Fasciolosis is a severe zoonosis responsible for major economic losses in livestock. The enhanced MM3-COPRO test (eMM3-COPRO) and the commercial version BIO K 201 (Bio-X Diagnostics, Rochefort, Belgium) are widely used as immunodiagnostic tools for the specific detection of coproantigens released by Fasciola during the late prepatent and patent stages of infection. However, performance of the eMM3-COPRO has never been evaluated under field conditions. To address this gap, a large number of ovine faecal samples, collected in a region where fasciolosis is endemic (Galicia, NW Spain), were analyzed. Two groups of sheep flocks were selected according to the Fasciola infection status: ‘Fasciola-free’ and ‘Fasciola-infected’ flocks. ‘Fasciola-free’ flocks were seronegative flocks with no history of fasciolosis detected by either coproscopy or necropsy in the last 5 years. Faecal samples from these sheep were used to calculate a cut-off value for infection (OD = 0.021). The cut-off was calculated using a bootstrap resampling method that enables estimation of the sampling distribution of the statistical parameters without making assumptions about the underlying data distribution. ‘Fasciola-infected’ flocks were characterized by high seroprevalence, a history of fasciolosis and periodical treatment with flukicides. Samples from these flocks were used to estimate the diagnostic accuracy of the eMM3-COPRO relative to coproscopy, which although limited by poor sensitivity is the only reference test available for diagnosing fasciolosis in vivo. To overcome this limitation, all animals classified positive by eMM3-COPRO were treated with triclabendazole and then retested. The eMM3-COPRO displayed higher sensitivity than coproscopy, as it detected coproantigens in all samples with positive coproscopy and in 12% of samples with negative coproscopy. Trematodes are widespread parasitic flatworms that significantly affect mankind either directly as human parasites, or indirectly via the infection of livestock and the related economic damage. The two most important trematode taxa are the blood flukes Schistosoma and the liver flukes Fasciola, but detection and differentiation of these parasites remains a challenge. Recently, microRNAs (miRNAs) were described from extracellular vesicles (EV) for both parasites secreted into respective hosts. These molecules have been proposed as mediators of parasite-host communication, and potential biomarkers for the detection of parasitic infections from host blood. Our aim here was to study similarities and differences in the miRNA complements of Schistosoma mansoni and Fasciola hepatica, EV-load in particular, to predict their targets and potential functions in the parasite-host interaction. We reanalyzed the known miRNA complements of S. mansoni and F. hepatica and found 16 and 4 previously overlooked, but deeply conserved miRNAs, respectively, further moving their complements closer together. We found distinct miRNA enrichment patterns in EVs both showing high levels of flatworm miRNAs with potential for the detection of an infection from blood. Two miRNAs of the protostome specific MIR-71 and MIR-277 families were highly expressed in EVs and could, therefore, have potential as biomarkers for trematode infection. Curiously, we identified nucleotide differences in the sequence of Mir-277-P2 between S. mansoni and F. hepatica that hold great promise for the distinction of both parasites. To test whether the EV-miRNAs of S. mansoni and F. hepatica could be modulating the expression of host genes, we predicted miRNA targets in 321 human and cattle messenger RNAs that overlapped between both hosts. El presente trabajo de investigación se efectuó durante los meses de septiembre y Diciembre del 2019, con el objetivo de conocer la prevalencia de fasciolosis en Ganado Vacuno Holstein en el Distrito de Vitor, Anexo Sotillo, Arequipa. La muestra empleada fue de 155, seleccionadas al azar, de la raza Hostein en todas las clases animales y de ambos sexos, para la realización de este trabajo se tomó las muestras de heces que se obtuvieron de la ampolla rectal, con guantes obstétricos y puestos en bolsas de polipropileno y agrupadas e identificadas con los datos del hato y fueron llevadas al laboratorio de análisis biológicos veterinarios y agrícolas majes el pedregal Los resultados que se obtuvieron fueron los siguientes: La prevalencia de fasciolosis en el ganado Vacuno Holstein es de 14.84%; la prevalencia según la clase animal representa, Toros 16.67%, Vacas 14.44%, Toretes 11.77%, Vaquillas 14.29%, Terneros 16.67%, Terneras 20.00%; Al aplicar la prueba estadística de Chi - Cuadrado, encontramos que no existe diferencia significativa entre las variables respecto a clase y sexo, demostrando que existe igual susceptibilidad. Introducción: La fascioliasis es un parasitismo producido por dos trematodos del género Fasciola, Fasciola hepatica y Fasciola gigantica. Este parasitismo tiene una especial relevancia en países como Asia, África y América del Sur, sin embargo, se han notificado casos en todos los continentes. Objetivos: Revisión del tratamiento del parasitismo y comparación entre los distintos métodos diagnósticos. Metodología: Se ha recurrido a diversas fuentes de información como libros, tesis doctorales, bases de datos, revistas y recursos electrónicos. Resultados y discusión: Triclabendazol es el tratamiento de elección del parasitismo, aunque debido a su uso masivo se ha notificado la aparición de resistencia. Por ello, en la actualidad se están buscando alternativas terapéuticas, como son Closantel y Oxiclozanida entre otros. En el diagnóstico contamos con métodos parasitológicos, inmunológicos y genético-moleculares. Los métodos parasitológicos resultan muy simples y baratos; no obstante, son laboriosos y su sensibilidad es especialmente baja, lo que obliga a tener que repetir numerosas veces las pruebas para poder obtener resultados fiables. Los métodos inmunológicos se basan en el uso de la técnica de Enzimoinmunoanálisis de Adsorción (ELISA) y estos suponen un aumento de la sensibilidad respecto a los anteriores. Existen dos tipos: ELISA para la detección de anticuerpos, que se considera la técnica más sensible, y ELISA para la detección de antígenos, que se consideran técnicas muy específicas. Los métodos genético-moleculares son los más recientes. Hay que destacar principalmente dos técnicas: Reacción en Cadena de la Polimerasa (PCR) y Amplificación Isotérmica Mediada por Bucle (LAMP). Ambas son técnicas de amplificación de material genético y presentan buenos resultados de sensibilidad, aunque, como en el caso de los métodos inmunológicos, presentan el inconveniente de la necesidad de laboratorios con instrumentación especializada. We report a European wide assessment of the economic burden of gastrointestinal nematodes, Fasciola hepatica (common liver fluke) and Dictyocaulus viviparus (bovine lungworm) infections to the ruminant livestock industry. The economic impact of these parasitic helminth infections was estimated by a deterministic spreadsheet model as a function of the proportion of the ruminant population exposed to grazing, the infection frequency and intensity, the effect of the infection on animal productivity and mortality and anthelmintic treatment costs. In addition, we estimated the costs of anthelmintic resistant nematode infections and collected information on public research budgets addressing helminth infections in ruminant livestock. The epidemiologic and economic input data were collected from international databases and via expert opinion of the Working Group members of the European Co-operation in Science and Technology (COST) action COMbatting Anthelmintic Resistance in ruminants (COMBAR). In order to reflect the effects of uncertainty in the input data, low and high cost estimates were obtained by varying uncertain input data arbitrarily in both directions by 20 %. The combined annual cost [low estimate-high estimate] of the three helminth infections in 18 participating countries was estimated at € 1.8 billion [€ 1.0–2.7 billion]. Eighty-one percent of this cost was due to lost production and 19 % was attributed to treatment costs. The cost of gastrointestinal nematode infections with resistance against macrocyclic lactones was estimated to be € 38 million [€ 11–87 million] annually. The annual estimated costs of helminth infections per sector were € 941 million [€ 488 – 1442 million] in dairy cattle, € 423 million [€ 205–663 million] in beef cattle, € 151million [€ 90–213 million] in dairy sheep, € 206 million [€ 132–248 million] in meat sheep and € 86 million [€ 67–107 million] in dairy goats. La fasciolosis es una trematodosis de gran relevancia en medicina humana y veterinaria. Los resultados de la presente tesis ponen de manifiesto la importancia de la selección adecuada de adyuvantes para obtener respuestas de anticuerpos frente a epitopos ocultos y sientan las bases para un diseño racional de antígenos quiméricos destinados a la vacunación de distintas especies. De manera complementaria, se evalúa una nueva metodología para mejorar la especificidad los métodos ELISA que emplean antígenos recombinantes expresados en E. coli.

Unfortunately, this important meat source is being infected by Fasciola, leading to increased production loss in cattle and human infection. This study therefore set out to determine the prevalence and intensity of Fasciola infection in cattle slaughtered at Bamenda abattoir. A total of 733 cattle(593 males and 140 females) brought from all seven divisions of the North West Region were sampled from January to June,2016. Faecal samples were collected and analyzed at the Regional Veterinary Laboratory by the formol ether concentration technique. Intensity was measured as eggs per gram. The overall prevalence of Fasciola infection was 20% (147/733). A higher prevalence of Fasciola infection 24.2% (90/371) was recorded during the rainy season compared with the dry season 15.7% (57/362). The prevalence of the infection was highest in cattle from Ngoketungia 37.5%(3/8) and lowest in those from Mezam Division 11.8%(24/203). Females had a higher prevalence of Fasciola infection 22.1%(31/140) than males 19.6(116/593). Intensity of Fasciola infection was highest in cattle from Bui Division (31.3eggs per gramme, EPG and lowest in Ngoketunjia (2.2 EPG). It was thus concluded that Fasciola infection was prevalent in cattle brought for slaughter at Bamenda abattoir with a higher prevalence in females than and greater occurrence in rainy season than dry season.

We report a case of a 39 year old male who presented with nausea and right upper quadrant pain. Marked eosinophilia and a hypoechoic liver lesion on ultrasound were identified. The differential diagnosis included neoplasms, infectious diseases and hepatic abscess. Indirect hemagglutination test using purified adult Fasciola hepatica f1Ag confirmed serologic diagnosis of fascioliasis. Radiologists should keep in mind the importance of correlating imaging, clinical and laboratory findings in order to reach the correct diagnosis.

Ante la escasa y poco actualizada información acerca de las trematodosis que afectan al ganado bovino en el Departamento de Salto (Uruguay), eminentemente agrícola, en el primer ensayo se recogieron muestras de sangre de 386 vacas de aptitud cárnica, láctea y mixta, pertenecientes a 13 explotaciones. Los sueros se analizaron con un ELISA y antígenos de excreción/secreción de adultos de F. hepatica, demostrándose una seropositividad individual del 67% (IC95% 62, 72), con los porcentajes más elevados en ganado de raza Angus y en los terneros menores de 2 años. Estos resultados coinciden con los obtenidos en otras regiones como el Noroeste de España (PAZ-SILVA y cols., 2007), Gales (McCANN y cols., 2010), Australia (MOLLOY y cols., 2005), Puerto Rico (HILLYER y cols., 1996) o el norte de Egipto (EL-AZIZ y cols., 2001). El análisis simultáneo demostró que la raza, y a continuación la edad son los factores más influyentes en el aumento de la seroprevalencia. Las diferencias observadas en función de la raza parecen atribuibles al manejo de los animales. La fasciolosis en los hospedadores definitivos se produce por la ingestión de metacercarias adheridas al forraje o vehiculadas por el agua de bebida. Fasciola hepatica es un digeneo que tiene un ciclo biológico complejo, que incluye un hospedador vertebrado en el que acontece su reproducción sexual, verificable en animales de experimentación, por lo que el objetivo del presente estudio fue estimar la potencia invasiva (PI), extensión de la invasión (EI) y el potencial biótico de este digeneo, en Rattus norvegicus albinus de la cepa Holtzman experimentalmente infectadas por F. hepatica, Para ello se obtuvieron metacercarias de caracoles infectados en la naturaleza de dos procedencias, las que fueron administradas en número de veinte en la dieta semisólida a seis ejemplares de R. norvegicus, mantenidas en bioterio bajo condiciones ambientales controladas. Realizándoles su coproparasitoscopía mediante la técnica de concentración simple por sedimentación desde los 86 hasta los 100 días post infección, finalmente procediendo con las necropsias. We made coproparasitological analysis by simple concentration and sedimentation technique from the 86 to 100 days post infection and then proceeded with necropsies. We observed that the average production of eggs by adult F. hepatica was 9135.70 ± 2196.8, invasive power from 6.67 to 8.33% and the extent of invasion of 66.67% in R. norvegicus.

A total of 598 buffaloes were sampled for both coprological (210) and haematological (388) investigations at the Large Animal Clinics, GADVASU, Ludhiana, Punjab, India. Coprological examination revealed that the overall prevalence of gastrointestinal (GI) parasitic infections was 23.33% (49/210). Among the revealed parasites, amphistomes, Fasciola spp., Eimeria spp., Balantidium coli and strongyles were in 4.29, 3.33, 0.95, 2.86 and 15.71% of the examined buffaloes. Except coccidiosis, there was no significant variation of GI infections in relation to sex. Eimeria spp. was significantly higher in males. The present work emphasized that strongyles were the most prevalent gastrointestinal parasites found during coprological examination of buffaloes in Punjab, India. Examination of Giemsa-stained peripheral blood smears exhibited that 4.9% (19/388) of buffaloes were infected with haemoparasites comprising Theileria annulata (2.32%), Trypanosoma evansi (1.8%), Babesia bigemina (0.26%) and Anaplasma marginale (0.77%). Mixed infection appeared in one (0.26%) animal. Trypanosomosis was predominant in elder animals with no infection recorded in males.

Fasciola's excretory-secretory material comprises chiefly cathepsin B and cathepsin L. These cysteine proteases are proposed as major mediators of parasitism, and are considered targets for vaccination. In order to assess the vaccine efficacy of these enzymes, single and multivalent recombinant protein vaccinations of adult-stage F. hepatica cathepsin L5, metacercarial-stage F. gigantica cathepsin L1 g and juvenile-stage F. hepatica cathepsin B were analysed in rats against F. hepatica challenge infection. The protective efficacy of anti-fluke vaccines was evaluated in terms of parasitological parameters (recovered fluke burden, fluke body size and wet weight) and pathological changes (liver damage score) in rats. The rats vaccinated with recombinant proteins were shown to have significantly fewer and smaller flukes than the control rats. A maximum protection of 83% was seen in the group vaccinated with a combination of cathepsin B and cathepsin L5.

Liver flukes produce cathepsin B and cathepsin L in their excretory-secretory material. Cathepsin B is predominately released in the juvenile stage of the life-cycle, while different cathepsin L's are released throughout the cycle. Phylogenetic analysis of the mature proteins shows that cathepsin L's and B form distinct clades, as expected. Cathepsin L proteases also form clades that reflect their predicted enzymatic activity, and their temporal expression. We have expressed examples of both these cathepsins in yeast, and purified them for both enzymatic analysis and for use as vaccines. Recently, a vaccination experiment was performed combining cathepsins B and L as immunogens, demonstrating that a multivalent vaccine may be optimal.

Traditional vaccines have many characteristics which pose logistical problems for both military and civilian vaccinations programs. Currently, vaccines require several booster injections to obtain maximum protection, they are often effective against only a single organism and most require refrigeration. Biodegradable microspheres for vaccine encapsulation could eliminate many of these problems. Microencapsulated vaccines offer the promise of extended antigen presentation, thus eliminating the need for booster injections. Microencapsulation also allows incorporation of multiple antigens in a single dose. This report describes the production and in vivo evaluation of a new microsphere vaccine approach. Several microsphere tetanus and botulinum vaccine formulations were examined in vivo using a BALB/c mouse model. Several formulations of microencapsulated vaccines provided both an elevated and prolonged serum IgG response. At 9 weeks post injection, the microsphere vaccine evidenced 47% of its initial (2 week) serum IgG levels, without a booster injection. This study has demonstrated that this biodegradable microsphere antigen delivery system has the potential for prolonged protection. The vaccine delivery system described in this report, if successfully developed, will benefit military and civilian health efforts and could be marketed around the world.

The literature on the identification of Fasciola spp. and their intermediate hosts, the general life cycle of Fasciola and aspects of the epidemiology, clinical signs, diagnosis and treatment of infections is reviewed. Two experiments were carried out. The first involved 18 weeks observations on 5 adult male goats each infected with 150 metacercartae of F.hepatica and 5 uninfected controls. The second involved groups of 10 sheep and 10 goats each infected with 200 metacercariae with 5 uninfected controls of each species. In both experiments, faecal, haematological, biochemical and pathological examinations were conducted. The animals were also weighed regularly. In the first experiment, although only 15-35 flukes were established, measurable and, in many cases, statistically significant changes in a variety of parameters were observed. A depression in packed cell volumes relative to controls of approximately 20% occurred. Though haemoglobin, mean corpuscular volume and mean corpuscular haemoglobin concentration levels remained within the normal ranges, erythrocyte levels in the infected group were significantly lower than in the controls and there was a tendency for the anaemia to become macrocytic. This suggests that goats may be particularly susceptible to the effects of blood loss associated with Fasciola infections though further work is needed to confirm this. A marked peripheral eosinophilia and elevation in fibrinogen levels were observed in infected animals. Albumin levels decreased, globulin levels increased and the A/G ratio decreased significantly relative to the control group but all levels remained within the normal ranges. In infected animals, gamma glutamyl transferase (GGT) and glutamyl dehydrogenase (GD) levels rose to beyond the limits of the normal ranges although aspartate aminotransferase (AST) levels, which were also significantly elevated, did not.
