Although nonsteroidal anti-inflammatory drugs are the first line of therapeutics for the treatment of mild to moderate somatic pain, they are not generally considered to be effective for neuropathic pain. In the current study, direct activation of spinal Toll-like 4 receptors (TLR4) by the intrathecal (IT) administration of KDO2 lipid A (KLA), the active component of lipopolysaccharide, elicits a robust tactile allodynia that is unresponsive to cyclooxygenase inhibition, despite elevated expression of cyclooxygenase metabolites in the spinal cord. Intrathecal KLA increases 12-lipoxygenase-mediated hepoxilin production in the lumbar spinal cord, concurrent with expression of the tactile allodynia. The TLR4-induced hepoxilin production was also observed in primary spinal microglia, but not in astrocytes, and was accompanied by increased microglial expression of the 12/15-lipoxygenase enzyme 15-LOX-1. Intrathecal KLA-induced tactile allodynia was completely prevented by spinal pretreatment with the 12/15-lipoxygenase inhibitor CDC or a selective antibody targeting rat 15-LOX-1. Similarly, pretreatment with the selective inhibitors ML127 or ML351 both reduced activity of the rat homolog of 15-LOX-1 heterologously expressed in HEK-293T cells and completely abrogated nonsteroidal anti-inflammatory drug-unresponsive allodynia in vivo after IT KLA. Finally, spinal 12/15-lipoxygenase inhibition by nordihydroguaiaretic acid (NDGA) both prevents phase II formalin flinching and reverses formalin-induced persistent tactile allodynia. Taken together, these findings suggest that spinal TLR4-mediated hyperpathic states are mediated at least in part through activation of microglial 15-LOX-1.

Lipoxins (LX) are proresolution lipid mediators that inhibit phlogistic neutrophil recruitment and promote wound-healing macrophage recruitment in humans via potent and specific signaling through the LXA4 receptor (ALX). One model of lipoxin biosynthesis involves sequential metabolism of arachidonic acid by two cell types expressing a combined transcellular metabolon. It is currently unclear how lipoxins are efficiently formed from precursors or if they are directly generated after receptor-mediated inflammatory commitment. Here, we provide evidence for a pathway by which lipoxins are generated in macrophages as a consequence of sequential activation of toll-like receptor 4 (TLR4), a receptor for endotoxin, and P2X7, a purinergic receptor for extracellular ATP. Initial activation of TLR4 results in accumulation of the cyclooxygenase-2-derived lipoxin precursor 15-hydroxyeicosatetraenoic acid (15-HETE) in esterified form within membrane phospholipids, which can be enhanced by aspirin (ASA) treatment. Subsequent activation of P2X7 results in efficient hydrolysis of 15-HETE from membrane phospholipids by group IVA cytosolic phospholipase A2, and its conversion to bioactive lipoxins by 5-lipoxygenase. Our results demonstrate how a single immune cell can store a proresolving lipid precursor and then release it for bioactive maturation and secretion, conceptually similar to the production and inflammasome-dependent maturation of the proinflammatory IL-1 family cytokines. These findings provide evidence for receptor-specific and combinatorial control of pro- and anti-inflammatory eicosanoid biosynthesis, and potential avenues to modulate inflammatory indices without inhibiting downstream eicosanoid pathways.

Eicosanoids are the most well characterized oxidized lipids, which are derived from a 20-carbon, 4-double bond- containing fatty acid, arachidonic acid. Several eicosanoids are essential bioactive regulators of various physiological processes that promote advantageous human health outcomes, with inflammation being the most prominent. Pain, swelling, fever, and a host of other adverse conditions naturally accompany inflammation and are partly due to the actions of two well studied classes of eicosanoids, prostaglandins and leukotrienes. For obvious reasons, prostaglandins and leukotrienes have been among the most intensely pursued drug targets for several decades, but existing therapies have yielded mixed results, often at the cost of problematic, or even detrimental side effects. How eicosanoids and other oxidized lipids are biologically produced in animal and human inflammatory contexts is currently understood mostly at the in vitro level, which describes a general pathway of enzymes and receptors. Some eicosanoid pathway enzymes produce multiple lipid species, and some enzyme isomers only produce a single lipid species. Presently, a biologically relevant cellular understanding of eicosanoid production is not fully well developed, but the central importance of the biological roles of eicosanoids and related compounds in health and disease demands continued progress in this pursuit. Macrophage cells play a pivotal role in orchestrating innate immunity, partly through their ability to produce eicosanoids in response to both pathogenic and sterile traumatic cues. These haematopoietic immune cells proficiently sense danger signals via pathogen- recognizing receptors, and rid the body of excessive cellular accumulation via phagocytosis of apoptotic leukocytes during inflammation and red blood cells during

Eicosanoid metabolism differs in profile and quantity between macrophages of different tissue origin and method of elicitation, as well as between primary and immortalized macrophages after activation with inflammatory stimuli. Using a lipidomic approach, we comprehensively analyzed the eicosanoids made by murine RPMs, TGEMs, BMDM, and the macrophage-like cell line RAW after stimulation with the TLR-4-specific agonist KLA. Direct correlation among total COX metabolites, COX side-products (11-HETE, 15-HETE), COX-2 mRNA, and protein at 8 h was found when comparing each cell type. Comprehensive qPCR analysis was used to compare relative transcript levels between the terminal prostanoid synthases themselves as well as between each cell type. Levels of PGE(2), PGD(2), and TxB(2) generally correlated with enzyme transcript expression of PGES, PGDS, and TBXS, providing evidence of comparable enzyme activities. PGIS transcript was expressed only in RPM and TGEM macrophages and at an exceptionally low level, despite high metabolite production compared with other synthases. Presence of PGIS in RPM and TGEM also lowered the production of PGE(2) versus PGD(2) by approximately tenfold relative to BMDM and RAW cells, which lacked this enzyme. Our results demonstrate that delayed PG production depends on the maximal level of COX-2 expression in different macrophages after TLR-4 stimulation. Also, the same enzymes in each cell largely dictate the profile of eicosanoids produced depending on the ratios of expression between them, with the exception of PGIS, which appears to have much greater synthetic capacity and competes selectively with mPGES-1.

Rationale: Sepsis, a life-threatening organ dysfunction caused by a dysregulated host response to infection, is a major public health concern with high mortality and morbidity. Although inflammatory responses triggered by infection are crucial for host defense against invading microbes, the excessive inflammation often causes tissue damage leading to organ dysfunction. Resolution of inflammation, an active immune process mediated by endogenous lipid mediators (LMs), is important to maintain host homeostasis. Objectives: We sought to determine the role of the nucleotidebinding domain, leucine-rich repeat-containing receptor, pyrin domain-containing-3 (NLRP3) inflammasome in polymicrobial sepsis and regulation of LM biosynthesis. Methods: We performed cecal ligation and puncture (CLP) using mice lacking NLRP3 inflammasome-associated molecules to assess mortality. Inflammation was evaluated by using biologic fluids including plasma, bronchoalveolar, and peritoneal lavage fluid. Local acting LMs in peritoneal lavage fluid from polymicrobacterial septic mice were assessed by mass spectrometry-based metabololipidomics. Measurements and Main Results: Genetic deficiency of NLRP3 inhibited inflammatory responses and enhanced survival of CLP-induced septic mice. NLRP3 deficiency reduced proinflammatory LMs and increased proresolving LM, lipoxin B-4 (LXB4) in septic mice, and in macrophages stimulated with LPS and ATP. Activation of the NLRP3 inflammasome induced caspase-7 cleavage and pyroptosis. Caspase-7 deficiency similarly reduced inflammation and mortality in CLP-induced sepsis, and increased LXB4 production in vivo and in vitro. Exogenous application of LXB4 reduced inflammation, pyroptosis, and mortality of mice after CLP. Conclusions: Genetic deficiency of NLRP3 promoted resolution of inflammation in polymicrobial sepsis by relieving caspase-7-dependent repression of LXB4 biosynthesis, and increased survival potentially via LXB4 production and inhibition of proinflamm

Dietary ingestion of (n-3) PUFA alters the production of eicosanoids and can suppress chronic inflammatory and autoimmune diseases. The extent of changes in eicosanoid production during an infection of mice fed a diet high in (n-3) PUFA, however, has not, to our knowledge, been reported. We fed mice a diet containing either 18% by weight soybean oil (SO) or a mixture with fish oil (FO), FO:SO (4:1 ratio), for 2 wk and then infected them with Borrelia burgdorferi. We used an MS-based lipidomics approach and quantified changes in eicosanoid production during Lyme arthritis development over 21 d. B. burgdorferi infection induced a robust production of prostanoids, mono-hydroxylated metabolites, and epoxide-containing metabolites, with 103 eicosanoids detected of the 139 monitored. In addition to temporal and compositional changes in the eicosanoid profile, dietary FO substitution increased the accumulation of 15-deoxy PGJ(2), an antiinflammatory metabolite derived from arachidonic acid. Chiral analysis of the mono-hydroxylated metabolites revealed they were generated from primarily nonenzymatic mechanisms. Although dietary FO substitution reduced the production of inflammatory (n-6) fatty acid-derived eicosanoids, no change in the host inflammatory response or development of disease was detected.

To facilitate research on these lipid mediators, we have developed a targeted high-throughput mass spectrometric based methodology to monitor and quantitate both eicosanoids and NAEs, and can be analyzed separately or together in series. Each methodology utilizes scheduled multiple reaction monitoring (sMRM) pairs in conjunction with a 25 min reverse-phase HPLC separation. The eicosanoid methodology monitors 141 unique metabolites and quantitative amounts can be determined for over 100 of these metabolites against standards. The analysis covers eicosanoids generated from cycloxygenase, lipoxygenase, cytochrome P450 enzymes, and those generated from non-enzymatic pathways. The NAE analysis monitors 36 metabolites and quantitative amounts can be determined for 33 of these metabolites against standards. The NAE method contains metabolites derived from saturated fatty acids, unsaturated fatty acids, and eicosanoids. The lower limit of detection for eicosanoids ranges from 0.1pg to 1pg, while NAEs ranges from 0.1pg to 1000pg. The rationale and design of the methodology is discussed.

This study is concerned with the measurement of the fundamental laryngeal frequency in speech. Visual location of the laryngeal period boundaries is employed as the most suitable reference for evaluating techniques of automatic laryngeal frequency analysis. A laryngeal frequency analyzer is considered to consist of three essential components: a preprocessor, a basic extractor, and a postprocessor. If the basic extractor of the laryngeal frequency analyzer is of the type which is sensitive only to zero crossings, the preprocessor output wave should have exactly two zero crossings per laryngeal period. It was found that the criterion of two zero crossings per period is satisfied if the amplitude of the first harmonic component is greater than the sum of the amplitudes of the other harmonic components when multiplied by their respective harmonic numbers. Harmonic analysis showed that squaring and full-wave rectification are more appropriate for increasing the relative level of the fundamental signal component than half-wave rectification or a logarithmic transformation. A set of speech waves was processed through several experimental analyzers, and three postprocessors employing different principles and having different characteristics of operation were compared. The Lincolnshire FA aims to support players with disabilities play football either as a part of a structured club, a team that plays in the school games, within the Lincolnshire Ability Counts League or more recreationally learning how to play. However, despite the FA’s best efforts and their disability football strategy 2010-2012, only 141,000 disabled players are involved in the game, from a total population of 11 million disabled people in the UK. (The Football Association, 2012, p.12). Indeed, although football is arguably the most popular sport in the country, the participation rates with Lincoln are relatively low. This project presents data from a programme evaluation (Rossi et al 2004, Evaluation: A systematic approach) of a project delivered in partnership with the Lincolnshire county F.A which aimed to investigate the barriers which prevent students which are partially sighted, blind, or those that suffer cerebral palsy, from participating in football. Furthermore, the study aimed to evaluate the impact of football taster sessions designed to increase participation among disabled children through the use of the ‘Mars play’ scheme. This scheme provides qualified coaches, who set up a football session for an hour, with no commitments for the players. However, the intention is that, if players were talented enough, they could be directed into of the disability player pathway in local player development centres. The primary aim of this research was to complete a programme evaluation of the scheme in order to investigate whether it met disabled students’ expectations of disability football during the ‘Mars Play’ scheme at the University of Lincoln. Participants completed questionnaires which aimed to discover why participants’ perceived barriers to participation in disability football, what is preventing them from playing, whether they were aware of how they can begin to participate and is whether they perceived disability football to be an appropriate activity
