Water temperature is key to the study of aquatic ectotherm ecology, but precise measurements of individual-based thermal experience remain difficult to validate. The stable isotope composition of oxygen in biominerals acts as a natural thermometer due to the temperature dependence of isotopic fractionation between water and mineral phases. Coefficients of published temperature-dependent fractionation equations, however, vary among taxa (the so-called ‘vital effect’) without apparent consistent predictors, implying that species-specific experimental validation may be needed before inferring temperature from biomineral oxygen isotope thermometry. Here, we describe a meta-analysis conducted to assess the influence of biological and experimental sources of variation on the coefficients of published isotope thermometry equations. We observed that the thermal sensitivity (equation slope) was resistant to any biological or experimental factors, while the isotopic spacing between water and biomineral (equation intercept) showed consistent variation. Experimental conditions and phylogeny were the two main sources of variation in equation coefficients, where experiment approaches influenced both equation intercepts and the fit of the linear regression. Our results suggest that the use of common equation slopes and generalized taxa-specific equation intercepts may be appropriate under some circumstances. We additionally suggest that processes related to oxygen balance and osmoregulation may influence equation intercepts, and suggest further experimental work in this area. Finally, our observations provide ground for improvement for future design and reporting of biomineral thermometry experiments.

Large-scale, climate-induced synchrony in the productivity of fish populations is becoming more pronounced in the world's oceans. As synchrony increases, a population's ‘portfolio’ of responses can be diminished, in turn reducing its resilience to strong perturbation. Here we argue that the costs and benefits of trait synchronization, such as the expression of growth rate, are context dependent. Contrary to prevailing views, synchrony among individuals could actually be beneficial for populations if growth synchrony increases during favourable conditions, and then declines under poor conditions when a broader portfolio of responses could be useful. Importantly, growth synchrony among individuals within populations has seldom been measured, despite well-documented evidence of synchrony across populations. Here, we used century-scale time series of annual otolith growth to test for changes in growth synchronization among individuals within multiple populations of a marine keystone species (Atlantic cod, Gadus morhua). On the basis of 74,662 annual growth increments recorded in 13,749 otoliths, we detected a rising conformity in long-term growth rates within five northeast Atlantic cod populations in response to both favorable growth conditions and a large-scale, multidecadal mode of climate variability similar to the East Atlantic Pattern. The within-population synchrony was distinct from the across-population synchrony commonly reported for large-scale environmental drivers. Climate-linked, among-individual growth synchrony was also identified in other Northeast Atlantic pelagic, deep-sea and bivalve species. We hypothesize that growth synchrony in good years and growth asynchrony in poorer years reflects adaptive trait optimisation and bet hedging, respectively, that could confer an unexpected, but pervasive and stabilizing, impact on marine population productivity in response to large-scale environmental change.

Population connectivity is an increasingly important focal area for the understanding of how marine fish populations respond to anthropogenic pressures like climate change and fisheries. Our model species, the saithe Pollachius virens (Linnaeus, 1758), was chosen because genetic analyses have documented a mismatch between the assessed stocks and the biological populations. We combined laboratory experiments of saithe egg buoyancy and temperature-modulated development time, genetic field data, and high-resolution oceanographic models to disentangle the mechanisms causing isolation and mixing between the management units and the biological populations. Saithe egg buoyancy and development data were included in an individual-based model to simulate transport from all known spawning grounds in the Northeast Atlantic. The results show that interannual variability in the transport of early life stages is strongly influenced by wider climate systems (e.g. the North Atlantic Oscillation). One sample (Rockall) showed genetic differences from the other samples, and this finding was supported by the model showing low mixing with other populations and strong local retention. Strong retention of early life stages around Iceland could indicate an isolated population; however, this possible isolation is counteracted by active migration of adults westward from the Norwegian coast, and no genetic differentiation from other populations was found. Overall, the dispersal modeling supports the genetic analysis, showing a large and well-connected Central Northeast Atlantic population distributed across several management units. This mismatch between population structure and management units can potentially increase the risk for overexploitation of saithe.

Background There is heterogeneity in the names and anatomical descriptions of regional anesthetic techniques. This may have adverse consequences on education, research, and implementation into clinical practice. We aimed to produce standardized nomenclature for abdominal wall, paraspinal, and chest wall regional anesthetic techniques. Methods We conducted an international consensus study involving experts using a three-round Delphi method to produce a list of names and corresponding descriptions of anatomical targets. After long-list formulation by a Steering Committee, the first and second rounds involved anonymous electronic voting and commenting, with the third round involving a virtual round table discussion aiming to achieve consensus on items that had yet to achieve it. Novel names were presented where required for anatomical clarity and harmonization. Strong consensus was defined as ≥75% agreement and weak consensus as 50% to 74% agreement. Results Sixty expert Collaborators participated in this study. After three rounds and clarification, harmonization, and introduction of novel nomenclature, strong consensus was achieved for the names of 16 block names and weak consensus for four names. For anatomical descriptions, strong consensus was achieved for 19 blocks and weak consensus was achieved for one approach. Several areas requiring further research were identified. Conclusions Harmonization and standardization of nomenclature may improve education, research, and ultimately patient care. We present the first international consensus on nomenclature and anatomical descriptions of blocks of the abdominal wall, chest wall, and paraspinal blocks. We recommend using the consensus results in academic and clinical practice.

Background: The Khorana score is a validated tool to identify cancer patients at higher risk of venous thromboembolism (VTE). Objective: We compared its predictive performance to that of the clinical PROTECHT and the polygenic 5-SNP scores in patients who participated in the Dutch CPCT-02 study. Patients/methods: Data on VTE and its risk factors were retrospectively collected for 2729 patients with advanced stage solid tumors planned for systemic cancer treatment. Patients were followed for 6 months. Overall discriminatory performance of the scores was evaluated by time-dependent c-indices. The scores were additionally evaluated dichotomously in competing risk models. Results: A total of 160 (5.9%) patients developed VTE during follow-up. Time-dependent c-indices at 6 months for the Khorana, PROTECHT, and 5-SNP scores were 0.57 (95% confidence interval [CI]: 0.55–0.60), 0.60 (95% CI: 0.57–0.62), and 0.54 (95% CI: 0.51–0.57), respectively. The dichotomous scores classified 9.6%, 16.8%, and 9.5% as high-risk, respectively. VTE risk was about 2-fold higher among high-risk patients than low-risk patients for the Khorana (subdistribution hazard ratio [SHR] 1.9, 95% CI: 1.3–3.0), PROTECHT (SHR 2.1, 95% CI: 1.5–3.0), and 5-SNP scores (SHR 1.7, 95% CI: 1.03–2.8). The sensitivity at 6 months was 16.6% (95% CI: 10.5–22.7), 28.9% (95% CI: 21.5–36.3), and 14.9% (95% CI: 8.5-21.2), respectively. Conclusions: Performance of the PROTECHT or 5-SNP score was not superior to that of the Khorana score. The majority of cancer patients who developed VTE during 6-month follow-up were not identified by these scores. Future directions for studies on cancer-associated VTE prediction may include combined clinical-genetic scores.

Connectivity rate, measured as the exchange of individuals between spawning and nursery grounds, is therefore a crucial determinant of stock size. However, connectivity of early-life stages is hard to explore due to sampling limitations and insufficient knowledge on potential larval sources. Here we present new insights into pre- and post-settlement dispersal of the common sole (Solea solea L.) at a spatial scale of 5–500 km in the Southern North Sea. Patterns at a scale of <100 km were considered local, whereas patterns further than 100 km were considered regional. Multi-elemental signatures of the otolith edge of 213 juvenile sole were used to discriminate at 79% of overall accuracy three main nursery grounds in the Southern North Sea, namely UK coast, Belgian coast and Dutch Wadden Sea. Interregional differences in otolith composition (especially for Mg, Mn and Ba) suggest that sole migration following settlement is limited in the Southern North Sea. Elemental signatures of the same fish indicated mixing during larval dispersal. Each nursery ground recruited an important mix of juveniles from three of the four chemically distinct natal sources identified from the larval otolith signatures. However the percentage of correct regional re-assignment varied from 67 to 80% with a maximum in the Wadden Sea. The results contributed to the validation of biophysical models of larval drift. Our findings support decision making for both fisheries management and marine spatial planning at the national and European level.

Fish otoliths have provided biologists with a treasure-house of valuable life history information across levels of biological organization from individuals to ecosystems. Otoliths have long been used to provide the age and growth estimates which are basic data that underpin scientific fisheries and fisheries ecology. The traditional use of calcified tissues (CS) has been augmented by new applications to address questions of population connectivity, migration, and trophic ecology. These tools have become increasingly important in meeting the demands for spatially explicit and ecosystem-level management tools. Analysis of CS is now extended to applications related to spatial use (essential habitats) and as a record of past and present environmental conditions. This special volume of the ICES Journal of Marine Science contains contributions from the 5th International Otolith Symposium (IOS 2014), an ICES Science Symposium held in October 2014, in Mallorca, Spain. IOS 2014 is a continuation of a series of symposia that started in 1993 to encourage the exchange of information and expertise and to promote the development of new techniques and applications for otolith-based analysis in ecology, management, and conservation. IOS 2014 was the largest meeting to date, with 329 participants from 45 countries. The aim of IOS 2014 was to explore the use of CS as tools to support management and define indicators at environmental, community, population, and individual levels. This international forum is the centre for the exchange of information about developments in the field. New techniques of analysis, statistical treatment, and indeed new areas of application are all presented in this forum. Regular attendees are often the prominent users of otolith analyses in their home institution, and many are active in ICES work. The dissolution of anthropogenically emitted excess carbon dioxide lowers the pH of the world's ocean water. The larvae of mass spawning marine fishes may be particularly vulnerable to such ocean acidification (OA), yet the generality of earlier results is unclear. Here we show the detrimental effects of OA on the development of a commercially important fish species, the Atlantic herring (Clupea harengus). Larvae were reared at three levels of CO2: today (0.0385 kPa), end of next century (0.183 kPa), and a coastal upwelling scenario (0.426 kPa), under near-natural conditions in large outdoor tanks. Exposure to elevated CO2 levels resulted in stunted growth and development, decreased condition, and severe tissue damage in many organs, with the degree of damage increasing with CO2 concentration. This complements earlier studies of OA on Atlantic cod larvae that revealed similar organ damage but at increased growth rates and no effect on condition.

The elemental composition of European hake otoliths was analyzed at the core, settlement mark and otolith edge zones corresponding to the periods of pelagic early life stage, settlement to the bottom, and juvenile life, respectively. Barium, Sr, Mg, Mn and Ca were measured using LA-ICPMS on the sagitta otoliths of hake sampled in 6 different Western Mediterranean areas, separated by tens to hundreds of kilometers. The otolith edge, which should have incorporated the water mass signal of the sampling area, showed a high degree of overlap between geographical areas. This precluded the discrimination of geographically distinct groups. However, there was a clear ontogenetic signal in all the fish, with higher Sr:Ca in the otolith core. Ba:Ca was higher at the settlement mark, while Mn:Ca was overlapped with the core values. The otolith edge had lower ratios for all elements except for Mg. The conflicting effects of the ontogenic patterns and the inconclusive allocation of a geographical signal at the edge, precluded to assign individual hake to a natal origin based on the composition of the core and settlement mark zones. Our results may be explained by high mobility of the juvenile hake in this area of the Mediterranean, or by the relative homogeneity in the water masses for the four elements considered in this study. While this subset is useful for distinguishing major water masses or regional differences between, for example, the Atlantic and the Mediterranean, a larger set of elements could be needed for smaller-scale studies. © 2014 Elsevier B.V.

The growth and development of the aragonitic CaCO3 otoliths of teleost fish could be vulnerable to processes resulting from ocean acidification. The potential effects of an increase in atmospheric CO2 on the calcification of the otoliths were investigated by rearing Atlantic cod Gadus morhua L. larvae in 3 pCO2 concentrations—control (370 µatm), medium (1800 µatm) and high (4200 µatm)—from March to May 2010. Increased otolith growth was observed in 7 to 46 d post hatch (dph) cod larvae at elevated pCO2 concentrations. Efforts to expand the quantity and diversity of fish produced are hampered by the extreme diversity of ontogenetic responses of fish, especially during larval development. This review looks at advances in molecular phylogeny, endocrine and nutrient influences and long-term studies of the phenotypes of commercially important fish to put the sources and consequences of this plasticity into context. This nested context of evolutionary forces of the fish-specific genome duplication, epigenetic influences, ontogenetically conserved processes like metamorphosis and cell determination is further presented in relation to how fish larvae translate the environment into somatic signals, the teleostian diversity of internal processes like sex differentiation and somatogenesis, and the long-term practical consequences of changes in timing or anthropogenic influences. This review aims to present a new baseline of knowledge of marine fish larvae which is useful to scientists, managers and producers.

The broad range of otolith research and applications being carried out in recent years, points to an active field of science with increasingly diverse applications beyond traditional age determination: larval drift and connectivity, population dynamics, fish physiology, population tracking and environmental reconstruction. The main information encoded in the otoliths is based on the growth increments and in the geochemistry tags. The necessity to validate the temporal significanceof observed growth increments (i.e. periodicity of formation) was recognized early in the 1980’s (Beamish & McFarlane 1983). The recognition that ageing errors are important has stimulated interest in the validation of age estimation methods. Validation means proving that a technique is accurate and should be an obligatory step in all sclerochronological studies, and should include the evaluation of both accuracy and precision (Wright et al 2002). Validation is also an imperative in microchemistry studies where methodology is critical to the results, mostly because the heterogeneity of the samples and the low concentrations of many of its constituents. In addition, the supporting matrix of calcium carbonate, which makes up the majority of the mineralized portion of the otolith, causes interference and presents particular problems for analytical determination. With the recent increase in the number of studies based on otolith microchemistry, it is increasingly important that methods are standardized so that results from different laboratories can be compared across distances and years. Validation and standardization should include postmortem contamination of otoliths (i.e. handling and storage methods, cleaning procedures; otolith preparation (i.e. coring and pulverizing; to detection limits and accuracy and precision of the hardware (Geffen et al. 2013). The “Time-Keeping” property of calcified tissues may be their most valuable characteristic, and one that is often taken for granted. The ability to related a measurement of growth increment, chemical composition, or isotopic value to a discreet point in time forms the underlying basis for most of our work. However, it is not always easy to detect or interpret a temporal signal in the growth of calcified tissues. This has led to two parallel lines of investigation - (1) experiments and observations designed to validate the periodicity of accretion, or (2) image analysis and “smart” systems to search for periodicities in visual patterns. But what if there are no patterns? Is it possible that biomineralization can proceed aperiodically? How can we accommodate any “non-patterns” into the studies that rely on calcified tissue analysis? The otoliths of hake (Merluccius merluccius) are a good example of the problem. Over the past four decades, an increasing amount of effort has been channelled into improving the quality of age estimation data that is derived from otolith analysis, to support sustainable management of hake fisheries. Identification of annual increments is difficult, and consensus on interpretation of features can be as low as 20-40%. Recent tagging experiments have shown that hake grow much faster than suspected, and also that many of the increments are not annual and have little link with seasonal variations. They appear to reflect random events or individual movements. Despite intense analysis, it might be that these increments are “non-periodic”, at least at the population level. Nevertheless, there are many sclerochronolgy techniques that do not rely on an absolute timeframe. Some otolith features are linked to life-history stages and can be used as habitat tags. Other questions can be addressed from measurements made of the otolith core or most recent growth. Most importantly, it is time to consider that this is only one example of a more widespread ph

Otolith microchemistry is a powerful tool for fisheries biology, but many applications stretch the limits of analytical techniques. Sources of measurement error were evaluated with a methodological comparison of solution and laser ablation inductively coupled plasma mass spectrometry at two laboratories, using otoliths and certified reference materials NIES-22 and FEBS-1. The intercalibration exercise was effective for Na, Ca, Zn, Cu, Sr, Ba, and Pb. Measurements of Sr were most robust, i.e. sensitivity, accuracy, and precision were least affected by laboratory, technique or sample type. Calcium and Ba measurements varied more between laboratories than between techniques. Lead and Cu measurements were more dependable across laboratory and technique while Zn measurements were more sensitive to these effects. Because the measurement of otolith elements may vary with methodology, it is imperative to consider the sensitivity, accuracy and precision for each application as the suite of elements of interest may differ. The development of reference materials has been the first step in standardization of the procedures, but there is a need for wider ranging calibration and comparison studies. © 2013 Elsevier B.V.

The genomic architecture underlying ecological divergence and ecological speciation with gene flow is still largely unknown for most organisms. One central question is whether divergence is genome‐wide or localized in ‘genomic mosaics’ during early stages when gene flow is still pronounced. Empirical work has so far been limited, and the relative impacts of gene flow and natural selection on genomic patterns have not been fully explored. Here, we use ecotypes of Atlantic cod to investigate genomic patterns of diversity and population differentiation in a natural system characterized by high gene flow and large effective population sizes, properties which theoretically could restrict divergence in local genomic regions. We identify a genomic region of strong population differentiation, extending over approximately 20 cM, between pairs of migratory and stationary ecotypes examined at two different localities. Furthermore, the region is characterized by markedly reduced levels of genetic diversity in migratory ecotype samples. The results highlight the genomic region, or ‘genomic island’, as potentially associated with ecological divergence and suggest the involvement of a selective sweep. Finally, we also confirm earlier findings of localized genomic differentiation in three other linkage groups associated with divergence among eastern Atlantic populations. Thus, although the underlying mechanisms are still unknown, the results suggest that ‘genomic mosaics’ of differentiation may even be found under high levels of gene flow and that marine fishes may provide insightful model systems for studying and identifying initial targets of selection during ecological divergence.

In response, international regulations have been imposed and many fisheries have been 'eco-certified' by consumer organizations, but methods for independent control of catch certificates and eco-labels are urgently needed. Here we show that, by using gene-associated single nucleotide polymorphisms, individual marine fish can be assigned back to population of origin with unprecedented high levels of precision. By applying high differentiation single nucleotide polymorphism assays, in four commercial marine fish, on a pan-European scale, we find 93-100% of individuals could be correctly assigned to origin in policy-driven case studies. We show how case-targeted single nucleotide polymorphism assays can be created and forensically validated, using a centrally maintained and publicly available database. Our results demonstrate how application of gene-associated markers will likely revolutionize origin assignment and become highly valuable tools for fighting illegal fishing and mislabelling worldwide.

Ocean acidification, caused by increasing atmospheric concentrations of CO2 (refs 1, 2, 3), is one of the most critical anthropogenicthreats to marine life. Changes in seawater carbonate chemistry have the potential to disturb calcification, acid–base regulation, blood circulation and respiration, as well as the nervous system of marine organisms, leading to long-term effects such as reduced growth rates and reproduction4, 5. In teleost fishes, early life-history stages are particularly vulnerable as they lack specialized internal pH regulatory mechanisms6, 7. So far, impacts of relevant CO2 concentrations on larval fish have been found in behaviour8, 9 and otolith size10, 11, mainly in tropical, non-commercial species. Here we show detrimental effects of ocean acidification on the development of a mass-spawning fish species of high commercial importance. We reared Atlantic cod larvae at three levels of CO2, (1) present day, (2) end of next century and (3) an extreme, coastal upwelling scenario, in a long-term ( months) mesocosm experiment. Exposure to CO2 resulted in severe to lethal tissue damage in many internal organs, with the degree of damage increasing with CO2 concentration. As larval survival is the bottleneck to recruitment, ocean acidification has the potential to act as an additional source of natural mortality, affecting populations of already exploited fish stocks.
