Abstract

Aim: We examined whether metabolic dysfunction-associated steatotic liver disease (MASLD) with or without significant fibrosis (assessed by validated non-invasive biomarkers) was associated with an increased risk of prevalent chronic kidney disease (CKD) or diabetic retinopathy in people with type 1 diabetes mellitus (T1DM). Methods: We performed a retrospective multicenter cross-sectional study involving 1,409 adult outpatients with T1DM, in whom hepatic steatosis index (HSI) and fibrosis (FIB)-4 index were calculated for non-invasively detecting hepatic steatosis (defined by HSI > 36), with or without coexisting significant fibrosis (FIB-4 index ≥ 1.3 or < 1.3). CKD was defined as an estimated glomerular filtration rate (eGFR) < 60 mL/min/1.73 m2 or urine albumin/creatinine ratio ≥ 3.0 mg/mmol. The presence of diabetic retinopathy was also recorded in all participants. Results: Patients with MASLD and significant fibrosis (n = 93) had a remarkably higher prevalence of CKD and diabetic retinopathy than their counterparts with MASLD without fibrosis (n = 578) and those without steatosis (n = 738). After adjustment for sex, diabetes duration, hemoglobin A1c, hypertension, and use of antihypertensive or lipid-lowering medications, patients with SLD and significant fibrosis had a higher risk of prevalent CKD (adjusted-odds ratio 1.76, 95 % confidence interval 1.05–2.96) than those without steatosis. Patients with MASLD without fibrosis had a higher risk of prevalent retinopathy (adjusted-odds ratio 1.49, 95 % CI 1.13–1.46) than those without steatosis. Conclusion: This is the largest cross-sectional study showing that MASLD with and without coexisting significant fibrosis was associated, independently of potential confounders, with an increased risk of prevalent CKD and retinopathy in adults with T1DM.

Abstract

We present the results of a fourth planetary defense exercise, focused this time on the small near-Earth asteroid (NEA) 2023 DZ2 and conducted during its close approach to the Earth in 2023 March. The International Asteroid Warning Network (IAWN), with support from NASA's Planetary Defense Coordination Office (PDCO), has been coordinating planetary defense observational campaigns since 2017 to test the operational readiness of the global planetary defense capabilities. The last campaign focused on the NEA Apophis, and an outcome of that exercise was the need for a short burst campaign to replicate a real-life near-Earth object impact hazard scenario. The goal of the 2023 DZ2 campaign was to characterize the small NEA as a potential impactor and exercise the planetary defense system including observations, hypothetical risk assessment and risk prediction, and hazard communication with a short notice of just 24 hr. The entire campaign lasted about 10 days. The campaign team was divided into several working groups based on the characterization method: photometry, spectroscopy, thermal IR photometry and optical polarimetry, radar, and risk assessment. Science results from the campaign show that 2023 DZ2 has a rotation period of 6.2745 ± 0.0030 minutes; visible wavelength color photometry/spectroscopy/polarimetry and near-IR spectroscopy all point to an E-type taxonomic classification with surface composition analogous to aubrite meteorites; and radar observations show that the object has a diameter of 30 ± 10 m, consistent with the high albedo (0.49) derived from polarimetric and thermal IR observations.

Abstract

In this paper, we study Polynomial Lawvere logic (PL), a logic on the quantale of the extended positive reals, developed for reasoning about metric spaces. PL is appropriate for encoding quantitative reasoning principles, such as quantitative equational logic. PL formulas include the polynomial functions on the extended positive reals, and its judgements include inequalities between polynomials. We present an inference system for PL and prove a series of completeness and incompleteness results relying and the Krivine-Stengle Positivstellensatz (a variant of Hilbert's Nullstellensatz) including completeness for finitely axiomatisable PL theories. We also study complexity results both for both PL and its affine fragment (AL). We demonstrate that the satisfiability of a finite set of judgements is NP-complete in AL and in PSPACE for PL; and that deciding the semantical consequence from a finite set of judgements is co-NP complete in AL and in PSPACE in PL.

4. MMSE Channel Estimation in Large-Scale MIMO: Improved Robustness with Reduced Complexity

Abstract

Large-scale MIMO systems with a massive number N of individually controlled antennas pose significant challenges for minimum mean square error (MMSE) channel estimation, based on uplink pilots. The major ones arise from the computational complexity, which scales with $N^3$, and from the need for accurate knowledge of the channel statistics. This paper aims to address both challenges by introducing reduced-complexity channel estimation methods that achieve the performance of MMSE in terms of estimation accuracy and uplink spectral efficiency while demonstrating improved robustness in practical scenarios where channel statistics must be estimated. This is achieved by exploiting the inherent structure of the spatial correlation matrix induced by the array geometry. Specifically, we use a Kronecker decomposition for uniform planar arrays and a well-suited circulant approximation for uniform linear arrays. By doing so, a significantly lower computational complexity is achieved, scaling as $N\sqrt{N}$ and $N\log N$ for squared planar arrays and linear arrays, respectively., Comment: Submitted to IEEE Transactions on Wireless Communications, 11 figures, 3 tables

Abstract

The Earth close approach of near-Earth asteroid 2005 LW3 on 2022 November 23 represented a good opportunity for a second observing campaign to test the timing accuracy of astrometric observation. With 82 participating stations, the International Asteroid Warning Network collected 1046 observations of 2005 LW3 around the time of the close approach. Compared to the previous timing campaign targeting 2019 XS, some individual observers were able to significantly improve the accuracy of their reported observation times. In particular, U.S. surveys achieved good timing performance. However, no broad, systematic improvement was achieved compared to the previous campaign, with an overall negative bias persisting among the different observers. Abstract

Context. The physical characterization of trans-Neptunian objects is essential for improving our understanding of the formation and evolution of our Solar System. Stellar occultation is a ground-based technique that can be successfully used to determine some of the TNOs’ fundamental physical properties with high precision, such as size and shape. Aims. This work is aimed at constraining the size, shape, and geometric albedo of the dwarf planet candidate (307261) 2002 MS4 through the analysis of nine stellar occultation events. Using multichord detection, we also study the object’s topography by analyzing the obtained limb and residuals between the observed chords and the best-fit ellipse. Metods. We predicted and organized the observational campaigns of nine stellar occultations by 2002 MS4 between 2019 and 2022, resulting in two single-chord events, four double-chord detections, and three events with between 3 and 61 positive chords. We derived the occultation light curves using differential aperture photometry, from which the star ingress and egress instants were calculated. Using 13 selected chords from the 8 August 2020 event, we determined the global elliptical limb of 2002 MS4. The best-fit ellipse, combined with the object’s rotational information from the literature, sets constraints on the object’s size, shape, and albedo. Additionally, we developed a new method to characterize the topography features on the object’s limb. Results. The global limb has a semi-major axis of a′ = 412 ± 10 km, a semi-minor axis of b′= 385 ± 17 km, and the position angle of the minor axis is 121° ± 16°. From this instantaneous limb, we obtained 2002 MS4’s geometric albedo of pV = 0.1 ± 0.025, using HV = 3.63 ± 0.05 mag and a projected area-equivalent diameter of 796 ± 24 km. Abstract

A growing number of scientific contributions suggest that COVID-19 infection can lead to impairment of cognition, mainly in executive functions and memory domains, even in the absence of frank neurological pathologies. The primary objective of this retrospective study is to evaluate the frequency and type of inefficiencies in a selection of cognitive tests administered to a sample of subjects who, following infection, required invasive assisted ventilation and were admitted to rehabilitation wards for the treatment of functional impairment. Fifty-seven subjects were enrolled. The recruited patients undergone an assessment of verbal and visuospatial memory and executive functions, upon entry into the rehabilitation department, after discharge from intensive care. The following tests were administered: Rey Auditory Verbal Learning Test (AVLT) (immediate and delayed recall), Rey-Osterrieth Complex Figure Test (ROCFT) (copy and delayed recall), Stroop Color-Word Test, and Trail Making Test (TMT, A and B). Deficient scores, in beyond 25% of subjects, were found in the copy of the ROCFT (32.1% of subjects), and in the delayed recall of ROCFT (27.2%). Between 10 and 20% of patients presented an abnormal result in delayed recall of AVLT (16.07%), and Stroop Test (time, 15.6%, error, 11.5%). Less than 10% of the sample had abnormal performances on TMT (A, 3.5%, and B, 9.4%), and in AVLT immediate recall (8.9%). Correlations of the performances with age, sex, and education were also found. This paper highlights the high incidence of abnormal cognitive performances in this specific subpopulation of patients with COVID-19 infection.

Abstract

The Earth close approach of near-Earth asteroid 2005 LW3 on 2022 November 23 represented a good opportunity for a second observing campaign to test the timing accuracy of astrometric observation. With 82 participating stations, the International Asteroid Warning Network collected 1046 observations of 2005 LW3 around the time of the close approach. Compared to the previous timing campaign targeting 2019 XS, some individual observers were able to significantly improve the accuracy of their reported observation times. In particular, U.S. surveys achieved good timing performance. However, no broad, systematic improvement was achieved compared to the previous campaign, with an overall negative bias persisting among the different observers. The calibration of observing times and the mitigation of timing errors should be important future considerations for observers and orbit computers, respectively.

Abstract

Aim: We examined whether different insulin administration modalities, i.e., multiple daily injections (MDI) or continuous subcutaneous insulin infusion (CSII by insulin pumps), are differently associated with the risk of having metabolic dysfunction-associated fatty liver disease (MAFLD), with or without coexisting significant liver fibrosis (assessed by validated non-invasive biomarkers), in adults with type 1 diabetes mellitus (T1DM). Methods: We conducted a retrospective, multicenter, cross-sectional study involving 1,417 adult individuals with established T1DM treated with MDI or CSII. We calculated hepatic steatosis index (HSI) and fibrosis (FIB)-4 index for non-invasively detecting MAFLD (defined by HSI >36), with or without coexisting significant fibrosis (defined by FIB-4 index ≥ 1.3 or <1.3, respectively). Results: Compared to the MDI group (n = 1,161), insulin-pump users (n = 256; 18.1%) were more likely to be younger (mean age: 40 vs. 48 years, P < 0.001), had better glycemic control (mean hemoglobin A1c: 7.7% vs. 7.9%, P = 0.025) and a markedly lower prevalence of MAFLD with coexisting significant fibrosis (2.7% vs. 8.1%, P = 0.010), but a comparable prevalence of MAFLD without fibrosis. In multinomial logistic regression analysis, CSII therapy was associated with a ∼70%-lower risk of MAFLD with significant fibrosis (unadjusted odds ratio 0.32, 95% confidence interval 0.14–0.70; P = 0.004), but this association was no longer significant after adjustment for age, hemoglobin A1c and other potential confounders. Conclusion: The lower prevalence of MAFLD with coexisting significant fibrosis we observed in adults with T1DM using CSII therapy, compared to those using MDI therapy, is primarily mediated by inter-group differences in age.

Abstract

Background: We assessed whether hepatic steatosis with or without significant fibrosis (determined by validated non-invasive biomarkers) is associated with an increased 10-year estimated risk for cardiovascular disease (CVD) in people with type 1 diabetes mellitus (T1DM). Methods: We conducted a retrospective, multicenter, cross-sectional study involving 1,254 adults with established T1DM without pre-existing CVD. We used the hepatic steatosis index (HSI) and fibrosis (FIB)-4 index for non-invasively detecting hepatic steatosis (defined as HSI > 36), with or without coexisting significant fibrosis (defined as FIB-4 index ≥ 1.3 or < 1.3). We calculated the Steno type 1 risk engine and the atherosclerotic CVD (ASCVD) risk score to estimate the 10-year risk of developing a first fatal or nonfatal CVD event. Results: Using the Steno type 1 risk engine, a significantly greater proportion of patients with hepatic steatosis and significant fibrosis (n = 91) had a high 10-year estimated CVD risk compared to those with hepatic steatosis alone (n = 509) or without steatosis (n = 654) (75.8% vs. 23.2% vs. 24.9%, p < 0.001). After adjustment for sex, BMI, diabetes duration, hemoglobin A1c, chronic kidney disease, and lipid-lowering medication use, patients with hepatic steatosis and significant fibrosis had an increased 10-year estimated risk of developing a first fatal or nonfatal CVD event (adjusted-odds ratio 11.4, 95% confidence interval 3.54–36.9) than those without steatosis. We observed almost identical results using the ASCVD risk calculator. Conclusions: The 10-year estimated CVD risk is remarkably greater in T1DM adults with hepatic steatosis and significant fibrosis than in their counterparts with hepatic steatosis alone or without steatosis.

Abstract

This work aims at constraining the size, shape, and geometric albedo of the dwarf planet candidate 2002 MS4 through the analysis of nine stellar occultation events. Using multichord detection, we also studied the object's topography by analyzing the obtained limb and the residuals between observed chords and the best-fitted ellipse. We predicted and organized the observational campaigns of nine stellar occultations by 2002 MS4 between 2019 and 2022, resulting in two single-chord events, four double-chord detections, and three events with three to up to sixty-one positive chords. Using 13 selected chords from the 8 August 2020 event, we determined the global elliptical limb of 2002 MS4. The best-fitted ellipse, combined with the object's rotational information from the literature, constrains the object's size, shape, and albedo. Additionally, we developed a new method to characterize topography features on the object's limb. The global limb has a semi-major axis of 412 $\pm$ 10 km, a semi-minor axis of 385 $\pm$ 17 km, and the position angle of the minor axis is 121 $^\circ$ $\pm$ 16$^\circ$. From this instantaneous limb, we obtained 2002 MS4's geometric albedo and the projected area-equivalent diameter. Significant deviations from the fitted ellipse in the northernmost limb are detected from multiple sites highlighting three distinct topographic features: one 11 km depth depression followed by a 25$^{+4}_{-5}$ km height elevation next to a crater-like depression with an extension of 322 $\pm$ 39 km and 45.1 $\pm$ 1.5 km deep. Our results present an object that is $\approx$138 km smaller in diameter than derived from thermal data, possibly indicating the presence of a so-far unknown satellite. However, within the error bars, the geometric albedo in the V-band agrees with the results published in the literature, even with the radiometric-derived albedo.

Abstract

Aims: Patients with type 1 diabetes (T1D) have higher cardiovascular disease (CVD) risk compared to the general population. This observational study aims to evaluate sex-related differences in CVD prevalence and CVD risk estimates in a large cohort of T1D adults. Materials and methods: We conducted a multicenter, cross-sectional study involving 2,041 T1D patients (mean age 46 years; 44.9% women). In patients without pre-existing CVD (primary prevention), we calculated the Steno type 1 risk engine to estimate the 10-year risk of developing CVD events. Results: CVD prevalence (n=116) was higher in men than in women aged ≥55 years (19.2 vs 12.8%, p=0.036), but comparable between the two sexes in those aged <55 years (p=0.91). In patients without pre-existing CVD (n=1,925), mean 10-year estimated CVD risk was 15.4±0.4% without any significant sex difference. However, stratifying this patient group by age, the 10-year estimated CVD risk was significantly higher in men than in women until age 55 years (p<0.001), but this risk equalized after this age. Carotid-artery plaque burden was significantly associated with age ≥55 years and with a medium and high 10-year estimated CVD risk, without any significant sex difference. Diabetic retinopathy and sensory-motor neuropathy were also associated with higher 10-year CVD risk and female sex. Conclusions: Both men and women with T1D are at high CVD risk. The 10-year estimated CVD risk was higher in men aged <55 years than in women of similar age, but these sex differences disappeared at age ≥55 years, suggesting that female sex was no longer protective.

17. Spherical Wavefronts Improve MU-MIMO Spectral Efficiency When Using Electrically Large Arrays

Abstract

Modern multiple-input multiple-output (MIMO) communication systems are almost exclusively designed assuming locally plane wavefronts over antenna arrays. This is known as the far-field approximation and is soundly justified at sub-6-GHz frequencies at most relevant transmission ranges. However, when higher frequencies and shorter transmission ranges are used, the wave curvature over the array is no longer negligible, and arrays operate in the so-called radiative near-field region. This letter aims to show that the classical far-field approximation may significantly underestimate the achievable spectral efficiency of multi-user MIMO communications operating in the 30-GHz bands and above, even at ranges beyond the Fraunhofer distance. For planar arrays with typical sizes, we show that computing combining schemes based on the far-field model significantly reduces channel gain and spatial multiplexing capability. When the radiative near-field model is used, interference rejection schemes, such as the optimal minimum mean-square-error combiner, appear to be very promising, when combined with electrically large arrays, to meet the requirements of next-generation networks., QC 20230803

18. Vaginal immunology and microbiome composition: bacterial vaginosis in a macaque model

Abstract

Nonhuman primates (NHPs) are important animal models for the study of human health and disease. In particular, the use of NHPs to study the vaginal microbiome and susceptibility to infections (such as HIV and herpesvirus) is exceptionally valuable due to the similarity in anatomy and physiology. An important aspect to this is maintaining a healthy vaginal microbiome which then minimizes colonization by pathogens and resulting inflammation along the mucosa. In women, conditions such as bacterial vaginosis (BV) are frequently treated with antibiotics such as metronidazole or clindamycin. Due to the excessive use of antimicrobials in medicine and agriculture, alternative compounds and therapies are highly desired to treat infections. Approaches that have been developed and used for vaginal infections includes the use of natural antimicrobials such as essential oils, probiotics, and live cultures, which mimic and function like antibiotics but lack development of resistance like classic antibiotics. However, these approaches have been minimally studied in humans and animals. Effectiveness of essential oils are anecdotal at best. Microbiome manipulation on the other hand has been investigated more thoroughly. Novel products are being distributed for medical use and are monotherapies containing Lactobacillus which colonize the vaginal mucosa (Ali et al., 2020; Brichacek et al., 2013; Lagenaur, Sanders-Beer, et al., 2011). Unfortunately, these therapies have limitations due to durability and individual response in women. By evaluating the extent by which the NHP vaginal mucosa can be colonized with exogenously delivered bacteria, this animal model will highlight the NHP for use in translational studies which use essential oils and beneficial microbiome bacteria for vaginal delivery.

Abstract

The dataset contains data about amoxicillin quantification in milk and blood collected from lactating sows and in piglets’ blood, obtained in the framework of the Horizon 2020 IMI funded project ConcePTION. The main goal of ConcePTION is to establish a trusted ecosystem that can efficiently, systematically, and in an ethically responsible manner, generate and disseminate reliable evidence-based information regarding effects of medications used during pregnancy and breastfeeding to women and their healthcare providers. This will be achieved by generating, cataloguing, linking, collecting and analysing data from pharmacovigilance, modelling, routine healthcare, breastmilk samples through a large network. Within such framework, task 3.3 of WP3 aims at developing and characterising a relevant in vivo model for drug passage from maternal blood to human breast milk. Based on a preliminary scan of existing literature, the chosen species to perform such non-clinical trial was the porcine one due to high anatomical and physiological similarities. In the initial trial, amoxicillin was chosen as the first molecule for a variety of reasons including alignment with other Work Packages (WPs) performing studies on human breastmilk. Additionally, since amoxicillin is commonly used in pigs, the choice of the route of administration, the administration interval and the doses were supported by reliable background data.

Task 3.3 of WP3 aims at developing and characterising a relevant in vivo model for drug passage from maternal blood to human breast milk. Based on a preliminary scan of existing literature, the chosen species to perform such non-clinical trial was the porcine one due to high anatomical and physiological similarities. After performing a pilot study using amoxicillin, 3 additional in vivo trials were scheduled using metofmin, levocetirizine and velnfaxine. The chosen medicines were in agreement with other tasks and work packages within the project.

Rainfall precipitation maps are usually derived based on the measurements collected by classical weather devices, such as rain gauges and weather stations. This article aims to show the benefits obtained by opportunistic rainfall measurements based on signal strength measurements provided by commercial-grade satellite terminals (e.g., used in TV broadcasting). Abstract

Effective integration of terrestrial and non-terrestrial segments is one of the key research avenues in the design of current and future wireless communication networks. To this aim, modern communication-satellite constellations intend to attain sufficiently high throughput in terms of bit rate per unit area on the ground by rather aggressive patterns of spatial frequency reuse. This goal calls for on-board narrow-beam antennas, whose size turns out to be in many cases incompatible with the size/mass and accommodation constraints of the hosting satellite. This paper investigates the attainable performance of large distributed arrays of antennas implemented as the ensemble of a few to many simpler sub-antennas of smaller sizes, carried by one (small) satellite each. The sub-antennas can in their turn be implemented like (regular) 2D arrays of simple radiating elements, realizing an overall (distributed) antenna architecture that we call "Formation of Arrays" (FoA). The satellites that implement this radiating architecture need to be relatively close to each other and constitute a formation of flying objects, to be coordinated and controlled as a whole. In this paper, we develop a theoretical analysis of an FoA antenna, and we show how to take advantage of this new technology to improve network throughput in a multi-beam S-band mobile communication network with low-earth or geostationary orbiting satellites directly providing 5G-like communication services to hand-held user terminals., Comment: to appear on IEEE Trans. 24. MM Algorithms to Estimate Parameters in Continuous-time Markov Chains

Abstract

Continuous-time Markov chains (CTMCs) are popular modeling formalism that constitutes the underlying semantics for real-time probabilistic systems such as queuing networks, stochastic process algebras, and calculi for systems biology. Prism and Storm are popular model checking tools that provide a number of powerful analysis techniques for CTMCs. These tools accept models expressed as the parallel composition of a number of modules interacting with each other. The outcome of the analysis is strongly dependent on the parameter values used in the model which govern the timing and probability of events of the resulting CTMC. However, for some applications, parameter values have to be empirically estimated from partially-observable executions. In this work, we address the problem of estimating parameter values of CTMCs expressed as Prism models from a number of partially-observable executions. We introduce the class parametric CTMCs -- CTMCs where transition rates are polynomial functions over a set of parameters -- as an abstraction of CTMCs covering a large class of Prism models. Then, building on a theory of algorithms known by the initials MM, for minorization-maximization, we present iterative maximum likelihood estimation algorithms for parametric CTMCs covering two learning scenarios: when both state-labels and dwell times are observable, or just state-labels are. We conclude by illustrating the use of our technique in a simple but non-trivial case study: the analysis of the spread of COVID-19 in presence of lockdown countermeasures.

25. Propositional Logics for the Lawvere Quantale

Abstract

Lawvere showed that generalised metric spaces are categories enriched over $[0, \infty]$, the quantale of the positive extended reals. The statement of enrichment is a quantitative analogue of being a preorder. Towards seeking a logic for quantitative metric reasoning, we investigate three $[0,\infty]$-valued propositional logics over the Lawvere quantale. The basic logical connectives shared by all three logics are those that can be interpreted in any quantale, viz finite conjunctions and disjunctions, tensor (addition for the Lawvere quantale) and linear implication (here a truncated subtraction); to these we add, in turn, the constant $1$ to express integer values, and scalar multiplication by a non-negative real to express general affine combinations. Quantitative equational logic can be interpreted in the third logic if we allow inference systems instead of axiomatic systems. For each of these logics we develop a natural deduction system which we prove to be decidably complete w.r.t. the quantale-valued semantics. The heart of the completeness proof makes use of the Motzkin transposition theorem. Consistency is also decidable; the proof makes use of Fourier-Motzkin elimination of linear inequalities. Strong completeness does not hold in general, even (as is known) for theories over finitely-many propositional variables; indeed even an approximate form of strong completeness in the sense of Pavelka or Ben Yaacov -- provability up to arbitrary precision -- does not hold. However, we can show it for theories axiomatized by a (not necessarily finite) set of judgements in normal form over a finite set of propositional variables when we restrict to models that do not map variables to $\infty$; the proof uses Hurwicz's general form of the Farkas' Lemma.

26. A comparison of numerical approaches for statistical inference with stochastic models

Abstract

Due to our limited knowledge about complex environmental systems, our predictions of their behavior under different scenarios or decision alternatives are subject to considerable uncertainty. As this uncertainty can often be relevant for societal decisions, the consideration, quantification and communication of it is very important. Due to internal stochasticity, often poorly known influence factors, and only partly known mechanisms, in many cases, a stochastic model is needed to get an adequate description of uncertainty. As this implies the need to infer constant parameters, as well as the time-course of stochastic model states, a very high-dimensional inference problem for model calibration has to be solved. This is very challenging from a methodological and a numerical perspective. To illustrate aspects of this problem and show options to successfully tackle it, we compare three numerical approaches: Hamiltonian Monte Carlo, Particle Markov Chain Monte Carlo, and Conditional Ornstein-Uhlenbeck Sampling. As a case study, we select the analysis of hydrological data with a stochastic hydrological model. We conclude that the performance of the investigated techniques is comparable for the analyzed system, and that also generality and practical considerations may be taken into account to guide the choice of which technique is more appropriate for a particular application.

28. Development of a porcine lactation model for the evaluation of mammary transfer of exogenous molecules - A contribution from the conception project

Abstract

Around 5 million women give birth each year in Europe and, while breastfeeding, the majority of them may need to take medications, either occasionally or continuously. Unfortunately, there is often scarce evidence of trustworthy information about how a specific molecule might affect the physiology of lactation. This is the reason that brought a European public-private partnership to fund the development of a reliable platform to provide women and health-care professionals a helpful instrument to reduce uncertainty about the effects of medication used during breastfeeding. On April 1st 2019, the ConcePTION project (Grant Agreement n°821520) started to develop such envisaged platform. The 3rd Work Package was in charge of the validation of in vitro, in vivo and in silico lactation models. Between the numerous species currently used in preclinical studies, pigs’ similarities with humans’ anatomy, physiology and genomics make them extremely useful as translational models, when proper veterinary expertise is applied. The ASA team from the University of Bologna, went first to characterize the translational lactation model using the swine species, chosen upon literature review. The aim of this work was to lay the foundations of a porcine lactation model that could be suitable for application within pharmaceutical tests, to study drug transfer through milk prior approval and commercialization. The obtained results highlighted both strengths and critical points of the study design, allowing a significant improvement in the knowledge of pharmacokinetic physiology in lactating mammals. Lastly, this project allowed the assessment of microbial changes in gut resident bacteria of newborns through an innovative in vitro colonic model. Indeed, even if there were no evident adverse effects determined by drug residues in milk, possible alterations in the delicate microbial ecology of newborns’ gastrointestinal tract was considered pivotal, giving its possible impact on the individual

29. DFT-Based Channel Estimation for Holographic MIMO

Abstract

Holographic MIMO (hMIMO) systems with a massive number of individually controlled antennas N make minimum mean square error (MMSE) channel estimation particularly challenging, due to its computational complexity that scales as $N^3$ . This paper investigates uniform linear arrays and proposes a low-complexity method based on the discrete Fourier transform (DFT) approximation, which follows from replacing the covariance matrix by a suitable circulant matrix. Numerical results show that, already for arrays with moderate size (in the order of tens of wavelengths), it achieves the same performance of the optimal MMSE, but with a significant lower computational load that scales as $N \log N$. Abstract

An 8-month-old female Lagotto Romagnolo dog was presented for a 1-month history of an initial severe reluctance to move, rapidly progressing to a marked stiff gait and progressive muscular weakness and evolving to tetraparesis, which persuaded the owner to request euthanasia. A primary muscle pathology was supported by necropsy and histopathological findings. Macroscopically, the muscles were moderately atrophic, except for the diaphragm and the neck muscles, which were markedly thickened. Histologically, all the skeletal muscles examined showed atrophy, hypertrophy, necrosis with calcification of the fibers, and mild fibrosis and inflammation. On immunohistochemistry, all three dystrophin domains and sarcoglycan proteins were absent. On Western blot analysis, no band was present for delta sarcoglycan. We sequenced the genome of the affected dog and compared the data to more than 900 control genomes of different dog breeds. Genetic analysis revealed a homozygous private protein-changing variant in the SGCD gene encoding delta- sarcoglycan in the affected dog. The variant was predicted to induce a SGCD:p.(Leu242Pro) change in the protein. In silico tools predicted the change to be deleterious. Other 770 Lagotto Romagnolo dogs were genotyped for the variant and all found to be homozygous wild type. Based on current knowledge of gene function in other mammalian species, including humans, hamsters, and dogs, we propose the SGCD missense variant as the causative variant of the observed form of muscular dystrophy in the index case. The absence of the variant allele in the Lagotto Romagnolo breeding population indicates a rare allele that has appeared recently.

Abstract

We develop a theory for the commutative combination of quantitative effects, their tensor, given as a combination of quantitative equational theories that imposes mutual commutation of the operations from each theory. As such, it extends the sum of two theories, which is just their unrestrained combination. Tensors of theories arise in several contexts; in particular, in the semantics of programming languages, the monad transformer for global state is given by a tensor. We show that under certain assumptions on the quantitative theories the free monad that arises from the tensor of two theories is the categorical tensor of the free monads on the theories. As an application, we provide the first algebraic axiomatizations of labelled Markov processes and Markov decision processes. Apart from the intrinsic interest in the axiomatizations, it is pleasing they are obtained compositionally by means of the sum and tensor of simpler quantitative equational theories.

Abstract

We introduce polynomial couplings, a generalization of probabilistic couplings, to develop an algorithm for the computation of equivalence relations which can be interpreted as a lifting of probabilistic bisimulation to polynomial differential equations, a ubiquitous model of dynamical systems across science and engineering. The algorithm enjoys polynomial time complexity and complements classical partition-refinement approaches because: (a) it implements a local exploration of the system, possibly yielding equivalences that do not necessarily involve the inspection of the whole system of differential equations; (b) it can be enhanced by up-to techniques; and (c) it allows the specification of pairs which ought not to be included in the output. Using a prototype, these advantages are demonstrated on case studies from systems biology for applications to model reduction and comparison. Notably, we report four orders of magnitude smaller runtimes than partition-refinement approaches when disproving equivalences between Markov chains.

Abstract

The lncRNA metastasis-associated lung adenocarcinoma transcript 1 (MALAT1) promotes growth and progression in prostate cancer (PCa); however, little is known about its possible impact in PCa metabolism. The aim of this work has been the assessment of the metabolic reprogramming associated with MALAT1 silencing in human PCa cells and in an ex vivo model of organotypic slice cultures (OSCs). Cultured cells and OSCs derived from primary tumors were transfected with MALAT1 specific gapmers. Cell growth and survival, gene profiling, and evaluation of targeted metabolites and metabolic enzymes were assessed. Computational analysis was made considering expression changes occurring in metabolic markers following MALAT1 targeting in cultured OSCs. MALAT1 silencing reduced expression of some metabolic enzymes, including malic enzyme 3, pyruvate dehydrogenase kinases 1 and 3, and choline kinase A. Consequently, PCa metabolism switched toward a glycolytic phenotype characterized by increased lactate production paralleled by growth arrest and cell death. Conversely, the function of mitochondrial succinate dehydrogenase and the expression of oxidative phosphorylation enzymes were markedly reduced. A similar effect was observed in OSCs. Based on this, a predictive algorithm was developed aimed to predict tumor recurrence in a subset of patients. MALAT1 targeting by gapmer delivery restored normal metabolic energy pathway in PCa cells and OSCs.

Abstract

Euclidean Markov decision processes are a powerful tool for modeling control problems under uncertainty over continuous domains. Finite state imprecise, Markov decision processes can be used to approximate the behavior of these infinite models. In this paper we address two questions: first, we investigate what kind of approximation guarantees are obtained when the Euclidean process is approximated by finite state approximations induced by increasingly fine partitions of the continuous state space. We show that for cost functions over finite time horizons the approximations become arbitrarily precise. Second, we use imprecise Markov decision process approximations as a tool to analyse and validate cost functions and strategies obtained by reinforcement learning. We find that, on the one hand, our new theoretical results validate basic design choices of a previously proposed reinforcement learning approach. On the other hand, the imprecise Markov decision process approximations reveal some inaccuracies in the learned cost functions.

Abstract

Objectives: Few studies have analyzed factors associated with delirium subtypes. In this study, we investigate factors associated with subtypes of delirium only in patients with dementia to provide insights on the possible prevention and treatments. Design: This is a cross-sectional study nested in the “Delirium Day” study, a nationwide Italian point-prevalence study. Setting and Participants: Older patients admitted to 205 acute and 92 rehabilitation hospital wards. Measures: Delirium was evaluated with the 4-AT and the motor subtypes with the Delirium Motor Subtype Scale. Dementia was defined by the presence of a documented diagnosis in the medical records and/or prescription of acetylcholinesterase inhibitors or memantine prior to admission. Results: Of the 1057 patients with dementia, 35% had delirium, with 25.6% hyperactive, 33.1% hypoactive, 34.5% mixed, and 6.7% nonmotor subtype. There were higher odds of having venous catheters in the hypoactive (OR 1.82, 95% CI 1.18-2.81) and mixed type of delirium (OR 2.23, CI 1.43-3.46), whereas higher odds of urinary catheters in the hypoactive (OR 2.91, CI 1.92-4.39), hyperactive (OR 1.99, CI 1.23-3.21), and mixed types of delirium (OR 2.05, CI 1.36-3.07). We found higher odds of antipsychotics both in the hyperactive (OR 2.87, CI 1.81-4.54) and mixed subtype (OR 1.84, CI 1.24-2.75), whereas higher odds of antibiotics was present only in the mixed subtype (OR 1.91, CI 1.26-2.87). Conclusions and Implications: In patients with dementia, the mixed delirium subtype is the most prevalent followed by the hypoactive, hyperactive, and nonmotor subtype. Motor subtypes of delirium may be triggered by clinical factors, including the use of venous and urinary catheters, and the use of antipsychotics. Future studies are necessary to provide further insights on the possible pathophysiology of delirium in patients with dementia and to address the optimization of the management of potential risk factors.

Abstract

Purpose: In presence of indeterminate lesions by fine needle aspiration (FNA), thyroid cancer cannot always be easily diagnosed by conventional cytology. As a consequence, unnecessary removal of thyroid gland is performed in patients without cancer based on the lack of optimized diagnostic criteria. Aim of this study is identifying a molecular profile based on long noncoding RNAs (lncRNAs) expression capable to discriminate between benign and malignant nodules. Methods: Patients were subjected to surgery (n = 19) for cytologic suspicious thyroid nodules or to FNA biopsy (n = 135) for thyroid nodules suspicious at ultrasound. Three thyroid-specific genes (TG, TPO, and NIS), six cancer-associated lncRNAs (MALAT1, NEAT1, HOTAIR, H19, PVT1, MEG3), and two housekeeping genes (GAPDH and P0) were analyzed using Droplet Digital PCR (ddPCR). Results: Based on higher co-expression in malignant (n = 11) but not in benign (n = 8) nodules after surgery, MALAT1, PVT1 and HOTAIR were selected as putative cancer biomarkers to analyze 135 FNA samples. Cytological and histopathological data from a subset of FNA patients (n = 34) were used to define a predictive algorithm based on a Naïve Bayes classifier using co-expression of MALAT1, PVT1, HOTAIR, and cytological class. This classifier exhibited a significant separation capability between malignant and benign nodules (P < 0.0001) as well as both rule in and rule out test potential with an accuracy of 94.12% and a negative predictive value (NPV) of 100% and a positive predictive value (PPV) of 91.67%. Conclusions: ddPCR analysis of selected lncRNAs in FNA biopsies appears a suitable molecular tool with the potential of improving diagnostic accuracy.

Abstract

Prostate cancer (PCa) is a sex-steroid hormone-dependent cancer in which estrogens play a critical role in both initiation and progression. Recently, several long non-coding RNAs (lncRNAs) have been associated with PCa and are supposedly playing a pivotal role in the biology and progression of this type of cancer. In this review, we focused on some lncRNAs that are known for their androgen and estrogen transcriptional responsiveness in PCa. Specifically, we summarized recent pieces of evidence about lncRNAs NEAT1, H19, MALAT1, and HOTAIR, in estrogen signaling, emphasizing their role in PCa progression and the acquisition of a castration-resistant phenotype. Here, the reader will find information about lncRNAs present in estrogen-dependent transcriptional complexes. The potential role of lncRNA/estrogen signaling as a novel pathway for PCa treatment will be discussed

39. Anti-retinal IgG antibodies in patients with early and advanced type 2 macular telangiectasia

Abstract

Type 2 idiopathic macular telangiectasia (MacTel-2) is a progressive adult-onset macular disease associated with bilateral perifoveal vascular changes, Muller cell degeneration and increased blood-retinal barrier permeability. The pathophysiological mechanisms of MacTel-2 remain unclear, however it was previously reported that anti-retinal antibodies in MacTel-2 patients are a significant feature of the disease. In this study, we aimed to compare the prevalence of anti-retinal antibodies in patients MacTel-2, healthy controls and patients with other retinal diseases. MacTel-2 patients diagnosed with multimodal imaging were enrolled and their disease severities were graded using spectral-domain optical coherence tomography. For comparison, patients with age-related macular degeneration (AMD), inherited retinal diseases (IRDs) or no retinal disease (healthy controls) were recruited as controls. Blood serum samples were screened for immunoglobulin G anti-retinal antibodies by western blotting, followed by densitometry analysis. Odds ratios (OR) with 95% confidence intervals (CI) were calculated and p < 0.05 considered statistically significant. Overall, anti-retinal antibody-positive cases were older (64 ± 15 vs 53 ± 17 years, p < 0.001) and females were more likely to develop anti-retinal antibodies (OR: 2.41, CI: 1.12–5.18). The frequency of anti-retinal antibody detection in MacTel-2 patients (n = 42, 36%) was not significantly different from healthy controls (n = 52, 25%) or IRD patients (n = 18, 25%) and the majority of MacTel-2 patients had no anti-retinal antibodies. In contrast, the frequency of anti-retinal antibody detection was significantly higher in patients with AMD (n = 15, 73%, p < 0.001). The lack of a greater anti-retinal antibody frequency or specificity in the MacTel-2 cohort suggests that antibody mediated immunological mechanisms may play a less significant role in MacTel-2 disease pathogenesis.

Abstract

Papillomavirus associated tumors are well recognized entities in humans as well as in animals. Here is reviewed the current understanding of human papillomavirus (HPV) associated cancers to better understand the oncogenic mechanisms of Equine papillomavirus (EcPV) and Bovine Papillomavirus (BPV) in horses. In the first part of this study the interactions between Equine papillomavirus 2 (EcPV-2) and cell cycle proteins are discussed. EcPV-2 has been recognized as the cause of genital squamous cell carcinomas (SCCs) in horses, but the exact mechanism of carcinogenesis is not fully understood. The aim of the first part of this study is to assess the expression of cell cycle proteins p53, p16, pRB and Cyclin D1 in a series of equine SCCs and papillomas. Results confirm the role of EcPV-2 in the pathogenesis of genital SCCs. Moreover, in a small subset of ocular SCCs, EcPV-2 was detected for the first time. By immunohistochemistry, p53 was mostly expressed in ocular SCCs with a suprabasal localization. Regarding p16, overexpression was associated with increased mitotic index but not with viral infection. Investigation on pRB and Cyclin D1 proteins did not show significant correlation with other variables. The second part of this study is focused on the carcinogenetic mechanisms of BPV in equine sarcoids. The aim of the second part of this study was to characterize the typical histomorphological features of equine sarcoids, assess the expression of cell cycle proteins and Ki-67 proliferation index. Our results confirm that the typical histological features of sarcoids cannot be used to correctly classify the clinical types. Moreover, in a subset of sarcoids low pRB-Cyclin D1 scores were associated with simultaneous high p16 expression. The Ki-67 proliferation index confirm the low proliferative activity of sarcoids, except for tumors displaying a fascicular pattern. Finally, a subset of sarcoids recurred after excision.

42. Sum and Tensor of Quantitative Effects

Abstract

Inspired by the seminal work of Hyland, Plotkin, and Power on the combination of algebraic computational effects via sum and tensor, we develop an analogous theory for the combination of quantitative algebraic effects. Quantitative algebraic effects are monadic computational effects on categories of metric spaces, which, moreover, admit an algebraic presentation in the form of quantitative equational theories, a logical framework introduced by Mardare, Panangaden, and Plotkin that generalises equational logic to account for a concept of approximate equality. As our main result, we show that the sum and tensor of two quantitative equational theories correspond to the categorical sum (i.e., coproduct) and tensor, respectively, of their effects qua monads. We further give a theory of quantitative effect transformers based on these two operations, essentially providing quantitive analogues to the following monad transformers due to Moggi: exception, resumption, reader, and writer transformers. Finally, as an application we provide the first quantitative algebraic axiomatizations to the following coalgebraic structures: Markov processes, labelled Markov processes, Mealy machines, and Markov decision processes, each endowed with their respective bisimilarity metrics. Apart from the intrinsic interest in these axiomatizations, it is pleasing they have been obtained as the composition, via sum and tensor, of simpler quantitative equational theories.

Abstract

GluN3A is an atypical glycine-binding subunit of NMDA receptors (NMDARs) whose actions in the brain are mostly unknown. Here, we show that the expression of GluN3A subunits controls the excitability of mouse adult cortical and amygdalar circuits via an unusual signaling mechanism involving the formation of excitatory glycine GluN1/GluN3A receptors (eGlyRs) and their tonic activation by extracellular glycine. eGlyRs are mostly extrasynaptic and reside in specific neuronal populations, including the principal cells of the basolateral amygdala (BLA) and SST-positive interneurons (SST-INs) of the neocortex. In the BLA, tonic eGlyR currents are sensitive to fear-conditioning protocols, are subject to neuromodulation by the dopaminergic system, and control the stability of fear memories. In the neocortex, eGlyRs control the in vivo spiking of SST-INs and the behavior-dependent modulation of cortical activity. GluN3A-containing eGlyRs thus represent a novel and widespread signaling modality in the adult brain, with attributes that strikingly depart from those of conventional NMDARs.

Abstract

Type 2 idiopathic macular telangiectasia (MacTel-2) is a progressive adult-onset macular disease associated with bilateral perifoveal vascular changes, Muller cell degeneration and increased blood-retinal barrier permeability. The pathophysiological mechanisms of MacTel-2 remain unclear, however it was previously reported that anti-retinal antibodies in MacTel-2 patients are a significant feature of the disease. In this study, we aimed to compare the prevalence of anti-retinal antibodies in patients MacTel-2, healthy controls and patients with other retinal diseases. MacTel-2 patients diagnosed with multimodal imaging were enrolled and their disease severities were graded using spectral-domain optical coherence tomography. For comparison, patients with age-related macular degeneration (AMD), inherited retinal diseases (IRDs) or no retinal disease (healthy controls) were recruited as controls. Blood serum samples were screened for immunoglobulin G anti-retinal antibodies by western blotting, followed by densitometry analysis. Odds ratios (OR) with 95% confidence intervals (CI) were calculated and p < 0.05 considered statistically significant. Overall, anti-retinal antibody-positive cases were older (64 ± 15 vs 53 ± 17 years, p < 0.001) and females were more likely to develop anti-retinal antibodies (OR: 2.41, CI: 1.12–5.18). The frequency of anti-retinal antibody detection in MacTel-2 patients (n = 42, 36%) was not significantly different from healthy controls (n = 52, 25%) or IRD patients (n = 18, 25%) and the majority of MacTel-2 patients had no anti-retinal antibodies. In contrast, the frequency of anti-retinal antibody detection was significantly higher in patients with AMD (n = 15, 73%, p < 0.001). The lack of a greater anti-retinal antibody frequency or specificity in the MacTel-2 cohort suggests that antibody mediated immunological mechanisms may play a less significant role in MacTel-2 disease pathogenesis.

46. Verified Probabilistic Policies for Deep Reinforcement Learning

Abstract

Deep reinforcement learning is an increasingly popular technique for synthesising policies to control an agent's interaction with its environment. There is also growing interest in formally verifying that such policies are correct and execute safely. Progress has been made in this area by building on existing work for verification of deep neural networks and of continuous-state dynamical systems. In this paper, we tackle the problem of verifying probabilistic policies for deep reinforcement learning, which are used to, for example, tackle adversarial environments, break symmetries and manage trade-offs. We propose an abstraction approach, based on interval Markov decision processes, that yields probabilistic guarantees on a policy's execution, and present techniques to build and solve these models using abstract interpretation, mixed-integer linear programming, entropy-based refinement and probabilistic model checking. 47. The Bounded Beam Search algorithm for the Block Relocation Problem

Abstract

In this paper we deal with the restricted Block Relocation Problem. We present a new lower bound and a heuristic approach for the problem. The proposed lower bound can be computed in polynomial time and it is provably better than some previously known lower bounds. We use it within a bounded beam search algorithm to solve the Block Relocation Problem and show that the considered heuristic approach outperforms the other existing algorithms on most of the instances in the literature. In order to test the approaches on real-size dimensions, new large instances of the Block Relocation Problem are also introduced.

48. Molecular analysis of Papillomavirus-induced cutaneous tumors in equids

Abstract

Papillomavirus associated tumors are well recognized entities in humans as well as in animals. Here is reviewed the current understanding of human papillomavirus (HPV) associated cancers to better understand the oncogenic mechanisms of Equine papillomavirus (EcPV) and Bovine Papillomavirus (BPV) in horses. In the first part of this study the interactions between Equine papillomavirus 2 (EcPV-2) and cell cycle proteins are discussed. EcPV-2 has been recognized as the cause of genital squamous cell carcinomas (SCCs) in horses, but the exact mechanism of carcinogenesis is not fully understood. The aim of the first part of this study is to assess the expression of cell cycle proteins p53, p16, pRB and Cyclin D1 in a series of equine SCCs and papillomas. Results confirm the role of EcPV-2 in the pathogenesis of genital SCCs. Moreover, in a small subset of ocular SCCs, EcPV-2 was detected for the first time. By immunohistochemistry, p53 was mostly expressed in ocular SCCs with a suprabasal localization. Abstract

The research from which this reflection article arose had the objective of taking a look at an idea of ​​territory that has taken a lot of strength in Colombia; this is a reference about to the concept of amazorinoquia. The research methodology focused on documentary bibliographic analysis, for which an investigation was made on the three key concepts developed in this study. As a result, it was perfected to delve into three separate concepts, development, Amazonia and Orinoquía, with the aim of observing how a territorial space can become a concept in a diversified economic objective. As a conclusion, it will be improved to generate a critical reflection in which the various concepts of territory are mixed, towards a possible justification for its exploitation., O objetivo da pesquisa a partir da qual este artigo surgiu foi dar uma olhada numa idéia de território que ganhou muita força na Colômbia; ele se refere ao conceito de Amazorinoquía. A metodologia de pesquisa se concentrou na análise bibliográfica documental, para a qual foi feita uma pesquisa - de várias fontes - sobre os conceitos-chave desenvolvidos neste estudo. Como resultado, três conceitos distintos - desenvolvimentismo, Amazônia e Orinoco - foram explorados em profundidade, com o objetivo de observar como um espaço territorial pode se tornar um objetivo econômico diversificado. Em conclusão, foi possível gerar uma reflexão crítica sobre a forma como os diferentes conceitos de território são misturados, para uma possível justificativa de sua exploração., La investigación de la cual surgió el presente artículo de reflexión tuvo por objetivo dar una mirada a una idea de territorio que ha tomado mucha fuerza en Colombia; se hace referencia al concepto de Amazorinoquía. La metodología de investigación se centró en el análisis bibliográfico documental, para lo cual se hizo una indagación —desde diversas fuentes—, sobre los conceptos claves desarrollados en el presente estudio Como resultado, se logró profundizar sobre tres conceptos por separado, desarrollismo, Amazonía y Orinoquía, ello con el objetivo de observar la forma como un espacio territorial, puede llegar a constituirse como concepto en un objetivo económico diversificado. Abstract

Perisomatic inhibition of pyramidal neurons (PNs) coordinates cortical network activity during sensory processing, and this role is mainly attributed to parvalbumin-expressing basket cells (BCs). However, cannabinoid receptor type 1 (CB1)-expressing interneurons are also BCs, but the connectivity and function of these elusive but prominent neocortical inhibitory neurons are unclear. We find that their connectivity pattern is visual area specific. Persistently active CB1 signaling suppresses GABA release from CB1 BCs in the medial secondary visual cortex (V2M), but not in the primary visual cortex (V1). Accordingly, in vivo, tonic CB1 signaling is responsible for higher but less coordinated PN activity in the V2M than in the V1. These differential firing dynamics in the V1 and V2M can be captured by a computational network model that incorporates visual-area-specific properties. Our results indicate a differential CB1-mediated mechanism controlling PN activity, suggesting an alternative connectivity scheme of a specific GABAergic circuit in different cortical areas.
