1. Upprätthållande av ett säkert flöde över en sterilbarriär under vattenlinjen för ex vivo organperfusion

- Abstract
Ex vivo organ perfusion is a technique that aims to increase the amount of organ donors by treating an otherwise nonfunctional organ with a perfusion fluid outside of the body to restore its function. UGLK Science is an ex vivo organ perfusion company outside of Lund specializing in kidney transplantation. The organ is perfused in a machine which restores kidney function, allowing the organ to survive longer outside of the body. The tube for circulation of fluid in the system pumps the fluid upwards over the edge of the container. However, this causes air bubbles to form which have to be removed in order for the system to proceed without interruption. The presented project has therefore been carried out in collaboration with UGLK Science with the aim of developing a possible outlet located at the bottom of the container, maintaining a safe flow through a sterile barrier below the waterline. Risk analysis, fluid flow simulations and a literature study were performed in order to evaluate potential solutions. This resulted in a prototype and a specification of requirements. The requirements included, among other things, that the outlet must be biocompatible with blood, not affect the flow and allow emptying of the fluid. The final idea was a silicone-based, external outlet designed similarly to a funnel. The rounded edge of the outlet was inspired by a V-ring seal, which enables a geometric attachment to the container. The solution meets all criteria in the specification of requirements and enables a safe flow over the sterile barrier. A fully developed product based on the presented prototype of the outlet could facilitate UGLK Sciences' work and in the long run contribute to an increased number of available kidney donors.
