© The Royal Society of Chemistry 2018. Lignin conversion to renewable chemicals is a promising means to improve the economic viability of lignocellulosic biorefineries. Alkaline aerobic oxidation of lignin has long been employed for production of aromatic compounds such as vanillin and syringaldehyde, but this approach primarily focuses on condensed substrates such as Kraft lignin and lignosulfonates. Conversely, emerging lignocellulosic biorefinery schemes enable the production of more native-like, reactive lignin. Here, we revisit alkaline aerobic oxidation of highly reactive lignin substrates to understand the impact of reaction conditions and catalyst choice on product yield and distribution. The oxidation of native poplar lignin was studied as a function of temperature, NaOH loading, reaction time, and oxygen partial pressure. Besides vanillin and syringaldehyde, other oxidation products include acetosyringone and vanillic, syringic, and p-hydroxybenzoic acids. Reactions with vanillin and syringaldehyde indicated that these compounds are further oxidized to non-aromatic carboxylic acids during alkaline aerobic oxidation, with syringaldehyde being substantially more reactive than vanillin. The production of phenolic compounds from lignin is favored by high NaOH loadings and temperatures, but short reaction times, as the products degrade rapidly, which is further exacerbated by the presence of oxygen. Under optimal conditions, a phenolic monomer yield of 30 wt% was obtained from poplar lignin. Testing a range of catalysts showed that Cu-containing catalysts, such as CuSO4 and LaMn0.8Cu0.2O3, accelerate product formation; specifically, the catalyst does not increase the maximum yield, but expands the operating window in which high product yields are obtainable. We also demonstrate that other native and isolated lignin substrates that are significantly chemically modified are effectively converted to phenolic compounds. © 2019, The Author(s). The ratio of syringyl (S) and guaiacyl (G) units in lignin has been regarded as a major factor in determining the maximum monomer yield from lignin depolymerization. This limit arises from the notion that G units are prone to C-C bond formation during lignin biosynthesis, resulting in less ether linkages that generate monomers. This study uses reductive catalytic fractionation (RCF) in flow-through reactors as an analytical tool to depolymerize lignin in poplar with naturally varying S/G ratios, and directly challenges the common conception that the S/G ratio predicts monomer yields. Rather, this work suggests that the plant controls C-O and C-C bond content by regulating monomer transport during lignin biosynthesis. Overall, our results indicate that additional factors beyond the monomeric composition of native lignin are important in developing a fundamental understanding of lignin biosynthesis.

Reductive catalytic fractionation (RCF) has emerged as an effective biomass pretreatment strategy to depolymerize lignin into tractable fragments in high yields. We investigate the RCF of corn stover, a highly abundant herbaceous feedstock, using carbon-supported Ru and Ni catalysts at 200 and 250 °C in methanol and, in the presence or absence of an acid cocatalyst (H₃PO₄ or an acidified carbon support). Three key performance variables were studied: (1) the effectiveness of lignin extraction as measured by the yield of lignin oil, (2) the yield of monomers in the lignin oil, and (3) the carbohydrate retention in the residual solids after RCF. The monomers included methyl coumarate/ferulate, propyl guaiacol/syringol, and ethyl guaiacol/syringol. The Ru and Ni catalysts performed similarly in terms of product distribution and monomer yields. The monomer yields increased monotonically as a function of time for both temperatures. At 6 h, monomer yields of 27.2 and 28.3% were obtained at 250 and 200 °C, respectively, with Ni/C. The addition of an acid cocatalysts to the Ni/C system increased monomer yields to 32% for acidified carbon and 38% for phosphoric acid at 200 °C. The monomer product distribution was dominated by methyl coumarate regardless of the use of the acid cocatalysts. The use of phosphoric acid at 200 °C or the high temperature condition without acid resulted in complete lignin extraction and partial sugar solubilization (up to 50%) thereby generating lignin oil yields that exceeded the theoretical limit. In contrast, using either Ni/C or Ni on acidified carbon at 200 °C resulted in moderate lignin oil yields of ca. 55%, with sugar retention values >90%. Notably, these sugars were amenable to enzymatic digestion, reaching conversions >90% at 96 h.
