Triplet dynamics in singlet fission depend strongly on the strength of the electronic coupling. Covalent systems in solution offer precise control over such couplings. Nonetheless, efficient free triplet generation remains elusive in most systems, as the intermediate triplet pair 1(T1T1) is prone to triplet-triplet annihilation due to its spatial confinement. In the solid state, entropically driven triplet diffusion assists in the spatial separation of triplets, resulting in higher yields of free triplets. Control over electronic coupling in the solid state is, however, challenging given its sensitivity to molecular packing. We have thus developed a hexameric system (HexPnc) to enable solid-state-like triplet diffusion at the molecular scale. This system is realized by covalently tethering three pentacene dimers to a central subphthalocyanine scaffold. Transient absorption spectroscopy, complemented by theoretical structural optimizations and steady-state spectroscopy, reveals that triplet diffusion is indeed facilitated due to intramolecular cluster formation. The yield of free triplets in HexPnc is increased by a factor of up to 14 compared to the corresponding dimeric reference (DiPnc). Thus, HexPnc establishes crucial design aspects for achieving efficient triplet dissociation in strongly coupled systems by providing avenues for diffusive separation of 1(T1T1), while, concomitantly, retaining strong interchromophore coupling which preserves rapid formation of 1(T1T1).

ABSTRACT In a recent editorial, the Editors-in-Chief of Journal of Experimental Biology argued that consensus building, data sharing, and better integration across disciplines are needed to address the urgent scientific challenges posed by climate change. We agree and expand on the importance of cross-disciplinary integration and transparency to improve consensus building and advance climate change research in experimental biology. We investigated reproducible research practices in experimental biology through a review of open data and analysis code associated with empirical studies on three debated paradigms and for unrelated studies published in leading journals in comparative physiology and behavioural ecology over the last 10 years. Nineteen per cent of studies on the three paradigms had open data, and 3.2% had open code. Similarly, 12.1% of studies in the journals we examined had open data, and 3.1% had open code. Previous research indicates that only 50% of shared datasets are complete and re-usable, suggesting that fewer than 10% of studies in experimental biology have usable open data. Encouragingly, our results indicate that reproducible research practices are increasing over time, with data sharing rates in some journals reaching 75% in recent years. Rigorous empirical research in experimental biology is key to understanding the mechanisms by which climate change affects organisms, and ultimately promotes evidence-based conservation policy and practice. We argue that a greater adoption of open science practices, with a particular focus on FAIR (Findable, Accessible, Interoperable, Re-usable) data and code, represents a much-needed paradigm shift towards improved transparency, cross-disciplinary integration, and consensus building to maximize the contributions of experimental biologists in addressing the impacts of environmental change on living organisms.

Ocean acidification—decreasing oceanic pH resulting from the uptake of excess atmospheric CO2—has the potential to affect marine life in the future. Among the possible consequences, a series of studies on coral reef fish suggested that the direct effects of acidification on fish behavior may be extreme and have broad ecological ramifications. Recent studies documenting a lack of effect of experimental ocean acidification on fish behavior, however, call this prediction into question. Indeed, the phenomenon of decreasing effect sizes over time is not uncommon and is typically referred to as the “decline effect.” Here, we explore the consistency and robustness of scientific evidence over the past decade regarding direct effects of ocean acidification on fish behavior. Using a systematic review and meta-analysis of 91 studies empirically testing effects of ocean acidification on fish behavior, we provide quantitative evidence that the research to date on this topic is characterized by a decline effect, where large effects in initial studies have all but disappeared in subsequent studies over a decade. The decline effect in this field cannot be explained by 3 likely biological explanations, including increasing proportions of studies examining (1) cold-water species; (2) nonolfactory-associated behaviors; and (3) nonlarval life stages. Furthermore, the vast majority of studies with large effect sizes in this field tend to be characterized by low sample sizes, yet are published in high-impact journals and have a disproportionate influence on the field in terms of citations. We contend that ocean acidification has a negligible direct impact on fish behavior, and we advocate for improved approaches to minimize the potential for a decline effect in future avenues of research.

Farmed aquatic animals represent an increasingly important source of food for a growing human population. However, the aquaculture industry faces several challenges with regard to producing a profitable, ethical and environmentally sustainable product, which are exacerbated by the ongoing intensification of operations and increasingly extreme and unpredictable climate conditions. Fortunately, bio-sensors capable of measuring a range of environmental, behavioural and physiological variables (e.g. temperature, dissolved gases, depth, acceleration, ventilation, heart rate, blood flow, glucose and l-lactic acid) represent exciting and innovative tools for assessing the health and welfare of farmed animals in aquaculture. Here, we illustrate how these state-of-the-art technologies can provide unique insights into variables pertaining to the inner workings of the animal to elucidate animal–environment interactions throughout the production cycle, as well as to provide insights on how farmed animals perceive and respond to environmental and anthropogenic perturbations. Using examples based on current challenges (i.e. sub-optimal feeding strategies, sub-optimal animal welfare and environmental changes), we discuss how bio-sensors can contribute towards optimizing the growth, health and welfare of farmed animals under dynamically changing on-farm conditions. While bio-sensors currently represent tools that are primarily used for research, the continuing development and refinement of these technologies may eventually allow farmers to use real-time environmental and physiological data from their stock as ‘early warning systems' and/or for refining day-to-day operations to ethically and sustainably optimize production. Abstract Many ectotherms have shown a reduction in maximum body size in the past decades in parallel with climate warming. Indeed, some models forecast a maximum body size decline of 14%–24% by 2050 for numerous fish species. The gill-oxygen limitation (GOL) hypothesis is perhaps the most prominent concept regarding the physiological mechanisms underlying the observed trends, implicating oxygen uptake limitations in driving the decline in fish body size with warming. Current scientific debates, however, demonstrate a clear need for a synthesis of existing empirical evidence to test the fundamental assumptions of the GOL hypothesis. Here, we perform a systematic literature review of the intraspecific allometry of gill surface area (GSA) and metabolic rate. Additionally, we introduce a new parameter, the ratio S, which provides a measure of GSA in relation to the metabolic requirements for maintenance (SSMR) and maximum activity (SAMR). Support for the GOL hypothesis would be evidenced by a universal decline in S with increasing body mass within each species, such that gills become less equipped to supply metabolic requirements as fish grow. In contrast to the predictions of the GOL hypothesis, we show that the scaling exponents for SSMR and SAMR are consistently close to zero, with only a few exceptions where S either increased or decreased. These findings suggest that the GSA of each species is sufficient to meet its oxygen requirements throughout life, and that growth is not universally restricted by oxygen uptake limitations across the gills. We identify the need to investigate hypotheses other than the GOL hypothesis to help explain the observed declines in maximum fish body sizes concurrent with climate warming, in order to facilitate accurate predictions of fish community structure and manage fisheries in the face of climate change.

Relevant and robust scientific knowledge is critical for providing decision-makers with the actionable evidence needed to inform conservation decisions. In the Anthropocene, science that leads to meaningful improvements in biodiversity conservation, restoration and management is desperately needed. Conservation Physiology has emerged as a discipline that is well-positioned to identify the mechanisms underpinning population declines, predict responses to environmental change and test different in situ and ex situ conservation interventions for diverse taxa and ecosystems. Here we present a consensus list of 10 priority research themes. Within each theme we identify specific research questions (100 in total), answers to which will address conservation problems and should improve the management of biological resources. The themes frame a set of research questions related to the following: (i) adaptation and phenotypic plasticity; (ii) human–induced environmental change; (iii) human–wildlife interactions; (iv) invasive species; (v) methods, biomarkers and monitoring; (vi) policy, engagement and communication; (vii) pollution; (viii) restoration actions; (ix) threatened species; and (x) urban systems. The themes and questions will hopefully guide and inspire researchers while also helping to demonstrate to practitioners and policy makers the many ways in which physiology can help to support their decisions.

Indirect effects of predators can manifest themselves as changes in prey behaviour and physiology. Given that digestion requires energy, it has been suggested that prey will choose to eat smaller meals under predation risk to reserve a larger portion of the aerobic metabolic scope they have available for energetically demanding tasks more critical than digestion, such as escape. To test this prediction, we quantified food consumption and growth of juvenile corkwing wrasses (Symphodus melops) over 11 days in the presence or absence of a predator (Atlantic cod, Gadus morhua). We then quantified behaviour and food consumption of the same wrasses in behavioural arenas with a predator. All food consumption was examined in the context of the aerobic scope that would have been available during the digestive period. Overall, there was no effect of predator exposure on food consumption or growth, yet predator-exposed wrasses were more consistent in their daily food consumption, lending some support to our prediction of prey bet-hedging on meal size under predation risk. The lack of a clear pattern may have resulted from a relatively low percentage of aerobic scope (similar to 20-27%) being occupied by digestion, such that fish retained ample capacity for activities other than digestion. In the subsequent behavioural trials, predator-exposed wrasses were more active and spent more time near the cod than predator-naive wrasses, suggesting the former had habituated to predation threat and were more risk-taking. Our results highlight the complex and often counter-intuitive effects that predator presence can have on prey populations beyond direct consumption. 1. Temperature has a dramatic effect on the physiology of ectothermic animals, impacting most of their biology. When temperatures increase above optimal for an animal, their growth gradually decreases. The main mechanism behind this growth rate reduction is unknown. 2. Here, we suggest the ‘aerobic scope protection’ hypothesis as a mechanistic explanation for the reduction in growth. 3. After a meal, metabolic rate, and hence oxygen consumption rate, transiently increases in a process called specific dynamic action (SDA). At warmer temperatures, the SDA response usually becomes temporally compressed, leading to a higher peak oxygen consumption rate. This peak in oxygen consumption rate risks taking up much of the animal’s aerobic scope (the difference between resting and maximum rates of oxygen consumption), which would leave little residual aerobic scope for other aerobic functions. 4. We propose that water‐breathing ectothermic animals will protect their postprandial residual aerobic scope by reducing meal sizes in order to regulate the peak SDA response during times of warming, leading to reductions in growth. 5. This hypothesis is consistent with the published literature on fishes, and we suggest predictions to test it.

The partial pressure of CO2 in the oceans has increased rapidly over the past century, driving ocean acidification and raising concern for the stability of marine ecosystems1,2,3. Coral reef fishes are predicted to be especially susceptible to end-of-century ocean acidification on the basis of several high-profile papers4,5 that have reported profound behavioural and sensory impairments—for example, complete attraction to the chemical cues of predators under conditions of ocean acidification. Here, we comprehensively and transparently show that—in contrast to previous studies—end-of-century ocean acidification levels have negligible effects on important behaviours of coral reef fishes, such as the avoidance of chemical cues from predators, fish activity levels and behavioural lateralization (left–right turning preference). Using data simulations, we additionally show that the large effect sizes and small within-group variances that have been reported in several previous studies are highly improbable. Together, our findings indicate that the reported effects of ocean acidification on the behaviour of coral reef fishes are not reproducible, suggesting that behavioural perturbations will not be a major consequence for coral reef fishes in high CO2 oceans.

Published by Wiley-VCH GmbH Three diketopyrrolopyrrole (DPP) dimers, linked via different dithienylphenylene spacers, ortho-DPP (o-DPP), meta-DPP (m-DPP), and para-DPP (p-DPP), are synthesized, characterized, and probed in light of intramolecular singlet fission (i-SF). Importantly, the corresponding DPP reference (DPP-Ref) singlet and triplet excited state energies of 2.22 and 1.04 eV, respectively, suggest that i-SF is thermodynamically feasible. The investigations focus on the impact of the relative positioning of the DPPs, and give compelling evidence that solvent polarity and/or spatial overlap govern i-SF dynamics and efficiencies. Polar solvents make the involvement of an intermediate charge transfer (CT) state possible, followed by the population of 1(T1T1) and subsequently (T1 + T1), while spatial overlap drives the mutual interactions between the DPPs. In o-DPP, the correct balance between polar solvents and spatial overlap leads to the highest triplet quantum yield (TQY) of 40%. Notable is the superimposition of CT and triplet excited states, preventing an accurate TQY determination. For m-DPP, poorer spatial overlap correlates with weaker CT character and manifests in a TQY of 11%. Strong CT character acts as a trap and prevents i-SF, as found with p-DPP. The DPP separation is decisive, enabling a symmetry-breaking charge-separated state rather than CT formation, shutting down the formation 1(T1T1).

Triplet-excited-state energies of perylene-monoimides (PMIs) lie in the range 1.12 eV ± 2 meV when compared to singlet-excited-state energies of about 2.39 eV ± 2 meV; therefore, the corresponding naphthalene-linked PMI-Dimer was investigated as a novel singlet-fission (SF) material. Ultrafast transient absorption measurements demonstrated the (S1S0)-to-1(T1T1) transformation and the involvement of a mediating step in the overall 1(T1T1) formation. The intermediate is a charge-transfer state that links the initial (S1S0) with the final 1(T1T1), and imposes charge-transfer character on both, which are thus denoted (S1S0)CT and 1(T1T1)CT. At room temperature, the decorrelation and stability of 1(T1T1)CT is affected by the geminate triplet-triplet recombination (G-TTR) of the two triplets. Independent confirmation for G-TTR to afford up-converted (S1S0)UC in fsTA and nsTA measurements with PMI-Dimer, came from probing PMI-Monomer (T1)s in triplet-triplet annihilation up-conversion (TTA-UC). The G-TTR channel, active in the PMI-Dimer at room temperature, is suppressed by working at either low temperatures (∼140 K) or in polar solvents (benzonitrile): Both scenarios assist in stabilizing (T1T1)CT. As a consequence, the triplet quantum yields are 4.2% and 14.9% at room temperature and 140 K, respectively, in 2-methyltetrahydrofuran.

Convergent and convenient regioselective synthesis of novel thiazolo[2,3-a]pyrimidine derivatives was accomplished using the one-pot reaction of 6-ethylthiouracil, bromoacetic acid, anhydrous sodium acetate, acetic anhydride, acetic acid and suitable aldehyde. X-ray crystallographic study reveals the presence of the Z configuration of only one regioisomer confirmed by computational studies as being the most likely isomer present.
