Background: Irritable bowel syndrome (IBS) is a functional disorder without any pathological alteration, in which the alterations of the Candida/Saccharomyces ratio of the gut microbiota, the balance of pro and anti-inflammatory cytokines and the brain-gut-microbiome axis are important for the development and progression of IBS. The aim of the study was to identify natural products, including essential oils or hydrolates, which were contextually harmless for the gut beneficial strains (e.g. Saccharomyces spp.) but inhibitory for the pathogenic ones (Candida spp.). Methods: The effectiveness of 6 essential oils and 2 hydrolates was evaluated using microbiological tests, carried out on 50 clinical isolates (Candida, Saccharomyces and Galattomyces species) and 9 probiotic strains (Saccharomyces cerevisiae, Lactobacillus species, Akkermansia muciniphila and Faecalibacterium prausnitzii) and immunological and antioxidant assays. Results: The study led to a mixture based on a 1/100 ratio of Citrus aurantium var. amara essential oil/Vitis vinifera cv Italia hydrolate able to contextually reduce, in a concentration-dependent manner, the ability of Candida species to form hyphal filaments and have an interesting immunomodulatory and anti-oxidant action. This mixture can potentially be useful in the IBS treatment promoting the restoration of the intestinal microbial and immunological balance.

Pyrethroid resistance is a significant threat to agricultural, urban and public health pest control activities. Because economic incentives for the production of novel active ingredients for the control of public health pests are lacking, this field is particularly affected by the potential failure of pyrethroid-based insecticides brought about by increasing pyrethroid resistance. As a result, innovative approaches are desperately needed to overcome insecticide resistance, particularly in mosquitoes that transmit deadly and debilitating pathogens. Numerous studies have demonstrated the potential of plant essential oils to enhance the efficacy of pyrethroids. The toxicity of pyrethroids combined with plant oils is significantly greater than the baseline toxicity of either oils or pyrethroids applied alone, which suggests there are synergistic interactions between components of these mixtures. The present study examined the potential of eight plant essential oils applied in one of two concentrations (1% and 5%) to enhance the toxicity of various pyrethroids (permethrin, natural pyrethrins, deltamethrin and β-cyfluthrin). The various plant essential oils enhanced the pyrethroids to differing degrees. The levels of enhancement provided by combinations of plant essential oils and pyrethroids in comparison with pyrethroids alone were calculated and synergistic outcomes characterized. Numerous plant essential oils significantly synergized a variety of pyrethroids; type I pyrethroids were synergized to a greater degree than type II pyrethroids. Eight plant essential oils significantly enhanced 24-h mortality rates provided by permethrin and six plant essential oils enhanced 24-h mortality rates obtained with natural pyrethrins. By contrast, only three plant essential plants significantly enhanced the toxicity of deltamethrin and β-cyfluthrin. Of the plant essential oils that enhanced the toxicity of these pyrethroids, some produced varying levels of synergism and antagonism

Momordica charantia L. or bitter gourd, a Cucurbitaceae family plant is a plant native to the semi-tropical climate of Thailand, Asia, India and Africa and has been traditionally used as a folk remedy and best known for its anti-diabetic, antiinflammatory, anti-microbial, anti-ulcer and antihelmintic properties. The aims of this study were to investigate the anti-ulcerogenic activities of Momordica charantia L. (MC) essential oil and polypeptide k on various rats model. The anti-ulcerogenic effects of MCEO and polypeptide k were studied against HCl/Ethanol and Indomethacin-induced ulcer in rats. Spraque Dawley rats were given treatment orally for 7 days consecutively. In pre-treatment for MCEO, total length for HCl/EtOH is significantly longer than indomethacin Generally, for negative control, total length is 48.2±19.9. Supplementation with 10 MCEO, it reduced to 19.3±13.1 and similar to Rantidine 100 mg/kg. When the dose was increased, the total length was decreased (19.3±13.1 to 6.2±6.2). Rantidine 100 mg/kg as reference drug reduced the length about half of the negative control group. Furthermore, polypeptide k showed significantly longer (ulcer length) in HCl/EtOH than indomethacin. Generally, for negative control, total length is 43.0±14.1. Supplementation with 10 PPK, it reduced to 29.3±18.6 and slightly similar to Rantidine 100 mg/kg. When the dose was increased, the total length was decreased (29.3±18.6 to 13.6±9.5). Rantidine 100 mg/kg as reference drug reduced the length about half of the negative control group (22.1±13.2). This has been supported by findings from pylorus-ligated model in rats. Pre-treatment with MCEO at 10 mg/kg, 50 mg/kg and 100 mg/kg failed to increase the volume of gastric acid secretion when compared to control group. The patchoulol synthase (PTS) is a multi-product sesquiterpene synthases which is the central enzyme for biosynthesis of patchouli essential oil in the patchouli plant. Sesquiterpene synthases catalyse the formation of various complex carbon backbones difficult to approach by organic synthesis. Here, we report the characterisation of a recombinant patchoulol synthase complementary DNA (cDNA) variant (PTS var. 1), exhibiting significant amino acid exchanges compared to the native PTS. The product spectrum using the natural substrate E,E-farnesyl diphosphate (FDP) as well as terpenoid products resulting from conversions employing alternative substrates was analysed by GC-MS. In respect to a potential use as a biocatalyst, important enzymatic parameters such as the optimal reaction conditions, kinetic behaviour and the product selectivity were studied as well. Adjusting the reaction conditions, an increased patchoulol ratio in the recombinant essential oil was achieved. Nevertheless, the ratio remained lower than in plant-derived patchouli oil. As alternative substrates, several prenyl diposphates were accepted and converted in numerous compounds by the PTS var. 1, revealing its great biocatalytic potential. Medicinal tea tree (Melaleuca alternifolia) leaves contain large amounts of an essential oil, dominated by monoterpenes. Several enzymes of the chloroplastic methylerythritol phosphate (MEP) pathway are hypothesised to act as bottlenecks to the production of monoterpenes. We investigated, whether transcript abundance of genes encoding for enzymes of the MEP pathway were correlated with foliar terpenes in M. alternifolia using a population of 48 individuals that ranged in their oil concentration from 39 -122 mg x g DM(-1). Our study shows that most genes in the MEP pathway are co-regulated and that the expression of multiple genes within the MEP pathway is correlated with oil yield. Using multiple regression analysis, variation in expression of MEP pathway genes explained 87% of variation in foliar monoterpene concentrations. The data also suggest that sesquiterpenes in M. alternifolia are synthesised, at least in part, from isopentenyl pyrophosphate originating from the plastid via the MEP pathway.

[EN] The essential oil composition of Cistus ladanifer grown in central Spain was analysed by gas chromatography and gas chromatography/mass spectrometry, revealing it to be rich in oxygenated compounds, with trans-pinocarveol (20.00%), bornyl acetate (7.03%) and terpinen-4-ol (6.37%) as the main monoterpene compounds. Viridiflorol (13.59%) and ledol (4.36%) were the main constituents of the oxygenated sesquiterpene fraction. Large amounts of-pinene (4.70%) were found in the hydrocarbonated fractions. To study its possible use as a natural herbicide, the oil was tested invitro against Amaranthus hybridus, Portulaca oleracea, Chenopodium album, Conyza canadensis and Parietaria judaica, completely inhibiting A. hybridus germination, and nearly blocking C. canadensis and P. judaica germination at all concentrations assayed. In P. oleracea, the oil showed less effect, inhibiting its germination at the higher doses studied. It did not control C. album germination. Regarding seedling length, the oil had strong phytotoxic activity, being effective at all concentrations tested. © 2012 Taylor & Francis.

More than 96% of the total oil components (43 and 54 compounds, respectively) were identified, with ascaridole (51.17 ± 9.51), p-cymene (16.31 ± 2.52) and 1,8-cineole (14.45 ± 2.99) as the main compounds in P. boldus and ¿-eudesmol (21.65 ± 0.41), followed of elemol (12.03 ± 0.34) and terpinen-4-ol (11.56 ± 1.06) in D. winterii. The herbicidal activity was tested against Amaranthus hybridus and Portulaca oleracea. P. boldus essential oil was the most phytotoxic against both weeds, inhibiting seed germination and seedling growth at all concentrations assayed (0.125-1 ¿L/mL). D. winterii essential oil did not show any effect on A. hybridus germination and only affected P. oleracea germination at the highest concentration. The classical methods of chromatographic identification of compounds were based on calculation of retention indices by using different stationary phases. The aim of the work was to differentiate essential oils extracted from different plant species by identification of some of their major compounds. The method of identification was based on the calculation of new retention indices of essential oils compounds fractionated on a polar chromatographic column with temperature programming system. Similar chromatograms have been obtained on the same column for one plant family with two different temperature gradients allowing the rapid identification of essential oils of different species, sub-species or chemotypes of Citrus, Mentha and Thymus.

The microbiological contamination of raw plant materials is common and may be adequately reduced by radiation processing. This study evaluated the effects of gamma- and e-beam ionizing radiations (25 kGy) on three plants used as food or as medicinal products (Thymus vulgaris L., Eucalyptus radiata D.C., and Lavandula angustifolia Mill.) as well as their effects on extracted or commercial essential oils and pure standard samples. Comparison between irradiated and nonirradiated samples was performed by GC/FID and GC/MS. At the studied doses, gamma and e-beam ionizing radiation did not induce any detectable qualitative or quantitative significant changes in the contents and yields of essential oils immediately after ionizing radiation of plants or commercial essential oils and standards. As the maximum dose tested (25 kGy) is a sterilizing dose (much higher than doses used for decontamination of vegetable drugs), it is likely that even decontamination with lower doses will not modify yields or composition of essential oils of these three plants.
