1. Exploring the Orphan GPCR GPR18 through Novel Synthetic Cannabidiol Derivatives

Abstract: GPR18 is an orphan GPCR highly expressed in lymphoid tissues and the central nervous system that regulates cellular migration, proliferation, nociociception, and immunomodulation. The endocannabinoid derivative N-Arachidonoylglycine (NAGly) has been proposed as the putative ligand. Several other cannabinoids also interact with GPR18, such as Abn-CBD and ¿9-THC. However, very few potent synthetic GPR18 ligands have been described so far. A new family of compounds with a cannabidiol scaffold were designed to target GPR18. Calcium mobilization imaging studies1 and docking studies in a in silico model2 were used to evaluate the activity of compounds and their mechanism of action. Here, two of the best compounds are exemplified: S5, a GPR18 agonist, and S4, a GPR18 antagonist.

Abstract: GPR55 is an orphan Class A G-protein coupled receptor that recognizes a sub-set of cannabinoid CB1 and CB2 ligands, suggesting that GPR55 could belong to the endocannabinoid system. Lysophosphatidylinositol (LPI) has been proposed to be endogenous ligand for GPR55. This receptor is involved in diverse physiological and pathological processes such as inflammatory and neuropathic pain, metabolic disorder, bone and neuronal development, and cancer. Few potent GPR55 ligands have been identified to date. High throughput screening of a large library of compounds from the Molecular Libraries Probe Production Centers Network (MLPCN) allowed the identification of different GPR55 chemical scaffolds, including ML192, a GPR55 antagonist. However, their potency and selectivity needs to be optimized in order to develop appropriate pharmacological tools or novel drugs to continue with the challenging goal of the validation of this receptor

Abstract: GPR55 is an orphan Class A G-protein coupled receptor that recognizes a sub-set of cannabinoid CB1 and CB2 ligands, suggesting that GPR55 could belong to the endocannabinoid system. Lysophosphatidylinositol (LPI) has been proposed to be endogenous ligand for GPR55. However, GPR55 is still considered orphan receptor due to the lack of in vivo efficacy. The interest of GPR55 ligands as therapeutic agents is supported by the fact that this receptor is involved in diverse physiological and pathological processes such as inflammatory and neuropathic pain, metabolic disorder, bone and neuronal development, and cancer. Few potent GPR55 ligands have been identified to date due to an absence of information about salient features of GPR55, such as residues importance for binding and residues implicated in the GPR55 signaling cascade. High throughput screening of a large library of compounds from the Molecular Libraries Probe Production Centers Network (MLPCN) allowed the identification of different GPR55 chemical scaffolds, including ML192, a GPR55 antagonist. However, their potency and selectivity needs to be optimized in order to develop appropriate pharmacological tools or novel drugs to continue with the challenging goal of the validation of this receptor.

Abstract: GPR18 is a G-protein-coupled receptor that belongs to the orphan class A family. Even though it shares low sequence homology with the cannabinoid receptors CBR and CBR, a growing body of research suggests its relationship with the endocannabinoid system, not only because it is able to recognize cannabinoid ligands but also because of its expression and ability to heteromerize with CBRs. In this review, we aim to analyze the biological relevance, reported modulators, and structural features of GPR18. In order to guide future drug design in this field, highlights from molecular modeling of GPR18 will be provided.

5. GPR6 structural insights: Homology model construction and docking studies

Abstract: GPR6 is an orphan G protein-coupled receptor that has been associated with the cannabinoid family because of its recognition of a sub-set of cannabinoid ligands. The high abundance of GPR6 in the central nervous system, along with high constitutive activity and a link to several neurodegenerative diseases make GPR6 a promising biological target. In fact, diverse research groups have demonstrated that GPR6 represents a possible target for the treatment of neurodegenerative disorders such as Parkinson’s disease, Alzheimer’s disease, and Huntington’s disease. Several patents have claimed the use of a wide range of pyrazine derivatives as GPR6 inverse agonists for the treatment of Parkinson’s disease symptoms and other dyskinesia syndromes. However, the full pharmacological importance of GPR6 has not yet been fully explored due to the lack of high potency, readily available ligands targeting GPR6. The long-term goal of the present study is to develop such ligands. In this paper, we describe our initial steps towards this goal. A human GPR6 homology model was constructed using a suite of computational techniques. This model permitted the identification of unique GPR6 structural features and the exploration of the GPR6 binding crevice. A subset of patented pyrazine analogs were docked in the resultant GPR6 inactive state model to validate the model, rationalize the structure-activity relationships from the reported patents and identify the key residues in the binding crevice for ligand recognition. We will take this structural knowledge into the next phase of GPR6 project, in which scaffold hopping will be used to design new GPR6 ligands.

12. Design of Novel GPR6 Inverse Agonists Using a Fragment Replacement Scaffold Hopping Approach

Abstract: The orphan G protein coupled receptor 6 (GPR6) is a cannabinoid-related Class A GPCR. It is highly expressed in the central nervous system and exhibits high constitutive activation of adenylyl cyclase. Several research groups have demonstrated that GPR6 represents a possible target for the treatment of neurodegenerative disorders such as Parkinson’s, Alzheimer’s, and Huntington’s diseases. Several patents claim the use of a wide range of pyrazine derivatives as GPR6 inverse agonists for the treatment of Parkinson’s disease and other dyskinesia syndromes. Using cyclic AMP accumulation assays in hGPR6-CHO cells as a readout, the most potent GPR6 pyridopyrazine inverse agonist compounds identified thus far have been found to display IC50 values in the low nanomolar range. A subset of these compounds was used here as starting points for the design of novel potent GPR6 inverse agonists using a core hopping approach. In parallel with the core hopping studies, we employed a recently constructed homology model of the GPR6 inactive state. The X-ray crystal structure of the Sphingosine-1-phosphate receptor 1 (S1P1) receptor structure was used as the template and the conformational memories technique was used to explore the conformational consequences of sequence differences between S1P1 and GPR6. The most potent GPR6 pyridopyrazine inverse agonists were docked in the resultant GPR6 inactive state model, first as tests of the model. Once we had identified the binding site of each inverse agonist, we used this site to identify amino acid sites in proximity that we can use to build additional interactions for ligands output from core hopping studies above. ADME properties and the absence of PAINS will also be considered for the hit selection process. These potential GPR6 chemotypes may serve as research tools for further understanding the biological role of this orphan receptor.

Abstract: The orphan G-protein coupled receptor (GPCR), GPR18, has been recently proposed as a potential member of the cannabinoid family as it recognizes several endogenous, phytogenic, and synthetic cannabinoids. Potential therapeutic applications for GPR18 include intraocular pressure, metabolic disorders, and cancer. GPR18 has been reported to have high constitutive activity, i.e., activation/signaling occurs in the absence of an agonist. This activity can be reduced significantly by the A3.39N mutation. At the intracellular (IC) ends of (transmembrane helices) TMH3 and TMH6 in GPCRs, typically, a pair of oppositely charged amino acids form a salt bridge called the “ionic lock”. Breaking of this salt bridge creates an IC opening for coupling with G protein. The GPR18 “ionic lock” residues (R3.50/S6.33) can form only a hydrogen bond. In this paper, we test the hypothesis that the high constitutive activity of GPR18 is due to the weakness of its “ionic lock” and that the A3.39N mutation strengthens this lock. To this end, we report molecular dynamics simulations of wild-type (WT) GPR18 and the A3.39N mutant in fully hydrated (POPC) phophatidylcholine lipid bilayers. Results suggest that in the A3.39N mutant, TMH6 rotates and brings R3.50 and S6.33 closer together, thus strengthening the GPR18 “ionic lock”.

Abstract: While many online graduate students are gaining academic and scholarly knowledge, the opportunities for students to develop and hone professional skills essential for the workplace are lacking. Given the virtual environment of distance learning, graduate students are often expected to glean professional skills such as analytical thinking, self-awareness, flexibility, team-building, and problem-solving inherently through informal means (Cleveland-Innes & Ally, 2012). The goal of this study was to evaluate the experiences of online graduate students participating in synchronous online professional skills workshops. Students attended the sessions from the various graduate programs at an online Canadian university. The discussions from the focus group held at the end of the project were used to achieve the research goals. This paper used a phenomenological lens to accomplish its research goals. Abstract: The three main objectives of the GCOS (Global Climate Observing System) Reference Upper-Air Network (GRUAN) are to provide long-term high quality climate records of vertical profiles of selected Essential Climate Variables (ECVs), to constrain and calibrate data from more spatially comprehensive global networks, and to fully characterize the properties of the atmospheric column. In the five years since the first GRUAN implementation and coordination meeting and the publication of a BAMS article (Seidel et al., 2009), GRUAN has matured to become a functioning network that provides reference-quality observations to a community of users. This article describes the achievements within GRUAN over the past five years towards making reference-quality observations of upper-air ECVs. Milestones in the evolution of GRUAN are emphasized, including development of rigorous criteria for site certification and assessment, the formal certification of the first GRUAN sites, salient aspects of the GRUAN Manual and Guide to Operations, public availability of GRUAN’s first data product, outcomes of a network expansion workshop, and key results of scientific studies designed to provide a sound scientific foundation for GRUAN operations. Two defining attributes of GRUAN are (1) that every measurement is accompanied by a traceable estimate of the measurement uncertainty and (2) that data quality and continuity are maximized because network changes are minimized and managed. This article summarizes how these imperatives are being achieved for existing and planned data products and provides an outlook for the future, including expected new data streams, network expansion, and critical needs for the ongoing success of GRUAN.

Abstract: A combination of molecular modeling and structure–activity relationship studies has been used to fine-tune CB2 selectivity in the chromenopyrazole ring, a versatile CB1/CB2 cannabinoid scaffold. Thus, a series of 36 new derivatives covering a wide range of structural diversity has been synthesized, and docking studies have been performed for some of them. Biological evaluation of the new compounds includes, among others, cannabinoid binding assays, functional studies, and surface plasmon resonance measurements. The most promising compound [43 (PM226)], a selective and potent CB2 agonist isoxazole derivative, was tested in the acute phase of Theiler’s murine encephalomyelitis virus-induced demyelinating disease (TMEV-IDD), a well-established animal model of primary progressive multiple sclerosis. Compound 43 dampened neuroinflammation by reducing microglial activation in the TMEV.

The longest available record from balloon observations over Boulder, Colorado, USA shows increases in stratospheric water vapour concentrations that cannot be fully explained by observed changes in the main drivers, tropical tropopause temperatures and methane. Satellite observations could help resolve the issue, but constructing a reliable long-term data record from individual short satellite records is challenging. Here we present an approach to merge satellite data sets with the help of a chemistry-climate model nudged to observed meteorology. We use the models' water vapour as a transfer function between data sets that overcomes issues arising from instrument drift and short overlap periods. In the lower stratosphere, our water vapour record extends back to 1988 and water vapour concentrations largely follow tropical tropopause temperatures. Lower and mid-stratospheric long-term trends are negative, and the trends from Boulder are shown not to be globally representative. In the upper stratosphere, our record extends back to 1986 and shows positive long-term trends. The altitudinal differences in the trends are explained by methane oxidation together with a strengthened lower-stratospheric and a weakened upper-stratospheric circulation inferred by this analysis. Our results call into question previous estimates of surface radiative forcing based on presumed global long-term increases in water vapour concentrations in the lower stratosphere.

Abstract: Several large-scale climate patterns influenced climate conditions and weather patterns across the globe during 2010. The transition from a warm El Nino phase at the beginning of the year to a cool La Nina phase by July contributed to many notable events, ranging from record wetness across much of Australia to historically low Eastern Pacific basin and near-record high North Atlantic basin hurricane activity. The remaining five main hurricane basins experienced below-to well-below-normal tropical cyclone activity. The negative phase of the Arctic Oscillation was a major driver of Northern Hemisphere temperature patterns during 2009/10 winter and again in late 2010. It contributed to record snowfall and unusually low temperatures over much of northern Eurasia and parts of the United States, while bringing above-normal temperatures to the high northern latitudes. The February Arctic Oscillation Index value was the most negative since records began in 1950. The 2010 average global land and ocean surface temperature was among the two warmest years on record. The Arctic continued to warm at about twice the rate of lower latitudes. The eastern and tropical Pacific Ocean cooled about 1 C from 2009 to 2010, reflecting the transition from the 2009/10 El Nino to the 2010/11 La Nina. Ocean heat fluxes contributed to warm sea surface temperature anomalies in the North Atlantic and the tropical Indian and western Pacific Oceans. Global integrals of upper ocean heat content for the past several years have reached values consistently higher than for all prior times in the record, demonstrating the dominant role of the ocean in the Earth's energy budget. Deep and abyssal waters of Antarctic origin have also trended warmer on average since the early 1990s. Lower tropospheric temperatures typically lag ENSO surface fluctuations by two to four months, thus the 2010 temperature was dominated by the warm phase El Nino conditions that occurred during the latter half of 2009 and early 201

Abstract: Several large-scale climate patterns influenced climate conditions and weather patterns across the globe during 2010. The transition from a warm El Nino phase at the beginning of the year to a cool La Nina phase by July contributed to many notable events, ranging from record wetness across much of Australia to historically low Eastern Pacific basin and near-record high North Atlantic basin hurricane activity. The remaining five main hurricane basins experienced below-to well-below-normal tropical cyclone activity. The negative phase of the Arctic Oscillation was a major driver of Northern Hemisphere temperature patterns during 2009/10 winter and again in late 2010. It contributed to record snowfall and unusually low temperatures over much of northern Eurasia and parts of the United States, while bringing above-normal temperatures to the high northern latitudes. The February Arctic Oscillation Index value was the most negative since records began in 1950. The 2010 average global land and ocean surface temperature was among the two warmest years on record. The Arctic continued to warm at about twice the rate of lower latitudes. The eastern and tropical Pacific Ocean cooled about 1 C from 2009 to 2010, reflecting the transition from the 2009/10 El Nino to the 2010/11 La Nina. Ocean heat fluxes contributed to warm sea surface temperature anomalies in the North Atlantic and the tropical Indian and western Pacific Oceans. Global integrals of upper ocean heat content for the past several years have reached values consistently higher than for all prior times in the record, demonstrating the dominant role of the ocean in the Earth's energy budget. Deep and abyssal waters of Antarctic origin have also trended warmer on average since the early 1990s. Lower tropospheric temperatures typically lag ENSO surface fluctuations by two to four months, thus the 2010 temperature was dominated by the warm phase El Nino conditions that occurred during the latter half of 2009 and early 201

Abstract: Several large-scale climate patterns influenced climate conditions and weather patterns across the globe during 2010. The transition from a warm El Nino phase at the beginning of the year to a cool La Nina phase by July contributed to many notable events, ranging from record wetness across much of Australia to historically low Eastern Pacific basin and near-record high North Atlantic basin hurricane activity. The remaining five main hurricane basins experienced below-to well-below-normal tropical cyclone activity. The negative phase of the Arctic Oscillation was a major driver of Northern Hemisphere temperature patterns during 2009/10 winter and again in late 2010. It contributed to record snowfall and unusually low temperatures over much of northern Eurasia and parts of the United States, while bringing above-normal temperatures to the high northern latitudes. The February Arctic Oscillation Index value was the most negative since records began in 1950. The 2010 average global land and ocean surface temperature was among the two warmest years on record. The Arctic continued to warm at about twice the rate of lower latitudes. The eastern and tropical Pacific Ocean cooled about 1 C from 2009 to 2010, reflecting the transition from the 2009/10 El Nino to the 2010/11 La Nina. Ocean heat fluxes contributed to warm sea surface temperature anomalies in the North Atlantic and the tropical Indian and western Pacific Oceans. Global integrals of upper ocean heat content for the past several years have reached values consistently higher than for all prior times in the record, demonstrating the dominant role of the ocean in the Earth's energy budget. Deep and abyssal waters of Antarctic origin have also trended warmer on average since the early 1990s. Lower tropospheric temperatures typically lag ENSO surface fluctuations by two to four months, thus the 2010 temperature was dominated by the warm phase El Nino conditions that occurred during the latter half of 2009 and early 201

Abstract: Several large-scale climate patterns influenced climate conditions and weather patterns across the globe during 2010. The transition from a warm El Nino phase at the beginning of the year to a cool La Nina phase by July contributed to many notable events, ranging from record wetness across much of Australia to historically low Eastern Pacific basin and near-record high North Atlantic basin hurricane activity. The remaining five main hurricane basins experienced below-to well-below-normal tropical cyclone activity. The negative phase of the Arctic Oscillation was a major driver of Northern Hemisphere temperature patterns during 2009/10 winter and again in late 2010. It contributed to record snowfall and unusually low temperatures over much of northern Eurasia and parts of the United States, while bringing above-normal temperatures to the high northern latitudes. The February Arctic Oscillation Index value was the most negative since records began in 1950. The 2010 average global land and ocean surface temperature was among the two warmest years on record. The Arctic continued to warm at about twice the rate of lower latitudes. The eastern and tropical Pacific Ocean cooled about 1 C from 2009 to 2010, reflecting the transition from the 2009/10 El Nino to the 2010/11 La Nina. Ocean heat fluxes contributed to warm sea surface temperature anomalies in the North Atlantic and the tropical Indian and western Pacific Oceans. Global integrals of upper ocean heat content for the past several years have reached values consistently higher than for all prior times in the record, demonstrating the dominant role of the ocean in the Earth's energy budget. Deep and abyssal waters of Antarctic origin have also trended warmer on average since the early 1990s. 27. Spatial distribution of Delta(CO2)-C-14 across Eurasia: measurements from the TROICA-8 expedition

Abstract: Because fossil fuel derived CO2 is the only source of atmospheric CO2 that is devoid of 14C, atmospheric measurements of delta14CO2 can be used to constrain fossil fuel emission estimates at local and regional scales. However, at the continental scale, uncertainties in atmospheric transport and other sources of variability in delta14CO2 may influence the fossil fuel detection capability. We present a set of delta14CO2 observations from the train-based TROICA-8 expedition across Eurasia in March-April 2004. Local perturbations in delta14CO2 are caused by easily identifiable sources from nuclear reactors and localized pollution events. The remaining data show an increase in delta14CO2 from Western Russia (40° E) to Eastern Siberia (120° E), consistent with depletion in 14CO2 caused by fossil fuel CO2 emissions in heavily populated Europe, and gradual dispersion of the fossil fuel plume across Northern Asia. Other trace gas species which may be correlated with fossil fuel CO2 emissions, including carbon monoxide, sulphur hexafluoride, and perchloroethylene, were also measured and the results compared with the delta14CO2 measurements. The sulphur hexafluoride longitudinal gradient is not significant relative to the measurement uncertainty. Carbon monoxide and perchloroethylene show large-scale trends of enriched values in Western Russia and decreasing values in Eastern Siberia, consistent with fossil fuel emissions, but exhibit significant spatial variability, especially near their primary sources in Western Russia. The clean air delta14CO2 observations are compared with simulated spatial gradients from the TM5 atmospheric transport model. We show that the change in delta14CO2 across the TROICA transect is due almost entirely to emissions of fossil fuel CO2, but that the magnitude of this delta14CO2 gradient is relatively insensitive to modest uncertainties in the fossil fuel flux. Abstract: The amount of chlorine in the stratosphere has a direct influence on the magnitude of chlorine-catalyzed ozone loss. A comprehensive suite of organic source gases of chlorine in the stratosphere was measured during the NASA Stratospheric Aerosol and Gas Experiment (SAGE) III Ozone Loss and Validation Experiment (SOLVE) campaign in the arctic winter of 2000. Measurements included chlorofluorocarbons (CFCs), hydrochlorofluorocarbons (HCFCs), halon 1211, solvents, methyl chloride, N2O, and CH4. Inorganic chlorine contributions from each compound were calculated using the organic chlorine measurements, mean age of air, tropospheric trends, and a method to account for mixing in the stratosphere. Total organic chlorine measured at tropospheric levels of N2O was on the order of 3500 ppt. Total calculated inorganic chlorine at a N2O mixing ratio of 50 ppb (corresponding to a mean age of 5.5 years) was on the order of 3400 ppt. CFCs were the largest contributors to total organic chlorine (55-70%) over the measured N2O range (50-315 ppb), followed by CH3Cl (15%), solvents (5-20%), and HCFCs (5-25%). CH3Cl contribution was consistently about 15% across the organic chlorine range. Contributions to total calculated inorganic chlorine at 50 ppb N2O were 58% from CFCs, 24% from solvents, 16% from CH3Cl, and 2% from HCFCs. Updates to fractional chlorine release values for each compound relative to CFC 11 were calculated from the SOLVE measurements. An average value of 0.58 was calculated for the fractional chlorine release of CFC 11 over the 3-4 year mean age range, which was lower than the previous value of 0.80. The fractional chlorine release values for HCFCs 141b and 142b relative to CFC 11 were significantly lower than previous calculations.

Abstract: The amount of chlorine in the stratosphere has a direct influence on the magnitude of chlorine-catalyzed ozone loss. A comprehensive suite of organic source gases of chlorine in the stratosphere was measured during the NASA Stratospheric Aerosol and Gas Experiment (SAGE) III Ozone Loss and Validation Experiment (SOLVE) campaign in the arctic winter of 2000. Measurements included chlorofluorocarbons (CFCs), hydrochlorofluorocarbons (HCFCs), halon 1211, solvents, methyl chloride, N2O, and CH4. Inorganic chlorine contributions from each compound were calculated using the organic chlorine measurements, mean age of air, tropospheric trends, and a method to account for mixing in the stratosphere. Total organic chlorine measured at tropospheric levels of N2O was on the order of 3500 ppt. Total calculated inorganic chlorine at a N2O mixing ratio of 50 ppb (corresponding to a mean age of 5.5 years) was on the order of 3400 ppt. CFCs were the largest contributors to total organic chlorine (55-70%) over the measured N2O range (50-315 ppb), followed by CH3Cl (15%), solvents (5-20%), and HCFCs (5-25%). CH3Cl contribution was consistently about 15% across the organic chlorine range. Contributions to total calculated inorganic chlorine at 50 ppb N2O were 58% from CFCs, 24% from solvents, 16% from CH3Cl, and 2% from HCFCs. Updates to fractional chlorine release values for each compound relative to CFC 11 were calculated from the SOLVE measurements. An average value of 0.58 was calculated for the fractional chlorine release of CFC 11 over the 3-4 year mean age range, which was lower than the previous value of 0.80. The fractional chlorine release values for HCFCs 141b and 142b relative to CFC 11 were significantly lower than previous calculations.

Abstract: Simultaneous balloon-borne observations of ozone (O3) and nitrous oxide (N2O), a long-livedtracer of dynamical motion, are used to quantify the chemical loss of ozone in the Arctic vortexduring the winter of 1999/2000. Chemical loss of ozone occurred between altitudes of about 14 and22 km (pressures from ~120 to 30 mbar) and resulted in a 61 ± 13 Dobson unit reduction in totalcolumn ozone between late November 1999 and 5 March 2000 (the date of the last balloon-bornemeasurement considered here). This loss estimate is valid for the core of the vortex during the timeperiod covered by the observations. It is shown that the observed changes in the O3 versus N2Orelation were almost entirely due to chemistry and could not have been caused by dynamics. Abstract: Large losses of Arctic ozone occur during winters with cold, stable stratospheric circulations that result in the extensive occurrence of polar stratospheric clouds (PSCs). Reactions on the surface of PSCs lead to elevated abundances of chlorine monoxide (ClO) that, in the presence of sunlight, destroys ozone. Here we show that PSCs were more widespread during the 1999/2000 Arctic winter than for any other winter in the past two decades. We have used three fundamentally different approaches to derive the degree of chemical ozone loss from ozone sonde, balloon, aircraft and satelite instruments. We show that the ozone losses derived from these different instruments and approaches agree very well, resulting in a high level of confidence in the results. Chemical processes led to a 70% reduction of ozone for a ~1 km thick region of the lower stratosphere, the largest degree of local loss ever reported for the Arctic. The chemical loss of ozone in the total column amounted to about 100 DU by the end of the winter. This total column loss was balanced by transport, resulting in relatively constant total ozone between early January and late March, which is in contrast to the climatological increase of the total ozone column during this period, that is observed during most years.

Abstract: OBJECTIVE AND DESIGN: The majority of patients with otitis media with effusion (OME) and atopy have been shown to have elevated levels of eosinophil cationic protein (ECP) in their middle ear fluid. The mechanism underlying these elevated levels of ECP is not clear. The purpose of this study was to investigate the feasibility of a quantitative determination of eosinophils and neutrophils in the middle ear lining by specific immunocytochemical markers, in order to study the extent of the involvement of these cells in patients with OME. METHODS: Bilateral middle ear biopsies from five children with persistent OME and atopy confirmed by in vitro testing were evaluated for the presence of eosinophils and neutrophils with monoclonal antibodies against specific granule proteins. Five subjects who had no signs of effusion or infection but were undergoing routine tympanoplasty for dry perforations served as controls. The biopsies were embedded in a plastic resin to improve the structural preservation of the target cells and to increase the resolution in the light microscope. Dual markers were used to determine which marker was better for eosinophils and neutrophils, respectively. The following markers were used: eosinophil cationic protein (EG2), and eosinophil peroxidase (EPO) for eosinophils and myeloperoxidase (MPO) and human neutrophil lipocalin (HNL) for neutrophils. RESULTS: Antibodies against EPO gave a more localized and intense staining than antibodies against EG2. Antibodies against HNL appear more specific to neutrophils than antibodies against MPO that also recognize monocytes. The number of cells was determined both in the tissue and in the mucus covering the epithelium. Eosinophils and neutrophils were present in the subepithelial connective tissue and in the mucus blanket in the middle ear of patients with OME in significantly higher number than in the control group. Abstract: The trimethylsilyl group on the 6-position of methyl 2,3,4,6-tetra-O-trimethylsilyl-alpha-D-glucopyranoside is preferentially removed by methanolysis, using basic or acidic catalysts, giving methyl 2,3,4-tri-O-trimethylsilyl-alpha-D-glucopyranoside in high yield (>80%). Reaction of the latter with acetic anhydride, benzoyl chloride, or phenyl isocyanate produces the corresponding 6-substituted derivatives in quantitative yield. The trimethylsilyl residues from the latter compounds are hydrolyzed by 50% aqueous methanol with the concomitant formation of 6-O-acetyl, 6-O-benzoyl, or 6-carbanilate derivatives of methyl alpha-D-glucopyranoside in excellent yield. Chemical and proton magnetic resonance (p.m.r.) studies established the structure of methyl 2,3,4-tri-O-trimethylsilyl-alpha-D-glucopyranoside, and of subsequent derivatives prepared from this compound. Infrared and p.m.r. evidence is offered to support conclusions regarding the conformation of the groups or atoms attached to C6 and C5 of the glucoside residue in some of these compounds. Abstract: In 2013, the vast majority of the monitored climate variables reported here maintained trends established in recent decades. ENSO was in a neutral state during the entire year, remaining mostly on the cool side of neutral with modest impacts on regional weather patterns around the world. This follows several years dominated by the effects of either La NiÃ±a or El NiÃ±o events. According to several independent analyses, 2013 was again among the 10 warmest years on record at the global scale, both at the Earths surface and through the troposphere. Some regions in the Southern Hemisphere had record or near-record high temperatures for the year. Australia observed its hottest year on record, while Argentina and New Zealand reported their second and third hottest years, respectively. In Antarctica, Amundsen-Scott South Pole Station reported its highest annual temperature since records began in 1957. At the opposite pole, the Arctic observed its seventh warmest year since records began in the early 20th century. At 20-m depth, record high temperatures were measured at some permafrost stations on the North Slope of Alaska and in the Brooks Range. In the Northern Hemisphere extratropics, anomalous meridional atmospheric circulation occurred throughout much of the year, leading to marked regional extremes of both temperature and precipitation. Cold temperature anomalies during winter across Eurasia were followed by warm spring temperature anomalies, which were linked to a new record low Eurasian snow cover extent in May. Minimum sea ice extent in the Arctic was the sixth lowest since satellite observations began in 1979. Including 2013, all seven lowest extents on record have occurred in the past seven years. Antarctica, on the other hand, had above-average sea ice extent throughout 2013, with 116 days of new daily high extent records, including a new daily maximum sea ice area of 19.57 million km2 reached on 1 October.
