Two recombinant barley cystatins, HvCPI5 and HvCPI6, have been tested in vitro against promastigotes and intracellular amastigotes of Leishmania infantum in the J774 monocytic cell line. Toxicity of cystatins for J774 cells was also determined. In addition, a comparison between direct counts of intracellular amastigotes and quantitation of burden by Q-PCR was carried out. Low concentrations (2 lM) from both cystatins were unable to inhibit promastigote replication. HvCPI5 was toxic for mammalian cells; 0.1 lM reduced by more than 50% the cell viability. On the contrary, HvCPI6 did not exhibit any toxicity for J774 cells up to 6 lM and inhibited the intracellular amastigote multiplication. Dose–response analysis showed that 4.8 lM HvCPI6 reduced by >90% the intracellular parasite load and had an approximate IC50 value of 1.5 lM. Comparable results were obtained by direct counting of intracellular amastigotes and Q-PCR. Ambient energy harvesting has great potential to contribute to sustainable development and address growing environmental challenges. Converting waste energy from energy-intensive processes and systems (e.g. combustion engines and furnaces) is crucial to reducing their environmental impact and achieving net-zero emissions. Compact energy harvesters will also be key to powering the exponentially growing smart devices ecosystem that is part of the Internet of Things, thus enabling futuristic applications that can improve our quality of life (e.g. smart homes, smart cities, smart manufacturing, and smart healthcare). To achieve these goals, innovative materials are needed to efficiently convert ambient energy into electricity through various physical mechanisms, such as the photovoltaic effect, thermoelectricity, piezoelectricity, triboelectricity, and radiofrequency wireless power transfer. By bringing together the perspectives of experts in various types of energy harvesting materials, this Roadmap provides extensive insights into recent advances and present challenges in the field. Additionally, the Roadmap analyses the key performance metrics of these technologies in relation to their ultimate energy conversion limits. Building on these insights, the Roadmap outlines promising directions for future research to fully harness the potential of energy harvesting materials for green energy anytime, anywhere.

The economic downturn caused by the COVID-19 pandemic and its subsequent restrictions has had an alarming effect on Australians' mental health. Particularly worrying is the increase in high mental distress among parents, especially among non-employed fathers and parents of primary school aged children. Financial stress and work-family conflict tend to be the two major sources of mental distress for parents. Using data from the Melbourne Institute's Taking the Pulse of the Nation survey and the Household, Income and Labour Dynamics in Australia (HILDA) Survey, this Research Insight examines the surge in mental distress in parents compared to pre-COVID. The study identifies who has been most impacted, looking at how it impacts mothers and fathers, and how employment status and the age of the child can influence mental distress.

The past five years has seen a steady decline in private health insurance (PHI) membership in Australia. With rising premium prices and out-of-pocket costs, private healthcare has become increasingly unaffordable in an era of low wage growth. Given the private health sector relies heavily on PHI funding, this Research Insight explores how the decline in private health membership has affected the private hospital sector in Australia.

Of the close to 5 million parents with children under 18 in Australia, 24 percent have reported high rates of mental distress since the start of the pandemic. This has persisted well beyond the end of local lockdowns.

Emerging pollutants represent a new global problem for water quality. As these compounds get into the environment, they cause severe threats to aquatic environments and human health and are typically resistant to conventional wastewater treatments. In this work, TiO2 nanoparticles surface was functionalized with silver (Ag) nanoparticles, and solvent cast and electrospun membranes of poly (vinylidene fluoride-co-hexafluoropropylene) (PVDF-HFP) were prepared with different concentrations of TiO2 and Ag–TiO2 to produce a multifunctional material. The photocatalytic activity of the nanocomposites was evaluated through the degradation of norfloxacin under ultraviolet (UV) and visible radiation. It is shown that nanocomposites with Ag–TiO2 show the highest degradation efficiencies: 64.2% under UV and 80.7% under visible radiation, for 90 and 300 min, respectively. Furthermore, the recyclability of the membranes has also been demonstrated. Finally, it is shown the antimicrobial activity of the nanocomposite membranes, demonstrating the suitability of the Ag–TiO2/PVDF-HFP nanocomposites as multifunctional photocatalytic and antimicrobial membranes for water remediation applications.

[EN] Thermal degradation of chitosan with varying deacetylation degree (DD) ranging between 50 and 85% was analyzed by dynamic thermogravimetric analysis at different heating rates. The present study focused on the temperature range between 500 and 800 K, above water evaporation. Thermal degradation showed a main degradation stage in this temperature interval with a second stage that appeared in the weight derivative curves as a shoulder in the high temperature side of the main peak with increasing intensity as the DD decreased. The Kissinger and isoconversional Ozawa-Flynn-Wall models were employed to evaluate the Ea of both thermal degradation processes. Different kinetic models were tested to computer simulate the thermogravimetric traces calculating the model parameters with a non-linear least squares fitting routine. The Sestack-Berggren model allowed reproducing accurately the overlapping of the two degradation mechanisms and calculating the mass fraction lost in each of them revealing the coupling between the two degradation mechanisms.

[EN] Titanium and its alloys are widely used in medical implants because of their excellent properties. However, bacterial infection is a frequent cause of titanium-based implant failure and also compromises its osseointegration. In this study, we report a new simple method of providing titanium surfaces with antibacterial properties by alternating antibacterial chitosan domains with titanium domains in the micrometric scale. Surface micro grooves were etched on pure titanium disks at intervals of 60 pm using a modified 3D printer and were then coated with chitosan antibacterial polysaccharide. The dimensions of the patterned microgrooves made it possible to fix the chitosan domains to the titanium substrate without the need for covalent bonding. These domains were stable after 5 days of immersion in water and reduced the surface contact angle. Preliminary cell adhesion assays demonstrated that MC3T3-E1 pre-osteoblasts preferentially adhered to the titanium regions, while C2C12 myoblasts were uniformly distributed over the whole surface.

Since the first magnetoelectric polymer composites were fabricated more than a decade ago, there has been a reluctance to use piezoelectric polymers other than poly(vinylidene fluoride) and its copolymers due to their well-defined piezoelectric mechanism and high piezoelectric coefficients that lead to superior magnetoelectric coefficients of >1 V cm-1 Oe-1. This is the current situation despite the potential for other piezoelectric polymers, such as natural biopolymers, to bring unique, added-value properties and functions to magnetoelectric composite devices. Here we demonstrate a cellulose-based magnetoelectric laminate composite that produces considerable magnetoelectric coefficients of ≈1.5 V cm-1 Oe-1, comprising a Fano resonance that is ubiquitous in the field of physics, such as photonics, though never experimentally observed in magnetoelectric composites. The work successfully demonstrates the concept of exploring new advances in using biopolymers in magnetoelectric composites, particularly cellulose, which is increasingly employed as a renewable, low-cost, easily processable and degradable material.Magnetoelectric materials by converting a magnetic input to a voltage output holds promise in contactless electrodes that find applications from energy harvesting to sensing. Zong et al. report a promising laminate composite that combines a piezoelectric biopolymer, cellulose, and a magnetic material.

[EN] Polymeric scaffolds used in regenerative therapies are implanted in the damaged tissue and submitted to repeated loading cycles. In the case of articular cartilage engineering, an implanted scaffold is typically subjected to long-term dynamic compression. The evolution of the mechanical properties of the scaffold during bioresorption has been deeply studied in the past, but the possibility of failure due to mechanical fatigue has not been properly addressed. Nevertheless, the macroporous scaffold is susceptible to failure after repeated loading-unloading cycles. In this work fatigue studies of polycaprolactone scaffolds were carried by subjecting the scaffold to repeated compression cycles in conditions simulating the scaffold implanted in the articular cartilage. The behavior of the polycaprolactone sponge with the pores filled with a poly(vinyl alcohol) gel simulating the new formed tissue within the pores was compared with that of the material immersed in water. Results were analyzed with Morrow's criteria for failure and accurate fittings are obtained just up to 200 loading cycles. It is also shown that the presence of poly(vinyl alcohol) increases the elastic modulus of the scaffolds, the effect being more pronounced with increasing the number of freeze/thawing cycles.

The degree of development and operability of the indicators for the Marine Strategy Framework Directive (MSFD) using Descriptor 1 (D1) Biological Diversity was assessed. To this end, an overview of the relevance and degree of operability of the underlying parameters across 20 European countries was compiled by analysing national directives, legislation, regulations, and publicly available reports. Marked differences were found between countries in the degree of ecological relevance as well as in the degree of implementation and operability of the parameters chosen to indicate biological diversity. The best scoring EU countries were France, Germany, Greece and Spain, while the worst scoring countries were Italy and Slovenia. No country achieved maximum scores for the implementation of MSFD D1. The non-EU countries Norway and Turkey score as highly as the top-scoring EU countries. On the positive side, the chosen parameters for D1 indicators were generally identified as being an ecologically relevant reflection of Biological Diversity. On the negative side however, less than half of the chosen parameters are currently operational. It appears that at a pan-European level, no consistent and harmonized approach currently exists for the description and assessment of marine biological diversity. The implementation of the MSFD Descriptor 1 for Europe as a whole can therefore at best be marked as moderately successful.

Up to three degradation steps were observed in the TG data, corresponding to water dehydration reaction at temperatures below 100 C, loss of side groups formed between the amine groups of chitosan and trifluoroacetic acid between 150 and 270 C and chitosan thermal degradation that starts around 250 C and goes up to 400 C. The Kissinger model was employed to evaluate the activation energies of the electrospun membranes during isothermal experiments and revealed that thermal degradation activation energy increases for the samples processed by electrospinning and subsequent neutralization and cross-linking treatments with respect to the neat chitosan powder.

Fiber mats of randomly and aligned fibers were obtained with fixed and rotating ground collectors and fibrils were composed by thin ribbons whose width depends on electrospinning conditions; fibrils with 721 nm up to 2.12 ¿m width were achieved. After cross-linking with glutaraldehyde, ¿-elastin can uptake as much as 1700 % of PBS solution and a slight increase on fiber thickness was observed. The glass transition temperature of electrospun fiber mats was found to occur at ~80 °C. Moreover, ¿-Elastin showed to be a perfect elastomeric material, and no mechanical hysteresis was found in cycle mechanical measurements. The elastic modulus obtained for random and aligned fibers mats in a PBS solution was 330±10 kPa and 732±165 kPa, respectively. Finally, the electrospinning and cross-linking process does not inhibit MC-3T3-E1 cell adhesion. Cell culture results showed good cell adhesion and proliferation in the cross-linked elastin fiber mats.

The influence of the polymer solution concentration and processing parameters, such as electric field, flow rate and inner needle diameter, on microparticle size and distribution has been studied. Polymer concentration is the most influential parameter on PVDF microparticle formation. Higher concentrations promote the formation of fibers while dilute or semi dilute concentrations favor the formation of PVDF microparticles with average diameters ranging between 0.81 +/- 0.34 and 5.55 +/- 2.34 mu m. Once the formation of microparticles is achieved, no significant differences were found with the variation of other electrospray processing parameters. The electroactive beta-phase content, between 63 and 74%, and the crystalline phase content, between 45 and 55%, are mainly independent of the processing parameters. Finally, MC-3T3-E1 cell adhesion on the PVDF microparticles is assessed, indicating their potential use for biomedical applications.

Taguchi's methodology was followed to determine which parameters have the strongest influence on mean fiber diameter and fiber homogeneity. Chitosan nanofibers obtained with this procedure were water soluble due to the protonation of amine side groups but were successfully neutralized by immersion in calcium carbonate solutions. It was established that fiber diameter is mainly determined by the solution concentration and the distance from the needle to the collector while other parameters have less influence. The set of parameters that produce the thinnest fibers were a concentration of 8 wt%, a TFA/DCM ratio of 80:20 (v/v%), a voltage of 30 kV, a flow rate of 6.0 mL/h, a gap distance of 10 cm, using a needle diameter of 0.5, allowing to produce randomly oriented mats with a mean fiber thickness of 66 nm. © 2012 Society of Plastics Engineers.

We developed hybrid materials comprising of silica and stimuli responsive polymers (SRPs) to demonstrate the feasibility of SRPs as actuation elements in functional nanostructured materials. We successfully demonstrated that the hybrid membranes comprising of silica and SRPs can function as molecular filters with reversible permeability and good stability. We dispersed these SRPs in a silica matrix at a molecular level using sol-gel process. The encapsulated SRP molecules to act as nano- valves whose permeability can be controlled by cycling through the lower critical solution temperature (LCST) of the SRPs. Micropatterning studies on hybrid membranes suggest that a wide variety of water-soluble species can be patterned without need for a specific chemisorption or affinity label using localized heating with a UV laser. Developed porous silica nanocomposite materials and modified the surface with SRPs using atom transfer radical polymerization and demonstrated that the size and surface energy of the pores can be externally and reversibly controlled to dynamically modulate the adsorption and transport of molecular species. These have potential applications in the areas of separations, controlled drug delivery, environmental remediation, bioassays, biosensing, tissue engineering. Using a model system of silica and PNIPAAM we have demonstrated design principles and synthetic methods are applicable to a wide variety of porous structures and polymers (or small molecules) which are reversibly sensitive to different external stimuli such as temperature, light, electricity, pH, solutes, or enzymatic transformations.

While the extension of aquaculture along the coasts of the Mediterranean islands is today an economic priority, no data is available on the impact of these facilities on the littoral environment in general, and in particular on the Posidonia oceanica meadows that are responsible for the richness and diversity of these biotopes. The impact of fish farming facilities in the marine littoral environment is assessed at three Mediterranean sites in Corsica and Sardinia. Various parameters are considered: (i) the amount of light available, (ii) the nature of the sediment, and (iii) the vitality of the Posidonia oceanica beds. The findings confirm the impact of the aquaculture facilities within the water column (increase in turbidity, enrichment of the sediment in organic matter and nutrients), and it would appear that the Posidonia oceanica beds also undergo significant alterations.
