Background: Asthma is a disease of varying severity and differing disease mechanisms. To date, studies aimed at stratifying asthma into clinically useful phenotypes have produced a number of phenotypes that have yet to be assessed for stability and to be validated in independent cohorts. The aim of this study was to define and validate, for the first time ever, clinically driven asthma phenotypes using two independent, severe asthma cohorts: ADEPT and U-BIOPRED. Methods: Fuzzy partition-around-medoid clustering was performed on pre-specified data from the ADEPT participants (n = 156) and independently on data from a subset of U-BIOPRED asthma participants (n = 82) for whom the same variables were available. Models for cluster classification probabilities were derived and applied to the 12-month longitudinal ADEPT data and to a larger subset of the U-BIOPRED asthma dataset (n = 397). High and low type-2 inflammation phenotypes were defined as high or low Th2 activity, indicated by endobronchial biopsies gene expression changes downstream of IL-4 or IL-13. Results: Four phenotypes were identified in the ADEPT (training) cohort, with distinct clinical and biomarker profiles. Phenotype 1 was "mild, good lung function, early onset", with a low-inflammatory, predominantly Type-2, phenotype. Phenotype 2 had a "moderate, hyper-responsive, eosinophilic" phenotype, with moderate asthma control, mild airflow obstruction and predominant Type-2 inflammation. Phenotype 3 had a "mixed severity, predominantly fixed obstructive, non-eosinophilic and neutrophilic" phenotype, with moderate asthma control and low Type-2 inflammation. Phenotype 4 had a "severe uncontrolled, severe reversible obstruction, mixed granulocytic" phenotype, with moderate Type-2 inflammation. These phenotypes had good longitudinal stability in the ADEPT cohort. They were reproduced and demonstrated high classification probability in two subsets of the U-BIOPRED asthma cohort. Cultivating contemporary photosynthetic producers to generate energy and capture carbon from the atmosphere is one potential approach to sustaining society without disrupting the climate. Algae, photosynthetic aquatic microorganisms, are the fastest growing primary producers in the world and can therefore produce more energy with less land, water, and nutrients than terrestrial plant crops. We review recent progress and challenges in developing bioenergy technology based on algae. A variety of high-value products in addition to biofuels can be harvested from algal biomass, and these may be key to developing algal biotechnology and realizing the commercial potential of these organisms. Aspects of algal biology that differentiate them from plants demand an integrative approach based on genetics, cell biology, ecology, and evolution. We call for a systems approach to research on algal biotechnology rooted in understanding their biology, from the level of genes to ecosystems, and integrating perspectives from physical, chemical, and social sciences to solve one of the most critical outstanding technological problems.

These include high susceptibility to invading aquatic consumers and weeds, as well as prodigious requirements for nutrients that may compete with the fertiliser demands of other crops. Most research on algal biofuel technologies approaches these problems from a cellular or genetic perspective, attempting either to engineer or select algal strains with particular traits. However, inherent functional trade-offs may limit the capacity of genetic selection or synthetic biology to simultaneously optimise multiple functional traits for biofuel productivity and resilience. We argue that a community engineering approach that manages microalgal diversity, species composition and environmental conditions may lead to more robust and productive biofuel ecosystems. We review evidence for trade-offs, challenges and opportunities in algal biofuel cultivation with a goal of guiding research towards intensifying bioenergy production using established principles of community and ecosystem ecology.

A cross-ecosystem comparison of data obtained from 20 French Mediterranean lagoons with contrasting eutrophication status provided the basis for investigating the variables that best predict chlorophyll a (Chl a) concentrations and nutrient limitation of phytoplankton biomass along a strong nutrient enrichment gradient. Summer concentrations of dissolved inorganic nitrogen (DIN) and phosphorus (DIP) comprised only a small fraction of total nitrogen (TN) and total phosphorus (TP). On the basis of inorganic nutrient concentrations, the most oligotrophic lagoons appeared to be phosphorus-limited, with a tendency towards the development of nitrogen limitation as eutrophication increased, as evidenced by decreasing DIN:DIP ratios. A weak but significantly positive relationship was found between dissolved silicate (DSi) and Chl a, reflecting DSi accumulation in the water column along the trophic state gradient and implying a progressive shift away from potential Si limitation of phytoplankton growth. Observed concentrations of Chl a were far better explained by TN and TP than by DIN and DIP concentrations, suggesting that a total nutrient based approach is likely to be the most appropriate for managing eutrophication in Mediterranean lagoons and other coastal waters. These results give credence to the idea that marine and freshwater environments respond in a similar fashion to nutrient enrichment., Une comparaison inter-sites des concentrations de nutriments et de chlorophylle a (Chl a) issues de 20 lagunes de Mediterranee francaise de degres divers d'eutrophisation a ete menee afin de rechercher la meilleure variable expliquant le niveau de Chl a et d'explorer la limitation de la production primaire par les nutriments au long d'un fort gradient d'enrichissement en azote et phosphore. En ete, l'azote et le phosphore inorganiques dissous (DIN et PID respectivement) constituaient une faible fraction des concentrations d'azote et de phosphore totaux (TN et TP respectivement). Sur la base des concentrations en nutriments inorganiques, la plupart des lagunes oligotrophes semblaient limitees par le phosphore avec une tendance vers des rapports DIN:DIP decroissants, et donc une limitation par l'azote, lorsque l'eutrophisation augmentait. Une correlation positive mais faible a aussi ete obtenue entre les concentrations de silicates (DSi) et Chl a, indiquant une accumulation de DSi dans la colonne d'eau et donc une diminution de la limitation de la croissance du phytoplancton par la silice au fur et a mesure que l'eutrophisation augmentait. Le fait que Chl a etait beaucoup mieux expliquee par TN et TP que par DIN et DIP suggere meme que l'approche limnologique basee sur l'ensemble des nutriments est vraisemblablement la meilleure pour gerer l'eutrophisation dans les lagunes mediterraneennes et dans d'autres ecosystemes marins cotiers. Ces constats renforcent l'idee que les ecosystemes marins et d'eau douce repondent de facon similaire a l'enrichissement en nutriments.

Implementation of measures to protect and improve the environment requires knowledge about people’s preferences, both to ensure economic means and to gain public support for the measures. Since environmental legislation and protection measures become increasingly cross-national, knowledge of benefit perception among people across countries is important. This study addresses the aggregated preferences of environmentally- concerned individuals in France, USA, Norway, Russia, China and Spain. The aggregated preferences in all groups showed emphasis on pollution issues (rank 1 out of six issues in all six countries). The groups were least concerned with animal rights, which here included the right for top predators like tigers and wolf to roam the wilderness in a way that may cause statistical fatalities (rank 4–6). The group’s concern for pollution decreased with the buying power of the country to which they belonged (r2 =0.967). Also, agreement among the individuals in the groups tended to be less when the buying power was large (r2 =0.940). The study shows that benefits accrued in one country may not have the same weight in another country, in particular if countries have different economic development status. It also suggests that efforts to preserve species diversity may require other types of public motivation than efforts to reduce pollution or to use non-renewable resources.
