Abstract: Neocortical layer 1 (L1) is a site of convergence between pyramidal-neuron dendrites and feedback axons where local inhibitory signaling can profoundly shape cortical processing. Evolutionary expansion of human neocortex is marked by distinctive pyramidal neurons with extensive L1 branching, but whether L1 interneurons are similarly diverse is underexplored. Using Patch-seq recordings from human neurosurgical tissue, we identified four transcriptomic subclasses with mouse L1 homologs, along with distinct subtypes and types unmatched in mouse L1. Subclass and subtype comparisons showed stronger transcriptomic differences in human L1 and were correlated with strong morphoelectric variability along dimensions distinct from mouse L1 variability. Accompanied by greater layer thickness and other cytoarchitecture changes, these findings suggest that L1 has diverged in evolution, reflecting the demands of regulating the expanded human neocortical circuit.

Proceedings of the National Academy of Sciences of the United States of America, 119(4), (2022): e2115635118, https://doi.org/10.1073/pnas.2115635118., November 2020 marked 2 y since the launch of the Earth BioGenome Project (EBP), which aims to sequence all known eukaryotic species in a 10-y timeframe. Since then, significant progress has been made across all aspects of the EBP roadmap, as outlined in the 2018 article describing the project’s goals, strategies, and challenges (1). The launch phase has ended and the clock has started on reaching the EBP’s major milestones. This Special Feature explores the many facets of the EBP, including a review of progress, a description of major scientific goals, exemplar projects, ethical legal and social issues, and applications of biodiversity genomics. In this Introduction, we summarize the current status of the EBP, held virtually October 5 to 9, 2020, including recent updates through February 2021. References to the nine Perspective articles included in this Special Feature are cited to guide the reader toward deeper understanding of the goals and challenges facing the EBP.

Since then, significant progress has been made across all aspects of the EBP roadmap, as outlined in the 2018 article describing the project’s goals, strategies, and challenges (1). The launch phase has ended and the clock has started on reaching the EBP’s major milestones. This Special Feature explores the many facets of the EBP, including a review of progress, a description of major scientific goals, exemplar projects, ethical legal and social issues, and applications of biodiversity genomics. In this Introduction, we summarize the current status of the EBP, held virtually October 5 to 9, 2020, including recent updates through February 2021. References to the nine Perspective articles included in this Special Feature are cited to guide the reader toward deeper understanding of the goals and challenges facing the EBP.

Abstract: November 2020 marked 2 y since the launch of the Earth BioGenome Project (EBP), which aims to sequence all known eukaryotic species in a 10-y timeframe. Since then, significant progress has been made across all aspects of the EBP roadmap, as outlined in the 2018 article describing the project’s goals, strategies, and challenges (1). The launch phase has ended and the clock has started on reaching the EBP’s major milestones. This Special Feature explores the many facets of the EBP, including a review of progress, a description of major scientific goals, exemplar projects, ethical legal and social issues, and applications of biodiversity genomics. In this Introduction, we summarize the current status of the EBP, held virtually October 5 to 9, 2020, including recent updates through February 2021. References to the nine Perspective articles included in this Special Feature are cited to guide the reader toward deeper understanding of the goals and challenges facing the EBP.

Abstract: The distribution and genetic structure of most plant species in Britain and Ireland bear the imprint of the last ice age. These patterns were largely shaped by random processes during recolonization but, in angiosperms, whole-genome duplication may also have been important. We investigate the distribution of cytotypes of Campanula rotundifolia, considering DNA variation, postglacial colonization, environmental partitioning and reproductive barriers. Cytotypes and genome size variation from across the species’ range were determined by flow cytometry and genetic variation was assessed using cpDNA markers. A common garden study examined growth and flowering phenology of tetraploid, pentaploid and hexaploid cytotypes and simulated a contact zone for investigation of reproductive barriers. Irish populations were entirely hexaploid. In Britain, hexaploids occurred mostly in western coastal populations which were allopatric with tetraploids, and in occasional sympatric inland populations. Chloroplast markers resolved distinct genetic groups, related to cytotype and geographically segregated; allopatric hexaploids were distinct from tetraploids, whereas sympatric hexaploids were not. Genome downsizing occurred between cytotypes. Progeny of open-pollinated clones from the contact zone showed that maternal tetraploids rarely produced progeny of other cytotypes, whereas the progeny of maternal hexaploids varied, with frequent pentaploids and aneuploids. The presence of distinctive hexaploid chloroplast types in Ireland, Scottish islands and western mainland Britain indicates that its establishment preceded separation of these land masses by sea-level rise c. 16 000 years BP. This group did not originate from British tetraploids and probably diverged before postglacial invasion from mainland Europe. The combination of cytotype, molecular, contact zone and common garden data shows an overall pattern reflecting postglacial colonization events, now maintained by geographic separat

20. The Diagram Disconnect: An Examination of Note-Taking Behaviors In CollegeStudents

Abstract: Note-taking in college courses is prevalent yet often ineffective. One potential reason is a disconnect between the infor-mation in lectures and that recorded in notes. Whereas science-based lectures frequently include diagrams, students notesoften fail to include them. This disconnect likely inhibits learning and may be exacerbated by digital note-taking. We in-vestigated students note-taking during two mini neuroscience lectures and its relation to recall. Students were assigned todiagram presence (diagram embedded in notes for first or second lecture) and note-taking method (typed or handwritten)conditions. Students recalled more in the diagram first condition. There was no recall difference based on note-takingmethod. Including diagrams in notes for the first lecture likely primed participants to attend to diagrams in the subsequentlecture, helping them realize the importance of the diagram. The lack of a note-taking method effect is inconsistent withpast research, but may reflect increasing use of digital note-taking.

Abstract: Trees are usually eliminated in field due their possible competition with crops for water and nutrients. Our work aimed to investigate how to manage S. senegal trees to optimize their association with crops. A field trial was conducted under natural conditions in a S. senegal plantation 10 years old. Investigations were conducted during two years to evaluate the effect of S. senegal trees management on gum arabic yield, associated cowpea yield, soil mycorrhizal fungi spores density and enzymes activities reflecting soil fertility such as fluorescein diacetate (FDA), acid phosphatase and dehydrogenase. Four treatments of pruning were applicated (control, shoots, roots, shoots and roots pruning) and tapped in november. Soil samples were collected during dry and wet seasons at 0-25 cm layer. Cowpeas were sown at the beginning of rainy season in rows 0.5 m apart. Results showed that shoots pruning significantly increased gum arabic while a negative effect of roots pruning was noted. The presence of S. senegal increased soil fertility and cowpea yield. Any significant effect of treatments was noted on pods yield during the first year. However, shoots pruning, shoot and roots pruning increased significantly pods yield during the second year. Shoots and roots pruning seemed to decrease FDA and spores density depending on the season. Our results showed that shoots and roots pruning increased the positive effect on soil fertility and the association of S. senegal with annual crops.

Investigations were conducted during two years to evaluate the effect of S. senegal trees management on gum arabic yield, associated cowpea yield, soil mycorrhizal fungi spores density and enzymes activities reflecting soil fertility such as fluorescein diacetate (FDA), acid phosphatase and dehydrogenase. Four treatments of pruning were applicated (control, shoots, roots, shoots and roots pruning) and tapped in november. Soil samples were collected during dry and wet seasons at 0-25 cm layer. Cowpeas were sown at the beginning of rainy season in rows 0.5 m apart. Results showed that shoots pruning significantly increased gum arabic while a negative effect of roots pruning was noted. The presence of S. senegal increased soil fertility and cowpea yield. Any significant effect of treatments was noted on pods yield during the first year. However, shoots pruning, shoot and roots pruning increased significantly pods yield during the second year. Shoots and roots pruning seemed to decrease FDA and spores density depending on the season. Our results showed that shoots and roots pruning increased the positive effect on soil fertility and the association of S. senegal with annual crops.

Abstract: Agroforestry systems, containing mixtures of trees and crops, are often promoted because the net effect of interactions between woody and herbaceous components is thought to be positive if evaluated over the long term. From a modelling perspective, agroforestry has received much less attention than monocultures. However, for the potential of agroforestry to impact food security in Africa to be fully evaluated, models are required that accurately predict crop yields in the presence of trees. The positive effects of the fertiliser tree gliricidia (Gliricidia sepium) on maize (Zea mays) are well documented and use of this tree-crop combination to increase crop production is expanding in several African countries. Simulation of gliricidia-maize interactions can complement field trials by predicting crop response across a broader range of contexts than can be achieved by experimentation alone. We tested a model developed within the APSIM framework. APSIM models are widely used for one dimensional (1D), process-based simulation of crops such as maize and wheat in monoculture. The Next Generation version of APSIM was used here to test a 2D agroforestry model where maize growth and yield varied spatially in response to interactions with gliricidia. The simulations were done using data for gliricidia-maize interactions over two years (short-term) in Kenya and 11 years (long-term) in Malawi, with differing proportions of trees and crops and contrasting management. Predictions were compared with observations for maize grain yield, and soil water content. Simulations in Kenya were in agreement with observed yields reflecting lower observed maize germination in rows close to gliricidia. Soil water content was also adequately simulated, except for a tendency for slower simulated drying of the soil profile each season. Abstract: The Line Islands Ridge (LIR), located south of the Hawaiian Islands between 7°N and 1°S, is one of the few large central Pacific regions shallower than the regional carbonate compensation depth. Thick sequences of carbonate sediments have accumulated around the LIR despite it being located in the sediment-starved central tropical Pacific. The LIR is an important source of carbonates to the surrounding region and deposition around the LIR has expanded the equatorial Pacific carbonate sediment tongue by about 5% of its total area. Furthermore, sediments on the ridge are potentially important paleoceanographic archives. A recent survey at the crest of the LIR finds evidence for high current activity, significant erosion, but overall net sediment deposition. Currents are strong enough to form sediment waves and lee drifts in the Palmyra Basin, at the northern terminus of the LIR. Sediments along the LIR are pelagic foraminiferal sands that are easily eroded and flow out into the surrounding abyssal plain in active submarine channel systems. As channels migrate, pelagic sediments fill in the abandoned channel arms. Despite significant sediment losses from the top of the ridge, 1.3 km of sediment has accumulated in the upper Palmyra Basin over basement formed 68 to 85 million years ago (Ma). Late Neogene erosion may be more extensive than earlier erosion cycles, in response to reduced sediment production as the Palmyra Basin exited the high productivity equatorial latitudes. Abstract: The Line Islands Ridge (LIR), located south of the Hawaiian Islands between 7°N and 1°S, is one of the few large central Pacific regions shallower than the regional carbonate compensation depth. Thick sequences of carbonate sediments have accumulated around the LIR despite it being located in the sediment-starved central tropical Pacific. The LIR is an important source of carbonates to the surrounding region and deposition around the LIR has expanded the equatorial Pacific carbonate sediment tongue by about 5% of its total area. Furthermore, sediments on the ridge are potentially important paleoceanographic archives. A recent survey at the crest of the LIR finds evidence for high current activity, significant erosion, but overall net sediment deposition. Currents are strong enough to form sediment waves and lee drifts in the Palmyra Basin, at the northern terminus of the LIR. Sediments along the LIR are pelagic foraminiferal sands that are easily eroded and flow out into the surrounding abyssal plain in active submarine channel systems. As channels migrate, pelagic sediments fill in the abandoned channel arms. Despite significant sediment losses from the top of the ridge, 1.3 km of sediment has accumulated in the upper Palmyra Basin over basement formed 68 to 85 million years ago (Ma). Late Neogene erosion may be more extensive than earlier erosion cycles, in response to reduced sediment production as the Palmyra Basin exited the high productivity equatorial latitudes. Sediments with good stratigraphic order needed for paleoceanographic study are limited in this dynamic sedimentary environment, but can be found with proper survey.

39. Nutrient requirements of in vitro cultured Halophila ovalis and Posidoniq coriacea: nitrogen source

Abstract: Axenic cultures of Posidonia coriacea and Halophila ovalis were established to investigate the effect of nitrogen addition and the optimum concentration of MS (Murashige and Skoog Physiol Plant 15:473–497, 1962) macronutrients, micronutrients and organics. Chlorophyll content and dissolved oxygen of H. ovalis was optimal at the control concentration of macronutrients (half strength MS). Below and above this concentration, growth of this species was compromised. A similar effect was observed for H. ovalis growth in response to MS micronutrient. However, changing the MS vitamins concentration had no impact on the growth of H. ovalis or P. coriacea. Leaf and root numbers of H. ovalis were highest in the treatment with the most nitrogen (50.6 mM). Root architecture of H. ovalis was affected by nitrogen source; shorter roots without root hairs were produced when NH4NO3 was present in the media. Longer roots with root hairs were produced when the cultures were grown in the absence of NH4NO3. P. coriacea growth did not exceed that of the control for any of the treatments. The response of H. ovalis to nutrient addition suggests its opportunistic nature, whereas the lack of response by P. coriacea suggests there may be some other limiting factor. This study shows that the proposed formulation of ½ strength MS can be suitable for both H. ovalis and P. coriacea.

40. Under Pressure: Does Osmotic Potential of the Culture Medium Affect Growth of Seagrasses?

Abstract: Seagrasses have large carbohydrate reserves in their rhizomes, which are used to survive over winter and other periods of reduced light. Seagrass seedlings also have carbohydrate reserves in an enlarged hypocotyl, which assist during establishment. Axenic seedling cultures of the seagrass Posidonia coriacea were used to test whether the osmotic potential of the culture media affects growth. Sucrose, glucose and mannitol were added to the culture media (individually and in combination) at O to 90 mM to determine whether they have a nutritive, osmotic or combined effect on growth. To ensure osmotic potentials of the culture media were similar, carbohydrate concentrations were compared in millimoles rather than g 1-1 . Osmotic potential was calculated using the formula [ps] -CiRT, where [ps] is osmotic potential, C is sugar concentration (mM), i is ionisation constant, R is gas constant and T is the temperature. Osmolality of the culture media was also determined using a one-ten Fiske osmometer. Glucose and sucrose were found to have a nutritive effect on growth as biomass and leaf number increased with their increasing concentrations. Mannitol had. a non-dose dependent effect on growth, suggesting an osmotic rather than nutritive role. Growth in the no carbohydrates treatments was significantly reduced compared with treatments containing glucose and/or sucrose. These findings suggest that P. coriacea can use carbohydrate reserves from the hypocotyl but will also assimilate carbohydrates from the culture medium.

41. Rangewide ploidy variation and evolution in Acacia senegal: a north-south divide?

Abstract: Knowledge of rangewide variation in DNA content and ploidy level may be valuable in understanding the evolutionary history of a species. Recent studies of Acacia senegal report diploids and occasional tetraploids in the Sudano-Sahelian region of sub-Saharan Africa, but nothing is known about the overall extent of DNA ploidy variation within the species. In this study, we determine the DNA content and ploidy level of A. senegal across its native range, and explore whether the variation is related to its evolutionary and colonization history. We used propidium iodide flow cytometry (FCM) to estimate DNA content (2C value) and infer ploidy in 157 individuals from 54 populations on various tissues, using seeds, fresh leaves, dried leaves and twigs and herbarium specimens. The mean 2C DNA (pg ± s.d.) contents detected were 1.47 ± 0.09, 2.12 ± 0.02, 2.89 ± 0.12, and a single individual with 4.51 pg, corresponding to a polyploid series of diploid, triploid, tetraploid and hexaploid individuals. Diploids were confirmed by chromosome counts (2n = 2x = 26). Most populations (90.7 %) were of single ploidy level, while mixed ploidy populations (9.3 %) comprising mostly diploids (2x+3x, 2x+4x and 2x+6x) were restricted to the Sudano-Sahelian and Indian subcontinent regions, its northern range. The species is predominantly diploid, and no mixed ploidy populations were detected in east and southern Africa, its southern range. The geographic pattern of ploidy variation in conjunction with existing phylogeographic and phylogenetic data of the species suggests that polyploids have occurred multiple times in its evolutionary and recent colonization history, including contemporary ecological timescales. The successful use of external tissues of dried twigs in FCM is new, and presents the opportunity to study numerous other dryland woody species.
