1. Adaptation strategies of giant viruses to low-temperature marine ecosystems.

Abstract

Microbes in marine ecosystems have evolved their gene content to thrive successfully in the cold. Although this process has been reasonably well studied in bacteria and selected eukaryotes, less is known about the impact of cold environments on the genomes of viruses that infect eukaryotes. Here, we analyzed cold adaptations in giant viruses (Nucleocytoviricota and Mirusviricota) from austral marine environments and compared them with their Arctic and temperate counterparts. We recovered giant virus metagenome-assembled genomes (98 Nucleocytoviricota and 12 Mirusviricota MAGs) from 61 newly sequenced metagenomes and metaviromes from sub-Antarctic Patagonian fjords and Antarctic seawater samples. When analyzing our data set alongside Antarctic and Arctic giant viruses MAGs already deposited in the Global Ocean Eukaryotic Viral (GOEV) database, we found that Antarctic and Arctic giant viruses predominantly inhabit sub-10°C environments, featuring a high proportion of unique phylotypes in each ecosystem. In contrast, giant viruses in Patagonian fjords were subject to broader temperature ranges and showed a lower degree of endemicity. However, despite differences in their distribution, giant viruses inhabiting low-temperature marine ecosystems evolved genomic cold-adaptation strategies that led to changes in genetic functions and amino acid frequencies that ultimately affect both gene content and protein structure. Such changes seem to be absent in their mesophilic counterparts. The uniqueness of these cold-adapted marine giant viruses may now be threatened by climate change, leading to a potential reduction in their biodiversity.

2. Model-driven design and validation of glucose supply control in a tubular photobioreactor operated under oxygen-balanced mixotrophy

Abstract

Scaling up bioprocesses remains challenging due to the partially unpredictable and suboptimal behavior of microbes during scale transition. Mathematical modelling serves as a valuable tool in navigating these challenges. In this study, we developed and validated a kinetic model based on the tank-in-series approach for a novel bioprocess known as oxygen-balanced mixotrophy with the microalga Galdieria sulphuraria. This model aims to facilitate the design of an effective glucose feeding control strategy tailored for two-phase tubular photobioreactors (TPBRs). The residence time distribution of our reference reactor was investigated by means of a tracer experiment. Qualitative analysis of the results indicated that our system was optimally represented by simulations with a 100 stirred-tank reactors (STRs). The controlled variable, oxygen concentration in the gas phase, showed a downward trend in simulations with simplified conditions: fixed oxygen production and balanced glucose supply. The decrease was caused by dominant net heterotrophic activity and accumulation of CO2 in the gas phase. Different configurations of a proportional-integral-derivative (PID) controller were designed by means of the ultimate gain method and compared under these simplified conditions. The most effective PID controller was implemented and refined in simulations with outdoor weather conditions. The model was validated successfully by simulating over time in a lab-scale STR the spatial fluctuations happening in a TPBR. The empirical oxygen consumption and production were faster and steeper than in the model, probably due to inaccuracies in parameter estimation or biomass adaptation.

3. De capitanes pobres y malones. La trayectoria de la conflictividad interétnica en Península Valdés (1779-1810, provincia de Chubut, República Argentina)

Abstract

El presente trabajo tiene por objeto abordar el carácter predominantemente conflictivo de las relaciones interétnicas en el contexto de los enclaves coloniales españoles establecidos en Península Valdés (provincia de Chubut, República Argentina): el Fuerte San José y el Puesto de la Fuente (1779-1810). A nivel específico, se busca presentar, describir y examinar a lo largo del tiempo los episodios de tensiones y violencia interétnica en dichos establecimientos. Para ello, nos centraremos en el análisis y la confrontación crítica de la información suministrada por las fuentes primarias a lo largo de los 31 años de ocupación de los asentamientos. La comprensión de la singularidad de las relaciones interculturales en estos escenarios, necesariamente implicó abordarlas desde una perspectiva comparativa en función de su trayectoria histórica y regional más amplia en Patagonia durante el lapso temporal considerado. En consecuencia, fue posible establecer un protagonismo destacado de grupos indígenas procedentes de Patagonia Austral, así como la identificación de múltiples causas en la génesis de los malones que experimentaron estos escenarios. Estos resultados ponen en evidencia la compleja trama de los vínculos interculturales y las relaciones coloniales durante el período colonial tardío en Patagonia.

Abstract

Polymer-based composites containing insulating inorganic fillers with different functionalities are attracting a great deal of interest for multiple applications, including embedded planar capacitors, flexible piezoelectric harvesters and portable devices for self-powered electronics. They combine typical properties of polymers, such as easy processing at low temperature, flexibility and high breakdown strength (BDS) with the dielectric or piezoelectric properties of some inorganic compounds and, in particular, of ferroelectric perovskites such as BaTiO3, 0.94 Na0.5Bi0.5TiO3 - 0.06 BaTiO3 and Ba1-yCayZrxTi1-xO3. A rational selection of components, good interface engineering and proper processing optimization allows a thorough comprehension of the complex process-structure-morphology-properties relationships of the multicomponent systems. Here, we present our most significant results on PVDF-ceramic composites for energy harvesting and storage. A careful multi-level characterization allows the optimization of the functional properties (dielectric behaviour, piezoelectricity, power density), performance and durability (mechanical strength, flexibility, fatigue resistance) as specifically required for the envisaged final devices.

7. Spontaneous parametric down-conversion in bottom-up grown lithium niobate microcubes

Abstract

Nonclassical light sources are highly sought-after as they are an integral part of quantum communication and quantum computation devices. Typical sources use bulk nonlinear crystals that rely on stringent phase-matching conditions, limiting the operating wavelength and bandwidth. In this work, we demonstrate the generation of photon pairs from a freestanding lithium niobate microcube at the telecommunication wavelength of 1.56 µm through the spontaneous parametric down-conversion process. The maximum photon pair generation rate obtained from a single microcube with the size of 3.6 µm is 490 Hz, resulting in an efficiency of 20.6 GHz/Wm, which is three orders of magnitude larger than the efficiency of biphoton generation in bulk nonlinear crystals. The microcubes are synthesized through a solvothermal method, offering the possibility for scalable devices via bottom-up assembly on any substrates. Our work constitutes an important step forward in the realization of compact nonclassical light sources with a wide bandwidth for various quantum applications.

11. BELIEFS ABOUT FALSE ALLEGATIONS OF SEXUAL VIOLENCE: INTERGROUP CONTACT THEORY AND RAPE MYTH ACCEPTANCE AT A ROCKY MOUNTAIN UNIVERSITY

Abstract

False allegations of sexual assault represent approximately 2% to 10% of all sexual assault allegations reported to law enforcement (Lisak et al., 2010). Despite this low prevalence rate, people tend to overestimate the occurrence of false allegations, and give false allegations excessive weight in arguments about rape myths and policy-making decisions. This excessive weight is given despite a lack of research evidence that false reports of sexual violence occur more frequently than those of any other crime (Ask, 2010; McMillan, 2018; Stabile et al., 2019). This overestimation is likely influenced by the overrepresentation and sensationalization of false allegations in the media, public discourse, and online social networking platforms (Banet-Weiser, 2021; Jones et al., 2021; Stabile et al., 2019). The current study seeks to examine this false allegation myth through quantitative study of young adults in a university setting, for whom sexual violence perpetration and victimization tend to be more prevalent than in the general population (Black et al., 2011; Sinozich & Langton, 2014; Rickert et al., 2004). Quantitative analysis of 1,060 participant responses to a university-wide student survey demonstrated that male gender, conservative political beliefs, false accuser acquaintance, lack of survivor acquaintance, lack of primary victimization experiences, and gaps in definitional knowledge of sexual violence and false allegations were associated with false allegation myth acceptance. This study highlights the importance of educational efforts in dispelling rape myths and evidences support for Intergroup Contact Theory as a model for educational intervention efforts.

Abstract

Background Complete closure of large mucosal defects following colorectal endoscopic submucosal dissection (ESD) with through-the-scope (TTS) clips is oftentimes not possible. We aimed to report our early experience of using a novel TTS suturing system for the closure of large mucosal defects after colorectal ESD.Methods We performed a retrospective multicenter cohort study of consecutive patients who underwent attempted prophylactic defect closure using the TTS suturing system after colorectal ESD. The primary outcome was technical success in achieving complete defect closure, defined as a < 5 mm residual mucosal defect in the closure line using TTS suturing, with or without adjuvant TTS clips.Results 82 patients with a median defect size of 30 (interquartile range 25-40) mm were included. Technical success was achieved in 92.7 % (n = 76): TTS suturing only in 44 patients (53.7 %) and a combination of TTS suturing to approximate the widest segment followed by complete closure with TTS clips in 32 (39.0 %). Incomplete/partial closure, failure of appropriate TTS suture deployment, and the need for over-the-scope salvage closure methods were observed in 7.3 % (n = 6). One intraprocedural bleed, one delayed bleed, and three intraprocedural perforations were observed. There were no adverse events related to placement of the TTS suture.Conclusion The TTS suture system is an effective and safe tool for the closure of large mucosal defects after colorectal ESD and is an alternative when complete closure with TTS clips alone is not possible.

13. Flexible composite films with enhanced piezoelectric properties for energy harvesting and wireless ultrasound-powered technology

Abstract

In the last years, ultrasound energy harvesting has emerged as the most promising technique for wireless power supply of implanted medical devices. These devices require flexible piezoelectric materials with high piezoelectric response in the ultrasonic range. Here we report on bio-compatible NBT-BT/PVDF flexible composites, with variable filler content up to 50 vol%, prepared by a properly designed and optimized process, which incorporates in a complex connectivity pattern fully sintered NBT-BT crystalline powders in a PVDF matrix. The dielectric constant of the flexible composites increased from 10 of pure PVDF polymer to 110 of composite films with 50 vol% NBT-BT content, while the high frequency piezoelectric d33 constant increased from 0.2 pC/N to 33 pC/N for the same samples. The composite with 50 vol% NBT-BT exhibits the figure of merit for the harvested ultrasound energy d33g33 ≅ 1.54 × 10−12 m3/J, which is comparable to the figure of merit for the NBT-BT piezoelectric ceramic (1.8 × 10−12 m3/J) and higher than other reported results for random composites. Based on these results, this study provides an easy method to fabricate random flexible piezoelectric composites with enhanced high frequency piezoelectric response and high energy density harvested from an ultrasound source.

Marine animal by-products of the food industry are a great source of valuable biomolecules. Skins and bones are rich in collagen, a protein with various applications in food, cosmetic, healthcare, and medical industries in its native form or partially hydrolyzed (gelatin). Salmon gelatin is a candidate of interest due to its high biomass production available through salmon consumption, its biodegradability, and its high biocompatibility. However, its low mechanical and thermal properties can be an obstacle for various applications requiring cohesive material. Thus, gelatin modification by cross-linking is necessary. Enzymatic cross-linking by microbial transglutaminase (MTG) is preferred to chemical cross-linking to avoid the formation of potentially cytotoxic residues. In this work, the potential of salmon skin gelatin was investigated, in a comparative study with porcine gelatin, and an enzymatic versus chemical cross-linking analysis. For this purpose, the two cross-linking methods were applied to produce three-dimensional, porous, and mechanically reinforced hydrogels and sponges with different MTG ratios (2%, 5%, and 10% w/w gelatin). Their biochemical, rheological, and structural properties were characterized, as well as the stability of the material, including the degree of syneresis and the water-binding capacity. The results showed that gelatin enzymatically cross-linked produced material with high cross-linking densities over 70% of free amines. The MTG addition seemed to play a crucial role, as shown by the increase in mechanical and thermal resistances with the production of a cohesive material stable above 40 °C for at least 7 days and comparable to porcine and chemically cross-linked gelatins. The coupled problem of hydrodynamics and solute transport for the Najafi-Golestanian three-sphere swimmer is studied, with the Reynolds number set to zero and P\'eclet numbers (Pe) ranging from 0.06 to 60. The adopted method is the numerical simulation of the problem with a finite element code based upon the FEniCS library. For the swimmer executing the optimal locomotion gait, we report the Sherwood number as a function of Pe in homogeneous fluids and confirm that little gain in solute flux is achieved by swimming unless Pe is significantly larger than 10. We also consider the swimmer as an learning agent moving inside a fluid that has a concentration gradient. The outcomes of Q-learning processes show that learning locomotion (with the displacement as reward) is significantly easier than learning chemotaxis (with the increase of solute flux as reward). The chemotaxis problem, even at low Pe, has a varying environment that renders learning more difficult. Further, the learning difficulty increases severely with the P\'eclet number. The results demonstrate the challenges that natural and artificial swimmers need to overcome to migrate efficiently when exposed to chemical inhomogeneities., Comment: 20 pages, 13 figures. Abstract

[EN] BaTiO has been extensively studied because of their microwave dielectrics properties. Traditionally, it is difficult to achieve this material as single-phase. Here, we report an effective method to obtain BaTiO powder with nanometer-scale crystals, by solid-state reaction at moderate temperatures and using as precursors nanostructured particles consisting of BaTiO and TiO. The main advantage is the intimate contact between the BaTiO and TiO that ensure, when the solid-state reaction takes place, the formation of complex solid compounds from three or more constituents. The formation mechanism of BaTiO has been studied as a function of both the thermal treatment and the time reaction. The reaction was monitored by Raman spectroscopy combined with Confocal microscopy, the aim of this characterization technique is to provide the description of the general strategy and design principles to obtain BaTiO powder. Consequently, this work is a challenging task for the compositional and structural study of complex inorganic nanoparticles.

20. A finite element method for simulating soft active non-shearable rods immersed in generalized Newtonian fluids

Abstract

We propose a finite element method for simulating one-dimensional solid models moving and experiencing large deformations while immersed in generalized Newtonian fluids. The method is oriented towards applications involving microscopic devices or organisms in the soft-bio-matter realm. By considering that the strain energy of the solid may explicitly depend on time, we incorporate a mechanism for active response. The solids are modeled as Cosserat rods, a detailed formulation being provided for the special case of a planar non-shearable rod. The discretization adopts one-dimensional Hermite elements for the rod and low-order Lagrange two-dimensional elements for the fluid's velocity and pressure. The fluid mesh is boundary-fitted, with remeshing at each time step. Several time marching schemes are studied, of which a semi-implicit scheme emerges as most effective. The method is demonstrated in very challenging examples: the roll-up of a rod to circular shape and later sudden release, the interaction of a soft rod with a fluid jet and the active self-locomotion of a sperm-like rod. The article includes a detailed description of a code that implements the method in the Firedrake library.

21. A multiscale Robin-coupled implicit method for two-phase flows in high-contrast formations

Abstract

In the presence of strong heterogeneities, it is well known that the use of explicit schemes for the transport of species in a porous medium suffers from severe restrictions on the time step. This has led to the development of implicit schemes that are increasingly favoured by practitioners for their computational efficiency. The transport equation requires knowledge of the velocity field, which results from an elliptic problem (Darcy problem) that is the most expensive part of the computation. When considering large reservoirs, a cost-effective way of approximating the Darcy problems is using multiscale domain decomposition (MDD) methods. They allow for the pressure and velocity fields to be computed on coarse meshes (large scale), while detailed basis functions are defined locally, usually in parallel, in a much finer grid (small scale). In this work we adopt the Multiscale Robin Coupled Method (MRCM, [Guiraldello, et al., J. Comput. Phys., 355 (2018) pp. 1-21], [Rocha, et al., J. Comput. Phys., (2020) 109316]), which is a generalization of previous MDD methods that allows for great flexibility in the choice of interface spaces. In this article we investigate the combination of the MRCM with implicit transport schemes. A sequentially implicit strategy is proposed, with different trust-region algorithms ensuring the convergence of the transport solver. The method is assessed on several very stringent 2D two-phase problems, demonstrating its stability even for large time steps. It is also shown that the best accuracy is achieved by considering recently introduced non-polynomial interface spaces, since polynomial spaces are not optimal for high-contrast channelized permeability fields., Comment: 41 pages, 23 figures. Abstract

A three dimensional parallel implementation of Multiscale Mixed Methods based on non-overlapping domain decomposition techniques is proposed for multi-core computers and its computational performance is assessed by means of numerical experimentation. As a prototypical method, from which many others can be derived, the Multiscale Robin Coupled Method is chosen and its implementation explained in detail. Numerical results for problems ranging from millions up to more than 2 billion computational cells in highly heterogeneous anisotropic rock formations based on the SPE10 benchmark are shown. The proposed implementation relies on direct solvers for both local problems and the interface coupling system. We find good weak and strong scalalability as compared against a state-of-the-art global fine grid solver based on Algebric Multigrid preconditioning in single and two-phase flow problems., Comment: 25 pages, 12 figures

23. Interface spaces based on physics for multiscale mixed methods applied to flows in fractured-like porous media

Abstract

It is well known that domain-decomposition-based multiscale mixed methods rely on interface spaces, defined on the skeleton of the decomposition, to connect the solution among the non-overlapping subdomains. Usual spaces, such as polynomial-based ones, cannot properly represent high-contrast channelized features such as fractures (high permeability) and barriers (low permeability) for flows in heterogeneous porous media. We propose here new interface spaces, which are based on physics, to deal with permeability fields in the simultaneous presence of fractures and barriers, accommodated respectively, by the pressure and flux spaces. Existing multiscale methods based on mixed formulations can take advantage of the proposed interface spaces, however, in order to present and test our results, we use the newly developed Multiscale Robin Coupled Method (MRCM) [Guiraldello, et al., J. Comput. Phys., 355 (2018) pp. 1-21], which generalizes most well-known multiscale mixed methods, and allows for the independent choice of the pressure and flux interface spaces. An adaptive version of the MRCM [Rocha, et al., J. Comput. Phys., 409 (2020), 109316] is considered that automatically selects the physics-based pressure space for fractured structures and the physics-based flux space for regions with barriers, resulting in a procedure with unprecedented accuracy. The features of the proposed approach are investigated through several numerical simulations of single-phase and two-phase flows, in different heterogeneous porous media. Abstract

Ceramics with perovskite structure and composition BaCexTi1-xO3 (x = 0.02-0.30) show a progressive evolution with increasing x, from the long-range polar order of ferroelectric BaTiO3 to the short-range polar order typical of relaxors. The ionic size mismatch between Ti4+ and Ce4+ determines strong local strains which have a significant impact on dielectric properties and phase transitions. The pair distribution function, coupled with transmission electron microscopy analysis, was applied to study the local structure. Because of the inner B-cation sizes, the superposition of rigid B—O octahedra with different volumes is not compatible with the construction of an ideal perovskite structure. In this light, local structure can be described by an original model which allows (i) different Ti—O and Ce—O distances and (ii) the typical distortions of the two end members: off-center displacement of Ti occurring in BaTiO3 and octahedral tilt in BaCeO3. The results show a clear difference, in terms of volumes, between oxygen octahedra with titanium and those related to cerium. In addition, the inclusion of cerium causes a tilt of its oxygen cage, as occurs in pure BaCeO3, creating contra-rotations and distortions of the octahedra containing titanium. This complex arrangement entails a substantial distortion, increasing as a function of cerium amount, which strongly influences the directions of titanium displacements, their local correlation and consequently their long-range cooperative effects.

Abstract

Objectives: The aim of this study was to investigate the safety and efficacy of coronary protection by preventive coronary wiring and stenting across the coronary ostia in patients at high risk for coronary obstruction after transcatheter aortic valve replacement (TAVR). Background: Coronary obstruction following TAVR is a life-threatening complication with high procedural and short-term mortality. Methods: Data were collected retrospectively from a multicenter international registry between April 2011 and February 2019. Results: Among 236 patients undergoing coronary protection with preventive coronary wiring, 143 had eventually stents implanted across the coronary ostia after valve deployment. At 3-year follow-up, rates of cardiac death were 7.8% in patients receiving stents and 15.7% in those not receiving stents (adjusted hazard ratio: 0.42; 95% confidence interval: 0.14 to 1.28; p = 0.13). There were 2 definite stent thromboses (0.9%) in patients receiving stents, both occurring after TAVR in “valve-in-valve” procedures. In patients not receiving stents, there were 4 delayed coronary occlusions (DCOs) (4.3%), occurring from 5 min to 6 h after wire removal. Three cases occurred in valve-in-valve procedures and 1 in a native aortic valve procedure. Distance between the virtual transcatheter valve and the protected coronary ostia <4 mm was present in 75.0% of patients with DCO compared with 30.4% of patients without DCO (p = 0.19). Conclusions: In patients undergoing TAVR at high risk for coronary obstruction, preventive stent implantation across the coronary ostia is associated with good mid-term survival rates and low rates of stent thrombosis. Patients undergoing coronary protection with wire only have a considerable risk for DCO.

Abstract

BACKGROUND: Lung ultrasound (LUS) is a useful tool for assessing lung status in critically ill patients, but is not widely applied in intensive care units (ICUs). The aim of this study was to investigate the use of LUS in Italian ICUs. METHODS: A questionnaire was distributed to 116 ICU representatives belonging to the Italian GiViTI network, participating in a national project on continuous quality of care assessment. The answers to the questionnaire were analyzed with the data collected in the course of said project. RESULTS: 87 ICUs took part in the study. LUS was performed in 94.3% of them, generally by an internal intensivist. The mean number of tests per bed per week was 0.97 (SD, 1.0). A standardized reporting format was present in only 11% of ICUs. LUS was generally used to investigate known or suspected diseases and as a screening tool in only 10%. The number of testes per bed per week was inversely correlated to the number of beds per physician (P<0.001) and to the provider performing the LUS (P<0.001), but not to other structural, organizational or performance data. CONCLUSIONS: LUS is performed in the majority of ICUs and is generally used to investigate known or suspected diseases. Frequency of use appears to be low and related to selected organizational factors. There is generally no standardized format for reporting LUS findings.

Se propone una reflexión teórica a partir del desarrollo de un plan maestro para un predio de 1.022 ha, ubicado en la periferia de Santiago de Chile.El Plan Maestro propone una serie de operaciones arquitectónicas y de urbanismo del paisaje, a diversas escalas, donde se consideran los cambios de uso de suelo a nivel comunal, otros proyectos de interés ambiental y social, la proximidad de áreas naturales y las condiciones propias del sitio, con especial atención a la laguna y al humedal asociado.

El tamaño tiene consecuencias prácticas y disciplinarias, también es un desafío para la imaginación. Cuando nos preguntamos cómo actuar hoy sobre lo vasto, necesariamente tenemos que conciliar y coordinar un enjambre de múltiples intereses, divisiones administrativas, regulaciones, condiciones ambientales y variables económicas; Tenemos también que incorporar el cambio y las medidas adaptativas, diseñando un proceso a lo largo del tiempo; conciliar los aparentemente contradictorios objetivos ecológicos y la programación de los usos. Preguntarnos, qué ocurre con las cualidades perceptuales y fenomenológicas de un sitio, cómo mantener y expresar la calidad del lugar y su significado cultural; cómo reconocer y diseñar con las huellas que han dejado usos anteriores. ¿Puede entonces, un diseño adaptarse a múltiples escalas, asumir la diversidad y adaptar las formas existentes a las nuevas prácticas sociales, para hacer que un lugar sea memorable?. Proponemos que los parques de gran tamaño pueden ser un verdadero laboratorio, que nos proporcione normas para actuar en el mundo más complejo de la ciudad. El proyecto que presentamos supone pensar el territorio bajo supuestos paisajísticos a múltiples escalas. Se trata del plan maestro para un predio de 1.022 hectáreas en la periferia de la ciudad de Santiago, que la Universidad de Chile, propone como un parque, un gran parque público.

34. An extended Elrod-Adams model to account for backpressure and blow-by inception

Abstract

The Piston-Ring-Liner system is the main tribological component of internal combustion engines. The Elrod-Adams model is customarily used to numerically assess the hydrodynamics of different ring designs and liner surface treatments. However, that model does not incorporate the backpressure boundary condition, which in this case corresponds to the combustion chamber pressure and is quite significant. In this contribution a model is proposed that imposes the combustion-chamber pressure in a mass-conserving way, together with an effective algorithm for its numerical approximation. The new model incorporates the pressure difference across the ring, which is shown to have a substantial effect on the predicted friction force and MFT. Abstract

The prompt availability of reliable epidemiological information on emerging pandemics is crucial for public health policy-makers. Early in 2013, a possible new H1N1 epidemic notified by an intensive care unit (ICU) to GiViTI, the Italian ICU network, prompted the re-activation of the real-time monitoring system developed during the 2009-2010 pandemic. Based on data from 216 ICUs, we were able to detect and monitor an outbreak of severe H1N1 infection, and to compare the situation with previous years. The timely and correct assessment of the severity of an epidemic can be obtained by investigating ICU admissions, especially when historical comparisons can be made.

Abstract

The interaction of carbohydrates with a variety of biological targets, including antibodies, proteins, viruses and cells are of utmost importance in many aspects of biology. Glycan microarrays are increasingly used to determine the binding specificity of glycan-binding proteins. In this study, a novel slide is reported for the fabrication of glycan arrays that combines the higher sensitivity of a layered Si-SiO2 with a novel approach to form a polymeric coating easily modifiable by subsequent click reaction. The alkyne-containing copolymer, adsorbed from an aqueous solution, produces a coating by a single step procedure and serves as a soft, tridimensional support for the oriented immobilization of carbohydrates via azide/alkyne Cu(I) catalyzed "click" reaction. The equilibrium and kinetics parameters of the interaction of Concanavalin A with eight synthetic glycans were determined using fluorescence microarray and Reflective Phantom Interface (RPI), a recently proposed optical label-free detection approach. The enhancement of fluorescence provided by the Si-SiO2 slides enabled to extend the limit of detection at lower surface densities of lectins, in turn enabling the study of the interaction for a wide range of glycans surface density. Equilibrium dissociation constants of a few nM were extracted for multivalent glycan-lectin binding, mimicking the conditions of biological membranes, whereas hundreds of nM were observed at the lower glycan surface densities.

Abstract

The object of this study is to analyze the use and adaptation of racialist ideology in the Afro-Hispanic Antilles following the start of the Revolution of Saint-Domingue in 1791, as it evolved to justify and reinforce plantation slavery and served to reinstitute and police the color line that was the central ideological premise supporting the economy of exchange and exploitation in the world of Atlantic coloniality. The renewed stigmatization of the racialized types in Creole population aimed to limit the echoes of the revolution against the plantation and it was an attempt to dismiss its political significance as a movement of self-emancipation and decolonization. The fear promoted by the colonial authorities, the planter class and Creole intellectuals, liberal and otherwise, aimed to establish a delicate balance between terror and profits wanting to justify the continuation of plantation slavery through the purposeful resemantization of the ideological tandem civilization/barbarity based on a racialized reading of history that championed European immigration and the systematic reduction of the population of Afro-descendants.

Abstract

Bioadhesion of marine organisms has been intensively studied over the last decade because of their ability to attach in various wet environmental conditions and the potential this offers for biotechnology applications. Many marine mollusc species are characterized by a two-phase life history: pelagic larvae settle prior to metamorphosis to a benthic stage. The oyster Crassostrea gigas has been extensively studied for its economic and ecological importance. However, the bioadhesive produced by ready to settle larvae of this species has been little studied. The pediveliger stage of oysters is characterized by the genesis of a specific organ essential for adhesion, the foot. Our scanning electron microscopy and histology analysis revealed that in C. gigas the adhesive is produced by several foot glands. This adhesive is composed of numerous fibres of differing structure, suggesting differences in chemical composition and function. Fourier transformed infrared spectroscopy indicated a mainly proteinaceous composition. Proteomic analysis of footprints was able to identify 42 proteins, among which, one uncharacterized protein was selected on the basis of its pediveliger transcriptome specificity and then located by mRNA in situ hybridization, revealing its potential role during substrate exploration before oyster larva settlement.

Abstract

Pancreatic cancer is the most lethal common solid malignancy. Systemic therapies are often ineffective, and predictive biomarkers to guide treatment are urgently needed. We generated a pancreatic cancer patient-derived organoid (PDO) library that recapitulates the mutational spectrum and transcriptional subtypes of primary pancreatic cancer. New driver oncogenes were nominated and transcriptomic analyses revealed unique clusters. PDOs exhibited heterogeneous responses to standard-of-care chemotherapeutics and investigational agents. In a case study manner, we found that PDO therapeutic profiles paralleled patient outcomes and that PDOs enabled longitudinal assessment of chemosensitivity and evaluation of synchronous metastases. We derived organoid-based gene expression signatures of chemosensitivity that predicted improved responses for many patients to chemotherapy in both the adjuvant and advanced disease settings. Finally, we nominated alternative treatment strategies for chemorefractory PDOs using targeted agent therapeutic profiling. We propose that combined molecular and therapeutic profiling of PDOs may predict clinical response and enable prospective therapeutic selection.Significance: New approaches to prioritize treatment strategies are urgently needed to improve survival and quality of life for patients with pancreatic cancer. Combined genomic, transcriptomic, and therapeutic profiling of PDOs can identify molecular and functional subtypes of pancreatic cancer, predict therapeutic responses, and facilitate precision medicine for patients with pancreatic cancer. Abstract

Bioadhesion of marine organisms has been intensively studied over the last decade because of their ability to attach in various wet environmental conditions and the potential this offers for biotechnology applications. Many marine mollusc species are characterized by a two-phase life history: pelagic larvae settle prior to metamorphosis to a benthic stage. The oyster Crassostrea gigas has been extensively studied for its economic and ecological importance. However, the bioadhesive produced by ready to settle larvae of this species has been little studied. The pediveliger stage of oysters is characterized by the genesis of a specific organ essential for adhesion, the foot. Our scanning electron microscopy and histology analysis revealed that in C. gigas the adhesive is produced by several foot glands. This adhesive is composed of numerous fibres of differing structure, suggesting differences in chemical composition and function. Fourier transformed infrared spectroscopy indicated a mainly proteinaceous composition. Proteomic analysis of footprints was able to identify 42 proteins, among which, one uncharacterized protein was selected on the basis of its pediveliger transcriptome specificity and then located by mRNA in situ hybridization, revealing its potential role during substrate exploration before oyster larva settlement.

Abstract

Background & aimsIt is unclear whether participation in competency-based fellowship programs for endoscopic ultrasound (EUS) and endoscopic retrograde cholangiopancreatography (ERCP) results in high-quality care in independent practice. We measured quality indicator (QI) adherence during the first year of independent practice among physicians who completed endoscopic training with a systematic assessment of competence.MethodsWe performed a prospective multicenter cohort study of invited participants from 62 training programs. In phase 1, 24 advanced endoscopy trainees (AETs), from 20 programs, were assessed using a validated competence assessment tool. We used a comprehensive data collection and reporting system to create learning curves using cumulative sum analysis that were shared with AETs and trainers quarterly. In phase 2, participating AETs entered data into a database pertaining to every EUS and ERCP examination during their first year of independent practice, anchored by key QIs.ResultsBy the end of training, most AETs had achieved overall technical competence (EUS 91.7%, ERCP 73.9%) and cognitive competence (EUS 91.7%, ERCP 94.1%). In phase 2 of the study, 22 AETs (91.6%) participated and completed a median of 136 EUS examinations per AET and 116 ERCP examinations per AET. Most AETs met the performance thresholds for QIs in EUS (including 94.4% diagnostic rate of adequate samples and 83.8% diagnostic yield of malignancy in pancreatic masses) and ERCP (94.9% overall cannulation rate).ConclusionsIn this prospective multicenter study, we found that although competence cannot be confirmed for all AETs at the end of training, most meet QI thresholds for EUS and ERCP at the end of their first year of independent practice. Abstract

Pancreatic cancer is the most lethal common solid malignancy. Systemic therapies are often ineffective, and predictive biomarkers to guide treatment are urgently needed. We generated a pancreatic cancer patient-derived organoid (PDO) library that recapitulates the mutational spectrum and transcriptional subtypes of primary pancreatic cancer. New driver oncogenes were nominated and transcriptomic analyses revealed unique clusters. PDOs exhibited heterogeneous responses to standard-of-care chemotherapeutics and investigational agents. In a case study manner, we found that PDO therapeutic profiles paralleled patient outcomes and that PDOs enabled longitudinal assessment of chemosensitivity and evaluation of synchronous metastases. We derived organoid-based gene expression signatures of chemosensitivity that predicted improved responses for many patients to chemotherapy in both the adjuvant and advanced disease settings. Finally, we nominated alternative treatment strategies for chemorefractory PDOs using targeted agent therapeutic profiling. We propose that combined molecular and therapeutic profiling of PDOs may predict clinical response and enable prospective therapeutic selection.Significance: New approaches to prioritize treatment strategies are urgently needed to improve survival and quality of life for patients with pancreatic cancer. Abstract

Recognizing and quantifying specific biomolecules in aqueous samples are constantly needed in research and diagnostic laboratories. As the typical detection procedures are rather lengthy and involve the use of labeled secondary antibodies or other agents to provide a signal, efforts have been made over the last 10 y to develop alternative label-free methods that enable direct detection. We propose and demonstrate an extremely simple, lowcost, label-free biodetector based on measuring the intensity of light reflected by the interface between a fluid sample and an amorphous fluoropolymer substrate having a refractive index very close to that of water and hosting various antibodies immobilized in spots. Under these index-matching conditions, the amount of light reflected by the interface allows straightforward quantification of the amount of antigen binding to each spot. Using antibodies targeting heterologous immunoglobulins and antigens commonly used as markers for diagnoses of hepatitis B and HIV, we demonstrate the limit of detection of a few picograms per square millimeter of surface-bound molecules. We also show that direct and real-time access to the amount of binding molecules allows the precise extrapolation of adhesion rates, from which the concentrations of antigens in solution can be estimated down to fractions of nanograms per milliliter.

Abstract

The nonlinear dielectric properties of dense BaTiO(3) ceramics with grain size of 1 mu m-90 nm were investigated. In the finest ceramics, the permittivity reduces below 1000 and a remarkable nonhysteretic linear dc-tunability [epsilon(E)] is obtained at high field, above 40 kV/cm. The observed behavior was explained by considering the nanostructured ceramic as a composite formed by ferroelectric grains, whose nonlinearity is reducing, and by low-permittivity nonferroelectric grain boundaries, whose volume fraction increases when decreasing the grain size.
