Tandem repeats (TRs) represent one of the largest sources of genetic variation in humans and are implicated in a range of phenotypes. Here we present a deep characterization of TR variation based on high coverage whole genome sequencing from 3550 diverse individuals from the 1000 Genomes Project and H3Africa cohorts. We develop a method, EnsembleTR, to integrate genotypes from four separate methods resulting in high-quality genotypes at more than 1.7 million TR loci. Our catalog reveals novel sequence features influencing TR heterozygosity, identifies population-specific trinucleotide expansions, and finds hundreds of novel eQTL signals. Finally, we generate a phased haplotype panel which can be used to impute most TRs from nearby single nucleotide polymorphisms (SNPs) with high accuracy. Overall, the TR genotypes and reference haplotype panel generated here will serve as valuable resources for future genome-wide and population-wide studies of TRs and their role in human phenotypes.

Recently, significant advances have been achieved to precisely program the response of liquid crystal elastomers (LCEs) through extrusion-based additive manufacturing techniques; however, important challenges remain, especially when well-defined scaffolds based on ultrafine fibers are required. Here the melt electrowriting of reactive liquid crystalline inks, leading, after ultraviolet-light-induced crosslinking, to digitally positioned uniform LCE fibers with diameters ranging from hundreds of nanometers to tens of micrometers is presented, which is hardly accessible with conventional extrusion-based printing techniques. The electrowriting process induces the preferential alignment of the mesogens parallel to the fiber's axis. Such an alignment, defined by the printing path, determines the mechanical response of the crosslinked material upon stimulation. This manufacturing platform allows the preparation of open square lattice scaffolds with ultrafine fibers (a few micrometers in diameter), periods as small as 90 µm, and well-defined morphology. Additionally, the combination of accurate fiber stacking (up to 50 layers) and fiber fusion between layers leads to unprecedented microstructures composed of high-aspect-ratio LCE thin walls. The possibility of digitally controlling the printing of fibers allows the preparation complex fiber-based scaffolds with programmed and reversible shape-morphing, thus opening new avenues to prepare miniaturized actuators and smart structures for soft robotics and biomedical applications.

Aims Data on the prognostic value of frailty to guide clinical decision-making for patients with myocardial infarction (MI) are scarce. To analyse the association between frailty classification, treatment patterns, in-hospital outcomes, and 6-month mortality in a large population of patients with MI. Methods and results An observational, multicentre study with a retrospective analysis of prospectively collected data using the SWEDEHEART registry. In total, 3381 MI patients with a level of frailty assessed using the Clinical Frailty Scale (CFS-9) were included. Of these patients, 2509 (74.2%) were classified as non-vulnerable non-frail (CFS 1-3), 446 (13.2%) were vulnerable non-frail (CFS 4), and 426 (12.6%) were frail (CFS 5-9). Frailty and non-frail vulnerability were associated with worse in-hospital outcomes compared with non-frailty, i.e. higher rates of mortality (13.4% vs. 4.0% vs. 1.8%), cardiogenic shock (4.7% vs. 2.5% vs. 1.9%), and major bleeding (4.5% vs. 2.7% vs. 1.1%) (all P < 0.001), and less frequent use of evidence-based therapies. In Cox regression analyses, frailty was strongly and independently associated with 6-month mortality compared with non-frailty, after adjustment for age, sex, the GRACE risk score components, and other potential risk factors [hazard ratio (HR) 3.32, 95% confidence interval (CI) 2.30-4.79]. A similar pattern was seen for vulnerable non-frail patients (fully adjusted HR 2.07, 95% CI 1.41-3.02). Conclusion Frailty assessed with the CFS was independently and strongly associated with all-cause 6-month mortality, also after comprehensive adjustment for baseline differences in other risk factors. Similarly, non-frail vulnerability was independently associated with higher mortality compared with those with preserved functional ability.

High-nitrogen-chromium alloyed powder metallurgy (PM) tool steels offer many attractive features including high strength and corrosion resistance. The PM route offers various advantages such as advanced alloy composition, high homogeneity, and well-defined size distribution of hard phase particles. This study presents microstructure and mechanical properties of a PM Cr-Mo-V-N alloy. The conventional manufacturing route for this alloy is hot isostatic pressing (HIP) followed by hot working. To investigate the possibility of near-net-shape manufacturing, a comprehensive comparison of the performance was made between steels produced by as-HIPed and HIPed followed by hot working. Sodium silicate was added to organic soil in order to study its effects on soil shear strength in this research. The different types of soil including sand, clay, peat have been used in this study. Different percentages of sodium silicate (5%, 9%, 12%, 15%) have been used for soil stabilization. In this study, the curing times were 3 days, 7 days, 28 days. Finally, after performing the necessary tests on soil shear strength and comparing the effects of sodium silicate on organic soils, it was observed that soil shear strength has increased after adding sodium silicate gradually. Therefore, lime can be replaced in organic soils to be mixed with sand and clay in with different percentages.

Lack of individual responsibility has resulted in disengagement in teaching. Based on a quality improvement project (QIP) at a District General Hospital, we suggest some ways this could be addressed. We suggest here that harnessing support from the medical education lead, incentivizing teaching, allocating student-junior doctor groups to harbor personal responsibility, providing a supportive framework, and educating about barriers to teaching can all be used to develop an effective teaching program.Keywords: bedside teaching, quality improvement, junior doctor

The study was conducted to enhance the dissolution rate of ketoconazole (KCZ) (a poorly water-soluble drug) using the liquisolid technique. Microcrystalline cellulose, colloidal silica, PEG400 and polyvinyl pyrrolidone (PVP) were employed as a carrier, coating substance, nonvolatile solvent and additive in the KCZ liquisolid compact formulation, respectively. The drug-to-PEG400 and carrier-to-coating ratio variations, PVP concentration and aging effects on the in vitro release behavior were assessed. Differential scanning calorimetry (DSC) and X-ray powder diffraction (XRD) data revealed no alterations in the crystalline form of the drug and the KCZ-excipient interactions within the process. The load factor and the drug release rate were significantly enhanced compared to directly compressed tablets in the presence of the additive. Increasing the PEG400-to-drug ratio in liquid medications enhanced the dissolution rate remarkably. The dissolution profile and hardness of liquisolid compacts were not significantly altered by keeping the tablets at 40 °C and relative humidity of 75 % for 6 months. With the proposed modification of the liquisolid process, it is possible to obtain flowable, compactible liquisolid powders of high-dose poorly-water soluble drugs with an enhanced dissolution rate.

Discontinuous Galerkin (DG) methods have emerged as a successful approach for non-linear hyperbolic problems and are widely regarded very promising for next generation CFD solvers. Their efficiency for high-order discretization makes them suitable for advanced physical models like DES and LES, while their stability in convection dominated regimes is also a merit of them. The compactness of DG methods, facilitate the parallelization and their element-by-element discontinuous nature is also helpful for adaptivity. This PhD thesis focuses on the development of an efficient and robust high-order Hybridizable Discontinuous Galerkin (HDG) Finite Element Method (FEM) for compressible viscous flow computations. HDG method is a new class of DG family which enjoys from merits of DG but has significantly less globally coupled unknowns compared to other DG methods. Its features makes HDG a possible candidate to be investigated as next generation high-order tools for CFD applications. The first part of this thesis recalls the basics of high-order HDG method. It is presented for the two-dimensional linear convection-diffusion equation, and its accuracy and features are investigated. Algunos fenómenos físicos sofisticados, por ejemplo en aeroacústica, presentan vórtices y turbulencias, y necesitan métodos de muy alta resolución para obtener resultados precisos. Since the development of industries, vehicles and the people in the cities and their suburban areas, carbon monoxide, sulfur dioxide, nitrogen dioxide and some other pollutants have been increasing. This study has been conducted to predict the air pollution, utilizing model of atmospheric pollutant dispersion for a large city of Ahwaz, with regard to land use, the amount of pollutants released from point, area and line sources, subsequently integrated mesoscale model was used to predict the dispersion and concentration of the air pollutants. The main purpose of the study was to determine the dispersion factors and the meteorological condition which cause the air to be polluted over the study area. To achieve the main goal of the study, establishment of the emission inventory, meteorological simulation and (CO), (NO, NO2) and SO2 dispersions sources in the Ahwaz city regions by using the WRF CAMx and evaluation of the model performance were done. The mean difference between prediction and observed data was obtained by Root Mean Square Error (RMSE) and the Peak results are as follow: SO2, 75 ppb, NO2, 100 ppb, NO, 85 ppb and CO, 40 ppb. A comparison of the observed peaks from the air pollution measurement stations at ground level, and the results of WRF / CAMx model show the error of the peaks is less than 30%, which this percentage indicates that input data to the model and to the real data are close to each other. Normalized error was ±20% and normalized bias was between -15% to +15%, this amount for the air pollution models are acceptable. Also, the BIAS ERROR calculates the relationship coefficient between the simulated prediction and actual data. The results showed that the Zergan Power Plant has more effect over the North East part of the Ahwaz city than the Ramin Power Plant, due to distances and the wind direction characteristics and that the pollutants concentration is equally related to emitted pollutants from their own sources. In this chapter we focus on slack reclamation and propose a new slack reclamation technique, Multiple Frequency Selection DVFS (MFS-DVFS). The key idea is to execute each task with a linear combination of more than one frequency such that this combination results in using the lowest energy by covering the whole slack time of the task. We have tested our algorithm with both random and real-world application task graphs and compared with the results in previous researches in [9] and [12-13].
