Global warming boosted by climate change affects grape quality, with increasing total soluble solids (TSS) content and decreasing total acidity (TA). However, current wine preferences increasingly include moderate alcohol content, higher acidity and the preservation of primary aromas reminiscent of grapes. Therefore, we hypothesised that applying phytohormones or mineral nutrients to leaves or carrying out defoliation can improve grape must properties in the face of climate warming and in accordance with current oenological trends. The effects of these three viticultural strategies were assessed independently from one another during three growing seasons in a Vitis vinifera L. cv. Tempranillo vineyard in northern Spain. Specifically, three 1-naphtaleneacetic acid (NAA) treatments, two early defoliations (ED; moderate and severe) and two foliar fertilisations with magnesium (Mg) were applied. Treatment with NAA was the most encouraging strategy for decreasing must TSS while increasing TA: it had slight effects on TSS in general and also slight effects on TA when applied close to veraison. The effects of the Mg treatments and moderate ED had null to slightly adverse effects. Finally, severe ED was clearly counter-productive. This study contributes to understanding the effects of both auxin and early defoliation treatments on grape must TSS, acidity and even yeast assimilable nitrogen (YAN) at harvest time. The favourable effects of NAA application are shown to be consistent though slight. Therefore, according to these results, the application of auxins may be an adequate choice for balancing sugars with acidity in grape musts. However, the results also suggest that more research needs to be undertaken to better characterise the effects of auxin treatments on grape must properties at harvest. In particular, different types of auxins, rates, concentrations and number of applications should be tested in the quest for more marked effects.

Using response surface methodology (RSM), this study investigates the effect of NaCl substitution (50%) with KCl, CaCl2, and MgCl2 in the packaging brines (controlled variables) on the characteristics (responses) of plain green Spanish-style Manzanilla olives, maintaining the salt-mixture level of 5%. The RSM showed that the increment of CaCl2 caused a linear significant (p-value ≤ 0.05) decrease in pH and a linear increase in firmness (instrumental), hardness (panel scores), and crunchiness. The models for bitterness and fibrousness also included quadratic (CaCl2·MgCl2) and cubic (the three salt) interactions, which led to areas of minimum and maximum scores around the central points of the CaCl2-MgCl2 and KCl-MgCl2 axes, respectively. In contrast, the increase in the KCl level linearly decreased bitterness scores. Optimisation resulted in a relatively low desirability (0.57) and the selection of a combination that may necessitate further refinement, such as increasing KCl or reducing CaCl2 levels, especially for markets sensitive to bitterness. Interestingly, the overall score and buying predisposition positively correlate with salty, smell, acid, and appearance and negatively with bitterness. Furthermore, PLS-R analysis found that the pivotal attributes influencing overall appreciation were smell and crunchiness while buying predisposition was promoted by crunchiness. Conversely, bitterness had a detrimental impact on these appreciations. Cluster analysis grouped the experimental runs into four categories, with sensory profiles predominantly diverging in bitterness, salty, and kinesthetic characteristics. Ultimately, this study elucidates four distinct sensory profiles that consumers experience.

Plasma potassium (p-K) in the high-normal range has been suggested to reduce risk of cardiovascular arrythmias and mortality through electrophysiological and mechanical effects on the myocardium. In this study, it was to investigated if increasing p-K to high-normal levels improves systolic- and diastolic myocardial function in patients with low-normal to moderately reduced left ventricular ejection fraction (LVEF). The study included 50 patients (mean age 58 years (SD 14), 81% men), with a mean p-K 3.95 mmol/l (SD 0.19), mean LVEF 48% (SD 7), and mean Global Longitudinal Strain (GLS) -14.6% (SD 3.1) patients with LVEF 35–55% from “Targeted potassium levels to decrease arrhythmia burden in high-risk patients with cardiovascular diseases trial” (POTCAST). Patients were given standard therapy and randomized (1:1) to an intervention that included guidance on potassium-rich diets, potassium supplements, and mineralocorticoid receptor antagonists targeting high-normal p-K levels (4.5-5.0 mmol/l). Echocardiography was done at baseline and after a mean follow-up of 44 days (SD 18) and the echocardiograms were analyzed for changes in GLS, mechanical dispersion, E/A, e’, and E/e’. At follow-up, mean difference in changes in p-K was 0.52 mmol/l (95%CI 0.35;0.69), P<0.001 in the intervention group compared to controls. GLS was improved with a mean difference in changes of -1.0% (-2;-0.02), P<0.05 and e’ and E/e’ were improved with a mean difference in changes of 0.9 cm/s (0.02;1.7), P = 0.04 and ? 1.5 (-2.9;-0.14), P = 0.03, respectively. Thus, induced increase in p-K to the high-normal range improved indices of systolic and diastolic function in patients with low-normal to moderately reduced LVEF.

The research study was conducted at the experimental farm of University for Development Studies, Nyankpala-Tamale in northern region, Ghana during the 2021 growing season to evaluate the effect of inorganic NPK fertilizer, Neem cake and neem oil as soil amendments on maize P uptake and grain yield. A 3 × 3 × 2 factorial experiment was laid out in a Randomized Complete Block Design with three replications. The treatments consisted of combinations of three levels of neem cake (0, 200 and 400 kg ha-1), three levels of neem oil (0, 10 and 20 L ha-1) and two levels of NPK (0 and 250 kg ha-1). Parameters examined were plant height, crop growth rate, chlorophil content (using spad), leaf area index (LAI), K uptake and grain yield. The study revealed plant height, uptake of K, grain yield and biological yield of maize increased with increased rate of neem cake (200 to 400 kg ha-1) when combined with 250 kg NPK/ha. Plant height, growth rate, uptake and uptake efficiency of K and grain yield of the crop decreased with increased rate of neem oil from 10 l/ha to 20 l/ha, when combined with 250 kg NPK/ha. The results showed combining 250 kg NPK/ha with 400 kg/ha neem cake increased K uptake by 34% (from 99 kg/ha to 133 kg/ha) compared to the recommended NPK rate. Application of 250 kg NPK/ha plus 400 kg/ha neem cake gave grain yield of 7637 kg/ha representing 61% increment over the application of only the inorganic fertilizer (4736 kg/ha), .and biological yield of 26873 kg/ha as maximum entries. The results of the study posited important implications for soil health management for enhanced maize production.

The electronic states producing the surface metallization have been studied for increasing K coverages up to room-temperature saturation. As K coverage increases, the interface undergoes a transition at a critical coverage, from a low-coverage semiconducting phase, to a saturation-coverage metallic phase. Two different surface states (F-1 and F-2) have been detected in the vicinity of the Fermi level. These two states are sequentially filled along the metallization process. The coverage dependence of both F-1 and F-2, and their symmetry properties indicate that the metallization is due to the filling of an initially empty surface band (appearance of F-2) We relate F-1 to the completion of K chains in the single-domain surface. Vineyard soils often contain mineralogy that can confound predictions of plant-available potassium (K). Too little or too much K uptake into wine-grape (Vitis varieties) tissues can negatively affect fruit chemistry, making it important to have soil tests that accurately quantify plant-available K. Our goal was to determine the best soil sampling, processing, and extraction methods for predicting K availability in vineyard soils. We sampled soil and grapevine tissue in 39 vineyard blocks from 22 vineyards in Virginia, Maryland, and New Jersey. Plant tissue sampling included petioles at bloom and both petioles and leaf blades at veraison. Soil samples were collected from 0 to 10- and 0 to 38-cm depths. We tested three soil extraction methods-Mehlich 1, Mehlich 3, and sodium tetraphenylboron (NaTPB)-on both oven-dried and field-moist samples. The different sampling, processing, and extracting procedures produced distinct K concentrations, with shallow 0-10 cm samples and NaTPB extraction resulting in higher K concentrations than their counterparts. Upon drying, three soils fixed K and 24 samples released K. Whole leaf (petiole plus leaf blade) samples collected at veraison had the best relationships with most soil K concentrations. The best soil testing method for predicting tissue K concentration in whole leaves at veraison was Mehlich 1 extractions of field-moist soils from 0 to 38-cm depth. This sampling combination appears to be best suited for growers to use when assessing K concentrations in vineyard soils.

Agricultural nutrient budgets help to identify an excess or an insufficiency in use of fertilizers and other nutrient sources. Nutrient budgets allow for indicators such as the nutrient balance (surplus or deficit) and nutrient use efficiency to be estimated. This can help in the monitoring of agricultural productivity and sustainability globally. The present dataset is a global database of country-level budget estimates for nitrogen (N), phosphorus (P) and potassium (K) in cropland. The database is disseminated in FAOSTAT and provides a global reference, synthesizing and continuously updating the state-of-the-art on this topic. The database covers the period 961 to 2020 for 205 countries and territories, as well as regional and global aggregates. Results indicate the wide range in nutrient use and use efficiencies across regions, nutrients, and time. This dataset introduces improvements over previous work in relation to key nutrient coefficients affecting nutrient budgets and use efficiency estimates. This is especially for nutrient removal in crop products, manure nutrient content, atmospheric deposition and crop biological N fixation rates.

Estimates of crop nutrient removal (as crop products and crop residues) are an important component of crop nutrient balances. Crop nutrient removal can be estimated through multiplication of the quantity of crop products or crop residues (removed) by the nutrient concentration of those crop products and crop residue components respectively. Data for quantities of crop products removed at a country level are available through FAOSTAT (https://www.fao.org/faostat/en/), but equivalent data for quantities of crop residues are not available at a global level. However, quantities of crop residues can be estimated if the relationship between quantity of crop residues and crop products is known. Harvest index (HI) provides one such indication of the relationship between quantity of crop products and crop residues. HI is the proportion of above-ground biomass as crop products and can be used to estimate quantity of crop residues based on quantity of crop products. Previously, meta-analyses or surveys have been performed to estimate nutrient concentrations of crop products and crop residues and harvest indices (collectively known as crop coefficients). The challenges for using these coefficients in global nutrient balances include the representativeness of world regions or countries. Moreover, it may be unclear which countries or crop types are actually represented in the analyses of data. In addition, units used among studies differ which makes comparisons challenging. To overcome these challenges, data from meta-analyses and surveys were collated in one dataset with standardised units and referrals to the original region and crop names used by the sources of data. Original region and crop names were converted into internationally recognised names, and crop coefficients were summarised into two Tiers of data, representing the world (Tier 1, with single coefficient values for the world) and specific regions or countries of the world (Tier 2, with single coefficient values for

Salinization of freshwater ecosystems due to anthropogenic sources will increasingly impact biodiversity. An example of point-source industrial salinization has occurred from historical activities at a U.S. Environmental Protection Agency Superfund Site near Saltville, Virginia USA and its associated chemical waste ponds adjacent to the North Fork Holston River. These point source discharges are documented contributors to mussel declines, partially due to high concentrations of chloride (Cl-, <= 26,000 mg Cl-/L) and potassium (K+, <= 97 mg K+/L). During a chronic 61-day laboratory study, Rainbow mussels, Villosa iris, were exposed to concentrations of Cl-(0, 416, 831, and 1,663 mg/L) and K+ (0, 4, 8, and 17 mg/L) to determine effects on survival and organ tissues. All test mussels died by day-2 in the 1,663 mg Cl-/L exposure, and 50% of mussels died by day-13 in the 17 mg K+/L concentration. Significantly greater abundances of tissue abnormalities were observed in digestive glands and kidneys with exposures to the 4 and 8 mg/L concentrations of K+ versus the control, and significantly greater abundances of lesions in kidneys were observed in the 416 and 831 mg Cl-/L concentrations compared to the control. The sublethal effects to digestive glands and kidneys were below reported effect (EC50, 20, 10 and LOEC) concentrations. Significant histological differences between control and baseline (day-0 sample) mussels were observed, suggesting the need for further study on the effects of captivity during longer-term laboratory experiments.

Under low-potassium (K+) stress, a Ca2+ signaling network consisting of calcineurin B-like proteins (CBLs) and CBL-interacting kinases (CIPKs) play essential roles. Specifically, the plasma membrane CBL1/9-CIPK pathway and the tonoplast CBL2/3-CIPK pathway promotes K+ uptake and remobilization, respectively, by activating a series of K+ channels. While the dual CBL-CIPK pathways enable plants to cope with low-K+ stress, little is known about the early events that link external K+ levels to the CBL-CIPK proteins. Here we show that K+ status regulates the protein abundance and phosphorylation of the CBL-CIPK-channel modules. Further analysis revealed low K+-induced activation of VM-CBL2/3 happened earlier and was required for full activation of PM-CBL1/9 pathway. Moreover, we identified CIPK9/23 kinases to be responsible for phosphorylation of CBL1/9/2/3 in plant response to low-K+ stress and the HAB1/ABI1/ABI2/PP2CA phosphatases to be responsible for CBL2/3-CIPK9 dephosphorylation upon K+-repletion. Further genetic analysis showed that HAB1/ABI1/ABI2/PP2CA phosphatases are negative regulators for plant growth under low-K+, countering the CBL-CIPK network in plant response and adaptation to low-K+ stress.

Multiple sclerosis (MS) is a progressive inflammatory demyelinating disease of the CNS. Increasing evidence suggests that vulnerable neurons in MS exhibit fatal metabolic exhaustion over time, a phenomenon hypothesized to be caused by chronic hyperexcitability. Axonal Kv7 (outward-rectifying) and oligodendroglial Kir4.1 (inward-rectifying) potassium channels have important roles in regulating neuronal excitability at and around the nodes of Ranvier. Here, we studied the spatial and functional relationship between neuronal Kv7 and oligodendroglial Kir4.1 channels and assessed the transcriptional and functional signatures of cortical and retinal projection neurons under physiological and inflammatory demyelinating conditions. We found that both channels became dysregulated in MS and experimental autoimmune encephalomyelitis (EAE), with Kir4.1 channels being chronically downregulated and Kv7 channel subunits being transiently upregulated during inflammatory demyelination. Further, we observed that pharmacological Kv7 channel opening with retigabine reduced neuronal hyperexcitability in human and EAE neurons, improved clinical EAE signs, and rescued neuronal pathology in oligodendrocyte-Kir4.1-deficient (OL-Kir4.1-deficient) mice. In summary, our findings indicate that neuron-OL compensatory interactions promoted resilience through Kv7 and Kir4.1 channels and identify pharmacological activation of nodal Kv7 channels as a neuroprotective strategy against inflammatory demyelination.

Hypokalemia is a commonly encountered electrolyte disturbance in clinical practice. As the predominant intracellular cation, potassium homeostasis ensures optimum cell functionality. Transient reductions in plasma potassium concentration depend on dietary intake and potassium excretion. An acceptable lower limit for serum potassium (K+) is 3.5 mmol/L. Although hypokalemia is well tolerated in healthy individuals, its severe cases can cause serious repercussions. The clinical manifestation of hypokalemia varies depending on the severity and duration of potassium loss from the body. Symptoms include heart palpitations, fatigue, muscle spasm, numbness, muscle twitches, and in severe cases cardiac arrhythmias and heart failure. The main route of K+ excretion is urine (90%) whereas a small amount (10%) is removed by GIT. External potassium losses are exacerbated in individuals with chronic diarrhea and prolonged sweating. Clinical history and examination are often sufficient to rule out the cause of hypokalemia. The overarching principle of hypokalemia therapy requires reducing potassium deficit, replenishing potassium reserves, and diagnosis and alleviation of causative factors to avoid future reoccurrences. Optimum recovery is reported in the majority of cases when underlying factors are eliminated. Potassium supplementation is paramount in all cases. The route of potassium administration should be based on the severity of the disease. Como el principal catión intracelular, la homeostasis del potasio asegura un funcionamiento celular óptimo. Reducciones transitorias en la concentración plasmática de potasio dependen de la ingesta dietética y su excreción. El límite inferior tolerable para la concentración de potasio sérico (K+) es 3.5 mmol/L. A pesar de que la hipocalemia es bien tolerada en individuos sanos, sus casos severos pueden causar repercusiones serias. Las manifestaciones clínicas de hipocalemia varían dependiendo de la severidad y la duración de la perdida de potasio del cuerpo. Síntomas incluyen palpitaciones cardiacas, fatiga, espasmos musculares, adormecimiento muscular y en casos severos arritmias cardiacas y falla cardiaca. La ruta principal de excreción de K+ es la orina (90%), mientras que un pequeño porcentaje (10%) es removido por el tracto gastrointestinal. Pérdidas externas de potasio se ven exacerbadas en individuos con diarrea crónica y sudoración prolongada. La historia clínica y el examen físico son usualmente suficientes para determinar la causa del déficit, reponer las reservas de potasio, diagnosticar y aliviar los factores causales para evitar futuras recurrencias. En la mayoría de las ocasiones, se reporta una recuperación óptima cuando los factores subyacentes son eliminados. Potasio suplementario resulta vital en todos los casos. La ruta de administración de potasio se debe basar en la severidad de la enfermedad. Quinoa is a crop with high economic and social potential in the Puno Region, Peru; however, its production is affected by low soil fertility. One way to deal with this problem, without affecting the environment, is through the use of beneficial microorganisms. The objective of the study was to evaluate the effect of Trichoderma sp. on N, P and K levels in plants and soil. The research was carried out in the Phytopathology Laboratory and greenhouse of the Professional School of Agricultural Engineering in the city of Puno, from January to July 2015. Five strains of Trichoderma sp. were inoculated in quinoa var. Salcedo-INIA plants by soil application or using pelleted seed. At the end of the crop cycle (six months), the levels of N, P and K were evaluated both in the plant tissue and in soil. No effects of Trichoderma inoculation on soil nutrient levels were detected, but the inoculated plants showed higher P and K contents than the control (p≤0.05). The results did not support the thesis of the solubilizing effect of Trichoderma on soil nutrients, but support the idea that the plant improved its efficiency in nutrient absorption attributed to the greater growth of the root system, La quinua es un cultivo con alto potencial económico y social en la Región Puno, Perú; sin embargo, su producción está afectada por la baja fertilidad de los suelos. Una forma de enfrentar este problema sin afectar el ambiente es mediante el uso de microorganismos benéficos. El objetivo de este estudio fue evaluar el efecto de cepas de Trichoderma sp. en la nutrición de la quinua var. Salcedo INIA y en las características químicas del suelo. La investigación se desarrolló en el Laboratorio de Fitopatología e instalaciones de la Escuela Profesional de Ingeniería Agronómica, ubicado en la ciudad de Puno durante los meses de enero a julio de 2015. Se emplearon cinco cepas de Trichoderma sp. para la inoculación en plantas de quinua mediante la aplicación al suelo o usando semilla peletizada. Al final del ciclo del cultivo (seis meses) se evaluaron los niveles de N, P y K tanto en el tejido vegetal como en el suelo. No se detectaron efectos de la inoculación con Trichoderma en los niveles de los nutrientes en el suelo, pero las plantas inoculadas mostraron contenidos de P y K superiores a los del testigo (P≤0.05). Los resultados no lograron sustentar la tesis del efecto solubilizador de Trichoderma sobre los nutrientes del suelo, pero sí la idea de que la planta mejoró su eficiencia en la absorción de nutrientes atribuido al mayor crecimiento del sistema radical.

Plant root traits play a crucial role in resource acquisition and crop performance when soil nutrient availability is low. However, the respective trait responses are complex, particularly at the field scale, and poorly understood due to difficulties in root phenotyping monitoring, inaccurate sampling, and environmental conditions. Here, we conducted a systematic review and meta-analysis of 50 field studies to identify the effects of nitrogen (N), phosphorous (P), or potassium (K) deficiencies on the root systems of common crops. Root length and biomass were generally reduced, while root length per shoot biomass was enhanced under N and P deficiency. Root length decreased by 9% under N deficiency and by 14% under P deficiency, while root biomass was reduced by 7% in N-deficient and by 25% in P-deficient soils. Root length per shoot biomass increased by 33% in N deficient and 51% in P deficient soils. The root-to-shoot ratio was often enhanced (44%) under N-poor conditions, but no consistent response of the root-to-shoot ratio to P-deficiency was found. Only a few K-deficiency studies suited our approach and, in those cases, no differences in morphological traits were reported. We encountered the following drawbacks when performing this analysis: limited number of root traits investigated at field scale, differences in the timing and severity of nutrient deficiencies, missing data (e.g., soil nutrient status and time of stress), and the impact of other conditions in the field. Nevertheless, our analysis indicates that, in general, nutrient deficiencies increased the root-length-to-shoot-biomass ratios of crops, with impacts decreasing in the order deficient P > deficient N > deficient K. Our review resolved inconsistencies that were often found in the individual field experiments, and led to a better understanding of the physiological mechanisms underlying root plasticity in fields with low nutrient availability., Peer Reviewed

In order to further close nutrient cycles of aquaponic systems, it could be possible to integrate a third trophic level in the form of insect larvae production (i.e., black soldier fly larvae) to recycle internal waste streams into valuable nutrients. This would present opportunities to formulate sustainable circular aquafeeds that combine these internally available nutrients with complementary external raw materials. The ingredient composition of feeds for such circular multitrophic food production systems (CMFS) may affect fish performance as well as excretion of important dissolved plant nutrients such as N, P and K. Hence, fish meal from catfish processing (CM) as base ingredient was combined with variable levels of poultry by-product meal (PM) and black soldier fly larvae meal (BSFM) into three marine-ingredient-free experimental diets corresponding to hypothetical production scenarios of a CMFS that aims to integrate aquaponics with insect larvae production. These experimental diets and a commercial diet (COM) were compared using isonitrogenous and isolipidic formulations. They were fed to African catfish (Clarias gariepinus) in recirculating aquaculture systems (RAS) and evaluated concerning growth performance and nutrient excretion. All diets resulted in similar total inorganic nitrogen (TIN) excretion, whereas the increase of dietary PM inclusion from 0% (BSF diet) to 20% (MIX diet) and to 41% (PM diet) and concomitant reduction of BSFM inclusion led to increasingly higher soluble reactive phosphorus (SRP) excretion per unit of feed compared to the COM diet. While the PM diet enabled the best growth and feed conversion performance, the MIX and especially the BSF diet produced more similar performance to the COM diet, which generated the highest dissolved K excretion. The MIX and the PM diet resulted in the highest Ca and P, yet lower N content in the fish feces. Syftet med denna studie var att undersöka om rörtransportsystemet på Universitetssjukhuset Örebro (USÖ) påverkar analysresultatet av kaliumprover och om förekomsten av hemolys ökade i kaliumprover som transporterades med rörpost. Studien utfördes med hjälp av direkt och indirekt potentiometrisk jonselektiv elektrodmetod (ISE) för att analysera nivåerna av kalium i plasma, samt spektrofotometri för att kvantifiera graden av hemolys i 129 prover som insamlades på akutmottagningen vid Universitetssjukhuset Örebro (USÖ). Ingen kliniskt relevant skillnad i hemolys eller kaliumkoncentration kunde påvisas i prover som transporterats med rörpost i jämförelse med manuell transport till laboratoriet. Samples were analyzed for hemolysis, potassium, and sodium using ion-selective electrode (ISE) methods. No clinically relevant difference in hemolysis or potassium concentration could be demonstrated in samples transported by pneumatic tube compared to manual transport to the laboratory. The use of tube transport is cost-effective and rapid despite the small difference observed, it should still be used. In conclusion, while hemolysis can affect analytes, the observed difference between tube and manual transport in this study had no clinical significance. Other factors such as different G- forces and disease states may have played a role in explaining divergent results.

Ultracold gases with strong interactions have been studied to a great extent with the help of Feshbach resonances, and have been used as precisely controllable models for other systems that are not easily accessible experimentally. Fermionic gases are especially interesting, because they can be used to simulate many-body physics present in primordial matter, neutron stars, atomic nuclei or condensed matter systems, particularly superconductors. Since interactions in cold fermions are in general only present if the particles are not identical, most experiments have been working with two spin states of a single atomic species. Changing the number ratio of the two spins can lead to interesting new pairing phenomena, some of which have been studied extensively in recent years. Mass-imbalanced systems have been theoretically predicted to exhibit new exotic interaction regimes and phases that go beyond the physics of spin mixtures. This thesis reports on the efforts to realize a strongly interacting mass-imbalanced Fermi-Fermi mixture of Dy and K that features collisional stability as well as tunability of interaction strength and trapping geometry. Feshbach resonances have become a ubiquitous tool to control the interaction strength in ultracold gases. With the identification of a broad Feshbach resonance close to 217 G, we were able to realize a resonantly interacting sample of Dy and K. A detailed characterization of the resonance was conducted to extract relevant parameters. In the expanding mixture, the resonant interspecies interaction causes a hydrodynamic behavior which leads to a bimodal density profile. The influence of mass-imbalance and other experimental parameters on the hydrodynamic expansion have been studied with the help of a Monte Carlo simulation, and the results have been found to be in good agreement with the experimental data. Biomass can be used to generate heat, power, or biofuels in thermal conversion processes such as combustion, gasification and pyrolysis. However, some types of biomass contain high levels of phosphorus, which can be released to the gas phase and cause operational or environmental problems. The mechanism(s) responsible for phosphorus release has not been convincingly established. Understanding the high-temperature phosphorus chemistry is also important in order to enable efficient recovery of phosphorus in residues from thermal conversion of biomass. In this work, the release of phosphorus from wheat bran char and sunflower seed char in different gas environments (100 % N2, 1–20 % O2, and 10 % CO2) and temperatures (900–1100 °C) was studied. The chars were converted in a horizontal tube reactor and characterized using ICP-OES, XRD, SEM-EDS, and 31P NMR. The release of ash-forming elements was determined using ICP-OES analysis of the char and sample residues, whereas the release of carbon was determined using CO and CO2 gas analysis. In both chars, phosphorus was present primarily together with potassium and magnesium, mainly as pyrophosphates in the wheat bran char, and largely as orthophosphates in the sunflower seed char. For wheat bran char, the release of phosphorus increased from 27 % at 900 °C to 71 % at 1100 °C in N2, whereas the release was at least 20 % lower in the oxidizing atmospheres (1–20 % O2, or 10 % CO2). The sunflower seed char reached a maximum release of 55 % at 1100 °C in N2. For wheat bran char, the molar ratio of released carbon/phosphorus was close to 2.5, which fits well with the theoretical value for carbothermic reduction of phosphates (P2O5(s, l) + 5C(s) → P2(g) + 5CO(g)). At 1100 °C, in N2, the release of phosphorus, potassium and sodium occurred mainly during the first 10 min. It was shown that KM

Potassium is one of the principal macronutrients required by all plants, but its mobility is restricted between soil compartments. Numerous studies have shown that Plant Growth Promoting Bacteria (PGPB) can facilitate nutrient uptake. The present work examined the effects of the PGPB (Bacillus megaterium) on rice plants subjected to potassium deprivation. To study only direct effects of B. megaterium, we first checked its lack of capacity to solubilize soil K. Rice plants were provided with 1.5 mM K (100%) or 0.015 mM K (1%) and growth related parameters, nutrient concentrations and gene expression of K transporters were determined. After two weeks, the 1% K treatment reduced growth of non-inoculated plants by about 50% compared with the 100% K treatment. However, there was no effect of reduced K nutrition on growth of inoculated plants. The reduction in growth in non-inoculated plants was accompanied by a similar reduction in K concentration in both roots and leaves and an overall 80% reduction of the plant potassium concentrations. In inoculated plants a 50% reduction occurred only in leaves. The expression of the K transporters HKT1;1, 1;2, 1;5, 2;2, 2;3 and 2;4 was up-regulated by the inoculation of B. megaterium under K deprivation conditions, explaining their higher K tissue concentrations and growth. Thus, the bacterial strain improved plant potassium nutrition without affecting K availability in the soil. The results demonstrate the potential of this bacteria for using as a biofertilizer to reduce the amount of potassium fertilizers to be applied in the field.

Last few decades, there has been a substantial advancement of geopolymer (GP) as a Portland cement substitute. It is vital to investigate potential building uses for geopolymer concrete (GPC). Six different mixes were cast for an alkaline to binder (A/B) ratio of 0.25-0.50 with an interval of 0.05. Metakaolin-based geopolymer were cured at ambient tem-perature and tested for 7, 14, 28, and 90 days. Metakaolin-Marble (MM00) mix was observed to have a maximum slump. For an A/B ratio of 0.35, maximum compressive, split tensile, flexural strength and modulus of elasticity was achieved. For elevated temperature resis-tance, geopolymer concrete cubes were exposed to temperatures (T) of 200, 400, to 600 C. As the temperature increased, compressive strength (CS) reduced. As the increase of the alkaline to binder (A/B) ratio, the strength of geopolymer concrete increases up to a specific limit beyond the limit strength decline. An empirical formula for split tensile (STS) value prediction using compressive strength values is proposed, valid for determining split tensile strength value. Pro zvýšenou teplotní odolnost byly geopolymerní betonové kostky vystaveny teplotám (T) 200, 400 až 600 C. Se zvyšující se teplotou se snižovala pevnost v tlaku (CS). Potassium ion (K+) plays a critical role as an essential electrolyte in all biological systems. Genetically-encoded fluorescent K+ biosensors are promising tools to further improve our understanding of K+-dependent processes under normal and pathological conditions. Here, we report the crystal structure of a previously reported genetically-encoded fluorescent K+ biosensor, GINKO1, in the K+-bound state. Using structure-guided optimization and directed evolution, we have engineered an improved K+ biosensor, designated GINKO2, with higher sensitivity and specificity. We have demonstrated the utility of GINKO2 for in vivo detection and imaging of K+ dynamics in multiple model organisms, including bacteria, plants, and mice.

The application of mechanically crushed silicate minerals to agricultural soils has been proposed as a method for both improving crop yields and sequestering inorganic carbon through enhanced mineral weathering. In Greenland, large quantities of finely grained glacial rock flour (GRF) are naturally produced by glacial erosion of bedrock and deposited in easily accessible lacustrine and marine deposits, without the need for energy-intensive grinding. To determine if this material can improve crop yields, we applied 10 and 50 t GRF ha−1 to a sandy, organic agricultural field in Denmark. Two field trials were carried out to test the first-year yield response to GRF in both maize and potatoes, residual effects on potato yields in the year after application, and second and third-year residual effects on spring wheat. Reference-K treatments were included for comparison to determine if the beneficial effects of GRF were primarily due to its K content (3.5% K2O). This alternative source of silicate minerals improved crop yields in the year of application. Though there was no improvement in yield with the reference-K treatments, for each additional ton of GRF applied, maize dry yield increased by 59 kg ha−1 and potato tuber yield by an additional 90 kg ha−1. No residual effects on crop yields were observed in the following years, but we suspect that benefits might persist over multiple seasons at sites with lower initial fertility. The increase in yields achieved with GRF could offset some of the costs of applying silicate minerals as a CO2 sequestration scheme.

Althought safflower is a tolerant crop against many environmental stresses, but its yield and performance reduce under stress. The aim of this experiment was to investigate the effect of silicon (Si) application on the possibility of increasing salinity resistance and related mechanisms in safflower. A greenhouse experiment was conducted to investigate the effects of Si spraying (0, 1.5 and 2.5 mM) on safflower plants grown under salt stress condition (non-saline and 10 dS m(-1)). Salinity reduced seedling emergence percent and rate, growth parameters and disrupted ion uptake but increased emergence time and specifc leaf weight. Spraying of Si increased plant height, fresh and dry weight, leaf area, relative water content (RWC), potassium, calcium and silicon content, while sodium absorption was decreased. As a result, the K+/Na+ and Ca2+/Na+ ratios were increased. Elevated ion contents and ratios indicate an enhanced selectivity of ion uptake following silicon application and may increase ion discrimination against Na+. Treatment with 2.5 mM Si showed the most positive effect on the measured growth traits. Decrement in leaf area ratio under salinity indicates a more severe effect of salinity on leaf area compared to biomass production. On the other hand, silicon reduced the specific leaf weight under stress and non-stress conditions, which revalues the positive effects of silicon on leaf area expansion. Improvement of RWC may a reason for the icrease in leaf area and biomass production. Data shows that spraying with Si especialy with 2.5 mM can reduce salinity stress damage to safflower and increase biomass production.

The aim of this study was to determine the content of physiologically active potassium (K2O) in the soil in the territory covered by the work of the Agricultural Advisory and Expert Service of Smederevo (PSSS Smederevo). Research was carried out on three locations: Velika Plana, the City of Smederevo and Smederevska Palanka, for the purpose of ensuring the optimal use of mineral and organic fertilizers, to preserve soil resources. The paper presents the results of testing the content of physiologically active potassium – K2O. The territory of Velika Plana had most areas with harmful content, while the lowest content of K2O was recorded in the territory of Smederevska Palanka. The results indicate that the obtained values were greatly influenced by different types of soil on the examined plots, and an anthropogenic factor was also evident. All the plots from which soil samples were taken for analysis of potassium content were mapped, and based on the obtained results farmers were given advice on the necessary remedial measures and optimal use of organic and mineral fertilizers to improve agricultural production and preserve biodiversity. BackgroundThe KBindER (K+ Binders in Emergency Room and hospitalized patients) clinical trial is the first head-to-head evaluation of oral potassium binders (cation-exchange resins) for acute hyperkalemia therapy.MethodsEmergency room and hospitalized patients with a blood potassium level ≥ 5.5 mEq/L are randomized to one of four study groups: potassium binder drug (sodium polystyrene sulfonate, patiromer, or sodium zirconium cyclosilicate) or nonspecific laxative (polyethylene glycol). Exclusion criteria include recent bowel surgery, ileus, diabetic ketoacidosis, or anticipated dialysis treatment within 4 h of treatment drug. Primary endpoints include change in potassium level at 2 and 4 h after treatment drug. Length of hospital stay, next-morning potassium level, gastrointestinal side effects and palatability will also be analyzed. We are aiming for a final cohort of 80 patients with complete data endpoints (20 per group) for comparative statistics including multivariate adjustment for kidney function, diabetes mellitus, congestive heart failure, metabolic acidosis, renin-angiotensin-aldosterone system inhibitor prescription, and treatment with other agents to lower potassium (insulin, albuterol, loop diuretics).DiscussionThe findings from our study will inform decision-making guidelines on the role of oral potassium binders in the treatment of acute hyperkalemia.Trial registrationClinicalTrials.gov Identifier: NCT04585542 . Current ware potato yields in the Netherlands are approximately 70% of their potential. It is hypothesised by several stakeholders within the potato value chain that part of the potato yield gap is caused by a lack of phosphorus (P) and potassium (K) application. In this study, we analysed for 46 farmers’ fields if increasing P and K fertiliser application rates led to a higher yield and yield quality. We found that, on average, increased P and K fertiliser application did not result in a significantly higher yield for two currently cropped potato cultivars on two different soil types in the Netherlands (Innovator on clay soils and Fontane on sandy soils) and in two years (2019 and 2020). However, on sandy soils at relatively lower farmer K application rates, our K application led to a small positive yield response up to 5 t ha−1. On clay soils, there was an average positive yield response to our K application at lower yield levels of the control. For P, we did not find any correlation between yield response to P application and the amount of P applied by farmers or any of the measured soil parameters. In terms of yield quality, the K application led to a slight reduction in underwater weight on sandy soils in 2019 and a slight increase in the yield of large tubers in 2020. We conclude that, although in some fields there was a small positive yield effect of increased K application, increasing P and K application rates will not narrow the potato yield gap and improve potato yield quality in the Netherlands. Instead, increasing the P and K application will decrease P and K use efficiency and hence is not recommended from an environmental and economic perspective.

Exhaled breath condensate (EBC) analysis is a recently developed, non-invasive method used to identify and quantify biomarkers, mainly those coming from the lower respiratory tract. It seems that diet can influence the airway’s inflammation and change the exhaled breath composition. This study aimed to assess the association between diet quality intake and markers in EBC among school-aged children. A cross-sectional analysis included 150 children (48.3% females, aged 7–12 years, mean age: 8.7 ± 0.8 years) from 20 schools across Porto, Portugal. We assessed diet quality through the Healthy Eating Index (HEI)-2015, which was estimated based on a single 24 h food recall questionnaire. EBC samples were collected, and we assessed their ionic content (Na+, K+) and conductivity. The association between diet quality and Na+, K+, Na+/K+ ratio and conductivity was estimated using logistic regression models adjusted for potential confounders. After adjustment, a higher quality diet score increases the odds of higher conductivity values of the EBC (aOR = 1.04, 95%CI 1.00; 1.08). Our findings suggest that a higher diet quality in school-aged children is associated with higher conductivity levels of the EBC.

The dormant state of bacterial spores is generally thought to be devoid of biological activity. We show that despite continued dormancy, spores can integrate environmental signals over time through a preexisting electrochemical potential. Specifically, we studied thousands of individual Bacillus subtilis spores that remain dormant when exposed to transient nutrient pulses. Guided by a mathematical model of bacterial electrophysiology, we modulated the decision to exit dormancy by genetically and chemically targeting potassium ion flux. We confirmed that short nutrient pulses result in step-like changes in the electrochemical potential of persistent spores. During dormancy, spores thus gradually release their stored electrochemical potential to integrate extracellular information over time. These findings reveal a decision-making mechanism that operates in physiologically inactive cells.

For persons with proteinuria, angiotensin-converting enzyme inhibitors (ACEis) and angiotensin II receptor blockers (ARBs) are treatment mainstays for reducing kidney disease progression. Guidelines for managing hypertension and chronic kidney disease recommend titrating to the maximum ACEi/ARB dose tolerated. Using deidentified national electronic health record data from the Optum Labs Data Warehouse, we examined ACEi/ARB dosing among adults with proteinuria-defined as either a urine albumin to creatinine ratio of 30 mg/g or greater or a protein to creatinine ratio of 150 mg/g or greater-who were prescribed an ACEi/ARB medication between January 1, 2017, and December 31, 2018. Among 100,238 included patients (mean age, 65.1 years; 49,523 [49.4%] female), 29,883 (29.8%) were taking maximal ACEi/ARB doses. Among 74,287 patients without potential contraindications to dose escalation (systolic blood pressure <120 mm Hg, estimated glomerular filtration rate <15 mL/min per 1.73 m2, serum potassium level greater than 5.0 mEq/L, or acute kidney injury within the prior year), the frequency of maximal ACEi/ARB dosing was 32.3% (24,025 patients). In adjusted analyses, age less than 40 years, female sex, Hispanic ethnicity, lower urine albumin to creatinine ratio, lack of diabetes, heart failure, lower blood pressure, higher serum potassium level, and prior acute kidney injury were associated with lower odds of maximal ACEi/ARB dosing. Having a prior nephrologist visit was not associated with maximal dosing. Our results suggest that greater attention toward optimizing the dose of ACEi/ARB therapy may represent an opportunity to improve chronic kidney disease care and reduce excess morbidity and mortality associated with disease progression.

Aims/Introduction: We evaluated the effect of co-administration of esaxerenone and a sodium–glucose cotransporter 2 (SGLT2) inhibitor on the magnitude of serum potassium elevation in Japanese patients with diabetic kidney disease. Materials and Methods: We carried out a prespecified subanalysis of data from two phase III studies: a multicenter, randomized, double-blind, placebo-controlled trial in patients with type 2 diabetes and microalbuminuria (J308); and a multicenter, single-arm, open-label trial in patients with type 2 diabetes and macroalbuminuria (J309). Changes in serum potassium levels during the studies and other measures were evaluated according to SGLT2 inhibitor use. Results: In both studies, time-course changes in serum potassium levels, and incidence rates of serum potassium elevation were lower in patients with co-administration of SGLT2 inhibitor in both the placebo and esaxerenone groups than those without the inhibitor. In contrast, time-course changes and mean percentage changes from baseline in urinary albumin-to-creatinine ratio, the proportion of patients with albuminuria remission and time-course changes in blood pressure did not change with or without SGLT2 inhibitor, whereas the albumin-to-creatinine ratio and blood pressure were reduced with esaxerenone. The blood glucose-lowering effect of SGLT2 inhibitor was not affected by esaxerenone. Conclusions: In Japanese patients with type 2 diabetes and albuminuria treated with esaxerenone, concomitant use of SGLT2 inhibitor reduced the magnitude of serum potassium elevation without any change of its antihypertensive and albuminuria-suppressing effects. Co-administration of esaxerenone and SGLT2 inhibitor might be a beneficial treatment option for patients with diabetic kidney disease.

Background and objectives Hyperkalemia after starting renin-angiotensin system inhibitors has been shown to be subsequently associated with a higher risk of cardiovascular and kidney outcomes. However, whether to continue or discontinue the drug after hyperkalemia remains unclear. Design, setting, participants, & measurements Data came from the Action in Diabetes and Vascular Disease: Preterax and Diamicron Modified Release Controlled Evaluation (ADVANCE) trial, which included a run-in period where all participants initiated angiotensin-converting enzyme inhibitor–based therapy (a fixed combination of perindopril and indapamide). The study population was taken as patients with type 2 diabetes with normokalemia (serum potassium of 3.5 to,5.0 mEq/L) at the start of run-in. Potassium was remeasured 3 weeks later when a total of 9694 participants were classified into hyperkalemia (≥5.0 mEq/L), normokalemia, and hypokalemia (,3.5 mEq/L) groups. After run-in, patients were randomized to continuation of the angiotensin-converting enzyme inhibitor–based therapy or placebo; major macrovascular, microvascular, and mortality outcomes were analyzed using Cox regression during the following 4.4 years (median). Results During active run-in, 556 (6%) participants experienced hyperkalemia. During follow-up, 1505 participants experienced the primary composite outcome of major macrovascular and microvascular events. Randomized treatment of angiotensin-converting enzyme inhibitor–based therapy significantly decreased the risk of the primary outcome (38.1 versus 42.0 per 1000 person-years; hazard ratio, 0.91; 95% confidence interval, 0.83 to 1.00; P=0.04) compared with placebo. BackgroundRefeeding hypophosphatemia (RH) in individuals with anorexia nervosa (AN) is a potentially fatal complication of nutrition restoration; yet, little is known about risk. This retrospective cohort study examined factors found in hospitalized youth with AN that may contribute to RH.MethodsWe reviewed medical records of 300 individuals diagnosed with AN admitted between the years of 2010 and 2016. Logistic regression examined factors associated with RH. Multivariate regression examined factors associated with phosphorus nadir.ResultsFor 300 participants, the mean (SD) age was 15.5 (2.5) years, 88.3% were White, and 88.3% were female. Participants lost an average of 11.3 (9.7) kg of body weight and were 82% (12.1) of median body mass index (BMI). Age (P = .022), nasogastric (NG) tube feeding (P = .054), weight gain (P = .003), potassium level (P = .001), and magnesium level (P = .024) were contributors to RH. Odds of RH were 13.7 times higher for each unit reduction in magnesium, 9.2 times higher for each unit reduction in potassium, three times higher in those who received NG feeding, 1.5 times higher for each kg of weight gain, and 1.2 times higher for each year of age. Regarding phosphorus nadir, serum magnesium level (P < .001) and admission BMI (P = .002) contributed significantly.ConclusionThe results indicate that age, NG feeding, weight gain, electrolyte abnormalities, and BMI on admission are potential indicators of the development of RH in youth. This study identifies clinical risk factors associated with RH and may guide further investigation.

Plants obtain nutrients from the soil via transmembrane transporters and channels in their root hairs, from which ions radially transport in toward the xylem for distribution across the plant body. We determined structures of the hyperpolarization-activated channel AKT1 from Arabidopsis thaliana, which mediates K+ uptake from the soil into plant roots. These structures of AtAKT1 embedded in lipid nanodiscs show that the channel undergoes a reduction of C4 to C2 symmetry, possibly to regulate its electrical activation.

We report two structures of the human voltage-gated potassium channel (Kv) Kv1.3 in immune cells alone (apo-Kv1.3) and bound to an immunomodulatory drug called dalazatide (dalazatide-Kv1.3). Both the apo-Kv1.3 and dalazatide-Kv1.3 structures are in an activated state based on their depolarized voltage sensor and open inner gate. In apo-Kv1.3, the aromatic residue in the signature sequence (Y447) adopts a position that diverges 11 Å from other K+ channels. The outer pore is significantly rearranged, causing widening of the selectivity filter and perturbation of ion binding within the filter. This conformation is stabilized by a network of intrasubunit hydrogen bonds. In dalazatide-Kv1.3, binding of dalazatide to the channel's outer vestibule narrows the selectivity filter, Y447 occupies a position seen in other K+ channels, and this conformation is stabilized by a network of intersubunit hydrogen bonds. These remarkable rearrangements in the selectivity filter underlie Kv1.3's transition into the drug-blocked state.

BackgroundHyperkalemia (HK) may be associated with poor clinical outcomes among COVID-19 patients. This study aimed to describe the prevalence of HK and evaluate the associations between HK and in-hospital mortality, intensive care unit (ICU) admission, length of hospital stay (LOS), and hospitalization cost among COVID-19 inpatients.MethodsA retrospective cohort study was conducted using a large hospital discharge database (PINC AI Healthcare Database) for COVID-19 inpatients discharged between April 1 and August 31, 2020. HK was defined with discharge diagnosis and potassium binder use.ResultsOf 192,182 COVID-19 inpatients, 12% (n = 22,702) had HK. HK patients were more likely to be older (median age 67 vs 63 years), male (63% vs 50%), black (30% vs 22%), and have a history of chronic kidney disease (45% vs 16%) or diabetes mellitus (55% vs 35%) than non-HK patients (all p<.001). A significantly higher proportion of patients with HK had in-hospital mortality (42% vs 11%, p<.001) than those without HK; this was persistent after adjusting for confounders (adjusted odds ratio [aOR] 1.69, 95% confidence interval [CI]1.62-1.77). Patients with HK were also more likely to be admitted to ICU (aOR 1.05, 95% CI 1.01-1.09), incur higher cost of care (adjusted mean difference $5,389) and have longer LOS (adjusted mean difference 1.3 days) than non-HK patients.ConclusionsPresence of HK was independently associated with higher in-hospital mortality, LOS, and cost of care among COVID-19 inpatients. Detecting and closely monitoring HK are recommended to improve clinical outcomes and reduce LOS and healthcare cost among COVID-19 patients.

Background Structural and electrophysiological remodeling characterize heart failure (HF) enhancing arrhythmias. PKD1 (protein kinase D1) is upregulated in HF and mediates pathological hypertrophic signaling, but its role in K+ channel remodeling and arrhythmogenesis in HF is unknown. Methods and Results We performed echocardiography, electrophysiology, and expression analysis in wild-type and PKD1 cardiomyocyte-specific knockout (cKO) mice following transverse aortic constriction (TAC). PKD1-cKO mice exhibited significantly less cardiac hypertrophy post-TAC and were protected from early decline in cardiac contractile function (3 weeks post-TAC) but not the progression to HF at 7 weeks post-TAC. Wild-type mice exhibited ventricular action potential duration prolongation at 8 weeks post-TAC, which was attenuated in PKD1-cKO, consistent with larger K+ currents via the transient outward current, sustained current, inward rectifier K+ current, and rapid delayed rectifier K+ current and increased expression of corresponding K+ channels. Conversely, reduction of slowly inactivating K+ current was independent of PKD1 in HF. Acute PKD inhibition slightly increased transient outward current in TAC and sham wild-type myocytes but did not alter other K+ currents. Sham PKD1-cKO versus wild-type also exhibited larger transient outward current and faster early action potential repolarization. Tachypacing-induced action potential duration alternans in TAC animals was increased and independent of PKD1, but diastolic arrhythmogenic activities were reduced in PKD1-cKO. Conclusions Our data indicate an important role for PKD1 in the HF-related hypertrophic response and K+ channel downregulation. Therefore, PKD1 inhibition may represent a therapeutic strategy to reduce hypertrophy and arrhythmias; however, PKD1 inhibition may not prevent disease progression and reduced contractility in HF.

Oxytocin (OXT) and OXT receptor (OXTR)-mediated signaling control excitability, firing patterns, and plasticity of hippocampal CA2 pyramidal neurons, which are pivotal in generation of brain oscillations and social memory. Nonetheless, the ionic mechanisms underlying OXTR-induced effects in CA2 neurons are not fully understood. Using slice physiology in a reporter mouse line and interleaved current-clamp and voltage-clamp experiments, we systematically identified the ion channels modulated by OXT signaling in CA2 pyramidal cells (PYRs) in mice of both sexes and explored how changes in channel conductance support altered electrical activity. Activation of OXTRs inhibits an outward potassium current mediated by inward rectifier potassium channels (I Kir) and thus favoring membrane depolarization. Concomitantly, OXT signaling also diminishes inward current mediated by hyperpolarization-activated cyclic-nucleotide-gated (HCN) channels (I h), providing a hyperpolarizing drive. The combined reduction in both I Kir and I h synergistically elevate the membrane resistance and favor dendritic integration while the membrane potential is restrained from quickly depolarizing from rest. As a result, the responsiveness of CA2 PYRs to synaptic inputs is highly sharpened during OXTR activation. Unexpectedly, OXTR signaling also strongly enhances a tetrodotoxin-resistant (TTX-R), voltage-gated sodium current that helps drive the membrane potential to spike threshold and thus promote rhythmic firing. This novel array of OXTR-stimulated ionic mechanisms operates in close coordination and underpins OXT-induced burst firing, a key step in CA2 PYRs' contribution to hippocampal information processing and broader influence on brain circuitry. BACKGROUND: In healthy adults, higher dietary potassium intake is recommended given that potassium-rich foods are major sources of micronutrients, antioxidants, and fiber. Yet among patients with advanced kidney dysfunction, guidelines recommend dietary potassium restriction given concerns about hyperkalemia leading to malignant arrhythmias and mortality. OBJECTIVES: Given sparse data informing these recommendations, we examined associations of dietary potassium intake with mortality in a nationally representative cohort of adults from the NHANES. METHODS: We examined associations between daily dietary potassium intake scaled to energy intake (mg/1000 kcal), ascertained by 24-h dietary recall, and all-cause mortality among 37,893 continuous NHANES (1999-2014) participants stratified according to impaired and normal kidney function (estimated glomerular filtration rates <60 and ≥60 mL · min-1 · 1.73 m-2, respectively) using multivariable Cox models. We also examined the impact of the interplay between dietary potassium, source of potassium intake (animal- compared with plant-based sources), and coexisting macronutrient and mineral consumption upon mortality. RESULTS: Among participants with impaired and normal kidney function, the lowest tertile of dietary potassium scaled to energy intake was associated with higher mortality (ref: highest tertile) [adjusted HR (aHR): 1.18; 95% CI: 1.02, 1.38 and aHR: 1.17; 95% CI: 1.06, 1.28, respectively]. Compared with high potassium intake from plant-dominant sources, participants with low potassium intake from animal-dominant sources had higher mortality irrespective of kidney function. Among participants with impaired kidney function, pairings of low potassium intake with high protein, low fiber, or high phosphorus consumption were each associated with higher death risk. ATP-sensitive potassium (KATP) channels couple the intracellular ATP concentration to insulin secretion. KATP channel activity is inhibited by ATP binding to the Kir6.2 tetramer and activated by phosphatidylinositol 4,5-bisphosphate (PIP2). Here, we use molecular dynamics simulation, electrophysiology and fluorescence spectroscopy to show that ATP and PIP2 occupy different binding pockets that share a single amino acid residue, K39. When both ligands are present, simulations suggest that K39 shows a greater preference to co-ordinate with PIP2 than with ATP. They also predict that a neonatal diabetes mutation at K39 (K39R) increases the number of hydrogen bonds formed between K39 and PIP2, potentially accounting for the reduced ATP inhibition observed in electrophysiological experiments. Our work suggests that PIP2 and ATP interact allosterically to regulate KATP channel activity. (Figure presented.). Background Patients with Type 1 diabetes mellitus have been shown to be at a two to ten-fold higher risk of sudden cardiac death (SCD) (Svane et al., Curr Cardiol 2020; 22:112) than the general population, but the underlying mechanism is unclear. Hyperglycaemia is a recognised cause of QTc prolongation; a state patients with type 1 diabetes are more prone to, potentially increasing their risk of ventricular arrhythmia. Understanding the QTc prolongation effect of both hyperglycaemia and the concomitant additive risk of commonly prescribed QTc-prolonging drugs such as Moxifloxacin may help to elucidate the mechanism of sudden cardiac death in this cohort. This single-blinded, placebo-controlled study investigated the extent to which hyperglycaemia prolongs the QTc in controlled conditions, and the potential additive risk of QTc-prolonging medications.Methods 21 patients with type 1 diabetes mellitus were enrolled to a placebo-controlled crossover study at a single clinical trials unit. Patients underwent thorough QTc assessment throughout the study. A 'hyperglycaemic clamp' of oral and intravenous glucose was administered with a target blood glucose of > 25 mM and maintained for 2 h on day 1 and day 3, alongside placebo on day 1 and moxifloxacin on day 3. Day 2 served as a control day between the two active treatment days. Thorough QTc assessment was conducted at matched time points over 3 days, and regular blood sampling was undertaken at matched time intervals for glucose levels and moxifloxacin exposure.Results Concentration-effect modelling showed that acute hyperglycaemia prolonged the QTc interval in female and male volunteers with type 1 diabetes by a peak mean increase of 13 ms at 2 h. Cilj rada bio je da se ispita da li potapanje semena može da poboljša klijavost semena soje u nepovoljnim uslovima, odnosno na nižoj temperaturi. Za istraživanje je korišćeno seme sorte soje NS Apolo, koja je selekcionisana na Institutu za ratarstvo i povrtarstvo u Novom Sadu. Seme je potapano u različite rastvore: kalijum nitrat - KNO3 (1%), askorbinska kiselina - AsA (100 mg l-1) i kalijum hlorid - KCl (1%). Nakon sušenja, seme je naklijavano na temperaturi 10°C, dok je seme naklijavano na optimalnoj temperaturi od 25°C poslužilo kao kontrola. Rezultati istraživanja su pokazali da je potapanje semena pre setve poboljšalo energiju klijanja, klijavost semena i brzinu klijanja u uslovima niske temperature. Upotreba rastvora KNO3 i AsA bila je efekasnija od primene rastvora KCl u uslovima niže temperature. Rastvor KNO3 i AsA su značajnije uticali na povećanje klijavosti i energije klijanja., The aim of this study was to examine whether priming seeds can improve the germination of soybean seeds in unfavorable conditions, i.e. at a lower temperature. For the research, the seeds of the NS Apolo soybean variety, which were selected at the Institute of Agriculture and Vegetables in Novi Sad, were used. The seeds were primed in different solutions: potassium nitrate - KNO3 (1%), ascorbic acid - AsA (100 mg l- 1) and potassium chloride - KCl (1%). After drying, the seeds were germinated at a temperature of 10°C, while the seeds germinated at an optimal temperature of 25°C served as a control. The research results showed that priming the seeds before sowing improved germination energy, seed germination and germination rate under low temperature conditions. The use of KNO3 and AsA solution was more effective than the use of KCl solution under lower temperature conditions. The solution of KNO3 and AsA significantly influenced the increase in germination and germination energy.

Reducing salt intake is one of the most cost-effective interventions to improve population health due to the subsequent reductions in blood pressure. Countries are introducing programs to lower salt consumption. Such programs usually focus on reducing salt in packaged foods and meals alongside campaigns to change consumer behavior. Thus, this paper provides an overview of the rationale for and evidence supporting the use of salt substitutes. Cur-rent approaches to salt reduction are insufficient, and more innovative solutions need to be identified. There is good evidence that salt substitutes, where some of the sodium is replaced with potassium, are effective to lower sodium total intake. The main challenge is to understand the pathways to market for salt substitutes. How do we implement programs to promote salt substitutes in different countries? What levels of government intervention are required? With more research and government investment, salt substitutes could be a game changer for increasing the impact of strategies to reduce population salt intake.

The public health issue of chronic kidney disease (CKD) is especially acute in India. Only 10% of patients with incident end stage renal disease (ESRD) cases receive therapy in India due to limited availability and expensive costs. To best manage CKD, ensure a sufficient intake of protein and energy, prevent or correct protein-energy wasting, and optimise nutritional management of CKD, a balanced and personalised low protein diet (LPD) approach should be developed. The relationship between mineral intake and chronic renal disease has only been briefly studied (CKD). In a cross-sectional analysis, the Health Examinee (HEXA) cohort of the Korean Genome and Epidemiologic Study was used to examine the relationship between mineral intake (calcium, phosphorus, sodium, potassium, iron, and zinc) and CKD (KoGES). A food frequency questionnaire was used to determine the mineral consumption of 159,711 participants. An estimated glomerular filtration rate (eGFR) of less than 60 mL/min/1.73 m2 was used to characterise CKD. Each mineral's dietary intake was separated into quartiles, with the quartile containing the acceptable intake (AI) or recommended dietary allowance (RDA) of each mineral serving as the reference. As a result, every effort should be taken to stop the progression of CKD. The study has shown to require analyses of food variance, nutritional demand changes, and variation in dietary intake in order to strengthen the links between nutrient consumption and renal function. This review article examines the function of low protein diets (LPD) in the treatment of CKD patients and offers advice on how to use them in clinical settings. It also discusses the relationship between inadequate mineral consumption and the prevalence of CKD in the general population.

The concerted function of the large number of ion channels expressed in excitable cells, including brain neurons, shapes diverse signaling events by controlling the electrical properties of membranes. It has long been recognized that specific groups of ion channels are functionally coupled in mediating ionic fluxes that impact membrane potential, and that these changes in membrane potential impact ion channel gating. Recent studies have identified distinct sets of ion channels that can also physically and functionally associate to regulate the function of either ion channel partner beyond that afforded by changes in membrane potential alone. Here, we review canonical examples of such ion channel partnerships, in which a Ca2+ channel is partnered with a Ca2+-activated K+ channel to provide a dedicated route for efficient coupling of Ca2+ influx to K+ channel activation. We also highlight examples of non-canonical ion channel partnerships between Ca2+ channels and voltage-gated K+ channels that are not intrinsically Ca2+ sensitive, but whose partnership nonetheless yields enhanced regulation of one or the other ion channel partner. We also discuss how these ion channel partnerships can be shaped by the subcellular compartments in which they are found and provide perspectives on how recent advances in techniques to identify proteins in close proximity to one another in native cells may lead to an expanded knowledge of other ion channel partnerships.

Analytical data of Roman and early Islamic glass established several primary glass production groups linked to glassmaking centres in the Levant and in Egypt. In contrast, the activities of secondary glass workshops are largely invisible in the compositional fingerprint of first millennium glass. Laser ablation inductively coupled plasma mass spectrometry (LA-ICP-MS) of 261 glass finds from the Visigothic settlement of Tolmo de Minateda (Spain) revealed a site-specific contamination pattern due to secondary glass processing and recycling, namely the enrichment of the glass batch by a unique combination of rare alkali elements (Li, K, Rb, Cs). With a median of 21 ppm, Li is particularly distinctive. Elevated lithium contents (Li>30 ppm) are also one of the characteristic features of Iberian plant ash glass from the Islamic period. The earliest known examples of this type of glass were found among the ninth-century remains from Tolmo.

As part of the NL Next Level manure valorization project, research has been carried out into emissions, mass balances and costs for various manure processing options on cattle farms. An emission model and a mass balance and cost estimate for manure processing have been drawn up for this purpose. Scenarios have been calculated, mainly looking at different housing systems (faster manure removal, emission-reducing measures and separation of urine and faeces) in combination with and without manure processing, and their effects on ammonia and greenhouse gas emissions from excretion of manure to the application of manure products., In het kader van het project NL Next Level mestverwaarden is onderzoek uitgevoerd naar emissies, massabalansen en kosten voor verschillende mestbewerkingsopties op rundveebedrijven. The maintenance of intracellular nitrogen-fixing bacteria causes changes in proteins' location and in gene expression that may be detrimental to the host cell fitness. We hypothesized that the nodule's high vulnerability toward salt stress might be due to alterations in mechanisms involved in the exclusion of Na+ from the host cytoplasm. Confocal and electron microscopy immunolocalization analyses of Na+/K+ exchangers in the root nodule showed the plasma membrane (MtNHX7) and endosome/tonoplast (MtNHX6) signal in non-infected cells; however, in mature infected cells the proteins were depleted from their target membranes and expelled to vacuoles. This mistargeting suggests partial loss of the exchanger's functionality in these cells. In the mature part of the nodule 7 of the 20 genes encoding ion transporters, channels, and Na+/K+ exchangers were either not expressed or substantially downregulated. In nodules from plants subjected to salt treatments, low temperature-scanning electron microscopy and X-ray microanalysis revealed the accumulation of 5-6 times more Na+ per infected cell versus non-infected one. Hence, the infected cells' inability to withstand the salt may be the integral result of preexisting defects in the localization of proteins involved in Na+ exclusion and the reduced expression of key genes of ion homeostasis, resulting in premature senescence and termination of symbiosis.
