Abstract

While the science supporting fisheries management has generally been dominated by the natural sciences, there has been a growing recognition that managing fisheries essentially means managing economic systems. Indeed, over the past seven decades, economic ideas and insights have increasingly come to play a role in fisheries management and policy. As an illustration of this, the International Council for the Exploration of the Sea (ICES) has been actively seeking to expand the scope of its scientific expertise beyond natural sciences [another inter-governmental marine science organization which has done this over the same period is the North Pacific Marine Science organization (PICES)]. In particular, the recently created ICES Working Group on Economics set out to review current work and key future needs relating to economic research and management advice on marine capture fisheries. This article presents the results of this review and addresses how economic research can be incorporated into the science of ICES to provide integrated perspectives on fisheries systems that can contribute to the provision of advice in support of policy development and management decision-making for sustainable uses of living marine resources.

Abstract

Non-coding genetic variants play an important role in driving susceptibility to complex diseases but their characterization remains challenging. Here, we employed a novel approach to interrogate the genetic risk of such polymorphisms in a more systematic way by targeting specific regulatory regions relevant for the phenotype studied. We applied this method to meningococcal disease susceptibility, using the DNA binding pattern of RELA - a NF-kB subunit, master regulator of the response to infection - under bacterial stimuli in nasopharyngeal epithelial cells. We designed a custom panel to cover these RELA binding sites and used it for targeted sequencing in cases and controls. Variant calling and association analysis were performed followed by validation of candidate polymorphisms by genotyping in three independent cohorts. We identified two new polymorphisms, rs4823231 and rs11913168, showing signs of association with meningococcal disease susceptibility. In addition, using our genomic data as well as publicly available resources, we found evidences for these SNPs to have potential regulatory effects on ATXN10 and LIF genes respectively. The variants and related candidate genes are relevant for infectious diseases and may have important contribution for meningococcal disease pathology. Finally, we described a novel genetic association approach that could be applied to other phenotypes.

Abstract

Fever is the most common reason that children present to Emergency Departments. Clinical signs and symptoms suggestive of bacterial infection are often non-specific, and there is no definitive test for the accurate diagnosis of infection. The 'omics' approaches to identifying biomarkers from the host-response to bacterial infection are promising. In this study, lipidomic analysis was carried out with plasma samples obtained from febrile children with confirmed bacterial infection (n = 20) and confirmed viral infection (n = 20). We show for the first time that bacterial and viral infection produces distinct profile in the host lipidome. Some species of glycerophosphoinositol, sphingomyelin, lysophosphatidylcholine and cholesterol sulfate were higher in the confirmed virus infected group, while some species of fatty acids, glycerophosphocholine, glycerophosphoserine, lactosylceramide and bilirubin were lower in the confirmed virus infected group when compared with confirmed bacterial infected group. A combination of three lipids achieved an area under the receiver operating characteristic (ROC) curve of 0.911 (95% CI 0.81 to 0.98). This pilot study demonstrates the potential of metabolic biomarkers to assist clinicians in distinguishing bacterial from viral infection in febrile children, to facilitate effective clinical management and to the limit inappropriate use of antibiotics.

BACKGROUND: Alcohol consumption is associated with enhanced Th2 immune responses. Aim: To investigate the frequency of false-positive serological tests of allergy in alcoholics.METHODS: A total of 138 alcoholic patients consecutively admitted to hospital underwent a panel of allergy tests that included serum total IgE, a multiallergen IgE test (UniCAP Phadiatop), and skin prick tests to relevant aeroallergens in the area, which were considered the standard reference for atopy. In selected cases with positive specific IgE (sIgE) to cross-reactive carbohydrate determinants (CCDs) on ImmunoCAP, we determined sIgE to Hymenoptera venom components (ADVIA Centaur) and a microarray of 103 allergen components (ISAC).RESULTS: Increased (>170 IU/mL) serum total IgE was observed in 59/110 (54%) of non-atopic (skin prick test-negative) alcoholics. Forty-six non-atopic alcoholics (42%) had a positive multiallergen IgE test. This finding was closely associated with high serum concentrations of total IgE and sIgE to CCDs. The vast majority of alcoholics with positive CCD-sIgE showed positivity to glycosylated plant and Hymenoptera allergen components on ISAC and ADVIA Centaur. Only one out of 26 patients with positive sIgE to CCD and Hymenoptera venoms developed honeybee venom allergy after a median follow-up of 166 months. Measurements of sIgE to CCD markers on ImmunoCAP, ADVIA Centaur, and ISAC showed imperfect correlation.CONCLUSIONS: Serological tests for allergy should be interpreted with caution in alcoholics, who frequently show increased levels of total IgE, CCD-sIgE and subsequent positivity of sIgE to glycosylated allergen components, irrespective of the method used.

Abstract

The comprehension of molecular recognition phenomena demands the understanding of the energetic and kinetic processes involved. General equations valid for the thermodynamic analysis of any observable that is assessed as a function of the concentration of the involved compounds are described, together with their implementation in the AFFINImeter software. Here, a maximum of three different molecular species that can interact with each other to form an enormous variety of supramolecular complexes are considered. The corrections currently employed to take into account the effects of dilution, volume displacement, concentration errors and those due to external factors, especially in the case of ITC measurements, are included. The methods used to fit the model parameters to the experimental data, and to generate the uncertainties are described in detail. A simulation tool and the so called kinITC analysis to get kinetic information from calorimetric experiments are also presented. An example of how to take advantage of the AFFINImeter software for the global multi-temperature analysis of a system exhibiting cooperative 1:2 interactions is presented and the results are compared with data previously published. Some useful recommendations for the analysis of experiments aimed at studying molecular interactions are provided.

Abstract

The progressive aging of the Galician population results in increasing costs of healthcare and appears as an emerging social challenge to address the care and needs of older patients. Currently, the fragmentation of the healthcare system and the scarce coordination between healthcare and sociosanitary systems should lead to development of tools and ways to optimize the resources towards integrated chronicity healthcare models. Over the last years, diverse chronicity care models to improve healthcare efficiency, have been defined. This article describes a comprehensive healthcare model focused on networking and cross-working between the different stakeholders involved in chronicity healthcare, as well as the opportunities to implement improvement strategies in chronic diseases management., O progresivo envellecemento da poboación galega supón un crecemento nos custos do sistema sanitario e un desafío para a sociedade no seu conxunto para abordar os coidados e necesidades destes pacientes. Na actualidade a fragmentación do sistema sanitario en niveis e a práctica ausencia dunha coordinación entre estes e os servizos sociais fai necesario o desenvolvemento de instrumentos e canles que optimicen os recursos e melloren de forma progresiva a atención integral das persoas en situación de cronicidade. Durante os últimos anos definíronse diversos modelos de atención á cronicidade nun intento de mellorar a eficiencia da xestión dos sistemas sanitarios. Neste artigo descríbese un modelo de atención integral centrado no traballo en rede e transversal entre os diferentes actores de sistema sanitario implicados na atención á cronicidade, así como as oportunidades de mellora do modelo de atención á cronicidade.

Magnetite (Fe3O4) is attracting much interest in the last years due to its robust ferrimagnetism down to nanometer thickness, good electrical conductivity and presumed half-metal character. In particular, Fe3O4 films are studied as ideal cases for the design of improved bulk magnets [1] and have been tentatively used in spin-valves and spin-LEDs. Fe3O4 presents a low-temperature metal-insulator transition, the Verwey transition (TV) which has also been proposed for spintronic applications. An open question is to what extent the preparation of Fe3O4 films can affect their detailed magnetic properties, such as the magnetic anisotropy axis. This information is required to efficiently apply Fe3O4 in technological multiphase magnets and spintronic applications [1]. Most of studies dealing with bulk and Fe3O4 thin film systems show room temperature (RT) in-plane <110> magnetic easy axis. By contrast, we show in this work the preparation of pure stoichiometric Fe3O4 thin films with RT easy axes along the in-plane <100> directions [2], i.e. rotated by 45º respect to previous studies. Fe3O4 films have been grown by ablation from a sintered hematite target using a nanosecond infrared (IR) laser at 1064 nm and a substrate temperature of 750 K [3]. Single crystal substrates of SrTiO3, MgAl2O4 and MgO have been used. The films were characterized using XRD, AFM, Raman and Mössbauer spectroscopies, vectorial magneto-optical Kerr effect microscopy (v-MOKE) and SQUID magnetometry. All films consisted of stoichiometric Fe3O4 and presented a Verwey transition at TV=115-118 K. RT in-plane hysteresis loops were measured by vectorial-MOKE as a function of the direction of the applied magnetic field in the 0º-360º range with an angular step of 5º. For all epitaxial films under study, the highest coercivity and remanence are found at 0º, 90º, 180º and 270º (i.e. <100> directions), thus orthogonal to each other, while the lowest coercivity values are found between them [Figures 1(a) and 1(b

25. Immunoglobulin-E reactivity to a glycosylated food allergen (peanuts) due to interference with cross-reactive carbohydrate determinants in heavy drinkers

IgE sensitization to N-glycans (cross-reactive carbohydrate determinants, CCDs) may be increased in heavy drinkers, who therefore show IgE reactivity to aeroallergens, latex, and Hymenoptera venoms. The peanut, a CCD-bearing allergen, is the leading cause of severe food allergic reactions in many populations. AIM OF THE STUDY: To investigate the potential interference of CCDs with determinations of IgE to peanuts in heavy drinkers. METHODS: We determined IgE to peanuts and IgE to a CCD marker (MUXF(3), the N-glycan from bromelain) in 41 heavy drinkers admitted to the hospital and 54 healthy controls. None of the participants reported symptoms of peanut allergy. In cases with positive (>or=0.35 kU/l) IgE to peanuts, we performed inhibition assays with a neoglycoprotein consisting of MUXF(3) molecules coupled to bovine serum albumin (MUXF(3)-BSA) and a similar neoglycoprotein lacking xylose and fucose (MM-BSA). In the same cases, we screened for IgE to a panel of recombinant nonglycosylated peanut allergens. SDS-PAGE immunoblotting and inhibition assays were performed in selected cases. RESULTS: The prevalence of positive IgE to peanuts was 22 and 3.7% in heavy drinkers and healthy controls, respectively (p < 0.001). Peanut-IgE positivity was closely related to the presence of IgE to CCDs. In most (8/9) heavy drinkers with positive IgE to peanuts, reactivity was inhibited by preincubation with MUXF(3)-BSA, but not with MM-BSA. IgE binding to multiple bands on immunoblotting studies was also inhibited by MUXF(3)-BSA preincubation. IgE to nonglycosylated recombinant peanut allergens was uniformly negative. CONCLUSION: Heavy drinking is associated with clinically asymptomatic IgE reactivity to peanuts, a relevant food allergen, in relation to CCD interference.

Abstract

The evaluation of the ROUT (Retrieval of Unformatted Text) document retrieval system consisted of a detailed examination of various retrieval errors in the context of different queryprocessing methods. Many of the methods used to extend the power of the basic coordinate indexing mechanism were compared. Three different evaluation techniques were used in the study. Based on the analysis, conclusions were drawn about the relative merits of several query- processing methods, and some general comments were made about the limitations of coordinate indexing as applied to the document retrieval problem.

28. METRIZED SCHEDULING RELATIONS AND APPLICATIONS

Abstract

The definitions of scheduling relations previously presented by the author are extended in this report to allow the translation of a relation net into an exact schedule. Time conditional conflicts in scheduling relation nets are detected and resolved by operating on extended versions of the bilateral implication and truth tables.
