The study of jet launching in AGN is an important research method to better understand supermassive black holes (SMBHs) and their immediate surroundings. The main theoretical jet launching scenarios invoke either magnetic field lines anchored to the black hole's (BH) accretion disc (Blandford & Payne 1982) or a magnetic field, which is directly connected to its rotating ergosphere (Blandford & Znajek 1977). The nearby and bright radio galaxy 3C84 (NGC1275) is a very suitable target for testing different jet launching mechanisms, as well as for the study of the innermost, sub-parsec scale AGN structure and the jet origin. Very long baseline interferometry (VLBI) - specifically at millimetre wavelengths - offers an unparalleled view into the physical processes in action, in the close vicinity of SMBHs. Utilising such mm-VLBI observations of 3C84, we study the jet kinematics of the VLBI core region of 3C84 by employing all available, high sensitivity 3 mm-VLBI data sets of this source. As part of this analysis we associate the component ejection events with the variability light-curves at different radio frequencies and in the $\gamma$-rays. The contemporary popularity of restorative justice, within youth justice, has expanded significantly in recent decades. Despite this, there is a considerable lack of research which explores girls’ experiences of restorative justice interventions. Drawing on the experiences of young female offenders, who have participated in restorative justice conferencing, this article presents research findings generated from interviews undertaken with 15 girls and 13 youth justice practitioners, in order to critically analyse their views and experiences through a gendered lens. The analysis and discussion presented provides a critical insight into the ways in which girls’ experience, internalise and engage in restorative justice conferencing and how these experiences fundamentally conflict with practitioners’ views on conferencing with girls in the youth justice system.

Background Most people have strong left-brain lateralisation for language, with a minority showing right- or bilateral language representation. On some receptive language tasks, however, lateralisation appears to be reduced or absent. This contrasting pattern raises the question of whether and how language laterality may fractionate within individuals. Building on our prior work, we postulated (a) that there can be dissociations in lateralisation of different components of language, and (b) these would be more common in left-handers. A subsidiary hypothesis was that laterality indices will cluster according to two underlying factors corresponding to whether they involve generation of words or sentences, vs. receptive language. Methods We tested these predictions in two stages: At Step 1 an online laterality battery (Dichotic listening, Rhyme Decision and Word Comprehension) was given to 621 individuals (56% left-handers); At Step 2, functional transcranial Doppler ultrasound (fTCD) was used with 230 of these individuals (51% left-handers). 108 left-handers and 101 right-handers gave usable data on a battery of three language generation and three receptive language tasks. Results Neither the online nor fTCD measures supported the notion of a single language laterality factor. In general, for both online and fTCD measures, tests of language generation were left-lateralised. In contrast, the receptive tasks were at best weakly left-lateralised or, in the case of Word Comprehension, slightly right-lateralised. The online measures were only weakly correlated, if at all, with fTCD measures. Most of the fTCD measures had split-half reliabilities of at least .7, and showed a distinctive pattern of intercorrelation, supporting a modified two-factor model in which Phonological Decision (generation) and Sentence Decision (reception) loaded on both factors. 3C84 (NGC1275) is one of the brightest radio sources in the mm radio-bands, which led to a plethora of VLBI observations at numerous frequencies over the years. They reveal a two-sided jet structure, with an expanding but not well-collimated parsec-scale jet, pointing southward. High resolution mm-VLBI observations allow the study and imaging of the jet base on the sub-parsec scale. This could facilitate the investigation of the nature of the jet origin, also in view of the previously detected two-railed jet structure and east-west oriented core region seen with RadioAstron at 22 GHz. We produce VLBI images of this core and inner jet region, observed during the past twenty years at 15, 43, and 86 GHz. We determine the kinematics of the inner jet and ejected features at 43 and 86 GHz and compare their ejection times with radio and $\gamma$-ray variability. For the moving jet features, we find an average velocity of $\beta^\textrm{avg}_\textrm{app} = 0.055-0.22$c ($\mu^\textrm{avg} = 0.04-0.18\,$mas/yr). From the time-averaged VLBI images at the three frequencies, we measure the transverse jet width along the bulk flow. On the $\leq 1.5$ parsec scale, we find a clear trend of the jet width being frequency dependent, with the jet being narrower at higher frequencies. This stratification is discussed in the context of a spine-sheath scenario, and is compared to other possible interpretations. From quasi-simultaneous observations at 43 and 86\,GHz we obtain spectral index maps, revealing a time-variable orientation of the spectral index gradient, due to structural variability of the inner jet., Comment: 23 pages, 12 figures, accepted for publication in A&A

When manufacturing free-form concrete parts, casting approaches are most commonly adopted. While casting allows good surface finish, geometric repeatability and easy replication of parts, it is inflexible and costly to setup. Expensive mould tools, with limited life, must be created before manufacturing can begin. These mould tools must then be safely stored to ensure future identical parts can be manufactured. If damaged, recreating exact replica mould tooling is an expensive and time-consuming process. The cost and difficulty of manufacturing new tooling therefore prohibits the economic manufacture of many bespoke parts, or results in significant lead time increases and project delays. An alternative approach, that is gaining increasing interest, is the use of 3D printing for concrete. A robot is used to guide a concrete deposition nozzle in a layer by layer deposition path, similar to polymer fused deposition modelling. However, in Concrete 3D Printing the combination of complex material rheological properties, and the need for high volumetric deposition rates, means achieving net shape precision parts from the deposition step is extremely challenging. To address this issue, a hybrid robotic 3D printing and milling system has been developed, with integrated in-process structured-light metrology, to enable the production of precision concrete parts with well controlled geometric features. The integrated robot manufacturing cell is described. Examples structures that demonstrate the ability to create and replicate complex concrete parts with improved surface properties are illustrated. Components with surface features that enable concrete assemblies are studied to illustrate the capability of the hybrid process to manufacture challenging parts with demanding geometric requirements. These structures are used to quantify the performance of the current system and illustrate the future potential for the approach.

The flat spectrum radio quasar (FSRQ) PKS 1510−089 is known for its complex multiwavelength behaviour and it is one of only a few FSRQs detected in very-high-energy (VHE, E > 100 GeV) γ rays. The VHE γ-ray observations with H.E.S.S. and MAGIC in late May and early June 2016 resulted in the detection of an unprecedented flare, which revealed, for the first time, VHE γ-ray intranight variability for this source. While a common variability timescale of 1.5 h has been found, there is a significant deviation near the end of the flare, with a timescale of ∼20 min marking the cessation of the event. The peak flux is nearly two orders of magnitude above the low-level emission. For the first time, a curvature was detected in the VHE γ-ray spectrum of PKS 1510–089, which can be fully explained by the absorption on the part of the extragalactic background light. Optical R-band observations with ATOM revealed a counterpart of the γ-ray flare, even though the detailed flux evolution differs from the VHE γ-ray light curve. Interestingly, a steep flux decrease was observed at the same time as the cessation of the VHE γ-ray flare. In the high-energy (HE, E > 100 MeV) γ-ray band, only a moderate flux increase was observed with Fermi-LAT, while the HE γ-ray spectrum significantly hardens up to a photon index of 1.6. A search for broad-line region (BLR) absorption features in the γ-ray spectrum indicates that the emission region is located outside of the BLR. Radio very-long-baseline interferometry observations reveal a fast-moving knot interacting with a standing jet feature around the time of the flare. As the standing feature is located ∼50 pc from the black hole, the emission region of the flare may have been located at a significant distance from the black hole. If this is indeed a true correlation, the VHE γ rays must have been produced far down in the jet, where turbulent plasma crosses a standing shock.

Jets which are powered by an AGN are a crucial element in the study of their central black holes (BH) and their immediate surroundings. The formation of such jets is the subject of intense research, mainly based on the dichotomy presented by the two main jet launching scenarios $-$ the one from Blandford & Payne (1982), and the one from Blandford & Znajek (1977). In this work we study the prominent and nearby radio galaxy 3C 84 (NGC 1275) with 15, 43, and 86 GHz quasi-simultaneous VLBI observations. From these we determine the jet apex to be located $83\pm7\,\mu$as ($0.028-0.11$pc) upstream of the 86 GHz VLBI core, applying a two dimensional cross-correlation analysis. A byproduct of this analysis are spectral index maps, in which we identify a robust spectral index gradient in the north-south direction, for the first time at such high resolution, for the 43-86 GHz pair. The magnetic field strength at distances from the VLBI core comparable to measurements from the literature ($\sim10$ Schwarzschild radii) for other prominent AGN, like NGC 1052 and M 87, is computed to be $70-600$G. We present the results of multi-epoch, multi-frequency monitoring of a blazar 4C +29.45, which was regularly monitored as part of the Interferometric Monitoring of GAmma-ray Bright Active Galactic Nuclei (iMOGABA) program - a key science program of the Korean Very long baseline interferometry Network (KVN). Observations were conducted simultaneously at 22, 43, 86, and 129 GHz during the 4 years from 5 December 2012 to 28 December 2016. We also used additional data from the 15 GHz Owens Valley Radio Observatory (OVRO) monitoring program. From the 15 GHz light curve, we estimated the variability time scales of the source during several radio flux enhancements. We found that the source experienced 6 radio flux enhancements with variability time scales of 9-187 days during the observing period, yielding corresponding variability Doppler factors of 9-27. From the multi-frequency simultaneous KVN observations, we were able to obtain accurate radio spectra of the source and hence to more precisely measure the turnover frequencies $\nu_{\rm r}$ of synchrotron self-absorbed (SSA) emission with a mean value of $\nu_{rm r}$ = 28.9 GHz. Using jet geometry assumptions, we estimated the size of the emitting region at the turnover frequency. We found that the equipartition magnetic field strength is up to two orders of magnitudes higher than the SSA magnetic field strength (0.001-0.1 G). This is consistent with the source being particle dominated. We performed a careful analysis of the systematic errors related to making these estimations. BACKGROUND: The BNT162b2 mRNA vaccine has been shown to be effective at preventing serious COVID-19 events in clinical trials. There is less evidence on effectiveness in real-world settings, especially for older people. Here, we aimed to estimate vaccine effectiveness in the context of the rapid NHS mass-vaccination programme in England, exploiting age-based vaccination eligibility thresholds to minimise and correct for selection bias. METHODS: We studied 170,226 individuals between the ages of 80 and 83 years from community settings outside care homes who received one dose of BNT162b2 mRNA between the 15 and 20 December 2020 and were scheduled a second dose 21 days later. We matched these vaccine recipients to slightly younger (aged 76-79 years) persons not yet eligible to receive the vaccine on gender, area of residence, area deprivation, health status, living arrangements, acute illness, and history of seasonal flu vaccination. We compared their rates of COVID-19 positivity and hospitalisation in the subsequent 45 days. We adjusted for the increasing concentration of COVID-19 positivity in the control population caused by the requirement to have no COVID-19 symptoms prior to vaccination. RESULTS: Emergency hospital admissions were 51.0% (95% confidence interval 19.9 to 69.5%) lower and positive COVID-19 tests were 55.2% (40.8 to 66.8%) lower for vaccinated individuals compared to matched controls 21 to 27 days after first vaccination. Emergency admissions were 75.6% (52.8 to 87.6%) lower, and positive COVID-19 tests were 70.1% (55.1 to 80.1%) lower 35 to 41 days after first vaccination when 79% of participants had received a second dose within 26 days of their first dose. CONCLUSIONS: Receipt of the BNT162b2 mRNA vaccine is effective at reducing COVID-19 hospitalisations and infections. The nationwide vaccination of older adults in England with the BNT162b2 mRNA vaccine reduced the burden of COVID-19.

Controversial studies on the jet collimation profile of BL Lacertae (BL Lac), the eponymous blazar of BL Lac objects class, complicate the scenario in this already puzzling class of objects. Understanding the jet geometry, in connection with the jet kinematics and the physical conditions in the surrounding medium, is fundamental to better constrain the formation, acceleration and collimation mechanisms in extragalactic jets. With the aim of investigating the jet geometry in the innermost regions of the jet of BL Lac, and solving the controversy, we explore the radio jet in this source, using high resolution millimeter-wave VLBI data. We collect 86GHz GMVA and 43GHz VLBA data to obtain stacked images that we use to infer the jet collimation profile by means of two comparable methods. We analyze the kinematics at 86GHz, and we discuss it in the context of the jet expansion. Finally we consider a possible implication of the Bondi sphere in shaping the different expanding region observed along the jet. We found that the jet in BL Lac expands with an overall conical geometry. A higher expanding rate region is observed between ~5 and 10 pc (de-projected) from the black hole. Such a region is associated with the decrease in brightness usually observed in high-frequency VLBI images of BL Lac. The jet retrieves the original jet expansion around 17 pc, where the presence of a recollimation shock is supported by both the jet profile and the 15GHz kinematics (MOJAVE survey). The change in the jet expansion profile occurring at ~5 pc could be associated with a change in the external pressure profile in correspondence of the Bondi radius (~3.3X10$^5$$R_s$)., Comment: 10 pages, 12 figures. The EXO-200 experiment searched for neutrinoless double-beta decay of $^{136}$Xe with a single-phase liquid xenon detector. It used an active mass of 110 kg of 80.6%-enriched liquid xenon in an ultra-low background time projection chamber with ionization and scintillation detection and readout. This paper describes the design and performance of the various support systems necessary for detector operation, including cryogenics, xenon handling, and controls. Novel features of the system were driven by the need to protect the thin-walled detector chamber containing the liquid xenon, to achieve high chemical purity of the Xe, and to maintain thermal uniformity across the detector., Comment: Manuscript updated in response to JINST reviewer comments

We measure the 10 and $18\mu$m silicate features in a sample of 67 local ($z<0.1$) type 1 active galactic nuclei (AGN) with available {\it Spitzer} spectra dominated by non-stellar processes. We find that the $10\mu$m silicate feature peaks at $10.3^{+0.7}_{-0.9}\mu$m with a strength (Si$_{p}$ = ln f$_{p}$(spectrum)/f$_{p}$(continuum)) of $0.11^{+0.15}_{-0.36}$, while the $18\mu$m one peaks at $17.3^{+0.4}_{-0.7}\mu$m with a strength of $0.14^{+0.06}_{-0.06}$. We select from this sample sources with the strongest 10$\mu$m silicate strength ($\sigma_{Si_{10\mu m}}>0.28$, 10 objects). We carry out a detailed modeling of the IRS/{\it Spitzer} spectra by comparing several models that assume different geometries and dust composition: a smooth torus model, two clumpy torus models, a two-phase medium torus model, and a disk+outflow clumpy model. We find that the silicate features are well modeled by the clumpy model of Nenkova et al. 2008, and among all models those including outflows and complex dust composition are the best (Hoenig et al. 2017). We note that even in AGN-dominated galaxies it is usually necessary to add stellar contributions to reproduce the emission at the shortest wavelengths., Comment: Accepted for publication in ApJ

BACKGROUND: Couples who receive a prenatal diagnosis of a fetal anomaly in Victoria, Australia, are generally offered a choice about whether or not to continue with the pregnancy. When a severe or 'lethal' abnormality is diagnosed, some couples decide to continue the pregnancy in the knowledge that their baby may die before or shortly after birth. Several Australian parents who published personal accounts of that experience describe a lack of clear clinical pathways, suggesting those who decide to continue a pregnancy following a diagnosis of a 'lethal fetal abnormality' (LFA) may not be receiving optimal care. AIMS: This study aimed to provide empirical Australian evidence of views and experiences of care provision from health professionals (HPs) and parents. MATERIALS AND METHODS: Two sequential phases of this qualitative study purposively recruited a range of key HPs and parents. Semi-structured interviews were thematically analysed. RESULTS: Findings reveal that current care provision following prenatal diagnosis of an LFA is 'ad hoc' with both participant groups identifying disparities between parents' needs and available care. However, the goodwill and good intentions of all HPs involved was apparent. There was strong support from both groups for considering a model of perinatal palliative care (PPC) based on existing programs overseas. CONCLUSIONS: Future care provision in this setting needs to be redefined. A formal PPC program could ensure better and more consistent experiences of support for parents as well as the HPs working in the field.

Interestingly, a steep flux decrease is observed at the same time as the cessation of the VHE flare. In the high energy (HE, $E>100\,$MeV) $\gamma$-ray band only a moderate flux increase is observed with Fermi-LAT, while the HE $\gamma$-ray spectrum significantly hardens up to a photon index of 1.6. A search for broad-line region (BLR) absorption features in the $\gamma$-ray spectrum indicates that the emission region is located outside of the BLR. Radio VLBI observations reveal a fast moving knot interacting with a standing jet feature around the time of the flare. As the standing feature is located $\sim 50\,$pc from the black hole, the emission region of the flare may have been located at a significant distance from the black hole. If this correlation is indeed true, VHE $\gamma$ rays have been produced far down the jet where turbulent plasma crosses a standing shock., Comment: 25 pages, 17 figures. We measure the 10 and $18\mu$m silicate features in a sample of 67 local ($z<0.1$) type 1 active galactic nuclei (AGN) with available {\it Spitzer} spectra dominated by non-stellar processes. We find that the $10\mu$m silicate feature peaks at $10.3^{+0.7}_{-0.9}\mu$m with a strength (Si$_{p}$ = ln f$_{p}$(spectrum)/f$_{p}$(continuum)) of $0.11^{+0.15}_{-0.36}$, while the $18\mu$m one peaks at $17.3^{+0.4}_{-0.7}\mu$m with a strength of $0.14^{+0.06}_{-0.06}$. We select from this sample sources with the strongest 10$\mu$m silicate strength ($\sigma_{Si_{10\mu m}}>0.28$, 10 objects). We carry out a detailed modeling of the IRS/{\it Spitzer} spectra by comparing several models that assume different geometries and dust composition: a smooth torus model, two clumpy torus models, a two-phase medium torus model, and a disk+outflow clumpy model. We find that the silicate features are well modeled by the clumpy model of Nenkova et al. 2008, and among all models those including outflows and complex dust composition are the best (Hoenig et al. 2017). We note that even in AGN-dominated galaxies it is usually necessary to add stellar contributions to reproduce the emission at the shortest wavelengths., Comment: Accepted for publication in ApJ

Extrusion cooking is widely used to improve the functional properties of dietary fibre of food processing by-products. This study investigated the capacity of extrusion cooking to increase the proportion of dietary fibre in the Australian sweet lupin seed coat which is soluble and modify its physicochemical properties (water binding capacity and water solubility). Fractional factorial design was used to identify which of the extrusion cooking factors most affected seed coat dietary fibre composition and physicochemical properties. Extrusion cooking significantly increased soluble dietary fibre content of lupin seed coat, from 29.03 g/kg dry basis to a maximum of 90.28 g/kg dry basis, while decreasing insoluble dietary fibre content from 898.91 g/kg dry basis to 828.85 g/kg dry basis. Water solubility increased (4.02–4.47% vs. 5.37–9.64% dry basis) whereas water binding capacity slightly decreased (3.84–3.88 g/g vs. 3.15–3.73 g/g dry basis) after extrusion cooking. The screw speed, total moisture content in barrel and barrel temperature were identified as the most important processing factors. These findings suggest that extrusion cooking could be a practical technology to increase the soluble dietary fibre content in lupin seed coat and enhance health benefits of the seed coat.

We report results from a deep polarization imaging of the nearby radio galaxy 3C$\,$84 (NGC$\,$1275). The source was observed with the Global Millimeter VLBI Array (GMVA) at 86$\,$GHz at an ultra-high angular resolution of $50\mu$as (corresponding to 250$R_{s}$). We also add complementary multi-wavelength data from the Very Long Baseline Array (VLBA; 15 & 43$\,$GHz) and from the Atacama Large Millimeter/submillimeter Array (ALMA; 97.5, 233.0, and 343.5$\,$GHz). At 86$\,$GHz, we measure a fractional linear polarization of $\sim2$% in the VLBI core region. The polarization morphology suggests that the emission is associated with an underlying limb-brightened jet. The fractional linear polarization is lower at 43 and 15$\,$GHz ($\sim0.3-0.7$% and $<0.1$%, respectively). This suggests an increasing linear polarization degree towards shorter wavelengths on VLBI scales. We also obtain a large rotation measure (RM) of $\sim10^{5-6}~{\rm rad/m^{2}}$ in the core at $\gtrsim$43$\,$GHz. Moreover, the VLBA 43$\,$GHz observations show a variable RM in the VLBI core region during a small flare in 2015. Faraday depolarization and Faraday conversion in an inhomogeneous and mildly relativistic plasma could explain the observed linear polarization characteristics and the previously measured frequency dependence of the circular polarization. Our Faraday depolarization modeling suggests that the RM most likely originates from an external screen with a highly uniform RM distribution. To explain the large RM value, the uniform RM distribution, and the RM variability, we suggest that the Faraday rotation is caused by a boundary layer in a transversely stratified jet. Located at the center of the Perseus cluster, 3C 84 is an extremely bright and nearby radio galaxy. Because of the strong diffuse thermal emission from the cluster in X-rays, the detailed properties and the origin of a power-law component from the central active galactic nucleus (AGN) remains unclear in the source. We report here the first NuSTAR observations of 3C 84. The source was observed for 24.2 and 32 ks on February 01 and 04, 2018, respectively. NuSTAR observations spectrally decompose the power-law AGN component above 10 keV. The power-law component dominates the spectrum above 20 keV with a photon index $\sim$1.9 and an energy flux F$_{20-30 keV}$ = 1.0 $\times$10$^{-11}$ erg cm$^{-2}$ s$^{-1}$, corresponding to an isotropic luminosity, $L_{20-30 keV}$ = 7.4$\times$10$^{42}$ erg s$^{-1}$. We discuss possible emitting sites for the power-law component. The expected thermal emission from the accretion disk is not hot enough to account for the hard X-rays detected from the source. Similar X-ray and $\gamma$-ray photon indices and long-term flux variations, the absence of cutoff energy in the hard X-ray spectrum of the source, correlated hard X-ray flux and hardness ratio variations, and the similarity of optical-X-ray slope to blazar rather than Seyfert galaxies supports the hard X-ray power-law component originating from the jet., Comment: accepted for publication in ApJ Letters

Understanding the launching, acceleration, and collimation of jets powered by active galactic nuclei remains an outstanding problem in relativistic astrophysics. This is partly because observational tests of jet formation models suffer from the limited angular resolution of ground-based very long baseline interferometry that has thus far been able to probe the transverse jet structure in the acceleration and collimation zone of only two sources. Here we report radio interferometric observations of 3C 84 (NGC 1275), the central galaxy of the Perseus cluster, made with an array including the orbiting radio telescope of the RadioAstron mission. The obtained image transversely resolves the edge-brightened jet in 3C 84 only 30 microarcseconds from the core, which is ten times closer to the central engine than what has been possible in previous ground-based observations, and it allows us to measure the jet collimation profile from ~ 100 to ~10000 gravitational radii from the black hole. The previously found, almost cylindrical jet profile on scales larger than a few thousand r_g is now seen to continue at least down to a few hundred r_g from the black hole and we find a broad jet with a transverse radius larger than about 250 r_g at only 350 r_g from the core. If the bright outer jet layer is launched by the black hole ergosphere, it has to rapidly expand laterally on scales smaller than 100 r_g. AIM: The aim of this study was to determine the barriers and enablers influencing the uptake of two recommendations from a tertiary paediatric hospital's clinical practice guidelines by maternal and child health nurses (MCHNs) and emergency department (ED) doctors: (i) explaining normal crying; and (ii) avoiding attributing crying to gastro-oesophageal reflux (GOR) and limiting anti-reflux medication use. METHODS: The study was designed as 1-h focus group discussions, guided by the Theoretical Domains Framework, and a short questionnaire, with a purposive sample of MCHNs and ED doctors in Victoria, Australia in (March to September) 2015. Analyses were conducted by inductive content analysis to identify key barriers and enablers. RESULTS: A total of 53 MCHNs and 25 ED doctors participated in 11 discussions. For explaining normal crying, key enablers were: adequate experience/competency, perceiving it was their role to explain and belief it prevented over-medicalisation. The main barriers were time restriction and beliefs about parents' perceptions. For MCHNs, key barriers to avoid attributing crying to GOR were: lack of knowledge and confusion around their role in diagnosing GOR. For ED doctors, key barriers to limiting anti-reflux medication were: parents requesting medication, concern about disrupting the parent-primary-care practitioner relationship and belief it was not their role to cease anti-reflux medication. CONCLUSIONS: Overall, MCHN and ED doctors were proficient in describing normal crying. However, several barriers to best practice were identified, including time constraints and belief about consequences of intervening. These results will be used to develop effective interventions to address the identified barriers and enablers to optimise the management of infant colic.

In the era of genomic medicine, expanded genetic screening may be offered to donors for the purpose of avoiding transmission of harmful genetic mutations. The objective of this study was to explore the attitudes of donors and recipients toward the expanded genetic screening of donors. METHODS: Qualitative interview study with thematic analysis, undertaken in a tertiary fertility centre. Semi-structured in-depth qualitative interviews were conducted with eleven recipients and nine donors from three different cohorts (sperm, egg and embryo donors/recipients). RESULTS: Donors and recipients acknowledged the importance of genetic information and were comfortable with the existing level of genetic screening of donors. Recipients recognised some potential benefits of expanded genetic screening of donors; however both recipients and donors were apprehensive about extended genomic technologies, with concerns about how this information would be used and the ethics of genetic selectivity. CONCLUSION: Participants in donor programs support some level of genetic screening of donors, but are wary of expanding genetic screening beyond current levels.

BACKGROUND: Tea consumption may improve endothelial function and blood pressure via increased bioavailability and bioactivity of nitric oxide. However, questions remain as to the impact of the common practice of adding milk. OBJECTIVE: To investigate the effect of regular consumption of black tea, with and without milk, on vascular function and blood pressure in healthy volunteers. DESIGN: A randomised, controlled, crossover study was performed in 17 healthy volunteers; 7 men and 10 women, mean age 22.4 ± 3.0 years. Participants received each of the following treatments in random order for 4 weeks, with no washout period in between, (i) hot water, (ii) black tea and (iii) black tea with milk. Vascular function was assessed using flow-mediated dilatation (FMD) of the brachial artery at the end of each treatment period. In addition, participants monitored their home blood pressure for the last 7 days of each treatment period. A blood and urine sample was also collected at the end of each treatment period. RESULTS: Black tea increased FMD compared to the hot water control group (1.00 ± 0.18%, P < 0.0001). Black tea with milk decreased FMD compared to both the hot water control (-0.64 ± 0.19%, P = 0.001) and black tea (-1.64 ± 0.19%, P < 0.0001). Compared with hot water, black tea did not alter blood pressure, while black tea with milk increased systolic (1.1 ± 0.5 mmHg, P = 0.03) and diastolic blood pressure (2.0 ± 0.5 mmHg, P < 0.0001). Black tea (-1.8 ± 0.5 bpm, P < 0.001) and black tea with milk (-1.8 ± 0.6 bpm, P < 0.001) lowered heart rate compared to hot water. No significant difference for plasma nitrate or nitrite was observed between treatment groups. CONCLUSIONS: The addition of milk to black tea alters the acute/short-term impact of regular tea consumption on vascular function and blood pressure in young healthy men and women. Background: In recognition of their multiple benefits on environment, food security, and human health, pulses are attracting worldwide attention. The seed coat is a major by-product of pulse processing, and its only markets are as low value ruminant feed and very limited use in high fibre foods. Recently, accumulating studies have suggested that this underutilised by-product has greater potential as a novel natural “nutritious dietary fibre” which can be used as a functional food ingredient. Scope and approach: This review discusses biochemical and physicochemical functionalities of seed coats of six globally important pulses: chickpea, field pea, faba/broad bean, lentil and mung bean with a special emphasis on the emerging food pulse lupin. Food process modification and recent human food applications of the seed coats are summarized. Bio-availability of the seed coat compounds, and phomopsins contaminated lupin seed coats as a typical example of safety issue are discussed. Key findings and conclusions: High levels of dietary fibre, minerals and potential health-promoting phytochemicals in the seed coats indicate their great potential to be used as a natural “nutritious dietary fibre”. However, further in-depth studies are required to improve their desirable nutritional, physiological and techno-functional properties whilst minimizing any undesirable ones.

Our purpose was to estimate dietary nitrate consumption and identify key dietary changes which have occurred over time within a representative sample of Australian women. Women from the 1946–1951 cohort of the Australian Longitudinal Study on Women’s Health with complete food frequency questionnaire data for both 2001 and 2013 were included for analysis. Dietary nitrate intakes were calculated using key published nitrate databases. Diet quality scores including the Australian Recommended Food Score, the Mediterranean Diet Score and the Nutrient Rich Foods Index were calculated along with food group serves as per the Australian Dietary Guidelines. Wilcoxon matched pairs tests were used to test for change in dietary intakes and Spearman’s correlations were used to examine associations. In our sample of 8161 Australian women, dietary nitrate intakes were on average 65–70 mg/day, and we detected a significant increase in dietary nitrate consumption over time (+6.57 mg/day). Vegetables were the primary source of dietary nitrate (81–83%), in particular lettuce (26%), spinach (14–20%), beetroot (10–11%), and celery (7–8%) contributed primarily to vegetable nitrate intakes. Further, increased dietary nitrate intakes were associated with improved diet quality scores (r = 0.3, p < 0.0001). Although there is emerging evidence indicating that higher habitual dietary nitrate intakes are associated with reduced morbidity and mortality, future work in this area should consider how dietary nitrate within the context of overall diet quality can facilitate health to ensure consistent public health messages are conveyed.

Seeds of the legume lupin (Lupinus spp.) are becoming increasingly important as human food. The seed coat, at ~25% of the whole seed of Lupinus angustifolius (Australian sweet lupin, ASL), is the main by-product of lupin kernel flour production. The primary market for lupin seed coat is low value feed with very limited use in foods. In this study, seed coats of six ASL commercial varieties from two growing sites were sampled for identification and quantification of polyphenols using a high-performance liquid chromatography (HPLC) with diode array detector (DAD) and coupled with a triple quadrupole mass spectrometer which equipped with electrospray ionization source (ESI-MS/MS). Three flavones (apigenin-7-O-ß-apiofuranosyl-6,8-di-C-ß-glucopyranoside, vicenin 2, and apigenin-7-O-ß-glucopyranoside), one isoflavone (genistein) and one dihydroflavonol derivative (aromadendrin-6-C-ß-D-glucopyranosyl-7-O-[ß-D-apiofuranosyl-(1 ? 2)]-O-ß-D-glucopyranoside), and several hydroxybenzoic and hydroxycinnamic acid derivatives were identified. Considerable variations in levels of individual polyphenols were found but apigenin-7-O-ß-apiofuranosyl-6,8-di-C-ß-glucopyranoside was the predominant polyphenol in all samples accounting for 73.08–82.89% of the total free polyphenols. These results suggest that ASL seed coat could be valuable dietary source of polyphenols.

Background: Dietary nitrate is an important source of nitric oxide (NO), a molecule critical for cardiovascular health. Nitrate is sequentially reduced to NO through an enterosalivary nitrate-nitrite-NO pathway that involves the oral microbiome. This pathway is considered an important adjunct pathway to the classical l-arginine-NO synthase pathway. Objective: The objective of this study was to systematically assess the evidence for dietary nitrate intake and improved cardiovascular health from both human and animal studies. Design: A systematic literature search was performed according to PRISMA (Preferred Reporting Items for Systematic Reviews and Meta-Analyses) guidelines by using key search terms in Medline and EMBASE databases and defined inclusion and exclusion criteria. Results: Thirty-seven articles on humans and 14 articles on animals were included from 12,541 screened references. Data on the effects of dietary nitrate on blood pressure, endothelial function, ischemic reperfusion injury, arterial stiffness, platelet function, and cerebral blood flow in both human and animal models were identified. Beneficial effects of nitrate on vascular health have predominantly been observed in healthy human populations, whereas effects in populations at risk of cardiovascular disease are less clear. Few studies have investigated the long-term effects of dietary nitrate on cardiovascular disease clinical endpoints. In animal studies, there is evidence that nitrate improves blood pressure and endothelial function, particularly in animal models with reduced NO bioavailability. Nitrate dose seems to be a critical factor because there is evidence of cross-talk between the 2 pathways of NO production. Conclusions Evidence for a beneficial effect in humans at risk of cardiovascular disease is limited. Furthermore, there is a need to investigate the long-term effects of dietary nitrate on cardiovascular disease clinical endpoints. Further animal studies are required to elucidate

Scope: The cardioprotective effects of apples are primarily attributed to flavonoids, found predominantly in the skin. This study aimed to determine if acute and/or chronic (4 weeks) ingestion of flavonoid-rich apples improves endothelial function, blood pressure (BP), and arterial stiffness in individuals at risk for cardiovascular diseases (CVD). Methods and results: In this randomized, controlled cross-over trial, acute and 4 week intake of apple with skin (high flavonoid apple, HFA) is compared to intake of apple flesh only (low flavonoid apple, LFA) in 30 participants. The primary outcome is endothelial function assessed using flow-mediated dilation (FMD) of the brachial artery, while main secondary outcomes are 24 h ambulatory BP and arterial stiffness. Other outcomes include fasting serum glucose and lipoprotein profile, plasma heme oxygenase-1 (Hmox-1), F2-isoprostanes, flavonoid metabolites, and plasma and salivary nitrate (NO3-) and nitrite (NO2-) concentrations. Compared to LFA control, the HFA results in a significant increase in FMD acutely (0.8%, p < 0.001) and after 4 weeks chronic intake (0.5%, p < 0.001), and in plasma flavonoid metabolites (p < 0.0001). Other outcomes are not altered significantly. Conclusion: A lower risk of CVD with higher apple consumption could be mediated by the beneficial effect of apple skin on endothelial function, both acutely and chronically.

Background: Emerging evidence suggests that increasing intakes of nitrate-rich vegetables may be an effective approach to reduce blood pressure. Objective: Our primary aim was to determine whether daily consumption of nitrate-rich vegetables over 4 wk would result in lower blood pressure. Design: Thirty participants with prehypertension or untreated grade 1 hypertension were recruited to a randomized controlled crossover trial with 4-wk treatment periods separated by 4-wk washout periods. Participants completed 3 treatments in random order: 1) increased intake (∼200 g/d) of nitrate-rich vegetables [high-nitrate (HN); ∼150 mg nitrate/d], 2) increased intake (∼200 g/d) of nitrate-poor vegetables [low-nitrate (LN); ∼22 mg nitrate/d], and 3) no increase in vegetables (control; ∼6 mg nitrate/d). Compliance was assessed with the use of food diaries and by measuring plasma nitrate and carotenoids. Nitrate metabolism was assessed with the use of plasma, salivary, and urinary nitrate and nitrite concentrations. The primary outcome was blood pressure assessed by using 24-h ambulatory, home, and clinic measurements. Secondary outcomes included measures of arterial stiffness. Results: Plasma nitrate and nitrite concentrations increased with the HN treatment in comparison to the LN and control treatments (P < 0.001). Plasma carotenoids increased with the HN and LN treatments compared with the control (P < 0.01). HN treatment did not reduce systolic blood pressure [24-h ambulatory—HN: 127.4 ± 1.1 mm Hg; LN: 128.6 ± 1.1 mm Hg; control: 126.2 ± 1.1 mm Hg (P = 0.20); home—HN: 127.4 ± 0.7 mm Hg; LN: 128.7 ± 0.7 mm Hg; control: 128.3 ± 0.7 mm Hg (P = 0.36); clinic—HN: 128.4 ± 1.3 mm Hg; LN: 130.3 ± 1.3 mm Hg; control: 129.8 ± 1.3 mm Hg (P = 0.49)] or diastolic blood pressure compared with LN and control treatments (P > 0.05) after adjustment for pretreatment values, treatment period, and treatment order. Understanding the formation of relativistic jets in active galactic nuclei remains an elusive problem1.This is partly because observational tests of jet formation models suffer from the limited angular resolution of ground-based very-long-baseline interferometry that has thus far been able to probe the structure of the jet acceleration and collimation region in only two sources2,3. Here, we report observations of 3C84 (NGC 1275)-the central galaxy of the Perseus cluster-made with an interferometric array including the orbiting radio telescope of the RadioAstron4mission. The data transversely resolve the edge-brightened jet in 3C84 only 30 µas from the core, which is ten times closer to the central engine than was possible in previous ground-based observations5and allows us to measure the jet collimation profile from ~102to ~104gravitational radii (rg) from the black hole. The previously found5, almost cylindrical jet profile on scales larger than a few thousand rgis seen to continue at least down to a few hundred rgfrom the black hole, and we find a broad jet with a transverse radius of â‰250 rgat only 350 rgfrom the core. This implies that either the bright outer jet layer goes through a very rapid lateral expansion on scales â‰102rgor it is launched from the accretion disk.

The recommendation to increase vegetable intake is part of most dietary guidelines. Despite widespread and long-running public health messages to increase vegetable intake, similar to other countries worldwide, less than 1 in 10 adult Australians manage to meet target advice. Dietary guidelines are predominantly based on studies linking diets high in vegetables with lower risk of chronic diseases. Identifying vegetables with the strongest health benefits and incorporating these into dietary recommendations may enhance public health initiatives around vegetable intake. These enhanced public health initiatives would be targeted at reducing the risk of chronic diseases, such as cardiovascular diseases (CVD). Specific vegetable types contain high levels of particular nutrients and phytochemicals linked with cardiovascular health benefits. However, it is not clear if increasing intake of these specific vegetable types will result in larger benefits on risk of chronic diseases. This review presents an overview of the evidence for the relationships of specific types of vegetables, including leafy green, cruciferous, allium, yellow-orange-red and legumes, with subclinical and clinical CVD outcomes in observational epidemiological studies.

We aimed to examine the relationship between total fluid and the sources of fluid consumption, risk of rapid renal decline, cardiovascular disease (CVD) mortality and all-cause mortality in elderly women. Design, setting and participants We conducted a longitudinal analysis of a population-based cohort study of 1055 women aged >=70? ...years residing in Australia. Main outcome measures The associations between total daily fluid intake (defined as total volume of beverage excluding alcohol and milk) and the types of fluid (water, black tea, coffee, milk and other fluids) measured as cups per day and rapid renal decline, CVD and all-cause mortality were assessed using adjusted logistic and Cox regression analyses. Results Over a follow-up period of 10? ...years, 70 (6.6%) experienced rapid renal decline and 362 (34.4%) died, of which 142 (13.5%) deaths were attributed to CVD. The median (IQR) intake of total fluid was 10.4 (8.5-12.5) cups per day, with water (median (IQR) 4 (2-6) cups per day) and black tea (median (IQR) 3 (1-4) cups per day) being the most frequent type of fluid consumed. Every cup per day higher intake of black tea was associated with adjusted HRs of 0.90 (95% CI 0.81 to 0.99) and 0.92 (95% CI 0.86 to 0.98) for CVD mortality and all-cause mortality, respectively. There were no associations between black tea intake and rapid renal decline, or between the quantity or type of other fluids, including water intake, and any clinical outcomes. Conclusions Habitual higher intake of black tea may potentially improve long-term health outcomes, independent of treating traditional CVD risk factors, but validation of our study findings is essential.

A diet rich in plant polyphenols has been suggested to reduce the incidence of cardiovascular disease and type 2 diabetes mellitus, in part, via improvements in endothelial function. Coffee is a rich source of phenolic compounds including the phenolic acid, chlorogenic acid (CGA). The aim of the study was to investigate the effect of coffee as a whole beverage on endothelial function, blood pressure and blood glucose concentration. Twelve healthy men and women were recruited to a randomised, placebo-controlled, cross-over study, with three treatments tested: (i) 18 g of ground caffeinated coffee containing 300 mg CGA in 200 mL of hot water, (ii) 18 g of decaffeinated coffee containing 287 mg CGA in 200 mL of hot water, and (iii) 200 mL of hot water (control). Treatment beverages were consumed twice, two hours apart, with the second beverage consumed simultaneously with a 75 g glucose load. Blood pressure was recorded and the finger prick glucose test was performed at time = 0 and then every 30 minutes up to 2 hours. Endothelial function, assessed using flow-mediated dilatation (FMD) of the brachial artery, was measured at 1 hour and a blood sample taken at 2 hours to measure plasma nitrate/nitrite and 5-CGA concentrations. The FMD response was significantly higher in the caffeinated coffee group compared to both decaffeinated coffee and water groups (P < 0.001). There was no significant difference in the FMD response between decaffeinated coffee and water. Blood glucose concentrations and blood pressure were not different between the three treatment groups. In conclusion, the consumption of caffeinated coffee resulted in a significant improvement in endothelial function, but there was no evidence for benefit regarding glucose metabolism or blood pressure.
