Manganese (Mn) is key to oxygenic photosynthesis as it catalyzes the splitting of water in photosystem II and functions as cofactor of multiple enzymes. A single ABC-type transporter, MntCAB, is so far established for the uptake of the metal under limiting conditions in cyanobacteria. It is unknown, how Mn is imported under replete conditions. We identified two proteins in the model cyanobacterium Synechocystis sp. PCC 6803, which are homologous to the unknown protein family 0016 (UPF0016) member manganese exporter (Mnx). In contrast to Mnx, which consists of six transmembrane domains, the new candidate proteins contain only three transmembrane domains. Hence, we named them hemi manganese exporter (Hmx) 1 and 2. Knock-out mutants in hmx1 and/or hmx2 showed sensitivity toward low Mn supplementation, and reduced intracellular Mn pools. Additional deletion of mntC hindered the cells to thrive unless external Mn was added and enhanced the depletion of their intracellular Mn pool. In accordance with the observed localization of Hmx1 and Hmx2 in the plasma membrane, we postulate a Mn uptake function for a heteromeric Hmx1/2 across the plasma membrane under a wide range of Mn concentrations and a supporting role for the MntCAB system under Mn-limiting conditions. On the basis of their phylogenies, we propose that Hmx1 and Hmx2 are the ancestral progenitors of eukaryote-type UPF0016 proteins with six transmembrane domains. The Mn transport function of Hmx1/2 underscores this as fundamental ancient feature of the UPF0016 family. Likely, Hmx1 and Hmx2 coevolved with the internalization of the oxygen-evolving complex.

Large datasets of greenhouse gas and energy surface-atmosphere fluxes measured with the eddy-covariance technique (e.g., FLUXNET2015, AmeriFlux BASE) are widely used to benchmark models and remote-sensing products. This study addresses one of the major challenges facing model-data integration: To what spatial extent do flux measurements taken at individual eddy-covariance sites reflect model- or satellite-based grid cells? We evaluate flux footprints—the temporally dynamic source areas that contribute to measured fluxes—and the representativeness of these footprints for target areas (e.g., within 250–3000 m radii around flux towers) that are often used in flux-data synthesis and modeling studies. We examine the land-cover composition and vegetation characteristics, represented here by the Enhanced Vegetation Index (EVI), in the flux footprints and target areas across 214 AmeriFlux sites, and evaluate potential biases as a consequence of the footprint-to-target-area mismatch. Monthly 80% footprint climatologies vary across sites and through time ranging four orders of magnitude from 103 to 107 m2 due to the measurement heights, underlying vegetation- and ground-surface characteristics, wind directions, and turbulent state of the atmosphere. Few eddy-covariance sites are located in a truly homogeneous landscape. Thus, the common model-data integration approaches that use a fixed-extent target area across sites introduce biases on the order of 4%–20% for EVI and 6%–20% for the dominant land cover percentage. These biases are site-specific functions of measurement heights, target area extents, and land-surface characteristics. We advocate that flux datasets need to be used with footprint awareness, especially in research and applications that benchmark against models and data products with explicit spatial information. Break-off of part of the down-going plate during continental collision occurs due to tensile stresses built-up between the deep and shallow slab, for which buoyancy is increased because of continental-crust subduction. Break-off governs the subsequent orogenic evolution but real-time observations are rare as it happens over geologically short times. Here we present a finite-frequency tomography, based on jointly inverted local and remote earthquakes, for the Hindu Kush in Afghanistan, where slab break-off is ongoing. We interpret our results as crustal subduction on top of a northwards-subducting Indian lithospheric slab, whose penetration depth increases along-strike while thinning and steepening. This implies that break-off is propagating laterally and that the highest lithospheric stretching rates occur during the final pinching-off. In the Hindu Kush crust, earthquakes and geodetic data show a transition from focused to distributed deformation, which we relate to a variable degree of crust-mantle coupling presumably associated with break-off at depth.

CaTeNA – Climatic and Tectonic Natural Hazards in Central Asia – is an interdisciplinary, international project funded by the German Ministry of Education and Research to study natural hazards in Central Asia. Central Asia is one of the most tectonically active regions of the world and is influenced by both the west wind zone and monsoon. CaTeNA is examining the two most serious natural hazards arising from these conditions: Earthquakes and mass movements. The project goal is to better understand the underlying processes and triggering factors and to better estimate the resulting risks. For this purpose, CaTeNA localises tectonic faults and determines deformation rates and their changes. Focus is put on two of the most active fault systems, the Main Pamir Thrust and the Darvaz Fault crossing Tajikistan and Kyrgyzstan. We try to estimate recurrence intervals of large earthquakes and to understand their relationship to mass movements using paleo-seismology, geomorphology and remote sensing. The current deformation field is characterised and quantified using the methods of space geodesy and seismology. The results will be incorporated into the openly accessible Central Asian Tectonic Database developed within the project, making it accessible to the public, stakeholders and decision-makers. They form the basis for a more accurate estimation of the risk for earthquakes and landslides. Another important project goal is the development and implementation of a dynamic risk assessment for landslides, including high-resolution, model-based precipitation and snowmelt maps. This allows for improved estimation of the effects of geological hazards on inhabited areas and traffic infrastructure. Direct and efficient risk communication is achieved through interactive visualisation based on a dynamic multilingual web GIS platform. Introduction: Cascades, which track the progressive stages of engagement on the path towards a successful outcome, are increasingly being employed to quantitatively assess progress towards targets associated with health and development responses. Maximizing the proportion of people with successful outcomes within a budget-constrained context requires identifying and implementing interventions that are not only effective, but also cost-effective. Methods: We developed a software application called the Cascade Analysis Tool that implements advanced analysis and optimization methods for understanding cascades, combined with the flexibility to enable application across a wide range of areas in health and development. The tool allows users to design the cascade, collate and enter data, and then use the built-in analysis methods in order to answer key policy questions, such as: understanding where the biggest drop-offs along the cascade are; visualizing how the cascade varies by population; investigating the impact of introducing a new intervention or scaling up/down existing interventions; and estimating how available funding should be optimally allocated among available interventions in order to achieve a variety of different objectives selectable by the user (such as optimizing cascade outcomes in target years). The Cascade Analysis Tool is available via a user-friendly web-based application, and comes with a user guide, a library of pre-made examples, and training materials. Discussion: Whilst the Cascade Analysis Tool is still in the early stages of existence, it has already shown promise in preliminary applications, and we believe there is potential for it to help make sense of the increasing quantities of data on cascades.

The use of open-Access, high-quality and multi-level data products by different user communities is crucial for the Integrated Carbon Observation System in order to achieve its scientific potential and societal value.

The EC method is central to the ecosystem component of the Integrated Carbon Observation System, a monitoring network for greenhouse gases across the European Continent. The data processing sequence applied to the collected raw data is complex, and multiple robust options for the different steps are often available. For Integrated Carbon Observation System and similar networks, the standardisation of methods is essential to avoid methodological biases and improve comparability of the results. We introduce here the steps of the processing chain applied to the eddy covariance data of Integrated Carbon Observation System stations for the estimation of final CO2, water and energy fluxes, including the calculation of their uncertainties. The selected methods are discussed against valid alternative options in terms of suitability and respective drawbacks and advantages. The main challenge is to warrant standardised processing for all stations in spite of the large differences in e.g. ecosystem traits and site conditions. The main achievement of the Integrated Carbon Observation System eddy covariance data processing is making CO2 and energy flux results as comparable and reliable as possible, given the current micrometeorological understanding and the generally accepted state-of-the-art processing methods.

The nucleation area of the series of M6 events in Parkfield has been shown to be characterized by low b-values throughout the seismic cycle. Since low b-values represent high differential stresses, the asperity structure seems to be always stably stressed and even unaffected by the latest main shock in 2004. However, because fault loading rates and applied shear stress vary with time, some degree of temporal variability of the b-value within stable blocks is to be expected. We discuss in this study adequate techniques and uncertainty treatment for a detailed analysis of the temporal evolution of b-values. We show that the derived signal for the Parkfield asperity correlates with changes in surface creep, suggesting a sensitive time resolution of the b-value stress meter, and confirming near-critical loading conditions within the Parkfield asperity

Interactions of desert dust and air pollution over the eastern Mediterranean (EM) have been studied, focusing on two distinct dust transport events on 22 and 28 September 2011. The atmospheric chemistry-climate model EMAC has been used at about 50 km grid spacing, applying an on-line dust emission scheme and calcium as a proxy for dust reactivity. EMAC includes a detailed tropospheric chemistry mechanism, aerosol microphysics and thermodynamics schemes to describe dust aging. The model is evaluated using ground-based observations for aerosol concentrations and aerosol optical depth (AOD) as well as satellite observations. Simulation results and back trajectory analysis show that the development of synoptic disturbances over the EM can enhance dust transport from the Sahara and Arabian deserts in frontal systems that also carry air pollution to the EM. The frontal systems are associated with precipitation that controls the dust removal. Our results show the importance of chemical aging of dust, which increases particle size, dust deposition and scavenging efficiency during transport, overall reducing the lifetime relative to non-aged dust particles. The relatively long travel periods of Saharan dust result in more sustained aging compared to Arabian dust. Sensitivity simulations indicate 3 times more dust deposition of aged relative to pristine dust, which significantly decreases the dust lifetime and loading.

Growth and differentiation of multicellular systems is orchestrated by spatially restricted gene expression programs in specialized subpopulations. The targeted manipulation of such processes by synthetic tools with high-spatiotemporal resolution could, therefore, enable a deepened understanding of developmental processes and open new opportunities in tissue engineering. Here, we describe the first red/far-red light-triggered gene switch for mammalian cells for achieving gene expression control in time and space. We show that the system can reversibly be toggled between stable on- and off-states using short light pulses at 660 or 740 nm. Red light-induced gene expression was shown to correlate with the applied photon number and was compatible with different mammalian cell lines, including human primary cells. The light-induced expression kinetics were quantitatively analyzed by a mathematical model. We apply the system for the spatially controlled engineering of angiogenesis in chicken embryos. The system’s performance combined with cell- and tissue-compatible regulating red light will enable unprecedented spatiotemporally controlled molecular interventions in mammalian cells, tissues and organisms.

Dust devils – convective vortices made visible by the dust and debris they entrain – are common in arid environments and have been observed on Earth and Mars. Martian dust devils have been identified both in images taken at the surface and in remote sensing observations from orbiting spacecraft. Observations from landing craft and orbiting instruments have allowed the dust devil translational forward motion (ground velocity) to be calculated, but it is unclear how these velocities relate to the local ambient wind conditions, for (i) only model wind speeds are generally available for Mars, and (ii) on Earth only anecdotal evidence exists that compares dust devil ground velocity with ambient wind velocity. If dust devil ground velocity can be reliably correlated to the ambient wind regime, observations of dust devils could provide a proxy for wind speed and direction measurements on Mars. Hence, dust devil ground velocities could be used to probe the circulation of the martian boundary layer and help constrain climate models or assess the safety of future landing sites. We present results from a field study of terrestrial dust devils performed in the southwest USA in which we measured dust devil horizontal velocity as a function of ambient wind velocity. We acquired stereo images of more than a 100 active dust devils and recorded multiple size and position measurements for each dust devil. We used these data to calculate dust devil translational velocity. The dust devils were within a study area bounded by 10 m high meteorology towers such that dust devil speed and direction could be correlated with the local ambient wind speed and direction measurements.

In situ (mobile) sampling of 33 natural dust devil vortices reveals very high total suspended particle (TSP) mean values of 296 mg m−3 and fine dust loadings (PM10) mean values ranging from 15.1 to 43.8 mg m−3 (milligrams per cubic meter). Concurrent three-dimensional wind profiles show mean tangential rotation of 12.3 m s−1 and vertical uplift of 2.7 m s−1 driving mean vertical TSP flux of 1689 mg m−3 s−1 and fine particle flux of ~1.0 to ~50 mg m−3 s−1. Peak PM10 dust loading and flux within the dust column are three times greater than mean values, suggesting previous estimates of dust devil flux might be too high. We find that deflation rates caused by dust devil erosion are ~2.5–50 μm per year in dust devil active zones on Earth. Similar values are expected for Mars, and may be more significant there where competing erosional mechanisms are less likely.

In contrast to bare silicone rubber, OptiChem samples did not become colonized upon reimplantation despite the fact that surrounding tissues were always culture-positive. We conclude that the commercial OptiChem coating considerably slows down bacterial biofilm formation both in vitro and in vivo, making it an attractive candidate for biomaterials implant coating.

However, bacterial adhesion remains of the order of 10(4)cm(-2). It is unknown whether this density of bacteria will eventually grow into a biofilm. This study investigates the kinetics of staphylococcal biofilm formation on a commercially produced, robust, cross-linked PEG-based polymer coating (OptiChem) in vitro and in vivo. OptiChem inhibits biofilm formation in vitro, and although adsorption of plasma proteins encourages biofilm formation, microbial growth kinetics are still strongly delayed compared to uncoated glass. In vivo, OptiChem-coated and bare silicone rubber samples were inserted into an infected murine subcutaneous pocket model. To elucidate human induced changes of aerosol load and composition in the atmosphere, a coupled aerosol and gas-phase chemistry transport model of the troposphere and lower stratosphere has been used. The present 3-D modeling study focuses on aerosol chemical composition change since preindustrial times considering the secondary organic aerosol formation together with all other main aerosol components including nitrate. In particular, we evaluate non-sea-salt sulfate (nss-SO4=), ammonium (NH4+), nitrate (NO3¿), black carbon (BC), sea-salt, dust, primary and secondary organics (POA and SOA) with a focus on the importance of secondary organic aerosols. Our calculations show that the aerosol optical depth (AOD) has increased by about 21% since preindustrial times. This enhancement of AOD is attributed to a rise in the atmospheric load of BC, nss-SO4=, NO3, POA and SOA by factors of 3.3, 2.6, 2.7, 2.3 and 1.2, respectively, whereas we assumed that the natural dust and sea-salt sources remained constant. The nowadays increase in carbonaceous aerosol loading is dampened by a 34¿42% faster conversion of hydrophobic to hydrophilic carbonaceous aerosol leading to higher removal rates. These changes between the various aerosol components resulted in significant modifications of the aerosol chemical composition. The relative importance of the various aerosol components is critical for the aerosol climatic effect, since atmospheric aerosols behave differently when their chemical composition changes. According to this study, the aerosol composition changed significantly over the different continents and with height since preindustrial times. The presence of anthropogenically emitted primary particles in the atmosphere facilitates the condensation of the semi-volatile species that form SOA onto the aerosol phase, particularly in the boundary layer. The SOA burden that is dominated by the natural component has increased by 24% while its contribution to the AOD has increased by 11%

Real-time measurements of ammonia, nitric acid, hydrochloric acid, sulfur dioxide and the water-soluble inorganic aerosol species, ammonium, nitrate, chloride, and sulfate were performed at a pasture site in the Amazon Basin (Rondônia, Brazil). The measurements were made during the late dry season (biomass burning), the transition period, and the onset of the wet season (clean conditions) using a wet-annular denuder (WAD) in combination with a Steam-Jet Aerosol Collector (SJAC). Measurements were conducted from 12 September to 14 November 2002 within the framework of LBA-SMOCC (Large-Scale Biosphere Atmosphere Experiment in Amazonia - Smoke Aerosols, Clouds, Rainfall, and Climate: Aerosols From Biomass Burning Perturb Global and Regional Climate). Real-time data were combined with measurements of sodium, potassium, calcium, magnesium, and low-molecular weight (LMW) polar organic acids determined on 12-, 24-, and 48-hours integrated filter samples. The contribution of inorganic species to the fine particulate mass (Dp = 2.5 µm) was frequently below 20% by mass, indicating the preponderance of organic matter. The measured concentration products of NH3 × HNO3 and NH3 × HCl persistently remained below the theoretical equilibrium dissociation constants of the NH3/HNO3/NH4NO3 and NH3/HCl/NH4Cl systems during daytime (RH <90%). The application of four thermodynamic equilibrium models (EQMs) indicates that the fine mode aerosol anions NO3 -, Cl-, and SO4 2- were balanced predominantly by mineral cations (particularly pyrogenic K+) during daytime. At nighttime (RH > 90%) fine-mode NH4NO3 and NH4Cl are predicted to be formed in the aqueous aerosol phase. Probably, Cl- was driven out of the aerosol phase largely by reaction of pyrogenic KCl with HNO3 and H2SO4. Numerical and statistical predictions of simplified models are linearly combined in a sensitivity study to identify the crucial parameters that are needed to enhance predictability of the El Nino-Southern Oscillation (ENSO) phenomenon. The results indicate that the ENSO prediction skill of the simplified models can be improved. The profit achieved strongly depends on the phase information that is utilized by the forecast combination and is inherent in predictions of a quasi-periodic process such as ENSO. The simplest forecast combination that still yields useful forecasts at longer lead times of about half of the ENSO period ( 18 - 24 months) is the combination of two persistence forecast schemes. For the prediction period 1982 - 2003, that is the persistence of a sea surface temperature anomaly (SSTA) index area at 60degreesS, 180degreesW and the Nino-3 index SSTA. The level of skill improvement critically depends on the prediction schemes and prediction period, as well as on the period from which the combination weights are derived. Differences between combination forecast and hindcast are minimized if the statistical weights are derived from a time period that is characterized by an ENSO statistic that is close to the prediction period. In this study the prediction period has simply been halved for the sake of simplicity to derive the statistical weights, which is sufficient for predicting the 1980s and 1990s ( with each other), but not for predicting, for example, the 1970s. A technique has been developed to apply force to the antibody-antigen complex in a solid-phase immunoassay. Force was applied to the immunochemical complex by labeling the secondary antibody with a magnetically susceptible, micrometer-size particle and placing the assay chamber in a magnetic field of defined magnitude and orientation. The force was strong enough to displace weakly bound particles but was not strong enough to rupture the immunochemical complex. The number of particles bound to the surface after applying the differentiation force was related to the analyte concentration, thus an optical detection scheme was developed for counting the number of particles on the surface. The sensitivity of the force differentiation assay was demonstrated to be one to two orders of magnitude higher than conventional solid-phase immunoassay techniques for model protein, virus, and bacterial analytes, with 99% specificity. The enhanced sensitivity of this assay appears to result from lowering the assay background through the identification of weakly adhesive, nonspecific interactions. (C) 2000 Academic Press.

Dust devils – convective vortices made visible by the dust and debris they entrain – are common in arid environments and have been observed on Earth and Mars. Martian dust devils have been identified both in images taken at the surface and in remote sensing observations from orbiting spacecraft. Observations from landing craft and orbiting instruments have allowed the dust devil translational forward motion (ground velocity) to be calculated, but it is unclear how these velocities relate to the local ambient wind conditions, for (i) only model wind speeds are generally available for Mars, and (ii) on Earth only anecdotal evidence exists that compares dust devil ground velocity with ambient wind velocity. If dust devil ground velocity can be reliably correlated to the ambient wind regime, observations of dust devils could provide a proxy for wind speed and direction measurements on Mars. Hence, dust devil ground velocities could be used to probe the circulation of the martian boundary layer and help constrain climate models or assess the safety of future landing sites. We present results from a field study of terrestrial dust devils performed in the southwest USA in which we measured dust devil horizontal velocity as a function of ambient wind velocity. We acquired stereo images of more than a hundred active dust devils and recorded multiple size and position measurements for each dust devil. We used these data to calculate dust devil translational velocity. The dust devils were within a study area bounded by 10 m high meteorology towers such that dust devil speed and direction could be correlated with the local ambient wind speed and direction measurements. Daily (10:00 to 16:00 local time) and two-hour averaged dust devil ground speeds correlate well with ambient wind speeds averaged over the same period. Unsurprisingly, individual measurements of dust devil ground speed match instantaneous measurements of ambient wind speed more poorly; a 20-minute smoothing windo

We present results from the first high-resolution near-surface profiles conducted on dust devil wind fields. These results are integrated with extensive geologic mapping to understand the factors that influence vortex generation and erosive efficacy.

The nucleation area of the series of M6 events in Parkfield has been shown to be characterized by low b-values throughout the seismic cycle. Since low b-values represent high differential stresses, the asperity structure seems to be always stably stressed and even unaffected by the latest main shock in 2004. However, because fault loading rates and applied shear stress vary with time, some degree of temporal variability of the b-value within stable blocks is to be expected. We discuss in this study adequate techniques and uncertainty treatment for a detailed analysis of the temporal evolution of b-values. We show that the derived signal for the Parkfield asperity correlates with changes in surface creep, suggesting a sensitive time resolution of the b-value stress meter, and confirming near-critical loading conditions within the Parkfield asperity
