We laconically describe the great contributions of Professor Francesco Altomare to mathematical research and Ph.D education, and his unique status in the mathematical community. In particular, we present and give examples of his innovative and great achievements related to the following areas of mathematics: Functional Analysis, Operator Theory, Potential Theory, Approximation Theory, Probability Theory, Function Spaces, Choquet's Theory, Dirichlet's Problem and Semigroup Theory. Moreover, we report on and give concrete examples of his unique way to work together with Ph.D students, both before and sometimes also after their dissertation. Finally, we shortly describe his remarkable "class travel" from "simple" conditions with no academic traditions in his family in the small town Giovinazzo to finally become the broad, ingenious, and powerful mathematician he is regarded to be today.

BACKGROUND: HIV-infected cell lines are widely used to study latent HIV infection, which is considered the main barrier to HIV cure. We hypothesized that these cell lines differ from each other and from cells from HIV-infected individuals in the mechanisms underlying latency. RESULTS: To quantify the degree to which HIV expression is inhibited by blocks at different stages of HIV transcription, we employed a recently-described panel of RT-ddPCR assays to measure levels of 7 HIV transcripts ("read-through," initiated, 5' elongated, mid-transcribed/unspliced [Pol], distal-transcribed [Nef], polyadenylated, and multiply-sliced [Tat-Rev]) in bulk populations of latently-infected (U1, ACH-2, J-Lat) and productively-infected (8E5, activated J-Lat) cell lines. To assess single-cell variation and investigate cellular genes associated with HIV transcriptional blocks, we developed a novel multiplex qPCR panel and quantified single cell levels of 7 HIV targets and 89 cellular transcripts in latently- and productively-infected cell lines. The bulk cell HIV transcription profile differed dramatically between cell lines and cells from ART-suppressed individuals. Compared to cells from ART-suppressed individuals, latent cell lines showed lower levels of HIV transcriptional initiation and higher levels of polyadenylation and splicing. ACH-2 and J-Lat cells showed different forms of transcriptional interference, while U1 cells showed a block to elongation. Single-cell studies revealed marked variation between/within cell lines in expression of HIV transcripts, T cell phenotypic markers, antiviral factors, and genes implicated in latency. Expression of multiply-spliced HIV Tat-Rev was associated with expression of cellular genes involved in activation, tissue retention, T cell transcription, and apoptosis/survival. CONCLUSIONS: HIV-infected cell lines differ from each other and from cells from ART-treated individuals in the mechanisms governing latent HIV infection. These differences

Psychological interventions in cardiac rehabilitation programs appear relevant in as much they significantly contribute to achieve the goals of rehabilitation, to reduce the risk of relapses and to improve patients' adherence to therapy. To this aim, motivational interviewing (MI) has shown promising results in improving motivation to change and individuals' confidence in their ability to do so.

Despite a decreasing trend in incidence and mortality, Cardiovascular Diseases (CVDs) still represent important causes of death and disability in developed countries, significantly affecting individuals’ quality of life and healthcare costs . Unhealthy lifestyle behaviors, such as poor diet, lack of physical activity and smoking status, constitute a challenge in contrasting the disease. Alternatively to the medical model, which rely on the traditional approach of information and advicegiving, evidence to date indicate the need of psychological actions able to address patients’ beliefs and concerns about their health status as well as to enhance confidence in their abilities to overcome barriers to adherence and achieve life‐style modifications in the long term. Even if the World Health Organization (WHO) Expert Committee stated that Cognitive‐Behavioural Therapy (CBT) is an important component of Cardiac Rehabilitation (CR) programs, it does not specifically focus on eliciting patients’ motivation, leading to the development of interventions aimed at enhancing health behavior change and among which Motivational Interviewing (MI) has obtained varying degrees of success. Also, the new mHealth (mobile health) approach could represent an important strategy in order to move motivational psychological support where necessary (outpatient settings), maximizing the results obtained from the CR in a long term among people suffering from CVD and other chronic conditions.

We have developed a sequencing assay for determining the usage of the genotypic HIV-1 co-receptor using peripheral blood mononuclear cell (PBMC) DNA in virologically suppressed HIV-1 infected patients. Our specific aims were to (1) evaluate the efficiency of V3 sequences in B versus non-B subtypes, (2) compare the efficiency of V3 sequences and tropism prediction using whole blood and PBMCs for DNA extraction, (3) compare the efficiency of V3 sequences and tropism prediction using a single versus a triplicate round of amplification.

The DIVA study is aimed at setting up a standardized genotypic tropism-testing on proviral-DNA for the routine clinical diagnostic-laboratory. Methods: Twelve local centres and 5 reference centres (previously cross-validated) were identified. For inter-center validation-procedure, 60 peripheral-blood mononuclear cells (PBMCs) aliquots from 45 HAART-treated patients were randomly chosen for population V3 sequencing on proviral-DNA at local HIV centre and at reference-laboratory. Viral tropism was predicted by Geno2Pheno algorithm (False Positive Rate [FPR] = 20%) as proposed by the European-Guidelines. Quantification of total HIV-1 DNA was based on a method described by Viard (2004). Results: Quantification of HIV-1 DNA was available for 35/45 (77.8%) samples, and gave a median value of 598 (IQR:252- 1,203) copies/10 6 PBMCs. A total of 56/60 (93.3%) samples were successfully amplified by both the reference and the local virological centers. The overall concordance of tropism prediction between local and reference centers was 54/56 (96.4%). Results of tropism prediction by local centers were: 33/54 (61.1%) R5 and 21/54 (38.9%) X4/DM. Conclusion: There was high concordance in the genotypic tropism prediction based on proviral DNA among different virological centers throughout Italy. Our results are in line with other European studies, and support the use of genotypic tropism testing on proviral DNA in patients with suppressed plasma HIV-1 RNA candidate to CCR5-antagonist treatment.
