Folate metabolism can be an effective target for cancer treatment. However, standard cell culture conditions utilize folic acid, a non-physiological folate source for most tissues. We find that the enzyme that couples folate and methionine metabolic cycles, methionine synthase, is required for cancer cell proliferation and tumour growth when 5-methyl tetrahydrofolate (THF), the major folate found in circulation, is the extracellular folate source. In such physiological conditions, methionine synthase incorporates 5-methyl THF into the folate cycle to maintain intracellular levels of the folates needed for nucleotide production. 5-methyl THF can sustain intracellular folate metabolism in the absence of folic acid. Therefore, cells exposed to 5-methyl THF are more resistant to methotrexate, an antifolate drug that specifically blocks folic acid incorporation into the folate cycle. Together, these data argue that the environmental folate source has a profound effect on folate metabolism, determining how both folate cycle enzymes and antifolate drugs impact proliferation.

To accelerate the translation of cancer nanomedicine, we used an integrated genomic approach to improve our understanding of the cellular processes that govern nanoparticle trafficking. We developed a massively parallel screen that leverages barcoded, pooled cancer cell lines annotated with multiomic data to investigate cell association patterns across a nanoparticle library spanning a range of formulations with clinical potential. We identified both materials properties and cell-intrinsic features that mediate nanoparticle-cell association. Using machine learning algorithms, we constructed genomic nanoparticle trafficking networks and identified nanoparticle-specific biomarkers. We validated one such biomarker: gene expression of SLC46A3 , which inversely predicts lipid-based nanoparticle uptake in vitro and in vivo. Our work establishes the power of integrated screens for nanoparticle delivery and enables the identification and utilization of biomarkers to rationally design nanoformulations.

BACKGROUND: In 2014, germline signal transducer and activator of transcription (STAT) 3 gain-of-function (GOF) mutations were first described to cause a novel multisystem disease of early-onset lymphoproliferation and autoimmunity. OBJECTIVE: This pivotal cohort study defines the scope, natural history, treatment, and overall survival of a large global cohort of patients with pathogenic STAT3 GOF variants. METHODS: We identified 191 patients from 33 countries with 72 unique mutations. Inclusion criteria included symptoms of immune dysregulation and a biochemically confirmed germline heterozygous GOF variant in STAT3. RESULTS: Overall survival was 88%, median age at onset of symptoms was 2.3 years, and median age at diagnosis was 12 years. Immune dysregulatory features were present in all patients: lymphoproliferation was the most common manifestation (73%); increased frequencies of double-negative (CD4-CD8-) T cells were found in 83% of patients tested. Autoimmune cytopenias were the second most common clinical manifestation (67%), followed by growth delay, enteropathy, skin disease, pulmonary disease, endocrinopathy, arthritis, autoimmune hepatitis, neurologic disease, vasculopathy, renal disease, and malignancy. Infections were reported in 72% of the cohort. A cellular and humoral immunodeficiency was observed in 37% and 51% of patients, respectively. Clinical symptoms dramatically improved in patients treated with JAK inhibitors, while a variety of other immunomodulatory treatment modalities were less efficacious. Thus far, 23 patients have undergone bone marrow transplantation, with a 62% survival rate. CONCLUSION: STAT3 GOF patients present with a wide array of immune-mediated disease including lymphoproliferation, autoimmune cytopenias, and multisystem autoimmunity. Patient care tends to be siloed, without a clear treatment strategy. Thus, early identification and prompt treatment implementation are lifesaving for STAT3 GOF syndrome.

Predators can have devastating impacts on biodiversity when introduced beyond their native range, and so are often lethally controlled. However, control of a dominant invasive predator can backfire if it frees subordinate invasive predators from top-down suppressive and competitive effects, a process referred to as ‘mesopredator release’. Identifying whether mesopredator release is occurring is important for biodiversity conservation, particularly as integrated management of co-occurring invasive predators is often costly or infeasible. Lethal control of the introduced red fox *Vulpes vulpes* (hereafter ‘fox’) is a common conservation strategy in Australia. Concerningly, there are reports of dramatic declines in native prey species following targeted fox control, frequently attributed to mesopredator release of the feral cat *Felis catus* (hereafter ‘cat’). However, evidence for the mesopredator release hypothesis is limited--both in regards to cats and more generally. The primary aim of my thesis was to test whether lethal fox control (1080 poison-baiting) causes mesopredator release of cats, in terms of their population density, occurrence, and behaviour. In the process, I derived the first estimates of cat density for Australia wet forest environments as well as investigated the relative importance of fox control, fire and other environmental drivers on the occurrence of these invasive predators and two threatened native prey species. My study was conducted across two regions in temperate south-eastern Australia, where foxes and cats are the only medium-large sized terrestrial predators (dingoes *Canis familiaris* being absent). In one region, I spatially replicated a landscape-scale control-impact experimental design three times within a long-term fox control program. There has been no previous comprehensive analysis of the potential impact of the introduced European red fox, Vulpes vulpes, on Australian reptiles.Aims: We seek to provide an inventory of all Australian reptile species known to be consumed by the fox, and identify characteristics of squamate species associated with such predation. Two introduced carnivores, the European red fox Vulpes vulpes and domestic cat Felis catus, have had, and continue to have, major impacts on wildlife, particularly mammals, across Australia. Based mainly on the contents of almost 50,000 fox dietary samples, we provide the first comprehensive inventory of Australian mammal species known to be consumed by foxes, and compare this with a similar assessment for cats. We recorded consumption by foxes of 114 species of Australian land mammal (40% of extant species), fewer than consumed by cats (173 species). Foxes are known to consume 42 threatened mammal species (50% of Australia's threatened land mammals and 66% of those within the fox's Australian range). Reflecting the importance of mammals in their diet, foxes are known to consume a far higher proportion of Australian mammal species (40%) than of Australian birds (24%) and reptiles (16%). Both foxes and cats were most likely to consume medium-sized mammals, with the likelihood of predation by foxes peaking for mammals of ca. 280 g and by cats at ca. 130 g. For non-flying mammals, threatened species had a higher relative likelihood of predation by foxes than non-threatened species. Using trait-based modelling, we estimate that many now-extinct Australian mammal species had very high likelihoods of predation by foxes and cats, although we note that for some of these species, extinction likely pre-dated the arrival of foxes. These two predators continue to have compounding and complementary impacts on Australian mammals. Targeted and integrated management of foxes and cats is required to help maintain and recover the Australian mammal fauna.

Two introduced carnivores, the European red fox Vulpes vulpes and domestic cat Felis catus, have had, and continue to have, major impacts on wildlife, particularly mammals, across Australia. Based mainly on the contents of almost 50,000 fox dietary samples, we provide the first comprehensive inventory of Australian mammal species known to be consumed by foxes, and compare this with a similar assessment for cats. We recorded consumption by foxes of 114 species of Australian land mammal (40% of extant species), fewer than consumed by cats (173 species). Foxes are known to consume 42 threatened mammal species (50% of Australia's threatened land mammals and 66% of those within the fox's Australian range). Reflecting the importance of mammals in their diet, foxes are known to consume a far higher proportion of Australian mammal species (40%) than of Australian birds (24%) and reptiles (16%). Both foxes and cats were most likely to consume medium-sized mammals, with the likelihood of predation by foxes peaking for mammals of ca. 280 g and by cats at ca. 130 g. For non-flying mammals, threatened species had a higher relative likelihood of predation by foxes than non-threatened species. Using trait-based modelling, we estimate that many now-extinct Australian mammal species had very high likelihoods of predation by foxes and cats, although we note that for some of these species, extinction likely pre-dated the arrival of foxes. These two predators continue to have compounding and complementary impacts on Australian mammals. Targeted and integrated management of foxes and cats is required to help maintain and recover the Australian mammal fauna.

Yet much of what is known about sharks has been inferred from catch records in industrial fisheries, whereas far less information is available about sharks that live in coastal habitats3. Here we address this knowledge gap using data from more than 15,000 standardized baited remote underwater video stations that were deployed on 371 reefs in 58 nations to estimate the conservation status of reef sharks globally. Our results reveal the profound impact that fishing has had on reef shark populations: we observed no sharks on almost 20% of the surveyed reefs. Reef sharks were almost completely absent from reefs in several nations, and shark depletion was strongly related to socio-economic conditions such as the size and proximity of the nearest market, poor governance and the density of the human population. However, opportunities for the conservation of reef sharks remain: shark sanctuaries, closed areas, catch limits and an absence of gillnets and longlines were associated with a substantially higher relative abundance of reef sharks. These results reveal several policy pathways for the restoration and management of reef shark populations, from direct top-down management of fishing to indirect improvement of governance conditions. Reef shark populations will only have a high chance of recovery by engaging key socio-economic aspects of tropical fisheries.

BET-bromodomain inhibition (BETi) has shown pre-clinical promise for MYC-amplified medulloblastoma. However, the mechanisms for its action, and ultimately for resistance, have not been fully defined. Here, using a combination of expression profiling, genome-scale CRISPR/Cas9-mediated loss of function and ORF/cDNA driven rescue screens, and cell-based models of spontaneous resistance, we identify bHLH/homeobox transcription factors and cell-cycle regulators as key genes mediating BETi’s response and resistance. Cells that acquire drug tolerance exhibit a more neuronally differentiated cell-state and expression of lineage-specific bHLH/homeobox transcription factors. However, they do not terminally differentiate, maintain expression of CCND2, and continue to cycle through S-phase. Moreover, CDK4/CDK6 inhibition delays acquisition of resistance. Therefore, our data provide insights about the mechanisms underlying BETi effects and the appearance of resistance and support the therapeutic use of combined cell-cycle inhibitors with BETi in MYC-amplified medulloblastoma.
