Im Jahr 2017 - 2018 wurden in Cottbus zwei solare Mehrfamilienhäuser (je 7 Wohneinheiten) gebaut und seit 2019 im bewohnten Zustand im Rahmen des Eversol-Projektes durch die TU Bergakademie Freiberg wissenschaftlich begleitet. Neben dem Energiekonzept (Solarthermie und Photovoltaik) war auch das Bewohnerverhalten aufgrund einer Pauschalmiete (Festpreis für Wohnen, Strom, Wärme und Kälte) von Interesse. Die im Detail untersuchten Gebäude in Cottbus erreichen die hohen planerischen solaren Deckungsgrade in der elektrischen Energieversorgung sehr gut (70 % - 77 %). Das Stromverbrauchsprofil ist vergleichbar mit Literaturwerten und weist keine Auffälligkeiten aufgrund von Pauschalmiete auf. Durch deutlichen Mehrverbrauch in der Wärmeversorgung wird der solare Wärmedeckungsgrad allerdings verfehlt (50 % - 55 %). Gründe sind hier neben einem hohen Komfortanspruch der Bewohner (hohe Raumtemperaturen) u. a. auch in einer fehlerhaften Thermostateinstellung und hohen Warmwasser-Vorlauftemperaturen zu sehen. Insgesamt weisen die Monitoringergebnisse jedoch nicht auf ein verschwenderisches Verhalten aufgrund der Pauschalmiete hin. Für zukünftige Projekte wird deutlich, dass sowohl elektrischer, als auch thermischer Speicher deutlich kleiner dimensioniert werden könnten, ohne einen großen Einfluss auf die Autarkie zu haben. Der Erfolg der Auskopplung von Strom- und Wärmeüberschüssen in das umliegende Quartier konnte nachgewiesen werden. Es besteht jedoch noch weiteres Potenzial durch eine intelligente Steuerung. Die in diesem Forschungsprojekt mit umgesetzte pauschalisierte Inklusivmiete / Pauschalmiete kann als ein wesentlicher zukünftiger Anreizfaktor für ein Umdenken im Neubau von gesetzlichen, energetischen Pflichtvorgaben hin zu ökologisch-wirtschaftlichen Lösungen, von denen Mieter wie Vermieter profitieren, angesehen werden. In addition to the high comfort requirements of the occupants (high room temperatures), this is e. g. also due to incorrect thermostat settings and high hot water temperatures. On the whole, however, the monitoring results do not indicate any wasteful behaviour as a result of the flat-rate rent. For future projects, it is clear that both the electrical and thermal storage tanks could be significantly smaller without having a major impact on self-sufficiency. The success of exporting surplus electricity and heat to the surrounding neighbourhood has been proven, but there is further potential through intelligent control. The all-inclusive rent / flat-rate rent implemented in this research project can be seen as a significant future motivator for a rethinking in new buildings from legislative, mandatory energy requirements to ecological and economical solutions that benefit both tenants and landlords. Combining new buildings with existing buildings to form a quarter, as well as energy-efficient renovation and integration of renewable energy into existing buildings, has great potential in Germany and should be explored further to develope new rapid realisation

Repurposing groundwater-filled mine cavities for thermal energy storage has demonstrated promising potential to buffer the imbalance of energy supply and demand. Fractured formations are widespread in old mines due to previous excavation activities, which dominates the performance and efficiency of the mine thermal energy storage (MTES) system. Therefore, understanding the mechanisms of fluid flow, heat, and solute transport in fractured reservoirs in the MTES system is essential for its application and the assessment of environmental impact. In this study, fluid flow, heat, and solute transport in a 3D fracture-matrix MTES system are modeled simultaneously based on site-specific data in Freiberg, Germany. The simulations are conducted with a coupled Hydro-Thermo-Component process newly developed in the open-source software OpenGeoSys (OGS). To stabilize the simulation in the fracture-matrix hybrid system, a flux-corrected transport scheme is specifically implemented and applied in the model. Thus, heat and mass exchange between the embedded fractures and the matrix in the formation of MTES can be accurately simulated on the basis of the conforming mesh. In a hypothetical short-term scenario of a 15-day heating and cooling cycle of the MTES operation, the solute is transported faster than the heat in the fractured formation, indicating that disturbance of mineral compositions in the original mine water can affect a larger region than heat. The embedded fracture network in the surrounding formation is also found to increase the thermal energy storage capacity by 44%, while decreasing the recovery ratio by 14% compared to those of the unfractured formation. More energy will be transported and stored in the more fractured formation from heated mine water, but less energy can be recovered. Aim: This observational study investigated the association between remdesivir treatment during hospitalization for COVID-19 and 30-day COVID-19-related and all-cause readmission across different variants time periods. Patients & methods: Hospitalization records for adult patients discharged from a COVID-19 hospitalization between 1 May 2020 to 30 April 2022 were extracted from the US PINC AI Healthcare Database. Likelihood of 30-day readmission was compared among remdesivir-treated and nonremdesivir-treated patients using multivariable logistic regression models adjusted for age, corticosteroid treatment, Charlson comorbidity index and intensive care unit stay during the COVID-19 hospitalization. Analyses were stratified by maximum supplemental oxygen requirement and variant time period (pre-Delta, Delta and Omicron). Results: Of the 440,601 patients discharged alive after a COVID-19 hospitalization, 248,785 (56.5%) patients received remdesivir. Overall, remdesivir patients had a 30-day COVID-19-related readmission rate of 3.0% and all-cause readmission rate of 6.3% compared with 5.4% and 9.1%, respectively, for patients who did not receive remdesivir during their COVID-19 hospitalization. After adjusting for demographics and clinical characteristics, remdesivir treatment was associated with significantly lower odds of 30-day COVID-19-related readmission (odds ratio 0.60 [95% confidence interval: 0.58-0.62]), and all-cause readmission (0.73 [0.72-0.75]). Significantly lower odds of 30-day readmission in remdesivir-treated patients was observed across all variant time periods. Conclusion: Treating patients hospitalized for COVID-19 with remdesivir is associated with a statistically significant reduction in 30-day COVID-19-related and all-cause readmission across variant time periods. These findings indicate that the clinical benefit of remdesivir may extend beyond the COVID-19 hospitalization.

Changes and disturbances to water diversity and quality are complex and multi-scale in space and time. Although in situ methods provide detailed point information on the condition of water bodies, they are of limited use for making area-based monitoring over time, as aquatic ecosystems are extremely dynamic. Remote sensing (RS) provides methods and data for the cost-effective, comprehensive, continuous and standardised monitoring of characteristics and changes in characteristics of water diversity and water quality from local and regional scales to the scale of entire continents. In order to apply and better understand RS techniques and their derived spectral indicators in monitoring water diversity and quality, this study defines five characteristics of water diversity and quality that can be monitored using RS. These are the diversity of water traits, the diversity of water genesis, the structural diversity of water, the taxonomic diversity of water and the functional diversity of water. It is essential to record the diversity of water traits to derive the other four characteristics of water diversity from RS. Furthermore, traits are the only and most important interface between in situ and RS monitoring approaches. The monitoring of these five characteristics of water diversity and water quality using RS technologies is presented in detail and discussed using numerous examples. Finally, current and future developments are presented to advance monitoring using RS and the trait approach in modelling, prediction and assessment as a basis for successful monitoring and management strategies.

Background: The SARS-CoV-2 pandemic is still ongoing and a major challenge for health care services worldwide. In the first WSES COVID-19 emergency surgery survey, a strong negative impact on emergency surgery (ES) had been described already early in the pandemic situation. However, the knowledge is limited about current effects of the pandemic on patient flow through emergency rooms, daily routine and decision making in ES as well as their changes over time during the last two pandemic years. This second WSES COVID-19 emergency surgery survey investigates the impact of the SARS-CoV-2 pandemic on ES during the course of the pandemic. Methods: A web survey had been distributed to medical specialists in ES during a four-week period from January 2022, investigating the impact of the pandemic on patients and septic diseases both requiring ES, structural problems due to the pandemic and time-to-intervention in ES routine. Results: 367 collaborators from 59 countries responded to the survey. The majority indicated that the pandemic still significantly impacts on treatment and outcome of surgical emergency patients (83.1% and 78.5%, respectively). As reasons, the collaborators reported decreased case load in ES (44.7%), but patients presenting with more prolonged and severe diseases, especially concerning perforated appendicitis (62.1%) and diverticulitis (57.5%). Otherwise, approximately 50% of the participants still observe a delay in time-to-intervention in ES compared with the situation before the pandemic. Relevant causes leading to enlarged time-to-intervention in ES during the pandemic are persistent problems with in-hospital logistics, lacks in medical staff as well as operating room and intensive care capacities during the pandemic. This leads not only to the need for triage or transferring of ES patients to other hospitals, reported by 64.0% and 48.8% of the collaborators, respectively, but also to paradigm shifts in treatment modalities to non-operative approach

The development of algorithms for remote sensing of water quality (RSWQ) requires a large amount of in situ data to account for the bio-geo-optical diversity of inland and coastal waters. The GLObal Reflectance community dataset for Imaging and optical sensing of Aquatic environments (GLORIA) includes 7,572 curated hyperspectral remote sensing reflectance measurements at 1 nm intervals within the 350 to 900 nm wavelength range. In addition, at least one co-located water quality measurement of chlorophyll a, total suspended solids, absorption by dissolved substances, and Secchi depth, is provided. The data were contributed by researchers affiliated with 59 institutions worldwide and come from 450 different water bodies, making GLORIA the de-facto state of knowledge of in situ coastal and inland aquatic optical diversity. Each measurement is documented with comprehensive methodological details, allowing users to evaluate fitness-for-purpose, and providing a reference for practitioners planning similar measurements. We provide open and free access to this dataset with the goal of enabling scientific and technological advancement towards operational regional and global RSWQ monitoring.

Diagnostic applications for skin in the microwave range have developed significantly in recent years, due the non-invasiveness of these applications and their ability to assess tissue water content. Despite their capabilities, however, there is still no appropriate clinically applicable microwave tool for the assessment of burn wounds. A common practice is the visual inspection and evaluation of burns by the doctor, which is a challenging task even for experienced medical professionals. An incorrect assessment can have far-reaching consequences, such as unnecessary surgery or surgery that is necessary but omitted. In this paper, two different approaches of millimeter-wave burn wound assessment are presented: millimeter-wave imaging and near-field spectroscopy. For imaging, a MIMO sparse array was used to assess ex vivo burns on porcine skin in the frequency range of 70–80 GHz. With a resonant millimeter-wave near-field probe, reflective spectroscopy at individual sites of an ex vivo burn on porcine skin in the frequency range of 75–110 GHz was performed. The results showed individual advantages and drawbacks for both approaches, with surprising benefits of the spectroscopic method. Nevertheless, both approaches were shown to be suitable for clinical usage in diagnosing burns.

Biostatistical evaluation of DNA profiles supports the jurisdiction in the assessment of the evidentiary value of a DNA stain. In order to ensure comparability of such calculations on the basis of established scientific standards, a set of recommendations have already been brought to national consensus in the past. With the introduction of fully continuous models (FCMs) for probabilistic genotyping, which among other things take account of an electropherogram's signal intensities, these recommendations have to be amended. Fully continuous models allow a biostatistical evaluation of complex DNA profiles with presumed allelic drop-in and drop-out events, as well as probability based deductions of individual DNA profiles contributing to a mixture (deconvolution). This publication provides recommendations on the use of FCM-based software and the reporting of fully continuous likelihood ratios (LRfc). It recommends an FCM-based calculation for evaluating the role of an alleged contributor to DNA evidence which is difficult to interpret. Thus it replaces the previous approach of a binary calculation with the exclusion of selected loci, which was previously considered acceptable in exceptional cases. The application of FCM-based software requires comprehensive user training as well as validation and verification according to the software developer's instructions. Furthermore, considerations on LRfc thresholds are described in order to ensure compatibility among probabilistic genotyping results of different origins.

Year-round observations of the physical snow and ice properties and processes that govern the ice pack evolution and its interaction with the atmosphere and the ocean were conducted during the Multidisciplinary drifting Observatory for the Study of Arctic Climate (MOSAiC) expedition of the research vessel Polarstern in the Arctic Ocean from October 2019 to September 2020. This work was embedded into the interdisciplinary design of the 5 MOSAiC teams, studying the atmosphere, the sea ice, the ocean, the ecosystem, and biogeochemical processes. The overall aim of the snow and sea ice observations during MOSAiC was to characterize the physical properties of the snow and ice cover comprehensively in the central Arctic over an entire annual cycle. This objective was achieved by detailed observations of physical properties and of energy and mass balance of snow and ice. By studying snow and sea ice dynamics over nested spatial scales from centimeters to tens of kilometers, the variability across scales can be considered. On-ice observations of in situ and remote sensing properties of the different surface types over all seasons will help to improve numerical process and climate models and to establish and validate novel satellite remote sensing methods; the linkages to accompanying airborne measurements, satellite observations, and results of numerical models are discussed. We found large spatial variabilities of snow metamorphism and thermal regimes impacting sea ice growth. We conclude that the highly variable snow cover needs to be considered in more detail (in observations, remote sensing, and models) to better understand snow-related feedback processes. The ice pack revealed rapid transformations and motions along the drift in all seasons. The number of coupled ice–ocean interface processes observed in detail are expected to guide upcoming research with respect to the changing Arctic sea ice.

In küstennahen Gewässern ist es von Vorteil, satellitengestützte optische Messungen des Meeres mit visuellen und sensorischen Beobachtungen von Tauchrobotern zu fusionieren. Obwohl Satelliten nur wenige Meter tief in Gewässer hineinschauen können, ist es möglich, generelle Wassereigenschaften oder den Bodenbewuchs von Küstengewässern zu bestimmen. Visuelle und sensorische Tauchroboterbeobachtungen sind hierzu komplementär und können auch tiefere Gewässer erreichen. Das mitgeführte künstliche Licht wird jedoch stark gestreut und erfordert andere Messmodelle. Zusätzlich sind die räumlichen und spektralen Auflösungen der Beobachtungen oftmals sehr unterschiedlich. Wir analysieren hier die damit verbundenen Problematiken und skizzieren Wege, wie die Fusion der grundverschiedenen Messungen dennoch gelingen könnte.

In engineering, machines are typically built after a careful conception and design process: All components of a system, their roles and the interaction between them is well understood, and often even digital models of the system exist before the actual hardware is built. This enables simulations and even feedback loops between the real-world system and a digital model, leading to a digital twin that allows better testing, prediction and understanding of complex effects. On the contrary, in Earth sciences, and particularly in ocean sciences, models exist only for certain aspects of the real world, of certain processes and of some interactions and dependencies between different “components” of the ocean. These individual models cover large temporal (seconds to millions of years) and spatial (millimetres to thousands of kilometres) scales, a variety of field data underpin them, and their results are represented in many different ways. A key to enabling digital twins in the oceans is fusion at different levels, in particular, fusion of data sources and modalities, fusion over different scales and fusion of differing representations. We outline these challenges and exemplify different envisioned digital twins employed in the oceans involving remote sensing, underwater photogrammetry and computer vision, focusing on optical aspects of the digital twinning process. In particular, we look at the holistic sensing scenarios of optical properties in coastal waters as well as seafloor dynamics at volcanic slopes and discuss road blockers for digital twins as well as potential solutions to increase and widen the use of digital twins.

In the first WSES COVID-19 emergency surgery survey, a strong negative impact on emergency surgery (ES) had been described already early in the pandemic situation. However, the knowledge is limited about current effects of the pandemic on patient flow through emergency rooms, daily routine and decision making in ES as well as their changes over time during the last two pandemic years. This second WSES COVID-19 emergency surgery survey investigates the impact of the SARS-CoV-2 pandemic on ES during the course of the pandemic. METHODS: A web survey had been distributed to medical specialists in ES during a four-week period from January 2022, investigating the impact of the pandemic on patients and septic diseases both requiring ES, structural problems due to the pandemic and time-to-intervention in ES routine. RESULTS: 367 collaborators from 59 countries responded to the survey. The majority indicated that the pandemic still significantly impacts on treatment and outcome of surgical emergency patients (83.1% and 78.5%, respectively). As reasons, the collaborators reported decreased case load in ES (44.7%), but patients presenting with more prolonged and severe diseases, especially concerning perforated appendicitis (62.1%) and diverticulitis (57.5%). Otherwise, approximately 50% of the participants still observe a delay in time-to-intervention in ES compared with the situation before the pandemic. Relevant causes leading to enlarged time-to-intervention in ES during the pandemic are persistent problems with in-hospital logistics, lacks in medical staff as well as operating room and intensive care capacities during the pandemic. This leads not only to the need for triage or transferring of ES patients to other hospitals, reported by 64.0% and 48.8% of the collaborators, respectively, but also to paradigm shifts in treatment modalities to non-operative approach

Seagrass meadows are one of the most important benthic habitats in the Baltic Sea. Nevertheless, spatially continuous mapping data of Zostera marina, the predominant seagrass species in the Baltic Sea, are lacking in the shallow coastal waters. Sentinel-2 turned out to be valuable for mapping coastal benthic habitats in clear waters, whereas knowledge in turbid waters is rare. Here, we transfer a clear water mapping approach to turbid waters to assess how Sentinel-2 can contribute to seagrass mapping in the Western Baltic Sea. Sentinel-2 data were atmospherically corrected using ACOLITE and subsequently corrected for water column effects. To generate a data basis for training and validating random forest classification models, we developed an upscaling approach using video transect data and aerial imagery. We were able to map five coastal benthic habitats: bare sand (25 km²), sand dominated (16 km²), seagrass dominated (7 km²), dense seagrass (25 km²) and mixed substrates with red/ brown algae (3.5 km²) in a study area along the northern German coastline. Validation with independent data pointed out that water column correction does not significantly improve classification results compared to solely atmospherically corrected data (balanced overall accuracies ˜0.92). Within optically shallow waters (0–4 m), per class and overall balanced accuracies (>0.82) differed marginally depending on the water depth. Overall balanced accuracy became worse (<0.8) approaching the border to optically deep water (˜ 5 m). The spatial resolution of Sentinel-2 (10–20 m) allowed delineating detailed spatial patterns of seagrass habitats, which may serve as a basis to retrieve spatially continuous data for ecologically relevant metrics such as patchiness. Thus, Sentinel-2 can contribute unprecedented information for seagrass mapping between 0 and around 5 m water depths in the Western Baltic Sea.

Along the Baltic coastline of Germany, drifting vegetation and beach cast create overlays at the otherwise sandy or stony beaches. These overlays influence the morphodynamics and structures of the beaches. To better understand the influence of these patchy habitats on coastal environments, regular monitoring is necessary. Most studies, however, have been conducted on spatially larger and temporally more stable occurrences of aquatic vegetation such as floating fields of Sargassum. Nevertheless, drifting vegetation and beach cast pose a particular challenge, as they exhibit high temporal dynamics and sometimes small spatial extent. Regular surveys and mappings are the traditional methods to record their habitats, but they are time-consuming and cost-intensive. Spaceborne remote sensing can provide frequent recordings of the coastal zone at lower cost. Our study therefore aims at the monitoring of drifting vegetation and beach cast on spatial scales between 3 and 10 m. We developed an automated coastline masking algorithm and tested six supervised classification methods and various classification ensembles for their suitability to detect small-scale assemblages of drifting vegetation and beach cast in a study area at the coastline of the Western Baltic Sea using multispectral data of the sensors Sentinel-2 MSI and PlanetScope. The shoreline masking algorithm shows high accuracies in masking the land area while preserving the sand-covered shoreline. We could achieve best classification results using PlanetScope data with an ensemble of a random forest classifier, cart classifier, support vector machine classifier, naïve bayes classifier and stochastic gradient boosting classifier. This ensemble accomplished a combined f1-score of 0.95. The accuracy of the Sentinel-2 classifications was lower but still achieved a combined f1-score of 0.86 for the same ensemble. Uterine fibroids are one of the most common diseases in female patients, lead mainly to bleeding disorders and lower abdominal pain, and reduce the chance of having children. In recent years we have seen a trend towards more and more pharmacotherapies and minimally invasive organ-preserving treatments. One novel and innovative procedure for an organ-preserving treatment of symptomatic uterine fibroids is the transcervical ultrasound-guided radiofrequency ablation (TRFA). TRFA has been used in Germany since 2013 and later found use in other German-speaking countries as well. There have now been more than 1200 TRFA treatments performed in Germany, Austria, and Switzerland. Experts from these three countries came together for a consensus meeting to analyze the significance of the procedure in the overall concept of the treatment of symptomatic uterine fibroids.

Year-round observations of the physical snow and ice properties and processes that govern the ice pack evolution and its interaction with the atmosphere and the ocean were conducted during the Multidisciplinary drifting Observatory for the Study of Arctic Climate (MOSAiC) expedition of the research vessel Polarstern in the Arctic Ocean from October 2019 to September 2020. This work was embedded into the interdisciplinary design of the 5 MOSAiC teams, studying the atmosphere, the sea ice, the ocean, the ecosystem, and biogeochemical processes. The overall aim of the snow and sea ice observations during MOSAiC was to characterize the physical properties of the snow and ice cover comprehensively in the central Arctic over an entire annual cycle. This objective was achieved by detailed observations of physical properties and of energy and mass balance of snow and ice. By studying snow and sea ice dynamics over nested spatial scales from centimeters to tens of kilometers, the variability across scales can be considered. On-ice observations of in situ and remote sensing properties of the different surface types over all seasons will help to improve numerical process and climate models and to establish and validate novel satellite remote sensing methods; the linkages to accompanying airborne measurements, satellite observations, and results of numerical models are discussed. We found large spatial variabilities of snow metamorphism and thermal regimes impacting sea ice growth. We conclude that the highly variable snow cover needs to be considered in more detail (in observations, remote sensing, and models) to better understand snow-related feedback processes. The ice pack revealed rapid transformations and motions along the drift in all seasons. The number of coupled ice–ocean interface processes observed in detail are expected to guide upcoming research with respect to the changing Arctic sea ice

In this study, we present a fused multi-scale approach to model habitat suitability index (HSI) maps for three different locust species. The presented methodology was applied for the Italian locust (Calliptamus italicus, CIT) in Pavlodar oblast, Northern Kazakhstan, for the Moroccan locust (Dociostaurus maroccanus, DMA) in Turkistan oblast, South Kazakhstan and for the desert locust (Schistocerca gregaria) in Awash river basin, Ethiopia, Djibouti, Somalia. The main novelty is based on implementing results from ecological niche modelling (ENM) with time-series analyses of high spatial resolution remote sensing data (Sentinel-2) and further auxiliary datasets in a fused HSI model. Within the ENM important climatic variables (e.g. temperature, rainfall) and edaphic variables (e.g. sand and moisture contents) are included at a coarse spatial resolution. The analyses of Sentinel-2 time-series data enables mapping locust breeding habitats based on recent remotely sensed land observation at high spatial resolution and mirror the actual vegetation state, land use, land cover and in this way identify areas with favorable conditions for egg survival and breeding. The fused HSI results for year 2019 were validated based on ground field observation and reach area under curve (AUC) performance of 0.747% for CIT, 0.850% for DMA and 0.801% for desert locust. The innovation of this study is a multi-scale approach which accounts not only for climatic and environmental conditions but also for current vegetation and land management situation. This kind of up-to-date spatial detailed information on breeding suitability could enable area prioritization for risk assessment, monitoring and early intervention of locust pests.

This work was embedded into the interdisciplinary design of the 5 MOSAiC teams, studying the atmosphere, the sea ice, the ocean, the ecosystem, and biogeochemical processes. The overall aim of the snow and sea ice observations during MOSAiC was to characterize the physical properties of the snow and ice cover comprehensively in the central Arctic over an entire annual cycle. This objective was achieved by detailed observations of physical properties and of energy and mass balance of snow and ice. By studying snow and sea ice dynamics over nested spatial scales from centimeters to tens of kilometers, the variability across scales can be considered. On-ice observations of in situ and remote sensing properties of the different surface types over all seasons will help to improve numerical process and climate models and to establish and validate novel satellite remote sensing methods; the linkages to accompanying airborne measurements, satellite observations, and results of numerical models are discussed. We found large spatial variabilities of snow metamorphism and thermal regimes impacting sea ice growth. We conclude that the highly variable snow cover needs to be considered in more detail (in observations, remote sensing, and models) to better understand snow-related feedback processes. The ice pack revealed rapid transformations and motions along the drift in all seasons. The number of coupled ice–ocean interface processes observed in detail are expected to guide upcoming research with respect to the changing Arctic sea ice.

Chronic HIV infection may exacerbate atherosclerotic vascular disease, which at advanced stages presents as necrotic plaques rich in crystalline cholesterol. Such lesions can catastrophically rupture precipitating myocardial infarct and stroke, now important causes of mortality in those living with HIV. However, in this population little is known about plaque structure relative to crystalline content and its chemical composition. Here, we first interrogated plaque crystal structure and composition in atherosclerotic SIV-infected macaques using non-linear optical microscopy. By stimulated Raman scattering and second harmonic generation approaches both amorphous and crystalline plaque lipid was detected and the crystal spectral profile indicated a cholesterol ester (CE) dominated composition. Versus controls, SIV+ samples had a greater number of cholesterol crystals (CCs), with the difference, in part, accounted for by crystals of a smaller length. Given the ester finding, we profiled HIV+ plaques and also observed a CE crystalline spectral signature. We further profiled plaques from Ldlr-/- mice fed a high fat diet, and likewise, found CE-dominate crystals. Finally, macrophage exposure to CCs or AcLDL induced auto-fluorescent puncta that co-stained with the LC3B autophagy sensor. In aggregate, we show that atheromatous plaques from mice, macaques and humans, display necrotic cores dominated by esterified CCs, and that plaque macrophages may induce autophagic vesicle formation upon encountering CCs. These findings help inform our knowledge of plaque core lipid evolution and how the process may incite systemic inflammation.

Seagrass meadows are one of the most important benthic habitats in the Baltic Sea. Nevertheless, spatially continuous mapping data of Zostera marina, the predominant seagrass species in the Baltic Sea, are lacking in the shallow coastal waters. Sentinel-2 turned out to be valuable for mapping coastal benthic habitats in clear waters, whereas knowledge in turbid waters is rare. Here, we transfer a clear water mapping approach to turbid waters to assess how Sentinel-2 can contribute to seagrass mapping in the Western Baltic Sea. Sentinel-2 data were atmospherically corrected using ACOLITE and subsequently corrected for water column effects. To generate a data basis for training and validating random forest classification models, we developed an upscaling approach using video transect data and aerial imagery. We were able to map five coastal benthic habitats: bare sand (25 km²), sand dominated (16 km²), seagrass dominated (7 km²), dense seagrass (25 km²) and mixed substrates with red/ brown algae (3.5 km²) in a study area along the northern German coastline. Validation with independent data pointed out that water column correction does not significantly improve classification results compared to solely atmospherically corrected data (balanced overall accuracies ˜0.92). Within optically shallow waters (0–4 m), per class and overall balanced accuracies (>0.82) differed marginally depending on the water depth. Overall balanced accuracy became worse (<0.8) approaching the border to optically deep water (˜ 5 m). The spatial resolution of Sentinel-2 (10–20 m) allowed delineating detailed spatial patterns of seagrass habitats, which may serve as a basis to retrieve spatially continuous data for ecologically relevant metrics such as patchiness. Thus, Sentinel-2 can contribute unprecedented information for seagrass mapping between 0 and around 5 m water depths in the Western Baltic Sea.

Aims The aim of this official guideline published and coordinated by the German Society of Gynaecology and Obstetrics (DGGG) in cooperation with the Austrian Society for Gynaecology and Obstetrics (OEGGG) and the Swiss Society for Gynaecology and Obstetrics (SGGG) was to provide consensusbased recommendations for the diagnosis and treatment of endometriosis based on an evaluation of the relevant literature. Methods This S2k guideline represents the structured consensus of a representative panel of experts with different professional backgrounds commissioned by the Guideline Committee of the DGGG, OEGGG and SGGG. Recommendations Recommendations on the epidemiology, aetiology, classification, symptomatology, diagnosis and treatment of endometriosis are given and special situations are discussed.

Over the last decades forensic DNA analysis has become an important tool in criminal investigations, aided by increasing sensitivity and a high level of technological standardization. Due to the continuing methodological improvements the successful genetic analysis of smaller and smaller latent DNA traces has been taken for granted. This leads to a shift in tasks for the forensic DNA expert regarding the evaluation of complex trace scenarios during a court trial. Rather than discussing an individual's DNA as the potential source of a DNA trace, questions put before the expert are now focussed on activities leading to potentially innocent depositions of biological material. The German Stain Commission recognizes the need to assess alternative scenarios of trace deposition. For the evaluation of forensic DNA findings within the context of further evidence in a given case scenario, we introduce the hierarchy of propositions, a well-established concept in the international literature of forensic science. Furthermore, the scientific basis of evaluating alternative DNA transfer scenarios and the possibilities and limitations of expressing an expert opinion are described.

Atmospheric correction over inland and coastal waters is one of the major remaining challenges in aquatic remote sensing, often hindering the quantitative retrieval of biogeochemical variables and analysis of their spatial and temporal variability within aquatic environments. The Atmospheric Correction Intercomparison Exercise (ACIX-Aqua), a joint NASA - ESA activity, was initiated to enable a thorough evaluation of eight state-of-the-art atmospheric correction (AC) processors available for Landsat-8 and Sentinel-2 data processing. Over 1000 radiometric matchups from both freshwaters (rivers, lakes, reservoirs) and coastal waters were utilized to examine the quality of derived aquatic reflectances ((rho) over cap (w)). This dataset originated from two sources: Data gathered from the international scientific community (henceforth called Community Validation Database, CVD), which captured predominantly inland water observations, and the Ocean Color component of AERONET measurements (AERONET-OC), representing primarily coastal ocean environments. This volume of data permitted the evaluation of the AC processors individually (using all the matchups) and comparatively (across seven different Optical Water Types, OWTs) using common matchups. We found that the performance of the AC processors differed for CVD and AERONET-OC matchups, likely reflecting inherent variability in aquatic and atmospheric properties between the two datasets. For the former, the median errors in (rho) over cap (w)(560) and (rho) over cap (w)(664) were found to range from 20 to 30% for best-performing processors. Using the AERONET-OC matchups, our performance assessments showed that median errors within the 15-30% range in these spectral bands may be achieved. The largest uncertainties were associated with the blue bands (25 to 60%) for best-performing processors considering both CVD and AERONET-OC assessments.
