We describe a dataset on the crustacean Order Tanaidacea from the coasts of the Iberian Peninsula and surrounding seas, including the archipelagos of the Azores, Madeira, Savage, and the Canary Islands. The dataset gathers the records from all available sources published between 1828 to 2019, which were collected following a standardized Google Scholar search and cross checking each article’s reference lists. For each record, the dataset includes taxonomic, geographical, and ecological information, as well as remarks regarding the sampling methods. The dataset was further completed with 52 additional unpublished records obtained from screening the collections of the University Complutense of Madrid gathered from 35 shallow water surveys. Furthermore, 698 records from different oceanographic deep-sea campaigns have also been included. In total, 3456 records from 186 species in 22 families have been compiled. The dataset organises the current published and unpublished knowledge on tanaidaceans in the area and, by making it open access, it will allow comparisons of the distribution of tanaidaceans in zoogeographic studies.&nbsp

Glioblastoma is one of the most common and lethal types of primary brain tumor. Despite aggressive treatment with chemotherapy and radiotherapy, tumor recurrence within 6–9 months is common. To overcome this, more effective therapies targeting cancer cell stemness, invasion, metabolism, cell death resistance and the interactions of tumor cells with their surrounding microen-vironment are required. In this study, we performed a systematic review of the molecular mechanisms that drive glioblastoma progression, which led to the identification of 65 drugs/inhibitors that we screened for their efficacy to kill patient-derived glioma stem cells in two dimensional (2D) cul-tures and patient-derived three dimensional (3D) glioblastoma explant organoids (GBOs). From the screening, we found a group of drugs that presented different selectivity on different patient-derived in vitro models. Moreover, we found that Costunolide, a TERT inhibitor, was effective in reducing the cell viability in vitro of both primary tumor models as well as tumor models pre-treated with chemotherapy and radiotherapy. These results present a novel workflow for screening a relatively large groups of drugs, whose results could lead to the identification of more personalized and effective treatment for recurrent glioblastoma.

Caveolae-associated protein 3 (cavin3) is inactivated in most cancers. We characterized how cavin3 affects the cellular proteome using genome-edited cells together with label-free quantitative proteomics. These studies revealed a prominent role for cavin3 in DNA repair, with BRCA1 and BRCA1 A-complex components being downregulated on cavin3 deletion. Cellular and cell-free expression assays revealed a direct interaction between BRCA1 and cavin3 that occurs when cavin3 is released from caveolae that are disassembled in response to UV and mechanical stress. Overexpression and RNAi-depletion revealed that cavin3 sensitized various cancer cells to UV-induced apoptosis. Supporting a role in DNA repair, cavin3-deficient cells were sensitive to PARP inhibition, where concomitant depletion of 53BP1 restored BRCA1-dependent sensitivity to PARP inhibition. We conclude that cavin3 functions together with BRCA1 in multiple cancer-related pathways. The loss of cavin3 function may provide tumor cell survival by attenuating apoptotic sensitivity and hindering DNA repair under chronic stress conditions.

Objectives: Targeted immunotherapies such as chimeric antigen receptor (CAR)-T cells are emerging as attractive treatment options for glioblastoma, but rely on identification of a suitable tumor antigen. We validated a new target antigen for glioblastoma, fibroblast activation protein (FAP), by undertaking a detailed expression study of human samples. Methods: Glioblastoma and normal tissues were assessed using immunostaining, supported by analyses of published transcriptomic datasets. Short-term cultures of glioma neural stem (GNS) cells were compared to cultures of healthy astrocytes and neurons using flow cytometry. Glioblastoma tissues were dissociated and analysed by high-parameter flow cytometry and single-cell transcriptomics (scRNAseq). Results: Compared to normal brain, FAP was overexpressed at the gene and protein level in a large percentage of glioblastoma tissues, with highest levels of expression associated with poorer prognosis. FAP was also overexpressed in several paediatric brain cancers. FAP was commonly expressed by cultured GNS cells but absent from normal neurons and astrocytes. Within glioblastoma tissues, the strongest expression of FAP was around blood vessels. In fact, almost every tumor vessel was highlighted by FAP expression, whereas normal tissue vessels and cultured endothelial cells (ECs) lacked expression. Single-cell analyses of dissociated tumors facilitated a detailed characterisation of the main cellular components of the glioblastoma microenvironment and revealed that vessel-localised FAP is because of expression on both ECs and pericytes. Conclusion: Fibroblast activation protein is expressed by multiple cell types within glioblastoma, highlighting it as an ideal immunotherapy antigen to target destruction of both tumor cells and their supporting vascular network.

Ancient evidence of human presence in Europe is recorded in several Early Pleistocene archaeopalaeontological sites from Spain, France and Italy. This is the case of Barranco Leon (BL) and Fuente Nueva-3 (FN-3), two localities placed near the town of Orce (depression of Baza and Guadix, SE Spain) and dated to similar to 1.4 Ma. At these sites, huge assemblages of Oldowan tools and evidence of defleshing, butchering and marrow processing of large mammal bones have been recovered together with a deciduous tooth of Homo sp. in the case of level BL-D. In this study, we: (i) describe in detail the anthropic marks found in the bone assemblages from these sites; (ii) analyse patterns of defleshment, butchery and marrow processing, based on the modifications identified in the cortical surface of the fossils; and (iii) discuss on the subsistence strategies of the first hominins that inhabited the European subcontinent during Early Pleistocene times.

The current work focuses on the jet-in-crossflow penetration and dispersion behavior of water-in-oil emulsions in a high pressure environment. Both fuel injection strategies of using a water-in-oil emulsion and a jet-in-crossflow have demonstrated unique benefits in improving gas turbine performance from an emissions and efficiency standpoint. A jet-in-crossflow is very practical for use in gas turbine engines, rocket propulsion, and aircraft engines since it utilizes already available crossflow air to atomize fuel. Injecting water into a combustion chamber in the form of a water-in-oil emulsion allows for pollutant emissions reduction while reducing efficiency loses that may result from using a separate water or steam injection circuit. Dispersion effects on oil droplets are expected, therefore investigating the distribution of both oil and water droplets in the crossflow is an objective in this work. Understanding the synchronization and injection behavior of the two strategies is of key interest due to their combined benefits. A water-to-oil ratio and an ambient pressure parameter are developed for emulsion jet-in-crossflow trajectories. To this end, a total of 24 emulsion jet-in-crossflow tests were performed with varying ambient pressures of 2-8 atm and momentum flux ratios of 50, 85, and 120. Sobel edge filtering was applied to each averaged image obtained from a high speed video of each test case. Averaged and filtered images were used to resolve top and bottom edges of the trajectory in addition to the overall peak intensity up to 40 mm downstream of the injection point. An optimized correlation was established and found to differ from literature based correlations obtained under atmospheric pressure conditions. Overall it was found that additional parameters were not necessary for the top edge and peak intensity correlations, but a need for a unique emulsion bottom edge and width trajectory correlation was recognized. We used a computational approach to analyze the biomechanics of epithelial cell aggregates-islands, stripes, or entire monolayers-that combines both vertex and contact-inhibition-of-locomotion models to include cell-cell and cell-substrate adhesion. Examination of the distribution of cell protrusions (adhesion to the substrate) in the model predicted high-order profiles of cell organization that agree with those previously seen experimentally. Cells acquired an asymmetric distribution of basal protrusions, traction forces, and apical aspect ratios that decreased when moving from the edge to the island center. Our in silico analysis also showed that tension on cell-cell junctions and apical stress is not homogeneous across the island. Instead, these parameters are higher at the island center and scale up with island size, which we confirmed experimentally using laser ablation assays and immunofluorescence. Without formally being a three-dimensional model, our approach has the minimal elements necessary to reproduce the distribution of cellular forces and mechanical cross-Talk, as well as the distribution of principal stress in cells within epithelial cell aggregates. By making experimentally testable predictions, our approach can aid in mechanical analysis of epithelial tissues, especially when local changes in cell-cell and/or cell-substrate adhesion drive collective cell behavior.

Signaling from JAK (Janus kinase) protein kinases to STAT (signal transducers and activators of transcription) transcription factors is key to many aspects of biology and medicine, yet the mechanism by which cytokine receptors initiate signaling is enigmatic. We present a complete mechanistic model for activation of receptor-bound JAK2, based on an archetypal cytokine receptor, the growth hormone receptor. For this, we used fluorescence resonance energy transfer to monitor positioning of the JAK2 binding motif in the receptor dimer, substitution of the receptor extracellular domains with Jun zippers to control the position of its transmembrane (TM) helices, atomistic modeling of TM helix movements, and docking of the crystal structures of the JAK2 kinase and its inhibitory pseudokinase domain with an opposing kinase-pseudokinase domain pair. Activation of the receptor dimer induced a separation of its JAK2 binding motifs, driven by a ligand-induced transition from a parallel TM helix pair to a left-handed crossover arrangement. This separation leads to removal of the pseudokinase domain from the kinase domain of the partner JAK2 and pairing of the two kinase domains, facilitating trans-activation. This model may well generalize to other class I cytokine receptors.

H.264 is a new video coding standard developed jointly by ITU – T and the Moving Picture Expert Group (MPEG). It outperforms MPEG-2 by requiring around 50 % of the bit rate for a similar perceptual quality. It allows low bitrate networks to supply video at a quality previously not possible. Much of the previous work on concealment focuses on losses on the block level and uses the surrounding area information which is highly correlated. This thesis on the other hand analyzes and proposes ways to conceal errors in areas affecting much larger areas of a frame and with little information available from neighboring macroblocks. The methods proposed in this thesis include using weighted interpolation to create the visual effect of smoothness for the user when doing spatial concealment. High frequencies are also removed from concealed areas after inter concealment using a spatial filter. A background detector is proposed to reduce background blurriness and a method is proposed to dynamically adjust the range of pixel values after the concealment is done. This thesis work introduces the use of a scene change detector when doing interframe concealment to avoid mixing two different scenes. The results show that the perceived video quality can be significantly improved partly by removing highly noticeable artifacts and partly by giving a smooth image.

The maguiladora program was established by the Mexican government to encourage foreign investment and promote industrial growth. The success of the program encourages more and more participation each year. The low cost of labor in Mexico has attracted companies in all types of manufacturing in the United States with the exception of shipbuilding. With the focus on the domestic shipbuilding market over the last 25 years, U.S. shipyards have lacked the impetus to establish a maguiladora operation. The world market for shipbuilding has been steadily improving, while the U.S. domestic market has been steadily decreasing. The opportunities for U.S. shipyards to focus their strategies on the world market may not get any better.
