The usage of nanomaterials for rheumatoid arthritis (RA) treatment can improve bioavailability and enable selective targeting. The current study prepares and evaluates the in vivo biological effects of a novel hydroxyapatite/vitamin B-12 nanoformula in Complete Freunds adjuvant-induced arthritis in rats. The synthesized nanoformula was characterized using XRD, FTIR, BET analysis, HERTEM, SEM, particle size, and zeta potential. We synthesized pure HAP NPs with 71.01% loading weight percentages of Vit B12 and 49 mg/g loading capacity. Loading of vitamin B-12 on hydroxyapatite was modeled by Monte Carlo simulation. Anti-arthritic, anti-inflammatory, and antioxidant effects of the prepared nanoformula were assessed. Treated arthritic rats showed lower levels of RF and CRP, IL-1 beta, TNF-alpha, IL-17, and ADAMTS-5, but higher IL-4 and TIMP-3 levels. In addition, the prepared nanoformula increased GSH content and GST antioxidant activity while decreasing LPO levels. Furthermore, it reduced the expression of TGF-beta mRNA. Histopathological examinations revealed an improvement in joint injuries through the reduction of inflammatory cell infiltration, cartilage deterioration, and bone damage caused by Complete Freunds adjuvant. MYC is overexpressed in approximately 70% of human cancers. Given its pervasive role in cancer biology, MYC down-regulation has become an attractive cancer treatment strategy. MethodsThe CRISPR/Cas9 method was used to produce KO cell models. Western blot was used to analyzed the expressions of MYC and TATA-binding proteinassociated factors 10 (TAF10) in cancer cells (MCF7, A549, HepG2 cells) Cell culture studies were performed to determine the mechanisms by which small molecules (Z363119456, Z363) affects MYC and TAF10 expressions and functions. Mouse studies were carried out to investigate the impact of Z363 regulation on tumor growth. ResultsZ363 activate Thyroid hormone Receptor-interacting Protein 12 (TRIP12), which phosphorylates MYC at Thr58, resulting in MYC ubiquitination and degradation and thereby regulating MYC target genes. As nanotechnology has advanced, a growing number of nanodrugs have been used in the treatment of RA due to their unique physical and chemical properties. The purpose of this study was to assess the therapeutic potential of a novel zeolite/vitamin B12 nanocomposite (Nano ZT/Vit B12) formulation in complete Freunds adjuvant (CFA)-induced arthritis. The newly synthesized Nano ZT/Vit B12 was fully characterized using various techniques such as XRD, FT-IR, BET analysis, HERTEM, SEM, practical size, zeta potential, XRF, and EDX. The anti-arthritic, anti-inflammatory, and antioxidant activities as well as the immunomodulation effect of Nano ZT/Vit B12 on the CFA rat model of arthritis were examined. Histopathologic ankle joint injuries caused by CFA intrapedal injection included synovium hyperplasia, inflammatory cell infiltration, and extensive cartilage deterioration. In the current study, a novel method to improve the nano-entrapment of enzymes into Ca-alginate gel was investigated to determine the synergistic effects of ultrasound combined with microwave shock (UMS). The effects of UMS treatment on dextranase enzymes loading effectiveness (LE) and immobilization yield (IY) were investigated. By using FT-IR spectra and SEM, the microstructure of the immobilized enzyme (IE) was characterized. Additionally, the free enzyme was used as a control to compare the reusability and enzyme-kinetics characteristics of IEs produced with and without UMS treatments. The results demonstrated that the highest LE and IY were obtained when the IE was produced with a US of 40 W at 25 kHz for 15 min combined with an MS of 60 W at a shock rate of 20 s/min for 20 min, increasing the LE and the IY by 97.32 and 78.25%, respectively, when compared with an immobilized enzyme prepared without UMS treatment. In comparison with the control, UMS treatment dramatically raised the Vmax, KM, catalytic, and specificity constant values for the IE. The outcomes suggested that a microwave shock and ultrasound combination would be an efficient way to improve the immobilization of enzymes in biopolymer gel.

Urtica dioica is a perennial herb from the family of Urticaceae that is commonly known as stinging nettle. This plant is widespread in Europe, Africa, America, and a part of Asia, as it adapts to different environments and climatic conditions. The leaves, stalk, and bark of U. dioica found applications in the field of nutrition, cosmetics, textile, pest control and pharmacology. In this connection, bioactive chemical constituents such as flavonoids, phenolic acids, amino acids, carotenoids, and fatty acids have been isolated from the plant. With this review, we aim at providing an updated and comprehensive overview of the contributions in literature reporting computational, in vitro, pre-clinical and clinical data supporting the therapeutic applications of U. dioica. Experimental evidence shows that U. dioica constituents and extracts can provide neuroprotective effects by acting through a combination of different molecular mechanisms, that are discussed in the review. These findings could lay the basis for the identification and design of more effective tools against neurodegenerative diseases.

The usage of nanomaterials for rheumatoid arthritis (RA) treatment can improve bioavailability and enable selective targeting. The current study prepares and evaluates the in vivo biological effects of a novel hydroxyapatite/vitamin B-12 nanoformula in Complete Freunds adjuvant-induced arthritis in rats. The synthesized nanoformula was characterized using XRD, FTIR, BET analysis, HERTEM, SEM, particle size, and zeta potential. We synthesized pure HAP NPs with 71.01% loading weight percentages of Vit B12 and 49 mg/g loading capacity. Loading of vitamin B-12 on hydroxyapatite was modeled by Monte Carlo simulation. Anti-arthritic, anti-inflammatory, and antioxidant effects of the prepared nanoformula were assessed. Treated arthritic rats showed lower levels of RF and CRP, IL-1 beta, TNF-alpha, IL-17, and ADAMTS-5, but higher IL-4 and TIMP-3 levels. In addition, the prepared nanoformula increased GSH content and GST antioxidant activity while decreasing LPO levels. Furthermore, it reduced the expression of TGF-beta mRNA. Histopathological examinations revealed an improvement in joint injuries through the reduction of inflammatory cell infiltration, cartilage deterioration, and bone damage caused by Complete Freunds adjuvant. As nanotechnology has advanced, a growing number of nanodrugs have been used in the treatment of RA due to their unique physical and chemical properties. The purpose of this study was to assess the therapeutic potential of a novel zeolite/vitamin B12 nanocomposite (Nano ZT/Vit B12) formulation in complete Freunds adjuvant (CFA)-induced arthritis. Nano-ZT/Vit B12 significantly declined the LPO level and increased antioxidant activities, such as GSH content and GST activity, in the arthritic rats. In the current study, a novel method to improve the nano-entrapment of enzymes into Ca-alginate gel was investigated to determine the synergistic effects of ultrasound combined with microwave shock (UMS). The effects of UMS treatment on dextranase enzymes loading effectiveness (LE) and immobilization yield (IY) were investigated. By using FT-IR spectra and SEM, the microstructure of the immobilized enzyme (IE) was characterized. Additionally, the free enzyme was used as a control to compare the reusability and enzyme-kinetics characteristics of IEs produced with and without UMS treatments. The results demonstrated that the highest LE and IY were obtained when the IE was produced with a US of 40 W at 25 kHz for 15 min combined with an MS of 60 W at a shock rate of 20 s/min for 20 min, increasing the LE and the IY by 97.32 and 78.25%, respectively, when compared with an immobilized enzyme prepared without UMS treatment. In comparison with the control, UMS treatment dramatically raised the Vmax, KM, catalytic, and specificity constant values for the IE. The outcomes suggested that a microwave shock and ultrasound combination would be an efficient way to improve the immobilization of enzymes in biopolymer gel.

Hepatocellular carcinoma (HCC) is the most common liver cancer and is featured with prominent disparity in incidence and mortality rate between males and females. Phospholipases are enzymes that hydrolyze phospholipids (PLs) either with acylhydrolase or phosphodiesterase activity. Phospholipases are involved in lipid metabolism, membrane dynamics, cellular signaling, migration, growth, and cell death. Alterations of phospholipases in hepatic tissues play important roles in pathogenesis, progression, and gender disparity of HCC. Thus, inhibition of phospholipases may be novel targets for HCC chemotherapeutics. This book chapter summarizes the different types of phospholipases and their expressions in cancers, focuses on the potential mechanisms by which these esterases mediate carcinogenesis, and explains how the inhibition of phospholipases may be targets for HCC treatment.

Anomaly Intrusion Detection Systems (ADSs) identify patterns of network data behaviour to determine whether they are normal or represent an attack using the learning detection model. Much research has been conducted on enhancing ADSs particularly in the area of data mining that focuses on intrusive behaviour detection. Unfortunately, the current detection models such as the support vector machine (SVM) is affected by high dimensional data which limits its ability to accurately classify data. Moreover, the data points which appear similar between intrusive and regular behaviours could be problematic as some innovated attack behaviours may not be detected. To overcome this SVM drawback, we propose a combination of weighted chi-square (WCS) as a feature selection (FS) and a Discretization process (D). The WCS method is used firstly to reduce the dimensionality of data following which the assembled records are transformed into interval values via the D process before the SVM is used to identify groups of samples that behave similarly and dissimilarly such as malicious and non-malicious activities. Experiments were performed with well-known NSL-KDD data sets and the results show that the proposed method namely WCS-D-SVM (weighted chi-square, discretization and support vector machine) significantly improved and enhanced accuracy and detection rates while decreasing the false positives which the single SVM classifier produces.

A major output of this activity is the development of new materials in the nanometre scale. In this thesis, primarily, a novel fabrication of metal chalcogenides in the nano region is presented. The synthetic route is possible in less than 2 minutes in an aqueous based system using mercury nitrate and a sulfide precursor; which has a rate of reaction almost tenfold quicker than previous literature. To stabilise the compound, a range of surfactants and solvents were used in order to form monodispersed colloidal chalcogenide solutions. Using this approach, it is shown for the first time that metal chalcogenides can be solution-stabilised for greater than 2 minutes using Nafion®. This thesis highlights the ways in which the newly found method of synthesis for metal chalcogenides can be extended onto a range of salts such as cadmium sulfide and bismuth sulfide. Nafion was also extended and implemented in stabilising these salts to show that the method is extendable to a wide range of solutions. This has so far eradicated the problem of coalescing in group II- VI semiconductors maintaining their 1:1 ratio (metal: precursor) ultimately maintaining their unique properties which is the basis behind their roles. Finally, the attachment of siRNA onto a well-known nanostructure was explored.

Intrusion detection systems (IDSs) effectively balance extra security appliance by identifying intrusive activities on a computer system, and their enhancement is emerging at an unexpected rate. Anomaly-based intrusion detection methods, which employ machine learning algorithms, are able to identify unforeseen attacks. Regrettably, the foremost challenge of this method is to minimize false alarm while maximizing detection and accuracy rate. We propose an integrated machine learning algorithm across K-Means clustering and Naïve Bayes Classifier called KMC+NBC to overcome the aforesaid drawbacks. K-Means clustering is applied to labeling and gathers the entire data into corresponding cluster sets based on the data behavior, i.e. , i.e. normal and attack, while Naïve Bayes Classifier (NBC) is applied to reorder the misclassified clustered data into correct categories. Experiments have been performed to evaluate the performance of KMC+NBC and NBC against ISCX 2012 Intrusion Detection Evaluation Dataset. The result shows that KMC+NBC significantly improves the accuracy, detection rate up to 99% and 98.8%, respectively, while decreasing the false alarm to 2.2%.

Nanotechnology is expected to be the basis of many of the main technological innovations of the twenty first century. Research and development in this field is growing rapidly throughout the world. A major output of this activity is the development of new materials in the nanometre scale. In this thesis, primarily, a novel fabrication of metal chalcogenides in the nano region is presented. Many machine learning methods have also been introduced by researcher recently to obtain high accuracy and detection rate. Unfortunately, a potential drawback of all those methods is the rate of false alarm. However, our proposed approach shows better results, by combining clustering (to identify groups of similarly behaved samples, i.e. malicious and non-malicious activity) and classification techniques (to classify all data into correct class categories). The approach, KM+1R, combines the k-means clustering with the OneR classification technique. The KDD Cup '99 set is used as a simulation dataset. The result shows that our proposed approach achieve a better accuracy and detection rate, particularly in reducing the false alarm.

Current anomaly detection is often associated with high false alarm with moderate accuracy and detection rates when it's unable to detect all types of attacks correctly. To overcome this problem, we propose an hybrid learning approach through combination of K-Means clustering and Naïve Bayes classification. The proposed approach will be cluster all data into the corresponding group before applying a classifier for classification purpose. An experiment is carried out to evaluate the performance of the proposed approach using KDD Cup'99 dataset. Result show that the proposed approach performed better in term of accuracy, detection rate with reasonable false alarm rate.
