-bacterial mediated decomposition). To test between these possibilities, we simulate standing dead trees with untreated wooden posts and follow them in the field across 5 yr at 12 sites, before measuring buried, soil–air interface and aerial post sections to quantify wood decomposition and organism activities. High termite activities at the warmer sites are associated with rates of postfall that are three times higher than at the cooler sites. Termites primarily consume buried wood, with decomposition rates greatest where termite activities are highest. However, where higher microbial and termite activities co-occur, they appear to compensate for one another first, and then to slow decomposition rates at their highest activities, suggestive of interference competition. If the range of microbial and termite codomination of wood decomposer communities expands under climate warming, our data suggest that expansion will accelerate snag fall with consequent effects on forest carbon cycling and biodiversity in forests previously dominated by microbial decomposers.

Stripe rust of wheat, caused by the obligate biotrophic fungus Puccinia striiformis f.sp. tritici, is a major threat to wheat production worldwide with an estimated yearly loss of US $1 billion. The recent advances in long-read sequencing technologies and tailored-assembly algorithms enabled us to disentangle the two haploid genomes of Pst. This provides us with haplotype-specific information at a whole-genome level. Exploiting this novel information, we perform whole-genome comparative genomics of two P. striiformis f.sp. tritici isolates with contrasting life histories. We compare one isolate of the old European lineage (PstS0), which has been asexual for over 50 years, and a Warrior isolate (PstS7 lineage) from a novel incursion into Europe in 2011 from a sexual population in the Himalayan region. This comparison provides evidence that long-term asexual evolution leads to genome expansion, accumulation of transposable elements, and increased heterozygosity at the single nucleotide, structural, and allele levels. At the whole-genome level, candidate effectors are not compartmentalized and do not exhibit reduced levels of synteny. Yet we were able to identify two subsets of candidate effector populations. About 70% of candidate effectors are invariant between the two isolates, whereas 30% are hypervariable. The latter might be involved in host adaptation on wheat and explain the different phenotypes of the two isolates. Overall, this detailed comparative analysis of two haplotype-aware assemblies of P. striiformis f.sp. tritici is the first step in understanding the evolution of dikaryotic rust fungi at a whole-genome level.

Jamur merupakan salah satu produk hasil hutan non kayu yang paling disukai karena dapat dicari dengan mudah. Tujuan penelitian ini adalah untuk mengetahui jenis-jenis jamur Basidiomycetes serta untuk mengetahui jenis jamur pangan dan jamur obat yang ada di kawasan penelitian Punggualas Taman Nasional Sebangau. Metode penelitian yang digunakan adalah dengan metode eksplorasi dan identifikasi dengan membuat plot  ukuran 150 m x 100 m dengan jalur pengamatan jamur sebanyak 5 jalur, jarak antar jalur adalah 5 m dan lebar jalur 20 m. Hasil penelitian ditemukan 15 famili jamur Basidiomycetes,  49 jenis jamur dan jumlah individu jamur sebanyak 789 buah. Berdasarkan jumlah individu maka jamur Aucularia auricula merupakan jamur yang dominan tumbuh di Taman Nasional Sebangau. Indeks keanekaragaman (H´)=2,6260 tergolong sedang. Indeks kekayaan R=7,1956 tergolong tinggi dan indeks kemerataan E1=0,6747 tergolong sedang. Jamur Auricularia auricula (jamur kuping/kulat bitak) dan Pleurotus sp. Method(s): Retrospective data over 10 years from four tertiary pediatric oncology and hematopoietic stem cell transplant (HSCT) units across Australia were analyzed to report demographic, clinical, and mycological characteristics of IFI episodes, and crude IFI prevalence in select oncology/HSCT groups. Kaplan-Meier survival analyses were used to calculate 180-day overall survival. Result(s): A total of 337 IFI episodes occurred in 320 children, of which 149 (44.2%), 51 (15.1%), and 110 (32.6%) met a modified European Organization for Research and Treatment of Cancer (mEORTC) criteria for proven, probable, and possible IFI, respectively. There were a further 27 (8.0%) that met a "modified possible IFI" criteria. Median age at IFI diagnosis was 8.4 years. Crude mEORTC IFI prevalence in acute lymphoblastic leukemia, acute myeloid leukemia, solid tumor, and allogeneic HSCT cohorts was 10.6%, 28.2%, 4.4%, and 11.7%, respectively. Non-Aspergillus species represented 48/102 (47.1%) molds identified, and non-albicans Candida represented 66/93 (71.0%) yeasts identified. There were 56 deaths among 297 children who met mEORTC criteria, with 180-day overall survival for proven, probable, and possible IFIs of 79.7%, 76.2%, and 84.4%, respectively. Conclusion(s): Non-Aspergillus molds and non-albicans Candida contributed substantially to pediatric IFI in our study, with high IFI prevalence in leukemia and allogeneic HSCT cohorts. Seventeen extracts and seven secondary metabolites isolated from basidiomycetes were tested in medium culture against promastigote forms of Leishmania spp. and bloodstream forms of Trypanosoma cruzi. Extracts from the culture filtrate or mycelium were generally inactive against the parasites except the Zucoagaricus genus mycelium extract which reduced by 47% the number of bloodstream forms. Striatin A, striatin B and podoscyphic acid exhibited in vitro activity at 10, 5 and 100 μg/mL, respectively. One compound showed activity against bloodstream forms of T. cruzi, the sesquiterpenoid naematolin, lysing the parasites by 79%. BALB/c mice infected with L. amazonensis were treated 3 weeks post-infection with striatin A and striatin B by subcutaneous route for 15 days at 10 mg/kg daily. The reference drug, N-methylglucamine antimonate, administered by subcutaneous injections at 28 mg Sbv/kg/day for 15 days reduced the parasite burden by 71.2% (p <0.05). Subcutaneous administration of straitin A at 10 mg/kg produced a weak decrease of the parasite burdens in the footpad by 17.6%. The treatment with striatin B had no effect and showed higher toxicity than striatin A.

Lentinus tigrinus is a species of wood-decaying fungi (Polyporales) that has an agaricoid form (a gilled mushroom) and a secotioid form (puffball-like, with enclosed spore-bearing structures). Previous studies suggested that the secotioid form is conferred by a recessive allele of a single locus. We sequenced the genomes of one agaricoid (Aga) strain and one secotioid (Sec) strain (39.53-39.88 Mb, with 15,581-15,380 genes, respectively). We mated the Sec and Aga monokaryons, genotyped the progeny, and performed bulked segregant analysis (BSA). We also fruited three Sec/Sec and three Aga/Aga dikaryons, and sampled transcriptomes at four developmental stages. Using BSA, we identified 105 top candidate genes with nonsynonymous SNPs that cosegregate with fruiting body phenotype. Transcriptome analyses of Sec/Sec versus Aga/Aga dikaryons identified 907 differentially expressed genes (DEGs) along four developmental stages. On the basis of BSA and DEGs, the top 25 candidate genes related to fruiting body development span 1.5 Mb (4% of the genome), possibly on a single chromosome, although the precise locus that controls the secotioid phenotype is unresolved. The top candidates include genes encoding a cytochrome P450 and an ATP-dependent RNA helicase, which may play a role in development, based on studies in other fungi.

A Citizen Science initiative by the Westerdijk Fungal Biodiversity Institute and the Utrecht University Museum gave rise to a project where fungal and yeast isolates were obtained and identified from Dutch soil samples. During the current study, 386 yeast strains were isolated from 157 different locations in the Netherlands. These strains were identified using sequence data of the large-subunit rRNA gene (D1/D2 region) and the internal transcribed spacer 1 and 2 regions. A total of 53 different yeast species were found as well as 15 potentially novel species. Six novel ascomycetous species are described during this study that include Hanseniaspora mollemarum sp. nov., Ogataea degrootiae sp. nov., Pichia gijzeniarum sp. nov., Saccharomycopsis oosterbeekiorum sp. nov., Trichomonascus vanleenenius sp. nov. and Zygoascus flipseniorum sp. nov. This study made it possible to incorporate numerous yeast isolates into the CBS collection without any restrictions, which make these isolates readily available for use by others. Many of the isolates represented species of which only a few isolates or even only a single ex-type strain were available. Therefore, it is a clear indication that such biodiversity-orientated Citizen Science projects can enrich the pool of available yeasts for future research projects.

Ganoderma lucidum, a renowned fungus for its medicinal properties and content of bioactive compounds include polysaccharides, triterpenoids, immune modulatory proteins, among others; it has generated a significant increase and interest in its production. Most species of fungi respond and adapt to various environmental signal including light, which affects it not only in productivity, but also in quality and quantity to be closely related to the fruiting bodies. Therefore, the objective of this study was evaluating the biological efficiency (EB) and the rate of production (TP) as production parameters of the solid culture of Ganoderma lucidum at under irradiation of the substrate with light emitting diodes blue (LED) with two levels of photo-stimulation constant 12 and 24 h during all steps of culture to induce mycelial growth and the formation of fruiting bodies. Conventional parameters were applied to stimulate the growth and development of the fungus in the stages of production. For the formulation of substrates agro-industrial waste and lignocellulosic materials were used. The diameter of the fruiting bodies under blue light treatments was higher than those exposed to white fluorescent light (control). The results show that the cultivation of Ganoderma lucidum with exposure to blue light is useful for induction of fruiting bodies of high quality, achieving a reduction of the period of fermentation in 16 days for the photo-stimulation of 24 h with EB of 28,04% and TP of 0,64., RESUMEN Ganoderma lucidum es un hongo macromiceto reconocido por sus propiedades medicinales y su contenido de compuestos bioactivos que incluyen polisacáridos, triterpenoides, proteínas inmunomoduladoras, entre otros, lo que ha generado un incremento notable en su producción. Por tal razón, el objetivo de este estudio fue evaluar la eficiencia biológica (EB) y la tasa de producción (TP) como parámetros de productividad del cultivo sólido de Ganoderma lucidum bajo irradiación de los sustratos con luz emitida por diodos azules (LED) con dos periodos de foto-estímulo de 12 y 24 h durante todas las fases de cultivo para inducir el crecimiento micelial y la formación de los cuerpos fructíferos. Se aplicaron parámetros convencionales para el crecimiento y desarrollo del hongo en las etapas de producción. Para la formulación de los sustratos, se emplearon residuos agroindustriales y materiales lignocelulósicos. El diámetro de los cuerpos fructíferos sometidos a tratamientos con luz azul fue mayor que los exhibidos a luz blanca fluorescente (Testigo). Los resultados muestran que el cultivo de Ganoderma lucidum con exposición a la luz azul es útil para la inducción de cuerpos fructíferos de alta calidad, logrando una disminución del periodo de fermentación en 16 días para el foto-estímulo de 24 h con EB de 28,04% y TP de 0,64.

Therefore, the objective of this study was evaluating the biological efficiency (EB) and the rate of production (TP) as production parameters of the solid culture of Ganoderma lucidum at under irradiation of the substrate with light emitting diodes blue (LED) with two levels of photo-stimulation constant 12 and 24 h during all steps of culture to induce mycelial growth and the formation of fruiting bodies. Conventional parameters were applied to stimulate the growth and development of the fungus in the stages of production. Wastewater is an environmental constraint for the development of the textile industry, since sometimes due to inappropriate management and/or ineffective treatment, it leads to contamination of biota. One of the main characteristics of textile wastes is the high amount of dye which, during the production process did not adhere to the fiber, thus appearing in the final waste. These wastes, when released into the aquatic environment reduce the availability of light, photosynthesis, as well as other environmental problems. Furthermore, these compounds are mostly toxic, even in small concentrations. Enzymes produced by Pleurotus sajor-caju are reported as efficient in color removal, dye degradation and detoxification of textile wastes. The objective of this study was to evaluate the ability of Pleurotus sajor-caju crude enzyme broth to remove color and reduce the toxicity of solutions containing Red 50 and Blue 56 dyes (100 mg L -1 ). For such, production of the enzyme broth was carried out in a bioreactor (working volume of 4 L), which was used directly on the dye solution, diluted in water (1:1) and diluted in ABTS (2,2’-azino-bis (3-ethylbenzthiazoline-6- sulfonic acid)) (1:1). Ecotoxicological tests were performed using the freshwater crustacean Daphnia magna. Higher efficiency in color removal by the crude enzymatic broth of P. sajor-caju diluted in ABTS solution was observed (removal rates equal to 1.3 and 1.9x10 -2 mg U -1 h -1 , for dyes Red 50 and Blue 56, respectively). The laccase enzyme was quantified in the enzyme broth (115 U L -1 ) and maintained its activity 100% throughout all assays. A statistically significant reduction in toxicity was observed for Blue 56 dye. Red 50 dye remained toxic even after the enzymatic treatment, suggesting the need for a longer reaction time for this dye. The P. sajor- caju crude enzyme broth proved to be effective and promising for use in textile wastewater treatment plants., As águas residuárias são normalmente um empecilho ambiental para o desenvolvimento da indústria têxtil em razão do gerenciamento inadequado e/ou tratamento ineficaz, que levam à contaminação da biota. Efluentes de indústrias têxteis têm como uma de suas principais características a alta carga de corantes, os quais durante o processo produtivo não se aderiram à fibra e, assim, encontram-se presentes no efluente final. Estes, quando lançados no ambiente aquático, reduzem a disponibilidade de luz e a realização da fotossíntese e ocasionam outros problemas ambientais. Além disso, a maioria dos corantes têxteis são tóxicos, mesmo em pequenas concentrações. Enzimas produzidas por Pleurotus sajor-caju são reportadas como eficientes na remoção de cor, degradação de corantes e detoxificação de efluentes têxteis. Assim, neste trabalho teve-se como objetivo avaliar a capacidade do caldo enzimático bruto de Pleurotus sajor-caju em remover a cor e, concomitantemente, diminuir a toxicidade de soluções que contêm os corantes têxteis Red 50 e Blue 56 (100 mg L-1). Para tal, produziu-se o caldo enzimático em biorreator (volume útil de 4 L), que foi adicionado diretamente sobre a solução de corantes sem diluição, diluído em água (1:1) e diluído em solução ABTS (2,2’-azino-bis(3-etilbenzotiazolina-6-sulfônico)) (1:1). O microcrustáceo Daphnia magna foi utilizado nos testes ecotoxicológicos. Zur Gewinnung von Biogas wird Biomasse in Biogasanlagen vergoren. So resultiert z.B. aus der Fermentation von Getreidestroh in der Biogasanlage der s.g. Strohgärrest“, der als fester Rückstand übrig bleibt. Da Strohgärreste hohe Gehalte an Lignin aufweisen, einem schwer abbaubaren Biopolymer, würde eine weitere anaerobe Fermentation trotz des vorhandenen Gärpotentials zu keiner wirtschaftlich nennenswerten Biogasausbeute-Steigerung führen. Ziel der Arbeit war es, den Ligninabbau mit Hilfe von ausgewählten Weißfäulepilzen in pflanzlichen Gärresten (mit einer Inkubationsdauer von höchstens 4 Wochen) zu maximieren und somit den Gärrest zur Weitervergärung aufzuschließen, um gesteigerte Biogasausbeuten zu erzielen. Da Biogas lediglich aus leicht fermentierbaren C-Quellen der pflanzlichen Substrate gebildet werden kann (Cellulose und Hemicellulose), galt es, geeignete Weißfäulepilze zu identifizieren, die Lignin möglichst selektiv und effizient abbauen, ohne selbst die Kohlenhydrate als Nahrungsquellen zu nutzen. Je mehr Lignin und je weniger Kohlenhydrate durch den Basidiomyceten während der Kultivierung in den Gärrestkulturen abgebaut werden, desto höher ist die erwartete Biogasausbeute. Der Vorteil dieser biologischen Nachbehandlung mit Pilzen im Vergleich zum physikalischen oder chemischen Aufschluss von Lignin ist der schonende und selektive Abbau aufgrund der Enzymspezifität. Dieser erlaubt es, extreme Temperaturen, extreme pH-Werte und energieintensive Zerkleinerungsverfahren zum Zweck des Ligninabbaus zu vermeiden. Es wurden in dieser Arbeit zunächst ausgewählte Weißfäulepilze auf Weizenstrohgärrest sowohl submers als auch emers kultiviert, um die vielversprechendsten Stämme zu identifizieren. As biogas is only formed from easily biodegradable carbon sources (cellulose and hemicellulose) of plant substrates, it was a main objective to identify white-rot fungi which are able to degrade lignin as selectively and efficiently as possible without using the carbohydrates as nutrient sources. The more lignin and the less carbohydrates are degraded by basidiomycetes during cultivation in the digestate cultures the higher is the expected biogas yield. The advantage of this biological post-treatment with fungi contrary to physical and chemical treatment methods of lignin degradation is the gentle and selective degradation of lignin due to the enzymatic specificity. In order to degrade lignin such a process makes it possible to avoid extreme temperatures, extreme pH values and energy-intensive mechanical grinding processes. In this work selected white-rot fungi were cultivated in presence of wheat straw digestate both in submerged and emerged cultivations to identify the most promising strains. In submerged cultivations several parameters were variated (pH value, cultivation duration, mass ratio of digestate and fungal inoculum etc.) in order to achieve maximal lignin degradation and thus maximal biogas yields as fast as possible. Production of basidiomycete atromentin-derived pigments like variegatic acid (pulvinic acid-type) and involutin (diarylcyclopentenone) from the brown-rotter Serpula lacrymans and the ectomycorrhiza-forming Paxillus involutus, respectively, is induced by complex nutrition, and in the case of S. lacrymans, bacteria. Pigmentation in S. lacrymans was stimulated by 13 different bacteria and cell-wall-damaging enzymes (lytic enzymes and proteases), but not by lysozyme or mechanical damage. The use of protease inhibitors with Bacillus subtilis or heat-killed bacteria during co-culturing with S. lacrymans significantly reduced pigmentation indicating that enzymatic hyphal damage and/or released peptides, rather than mechanical injury, was the major cause of systemic pigment induction. Conversely, no significant pigmentation by bacteria was observed from P. involutus. We found additional putative transcriptional composite elements of atromentin synthetase genes in P. involutus and other ectomycorrhiza-forming species that were absent from S. lacrymans and other brown-rotters. Variegatic and its precursor xerocomic acid, but not involutin, in return inhibited swarming and colony biofilm spreading of Bacillus subtilis, but did not kill B. subtilis. We suggest that dissimilar pigment regulation by fungal lifestyle was a consequence of pigment bioactivity and additional promoter motifs. The focus on basidiomycete natural product gene induction and regulation will assist in future studies to determine global regulators, signalling pathways and associated transcription factors of basidiomycetes.

A Citizen Science initiative by the Westerdijk Fungal Biodiversity Institute and the Utrecht University Museum gave rise to a project where fungal and yeast isolates were obtained and identified from Dutch soil samples. During the current study, 386 yeast strains were isolated from 157 different locations in the Netherlands. These strains were identified using sequence data of the large-subunit rRNA gene (D1/D2 region) and the internal transcribed spacer 1 and 2 regions. A total of 53 different yeast species were found as well as 15 potentially novel species. Six novel ascomycetous species are described during this study that include Hanseniaspora mollemarum sp. nov., Ogataea degrootiae sp. nov., Pichia gijzeniarum sp. nov., Saccharomycopsis oosterbeekiorum sp. nov., Trichomonascus vanleenenius sp. nov. and Zygoascus flipseniorum sp. nov. This study made it possible to incorporate numerous yeast isolates into the CBS collection without any restrictions, which make these isolates readily available for use by others. Many of the isolates represented species of which only a few isolates or even only a single ex-type strain were available. Therefore, it is a clear indication that such biodiversity-orientated Citizen Science projects can enrich the pool of available yeasts for future research projects.

Lentinus tigrinus is a species of wood-decaying fungi (Polyporales) that has an agaricoid form (a gilled mushroom) and a secotioid form (puffball-like, with enclosed spore-bearing structures). Previous studies suggested that the secotioid form is conferred by a recessive allele of a single locus. We sequenced the genomes of one agaricoid (Aga) strain and one secotioid (Sec) strain (39.53-39.88 Mb, with 15,581-15,380 genes, respectively). We mated the Sec and Aga monokaryons, genotyped the progeny, and performed bulked segregant analysis (BSA). We also fruited three Sec/Sec and three Aga/Aga dikaryons, and sampled transcriptomes at four developmental stages. Using BSA, we identified 105 top candidate genes with nonsynonymous SNPs that cosegregate with fruiting body phenotype. Transcriptome analyses of Sec/Sec versus Aga/Aga dikaryons identified 907 differentially expressed genes (DEGs) along four developmental stages. On the basis of BSA and DEGs, the top 25 candidate genes related to fruiting body development span 1.5 Mb (4% of the genome), possibly on a single chromosome, although the precise locus that controls the secotioid phenotype is unresolved. The top candidates include genes encoding a cytochrome P450 and an ATP-dependent RNA helicase, which may play a role in development, based on studies in other fungi.

The aim of this paper was to study the degradation and growth patterns of brown rot fungi in modified wood and to measure moisture content (MC), structural integrity and the acetyl content by frequent monitoring over 300 days. Mass loss (ML) in the modified wood materials increased slowly up to 3% for 50–100 days after which it flattened out and remained constant during the remainder of the test. Structural integrity and acetyl content were maintained in the modified wood materials and MC was lower compared to untreated wood throughout the decay test. ML results of untreated wood indicate that fungi in solid wood go through distinct phases; the degradation patterns in the modified wood materials were more difficult to interpret.

The gradual elimination of chromium from wood preservative formulations results in higher Cu leaching and increased susceptibility to wood decay fungi. Finding a sustainable strategy in wood protection has become of great interest among researchers. The objective of these in vitro studies was to demonstrate the effect of T-720-enriched organic charcoal (biochar) against five wood decay basidiomycetes isolated from strongly damaged poles. For this purpose, the antagonistic potential of Trichoderma harzianum (strain T-720) was confirmed among other four Trichoderma spp. against five brown-rot basidiomycetes in dual culture tests. T-720 was genetically transformed and tagged with the green fluorescent protein (GFP) in order to study its antagonistic mechanism against wood decay basidiomycetes. It was also demonstrated that T-720 inhibits the oxalic acid production by basidiomycetes, a well-known mechanism used by brown-rot fungi to detoxify Cu from impregnated wood. Additionally, this study evaluated the effect of biochar, alone or in combination with T-720, on Cu leaching by different preservatives, pH stabilization and prevention of wood decay caused by five basidiomycetes. Addition of biochar resulted in a significant Cu binding released from impregnated wood specimens. T-720-enriched biochar showed a significant reduction of wood decay caused by four basidiomycetes. The addition of T-720-enriched biochar to the soil into which utility poles are placed may improve the efficiency of Cr-free wood preservatives.
