Abstract

The coalescence of a drop into a fluid bath is a subject of continuing interest in fluid dynamics, owing to its dynamical complexity, its accessibility in the laboratory, and its aesthetic appeal. It has been known for some time that a drop gently placed on a static bath of the same fluid may initially combine only partially with the underlying bath, resulting in a daughter droplet of approximately half the diameter of the parent droplet. This process can then repeat, giving rise to the so-called “coalescence cascade” until the final droplet is small enough to be completely absorbed., National Science Foundation (U.S.) (Grant CMMI-1333242), MIT-France Program

3. Hydrodynamics of new quantum analogs

Abstract

This report summarizes the results obtained during a four month-long internship at the Fluid Dynamics Laboratory at MIT. The work presented here is a result of the joint effort of Victor Prost and myself. During these four months we had the opportunity of working on diverse topics, from developing a high-precision experiment to creating stunning videos of bouncing drops with a high-speed video camera. This variety has allowed us to get a wide perspective on the daily life of a researcher. We present here the main scientific results that we obtained in this period. We consecrated the first part of this period to fluid visualization. Two main experiments were developed; these are presented and described hereafter. An important part of our time was dedicated to the Dynamics of a forced Soap Film. The results and strategies we carried on to understand the underlying mechanics are presented here. Towards the end of our research period I had the opportunity of concentrating my efforts in analyzing a new analog of the Casimir effect, whereas Victor developed a more refined modeling of the Soap Film through CFD. An annex includes an user-friendly Bouncing Droplets Demonstrator we developed to show the richness of the Hydrodynamic Quantum Analogs field.

4. Etude de la propulsion de l'aeromodelisme

Abstract

Étant passionnés d’aéronautique, nous nous sommes intéressés à la reproduction d’un problème de cette discipline en échelle réduite pour ce Module Appliqué en Laboratoire. De nos jours, l’un des principaux problèmes de la conception des avions conventionnels est la contrainte du diamètre des turbines, qui est directement corrélé au flux de quantité de mouvement et donc à la poussée. Dans le but d’optimiser le design des aéronefs, les ingénieurs ont trouvé diverses méthodes pour maximiser les dimensions de la turbine compte tenu des contraintes mécaniques de l’avion, notamment en rapprochant les turbines de l’aile, ce qui peut présenter éventuellement des contraintes thermiques dans la structure de celle-ci. Nous nous intéressons dans la présente étude aux effets de la position du moteur sur les valeurs dynamiques de l’avion. Pour cela nous avons développé un modèle d’aile et une turbine modélisées avec un moteur d’aéromodélisme, qui nous ont permis de réaliser un modèle d’aile en deux dimensions visant à caractériser l’effet sur la portance et la traînée de la position du moteur, celui-ci pouvant être à la fois placé en dessus et en dessous de l’aile. Cette idée principale nous a permis de faire une étude élargie de l’écoulement du jet produit par un moteur d’aéromodélisme, qui présente des caractéristiques particulières étant donné son système de génération de la poussée par rapport à un jet classique., Outgoing

7. Phage lytic enzymes as therapy for antibiotic-resistant Streptococcus pneumoniae infection in a murine sepsis model

Abstract

Objectives: Phage-coded lysins, i.e. murein hydrolases, are enzymes that destroy the cell wall of bacteria. A rapid killing of Streptococcus pneumoniae in the nasopharynx of mice has been described recently using a phage-coded murein hydrolase (enzybiotic). The in vivo effects of a dose-ranging treatment, using either of the phage-coded lytic enzymes Cpl-1 lysozyme or the Pal amidase, have been investigated here in a murine sepsis model., Methods: Purified Pal amidase and/or Cpl-1 lysozyme were used alone or in combination. These enzymes were injected intraperitoneally at different times after challenge with 5 × 107 cfu of a type 6B, antibiotic-resistant S. pneumoniae clinical isolate., Results: Animals challenged with 5 × 107 cfu of this strain alone died within 72 h, whereas a single intraperitoneal injection of Cpl-1 or Pal (200 µg; 1100 U) administered 1 h after the bacterial challenge was sufficient to effectively protect the mice, according to unpaired t-test (P < 0.0001). Bacteraemia in unprotected mice reached colony counts >107 cfu/mL, whereas the mean colony count in lysin-protected animals was <106 cfu/mL over time and ultimately became undetectable. Interestingly, a synergic effect in vivo was observed with the combined use of 2.5 µg each of Cpl-1 and Pal., Conclusions: Our findings suggest strongly that phage lysins protect animals from bacteraemia and death. Moreover, the simultaneous attack of the pneumococcal peptidoglycan by a lysozyme and an amidase leads to a remarkable effect through enhanced destruction of the bacterial cell wall. The benefits of therapy with enzybiotics against pneumococcus reported here might warrant the examination of alternative strategies for the treatment of diseases caused by clinically relevant pathogens.

Abstract

Isolates of serotype 23F Streptococcus pneumoniae with high levels of resistance to penicillin have been commonly recovered in Spain for more than a decade. Recently penicillin-resistant serotype 23F S. pneumoniae strains were also isolated from children attending a day-care center in Cleveland. A number of Spanish and Cleveland isolates were compared by electrophoretic analysis of penicillin-binding protein (PDP) profiles and DNA restriction endonuclease cleavage profiles of the PDP 2X and 2D genes amplified with the polymerase chain reaction and by multilocus enzyme electrophoresis. All strains were identical by these criteria. The findings demonstrate that the Spanish and Cleveland isolates are clonally related and suggest that this antibiotic resistant clone of serotype 23F S. pneumoniae has spread intercontinentally from Spain to the United States.
