Abstract: Kidney disease is often progressive, and patients experience diminished health-related quality of life. In addition, the impact of the coronavirus (COVID-19) pandemic, and its associated restrictions, has brought many additional burdens. It is therefore essential that effective and affordable systems are explored to improve the psychological health of this group that can be delivered safely during the COVID-19 pandemic. The aim of this study is to support a new service development project in partnership with the UK’s leading patient support charity Kidney Care UK by implementing the four-session Compassionate Mindful Resilience (CMR) programme, developed by MindfulnessUK, and explore its effectiveness for patients with stage 4 or 5 chronic kidney disease or have received a kidney transplant. The study will utilise a quasi-experimental, pretest/posttest design to measure the effect of the CMR programme on anxiety, depression, self-compassion, the ability to be mindful, wellbeing, and resilience, using pre- and posttests, alongside a qualitative exploration to explore factors influencing the feasibility, acceptability, and suitability of the intervention, with patients (and the Mindfulness Teacher) and their commitment to practice. Outcomes from this study will include an evidence-based mindfulness and compassion programme for use with people with kidney disease, which is likely to have applicability across other chronic diseases.

2. Quasi-periodic whistler mode emission in the plasmasphere as observed by the DSX spacecraft

Abstract: We describe the quasi-periodic (QP) whistler-mode emissions found in the plasmasphere as detected by electric and magnetic instrumentation onboard the Demonstration and Science Experiments (DSX) spacecraft in medium Earth orbit. Over the course of the nearly 2-year mission, at least 45 episodes of whistler mode QP emissions were detected by the Broad Band Receiver (BBR) onboard DSX. Episodes of QP emissions were identified by discrete events having a clear unambiguous periodic nature as detected by both the electric antennae and search coil magnetic sensors in the BBR survey data at 30 s temporal resolution. Most of the QP episodes occurred in a frequency range between 1 and 4 kHz, in a band previously identified by Van Allen Probes and Cluster investigators. However, episodes were also detected by DSX at higher frequencies - events in these episodes extending all the way to 15 kHz. We present our findings on these unusual high frequency events in the presentation herein. Specifically, these high frequency QP episodes tended to be observed near dawn/dusk when the spacecraft was at relatively high magnetic latitudes and on magnetic L-shells between 3 and 5. Another unusual feature of these episodes is that individual up-drifting events making up the episode were found to sometimes occur concurrently in time: The high frequency portion of one up-drifting ""polliwog-shaped"" event overlapped in time with the low frequency portion of the subsequent event. This behavior of the QP emissions has not been previously emphasized and we consider how this temporal concurrence relates to the source processes., We are very grateful from the support of the DSX project team and especially the operations team that kept the complex spacecraft flying successfully for 2 years. Abstract: To evaluate safety, dose response, and preliminary efficacy of reldesemtiv over 12 weeks in patients with amyotrophic lateral sclerosis (ALS). Methods: Patients (≤2 years since diagnosis) with slow upright vital capacity (SVC) of ≥60% were randomized 1:1:1:1 to reldesemtiv 150, 300, or 450 mg twice daily (bid) or placebo; active treatment was 12 weeks with 4-week follow-up. Primary endpoint was change in percent predicted SVC at 12 weeks; secondary measures included ALS Functional Rating Scale-Revised (ALSFRS-R) and muscle strength mega-score. Results: Patients (N = 458) were enrolled; 85% completed 12-week treatment. The primary analysis failed to reach statistical significance (p = 0.11); secondary endpoints showed no statistically significant effects (ALSFRS-R, p = 0.09; muscle strength mega-score, p = 0.31). Post hoc analyses pooling all active reldesemtiv-treated patients compared against placebo showed trends toward benefit in all endpoints (progression rate for SVC, ALSFRS-R, and muscle strength mega-score (nominal p values of 0.10, 0.01 and 0.20 respectively)). Reldesemtiv was well tolerated, with nausea and fatigue being the most common side effects. A dose-dependent decrease in estimated glomerular filtration rate was noted, and transaminase elevations were seen in approximately 5% of patients. Both hepatic and renal abnormalities trended toward resolution after study drug discontinuation. Conclusions: Although the primary efficacy analysis did not demonstrate statistical significance, there were trends favoring reldesemtiv for all three endpoints, with effect sizes generally regarded as clinically important. Tolerability was good; modest hepatic and renal abnormalities were reversible. The impact of reldesemtiv on patients with ALS should be assessed in a pivotal Phase 3 trial. Abstract: Spacecraft materials on orbit are subjected to the harsh weather of space. In particular, high-energy electrons alter the chemical structure of polymers and cause charge accumulation. Understanding the mechanisms of damage and charge dissipation is critical to spacecraft construction and operational anomaly resolution. Energetic particles in space plasma break molecular bonds in polymers and create radicals that can act as space charge traps. These electron-induced chemical changes also result in changes to the spectral absorption profile of polymers on orbit. Radicals react over time, either recreating identical bonds to those in the pristine material, leading to material recovery, or creating new bonds, resulting in a new material with new physical properties. Lack of knowledge about this dynamic aging is a major impediment to accurate modeling of spacecraft behavior over its mission life. This chapter first presents an investigation of the chemical and physical properties of polyimide films (PI, Kapton-H®) during and after irradiation with high-energy (90 keV) electrons. Second, the deleterious effects of space plasma on a spacecraft component level are presented. The results of this physical/chemical collaboration demonstrate the correlation of chemical changes in PI with the dynamic nature of spacecraft material aging.

Abstract: © Overman et al. Pharmacological targeting of transcription factors holds great promise for the development of new therapeutics, but strategies based on blockade of DNA binding, nuclear shuttling, or individual protein partner recruitment have yielded limited success to date. Transcription factors typically engage in complex interaction networks, likely masking the effects of specifically inhibiting single protein-protein interactions. Here, we used a combination of genomic, proteomic and biophysical methods to discover a suite of protein-protein interactions involving the SOX18 transcription factor, a known regulator of vascular development and disease. We describe a small-molecule that is able to disrupt a discrete subset of SOX18-dependent interactions. This compound selectively suppressed SOX18 transcriptional outputs in vitro and interfered with vascular development in zebrafish larvae. In a mouse pre-clinical model of breast cancer, treatment with this inhibitor significantly improved survival by reducing tumour vascular density and metastatic spread. Our studies validate an interactome-based molecular strategy to interfere with transcription factor activity, for the development of novel disease therapeutics.

Abstract: The radiation belts and plasma in the Earth’s magnetosphere pose hazards to satellite systems which restrict design and orbit options with a resultant impact on mission performance and cost. For decades the standard space environment specification used for spacecraft design has been provided by the NASA AE8 and AP8 trapped radiation belt models. There are well-known limitations on their performance, however, and the need for a new trapped radiation and plasma model has been recognized by the engineering community for some time. To address this challenge a new set of models, denoted AE9/AP9/SPM, for energetic electrons, energetic protons and space plasma has been developed. The new models offer significant improvements including more detailed spatial resolution and the quantification of uncertainty due to both space weather and instrument errors. Fundamental to the model design, construction and operation are a number of new data sets and a novel statistical approach which captures first order temporal and spatial correlations allowing for the Monte-Carlo estimation of flux thresholds for user-specified percentile levels (e.g., 50th and 95th) over the course of the mission. 10. Impact of lymph node status in patients with intrahepatic cholangiocarcinoma treated by major hepatectomy: a review of the National Cancer Database.

Abstract: IntroductionRoutine lymphadenectomy in the surgical treatment of intrahepatic cholangiocarcinoma (ICC) is not routinely performed. We aim to define predictive indicators of survival in patients with positive lymph nodes.MethodsThe National Cancer Data Base (NCDB) was queried for patients who underwent major hepatectomy for ICC between 1998 and 2011. Clinical and pathologic data were assessed using uni- and multi-variate analyses. A sub-analysis was performed on the 160 patients with positive lymph nodes.ResultsOf 849 patients with lymph node data, 57% had at least one lymph node examined. Median survival for lymph node negative patients was 37 months versus 15 months for lymph node positive patients. In lymph node positive patients, poorer survival was associated with not receiving chemotherapy (HR 1.83, p = 0.003), tumor size > 5 cm (p = 0.029), and older age (p < 0.0001). Lymph node positive patients age less than 45 had a median survival of 27 months.ConclusionsOverall survival in patients with lymph node metastases from ICC is poor. Adjuvant therapy was associated with a longer survival in lymph node positive patients, although prospective data are needed. Routine lymphadenectomy should be strongly considered to provide prognostic information and guidance for adjuvant therapy.

11. Marketing and sales: Optimization of a neglected relationship

Abstract: Purpose The purpose of this paper is to establish the importance of good coordination between marketing and sales. It aims to examine the research conducted to date, the area of marketing and sales integration and provide guidance to help integrate these functional areas better in industrial marketing firms. The paper seeks to develop a research agenda to continue this research. Design/methodology/approach A total of 21 interviews were collected through the use of qualitative research techniques and intensive interviews. The interactions between marketing and sales were modeled for during the marketing management process and the sales management process. Research limitations/implications More extensive sample surveys need to be conducted. Exploratory findings should be tested and possible situational differences identified. Practical implications Several required actions were identified for management. These fall into three categories: organizational design; communication; and human resources management. The primary suggestions are to make sure: product management functions effectively as a coordination mechanism; there is central coordination; the organizational structure is customer-centric; communication is timely and of high quality; and everyone in sales and marketing has an appreciation of the other role. Abstract: Reproducibility is a defining feature of science, but the extent to which it characterizes current research is unknown. We conducted replications of 100 experimental and correlational studies published in three psychology journals using high-powered designs and original materials when available. Replication effects were half the magnitude of original effects, representing a substantial decline. Ninety-seven percent of original studies had statistically significant results. Thirty-six percent of replications had statistically significant results; 47% of original effect sizes were in the 95% confidence interval of the replication effect size; 39% of effects were subjectively rated to have replicated the original result; and if no bias in original results is assumed, combining original and replication results left 68% with statistically significant effects. Correlational tests suggest that replication success was better predicted by the strength of original evidence than by characteristics of the original and replication teams.

Abstract: This report summarizes cross calibration of energetic electron observations between pairs of LANL geosynchronous (GEO) satellites. Of the GEO satellites considered, seven carried Synchronous Orbit Particle Analyzer (SOPA) detectors and two carried the Charged Particle Analyzer (CPA) detector. Discussion covers the comparison method used, information on available data and results for cross calibration offsets and residual errors. These cross calibrations were performed as part of overall intercalibration efforts for development of the AE9 radiation belt model., The original document contains color images.

Abstract: A new cross-calibrated data set of energetic electron observations from the Medium Electron Sensor A (MEA) and High Energy Electron Fluxmeter (HEEF) instruments on the Combined Release and Radiation Effects Satellite (CRRES) has been completed. Processing of these data sets includes cleaning for proton contaminated MEA data, cleaning for MEA/HEEF data with incomplete pitch angle data, spectral correction of MEA data, and adjustment of HEEF data at high flux levels. Description of the data set contents is provided. Abstract: In mammalian cells three closely related cavin proteins cooperate with the scaffolding protein caveolin to form membrane invaginations known as caveolae. Here we have developed a novel single-molecule fluorescence approach to directly observe interactions and stoichiometries in protein complexes from cell extracts and from in vitro synthesized components. We show that up to 50 cavins associate on a caveola. However, rather than forming a single coat complex containing the three cavin family members, single-molecule analysis reveals an exquisite specificity of interactions between cavin1, cavin2 and cavin3. Changes in membrane tension can flatten the caveolae, causing the release of the cavin coat and its disassembly into separate cavin1-cavin2 and cavin1-cavin3 subcomplexes. Each of these subcomplexes contain 9 ± 2 cavin molecules and appear to be the building blocks of the caveolar coat. High resolution immunoelectron microscopy suggests a remarkable nanoscale organization of these separate subcomplexes, forming individual striations on the surface of caveolae. © Gambin et al.

Abstract: The preponderance of matter over antimatter in the early Universe, the dynamics of the supernova bursts that produced the heavy elements necessary for life and whether protons eventually decay --- these mysteries at the forefront of particle physics and astrophysics are key to understanding the early evolution of our Universe, its current state and its eventual fate. The Long-Baseline Neutrino Experiment (LBNE) represents an extensively developed plan for a world-class experiment dedicated to addressing these questions. LBNE is conceived around three central components: (1) a new, high-intensity neutrino source generated from a megawatt-class proton accelerator at Fermi National Accelerator Laboratory, (2) a near neutrino detector just downstream of the source, and (3) a massive liquid argon time-projection chamber deployed as a far detector deep underground at the Sanford Underground Research Facility. This facility, located at the site of the former Homestake Mine in Lead, South Dakota, is approximately 1,300 km from the neutrino source at Fermilab -- a distance (baseline) that delivers optimal sensitivity to neutrino charge-parity symmetry violation and mass ordering effects. This ambitious yet cost-effective design incorporates scalability and flexibility and can accommodate a variety of upgrades and contributions. With its exceptional combination of experimental configuration, technical capabilities, and potential for transformative discoveries, LBNE promises to be a vital facility for the field of particle physics worldwide, providing physicists from around the globe with opportunities to collaborate in a twenty to thirty year program of exciting science. In this document we provide a comprehensive overview of LBNE's scientific objectives, its place in the landscape of neutrino physics worldwide, the technologies it will incorporate and the capabilities it will possess.

Abstract: The preponderance of matter over antimatter in the early Universe, the dynamics of the supernova bursts that produced the heavy elements necessary for life and whether protons eventually decay --- these mysteries at the forefront of particle physics and astrophysics are key to understanding the early evolution of our Universe, its current state and its eventual fate. The Long-Baseline Neutrino Experiment (LBNE) represents an extensively developed plan for a world-class experiment dedicated to addressing these questions. LBNE is conceived around three central components: (1) a new, high-intensity neutrino source generated from a megawatt-class proton accelerator at Fermi National Accelerator Laboratory, (2) a near neutrino detector just downstream of the source, and (3) a massive liquid argon time-projection chamber deployed as a far detector deep underground at the Sanford Underground Research Facility. This facility, located at the site of the former Homestake Mine in Lead, South Dakota, is approximately 1,300 km from the neutrino source at Fermilab -- a distance (baseline) that delivers optimal sensitivity to neutrino charge-parity symmetry violation and mass ordering effects. This ambitious yet cost-effective design incorporates scalability and flexibility and can accommodate a variety of upgrades and contributions. With its exceptional combination of experimental configuration, technical capabilities, and potential for transformative discoveries, LBNE promises to be a vital facility for the field of particle physics worldwide, providing physicists from around the globe with opportunities to collaborate in a twenty to thirty year program of exciting science. In this document we provide a comprehensive overview of LBNE's scientific objectives, its place in the landscape of neutrino physics worldwide, the technologies it will incorporate and the capabilities it will possess.

Abstract: Reproducibility is a defining feature of science. However, because of strong incentives for innovation and weak incentives for confirmation, direct replication is rarely practiced or published. The Reproducibility Project is an open, large-scale, collaborative effort to systematically examine the rate and predictors of reproducibility in psychological science. So far, 72 volunteer researchers from 41 institutions have organized to openly and transparently replicate studies published in three prominent psychological journals in 2008. Multiple methods will be used to evaluate the findings, calculate an empirical rate of replication, and investigate factors that predict reproducibility. Whatever the result, a better understanding of reproducibility will ultimately improve confidence in scientific methodology and findings.

Abstract: In early 2010, the Long-Baseline Neutrino Experiment (LBNE) science collaboration initiated a study to investigate the physics potential of the experiment with a broad set of different beam, near- and far-detector configurations. Nine initial topics were identified as scientific areas that motivate construction of a long-baseline neutrino experiment with a very large far detector. We summarize the scientific justification for each topic and the estimated performance for a set of far detector reference configurations. We report also on a study of optimized beam parameters and the physics capability of proposed Near Detector configurations. This document was presented to the collaboration in fall 2010 and updated with minor modifications in early 2011.

Abstract: In early 2010, the Long-Baseline Neutrino Experiment (LBNE) science collaboration initiated a study to investigate the physics potential of the experiment with a broad set of different beam, near- and far-detector configurations. Nine initial topics were identified as scientific areas that motivate construction of a long-baseline neutrino experiment with a very large far detector. We summarize the scientific justification for each topic and the estimated performance for a set of far detector reference configurations. We report also on a study of optimized beam parameters and the physics capability of proposed Near Detector configurations. Abstract: In early 2010, the Long-Baseline Neutrino Experiment (LBNE) science collaboration initiated a study to investigate the physics potential of the experiment with a broad set of different beam, near- and far-detector configurations. Nine initial topics were identified as scientific areas that motivate construction of a long-baseline neutrino experiment with a very large far detector. We summarize the scientific justification for each topic and the estimated performance for a set of far detector reference configurations. We report also on a study of optimized beam parameters and the physics capability of proposed Near Detector configurations. Abstract: The calorimeter, range detector and active target elements of the T2K near detectors rely on the Hamamatsu Photonics Multi-Pixel Photon Counters (MPPCs) to detect scintillation light produced by charged particles. Detailed measurements of the MPPC gain, afterpulsing, crosstalk, dark noise, and photon detection efficiency for low light levels are reported. In order to account for the impact of the MPPC behavior on T2K physics observables, a simulation program has been developed based on these measurements. Abstract: For samples of polymethylmethacrylate with and without quartz filler, the inverse of the contrast-gloss ratio is shown to be related to surface roughness and to the optical scattering coefficient. This finding adds to the importance of optical scattering, which has been widely studied because of its relation to color and translucency of materials. Furthermore, optical scattering by composite fillers is shown to be linearly related to the concentration of the filler material within the range of concentrations studied. Quartz fillers were incorporated at concentrations from 5 to 20 weight percent and were short fibers or granular powder, with the granular particles ranging in median equivalent spherical diameter from 15 to 3.3 μm. The efficiency of optical scattering for the granular quartz filler increased as the size of the filler decreased.

Abstract: The purpose of this study was to determine the increase in color contrast produced by the use of a tracer dye in detection of incipient caries lesions with transillumination. Twenty four caries-free first premolars were immersed in an acid gelatin for production of artificial incipient caries lesions. After the lesions had developed, these teeth were photographed by transillumination. Two photographs were taken of each tooth. The first photograph showed the lesion without dye. A blue tracer dye was then added and absorbed by the lesion, and a second photograph was taken. The data on the color difference were obtained by use of a reflectance colorimeter and showed a four-fold increase between the lesion and surrounding area with the dye. A two-way analysis of variance was used for the statistical interpretation. The color difference between the lesion without the dye and then with the dye was significant. The use of the blue tracer dye, therefore, significantly increased the contrast in the images of the artificial incipient lesions.

Abstract: Sintered samples of modifying porcelains of various colors and manufacturers were analyzed using reflectance spectrophotometry. Color designations are reported according to practices of the Commission Internationale de l'Eclairage (International Commission for Illumination), and color names were assigned based on a method developed through a joint effort of the Inter-Society Color Council and the United States National Bureau of Standards (ISCC.NBS). Within samples labeled by the same color, differences among manufacturers were found in the color designations and names. These differences were noted in part by plots of the chromaticity coordinates of the samples. The ISCC-NBS method of designating colors is proposed as a uniform and descriptive color-naming method for modifier porcelain. Consistent color naming is a step in improving communication over the use of modifying porcelains.

Abstract: The contrast gloss and the average roughness were measured for four commercial composite filling materials. Using a factorial design, each material was subjected to four available finishing methods. A significant difference was found in the contrast gloss among finishing methods, and a significant linear regression is given which relates the inverse of the contrast gloss to the average roughness. Surface gloss is proposed as playing a major role in the esthetic appearance of composite restorations.

Abstract: The first science run of the LIGO and GEO gravitational wave detectors presented the opportunity to test methods of searching for gravitational waves from known pulsars. Here we present new direct upper limits on the strength of waves from the pulsar PSR J1939+2134 using two independent analysis methods, one in the frequency domain using frequentist statistics and one in the time domain using Bayesian inference. Both methods show that the strain amplitude at Earth from this pulsar is less than a few times 10−22.
